The influx of K+(86Rb+) into intact roots of rye (Secale cereale L. cv. Rheidal) exposed to a differential temperature (DT) between the root (8° C) and shoot (20° C) is initially reduced compared with warm-grown (WG) controls with both shoot and root maintained at 20° C. Over a period of 3 d, however, K+-influx rates into DT plants are restored to levels similar to or greater than those of the WG controls, the absolute rates of K+ influx being strongly dependent upon the shoot/root ratio. Acclimation in DT plants results in a reduction of K+ influx into the apical (0–2 cm) region of the seminal root which is associated with a compensatory increase in K+ influx into the more mature, basal regions of the root. Values of V max and apparent K m for K+ influx into DT plants were similar to those for WG plants at assay temperatures of 8° C and 20° C except for an increase in the apparent K m at 8° C. The influx of K+ from solutions containing 0.6 mol·m-3 K+ into both WG and DT plants was found to be linearly related to assay temperature over the range 2–27° C, and the temperature sensitivity of K+ influx to be dependent upon shoot/root ratio. At high shoot/root ratios, the ratio of K+ influx at 20° C:K+ influx at 8° C for WG plants approached a minimum value of 1.9 whereas that for DT plants approached unity indicating that K+ influx into DT plants has a large temperature-insensitive component. Additionally, when plants were grown in solutions of low potassium concentration, K+ influx into DT plants was consistently greater than that into WG plants, in spite of having a greater root potassium concentration ([K+]int). This result indicates some change in the regulation of K+ influx by [K+]int in plants exposed to low root temperatures. We suggest that K+ influx into rye seedlings exposed to low root temperatures is regulated by the increased demand placed on the root system by a proportionally larger shoot and that the acclimation of K+ influx to low temperatures may be the result of an increased hydraulic conductivity of the root system.Abbreviations DT differential temperature pretreatment - [K+]int root potassium concentration - [K+]ext potassium concentration of nutrient medium - WG warm-grown pretreatment  相似文献

Abstract

Uptake of metals by whole Eichhornia crassipes plants and excised roots was studied using a bioassay system. Results indicate that in time-series bioassay at concentration of 10 mg L?1 of either Zn or Cu, rapid uptake by whole plants occurred during the first 4 h, but subsequently levelled off after 48 h. A greater proportion of metals absorbed remain in the root system rather than being translocated to other parts of plant. When treated with different combinations of both Cu and Zn, some synergistic effect of metals appeared to have occurred as indicated by enhanced uptake when bioassays involved combination of metals. Absorption kinetics of monovalent K+ by excised Eichhornia roots after a 24-h treatment, indicated an initial linear trend over the range of 1–10 mg K L?1, but subsequently levelled off at 15 mg K L?1 concentration with Vmax of 7.5 × 10–6 M K g?1 dry tissue h?1 and Km of 1.1 × 10?3 M K. Potentiometric titrations revealed existence of pH-dependent charge densities on root system which have variable affinities for cations and helps explain the ability of Eichhornia roots to absorb and accumulate large amounts of metal ions especially at high pH of media.  相似文献

The impact of two constant non-toxic levels of Al stress (0.2 and 0.4 mM) on growth and 32P uptake capacity on sub-optimal (P-limited) Betula pendula seedlings grown in sand culture was examined.Seedling growth was under optimum controlled conditions in a growth chamber where nutrient additions were made at a predetermined relative addition rate (RA) of 10% day-1. Three treatment groups of seedlings 0, 0.2 and 0.4 mM Al were harvested at 15, 29 and 42 days. The excised roots were exposed to a 32P-labelled solution for 15 minutes to measure their capacity for P uptake. Growth was determined by weighing the roots, stems and leaves of the seedlings.Growth data showed that relative growth rate (RG) should equal the RA of P the most limiting nutrient, which was supplied at P/N 3% instead of an optimal 15%. Therefore, Ingestad's theory can also be used succesfully in sand culture and this may be particularly important for future studies of root and rhizosphere exudates. Low levels of Al (< 0.2 mM) in combination with low P supply significantly lowered the RG of the birch seedlings by further reducing P supply. However, previous studies of birch seedling growth and nutrient uptake using Ingestad's solution culture technique with optimumal P supply did not show any effect of Al on growth untill the Al was in excess of 3 mM. Aluminium was not directly toxic to the plants and therefore roots could respond to the 32P bioassay.  相似文献

