The relationship between active extrusion of Ca++ from red cell ghosts and active uptake of Ca++ by isolated red cell membrane fragments was investigated by studying the Ca++ uptake activities of inside-out and right side-out vesicles. Preparations A and B which had mainly inside-out and right side-out vesicles, respectively, were isolated from red cell membranes and were compared with respect to Ca++ adenosine triphosphatase (ATPase) and ATP-dependent Ca++ uptake activities. Preparation A had nearly eight times more inside-out vesicles and took up eight times more 45Ca in the presence of ATP compared to preparation B. Separation of the 45Ca-labeled membrane vesicles by density gradient centrifugation showed that the 45Ca label was localized to the inside-out vesicle fraction. In addition, the 45Ca taken up in the presence of ATP was lost during a subsequent incubation in the absence of ATP. The rate of 45Ca loss was not influenced by the presence of EGTA, but was slowed in the presence of La+8 (0.1 mM) in the efflux medium. The results presented here support the thesis that the active uptake of Ca++ by red cell membrane fragments is due to the active transport of Ca++ into inside-out vesicles.  相似文献

The quality of rat liver plasma membranes was examined using three different methods of preparation. Hypotonic Ca++-free preparation media were compared with hypotonic Ca++ media and with isotonic Ca++ media. The plasma membranes prepared in hypotonic Ca++ free media were as pure or better than the other two preparation types, while the (Na+K+)-MgATPase, MgATPase, and the ADPase specific activities were higher. The AMPase specific activities were of similar value for all three types of preparation.  相似文献

The present study aimed to characterize the role of protein kinase C (PKC) on the dynamics of tight junction (TJ) opening and closing in the frog urinary bladder. The early events of TJ dynamics were evaluated by the fast Ca++ switch assay (FCSA), which consisted in opening the TJs by removing basolateral Ca++ ([Ca++] bl ), and closing them by returning [Ca++] bl to normal values. Changes in TJ permeability can be reliably gauged through changes of transepithelial electrical conductance (G) determined in the absence of apical Na+. The FCSA allows the appraisal of drugs and procedures acting upon the mechanism controlling the TJs. The time courses of TJ opening and closing in an FCSA were shown to follow single exponential time courses. PKC inhibition by H7 (100 μm) caused a reduction of the rate of junction opening in response to removing [Ca++] bl , without affecting junction closing, indicating that PKC is a key element in the control of TJ opening dynamics in this preparation. H7 at 250 μm almost completely inhibits TJ opening in response to basolateral Ca++ withdrawal. Subsequent H7 removal caused a prompt inhibition release characterized by a sharp G increase which, however, once started cannot be stopped by H7 reintroduction, Ca++ being necessary to allow TJ recovery. A step rise of apical Ca++ concentration ([Ca++] ap ) causes a reduction of the rate of TJ opening in a FCSA, an effect that is believed to be mediated by apical Ca++ entering the open TJs. The specific condition of having Ca++ only in the apical solution and the TJs located midway between the Ca++ source (apical solution) and the Ca++-binding sites presumably located at the zonula adhaerens, might configure a situation in which a control feedback loop is set up. A rise of [Ca++] ap during the phase of G increase in an FCSA causes a transient recovery of G followed by a subsequent escape phase where G increases again. Oscillations of G also appear in response to a rise of apical Ca++. Both escape and oscillations result from the properties of the TJ regulatory feedback loop. In conclusion, the present results indicate that PKC plays a key role in TJ opening in response to extracellular Ca++ withdrawal without major effect on the reverse process. In addition, PKC inhibition by H7 not only prevents TJ opening in response to basolateral Ca++ removal but induces a prompt blockade of TJ oscillations induced by apical Ca++, oscillations which reappear again when H7 is removed. White erythrocyte membranes, or ghosts, were monoconcave discocytes when incubated in 50mM N-tris (hydroxymethyl) methyl-2-aminoethane sulfonic acid titrated to pH 7.4 with triethanolamine. If 3mM MgCl2 was included in the incubation medium, the ghosts were predominantly echinocytes. The echinocytic form could also be induced by Co++, Ni++, Li+, Na+, K+, NH4+ and tetramethylammonium ion, all as chloride salts. The concentration of cation necessary for 50% of the ghosts to be echinocytes was correlated with the hydrated charge density of the cation with the most highly charged cations being the most effective. The cations Ca++, Sr++, Ba++ and La+++, (also as chloride salts) did not induce the normal echinocytic form, but at high levels induced a few misshapen forms with some resemblance to echinocytes. Instead Ca++, Sr++, Ba++ and La+++ suppressed the formation of echinocytes in the presence of Mg++ and other ions. This suggests the presence of a specific Ca++ binding site important to shape control in the erythrocyte membrane.  相似文献

