We have modelled a phospholipid bilayer as two monolayer sheets which interact with each other by a coupling which depends upon the states of the lipid hydrocarbon chains in each sheet. We make use of a model (Georgallas and Pink 1982a) and its parameters, already used to study monolayer phase changes at the LC-LE transition, in order to study the lipid main transition. Although the monolayer coexistence curve can be calculated exactly, we have made use of high-temperature series expansions to calculate the critical point of the bilayer. We also present the results of computer simulations on triangular lattices for the pressure-area isotherms. We find: (i) the interaction between the sheets of a DPPC bilayer is about 1.5–2% of the maximum interaction within the plane of each sheet; (ii) the internal lateral pressure of a DPPC bilayer is about 30.5 dyne/cm; (iii) the bilayer transition enthalpy depends sensitively upon the coupling between the sheets. Should this coupling vary from sample to sample (due, possibly, to its preparation) then very different values of transition enthalpy may be measured. Summary A mathematical treatment is given for the flux of ions of one charge sign across lipid bilayer membranes. This treatment is a generalization of a previous analysis of the membrane conductance by D. Walz, E. Bamberg and P. Läuger which was restricted to systems with negligible space charge in the membrane. The present theory includes space charge effects, and it is no longer assumed that the electric field strength in the membrane is constant. It is found that the ohmic membrane conductivity 0 is reduced by space charges; if only ions of one charge sign are soluble in the membrane, 0 approaches a limiting value for increasing concentration of the permeable ion in the aqueous solution. The theory also predicts the range in which the constant field approximation is valid. It is found that space charge effects become predominant when the mean concentration of the permeable ion in the membrane exceeds 5×10–5 m. The currentvoltage characteristic of the membrane remains practically linear even in the presence of a high space charge. It is therefore concluded that the experimentally observed nonlinearity is caused mainly by the distortion of the potential energy profile of an ion due to image forces.  相似文献

Summary The kinetic parameters for the steady-state rate of hydrolysis of egg phosphatidylcholine in multilamellar vesicles by bee venom phospholipase A2 are measured in the presence of 27 alkanols and several organic solvents. In general, small nonpolar solutes like enflurane, tetrahydrofuran, benzene, chloroform and diethylether do not promote the hydrolysis of multilamellar vesicles. The rate of hydrolysis shows a biphasic dependence upon the alkanol concentration for all higher (C5–C9) alcohols examined, i.e., an optimal rate of hydrolysis is observed at a characteristic concentration for each alcohol. The alkanol to lipid mole ratio (D/L ratio) in the bilayer at the peak activating concentration of an alkanol was computed from its bilayer/water partition coefficient. The branched chain alcohols induce peak activation of hydrolysis at lowerD/L ratios in the bilayer than the corresponding straight chain analogs. Similarly, the longer chainn-alkanols at peak activating concentration have a lowerD/L ratio than the corresponding lower alcohols. Both theK m andV m for phosphatidylcholine increase as a function of the chain length of the activating alcohol. These kinetic parameters also depend upon the position of the substituents on the activating alcohols. Both theD/L ratio andV m for an alcohol are found to change with the cross-sectional area of the activating alcohol across its long axis: alcohols with a more asymmetric cross-section exhibit higherV m and a lowerD/L ratio. Such correlations ofV m andD/L ratio with the molecular parameters of the alkanols are interpreted to suggest that the accessibility of the substrate molecule in the bilayer to the phospholipase is modulated by the free space introduced by the alkanols in the bilayer.Effect of tetradecane derivatives and A2C (a membrane fluidizing agent) on the phase transition characteristics of DPPC bilayers, and their susceptibility to phospholipase A2 from bee venom and pig pancreas is also reported. These solutes cause a broadening of the transition profile and reduce the size of the cooperative unit and the enthalpy of transition. These effects depend upon the mole fraction of a solute in the bilayer; however, equal concentrations of these solutes do not induce equal response. Susceptibility of the modified bilayers to phospholipase A2 depends not only upon the structure of the solute but also upon the source of the enzyme. The data show that the activity of the membrane-bound enzyme is modulated to different extents by different solutes, and the bilayer perturbing ability of these solutes may be related to the asymmetry of their cross-sectional area and to the free space introduced by the alkanols in a bilayer.  相似文献

Summary We present a quantitative theory that relates the fluorescence intensityvs. temperature (I vs. T) profile of a fluorescent-labeled two-component lipid bilayer to the phase diagram of the bilayer and the partition coefficientK of the fluorophore between fluid and solid phases of the bilayer. We show how the theory can be used to evaluateK from experimentalI vs. T profiles and the appropriate phase diagrams as well as to understand the different shapes ofI vs. T profiles obtained with particular fluorophores and phase diagrams. Using calculatedI vs. T graphs, we discuss the meaning of parameters, such as midpoint of the phase transition and onset and termination of a transition, which are often used to characterize phase transitions on the basis of fluorescence intensityvs. temperature profiles.  相似文献