The influence of drought stress (DS) upon whole-plant water content, water relations, photosynthesis, and water-use efficiency of Hibiscus rosa-sinensis cv. Leprechaun (Hibiscus) plants at three levels of potassium (K) nutritional status were determined after a 21-d gradually imposed DS treatment. Compared to K-deficient plants, adequate K supply improved the leaf water content (LWC) and leaf water relations of Hibiscus by decreasing the , and generally sustained rates of net photosynthesis (P N) and transpiration (E), and stomatal conductance (g s), both in DS and non-DS plants. In K-deficient Hibiscus, LWC, turgor potential ( P), and P N, E, and g s as well as instantaneous water-use efficiency, WUE (P N/E) were consistently lower, compared to K-sufficient plants. Carbon isotope discrimination () was lower (i.e. longterm WUE was greatest) in DS than non-DS plants, but K had no effect on during the 21-d drought treatment period under glasshouse conditions. However, the trend in the value of DS plants suggests that could be a useful index of the response of Hibiscus to DS under glasshouse growing conditions. Thus the incorporation of a properly controlled fertilization regime involving sufficient levels of K can improve the acclimation of P N to low leaf, increase P N/E of Hibiscus, and may have potential benefit for other woody plants species.  相似文献

Recently, two K+-transport cDNAs, KAT1 and AKT1, were cloned in Arabidopsis thaliana. These cDNAs had structural similarities to K+ channel genes in animals, and also conferred the ability for growth on micromolar levels of K+ when expressed in K+ transport-defective yeast mutants. In this study, we examined the possibility that KAT1 encodes the high-affinity K+ transport system that has been previously characterized in plant roots, by studying the concentration-dependent kinetics of K+ transport for KAT1 expressed in Xenopus oocytes and Saccharomyces cerevisiae. In both organisms, the K+ transport system encoded by KAT1 yielded Michaelis-Menten kinetics with a high Km for K+ (35 mM in oocytes, 0.6 mM in yeast cells). Furthermore, Northern analysis indicated that KAT1 is expressed primarily in the Arabidopsis shoot. These results strongly suggest that the system encoded by KAT1 is not a root high-affinity K+ transporter.  相似文献

The relative importance of endogenous abscisic acid (ABA), as well as Na+ and Cl in NaCl-induced responses related to growth in roots of rice seedlings were investigated. The increase in ammonium, proline and H2O2 levels, and cell wall peroxidase (POD) activity has been shown to be related to NaCl-inhibited root growth of rice seedlings. Increasing concentrations of NaCl from 50 to 150 mM progressively decreased root growth and increased both Na+ and Cl. Treatment with NaCl in the presence of 4,4-diisothiocyano-2,2-disulfonic acid (DIDS, a nonpermeating amino-reactive disulfonic acid known to inhibit the uptake of Cl) had less Cl level in roots than that in the absence of DIDS, but did not affect the levels of Na+, and responses related to growth in roots. Treatment with 50 mM Na-gluconate (the anion of which is not permeable to membrane) had similar Na+ level in roots as that with 100 mM NaCl. It was found that treatment with 50 mM Na-gluconate effected growth reduction and growth-related responses in roots in the same way as 100 mM NaCl. All these results suggest that Cl is not required for NaCl-induced responses in root of rice seedlings. Endogenous ABA level showed no increase in roots of rice seedlings exposed to 150 mM NaCl. It is unlikely that ABA is associated with NaCl-inhibited root growth of rice seedlings.  相似文献

