qazTin-119 and phosphorus-31 NMR spectra have been recorded for a series of adducts of RSnX3 (R  Me, Ph; X  Cl, Br) with halide, tributylphosphine (P) and tributylphosphine oxide (L). The adducts were either 1:1 five coordinate or 1:2 six coordinate complexes. The tin-ll9 NMR spectra of mixtures of corresponding chloro and bromo complexes reveal, in most cases, all possible mixed halide species but much additional structural information is obtained from these spectra which could not be extracted from the spectra of individual compounds themselves. Thus in some cases, in the five coordinate species the Berry pseudorotation between isomers within a particular stoichiometry could be slowed on the NMR timescale which allowed a determination of the molecular structure. An equimolar mixture of [PhSnCl5]2− and [PhSnBr5]2− shows eleven of the twelve geometries possible for [PhSnClxBr5−x]2−. In the six coordinate series [RSnX4P] the tin-119 NMR spectra of the mixtures of [RSnCl4P] and [RSnBr4P] allow the geometry to be determined as trans. Application of the pairwise additivity model for calculation of the tin-119 chemical shift positions for the mixed halide systems are discussed.  相似文献

The enzymatic conversion of leukotriene A4 into 5,6-dihydroxy-7,9,11,14-eicosatetraenoic acid, catalyzed by mouse liver cytosolic epoxide hydrolase (EC 3.3.2.3), was recently described (Haeggstr?m, J., Meijer, J. and R?dmark, O. (1986) J. Biol. Chem. 261, 6332-6337). In the present study, we report analytical data confirming the stereochemistry of this novel enzymatic metabolite of leukotriene A4. By steric analysis of the vicinal diol and comparison with synthetic material, the structure was established as (5S,6R)-dihydroxy-7,9-trans-11,14-cis-eicosatetraenoic acid. Apparent kinetic constants of this reaction were determined and found to be 5 microM and 550 nmol.mg-1.min-1, for Km and Vmax, respectively. Also, a semipurified preparation of human liver cytosolic epoxide hydrolase avidly catalyzed the same hydrolysis of leukotriene A4 (apparent Km was 8 microM). The enzyme was not inactivated by leukotriene A4, as judged by time-course experiments with a second substrate addition.  相似文献

Methanolysis of radioactively labelled D-fructose gives, finally, the α-pyranoside, β-pyranoside, α-furanoside and β-furanoside in the ratios 3:46:25:26. The corresponding ratios for the L-sorbosides are 92:1:5:2. As with the aldoses, these ketoses mainly give furanosides in kinetically controlled reactions; the pyranosides are then formed as thermodynamic products. A primary objective of the work was to examine the role played in the reactions by the acyclic dimethyl acetals; with neither sugar was any such product detected, and it is estimated that their concentrations were never in excess of 0.05% in the reaction components.  相似文献

1. (2R,3S)-[U-14C,3-3H1]- and (2R,3R)-[U-14C,2,3-3H2] Cysteine hydrochlorides have been separately synthesised. The latter compound has been shown to have uniform distributions of tritium between C-2 and C-3. 2. The abvoe cysteines and (2R)-[U-14C,3,3,3',3'-3H4]cystine have been converted to samples of penicillin G by Penicillium chrysogenum. 3. Incorporation results indicate that all but 14% of the tritium is lost from the (2R,3S)-[3-3H1]isomer; that 42% of tritium is retained by the non-stereospecifically C-3 tritiated cystine; and that 58% of tritium is retained by the (2R,3R)-[2,3-3H2]isomer on conversion to penicillin G. 4. Degradation of the penicillin G derived from (2R,3R)-[U-14C,2,3-3H2]cysteine hydrochloride has indicated that in fact about 87% of the original C-3 tritium of cysteine is retained at C-5 of penicillin G. 5. The results indicate stereospecificity in the cyclisation giving rise to the beta-lactam ring in penicillin G in nature with loss of the 3-pro-S-hydrogen and rentention of the 3-pro-R-hydrogen of cysteine. Thus there is net retention of stereochemistry in the cyclisation.  相似文献

3H and 14C from (2R,3S)[U-14C,3-3H1]cysteine and (2R,3R)-[U-14C,2,3-3H2]cysteine were incorporated into cephalosporin C by Cephalosporium acremonium. Analysis of the radioactive cephalosporin C indicated that the formation of its beta-lactam ring occurs stereospecifically and with retention of configuration at C-3 of cysteine.  相似文献

Rhizopus delemar lipase catalysed ester hydrolysis of the alpha-methoxy-beta-phenylpropanoate 1 affords the (R)-(+) and (S)-(-) isomers in > 84% enantiomeric excess. Absolute stereochemistry was determined by a single crystal X-ray analysis of a related synthetic analogue. The activity of these two enantiomers on glucose transport in vitro and as anti-diabetic agents in vivo is reported and their unexpected equivalence attributed to an enzyme-mediated stereospecific isomerisation of the (R)-(+) isomer. Binding studies using recombinant human PPARgamma (peroxisomal proliferator activated receptor gamma), now established as a molecular target for this compound class, indicate a 20-fold higher binding affinity for the (S) antipode relative to the (R) antipode.  相似文献

1. A specimen of stereospecifically 2-tritiated 3-hydroxybutyric acid was prepared by hydroboration of ethyl crotonate. It was assumed that the hydroboration reaction took a syn course and hence that (2R,3S) plus (2S,3R)-3-hydroxy[2 minus 3H1]butyric acid was formed after oxidation and hydrolysis. 2. 3RS-3-Hydroxy[2 minus 3H2]butyric acid, symmetrically tritiated at C-2, was prepared by isotopic exchange of ethyl acetoacetate in tritiated water, followed by reduction and hydrolysis. 3. The 3R-enantiomers of the acids listed under paragraphs (1) and (2) were destroyed enzymically by use of 3R-specific 3-hydroxybutyrate dehydrogenase and the residual 3S-enantiomers were isolated. 4. The resulting specimens of 2R,3S-3-hydroxy[2 minus 3H1]butyric acid and 3S-3hydroxy[2 minus 3H2]-butyric acid were converted chemically to the acyl-CoA derivatives. These were incubated with enoyl-CoA hydratase. 5. In the presence of the enoyl-CoA hydratase symmetrically labelled 3S-3-hydroxy[2 minus 3H2]BUTYRYL-CoA lost nearly 50% of its tritium label; 2R,3S-3hydroxy [2-3-H1]butyryl-CoA lost about 78%. 6. It was concluded that the elimination of the elements of water from 3-hydroxybutyryl-CoA on the hydratase occurs stereospecifically with syn geometry.  相似文献
