In order to determine ileal losses of nitrogen (N) and amino acids (AA) and the coefficients of apparent and true ileal digestibility (AID, TID) of N and AA from casein in piglets and pigs, two experiments were conducted. In Experiment 1, 24 piglets were used. The piglets were weaned at 17 days of age, weighing 6.4 kg and cannulated at terminal ileum. Ileal digesta was collected at 28-29 and 35-36 days of age in period 1 and 2, respectively. Feed intake was 150 and 300 g x d(-1) during the first and second period. In Experiment 2, 16 castrates weighing 52.5 kg and cannulated at terminal ileum were used. The intake level of digestible energy was 2.5 times their maintenance requirement. The experiment lasted 7 days and ileal digesta was collected on day 6-7. Treatments consisted of four levels of N from casein: 8, 16, 24 and 32 g N x kg(-1) feed, respectively. Results showed that N level did not increase N or AA ileal losses. In piglets, N and AA ileal losses were similar between periods, except for period 2, where losses per kg DMI were about 47 and 64% higher for glycine and proline, respectively (p < 0.05). When ileal losses from pigs and piglets were compared, piglets had higher (p < 0.05) ileal losses of N and AA (excepted glutamic acid and alanine). A lower (p < 0.05) AID was observed in piglets in period 2 for N, methionine, glutamic acid, glycine and proline. With exception of glycine in pigs, all values for TID of N and AA of casein were superior to 0.90. Piglets had higher TID of N, leucine, isoleucine, valine and phenylalanine. These results showed that piglets have higher ileal losses than pigs.  相似文献

Abstract

Four pigs prepared with re-entrant cannulas in the proximal duodenum and terminal ileum were used to study flow rates of total digesta, insoluble dry matter, nitrogen, and amino acids entering and leaving the small intestine. The pigs received a semipurified diet, a hard wheat diet, or a soft wheat diet. These were approximately isonitrogenous. A higher rate of passage of digesta through the proximal duodenum and terminal ileum were measured in pigs receiving the hard wheat diet. Peak flow of digesta at the duodenum of all pigs occurred at 1 h post feeding. Peak flow of digesta at the ileum occurred at 9 h post feeding on the soft wheat diet, but somewhat earlier on the hard wheat and semipurified diet. More nitrogen and essential amino acids flowed in the solid fraction of duodenal digesta during the first 2 h post feeding for the wheat diets and 4 h post feeding for the semipurified diet. It was concluded that flow rate of most nutrients from the stomach and through the small intestine of pigs is modified by the composition and texture of the food ingested. It is postulated that efficiency of mixing of digesta with digestive secretions in the stomach is a major factor influencing rate of flow.  相似文献

A 4 ‐ 4 Latin square design was used to determine ileal apparent digestibility of amino acids (AAs) in corn, soybean meal, feather meal and dried whey in young pigs. The data were then to be used in formulating a basal diet for studies on AA metabolism in young pigs. Eight castrates T-cannulated at terminal ileum (average initial body weight 12.5 ± 0.62kg) were divided into 4 groups on the basis of body weight and transferred to individual metabolism crates. They were then fed four experimental diets containing the four feedstuffs to be tested (corn, soybean meal, feather meal and dried whey). The trial lasted 20 days, which included 4 five-day periods for ileal digesta collection. It was found that the digestibility of the AAs was similar to that reported in literature. Based on the findings a basal diet for this research was formulated according to an ideal protein model for the 10 to 20kg piglet, on the basis of digestible AAs and containing 14.13MJ/kg digestible energy, 18.22% crude protein, 1.04% digestible lysine and 0.23% digestible histidine.  相似文献

