To study the origin and speciation of plants in oceanic islands, electrophoretic analyses have been done on three endemic species ofSymplocos in the Bonin Islands as well as on three other species;S. kuroki, S. nakaharae andS. tanakae which are considered to be closely related to the Bonin endemics. There occur three species:S. kawakamii, S. pergracilis andS. boninensis in Bonin. The genusSymplocos is one which is considered to be diversified in the Bonin Islands. Seven enzyme systems presumed to be encoded by 18 loci were examined. The genetic diversity was low in the island species, as reported in some oceanic island plants of Hawaii and the Bonin Islands. The three endemics share high genetic identities and they clustered together in the tree drawn by the UPGMA method, suggesting that they are a monophyletic group, that is, they result from a single introduction.  相似文献

The genusPittosporum includes about 160 species. Four species ofPittosporum occur in the Bonin Islands, and all of these are endemic to the islands. Electrophoretic studies of the four endemic species,P. tobira, from the Japanese mainland, andP. lutchuense var.denudatum from the Ryukyu Islands, were used to determine the origin and speciation pattern of the endemic species. 259 individuals were sampled from ten populations. Twenty loci in nine enzyme systems were resolved and used to calculate the gene frequencies for each population. A low genetic diversity was observed in three of the Bonin Island species, as is reported for other oceanic island plants. The exception,P. boninense, has the largest population size and widest distribution. A dendrogram generated by the UPGMA method shows two clusters. One consists of only the Bonin endemics, suggesting a monophyletic origin for these species.  相似文献

The genusCrepidiastrum is distributed in East Asia and includes 7 species. In the Bonin Islands, three species ofCrepidiastrum occur, and all of them are endemic to the islands. For detecting the origin and speciation of these endemic species, electrophoretic studies have been done in three endemic species of the Bonin Islands as well as in the remaining four species ofCrepidiastrum, andYoungia denticulata which is considered to be closely related toCrepidiastrum. A total of 386 individuals were sampled from 14 populations. As a result, 17 loci of 10 enzyme systems were resolved and gene frequencies for each population were calculated. The genetic variability was low in island species, as reported in some oceanic island plants. Four groups were recognized in the dendrogram generated by the UPGMA method. The Bonin endemics were clustered together, suggesting a monophyletic origin.C. ameristophyllum andC. linguaefolium were found to be genetically very similar, and this may suggest recent and rapid speciation within the islands.  相似文献

The interrelationship of the ten species of the genusTyphonium and related genera in subtribe Arinae of the Araceae was inferred by chloroplast DNA restriction fragment analysis. A total of 42 site mutations were observed and 26 site mutations were shared by two or more species. A majority rule consensus tree was made by performing 100 bootstrap replicates using Wagner Parsimony. Two groups ofTyphonium were recognized significantly as monophyletic groups, i.e. 1)Typhonium larsenii andT. kunmingense, and 2)T. trilobatum, T. blumei andT. flagelliforme.  相似文献

Restriction site mutations in the chloroplast (cpDNA) and ribosomal DNA (rDNA) were examined in 41 populations representing five of the seven recognized species of the genusRobinsonia, which is endemic to the Juan Fernandez Islands. No intraspecific variation was detected for cpDNA but one population of one of the species (R. evenia) had a restriction site mutation in rDNA not detected elsewhere. No restriction site mutations were unique to all species ofRobinsonia relative to the species ofSenecio used as outgroups. All 13 mutations (eight from cpDNA and five from rDNA) are restricted to single species, and thus provide no cladistically useful information within the genus. The distribution of mutations is concordant with the hypothesis of a rapid adaptive radiation ofRobinsonia subsequent to the dispersal of its ancestor to Masatierra.  相似文献