Sporidesmin, a mycotoxin fromPithomyces chartarum is a hydrophobic molecule. It can therefore be easily incorporated in the cell membrane, where it is likely to cause changes in the bilayer organization and the properties of membrane proteins. In order to understand the redox behaviour of sporidesmin in a hydrophobic environment, we have investigated the effects of oxidized and reduced sporidesmin on the phase transition properties of bilayers and on the susceptibility of bilayers to pancreatic phospholipase A2 (PLA2). The changes induced by sporidesmin in the thermotropic phase transition profiles of dimyristoyl-sn-3-phosphatidyl choline (DMPC) bilayers were similar to those caused by solutes known to localize in the glycerol-backbone region of the lipid bilayer, suggesting a similar localization for oxidized and reduced sporidesmin. Neither form of toxin disrupt the bilayer or membrane organization even at relatively high mole fractions. At concentrations <10 mole% both forms partitioned equally well in the gel and liquid-crystalline phases, whereas at higher concentrations (30 mole%) reduced sporidesmin is preferentially localized in the liquid-crystalline phase. These effects of sporidesmin on the phase properties of DMPC vesicles were also reported by the fluorescence behavior of 10-pyrenedecanoic acid (PDA). The effects of oxidized and reduced sporidesmins on PLA2 kinetics are consistent with their ability to perturb bilayer organisation.  相似文献

Summary The cyclic polyether XXXII, a neutral, lipid soluble molecule, produces large increases in the conductance of bilayer membranes formed from a variety of lipids. The conductance increases linearly with the concentration of alkali metal cation but with the square, and at higher concentrations the cube, of the polyether concentration. This implies that two or three polyether molecules combine with a single cation to carry it across the membrane. In the presence of XXXII the bilayer is permeable solely to cations and the membrane potential is described by an equation of the Goldman-Hodgkin-Katz type. The permeability ratios determined from potential measurements are independent of salt concentration, decrease in the sequence Cs>Rb>K>NH4>Na>Li(1.0,0.25, 0.15, 0.075, 0.007, 0.0013) and are equal to the conductance ratios at low (e.g. 10–3 m) salt concentration. At higher salt concentrations, the permeability and conductance ratios are not equal and maxima in the conductancevs. salt concentration curves are observed. Both these phenomena are postulated to be caused by the formation of relatively impermeant 11 polyether cation complexes in the aqueous phase. The 11 aqueous association constants deduced from bilayer measurements decrease in the sequence K>Rb>Na>NH4>Cs>Li (120, 34, 26, 19, 12, 4 liters per mole) and agree quantitatively with the literature values for the more water soluble polyether XXXI, which lacks only thet-butyl groups of XXXII.  相似文献

Summary Charge-pulse relaxation studies with the positively charged PV-K+ complex (cyclo-(d-Val-l-Pro-l-Val-d-Pro)3) and the negatively charged lipophilic ion dipicrylamine (DPA) have been performed in order to study the influence of structural properties on ion transport through lipid bilayer membranes. First, the thickness of monoolein membranes was varied over a wide range using differentn-alkanes and slovent-free membranes. The thickness (d) of the hydrocarbon core of these membranes varied between 4.9 and 2.5 nm. For both transport systems the partition coefficient was found to be rather insensitive to variations ind. The same was valid for the translocation rate constantk MS of PV-K+, whereas a strong increase of the translocation rate constantk i of DPA-with decreasingd was observed. In a further set of experimental conditions the structure of the lipids, such as number and position of the double bonds in the hydrocarbon chain and its chain length as well as the nature of the polar head group, was varied. The translocation constantk MS of PV-K+ transport was found to be much more sensitive to these variations thank i of DPA-.Much larger variations ink i andk MS were observed in membranes made from lipids with ether instead of ester linkages between glycerol backbone and hydrocarbon chain. The results are in qualitative agreement with the surface potentials of monolayers made from corresponding lipids. Increasing amounts of cholesterol in membranes of dioleoylphosphatidylcholine caused a strong decrease ofk MS (PV-K+), whereask i was found to be rather insensitive to this variation.In monoolein membranes cholesterol causes a decrease ofk MS up to sixfold and a increase ofk i up to eightfold. The partition coefficient of DPA was insensitive to cholesterol, whereas of PV-K+ was found to decrease about eightfold in these membranes. The influence of cholesterol onk MS is discussed on the basis of viscosity changes in the membrane and the change ink i of DPA and of PV-K+ on the basis of a possible change of the dipole potential of the membranes. The other sterols, epicholesterol and ergosterol cause no change in the kinetics of the two probes.The different influence of membrane properties like thickness, viscosity, and dipole potential on the two transport systems is discussed under the assumption that the adsorption planes of the two probes have different positions in a membrane. Possibly because of a larger hydrophobic interaction, the adsorption plane of PV-K+ is located more towards the hydrocarbon side and that of DPA more towards the aqueous side of the dipole layer.  相似文献