Sixteen male growing pigs of about 24kg BW were fitted with both a duodenal re-entrant and a post-valve T-shaped cannula inserted in the caecum. The animals were divided into four groups. Each group received one of the following diets: corn starch-soybean protein isolate-based diet without (diet C) and with carboxymethylcellulose (diet CMC) or a rye-wheat-based diet without (diet RW) and with xylanase addition (diet RWX). The diets provided similar levels of apparent precaecal digestible crude protein (CP), lysine, methionine+cystine, threonine and tryptophan. Additionally, [15N]-yeast was given with the diets during the first 10 days of the experiment. For estimation of digesta viscosity, N-flow of dietary and endogenous origin, apparent precaecal digestibilities of dry matter (DM), CP, amino acids and non starch polysaccharides (NSP) (only in pigs fed diets RW and RWX), ileal and duodenal digesta were quantitatively collected on day 16 and 17, respectively. The endogenous N-proportion was measured by the ratio of 15N enrichment in the digesta and urine. The duodenal and ileal digesta supernatant viscosity increased as carboxymethylcellulose was included into the diet. Xylanase addition to the rye-wheat based diet reduced the viscosity in the ileal digesta. There were no differences in precaecal digestibilities of DM, CP and amino acids between diet C and CMC. The precaecal digestibilities of DM and soluble and insoluble NSP increased from 69.5% to 73.9%, from 1.3% to 47.9% and from 17.0% to 35.4%, respectively, as xylanase was added to the rye-wheat-based diet. The apparent precaecal digestibility of most essential amino acids increased by 2 to 5 percent units. The amounts of endogenous N at the duodenal level were estimated to be 158, 233, 313 and 276mg per 12h per kg0.75 BW of pigs fed diets C, CMC, RW and RWX, respectively. The corresponding values at the ileal level were 95, 107, 164 and 150mg per 12h per kg0.75 BW. For endogenous N amounts, significant differences were observed between diets C and CMC (duodenum) and also between semi-purified and cereal-based diets (duodenum and ileum). Methodological aspects for the estimation of endogenous N using the isotope dilution technique are discussed. Obviously, the digesta viscosity per se does not affect the nutrient absorption and endogenous N flow within the small intestine of pigs. Other properties of complex dietary fibre, digesta passage rate or bacterial activity probably contribute to the observed changes.  相似文献

This study used a digesta transfer protocol to determine the site of absorption of lysine synthesized by the gastrointestinal microflora of pigs. Eight pigs were used, four with reentrant cannulas in the terminal ileum, two with simple T cannulas in the terminal ileum, and two intact. All pigs were given, for 5 days, the same low-protein diet that included fermentable carbohydrates. The diet of two pigs with reentrant cannulas (donor) and of the two intact (control) pigs was supplemented with (15)NH(4)Cl. The two other pigs with reentrant cannulas (acceptor pigs) and those with simple cannulas (used to supply unlabeled digesta) were given the same diet but unlabeled NH(4)Cl. Ileal digesta were collected continuously from all of the reentrant cannulas and kept on ice. All digesta from each donor pig were reheated and returned to the distal cannula of its companion acceptor, whose ileal digesta were discarded. Unlabeled ileal digesta from the pigs with simple cannulas were instilled into the distal cannulas of the donor pigs. At the end of the experiment, the average (15)N enrichment in the plasma free lysine of control pigs was 0.0407 atom % excess (APE); that of donor pigs was 0.0322 APE (79% of controls), whereas that of acceptor pigs was only 0.0096 APE (24% of controls). Due to nitrogen recycling, acceptor pigs had labeled lysine in the digesta of the stomach and small intestine, and donor pigs had labeled lysine in the digesta of the large intestine. If account is taken of the higher (15)N enrichment of microbial lysine in the large compared with the small intestine, it can be estimated that >90% of the absorption of microbial lysine took place in the small intestine.  相似文献

Six female rabbits fitted with a simple glass cannula in terminal ileum and a further 6 non-cannulated rabbits were used to perform digesta flow and transit measurements. The animals received ad libitum a diet based mainly on lucerne meal. Flow and transit measurements were carried out using two particulate markers: ytterbium (Yb) fixed on lucerne meal cell-walls by soaking, and chromium (Cr) fixed by mordancing. Both markers were incorporated in the same diet before pelleting. The digestibility coefficient or total mean retention time (MRTt) in the whole tract was not affected by cannulation. MRT values between cecum and rectum could be obtained in cannulated rabbits: 9.6 h. The MRTt values estimated with Yb were higher than those obtained with Cr because the size of particles labelled with Cr was probably greater and thus the particles were excreted more rapidly in hard feces. Flow measurements were not affected by the choice of marker, confirming indirectly the validity of ileal digesta samples. These first ileal flow measures obtained on cannulated rabbits indicate that about half (49%) of the digestible organic matter and two-thirds of the digestible crude protein apparently disappeared before the cecum. Cell-walls (CW), determined as NDF fraction, were totally recovered at the terminal ileum, although hemicellulose (NDF-ADF) digestion could occur in both the small intestine and the cecocolic parts. These results are discussed in relation to the specificity of the Van Soest method for digesta cell-wall analysis.  相似文献