Summary The exposure of red cell ghosts to external Ca++ and K+ leads to a rapid net K+ efflux. Preincubation of the ghosts for various lengths of time in the absence of K+ in the external medium prior to a challenge with maximally effective concentrations of Ca++ and K+ renders the ghosts unresponsive to that challenge with a half-time of about 7–10 min. Preincubation at a range of K+ concentrations for a fixed length of time (60 min) prior to the challenge revealed that K+ concentrations of about 500 m or more suffice to maintain the K+ channel in a maximally responsive state for at least 60 min. These K+ concentrations are considerably lower than the K+ concentrations required to make the responsive channel respond with a maximal rate of K+ efflux. Thus, external K+ is not only necessary to induce the permeability change but also to maintain the transport system in a functional state.The presence of Mg++ or ethylenediamine-tetraacetic acid (EDTA) in the K+-free preincubation media preserves the responsiveness to a challenge with Ca++ plus K+. In contrast to external K+, the presence of external Ca++ does not reduce but rather enhances the loss of responsiveness. An excess of EDTA prevents the effects of Ca++ while washes with EDTA after exposure to Ca++ do not reverse them.In red cell ghosts that contain Ca++ buffers, the transition from a responsive to a nonresponsive state incubation in the absence of external K+ is enhanced. The effects of incubation in the presence of Ca++ in K+-free media are reversed; external Ca++ now reduces the rate at which the responsiveness is lost. The loss of responsiveness after incubation in K+-free media prior to a challenge with external K+ and internal Ca++ does also take place when K+-efflux from red cell ghosts is measured by means of42K+ into media that have the same K+ concentrations as the ghost interior. The relationship between uptake of Ca++ and incorporation of sn-[14C]-glycerol-3-phosphate into phosphatidate, diglyceride, and triglyceride was evaluated in microsomes isolated from livers of normal fed male rats. Uptake of Ca++ was dependent on concentration of Ca++ (0.1 – 2.5 mM), and accompanied by a decrease in the rate of glycerolipid synthesis. The quantity of Ca++ ion taken up at 20 μM CaCl2 in the presence of ATP was equivalent to that observed with 2.5 mM CaCl2 in the absence of ATP. The ATP dependent uptake of Ca++, like the passive uptake at higher concentrations of Ca++, was correlated with inhibition of incorporation of sn-glycerol-3-phosphate into phosphatidate. Accumulation of Ca++ in hepatic microsomes, therefore, appears to result in a calcium-dependent decrease in biosynthesis of phosphatidate and other glycerolipids.  相似文献

Glucose-6-phosphate dehydrogenase [d-glucose-6-phosphate: NADP oxidoreductase, EC. 1. 1. 1. 49] obtained from spores of Bacillus subtilis PCI 219 strain was partially purified by filtration on Sephadex G-200, ammonium sulfate fractionation and chromatography on DEAE-Sephadex A-25 (about 54-fold). The optimum pH for stability of this enzyme was about 6.3 and the optimum pH for the reaction about 8.3. The apparent Km values of the enzyme were 5.7 × 10–4 M for glucose-6-phosphate and 2.4 × 10–4 M for nicotinamide adenine dinucleotide phosphate (NADP). The isoelectric point was about pH 3.9. The enzyme activity was unaffected by the addition of Mg++ or Ca++. The inactive glucoses-6-phosphate dehydrogenase obtained from the spores heated at 85 C for 30 min was not reactivated by the addition of ethylenediaminetetraacetic acid, dipicolinic acid or some salts unlike inactive glucose dehydrogenase.  相似文献