Abstract The effects of gibberellic acid (GA3) on whole sunflower (Helianthus annuus L.) plants grown at three potassium (K) levels (0.0, 0.5 and 5.0 mM) were studied. A tenfold increase in the length of the first internode was observed when plants grown without K were treated with GA3. The uneven K distribution along the plant (higher K content in the higher internodes) was enhanced by GA3 treatment. Gibberellic acid increased the content of reducing sugars, especially in K-deficient plants. An increase in the K level in the nutrient solution resulted in a decrease of the osmotic potential of stem segments. Osmotic potential differences within the elongating first internode were increased by GA3 treatment.  相似文献

The effects of different intensities and durations of soil drought and re-watering on the nitrate uptake ability of maize roots were studied. Plants were grown in split-root containers with one part of the root system subjected to different intensities and durations of soil drought and re-watering while the other part of the root system was continuously watered to 23% (w/w) soil water content (70% water capacity). Experiments were performed in split-root containers to maintain a high growth rate, thus ensuring high nutrient demand of the shoot irrespective of the soil water regime. To avoid limitation of nitrate uptake by transport processes in the dry soil, and to ensure a uniform 14N/15N ratio at the root surface, 15N was applied to the roots by placing them into an aerated nutrient solution with 0.5 mM Ca(15NO3)2. Shoot elongation and biomass were only slightly affected by drought in one root compartment when the soil in the other root compartment was kept wet. Therefore, the growth-related nutrient demand of the shoot remained at a high level. At moderate levels of soil drought (10% w/w water content) the ability of the roots for N-uptake was not affected even after 10 d of drought. N-uptake ability was reduced to about 20% of the well-watered control only when the soil water content was decreased to 5%. Total soluble sugar content of the roots increased with increasing soil drought, indicating that low N-uptake ability of roots subjected to severe soil drought was not caused by low assimilate supply from the shoot. Nitrate uptake ability of roots maintained in very dry soil (5% soil water content w/w) even for a prolonged period of 8 d, recovered within 3 d following re-watering. Root growth increased one day after re-watering. A short-term experiment with excised roots formerly subjected to severe soil drought showed that nitrate uptake ability recovered in old and young root segments after 2 d of re-watering. Obviously, the increase in N-uptake ability after re-watering was caused not only by new root growth but also by recovery of the uptake ability of formerly stressed roots.  相似文献

The carbon and nitrogen partitioning characteristics of wheat (Triticum aestivum L.) and maize (Zea mays L.) grown hydroponically at a constant pH on either 4 mM or 12 mM NO3 - or NH4 + nutrition were investigated using either 14C or 15N techniques. Greater allocation of 14C to amino-N fractions occurred at the expense of allocation of 14C to carbohydrate fractions in NH4 +-compared to NO3 --fed plants. The [14C]carbohydrate:[14C]amino-N ratios were 1.5-fold and 2.0-fold greater in shoots and roots respectively of 12 mM NO3 --compared to 12 mM NH4 +-fed wheat. In both 4 mM and 12 mM N-fed maize the [14C]carbohydrate:[14C]amino-N ratios were approximately 1.7-fold and 2.0-fold greater in shoots and roots respectively of NO3 --compared to NH4 +-fed plants. Similar results were observed in roots of wheat and maize grown in split-root culture with one root-half in NO3 --and the other in NH4 +-containing nutrient media. Thus the allocation of carbon to the amino-N fractions occurred at the expense of carbohydrate fractions, particularly within the root. Allocation of 14N and 15N within separate sets of plants confirmed that NH4 --fed plants accumulated more amino-N compounds than NO3 --fed plants. Wheat roots supplied with 15NH4 + for 8 h were found to accumulate 15NH4 + (8.5 g 15N g-1 h-1) whereas in maize roots very little 15NH4 + accumulated (1.5 g 15N g-1 h-1)It is proposed that the observed accumulation of 15NH4 + in wheat roots in these experiments is the result of limited availability of carbon within the roots of the wheat plants for the detoxification of NH4 +, in contrast to the situation in maize. Higher photosynthetic capacity and lower shoot: root ratios of the C4 maize plants ensure greater carbon availability to the root than in the C3 wheat plants. These differences in carbon and nitrogen partitioning between NO3 --and NH4 +-fed wheat and maize could be responsible for different responses of wheat and maize root growth to NO3 - and NH4 + nutrition.  相似文献