Origin and evolution of endemic plants of the Bonin (Ogasawara) Islands   总被引:1,自引:0,他引:1  
The Bonin Islands are typical oceanic islands, located at the western part of the North Pacific Ocean and approximately 1,000 km south of mainland Japan. This archipelago consists of about 20 small islands. Although floristic diversity is low due to the small area and limited environmental diversity, the Bonin Islands harbor unique endemic flora as in other well-known oceanic islands. This paper presents a brief summary of the results obtained from recent studies on the endemic flora of the Bonin Islands. The results are reviewed in relation to the four stages of the evolution of endemic flora in the oceanic islands; migration, establishment, enlargement and diversification. The ancestors of the flora originated mostly from tropical and subtropical Southeast Asia or mainland Japan by rare events of long distance dispersal. The proportion of bird-dispersed species is relatively high as for other oceanic islands. Genetic data sets obtained from allozyme variation in some endemic species suggest that migration occurred several million years ago and genetic diversity is correlated with current population size. At the time of establishment, self-compatible plants are expected to have an advantage. However, the percentage of dioecious plants is relatively high. This is partly due to evolutionary changes from hermaphroditic ancestors to dioecy which occurred in two genera in the Bonin Islands. In addition, there are some examples of evolutionary changes from herbaceous ancestors to woody endemics. Adaptive radiation is found in some genera, although the number of congeneric endemic species is less than five. Studies of allozyme variation inPittosporum, Symplocos andCrepidiastrum showed that genetic identity is generally very high between congeneric species in spite of their distinct morphologies. This result suggests that divergence of these species occurred rather recently and distinct morphological differences are based on a limited number of genetic changes.  相似文献

Chloroplast DNA (cpDNA) restriction site variation was examined in five species ofDesmodium subgenusPodocarpium (Leguminosae; Papilionoideae; Desmodieae). Twenty four phylogenetically informative cpDNA mutations were scored. The cladistic analysis of characters based on the 24 mutations resulted in the most parsimonious tree which supports the monophyly of the subgenus.Desmodium elegans of subgenusDollinera was the sister group of subgenusPodocarpium in this tree. The groupings obtained from the cpDNA characters were consistent with the present infrageneric classification system for the subgenus except for the infraspecific taxa ofD. podocarpum. Three groups withinD. podocarpum, which were incongruent with the infraspecific classification of the species, were distinguished by a total of four site mutations. The first group consisted of subsp.podocarpum, subsp.fallax, and subsp.oxyphyllum var.oxyphyllum; the second subsp.oxyphyllum var.oxyphyllum; and the last subsp.oxyphyllum var.oxyphyllum and var.mandshuricum.  相似文献

The examination of morphological traits has failed to resolve the tribal placement of Marshallia. Suggested relationships for this anomalous genus have, at various times, included Eupatorieae, Heliantheae, Vemonieae, and Inuleae. Chloroplast DNA restriction site mapping, using Bamadesiinae (Mutisieae) as the outgroup, revealed 981 restriction site mutations, 332 of which were phylogenetically informative, for 60 genera representing 15 tribes of Asteraceae. Wagner parsimony produced 36 equally parsimonious tress of 729 steps, and Dollo parsimony produced 34 equally parsimonious trees of 759 steps. Monophyletic groups, resulting from the Wagner analysis, were further tested with the bootstrap method. The placement of Marshallia in the Heliantheae-Tageteae-Coreopsideae-Eupatorieae complex was consistent for all trees produced. Tageteae and Coreopsideae form the sister group to paraphyletic Heliantheae, with Marshallia sharing its most recent common ancestor with Galinsoga, Palafoxia, and Bahia. The Eupatorieae form a monophyletic clade that is derived from helianthoid ancestors.  相似文献

The morphology and function of the flowers of three species of the genusCallicarpa, C. glabra, C. nisnimurae andC. subpubescens, endemic to the Bonin Islands, revealed that all three species were dioecious. Male plants had short-styled flowers with sterile ovaries, while female plants had long-styled flowers with non-germinating inaperturate pollen grains. Such dioecism has never been reported from the family Verbenaceae. This curious sexual system with non-germinating pollen grains may have been maintained as a reward to insect pollinators.  相似文献

Restriction sites for six enzymes were mapped for the plastid DNAs of 25 species of Eragrostideae, one species of Cynodonteae (Eustachys distichophylla), and one species of Pooideae. Of the 124 restriction sites observed, 67 were variably present and shared by two or more species. These data were analyzed by the parsimony method using equal and unequal weights and by bootstrap analysis. The cladistic analyses established that members of the Muhlenbergiinae, including the genera Muhlenbergia, Blepharoneuron, Bealia, Chaboissaea, Lycurus, and Pereilema, share seven restriction site mutations and are strongly supported by the data as a monophyletic subtribe. Surprisingly, Redfieldia flexuosa also clustered with the Muhlenbergiinae in the analysis, perhaps indicative of a past interspecific hybridization event. The restriction sites data also weakly support a relationship (six shared mutations) between Erioneuron, Munroa, and Dasyochloa.  相似文献

