The transformation of (5R)-2,6-di-O-benzyl-5-C-methoxy-β-d-galactopyranosyl-(1→4)-2,3:5,6-di-O-isopropylidene-aldehydo-d-glucose dimethyl acetal (8) into partially protected derivatives of d-xylo- and l-lyxo-aldohexos-5-ulose has been reported, applying appropriate epimerisation methods to its 3′-O- and 4′-O-protected alcoholic derivatives.  相似文献

Silver carbonate on Celite (the Fetizon reagent) was shown to be selective as an oxidizing agent, and convenient for the preparation of various aldonolactones. Whereas substituted aldoses having the 1-hydroxyl group free were readily converted into the corresponding lactones, partially protected 2-acetamido-2-deoxypyranoses having more than one free hydroxyl group were selectively oxidized at C-1. The oxidation was carrried out in boiling benzene or 1,4-dioxane. A series of partially protected 2-acetamido-2-deoxy-1,5-aldonolactones [2-acetamido-4,6-O-benzylidene-2-deoxy-D-mannono-1,5-lactone (13),2-acetamido-4,6-O-benzylidene-2-deoxy-D-glucono-1,5-lactone (15), 2-acetamido-2-deoxy-4,6-O-isopropylidene-D-glucono-1,5-lactone (18), 2-acetamido-2-deoxy-4,6-O-isopropylidene-D-mannono-1,5-lactone (20), 2-acetamido-2-deoxy-3,4-di-O-methyl-D-mannono-1,5-lactone (24), and 2-acetamido-2-deoxy-3,4-di-O-methyl-D-glucono-1,5-lactone (25)] was thus prepared; for these, the oxidation was accompanied by two side-reactions: (a) an elimination (dehydration) that gave the unsaturated lactones [2-acetamido-4,6-O-benzylidene-2,3-dideoxy-D-erythro-hex-2-enono-1,5-lactone (12), 2-acetamido-2,3-dideoxy-4,6-O-isopropylidene-D-erythro-hex-2-enono-1,5-lactone (17), and 2-acetamido-2,3-dideoxy-4-O-methyl-D-erythro-hex-2-enono-1,5-lactone (23)], and (b) partial gluco-to-manno epimerization occurring during the oxidation of 2-acetamido-4,6-O-benzylidene-2-deoxy-D-glucopyranose (14), 2-acetamido-2-deoxy-4,6-O-isopropylidene-D-glucopyranose (16), and 2-acetamido-2-deoxy-3,4-di-O-methyl-D-glucopyranose (22).The free unsaturated lactone, 2-acetamido-2,3-dideoxy-D-erythro-hex-2-enono-1,5-lactone (26), was obtained on hydrolysis of the isopropylidene group in lactone 17.  相似文献

d-Ribose (1) in N,N-dimethylformamide containing a trace of p-toluenesulfonic acid is acetonated under kinetic control by ethyl (or methyl) isopropenyl ether (2) to give mainly 3,4-O-isopropylidene-β-d-ribopyranose (3), together with lesser proportions of 2,3-O-isopropylidene-d-ribofuranose (4), its 5-(2-alkoxy-2-propyl) ether (5 or 5a), and 1,5:2,3-di-O-isopropylidene-β-d-ribofuranose (6). Similar treatment of d-arabinose (10) gives mostly 3,4-O-isopropylidene-β-d-arabinopyranose (11) together with a minor proportion of 1,2:3,4-di-O-isopropylidene-β-d-arabinopyranose (12). The strucutres of the monoacetals 3 and 11 were confirmed by an acetylation-deacetonation-acetylation sequence.  相似文献

2-Deoxy-d-arabino-hexose and some N-protected 2-amino-2-deoxy-d-glucose derivatives were each treated with 2,2-dimethoxy- or 2,2-dibenzyloxy-propane in 1,4-dioxane in the presence of p-toluenesulfonic acid at 60–70°. The major products were acyclic, dimethyl and dibenzyl acetals of 2-deoxy-3,4:5,6-di-O-isopropylidene-aldehydo-d-arabino-hexose or of N-protected 2-amino-2-deoxy-3,4:5,6-di-O-isopropylidene-aldehydo-d-glucose. Some of the dibenzyl acetals were converted into the corresponding 3,4:5,6-di-O-isopropylidene-aldehydo-d-hexoses in good yield.  相似文献