Possible Involvement of Cytokinin in Nitrate-mediated Root Growth in Maize   总被引:1,自引:1,他引:0  
Response of root system architecture to nutrient availability in soils is an essential way for plants to adapt to soil environments. Nitrate can affect root development either as a result of changes in the external concentration, or through changes in the internal nutrient status of the plant. Nevertheless, less is known about the physiological mechanisms. In the present study, two maize (Zea mays L.) inbred lines (478 and Wu312) were used to study a possible role of cytokinin in nitrate-mediated root growth in nutrient solutions. Root elongation of 478 was more sensitive to high nitrate supply than that of Wu312. Medium high nitrate (5 mM) inhibited root elongation in 478, while, root elongation in Wu312 was only inhibited at high NO 3 supply (20 mM). Under high nitrate supply, the root elongation zone in 478 became swollen and the site of lateral root elongation was close towards the root tip. Both of the phenomena are typical of root growth induced by exogenous cytokinin treatments. Correspondingly, zeatin and zeatin nucleotide (Z + ZR) concentrations were increased at higher nitrate supply in 478, whereas they were constant in Wu312. Furthermore, exogenous cytokinin 6-benzylaminopurine (6-BA) completely reversed the stimulatory effect of low nitrate on root elongation. Therefore, it is supposed that the inhibitory effect of high concentration of nitrate on root elongation is, at least in part, mediated by increased cytokinin level in roots. High nitrate supply may have negative influences on root apex activity by affecting cytokinin metabolism so that root apical dominance is weakened and, therefore, root elongation is suppressed and lateral roots grow closer to the root apex. Nitrate suppressed lateral root elongation in Wu312 at concentration higher than 5 mM. In 478, however, this phenomenon was not significant even at 20 mM nitrate. Although exogenous 6-BA (20 nM) could suppress lateral root elongation as well, the inhibitory effect of high NO 3 concentration of nitrate on lateral root growth cannot be explained by changes in endogenous cytokinin alone.  相似文献

Summary The leaf and root nitrate reductase activities were measured in 7 day-old barley seedlings by anoxic nitrite accumulation in darkness, during 48h after the transfer from a N-starved medium to a 1.5 mM K15NO3 medium. Thisin situ nitrate reduction was compared with the15N incorporation in the reduced N fraction of the whole seedlings.The nitrate reduction integrated fromin situ measurements was lower than the reduced15N accumulation. The rootin situ nitrate reductase activity seemed to account for only the third of the real root nitrate reduction, which may have been responsible for the overall underestimation. This discrepancy was partly explained by the ability of the root to reduce nitrite in an anoxic environment.These results suggest that, after correction of thein situ estimation of the nitrate reduction. the roots contribute to about 50% of the total assimilation.  相似文献