To elucidate the relationships among Diospyros kaki and species closely related in previous studies, the nuclear ribosomal internal transcribed spacer (ITS) DNA sequence and the chloroplast matK gene were sequenced and compared with those of nine Diospyros species from Thailand, four species from temperate regions, and one species of southern Africa, D. lycioides. Maximum parsimony, maximum likelihood, and neighbor joining analyses of the matK and ITS data sets revealed that D. kaki is closely related to two diploid species, D. oleifera and D. glandulosa. D. kaki, D. glandulosa, and D. oleifera were placed differently in the trees obtained from ITS and matK data sets, suggesting that hybridization and/or introgression may have occurred during the development of these species. D. kaki was not found to be closely related to D. ehretioides, a diploid species from Thailand. These results differed from a prior analysis of this genus performed with chloroplast DNA (cpDNA) restriction site mutations in 3.2- and 2.1-kb amplified sequences. The results supported Ng’s hypothesis that D. glandulosa and D. kaki may share a common ancestor. D. oleifera was also closely associated with D. kaki.  相似文献

Summary Restriction fragment patterns of mitochondrial DNAs (mtDNAs) from 13 carrot cultivars (Daucus carota ssp. sativus), wild carrot (ssp. carota), ssp. gummifer, and D. capillifolius were compared with each other using four restriction endonucleases. The mtDNAs of the 13 carrot cultivars could be classified into three distinct types — I, II and III — and were also clearly distinguishable from the mtDNAs of wild carrot (type IV), gummifer (V) and D. capillifolius (VI). The proportions of common restriction fragments (F values) shared by two of the three mtDNA types (I, II and III) of carrot cultivars were approximately 0.5–0.6. The F values were 0.4–0.5 for mitochondrial genomes between wild carrot, ssp. gummifer and D. capillifolius. The mitochondrial genomes between wild carrot and the carrot cultivars showed closer homologies those between wild carrot, ssp. gummifer, and D. capillifolius. The diversity of the mitochondrial genomes among the carrot cultivars is too high to presume that it was generated from the cytoplasm of only one common ancestor during the relatively short history of carrot breeding. These results suggested that the three types of cytoplasms found in the carrot cultivars might have existed in a prototype of D. carota in pre-historical times.  相似文献

Intraspecific chloroplast DNA polymorphisms were examined for 51 populations of seven species in the genus Krigia. A total of 1,100 restriction sites was surveyed and 46 of these were variable at the intraspecific level. Twenty-two of the variable sites were found within K. virginica, giving this species one of the highest levels of intraspecific chloroplast DNA divergence of any examined species. In contrast, no restriction site variation was detected within K. dandelion, K. wrightii, and K. occidentalis. Five polymorphisms were identified from the 16 populations of the K. cespitosa-gracilis complex, but no mutations distinguished the K. cespitosa and K. gracilis types. Krigia montana and K. biflora showed 11 and eight restriction site polymorphisms, respectively. The chloroplast genome of the hexaploid K. montana was derived from the diploid K. biflora rather than the tetraploid K. montana. High levels of polymorphism were found in species having different ploidy levels, such as K. virginica, K. biflora, and K. montana. Furthermore, most mutations found in these three species were recorded from the tetraploid lineages. As a result, evolutionary rates between different ploidy levels differ significantly. The chloroplast DNA restriction site data suggest that all surveyed populations of the autotetraploid K. virginica originated from a common ancestor. Our results also indicate that certain regions of the chloroplast genome have changed more rapidly than others and have the potential to resolve evolutionary questions at the population level.  相似文献

A 4,100-base pair (bp) region of the chloroplast genome, amplified via the polymerase chain reaction, was obtained from 14 species of the genus Astragalus and mapped with 23 restriction enzymes. The amplified region encompassed the chloroplast genes RNA polymerase Cl (rpoCl; 90.8% of the gene) and RNA polymerase C2 (rpoC2; 32.7% of the gene) including the intron in rpoC1 and the intergenic spacer between the two genes. Approximately 144 sites (615 bp) were identified; 37 restriction site mutations and one 10-bp length mutation were detected. Estimated interspecific sequence divergence values ranged from 0.00% to 3.92%. Phylogenetic analysis with Wagner and Dollo parsimony both resulted in a single 41-step tree with a consistency index of 0.951. The relative positions of 115 restriction sites were mapped. The insertion and ten of the restriction site mutations mapped to the intron in rpoC1, 18 site mutations mapped to the rpoC1 exons, three site mutations mapped to rpoC2, three site changes mapped to the intergenic spacer, and four site changes were not mapped. This study demonstrates the utility of restriction site analysis of PCR-amplified chloroplast DNA to the study of plant phylogenetic relationships and molecular evolution.  相似文献

