Summary Organic matter was extracted with a mixture of 0.1M Na4P2O7 and 0.1M NaOH from soil and a commercial organic matter product, Aqua Humus, and separated into humic and fulvic acids according to their solubility in acid and alkaline solutions. They were purified by passage through a cation exchange resin, freeze-dried, and used for characterization and seedling growth studies. The CEC increased with pH for both humic and fulvic acids. Data from tobacco seedling studies show that humic and fulvic acids from both sources increased root number and length at low concentrations while at high concentrations, humic acids inhibited length and number of roots but fulvic acids had no effect. The results support the proposition that fractions of organic matter referred to as humic and fulvic acids affect root development by means other than as a source of nutrients.  相似文献

Some effects of humic acids, formed during the breakdown of organic wastes by earthworms (vermicomposting), on plant growth were evaluated. In the first experiment, humic acids were extracted from pig manure vermicompost using the classic alkali/acid fractionation procedure and mixed with a soilless container medium (Metro-Mix 360), to provide a range of 0, 50, 100, 150, 200, 250, 500, 1,000, 2,000, and 4,000 mg of humate per kg of dry weight of container medium, and tomato seedlings were grown in the mixtures. In the second experiment, humates extracted from pig manure and food wastes vermicomposts were mixed with vermiculite to provide a range of 0, 50, 125, 250, 500, 1,000, and 4,000 mg of humate per kg of dry weight of the container medium, and cucumber seedlings were grown in the mixtures. Both tomato and cucumber seedlings were watered daily with a solution containing all nutrients required to ensure that any differences in growth responses were not nutrient-mediated. The incorporation of both types of vermicompost-derived humic acids, into either type of soilless plant growth media, increased the growth of tomato and cucumber plants significantly, in terms of plant heights, leaf areas, shoot and root dry weights. Plant growth increased with increasing concentrations of humic acids incorporated into the medium up to a certain proportion, but this differed according to the plant species, the source of the vermicompost, and the nature of the container medium. Plant growth tended to be increased by treatments of the plants with 50-500 mg/kg humic acids, but often decreased significantly when the concentrations of humic acids derived in the container medium exceeded 500-1,000 mg/kg. These growth responses were most probably due to hormone-like activity of humic acids from the vermicomposts or could have been due to plant growth hormones adsorbed onto the humates.  相似文献

Humic acid in the form of sodium humate and fulvic acid markedly increased the growth and efficiency of nitrogen fixation ofAzotobacter chroococcum. Fulvic acid proved more effective than the sodium humate. The effectiveness of these substances was more pronounced in increasing the number of cells than affecting the nitrogen fixation. The increases in growth and nitrogen fixation were in direct proportion to the quantities of sodium humate and fulvic acid applied up to 500 and 700 ppm respectively. Thereafter the growth was restricted, although it was not inhibited even at 1,400 ppm.  相似文献

Small-angle neutron scattering experiments have been made on solutions of humic acid aggregates with an acidity corresponding to pH 5.0 and at 0.1 M ionic strength. We observe power-law decay of the intensity over one decade of the scattering vector, Q, indicating that the aggregates are fractal. We explain the normalized intensity in the entire Q-range by assuming that the humic acid particles can be described by building units of a radial size, 25 Å, aggregated into clusters with an average radius of 400–500 Å. For humic acids obtained from two different sources, we determine the fractal dimension, D = 2.3 ± 0.1. For small values of Q, the measured data of one of the samples extend into the Guinier range giving an average radius of gyration of 320 ± 20 Å. Correspondence to: R. Österberg  相似文献

Fractions of humic acids (HA-K+) of molecular mass between 500 and 300 000 and exceeding 300 000 showed a very high nitrite depleting ability, whereas the fraction of HA-K+ with molecular mass lower than 500 had little or no such effect. Autoclaving HA-K+ (121 °C, 20 min) decreased the nitrite-depleting ability to about a half. This observation correlates with the results of mutagenic studies in Arabidopsis thaliana, demonstrating the inhibitory effect of HA-K+ and its fractions on the formation of mutagenic N-methyl-N-nitrosourea (MNU) from the nitrosation mixture of N-methylurea and nitrite. Nonfractionated HA-K+ had no inhibitory effect towards the mutagenic activity of preformed direct acting MNU or towards the activation of the promutagen N-nitrosodimethylamine to a mutagenic product.  相似文献