The ileal digestibility coefficient (CSID) of amino acids (AA) and crude protein (CP) in 40 feedstuffs for growing pigs were determined with the protein-free (PF) and enzyme-hydrolyzed casein (EHC) methods. The 40 feedstuffs that were used earlier were 10 samples of cereals and cereal by-products, 12 samples of legumes, 6 samples of animal protein feedstuff and 12 samples of oil seed meals. Six growing pigs (initial body weight of 35 ± 1.5 kg), fitted with T-cannula at the terminal ileum, were randomly allocated to either a PF or a EHC diet according to a crossover design during two ileal digesta collection periods. In each period, pigs were adjusted to the experimental diets for 5 days. On days 6 and 8, ileal digesta were collected continuously for 24 h to determine ileal endogenous AA and CP losses. Pigs fed the EHC diet had a higher ileal flow of endogenous CP and of most of AA (P<0.05) than pigs fed the PF diet. Among the ileal endogenous AA flows (g/kg dry matter intake for pigs), methionine excretion was the lowest in pigs (0.09 and 0.25 g/kg dry matter intake) fed the PF and EHC diet, respectively, whereas glutamate (1.83 g/kg dry matter intake) and proline (1.22 g/kg dry material intake) excretion were the highest in pigs fed the EHC and the PF diet, respectively. Endogenous losses of CP and AA determined in the current study and previously published data on apparent ileal digestibility [Yin, Y.L., Huang, R.L., Zhong, H.Y., Chen, C.M., Li, T.J., Pan, Y.F., 1993. Nutritive value of feedstuffs and diets for pigs: 1. Chemical composition, apparent ileal and fecal digestibilities. Anim. Feed Sci. Technol. 44, 1–27] were used to calculate CSID coefficients. For most cereals and cereal by-products, the CSID coefficients of CP determined by the EHC method were higher than those determined by the PF method. Arginine, lysine, methionine, threonine, valine, alanine, aspartate, glutamate, glycine, proline and serine in some cereals and cereal by-products; methionine, valine, alanine and proline in some legumes; and methionine, alaline and proline in some oilseed meals had higher CSID determined by the EHC method than the PF method indicating that there are methodological differences when evaluating the CSID of feed ingredients.  相似文献

Measurement of ileal amino acids (AA) bioavailability is recommended to evaluate protein quality. A dual isotope tracer method, based on plasma isotopic enrichment ratios, has been proposed to determine true digestibility in humans. In a pilot study, we aimed to evaluate whether this method could be implemented in rats to determine AA bioavailability based on isotopic enrichment ratios measured in cecal digesta or plasma samples. Goat milk proteins were intrinsically labeled with 15N and 2H. Wistar rats were fed a meal containing the doubly labeled goat whey proteins and a tracer dose of 13C-spirulina. Blood samples were collected 0, 1 h and 3 h after meal ingestion from the tail vein. The rats were euthanized 4 h (n?=?6) or 6 h (n?=?6) after meal to collect plasma and intestinal contents. True orocecal protein digestibility and AA bioavailability were assessed by means of 15N and 2H enrichment in cecum content and compared with absorption indexes determined at the plasma or cecum level using isotopic ratios. Plasma kinetics of isotopic enrichment could not be completed due to the limited quantity of plasma obtained with sequential blood collection. However, the absorption indexes determined from cecal 15N or 2H/13C ratios gave coherent values with true orocecal AA bioavailability. This dual isotope approach with measurements of isotopic ratios in digestive content could be an interesting strategy to determine true AA bioavailability in ileal digesta of rats.