Twelve microsatellite loci were isolated and characterized for Melicope quadrilocularis, an insular endemic tree species of the Bonin Islands. The observed number of alleles at each locus ranged from 1 to 18. The range of expected heterozygosity was 0.0000–0.9445. The inter-specific applicability of these loci was evaluated by analyzing two other endemic species and one endemic variety of Melicope that are also distributed on the Bonin Islands. All primer pairs for the 12 loci tested successfully amplified the loci in all taxa, except for primers for four loci in M. nishimurae.  相似文献

Thirty species and one variety of Symplocos (Symplocaceae), including all taxa distributed in Japan, were phylogenetically analyzed with DNA sequence data. The evolution of morphological characters is discussed on the basis of the phylogenetic relationships obtained. All species were Asian, except one, S. austromexicana, from Mexico. The nuclear ribosomal internal transcribed spacer (ITS) region and two intergenic spacer regions, between trnL and trnF, and between trnH and psbA of chloroplast DNA were used. The topologies of trees obtained from ITS and the chloroplast intergenic spacers are largely congruent. S. sonoharae, the representative of the subgenus Symplocos in Asia, was sister to all other species. The position of the American species, S. austromexicana, which also belongs to the subgenus Symplocos, was not well resolved. The phylogenetic tree based on combined sequence data largely supports the monophyletic origin of the infrageneric sections proposed earlier. However, the phylogenetic relationship between them is not well resolved, probably due to rapid diversification. The section Palura, a deciduous group, is well defined in the DNA analysis, suggesting its independent status in the genus Symplocos. In spite of their morphological divergence, the three endemic species of the Bonin Islands are monophyletic. The occurrence of curved seeds seems to be homoplastic, scattered over the phylogenetic tree without showing a particular infrageneric relationship.  相似文献

After more than 50 years, the rare autochthonous species, Capsicum lanceolatum, of Mesoamerica was rediscovered in a virgin remnant of the Guatemala cloud forest, Mario D'Arcy Avila Biotopo el Quetzal, a nature reserve for the resplendent quetzal. At all previous known locations C. lanceolatum was extirpated. Specific variation in flower and seed traits of the C. lanceolatum from the Biotopo el Quetzal population suggests it is a heretofore undiscovered population of this species. The usefulness of nature preserves in saving designated and cryptic species is illustrated.  相似文献

Chloroplast DNA restriction site variation inHemerocallis middendorfii Trautv. & Meyer was investigated to deduce the historical differentiation process that had taken place in populations among three districts Hokkaido, Honshu and the Island of Tobishima in Japan as distinct from the morphological data. The populations selected were eight in total, consisting of two populations from Honshu, one each from the Islands of Sado and Tobishima and four from Hokkaido. Twelve endonuclease restriction enzymes were used. Six restriction site mutations were found among the populations investigated. A single, most parsimonious phylogenetic tree is generated, and revealed that the four populations of Hokkaido and two populations of the Islands of Sado and Tobishima are monophyly, respectively.  相似文献

Restriction site variation in chloroplast DNAs (cpDNAs) of Coreopsis section Coreopsis was employed to assess divergence and phylogenetic relationships among the nine species of the section. A total of fourteen restriction site mutations and one length mutation was detected. Cladistic analysis of the cpDNA data produced a phylogeny that is different in several respects from previous hypotheses. CpDNA mutations divide the section into two groups, with the two perennial species C. auriculata and C. pubescens lacking any derived restriction site changes. The other seven species are united by five synapomorphic restriction site mutations and the one length mutation. These seven species fall into three unresolved clades consisting of 1) the remaining three perennial species, C. grandiflora, C. intermedia, and C. lanceolata; 2) three annual species, C. basalis, C. nuecensoides, and C. nuecensis; and 3) the remaining annual, C. wrightii. The cpDNA data suggest that, although the perennial habit is primitive within the section, the annual species of section Coreopsis have likely not originated from an extant perennial species. The estimated proportion of nucleotide differences per site (given as 100p) for the cpDNAs of species in the section ranges from 0.00 to 0.20, which is comparable to or lower than values reported for other congeneric species. The low level of cpDNA divergence is concordant with other data, including cross compatibility, interfertility and allozymes, in suggesting that species of the section are not highly divergent genetically.  相似文献