Subcellular localization of 45Ca++ in brain was determined after intracerebroventricular injection of the isotope in mice. Acute morphine injection selectively depleted 45Ca++ from synaptic vesicles while chronic morphine treatment increased the 45Ca++ in vesicular fractions. The elevated vesicular 45Ca++ found in tolerant-dependent animals rapidly declined during naloxone precipitated abstinence. These effects of morphine on brain Ca++ localization are discussed in terms of their possible relationship to neurotransmitter release and tolerance and dependence development.  相似文献

Oscillations of cytosolic Ca2+ activity ([Ca2+]i) induced by stimulation with ATP in rat astrocytes in primary cultures were analysed. Astrocytes, prepared from the brains of newborn rats, loaded with the fluorescent Ca2+ indicator fura-2/AM, were continuously stimulated with ATP (10 M). ATP caused a large initial [Ca2+ peak, followed by regular [Ca2+]i oscillations (frequencies 1–5/min). Astrocytes were identified by glial fibrillary acidic protein staining of cells after [Ca2+]i recording. The oscillations were reversibly blocked by the P2 purinoceptor antagonist suramin (30 M). Influx of extracellular Ca2+ and mobilization of Ca2+ from intracellular stores both contributed to the oscillations. The effects of hypertonic and hypotonic superfusion medium on ATP-induced [Ca2+]i oscillations were examined. Hypertonic medium (430 mOsm) reversibly suppressed the ATP-induced oscillations. Hypotonic medium (250 mOsm), in spite of having heterogeneous effects, most frequently induced a rise in [Ca2+]i, or reversibly increased the frequency of the oscillations. Thus, a change in cell volume might be closely connected with [Ca2+]i oscillations in astrocytes indicating that [Ca2+]i oscillations in glial cells play an important role in regulatory volume regulation in the brain.  相似文献