This study aimed to compare the effects of organic (proteinate) and inorganic (sulphate) copper (Cu) and zinc (Zn) supplements, in presence or absence of a mannan oligosaccharide (MOS) supplement, on mineral solubility and digestibility in pigs. Twenty-eight barrows (25 ± 4 kg) assigned randomly to four treatment groups were fed a corn-wheat-soya bean meal diet with 10 mg/kg of Cu and 100 mg/kg of Zn supplied as organic or inorganic supplement, and supplemented or not with 0.1% MOS. After an adaptation period, total faeces and urine were collected for a period of 6–7 days. Pigs were then euthanatised and digesta from ileum and caecum were collected. Apparent digestibility was calculated in ileum and caecum using titanium dioxide. The organic mineral supplement improved total (faecal) digestibility and retained/ingested ratio of Cu (p < 0.05) while reducing apparent digestibility of Zn in the ileum (p < 0.05) without effect on total digestibility of Zn. Solubilities of Cu and Zn in liquid fraction of ileum and caecum were not affected by mineral sources. Although MOS supplement increased Cu solubility in the ileum (< 0.05), it had no effect on digestibility of Zn and Cu in ileum, caecum and faeces, retained/ingested ratio of Zn and Cu, or pH and volatile fatty acid concentration in ileal and caecal digesta. In conclusion, organic mineral supplement improved total digestibility and retained/ingested ratio of Cu in pigs but this cannot be attributed to its solubility in ileal and caecal digesta. The MOS supplement did not interfere with digestibility or dietary utilisation of Zn and Cu in pigs fed above the Zn and Cu requirements.  相似文献

The time course of AA digestion, AA balance (sV AS), and AA absorption (wV AS) was estimated on growing rats (Wistar rats, LW= 124 g) in different sections of the intestinal tract using the combination of 15N tracer and TiO2 marker techniques. The animals received once a diet of 15N labelled wheat and yeast as protein sources supplemented by TiO2 as a marker. Up to 6 h after feeding the amino acid composition the 15N excess and the TiO2 content in the digesta of stomach, small and large intestine were determinated in the relation of amino acids resp. of 15N labelled amino acids to the marker. In addition the content of amino acids and the 15N excess of these amino acids were estimated in plasma. From these data the disappearance rates and the relation of exogenous to endogenous amino acids as well as the sV and the wV values of the different amino acids were calculated for the different gut sections.

The following results were obtained:

- The relative disappearance rate for N and TiO2 marker out of the stomach went approximately parallel but with a delay for TiO2 of about 30 minutes.

- The AA composition of the stomach content, the small and the large intestine content did not vary in dependence of the time.

- The AA composition of the stomach digesta was nearly identical to that of the diet, while that of the small intestine was between exogenous AA composition (feed) and endogenous AA composition (digesta on protein free feeding). AA composition of the large intestine digesta showed quite big differences (bacterial AA break down and AA synthesis).

- Considering a delay time (small intestine: 1 h, large intestine: 4 h) the exogenous portion of the different AA remained constant in both of these intestinal sections during the whole experimental time.

- The exogenous AA part varied for small intestine digesta between 31 and 69% (mean value: 41%), and for large intestine digesta between 13 and 39% (mean value: 22%).

- The sV AS values in the small intestine (AA balance resp. precaecal digestibility) differed from 61% (threonine) to 86% (proline) with an average of 73.4 ± 7.4%, those for wV AS (AA absorption) from 81% (lysine) to 94% (proline) with an average of 88.1±4.1%. There were significant differences between AA, but they are negligible for practical purposes.

- In the small intestine the estimated values for postprandial absorption of the exogenous AA accounted after 1 h, 2 h, 4 h and 6 h for 21%, 33.7%, 46.5%, and 70.7% of the AA intake respectively, mean absorption rate = 9.1 ± 0.5%/h.

- The AA balance in the whole tract (sV AS) was 75 to 94% (on average 82 ± 5.3%) and the wV AS (balance corrected by the 15N method) ranged from 92 to 99% (on average 96±1.8%). These values correspond to the faecal AA digestibility in conventional experiments.