Deprivation of nitrogen (N) increases assimilate partitioning towards roots at the expense of that to shoots. This study was done to determine the physiological basis of increased root growth of tea (sCammellia sinensis L.) under N shortage. Nine-month-old clonal tea (clone TRI2025) was grown in quartz sand under naturally lit glasshouse conditions. Three levels of N (0, 3.75 and 7.5 mM N) were incorporated in to the nutrient solution and applied daily. Plant growth, photosynthesis, root respiration and plant N contents were measured at 10-day intervals over a 45-day period. Root dry weight showed a sharp increase during the first 15 days after the plants were transferred to 0 mM N, whereas no such increase was shown in plants transferred to 7.5 mM N. In contrast, shoot dry weight increased at 7.5 mM N and was significantly greater than at 0 mM N, where no increase was observed. Due to the above changes, root weight ratio increased and leaf weight ratio decreased during the first 15 days of N deprivation. Leaf photosynthetic rates did not vary between N levels during the initial 15-day period. Thereafter, photosynthetic rates were greater at 7.5 mM and 3.75 mM N than at 0 mM N. Root respiration rate decreased at 0 mM N, whereas it increased at 3.75 and 7.5 mM N, probably because of the greater respiratory cost for nitrate uptake. Root respiratory costs associated with maintenance (R m) and nitrate uptake (R u) were calculated to investigate whether the sharp increase of root growth observed upon nitrogen deprivation was solely due to the reduced respiratory costs for nitrate uptake. The estimated values for R m and R u were 3.241 × 10–4 mol CO2 g–1 (root dry matter) s–1 and 0.64 mol CO2 (mol N)–1, respectively. Calculations showed that decreased respiratory costs for nitrate uptake could not solely account for the significant increase of root biomass upon N deprivation. Therefore, it is concluded that a significant shift in assimilate partitioning towards roots occurs immediately following N deprivation in tea.  相似文献

Summary The effects of aluminium concentrations between 0.2 and 30 mM at pH 3.8 ±0.2 on small plants of Norway spruce [(Picea abies (L.) Karst], Scots pine (Pinus sylvestris L.), and Scots pine infected with the ectomycorrhizal fungus Suillus bovinus (L. ex Fr.) O. Kuntze were investigated. The plants were grown at maximum relative growth rate (RG % day–1) with free access but very low external concentrations of nutrients. Steady-state conditions with respect to relative growth rate (RG) and internal nutrient concentrations were achieved before addition of aluminium, which was added as AlCl3 and/or Al(NO3)3. There were reductions in rg at aluminium concentrations of 0.3 mM in spruce, 6 mM in pine and 10 mM in ectomycorrhizal pine, i. e. at aluminium concentrations considerably higher than those normally occurring in the top layer of the mineral soil where most fine roots are found. Nutrient uptake rate per unit root growth rate was calculated for different nutrient elements. The uptake rate of calcium and magnesium was reduced at aluminium concentrations of 0.2 mM (spruce), 1 mM (pine) and 3 mM (ectomycorrhizal pine), without influencing Rg. The results question the validity of the hypothesis of aluminium toxicity to forest tree species at low external concentrations.  相似文献

We investigated the influence of an increased inorganic carbon supply in the root medium on NO?3 uptake and assimilation in seedlings of Lycopersicon esculentum (L.) Mill. cv. F144. The seedlings were pre-grown for 4 to 7 days with 0 or 100 mM NaCl in hydroponic culture using 0.2 mM NO?3 (group A) or 0.2 mM NH+4 (group B) as nitrogen source. The nutrient solution for group A plants was aerated with air or with air containing 4 800 μumol mol?1 CO2. Nitrate uptake rate and root and leaf malate contents in these plants were determined. The plants of group B were subdivided into two sets. Plants of one set were transferred either to N-free solution containing 0 or 5 mM NaHCO3, or to a medium containing 2 mM NO?3 and 5 mM NaHCO3. Both sets of group B plants were grown for 12 h in darkness prior to 2 h of illumination, and were assayed for malate content and NO?3 uptake rate (only for plants grown in N-free solution). The second set of group B plants was labeled with 14C by a 1-h pulse of H14CO?3 which was added to a 5 mM NaHCO3 solution containing 0 or 100 mM NaCl and 0 or 2 mM NO?3, and 14C-assimilates were extracted and fractionated. The roots of group B plants growing in carbonated medium accumulated twice as much malate as did control plants. This malate was accumulated only when NO?3 was absent from the root medium. Both a high level of root malate and aeration with CO2-enriched air stimulated NO?3 uptake. Analysis of 14C-assimilates indicated that with no NO?3 in the medium, the 14C was present mainly in organic acids, whereas with NO?3, a large proportion of 14C was incorporated into amino acids. Transport of root-incorporated 14C to the shoot was enhanced by NO?3, while the amino acid fraction was the major 14C-assimilates in the shoot. It is concluded that inorganic carbon fixed through phosphoenolpyruvate carboxylase (EC 4.1.1.31) in roots of tomato plants may have two fates: (a) as a carbon skeleton for amino acid synthesis; and (b) to accumulate, mainly as malate, in the roots, in the absence of a demand for the carbon skeleton. Inorganic carbon fixation in the root provides carbon skeletons for the assimilation of the NH+4 resulting from NO3 reduction, and the subsequent removal of amino acids through the xylem. This ‘removal’ of NO?3 from the cytoplasm of the root cells may in turn increase NO?3 uptake.  相似文献