3,4-Di-O-acetyl-2-O-benzyl-α-d-xylopyranosyl bromide (1) reacts with methyl 2,3-anhydro-α-d-ribopyranoside (2) to afford, in high yield, methyl 2,3-anhydro-4-O- (3,4-di-O-acetyl-2-O-benzyl-β-d-xylopyranosyl)-β-d-ribopyranoside (3). Deacetylation of 3 gave 4, which reacted with 2,3,4-tri-O-acetyl-α-d-xylopyranosyl bromide to give the branched tetrasaccharide derivative 5, which, in turn, was converted by a series or conventional reactions into methyl 4-O-[3,4-di-O-(β-d-xylopyranosyl)-β-d- xylopyranosyl]-β-d-xylopyranoside. The reaction of 1 with its hydrolysis product gave 3,4-di-O-acetyl-2-O-benzyl-α-d-xylopyranosyl 3,4-di-O-acetyl-2-O-benzyl-β-d-xylopyranoside, which was also isolated after the reaction of 1 with 2.  相似文献

The title disaccharide (16) has been synthesized in 50% overall yield by way of condensation of 4,6-di-O-acetyl-2,3-O-carbonyl-α-D-mannopyranosyl bromide 5 with methyl 2,3-O-isopropylidene-α-L-rhamnopyranoside (1) in chloroform solution, in the presence of silver oxide. The disaccharide was characterized as the crystalline isopropyl alcoholate of methyl 4-O-β-D-mannopyranosyl-α-L-rhamnopyranoside (11) and as 1,2,3-tri-O acetyl-4-O- (2,3,4,6-tetra-O-acetyl-β-D-mannopyranosyl)-α-L-rhamnopyranose (15). Methyl β-D-mannopyranoside isopropyl alcoholate 7 was readily obtained in 85% yield via the reaction of bromide 5 with methanol.Reduction of 2,3-di-O-methyl-L-rhamnose with sodium borohydride, followed by acetylation, may result in the formation of an appreciable proportion of a boric ester, namely 1,5-di-O-acetyl-4-deoxy-2,3-di-O-methyl-L-rhamnitol-4-yl dimethyl borate, depending on the procedure used.  相似文献

D-Glucose diethyl dithioacetal (1), its penta-O-acetyl derivative (2), penta-O-acetyl-aldehydo-D-glucose (3), L-xylo-hexulose phenylosotriazole (4), 1,2:5,6-di-O-isopropylidene-D-mannitol (5), 1,2:4,5-di-O-isopropylidene-β-D-fructopyranose (6), 1,2-O-isopropylidene-α-D-glucofuranose (7) and its triacetate (8), 1,6-anhydro-β-D-galactopyranose (9) and its triacetate (10), D-glucopyranose (11), methyl β-D-glucopyranoside tetraacetate (12), 1-thio-β-D-glucopyranose pentaacetate (13), β-D-fructofuranose pentaacetate (14), and raffinose hendecaacetate (15) have been examined by chemical-ionization mass-spectrometry with both isobutane and ammonia as ionizing intermediates. Extreme simplicity characterizes these spectra, and, in most instances, molecular-weight data are available from intact, protonor NH4+capture ions; the limited fragmentation that occurs corresponds in large measure to simple dehydration or substituent-cleavage processes, and is strongly dependent upon the groups present, so that considerable information about the substituent groups in the sugar molecule may be inferred.  相似文献