Summary Secretagogues of pancreatic enzyme secretion, the hormones pancreozymin, carbamylcholine, gastrin I, the octapeptide of pancreozymin, and caerulein as well as the Ca++-ionophore A 23187 stimulate45Ca efflux from isolated pancreatic cells. The nonsecretagogic hormones adrenaline, isoproterenol, secretin, as well as dibutyryl cyclic adenosine 3,5-monophosphate and dibutyryl cyclic guanosine 3,5-monophosphate have no effect on45Ca efflux. Atropine blocks the stimulatory effect of carbamylcholine on45Ca efflux completely, but not that of pancreozymin. A graphical analysis of the Ca++ efflux curves reveals at least three phases: a first phase, probably derived from Ca++ bound to the plasma membrane; a second phase, possibly representing Ca++ efflux from cytosol of the cells; and a third phase, probably from mitochondria or other cellular particles. The Ca++ efflux of all phases is stimulated by pancreozymin and carbamylcholine. Ca++ efflux is not significantly effected by the presence or absence of Ca++ in the incubation medium. Metabolic inhibitors of ATP production, Antimycin A and dinitrophenol, which inhibit Ca++ uptake into mitochondria, stimulate Ca++ efflux from the isolated cells remarkably, but inhibit the slow phase of Ca++ influx, indicating the role of mitochondria as an intracellular Ca++ compartment. Measurements of the45Ca++ influx at different Ca++ concentrations in the medium reveal saturation type kinetics, which are compatible with a carrier or channel model. The hormones mentioned above stimulate the rate of Ca++ translocation.The data suggest that secretagogues of pancreatic enzyme secretion act by increasing the rate of Ca++ transport most likely at the level of the cell membrane and that Ca++ exchange diffusion does not contribute to the45Ca++ fluxes.With the technical assistance of C. Oscillations in cytoplasmic Ca2+ concentration are a universal mode of signaling following physiological levels of stimulation with agonists that engage the phospholipase C pathway. Sustained cytoplasmic Ca2+ oscillations require replenishment of the membrane phospholipid phosphatidylinositol 4,5-bisphosphate (PIP2), the source of the Ca2+-releasing second messenger inositol trisphosphate. Here we show that cytoplasmic Ca2+ oscillations induced by cysteinyl leukotriene type I receptor activation run down when cells are pretreated with Li+, an inhibitor of inositol monophosphatases that prevents PIP2 resynthesis. In Li+-treated cells, cytoplasmic Ca2+ signals evoked by an agonist were rescued by addition of exogenous inositol or phosphatidylinositol 4-phosphate (PI4P). Knockdown of the phosphatidylinositol 4-phosphate 5 (PIP5) kinases α and γ resulted in rapid loss of the intracellular Ca2+ oscillations and also prevented rescue by PI4P. Knockdown of talin1, a protein that helps regulate PIP5 kinases, accelerated rundown of cytoplasmic Ca2+ oscillations, and these could not be rescued by inositol or PI4P. In Li+-treated cells, recovery of the cytoplasmic Ca2+ oscillations in the presence of inositol or PI4P was suppressed when Ca2+ influx through store-operated Ca2+ channels was inhibited. After rundown of the Ca2+ signals following leukotriene receptor activation, stimulation of P2Y receptors evoked prominent inositol trisphosphate-dependent Ca2+ release. Therefore, leukotriene and P2Y receptors utilize distinct membrane PIP2 pools. Our findings show that store-operated Ca2+ entry is needed to sustain cytoplasmic Ca2+ signaling following leukotriene receptor activation both by refilling the Ca2+ stores and by helping to replenish the PIP2 pool accessible to leukotriene receptors, ostensibly through control of PIP5 kinase activity.  相似文献

Human erythrocytes, white and resealed erythrocyte ghosts, and hemolysates were analysed for the occurrence of circadian and ultradian rhythms in vitro. Although the experimental conditions were extensively varied, we did not find any indication for the existence of rhythmicity. These results are at variance with the reports by others.  相似文献

A study of the intracellular transport of calcium in rat heart   总被引:4,自引:0,他引:4  
The distribution of in vivo injected 45Ca++ in the subcellular fractions of rat heart has been studied. Most of the radioactivity of the cell was found to be associated with the subcellular organelles; only a small fraction was recovered in the soluble phase. Mitochondria contained the greatest part of the total radioactivity associated with the subcellular organelles. After injection of 45Ca++ the specific activity of the mitochondrial calcium pool was several times higher than that of the calcium of the sarcoplasmic reticulum. Pentachlorophenol has been administered to rats to uncouple oxidative phosphorylation in heart mitochondria in vivo and its effect on the distribution of 45Ca++ in the heart studied. Under these conditions, it has been found that mitochondria contained much less 45Ca++ than the controls; this decrease was paralleled by an increase of the radioactivity associated with the microsomes and with the final supernatant. Experiments in which 45Ca++ was added to heart homogenates at 0° indicated that 45Ca++ also became bound to mitochondria and the other subcellular structures at 0°. However, PCP had no effect on the distribution of radioactivity among the subcellular fractions under these conditions. The results suggest that (1) energy-linked movements of Ca++ take place in mitochondria of the intact rat heart, (2) a part of the uptake of 45Ca++ by mitochondria does not depend on metabolism, and, (3) the movements of Ca++ in heart mitochondria in vivo are probably more active than those in the sarcoplasmic reticulum.  相似文献