Soybean cultivars (Glycine max(L.) Merr.) infected with soybean cyst nematode (SCN; Heterodera glycinesIchinohe) often show symptoms similar to K deficiency. The objectives of this experiment were to determine if SCN infection affected macronutrient concentrations in soybean seedling vegetative tissues, determine whether increased K fertility can overcome these possible effects, and to determine if these possible effects are localized at the site of infection or expressed systemically throughout the root system. Soybean plants were grown with root systems split into two halves. This allowed differential K (0.2, 2.4 and 6.0 mM K nutrient solutions) and SCN (0 and 15 000 eggs/plant) treatments to be applied to opposite root-halves of the same plant. Thirty days after plants were inoculated with SCN, macronutrient concentrations of shoot and root tissues were determined. Potassium concentration in leaf blades was not affected; but K concentrations in leaf-petiole and stem tissues were increased with SCN infection. Roots infected with SCN contained lower K concentrations than uninfected roots, but only for the 2.4 mM K treatment. Thus, at the medium level of K fertility, SCN reduced K concentration in soybean roots, and increasing K fertility to the high level overcame the effect. Because K concentrations in the shoot tissues were not reduced by SCN infection, above ground portions of the plant may be able to overcome limitations that occur in roots during the first 30 days of infection. Increasing K fertility level in soybean fields may not benefit vegetative growth of soybean infected with SCN.  相似文献

The influx of Rb+ into the roots of two barley varieties (Hordeum vulgare L. cv. Salve and cv. Ingrid) from a K+-free 86Rb-labelled nutrient solution with 2.0 mM Rb+, was checked at intervals from day 6 to day 18. The control plants were continuously grown in complete nutrient solution containing 5.0 mM K+, while two other groups of plants were grown in K+-free nutrient solution starting on day 6 and between day 6 and day 9, respectively. The pattern of Rb+ influx was similar for both varieties, although their efficiencies in absorbing Rb+ were different. The relationship between Rb+ influx and K+ concentration of the root could be interpreted in terms of negative feedback through allosteric control of uptake across the plasmalemma of the root cells. Hill plots were bimodal, but in the opposite direction. The Hill coefficients, reflecting the minimum number of interacting allosteric binding sites for K+ (Rb+), were low (≤–3.0). It is discussed whether the threshold value, that is the breaking point in the Hill plot, is indicative of a changed efficiency of transporting units for K+ (Rb+) transport to the xylem. Moreover, feedback regulation might be involved in transport of K+ between root and shoot. The variation in K+ concentrations in the roots and shoots of control plants were cyclic but in phase opposition despite an exponential growth. The average K+ concentration varied only slightly with age.  相似文献