The preparation of 2,3-di-O-benzoyl-4,6-O-benzylidene-α-d-glucopyranosyl-2-O-benzoyl-4,6-O-benzylidene-α-d-ribo-hexopyranosid-3-ulose (3) from 4,6:4′,6′-di-O-benzylidene-α,α-trehalose (1) via the 2,3,2′-tribenzoate 2 has been improved. Reduction of 3 with sodium borohydride gave 2-O-benzoyl-4,6-O-benzylidene-α-d-allopyranosyl 2,3-di-O-benzoyl-4,6-O-benzylidene-α-d-glucopyranoside (4), which was converted into the methanesulfonate 5 and trifluoromethanesulfonate 6. Displacement of the sulfonic ester group in 6 with lithium azide was very facile and afforded a high yield of 3-azido-2-O-benzoyl-4,6-O-benzylidene-3-deoxy-α-d-glucopyranosyl 2,3-di-O-benzoyl-4,6-O-benzylidene-α-d-glycopyranoside (7), whereas similar displacement in 5 proceeded sluggishly, giving a lower yield of 7 together with an unsaturated disaccharide (8). The azido sugar 7 was converted by conventional reactions into the analogous 2,3,2′-triacetate 9, the corresponding 2,3,2′-triol 10, and deprotected 3-azido-3-deoxy-α-d-glucopyranosyl α-d-glucopyranoside (11). Hydrogenation of 11 over Adams' catalyst furnished crystalline 3-amino-3-deoxy-α,α-trehalose hydrochloride (12), the overall yield from 3 being 35%.  相似文献

Reaction of methyl 4′,6′-di-O-mesyl-β-lactoside pentabenzoate (8), synthesised via the 4′,6′-O-benzylidene derivative (6), with sodium azide in hexamethylphosphoric triamide gave three products. In addition to the required 4′,6′-diazidocellobioside (9), an elimination product, methyl 4-O-(6-azido-2,3-di-O-benzoyl-4,6-dideoxy-α-L-threo-hex-4-enopyranosyl)-2,3,6-tri-O-benzoyl-β-D-glucopyranoside (12), and an unexpected product of interglycosidic cleavage, methyl 2,3,6-tri-O-benzoyl-β-D-glucopyranoside (13), were formed. The origin of the latter product is discussed. The diazide 9 was converted into 4′,6′-diacetamido-4′,6′-dideoxycellobiose hexa-acetate (16) by sequential debenzoylation, catalytic reduction, acetylation, and acetolysis.  相似文献

Several 2-O-benzoyl-4,6-di-O-benzyl-3-O-R-α-d-galactopyranosyl chlorides, designed as general precursors of β-linked, interior d-galactopyranosyl residues in oligosaccharides, were tested in a sequential synthesis of the galactotriose β-d-Galp-(1→3)-β-d-Galp-(1→3)-d-Gal (19). The chlorides having R = tetrahydro-2-pyranyl and tert-butyldimethylsilyl gave excellent results whereas those having = 3-benzoylpropionyl and chloroacetyl were unsatisfactory. An activated disaccharide block (17), having R = 2,3-di-O-benzoyl-4,6-di-O-benzyl-β-d-galactopyranosyl, was also prepared and tested as a glycosyl donor. The coupling of 17 to 1-propenyl 2-O-benzoyl-4,6-di-O-benzyl-α-d-galactopyranoside (14), in the molar ratio 1.13:1, gave 64% of a trisaccharide derivative (18) that could be converted into 19. This latter synthesis of 19 is efficient because all three galactose units are derived from 14 or its immediate precursor.  相似文献