- In the large intestine the postileal disappearance of the AA (sV) was on average 9.7% with a maximum value of 26% for glycine, and with a minimum for methionine of ‐2.1% caused by bacterial synthesis of methionine. Te postileal wV in the large intestine amounted to 7.5%.

- The time course of the disappearance rate of the 15N labelled AA in the small intestine and the appearance rate of these AA in the plasma showed an analogous behavior. Both of them characterize the postprandial absorption.

The following conclusion can be drawn:

The method used (combination of 15N tracer and TiO2 marker techniques) enables determining the time course of transit and the variation of exogenous AA: endogenous AA proportion in the different intestinal sections and estimating the faecal and precaecal digestibility of the different AA.

The course of secretion and absorption of the different AA should be specified in further experiments using the more precise analysis of 15N by GC‐MS resp. GC‐C‐IRMS technique. An apply of this method to farm animals (pigs) seems to be possible.  相似文献

The objective of the present study was to compare the isotope dilution, the difference and the amino acid profile (AAP) methods for the quantification of duodenal and ileal flows of endogenous nitrogen (N) and amino acids (AA) in growing goats. Nine growing goats were fed the same diet containing maize stover, ground corn and soybean meal. The duodenal flow of endogenous N determined by the isotope dilution method was significantly (p < 0.01) higher than that determined by the difference and AAP methods, while there was no difference between the difference and the AAP methods. The duodenal flows of individual endogenous AA determined by the isotope dilution method exceed those determined by the difference and the AAP method by 10 to 106%. The endogenous flow at the ileum determined by the isotope dilution method was not significantly different to those determined by the water-soluble method, but tended to be lower for N and all amino acids. It is concluded that the difference method and AAP method underestimate the duodenal flow of endogenous N and AA compared to the isotope dilution method.  相似文献

The common usage of protein-free diets to estimate unspecific AA losses has been criticised as unphysiological and incorrect. Therefore, in this study different diets were tested for the determination of endogenous losses (EL) of amino acids (AA) and nitrogen (N) assuming a complete absorption in the small intestine. Seven cannulated gilts received a protein-free diet (Diet PF) or diets with 3%, 6% or 10% crude protein (CP) from crystalline AA (Diets CA) or casein (Diets CAS) according to a 7 × 7 Latin square design. After 6 d adaptation to the diet, ileal digesta was collected for 24 h and thereafter analysed for AA, N and the digestibility markers Cr2O3 and acid insoluble ash (AIA). Generally, among all AA, the highest amounts of EL were found for Pro, Glu and Gly, and the smallest for Met. Different levels of CP in Diets CA and CAS had no effect on EL. Significant differences between treatments were observed only for the EL of Glu, Ile, Ser (higher in Diets CA and PF), Pro and Tyr (higher in Diet PF) (p < 0.05). There were no differences in determined EL using Cr2O3 or AIA as digestibility markers.  相似文献