Using the two-microelectrode voltage clamp technique in Xenopus laevis oocytes, we estimated Na+-K+-ATPase activity from the dihydroouabain-sensitive current (I DHO) in the presence of increasing concentrations of tetraethylammonium (TEA+; 0, 5, 10, 20, 40 mm), a well-known blocker of K+ channels. The effects of TEA+ on the total oocyte currents could be separated into two distinct parts: generation of a nonsaturating inward current increasing with negative membrane potentials (V M) and a saturable inhibitory component affecting an outward current easily detectable at positive V M. The nonsaturating component appears to be a barium-sensitive electrodiffusion of TEA+ which can be described by the Goldman-Hodgkin-Katz equation, while the saturating component is consistent with the expected blocking effect of TEA+ on K+ channels. Interestingly, this latter component disappears when the Na+-K+-ATPase is inhibited by 10 m DHO. Conversely, TEA+ inhibits a component of I DHO with a k d of 25±4 mm at +50 mV. As the TEA+-sensitive current present in I DHO reversed at –75 mV, we hypothesized that it could come from an inhibition of K+ channels whose activity varies in parallel with the Na+-K+-ATPase activity. Supporting this hypothesis, the inward portion of this TEA+-sensitive current can be completely abolished by the addition of 1 mm Ba2+ to the bath. The influence of cytochalasin B (CB), a potent inhibitor of cytoplasmic streaming, on 86Rb-labelled K+ translocation by detopped Lycopersicon esculentum Mill., Cucumis sativus L. and Zea mays L. plants was examined by measuring the radioactivity in xylem exudate before and after the addition of CB to the medium bathing the roots. CB caused complete cessation of cytoplasmic streaming in root segments within 15 min but was without effect on either total 86Rb uptake or exudation. Thus factors other than cytoplasmic streaming limit the movement of K+ across the symplast of the root of higher plants.  相似文献

The efficacy and mechanism of -dendrotoxin (DTX) block of K+ channel currents in Vicia stomatal guard cells was examined. Currents carried by inward- and outward-rectifying K+ channels were determined under voltage clamp in intact guard cells, and block was characterized as a function of DTX and external K+ (K+) concentrations. Added to the bath, 0.1-30 nM DTX blocked the inward-rectifying K+ current (IK,in), but was ineffective in blocking current through the outward-rectifying K+ channels (IK,out) even at concentrations of 30 nM. DTX block was independent of clamp voltage and had no significant effect on the voltage-dependent kinetics for IK,in, neither altering its activation at voltages negative of –120 mV nor its deactivation at more positive voltages. No evidence was found for a use dependence to DTX action. Block of IK,in followed a simple titration function with an apparent K1/2 for block of 2.2 nM in 3 mm K o + . However, DTX block was dependent on the external K+ concentration. Raising K+ from 3 to 30 mm slowed block and resulted in a 60–70% reduction in its efficacy (apparent K i = 10 mm in 10 nm DTX). The effect of K+ in protecting I K,in was competitive with DTX and specific for permeant cations. A joint analysis of IK,in block with DTX and K+ concentration was consistent with a single class of binding sites with a K d for DTX of 240 pm. A K d of 410 m for extracellular K+ was also indicated. These results complement previous studies implicating a binding site requiring extracellular K+ (K1/2 1 mm) for IK,in activation; they parallel features of K+ channel block by DTX and related peptide toxins in many animal cells, demonstrating the sensitivity of plant plasma membrane K+ channels to nanomolar toxin concentrations under physiological conditions; the data also highlight one main difference: in the guard cells, DTX action appears specific to the K+ inward rectifier.We thank J.O. The effects of varying CaSO4 and NaCl levels on the nutrient content ofLeucaena leucocephala were established by examining the concentrations of Na, Ca, Cl, K and Mg in leucaena roots, stems and leaves. Leucaena was grown in nutrient solution at four levels of CaSO4 (0.5, 1.0, 2.5 and 5.0 mM) and NaCl (1, 25, 50 and 100 mM), in randomized blocks with five replications. Leucaena excluded sodium from stems and leaves when NaCl concentration was 50 mM or less. Sodium uptake decreased as CaSO4 concentration increased. Calcium uptake was affected by NaCl concentration when substrate CaSO4 concentration was 0.5 mM. At this level, 100 mM NaCl caused a marked decrease in leaf calcium and a marked increase in leaf Cl. In all other treatments, Cl uptake was not affected by CaSO4 concentration. Potassium uptake was strongly depressed as NaCl concentration increased at low Ca concentration, but this effect was offset at high Ca. Magnesium uptake decreased as CaSO4 levels increased.  相似文献