Summary Rabbit aortic smooth muscle cells were prepared by enzymatic digestion of the aortic smooth muscle layer. The cells were subcultured up to Passage 22 starting from a cryogenically preserved stock (approximately 1010cells, Passage 8) and characterized morphologically and for45Ca++ uptake. Microscopically the cells demonstrated the characteristics of vascular smooth muscle cells.45Ca++ uptake by the cells plated on tissue culture flasks (25 cm2) was determined at 25°C in physiological salt solution (PSS) containing45Ca++ in low (5 mM) or high (50mM) KCl concentrations. At the end of the incubation period (0 to 30 min), PSS was aspirated and the cells quickly washed, digested with 0.5N NaOH, and counted for45Ca++. High K+ increased the45Ca++ uptake by 100% or more compared to the low K+ uptake of45Ca++. This K+-induced45Ca++ uptake was eliminated in osmotically shocked cells, and inhibited by nifedipine, verapamil, and diltiazem, in a dose-dependent manner. The extent of45Ca++ uptake and the inhibitory activity of nifedipine were retained up to Passage 22. It is concluded that the developed methodology for scaled-up cultures of rabbit aortic smooth muscle cells provides morphologically intact and biochemically functioning cells suitable for calcium channel studies.  相似文献

The stimulation of the (Ca2+ + Mg2+)ATPase of erythrocyte ghosts by calmodulin was observed not only in intact ghosts, but also in the solubilized (Triton X-100) and partially purified, reconstituted (phosphatidylserine liposomes) forms. Since the solubilized form of the enzyme migrated on Sepharose 6B at a position corresponding to a molecular weight of about 150,000, these results show that calmodulin stimulates by direct interaction with the ATPase complex. Additionally, the effects of calmodulin on erythrocyte ghosts prepared by the Dodge-EDTA method (hypotonic ghosts) and by the method of Ronner et al. (involving lysis followed by an isotonic wash repeated several times) were compared (P. Ronner, P. Gazzotti, and E. Carafoli, 1977, Arch. Biochem. Biophys. 179, 578–583). The (Ca2+ + Mg2+)ATPase of the hypotonic ghosts was low and was stimulated by added calmodulin while that of the isotonic ghosts was high and changed only slightly upon calmodulin addition; this difference in response to calmodulin persisted in the solubilized and reconstituted forms. Hypotonic ghosts bound 125I-labeled calmodulin, while isotonic ghosts did not. This comparison of two types of ghosts showed that isotonic ghosts possess an intact calmodulin-(Ca2+ + Mg2+)ATPase complex, and that the calmodulin remained with the ATPase during solubilization and reconstitution. The isotonic preparation is a particularly useful method of preparing ghosts with an intact calmodulin-ATPase complex, since it requires no special equipment and produces an enzyme activity which is stable to freezing.  相似文献

After injection of 45Ca++ or 89Sr++ into rats, the largest part of the radioactivity in the liver cell is associated with the subcellular structures, only negligible amounts of it being found in the soluble hyaloplasm. 50 % or more of the 45Ca++ and 89Sr++ in the liver cell is recovered in the mitochondrial fraction. The specific activity of Ca++ after injection of 45Ca++ is far greater in mitochondria than in microsomes. Pretreatment of the rats with uncouplers of oxidative phosphorylation markedly decreases the amount of radioactivity associated with the mitochondrial fraction. The amount of radioactivity recovered in the microsomes and in the final supernatant on the contrary increases. These effects are present only when mitochondrial oxidative phosphorylation is completely uncoupled. The Ca++ content of mitochondria from the livers of rats pretreated with uncouplers is sharply decreased with respect to the controls. It is concluded that in the liver cells of the intact animal energy-linked movements of Ca++ and Sr++ take place in mitochondria.  相似文献