Reinvestigation of the reaction of methyl 2,3-O-isopropylidene-4-O-toluene-p-sulphonyl-α-d-lyxopyranoside (4) with azide ion has shown that methyl 4-deoxy-2,3-O-isopropylidene-β-l-erythro-pent-4-enopyranoside (8, ~51.5%) is formed, as well as the azido sugar 7 (~48.5%) of an SN2 displacement. The unsaturated sugar 8 was more conveniently prepared by heating the sulphonate 4 with 1,5-diazabicyclo-[5.4.0]undec-5-ene. An azide displacement on methyl 2,3-O-isopropylidene-4-O-toluene-p-sulphonyl-β-l-ribopyranoside (12) furnished methyl 4-azido-4-deoxy-2,3-O-isopropylidene-α-d-lyxopyranoside (13, ~66%) and the unsaturated sugar 14 (~28.5%), which was also prepared by heating the sulphonate with 1,5-diazabicyclo[5.4.0]undec-5-ene. Deamination of methyl 4-amino-4-deoxy-2,3-O-isopropylidene-α-d-lyxopyranoside (5), prepared by reduction of 13, with sodium nitrite in 90% acetic acid at ~0°, yielded methyl 2,3-O-isopropylidene-α-d-lyxopyranoside (10a, 26.2%), methyl 2,3-O-isopropylidene-β-l-ribofuranoside (21a, 18.4%), and the corresponding acetates 10b (34.5%) and 21b (21.3%). These products are considered to arise by solvolysis of the bicyclic oxonium ion 29, formed as a consequence of participation by the ring-oxygen atom in the deamination reaction. Similar deamination of methyl 4-amino-4-deoxy-2,3-O-isopropylidene-β-l-ribopyranoside (6) afforded, exclusively, the products 10a (34.4%) and 10b (65.6%) of inverted configuration. Deamination of methyl 5-amino-5-deoxy-2,3-O-isopropylidene-β-d-ribofuranoside (20) gave 22ab, but no other products. An alternative synthesis of the amino sugars 5 and 6 is available by conversion of 10a into methyl 2,3-O-isopropylidene-β-l-erythro-pentopyranosid-4-ulose (11), followed by reduction of the derived oxime 15 with lithium aluminium hydride.  相似文献

Three different approaches starting from 1,2-O-isopropylidene-α-d-glucofuranose were tested for the synthesis of daunosamine hydrochloride (24), the sugar constituent of the antitumor antibiotics daunomycin and adriamycin. The third route, affording 24 in ~5% overall yield in 11 steps, constitutes a useful, preparative synthesis, 3,5,6-Tri-O-benzoyl-1,2-O-isopropylidene-α-d-glucofuranose was converted via methyl 2,3-anhydro-β-d-mannofuranoside into methyl 2,3:5,6-dianhydro-α-l-gulofuranoside, the terminal oxirane ring of which was split selectively on reduction with borohydride, to afford methyl 2,3-anhydro-6-deoxy-α-l-gulofuranoside (31). Compound 31 was converted into methyl 2,3-anhydro-5-O-benzyl-6-deoxy-α-l-gulofuranoside, which was selectively reduced at C-2 on treatment with lithium aluminum hydride, affording methyl 5-O-benzyl-2,6-dideoxy-α-l-xylo-hexofuranoside. Subsequent mesylation, and replacement of the mesoloxy group by azide, with inversion, afforded methyl 3-azido-5-O-benzyl-2,6-dideoxy-α-l-lyxo-hexofuranoside, which could be converted into either 24 or methyl 3-acetamido-5-O-acetyl-2,3,6-trideoxy-α-l-lyxo-hexofuranoside, which can be used as a starting material for the synthesis of daunomycin analogs.  相似文献

Methyl 3-azido-2-O-benzoyl-3,4-dideoxy-β-dl-erythro-pentopyranoside (6) was synthesized through two routes in five steps from methyl 2,3-anhydro-4-deoxy-β-dl-erythro-pentopyranoside (1). The first route proceeded via selective azide displacement of the 3-tosyloxy group of methyl 4-deoxy-2,3-di-O-tosyl-α-dl-threo-pentopyranoside, followed by detosylation and benzoylation. The second route consisted, with a better overall yield, in the azide displacement of the mesyloxy group of methyl O-benzoyl-4-deoxy-3-O-methylsulfonyl-α-dl-threo-pentopyranoside (10), obtained by benzylate opening of 1, followed by benzoylation, debenzylation, and mesylation. Compound 6 was transformed into its glycosyl chloride, further treated by 6-chloropurine to give the nucleoside 9-(3-azido-2-O-benzoyl-3,4-dideoxy-β-dl-erythro-pentopyranosyl)-6-chloropurine (13). When treated with propanolic ammonia, 13 yielded 9-(3-azido-3,4-dideoxy-β-dl-erythro-pentopyranosyl)adenine.  相似文献