The alignment of the sn-1 and sn-2 acyl chains at the terminal methyl ends generally produces significant influence on the thermodynamic properties of the bilayer phase transitions. We investigated the bilayer phase behavior of asymmetric phospholipids, myristoylpalmitoylphosphatidylcholine and palmitoylmyristoylphosphatidylcholine, by high-pressure light-transmittance and Prodan-fluorescence techniques and differential scanning calorimetry. Constructed temperature-pressure phase diagrams revealed that no stable phase can exist in the whole pressure range because of the formation of the most stable Lc phase. Nevertheless, the pretransition, the detection of which is severely hampered by the exceptionally prompt formation of the Lc phase, was successfully observed. Moreover, the effect of the total and difference of the sn-1 and sn-2 acyl chain lengths on minimal interdigitation pressure (MIP) was summarized in a MIP vs. chain-length inequivalence parameter plot, where the effect was proved to be classified in three zones depending on the alignment of both terminal methyl ends.  相似文献

Summary Southern Corn Leaf Blight is caused by a toxin produced by a virulent form ofHelminthosporium maydis (Race T). The toxin has been shown to uncouple oxidative phosphorylation and dissipate Ca2+ gradients in mitochondria isolated from susceptible, but not resistant, corn. The possibility that the toxin acted by increasing the permeability of membranes to ions was tested using a planar bilayer membrane system. Addition of the toxin to the bilayer system, under voltage-clamp conditions, resulted in stepwise increases in current across the phospholipid bilayer, a response characteristic for channel formers. Single-channel conductance in 1m KCl is 27 pS which corresponds to 1.7×107 ions sec–1 channel–1 at 100 mV applied potential. The toxin channels are: (i) fairly uniform in conductance, (ii) ideally selective for K+ over Cl, and (iii) most conductive to H+. The channel showed the following selectivity for alkali metal cations: Rb+>K+>Cs+>Na+>Li+ (169731) based on the most frequently observed conductance in 1m chloride salts. The toxin showed no voltage dependence over the range of –100 to +100 mV. Channel formation was also a property of a synthetic analog (Cmpd IV) of the toxin. The ability of the native toxin to form channels may be a mode of toxin action on mitochondrial membranes from susceptible corn.  相似文献

Summary The effect of endotoxin fromSalmonella typhimurium orEscherichia coli was studied on bilayer lipid membranes (1% lecithin–1% cholesterol in n-decane) formed in buffered 0.1m NaCl solution (pH 6.8). Endotoxin was added to the buffered solution either prior to membrane formation or after stable membranes were formed. In both cases, concentrations of 110 to 720 g/ml endotoxin initiated a decrease in the electrical resistance of the membranes followed by their rupture. A 50 g/ml concentration of the agent was unable to elicit any response. Also, the addition of an equal volume of buffer solution, serving as a control, caused no decrease in membrane resistance or survival time. Treatment of the endotoxin with alkaline hydroxylamine to remove esterand amide-bound fatty acids likewise abolished the membrane effect. This is the first report of an endotoxin effect on lipid bilayer membranes. The potential correlation of this interaction of bilayer and endotoxin with the diverse biologic effects of endotoxin is discussed.  相似文献