Primary cultures of bone cells and skin fibroblasts were examined for their Ca++ content, intracellular distribution and Ca++ fluxes. Kinetic analysis of 45Ca++ efflux curves indicated the presence of three exchangeable Ca++ compartments which turned over at different rates: a “very fast turnover” (S1), a “fast turnover” (S2), and a “slow turnover” Ca++ pool (S3). S1 was taken to represent extracellular membrane-bound Ca++, S2 represented cytosolic Ca++, and S3 was taken to represent Ca++ sequestered in some intracellular organelles, probably the mitochondria. Bone cells contained about twice the amount of Ca++ as compared with cultured fibroblasts. Most of this extra Ca++ was localized in the “slow turnover” intracellular Ca++ pool (S3). Serum activation caused the following changes in the amount, distribution, and fluxes of Ca++: (1) In both types of cells serum caused an increase in the amount of Ca++ in the “very fast turnover” Ca++ pool, and an increase in the rate constant of 45Ca++ efflux from this pool, indicating a decrease in the strength of Ca++ binding to ligands on cell membranes. (2) In fibroblasts, serum activation also caused a marked decrease in the content of Ca++ in the “slow turnover” Ca++ pool (S3), an increase in the rates of Ca++ efflux from the cells to the medium, and from S3 to S2, as well as a decrease in the rate of influx into S3. (3) In bone cells the amount of Ca++ in S3 remained high in “serum activated” cells, the rate of efflux from S3 to S2 increased, and the rate of influx into S3 also increased. The rate of efflux from the cells to the medium did not change. The results suggest specific properties of bone cells with regard to cell Ca++ presumably connected with their differentiation. Following serum activation we investigated the time course of changes in the amount of exchangeable Ca++ in bone cells and fibroblasts, in parallel with measurements of 3H-thymidine incorporation and cell numbers. Serum activation caused a rapid decrease in the content of cell Ca++ which was followed by a biphasic increase lasting until cell division.  相似文献

The technique of reversible hemolysis represents one approach which may be used to study transport regulation in nucleated red cells. After 1 h of incubation at 37°C, 88% of the ghosts regained their permeability barrier to l-glucose. In these ghosts, the carrier-mediated rate of entry of 3-O-methylglucose was more than 10-fold greater than the rate in intact cells. Glyceraldehyde-3-phosphate dehydrogenase prevented ghosts from resealing when it was present at the time of hemolysis. Albumin, lactic dehydrogenase and peroxidase did not have this effect. Sugar transport rate could not be tested in the unsealed ghosts. Two possible mechanisms for the effect of hypotonic hemolysis on sugar transport rate were discussed: (1) altered membrane organization and (2) loss of intracellular compounds which bind to the membrane and inhibit transport in intact cells.  相似文献

The Ca++ ionophore A23187 had no effect on the release of amylase by mouse pancreas fragments in the absence of Ca++ but when Ca++ was re-added to the medium amylase release was observed in a pattern which mimicked that produced by normal stimulants. Uptake of 45Ca++ by pancreatic fragments was increased by A23187. Tetracaine and dinitrophenol at concentrations which block cholinergic stimulated enzyme release blocked ionophore induced release whereas atropine did not. None of the inhibitors studied affected the ionophore induced Ca++ uptake.  相似文献

The effect of the monovalent cations Na+, Li+, and K+ on Ca++ fluxes has been determined in resealed synaptic plasma membrane vesicle preparations from rat brain. Freshly isolated synaptic membranes, as well as synaptic membranes which were frozen (?80°C), rapidly thawed, and passively loaded with K2/succinate and 45CaCl2, rapidly released approximately 60% of the intravesicular Ca++ when exposed to NaCl or to the Ca++ ionophore A 23187. Incubation of these vesicles with LiCl caused a lesser release of Ca++. The EC50 for Na+ activation of Ca++ efflux from the vesicles was approximately 6.6mM. exposure of the Ca++-loaded vesicles to 150 mM KCl produced a very rapid (?1 sec) loss of Ca++ from the vesicles, but the Na+-induced efflux could still be detected above this K+ - sensitive effect. Vesicles pre-loaded with NaCl (150 mM) exhibited rapid 45Ca uptake with an estimated EC50 for Ca++ of 7–10 μM. This Ca++ uptake was blocked by dissipation of the Na+ gradient. These observations are suggestive of the preservation in these purified frozen synaptic membrane preparations of the basic properties of the Na+Ca++ exchange process and of a K+ - sensitive Ca++ flux across the membranes.  相似文献