Amylose (1) was tritylated at O-6, the ether p-toluenesulfonylated at O-2 and O-3, and the product (3) treated with sodium iodide and zinc dust in N,N-dimethyl-formamide, to give 2,3-dideoxy-6-O-trityl-α-D-erythro-hex-2-enopyranoglycan (4). This 2,3-unsaturated polysaccharide could be converted into a 2,3-dibromo derivative (5), and hydrogenated with concomitant detritylation to the saturated analogue (6), and, on treatment with aqueous acetic acid, it gave 2-(D-glycero-1,2-dihydroxyethyl)-furan (8). The 2,3-bis(p-toluenesulfonate) (10) of β-D-xylan (9) was similarly converted into the 2,3-unsaturated polysaccharide, 2,3-dideoxy-β-D-glycero-pent-2-enopyranoglycan (11), which, with aqueous acetic acid, gave 2-(hydroxymethyl)furan (12a).  相似文献

The following primary sulphonates have been converted into the corresponding deoxyfluoro derivatives by reaction with potassium fluoride in ethylene glycol:1,2:3,4-di-O-isopropylidene-6-O-tosyl α-D-galactopyranose (1), methyl 2,3-O2-isopropyliden-5-O-tosyl-α,β-D-ribofuranoside (2), 1,2:3,4-di-O-methylene-6-O-tosyl-α-D-glucofuranose (3), 3,5-di-O-benzylidene-1,2-O-isopropylidene-6-O-tosyl-α-D-glucofuranose (4), and 1,2:3,5-di-O-isopropylidene-6-O-tosyl-α-D-glucofuranose (5). The yields were generally poor; in the reaction of 1, a major by-product was 6-O-(2-hydroxyethyl)-1,2:3,4-di-O-isopropylidene-α-D-galactopyranose (11). The reaction of the primary hydroxyl precursor of each of the above tosylates with N2-(2-chloro- 1,1,2-trifluoroethyl)-N,N-diethylamine generally yielded the O-chlorofluoroacetyl derivative; however, 1,2:3,5-di-O-methylene-α-D-glucofuranose (12) was converted into the 6-deoxy-6-fluoro derivative (8). The 19F resonances of compounds containing the CH2F moiety fall between φC +213 and φC +235 p.p.m. The differences between the vicinal19F-1H couplings of compounds having the D-gluco and D-galacto configurations clearly reflect the influence of the C-4O-4 substitutents on the populations of the C-5C-6 rotamers. A novel type of noise-modulated, heteronuclear decoupling experiment is described.  相似文献

Decarboxylative elimination of methyl 2,3-di-O-benzyl-α-D-glucopyranosiduronic acid (1) with N,N-dimethylformamide dineopentyl acetal in N,N-dimethylformamide gave methyl 2,3-di-O-benzyl-4-deoxy-β-L-threo-pent-4-enopyranoside (3). Debenzylation of 3 was effected with sodium in liquid ammonia to give methyl 4-deoxy-β-L-threo-pent-4-enopyranoside (4). Hydrogenation of 3 catalyzed by palladium-on-barium sulfate afforded methyl 2,3-di-O-benzyl-4-deoxy-β-L-threo-pentopyranoside (5), whereas hydrogenation of 3 over palladium-on-carbon gave methyl 4-deoxy-β-L-threo-pentopyranoside (6). An improved preparation of methyl 4,6-O-benzylidene-α-D-glucopyranoside is also described.  相似文献