Summary Charge-pulse experiments were performed with lipid bilayer membranes from oxidized cholesterol/n-decane at relatively high voltages (several hundred mV). The membranes show an irreversible mechanical rupture if the membrane is charged to voltages on the order of 300 mV. In the case of the mechanical rupture, the voltage across the membrane needs about 50–200 sec to decay completely to zero. At much higher voltages, applied to the membrane by charge pulses of about 500 nsec duration, a decrease of the specific resistance of the membranes by nine orders of magnitude is observed (from 108 to 0.1 cm2), which is correlated with the reversible electrical breakdown of the lipid bilayer membrane. Due to the high conductance increase (breakdown) of the bilayer it is not possible to charge the membrane to a larger value than the critical potential differenceV c. For 1m alkali ion chloridesV c was about 1 V. The temperature dependence of the electrical breakdown voltageV c is comparable to that being observed with cell membranes.V c decreases between 2 and 48°C from 1.5 to 0.6 V in the presence of 1m KCl.Breakdown experiments were also performed with lipid bilayer membranes composed of other lipids. The fast decay of the voltage (current) in the 100-nsec range after application of a charge pulse was very similar in these experiments compared with experiments with membranes made from oxidized cholesterol. However, the membranes made from other lipids show a mechanical breakdown after the electrical breakdown, whereas with one single membrane from oxidized cholesterol more than twenty reproducible breakdown experiments could be repeated without a visible disturbance of the membrane stability.The reversible electrical breakdown of the membrane is discussed in terms of both compression of the membrane (electromechanical model) and ion movement through the membrane induced by high electric field strength (Born energy).  相似文献

FTIR spectroscopy and fluorescence polarization were used to show that a bacterial dirhamnolipid interacts with phospholipid membranes composed of DPPC, altering both the acyl chain and the interfacial region of the bilayer. Incorporation of increasing amounts of dirhamnolipid into 2H-DPPC membranes broadened the transition and shifted the transition temperature toward lower values, according to the effect on the CD2 stretching vibration. Examination of the 13CO stretching band of 13C-DPPC indicated that, both below and above the phase transition, dirhamnolipid produced a shift of the band frequency toward higher values, indicating a strong dehydration of the phospholipid CO groups, and therefore of the interfacial region of the membrane. The effects on DPH and TMA-DPH fluorescence polarization provided additional support to hypothesize on the location of trehalose lipid within the bilayer. The results shown here could help to explain some of the interesting membrane-related biological actions of rhamnolipids reported before.  相似文献

A number of carbobenzoxy-dipeptide-amides raise the bilayer to hexagonal phase transition temperature of dielaidoylphosphatidylethanolamine (stabilizes the bilayer). The potency of the peptides in stabilizing the bilayer phase is Z-Tyr-Leu-NH2= Z-Gly-Phe-NH2>Z-Ser-Leu-NH2>Z-Gly-Leu-NH2>Z-Gly-Gly-NH2. A linear correlation was found between the respective HPLC retention time parameterk for the peptide and the slope of the bilayer stabilization curve determined with model membranes by differential scanning calorimetry. One dipeptide, Z-Ser-Leu-NH2, reduces measles virus cytopathic effect (CPE) in Vero cells. Summary The modifications of the electrical properties of bilayer lipid membranes (BLM) composed of cholesterol and an ionic surfactant upon interaction with charged polypeptides were studied. The addition of 10–8 m polylysine (Ps+) to one side of anionic cholesterol dodecylphosphate BLM increases the specific membrane conductance over 1000-fold (from 10–8 to 10–5 mho/cm2) and develops a cationic transmembrane potential larger than 50 mV. This potential is reverted by addition of polyanions such as RNA, polyglutamic or polyadenilic acid to the same side on which Ps+ is present, by addition of Ps+ to the opposite side, or by addition of trypsin to either side. Both conductance and potential changes are hindered by increasing the ionic strength or by raising the pH of the bathing medium, disappearing above pH 11.5 where it is known that Ps+ folds into an -helix. The interaction of polyglutamic acid (PGA) with a cationic cholesterol-hexadecyltrimethylammonium bromide BLM results in increased membrane conductance and development of an anionic transmembrane potential which is reverted by addition of polycations to the same aqueous phase where PGA is present. Addition of either Ps+ or PGA to one or both sides of a neutral BLM composed of 7-dehydrocholesterol induces no significant change. The observations suggest the formation of a lipid polymer membrane resultant from the interaction, predominantly electrostatic, of the isolated components. The implications of these results are discussed in terms of the current models of membrane structure.  相似文献