Humic acids are known for their overall positive health and productivity effects in animal feeding trials and, controversially, as an aetiological factor of cancer. We tried to assess the in vitro effect of humic acids from a selected source in Slovakia when used at recommended prophylactic dosage. We investigated antioxidant properties, enzymatic and non-enzymatic antioxidant defence system in liver mitochondria and cultured cancer cell lines in vitro. We observed a significant decrease in superoxide dismutase activity after humic acids treatment irrespective of dissolving in dimethyl sulphoxide or direct addition to mitochondria suspension in a respiration medium. Activities of other antioxidant enzymes measured, such as glutathione peroxidase and glutathione reductase, showed no significant differences from the control as well as the reduced glutathione content. Percentage of inhibition by humic acids of superoxide radical indicated lower efficacy compared with that of hydroxyl radical. Survival of six different cancer cells lines indicated that only the acute T lymphoblastic leukaemia cell line was sensitive to the tested humic acids. Despite relatively low solubility in aqueous solutions, humic acids from the selected source participated in redox regulation. By recapturing the radicals, humic acids reloaded the antioxidant defensive mechanism. Results from in vitro study conducted with humic acids from the natural source showed potential of these substances as promising immunity enhancing agents.  相似文献

Summary Functional group content of undegraded humic acids was compared with that of humic acids which had been partially degraded by a strain ofPenicillium frequentans. It was found that cleavage of humic acids by this organism did not involve reduction of the carboxyl groups. It was suggested that one of the steps involved was hydrolysis of the ether bonds present in humic acids.  相似文献

Capacity for sorption of humic acid (HA) from water solutions was shown for 38 bacterial strains. Isotherms of HA sorption were determined for the cells of 10 strains. The bonding strength between the cells and HA (k) and the terminal adsorption (Q max) determined from the Langmuir equation for gram-positive and gram-negative bacteria were reliably different. Gram-positive bacteria sorbed greater amounts of HA than gram-negative ones (Q max = 23 ± 10 and 5.6 ± 1.2 mg/m2, respectively). The bonding strength between HA and the cells was higher in gram-negative bacteria than in gram-positive: k = 9 ± 5 and 3.3 ± 1.1 mL/mg, respectively.  相似文献

The inhibitory effect of peat humic acids on the hydrolysis of N-acetyl-L-tyrosine ethyl ester and N-benzoyl-L-leucine methyl ester by alpha-chymotrypsin and subtilism has been studied. Samples of humic acids with M(W) approximately 18,000 have been used in experiments. The results of kinetic studies indicates the mixed type of inhibition of proteinase activity by humic acids. The meanings of inhibition constants under the action of humic acids on alpha-chymotrypsin and subtilism have been calculated.  相似文献

Summary The extent of contamination of soil humic fractions by silica gel, sodium bicarbonate and sodium carbonate was investigated by infrared spectroscopy. Humic fractions were extracted with 0.1 N NaOH from a surface sample of a Davidson soil (Rhodic Paleudult, Reddish-brown Lateritic soil). After careful purification, the humic preparations were characterized by scanning with the Beckman IR-18A spectrophotometer using the micro KBr pellet technique. The spectra of the various humic fractions corresponded to the respective spectra reported in the literature. No indications were obtained for the presence of silica gel, sodium bicarbonate or sodium carbonate. Strong absorption at 1000 cm-1, as exhibited by non-purified humic acid, could be caused by Si-O vibrations. Humus formation and the elemental composition of macromolecules of humic acids of automorphic Siberian soils (gray forest soils, chernozems, and chestnut soils) have been considered in comparison with those of the analogous soils of the European part of Russia. It has been found that an increase in the continentality of climate leads to significant changes in the structure of humic acids in soils of similar origin in the meridional direction.  相似文献

This paper considers the informative importance of one of the main features that characterize humic acids of soils at the diagnosis of the natural environment of their formation in both the present and the past. It is shown that the ratio of elements in the humic acids clearly corresponds to the landscape conditions and quantitative characteristics of climatic parameters. They persist over time and can serve as one of the reliable indicators in assessing the environment of different periods of paleogeographic history.  相似文献

Aqueous mixtures of oppositely charged polyelectrolytes undergo associative phase separation, resulting in coacervation, gelation, or precipitation. This phenomenon has been exploited here to form DNA gel particles by interfacial diffusion. We report on the formation of DNA gel particles by mixing solutions of DNA (either single-stranded (ssDNA) or double-stranded (dsDNA)) with solutions of cationic surfactant cetyltrimetrylammonium bromide (CTAB). By using CTAB, the formation of DNA reservoir gel particles, without adding any kind of cross-linker or organic solvent, has been demonstrated. Particles have been characterized with respect to the degree of DNA entrapment, surface morphology, and secondary structure of DNA in the particles. The swelling/deswelling behavior and the DNA release have been investigated in response to salt additions. Analysis of the data has suggested a higher degree of interaction between ssDNA and the cationic surfactant, confirming the stronger amphiphilic character of the denatured DNA. Fluorescence microscopy studies have suggested that the formation of these particles is associated with a conservation of the secondary structure of DNA.  相似文献