The objective of the study was to determine standardized ileal digestibilities (SID) of crude protein (CP) and amino acids (AA) in six ingredients commonly used in weanling pigs diets. The ingredients consisted of extruded soybeans (SBe), high-protein soybean meal (SBMhp), soy protein concentrate (SPC), hydrolyzed wheat gluten (WGh), conventional fish meal 1 (FM1) and extracted fish meal 2 (FM2e). Each assay feed ingredient was added to a purified cornstarch-based basal diet supplemented with casein and crystalline AA, and SID of CP and AA of assay ingredients were estimated using the difference method. A total of 12 3-week-old barrows were surgically fitted with T-cannulas at the distal ileum. The piglets were allocated to four simultaneous 3 × 3 Latin-square designs with three animals and three periods each, resulting in six observations per assay diet. After 3 to 5 days recovery from surgery, each assay diet was fed at a level of 30 g/kg body weight. Each experimental period consisted of 5 days for adaptation to the assay diets and 2 days for ileal digesta collection. The SID of CP, Lys, Met, Thr and Trp were 73%, 78%, 76%, 66% and 71% in SBe; 80%, 84%, 87%, 75% and 81% in SBMhp; 86%, 89%, 89%, 78% and 83% in SPC; 87%, 60%, 88%, 76% and 79% in WGh; 87%, 92%, 94%, 86% and 86% in FM1; and 79%, 86%, 89%, 80% and 74% in FM2e, respectively. The greatest SID of indispensable AA were observed for FM1 and SPC (SID 85% for most indispensable AA), followed by WGh, FM2e and SBMhp (SID 80% for most indispensable AA) and the smallest SID were obtained for SBe (SID < 80% for most indispensable AA). The SID of CP and indispensable AA in both types of fish meal, SBMhp and WGh were similar to SPC (P = 0.063 to 0.855), except for Arg (P = 0.010) and Lys (P = 0.001) in WGh, and Phe (P = 0.044) and Trp (P = 0.037) in FM2e. The SID of CP and indispensable AA were smaller in SBe compared with SPC (P < 0.001 to P = 0.017), except for Lys (P = 0.136). The SID of CP and indispensable AA were consistently smaller in FM2e compared with FM1 with significant differences for CP (P = 0.035), Phe (P = 0.028) and Trp (P = 0.008). Digestibility values measured in the present study can be used to formulate diets for piglets based on standardized ileal digestible contents of CP and AA.  相似文献

This study was designed to determine the effect of ultra-fine Chinese herbal powder as a dietary additive on serum concentrations and apparent ileal digestibilities (AID) of amino acids (AA) in young pigs. In Experiment 1, 60 Duroc x Landrace x Yorkshire piglets weaned at 21 days of age were randomly assigned to one of three treatments, representing supplementation with 0 or 2 g/kg of the powder, or 0.2 g/kg of colistin (an antibiotic) to corn- and soybean meal-based diets (n = 20 per group). Blood samples from five piglets per group were collected on days 7, 14, and 28 to determine serum AA concentrations. In Experiment 2, 12 barrows with an average initial body weight of 7.64 kg were randomly assigned to one of the three dietary treatments, followed by surgical placement of a simple T-cannula at the terminal ileum. All of the diets contained 0.1% titanium oxide as a digestibility marker. The samples of terminal ileal digesta were collected on day 7 for determining AID of AA. Results show that dietary supplementation with the herbal powder increased (P < 0.05) serum concentrations and AID of most AA by 10-50% and 10-16%, respectively. As an indicator of improved intestinal function, AID values of calcium were also enhanced in piglets supplemented with the herbal powder. Dietary supplementation of colistin increased serum concentrations and AID values of some AA by 8-44% and 10-15%, respectively, in comparison with the non-supplemented group. These novel findings demonstrate that the herbal powder can enhance the digestibility of dietary protein and the intestinal absorption of AA into the systemic circulation in post-weaning pigs, therefore providing a new mechanism for its growth- and immunity-promoting efficacy.  相似文献

An isotope dose technique was utilized (i) to determine endogenous amino acid (AA) and protein losses and (ii) to propose adjusted values for AA requirements. The endogenous flow rate was calculated from the pool of enrichment in plasma AA, assuming similitude to enrichment of endogenous AA. In experiment 1, chicks were orally administered D4-lysine at 2% of estimated lysine intake from 16 to 24 days to find the isotopic steady state of the atom percent excess (APE) of lysine for plasma and jejunal and ileal digesta. The APE of D4-lysine in plasma, jejunal digesta and ileal digesta reached the isotopic steady state at 5.5, 3.4 and 2.0 days, respectively, by using the broken-line model. It was assumed that the isotopic steady state at 5 days identified for D4-lysine is also representative for the 15N-labeled AA. In experiment 2, chicks were fed diets from 1 to 21 days with increasing levels of fat (6%, 8%, 12%, 13% extract ether), protein (26%, 28.5%, 31% CP) or fiber (14%, 16%, 18% NDF) by adding poultry fat, soybean meal, blended animal protein or barley. Chicks were orally administered 15N-threonine, 15N-cysteine, 15N-methionine, 15N-lysine and 15N-leucine at 2% of estimated daily intake for 5 days from 17 to 21 days of age. Dietary nutrients influenced endogenous losses (EL), where dietary fat stimulated EL of lysine (P=0.06), leucine and protein (P=0.07); dietary protein enhanced EL of leucine and protein; and finally the dietary fiber increased EL of leucine. Dietary nutrients also affected apparent ileal digestibility (AID). Dietary fat increased AID of cysteine but decreased AID of lysine. Dietary protein reduced AID of protein, threonine, lysine and leucine, and similarly dietary fiber decreased AID of protein, threonine, methionine, lysine and leucine. In contrast, dietary fat or protein did not affect real ileal digestibility (RID) of protein and AA except threonine and leucine. The dietary fiber reduced the RID of protein, threonine and leucine. This indicate that variations of some endogenous AA and protein losses due to dietary nutrients almost eliminates the effects of RID, and thus the EL coming from the body should be utilized to adjust the AA requirement instead of changing the true digestible nutrients of ingredients. The present data suggest that 5 days’ feeding labeled AA was enough to reach the isotopic steady state and AA requirements should be adjusted when additional dietary protein, fat or fiber is fed.  相似文献