Summary A general approach is developed to interpret linear dichroism (LD) spectra of ubiquinones (Q n) in host bilayers. Information is reported in terms of guest-host mutual orientation and localization. The overall orientational anisotropy of guest ubiquinone molecules is described by a basic set of limiting orientation/localization modes. Assignments of the UV transitions of the ubiquinone chromophore were obtained by the liquid crystal-linear dichroism technique and molecular orbital (CNDO/S) calculations. The LD spectra of Q n in the bilayers provided by the lyotropic nematic mesophase exhibited by water solutions of potassium laurate and decanol were interpreted on the basis of the above assignments. The resulting experimental evidence showed a multisite distribution in the host bilayer for the aromatic heads of all the investigated Q n derivatives except Q0. The orientational distribution suggested by the LD spectra fits the solubilization model recently proposed by G. Lenaz [J. Membrane Biol. (1988) 104:193–209] for ubiquinone in lipid membranes. Within this model Q n molecules are located in the midplane and their headgroups oscillate transversally across the membrane. Q 0 instead has a single site location, close to the polar bilayer interface. Experimental evidence that the headgroup carbonyls tend to grasp the polar interface of the host bilayer was also obtained. Summary The membrane permeability coefficients for the homologous monocarboxylic acids, formic through hexanoic, as well as benzoic and salicylic, were determined for egg phosphatidylcholine-decane planar bilayer membranes. The permeabilities of formic, acetic and propionic acid were also determined for solvent-free phosphatidylethanolamine bilayers. Permeability coefficients were calculated from tracer fluxes measured under otherwise symmetrical conditions, and precautions were taken to ensure that the values were not underestimated due to unstirred layer effects. The relation between the nonionic (HA) permeability (P m ) and the hexadecane/water partition coefficient (K p ) was: log m =0.90 log Kp+0.87 (correlation coefficient=0.996). Formic acid was excluded from the analysis because its permeability was sixfold higher than predicted by the other acids. The permeabilities for solvent-free membranes were similar to those for decanecontaining membranes. The exceptionally high permeability of formic acid and the high correlation of the other permeabilities to the hexadecane/water partition coefficient is a pattern that conforms with other nonelectrolyte permeabilities through bilayers. Similarly, the mean incremental free energy change per methylene group (G-CH2-) was –764 cal mol–1, similar to other homologous solutes in other membrane systems. However, much less negative G values (–120, to –400 cal mol–1) were previously reported for fatty acids permeating bilayers and biological membranes. These values are due primarily to unstirred layer effects, metabolism and binding to membranes and other cell components.  相似文献

Low-voltage-activated (1-v-a) and high-voltage-activated (h-v-a) Ca2+ currents I Ca were recorded in whole-cell voltage clamped NG108-15 neuroblastoma x glioma hybrid cells. We studied the effects of arachidonic acid (AA), oleic acid, myristic acid and of the positively charged compounds tetradecyltrimethyl-ammonium (C14TMA) and sphingosine. At pulse potentials >–20 mV, AA (25-100 m) decreased 1-v-a and h-v-a I Ca equally. The decrease developed slowly and became continually stronger with increasing time of application. It was accompanied by a small negative shift and a slight flattening of the activation and inactivation curves of the 1-v-a I Ca. The shift of the activation curve manifested itself in a small increase of 1-v-a I Ca at pulse potentials <–30 mV. The effects were only partly reversible. The AA effect was not prevented by 50 m 5, 8, 11, 14-eicosatetraynoic acid, an inhibitor of the AA metabolism, and not mimicked by 0.1–1 m phorbol 12, 13-dibutyrate, an activator of protein kinase C. Probably, AA directly affects the channel protein or its lipid environment. Oleic and myristic acid acted similarly to AA but were much less effective. The positively charged compounds C14TMA and sphingosine had a different effect: They shifted the activation curve of 1-v-a I Ca in the positive direction and suppressed 1-v-a more than h-v-a I Ca; their effect reached a steady-state within 5–10 min and was readily reversible. C14TMA blocked 1-v-a I Ca with an IC50 of 4.2 m while sphingosine was less potent.  相似文献

The interactions of the antibiotic polymixin B, a polycationic cyclic polypeptide containing a branched acyl side chain, with dimyristoylphosphatidylcholine (DMPC) and dimyristoylphosphatidic acid (DMPA) bilayers were investigated by Raman spectroscopy for a wide range of lipid/polypeptide mole fractions. Temperature profiles, constructed from peak height intensity ratios derived from the lipid methylene C-H stretching and acyl chain C-C stretching mode regions, reflected changes originating from lateral chain packing effects and intrachain trans / gauche rotamer formation, respectively. For DMPC/polymyxin B bilayers the temperature dependent curves indicate a broadening of the gel-liquid crystalline phase transition accompanied by an approx. 3 C deg. increase in the phase transition temperature from 22.8°C for the pure bilayer to 26°C for the polypeptide complex. For a 10:1 lipid/polypeptide mole ratio the temperature profile derived from the C-C mode spectral parameters displays a second order/disorder transition, at approx. 35.5°C, associated with the melting behavior of approximately three bilayer lipids immobilized by the antibiotic's charged cyclic headgroup and hydrophobic side chain. For the 10:1 mole ratio DMPA/polypeptide liposomes, the temperature profiles indicate three order/disorder transitions at 46, 36 and 24°C. Pure DMPA bilayers display a sharp lamellar-micellar phase transition at 51°C.  相似文献
