Diarrhea may be primarily infectious in origin. Causes can be conveniently classified according to the etiologic agent, which may be viral, chlamydial, bacterial, protozoal, helminthic or fungal. The most common type of infectious diarrhea in Canada is viral. Bacterial infection, particularly staphylococcal and salmonellal, also is relatively common.  相似文献

Electron microscopy of zoocin A-treated sensitive streptococcus cells revealed cytoplasmic disruption and ultimately complete rupture of the cell wall. Culture viability and optical density were shown to decrease rapidly and simultaneously in Streptococcus pyogenes FF22 but less quickly in the relatively more resistant Streptococcus mutans 10449. Zoocin A was shown to cleave hexaglycine in a colorimetric cell-free microtiter assay system, and it is concluded that the killing action of zoocin A, like that of lysostaphin, is most probably the result of direct cleavage of the peptidoglycan cross-links in the cell wall. The relationship between sensitivity to zoocin A and the peptidoglycan cross-linkage structure of Streptococcus zooepidemicus, Lactococcus spp., S. pyogenes, Streptococcus gordonii, Streptococcus oralis, S. mutans, and Streptococcus rattus has been evaluated.  相似文献

Abstract Serratia marcescens cultivated at 30°C was shown to produce a large amount of wetting agent, in contrast to bacteria cultivated at 37°C. The wetting agent, isolated by thin-layer chromatography was examined for surface activaties and was identified, by analysis of hydrolysates and methanolysates, and by infra-red and mass spectrometry, as an aminolipid identical to serratamolide.  相似文献

An enteropathogenic Enterococcus-like agent was isolated from a spontaneous outbreak of diarrhea that occurred in a colony containing neonatal rats. Diarrhea was experimentally reproduced in virus-antibody-free neonatal rats inoculated with this purified "enterococcus." Gram-positive cocci were adhered to the small intestinal villi of affected animals from which the organism was reisolated. The isolate's classification in the genus Enterococcus was confirmed by genetic probe; however, because of its unique fermentation pattern, it could not be definitively speciated. Indirect immunofluorescence assays indicate that this strain of enterococcus and Enterococcus hirae, another strain pathogenic for neonatal rats, differ antigenically. Enterococci should be considered as potential etiologic agents in outbreaks of diarrhea involving neonatal rats and future efforts directed to increasing our understanding of the pathophysiology of this disease.  相似文献

There is a lack of accurate and practical methods that require only small amounts of sample for quantifying virus-like particles (VLP). In this work, gel permeation (GP) HPLC was used to quantify double-layered rotavirus-like particles (dlRLP) produced in insect cells. The proposed methodology utilized two columns in series (pore sizes of 200 and 50 nm) and had a high precision (relative standard deviation below 5%). GP-HPLC not only allowed the routine quantification of dlRLP, but also of assembly intermediaries and other viral structures present in the samples. For the first time, kinetics of dlRLP accumulation could be followed. This methodology is valuable for designing new production processes and for optimizing dlRLP monitoring.  相似文献

Coprophilin, a decalin pentanedienoic acid methyl ester, was isolated from an unidentified fungus by bioassay guided separation. A glass metabolic cage was designed for studies involving newborn rats and small animals up to about 15 g. The metabolic chamber is a jacketed tube, open on both ends and closed with 0-ring clamps. Water is circulated around the cage to maintain a constant and physiologic chamber temperature. Information about the metabolic pathway of nutrients and drugs can thus be studied, and the effects of drugs on metabolism of substrates to CO2 can also be assessed in the newborn animal.  相似文献

Manassantin A (MNS-A), a novel neolignoid, neutral compound shown to possess neuroleptic properties, causes hypothermic response in male and female mice of CD-1 strain when administered by the intra-cerebroventricular (icv), (0.1, 1.0, 3.2, 10 micrograms/mouse), intraperitoneal (ip), (0.1, 0.32, 1.0, 3.2 mg/kg) and oral (0.5, 1.6, 5.0, 16 mg/kg) routes. The hypothermia was found to be dose and time dependent, the maximum decrease of temperature being observed by the icv route (P less than 0.001) after 2 hours. However, ip and oral administration of lower and middle order doses were not very effective but higher doses caused significant (P less than 0.001) reduction of body temperature. The centrally-induced hypothermic response by MNS-A may give future leads as a screening model for antidepressant drugs and can be a useful tool for manipulating physiological and pharmacological processes to understand the central thermoregulatory functions.  相似文献

An assay (enteropooling assay) to test the diarrheogenic property of prostaglandins is described. Fasted rats are given a prostaglandin either orally or subcutaneously, and are killed 30 min later. The entire small intestine is removed and its contents collected into a test tube. The greater the volume of this intestinal fluid, the more diarrheogenic is the prostaglandin. The assay is simple, rapid, quantitative, and predictive of diarrhea. It can be used to grade the relative diarrheogenic activity of prostaglandins as well as to test agents that may block this effect.The accumulation of fluid into the small intestine is called “enteropooling”. It is the sum of (a) the fluid being excreted from the blood into the lumen, and (b) to a lesser extent, the portion of fluid already into the lumen but whose absorption is inhibited by the prostaglandin. The degree of enteropooling depends also on how much fluid flows from the small to the large intestine. Our results support the hypothesis that the diarrhea observed after administration of high doses of prostaglandins is due to accumulation of abundant fluid into the small intestine, and not intestinal hypermotility. This fluid is then carried into the large intestine and eventually expelled as diarrhea.Agents other than prostaglandins were tested for enteropooling activity. Laxatives such as castor oil, hypertonic solutions and bile salts caused enteropooling.  相似文献