A 5-stage synthesis of the title compound (11), the first example of a secondary deoxyfluoroketose, is described. The synthesis comprised the following reaction sequence: D-fructose→1,2:4,5-di-O-isopropylidene-β-D-fructopyranose (4)→1,2:4,5-di-O-isopropylidene-3-O-tosyl-β-D-fructopyranose (3)→ 3,4-anhydro-1,2-O-isopropylidene-β-D-ribo-hexulopyranose (9)→4-deoxy-fluoro-1,2-O-isopropylidene-β-D-xylo-hexulopyranose (11). Fluoride displacement at C-4 in 9 was effected with tetrabutyl-ammonium fluoride in methyl cyanide. Similar treatment of either 3 or 1,2:4,5-di-O-isopropylidene-3-O-tosyl-β-D-ribo-hexulopyranose (5) failed to yield a fluoro derivative. Compound 5 was prepared by the sequence 4→1,2:4,5-di-O-isopropylidene-β-D-erythro-hexo-2,3-diulopyranose (6)→1,2:4,5-di-O-isopropylidene-β-D-ribo-hexulopyranose (7)→5.  相似文献

Oxidative dimerization of 7,8-dideoxy-1,2:3,4-di-O-isopropylidene-d-glycero-α-d-galacto-oct-7-ynopyranoside (1) gave a high yield of the diyne 2, readily reduced by lithium aluminum hydride to the trans,trans-diene (4). The structures of 2 and 4 were established spectroscopically and by degradation of 4 to d-glycero-d-galacto-heptitol (perscitol). A mixture of the alkyne 1 and its 7-epimer 10 was readily oxidized by dimethyl sulfoxide-acetic anhydride to the 6-ketone 11, and the 8-alkene analog was similarly prepared from the alkenes derived from 1 and 10. Likewise, oxidation of 6,7-dideoxy-1,2-O-isopropylidene-α-d-gluco(and β-L-ido)-hept-6-enopyranose gave the corresponding 5-ketone. The acetylenic ketone 11 gave a crystalline oxime and (2,4-dinitrophenyl)hydrazone, the latter being accompanied by the product of attack of the reagent at the acetylene terminus (C-8). Previous work had shown that formyl-methylenetriphenylphosphorane did not convert 1,2:3,4-di-O-isopropylidene-6-aldehydo-α-d-galacto-hexodialdo-1,5-pyranose into the corresponding C8 unsaturated aldehyde, although the latter was obtainable via1 and 10 by an ethynylation-hydroboration sequence. The Wittig route with formylmethylenetriphenylphosphorane is shown to be satisfactory for obtaining C7 unsaturated aldehydes from 3-O-benzyl-1,2-O-isopropylidene-5-aldehydo-α-d-xylo-pentodialdo-1,4-furanose (22) and the 3-epimer of 22, respectively. These reactions provide convenient access to higher-carbon sugars and chiral dienes for synthesis of optically pure products of cyclo-addition reactions.  相似文献

The reaction of 2,3-di-O-acetyl-4-O-benzyl-α,β-d-xylopyranosyl bromide (2) with methyl 2,3-di-O-acetyl-β-d-xylopyranoside gave methyl O-(2,3-di-O-acetyl-4-O-benzyl-β-d-xylopyranosyl)-(1→4)-2,3-di-O-acetyl-β-d-xylopyranoside (22). Catalytic hydrogenolysis of 22 exposed HO-4′ which was then condensed with 2. This sequence of reactions was repeated three more times to afford, after complete removal of protecting groups, a homologous series of methyl β-glycosides of (1→4)-β-d-xylo-oligosaccharides. 13C-N.m.r. spectra of the synthetic methyl β-glycosides (di- to hexa-saccharide) are presented together with data for six other, variously substituted, homologous series of (1→4)-d-xylo-oligosaccharides.  相似文献

The photobromination of 1,5-anhydro-2,3-O-isopropylidene-β-d-ribofuranose gave the corresponding (5S)-5-bromo compound. The reduction of the bromide with triphenyltindeuteride gave (5S)-(5-2H1)-1,5-anhydro-2,3-O-isopropylidene-β-d-ribofuranose, with a chiral purity of 76% at C-5, which was converted to (5R)- and (5S)-(5-2H1)-d-riboses and other ribofuranose derivatives.  相似文献