This study was conducted to evaluate the effects of the dietary ratio of ruminal degraded protein (RDP) to ruminal undegraded protein (RUP) and the dry matter intake (DMI) on the intestinal flows of endogenous nitrogen (N) and amino acids (AA) in goats. The experiment was designed as a 4 × 4 Latin square using four ruminally, duodenally and ileally cannulated goats. The treatments were arranged in a 2 × 2 factorial design; two ratios of RDP to RUP (65:35 and 45:55, RDP1 and RDP2, respectively) and two levels at 95% and 75% of voluntary feed intake (DMI1 and DMI2, respectively) were fed to the goats. There were no significant differences in the N intake, duodenal flow of total N, undegraded feed N, microbial N, endogenous N or ileal flow of endogenous N, but the duodenal and ileal flow of endogenous N numerically decreased by approximately 22% and 9%, respectively, when the feed intake changed from DMI1 (0.63 kg/d) to DMI2 (0.50 kg/d). The dietary ratio of RDP to RUP had significant effects (p < 0.05) on the ileal flows of endogenous leucine, phenylalanine and cysteine. The present results implied that the duodenal flows of endogenous N and AA decreased when the dietary RDP to RUP ratio and DMI decreased, and the flow of endogenous AA at the ileum also decreased when the DMI decreased but increased with decreasing RDP to RUP ratios.  相似文献

The pancreatic duct-ligated minipig (PL) is an established model of pancreatic exocrine insufficiency (PEI) with a significant decrease of nutrient digestibility. This study aimed to quantify and compare endogenous losses of nitrogen (N) (ileal and faecal) in minipigs receiving an almost N-free diet. Altogether, 12 Göttingen minipigs (7 PL and 5 control animals) fitted with an re-entrant ileo-caecal fistula were used. In Study 1, ileal digesta was collected over a period of 12 h on seven consecutive days, including one 24 h collection, when animals were fed a diet containing 0.49 g N/kg dry matter (DM). In Study 2, faeces were collected for 10 consecutive days. In Group PL, the amount and DM content of ileal digesta were higher (p < 0.05), while N concentration was lower than in the Control. The ileo-caecal N flux [g/kg DM intake] was about 2.5 times higher in Group PL (5.47 ± 1.15) than in the Control (1.91 ± 0.59) (p < 0.05). The amount of faeces did not differ, but faecal N losses were higher in Group PL (p < 0.05). Endogenous faecal N losses [g N/kg DM intake] of the Control group (1.17 ± 0.72) were comparable with earlier studies, while those of Group PL were 2.6 times higher (3.09 ± 1.34). In contrast, urinary excretion of N did not differ between the Control and Group PL. In conclusion, PEI caused markedly increased endogenous N losses. Therefore, the impact of reduced digestibility of nutrients on endogenous N losses might be relevant for apparent protein digestibility rates and should be taken into account.  相似文献