AIM: To partially characterize the bacteriocin produced by the GM-1 strain of Enterococcus faecium, isolated from the faeces of a newborn human infant. METHODS AND RESULTS: The bacteriocin produced by E. faecium GM-1 showed a broad spectrum of activity against indicator strains of Escherichia coli, Staphylococcus aureus, Vibrio spp., Salmonella typhimurium, Listeria monocytogenes, Lactobacillus acidophilus, and Streptococcus thermophilus. Treatment of the GM-1 bacteriocin with proteolytic enzymes reduced its inhibitory activities. The bacteriocin was stable at 100 degrees C for 20 min and displayed inhibitory activity at neutral pH. The optimal production of bacteriocin from E. faecium GM-1 was obtained when the culture conditions were pH 6.0-6.5 and 35-40 degrees C. The inhibitory activity of the bacteriocin was not substantially changed by the use of different carbon sources in the media, except when galactose was substituted for glucose. The use of a sole nitrogen source caused a decrease in inhibitory activity. A bacteriocin gene similar to enterocin P was identified from the total DNA of E. faecium GM-1 by PCR and direct sequencing methods. CONCLUSION: E. faecium GM-1, which was isolated from the faeces of a newborn baby, produces an enterocin P-like bacteriocin with inhibitory activity against Gram-positive and Gram-negative bacteria, including food-borne pathogens. SIGNIFICANCE AND IMPACT OF THE STUDY: E. faecium GM-1, isolated from infant faeces, produces a new bacteriocin that is similar to enterocin P. This bacteriocin is heat stable and has a broad antibacterial spectrum that includes both Gram-positive and Gram-negative bacteria.  相似文献

An epizootic of fatal diarrhea occurred in mice which were approximately 10 days of age. The enteric lesions were similar to those reported in lethal intestinal virus of infant mice, but many diseased mice also had necrotic hepatitis. Mouse hepatitis virus antigen was demonstrated in the affected intestine and liver, and a virus that produced syncitium formation in mouse brain tumor cell culture was isolated from the intestines, livers, and brains. The virus was capable of producing intestinal and hepatic lesions similar to the naturally occurring disease after inoculation into suckling mice. Electron microscopy revealed viral particles within affected intestinal epithelial cells of the inoculated mice.  相似文献

The mechanism of liver splanchnomegaly developed in rats bearing a transplantable pituitary mammotropic tumor which secretes large amounts of ACTH and prolactin has been studied. The results indicate that in the first phase of tumor growth liver enlargement could be due to hypertrophy, and later mainly to hyperplasia which overcomes hypertrophy. The adrenal glands were found to play an essential role in the process of liver splanchnomegaly because adrenalectomy prevented the disproportional growth of the liver. Evidence is presented showing that glucocorticoids are the dominant hormones responsible for the development of liver splanchnomegaly.  相似文献

目的研究引起幼儿腹泻的腺病毒的型别。方法取因腹泻住院的幼儿粪便标本.经PCR检测为腺病毒F组(Ad40或Ad41)阳性后,用293细胞分离病毒并培养,当细胞发生典型病变效应,刮下细胞备用:(1)固定,送电镜室检测;(2)按照湖北中医学院检验系实验室常用的方法提取病毒DNA。用SmaⅠ和HindⅢ2种限制性核酸内切酶消化腺病毒DNA并与文献报道的Ad40和Ad41的酶切图谱比较。结果5例阳性标本的PCR产物的大小是519bp.电镜可见细胞核内腺病毒晶格状排列;并且SmaⅠ和HindⅢ的酶切图与文献报道的Ad41图谱一致。结论用PCR和限制性核酸内切酶组合方法检测肠道腺病毒是可行的。  相似文献

The recognition that rotaviruses are the major cause of life-threatening diarrheal disease and significant morbidity in young children has focused efforts on disease prevention and control of these viruses. Although the correlates of protection in children remain unclear, some studies indicate that serotype-specific antibody is important. Based on this premise, current live attenuated reassortant rotavirus vaccines include the four predominant serotypes of virus. We are evaluating subunit rotavirus vaccines, 2/6/7-VLPs and 2/4/6/7-VLPs, that contain only a single VP7 of serotype G1 or G3. In mice immunized parenterally twice, G3 virus-like particles (VLPs) induced a homotypic, whereas G1 VLPs induced a homotypic and heterotypic (G3) serum neutralizing immune response. Administration of three doses of G1 or G3 VLPs induced serum antibodies that neutralized five of seven different serotype test viruses. The inclusion of VP4 in the VLPs was not essential for the induction of heterotypic neutralizing antibody in mice. To confirm these results in another species, rabbits were immunized parenterally with two doses of 2/4/6/7-VLPs containing a G3 or G1 VP7, sequentially with G3 VLPs followed by G1 (G3/G1) VLPs, or with live or psoralen-inactivated SA11. High-titer homotypic serum neutralizing antibody was induced in all rabbits, and low-level heterotypic neutralizing antibody was induced in a subset of rabbits. The rabbits immunized with the G1 or G3/G1 VLPs in QS-21 were challenged orally with live G3 ALA rotavirus. Protection levels were similar in rabbits immunized with homotypic G3 2/4/6/7-VLPs, heterotypic G1 2/4/6/7-VLPs, or G3/G1 2/4/6/7-VLPs. Therefore, G1 2/4/6/7-VLPs can induce protective immunity against a live heterotypic rotavirus challenge in an adjuvant with potential use in humans. Following challenge, broad serum heterotypic neutralizing antibody responses were detected in rabbits parenterally immunized with G1, G3/G1, or G3 VLPs but not with SA11. Immunization with VLPs may provide sufficient priming of the immune system to induce protective anamnestic heterotypic neutralizing antibody responses upon subsequent rotavirus infection. Therefore, a limited number of serotypes of VLPs may be sufficient to provide a broadly protective subunit vaccine.  相似文献