The relationship between ileal endogenous nitrogen (N) losses and N retention was studied in two experiments with growing pigs of 40 to 60 kg. In Experiment 1, 13 ileal cannulated castrated males were fed diets based on maize starch, containing either soyabean meal (SBM) with a low trypsin inhibitor activity (TIA), a mixture of toasted and untoasted soyabean meal with a high TIA (mSBM), a commercial batch of peas, or rapeseed expeller cake (RC). Ileal endogenous N recovery was measured using the 15N-isotope dilution technique. Apparent ileal digestibility of crude protein (CP) for the SBM, mSBM, pea and RC diets were 82.8, 72.0, 76.7 and 68.7% (P < 0.05). True ileal CP digestibility for the diets was 96.5, 93.0, 94.0 and 87.5% (P < 0.05), and the recovery of ileal endogenous N was 3.08, 6.01, 4.55 and 5.36 g/kg DMI (P < 0.05). In Experiment 2, sixteen castrated males were used to determine N retention, using almost similar diets as in Experiment 1. The diets contained either SBM, mSBM or peas and were balanced for the contents of apparent ileal digestible (ID) CP (96 g/kg) and ID essential amino acids (EAA; at least 85% of requirement values). The fourth treatment was a diet with mSBM as protein source, but supplemented with EAA to the level of 95% of the requirement values (diet mSBMs). Apparent faecal CP digestibility for the SBM, mSBM, pea and mSBMs diets was 88.6, 87.2, 86.1 and 86.0% (P < 0.05). Urinary N excretion and N retention for these treatments were 0.39, 0.59, 0.40, 0.53 (P < 0.05) and 0.87, 0.80, 0.85, 0.84 g/kg0.75/day (P < 0.05), respectively. Utilization of dietary ID N for N retention were 79.8, 73.3, 78.2 and 77.6% (P < 0.05), respectively. The study showed that increased ileal endogenous N losses are associated with higher losses of urinary N and with a lower N retention. Supplementation of extra essential AA to a diet causing a relatively high flow of ileal endogenous N, may compensate for the lower N utilization under these conditions, and thus limit effects on N retention.  相似文献

Effects of the use of three different soyabean products (soya concentrate and two different soyabean meals), peas or rapeseed hulls in the diet on the apparent ileal digestibility of CP (N × 6.25) and recovery of ileal endogenous nitrogen (N) in weanling pigs were investigated. Ileal endogenous N was measured using the 15N-isotope dilution method. Thirteen castrated male pigs (BW of 12–23 kg) were fed maize starch-based diets containing either soya concentrate (SC), purified rapeseed hulls (pRH) and SC (SRH diet), soyabean meal (SBM), a mixture of toasted and untoasted soyabean meal (mSBM) or peas. The apparent ileal digestibility (AID) of dry matter (DM) for the SC, SRH, SBM, mSBM and pea diets was 82.1, 74.3, 83.3, 80.0 and 74.9% (P < 0.05), respectively. The AID of CP for these diets was 82.4, 67.6, 81.6, 68.0 and 76.9% (P < 0.05), respectively. Similar differences in the AID for amino acids (AA) in the diets were found. The AID for CP and for the sum of AA in the pRH, as calculated by the difference between the SC and SRH diet, were 26 and 41%, respectively. For the SC, SRH, SBM, mSBM and pea diet, ileal recovery (g/kg of DM intake) for endogenous N were 2.79, 3.46, 2.73, 4.89 and 3.29 (P < 0.05), respectively, and for dietary N 1.16, 4.11, 1.64, 4.15 and 2.53 (P < 0.05), respectively. For the SC, SBM and mSBM diets, differences in AID of CP and AA were associated with differences in both the ileal recovery of endogenous N and the recovery of undigested dietary N. Differences in trypsin inhibitor activity (TIA) of the soya products were likely associated with these observations. It was also concluded that fibre (Neutral Detergent Fibre; NDF) in pea could be at least in part responsible for the relatively low AID of CP and AA and the high recovery of ileal endogenous N in the pea diet. The low AID of CP and AA of pRH was likely related to the association of protein to the fibre matrix in the fibre-rich rapeseed hulls.  相似文献
