A series of dinuclear copper(II) complexes involving 6-(benzylamino)purine derivatives, (HLn), as bridging ligands were synthesized, characterized and tested for both their in vitro and in vivo antioxidant activities. Based on results of elemental analyses, temperature dependence of magnetic susceptibility measurements, UV-vis, FTIR, EPR, NMR and MALDI-TOF mass spectroscopy, conductivity measurements and thermal analyses, the complexes with general compositions of [Cu2(μ-HLn)4Cl2]Cl2 · 2H2O (1-4) and [Cu2(μ-HLn)2(μ-Cl)2Cl2] (5-7) were prepared {where n = 1-4; HL1 = 6-[(2-methoxybenzyl)amino]purine, HL2 = 6-[(4-methoxybenzyl)amino]purine, HL3 = 6-[(2,3-dimethoxybenzyl)amino]purine and HL4 = 6-[(3,4-dimethoxybenzyl)amino]purine}. In the case of complexes 2, 3, 5 and 7, the antioxidant activities were studied by both in vitro {superoxide dismutase-mimic (SOD-mimic) activity} and in vivo {cytoprotective effect against the alloxan-induced diabetes (antidiabetic activity)} methods. The obtained IC50 value of the SOD-mimic activity for the complex 5 (IC50 = 0.253 μM) was shown to be even better than that of the native bovine Cu,Zn-SOD enzyme (IC50 = 0.480 μM), used as a standard. As for the antidiabetic activity, the pretreatment of mice with complexes 3 and 7 led to the complete elimination of cytotoxic attack of alloxan and its free radical metabolites, used as a diabetogenic agent. The cytoprotective effect of these compounds was proved by the preservation of the initial blood glucose levels of the pretreated animals, as against the untreated control group.  相似文献

Syntheses of two novel ligand precursors O,O'-diisopropyl- (1a) and O,O'-diisobutyl-(S,S)-ethylenediamine-N,N'-di-2-propanoate dihydrochloride monohydrate (1b) and the corresponding dichloroplatinum(II) (2a and 2b) and tetrachloroplatinum(IV) complexes (3a and 3b) are described here. The substances were characterized by IR, (1)H and (13)C spectroscopy and elemental analysis. Crystal structures were determined for 1a and the corresponding platinum(IV) complex, 3a. In vitro antiproliferative activity was determined against tumor cell lines: human adenocarcinoma HeLa, human myelogenous leukemia K562, human malignant melanoma Fem-x, rested and stimulated normal immunocompetent cells (human peripheral blood mononuclear PBMC cells) using KBR test (Kenacid Blue Dye binding test). The IC(50)(microM) values for the most active compound 3a were: 30.48+/-2.54; 12.26+/-2.60; 13.68+/-3.22; 80.18+/-24.07 and 71.30+/-21.70, respectively.  相似文献

In this work we present the synthesis and characterization of the complex dichloro[N-propanoate-N,N-bis-(2-pyridylmethyl)amine]iron(III) [FeIII(PBMPA)Cl2]. The ligand LiPBMPA was synthesized through the Michael reaction of BMPA with methylacrylate, followed by alkaline hydrolysis. The complex [FeIII(PBMPA)Cl2] has been synthesized by the reaction of the ligand with FeCl3 · H2O and was mainly characterized by cyclic voltammetry, conductivimetry, and electronic, infrared and Mössbauer spectroscopies, and by X-ray structural analysis, which showed an iron center coordinated by one carboxylate oxygen in a monodentate way, one tertiary amine, two pyridine groups and two chloride ions. It has been proposed that in water the chloride ligands are shifted by the solvent molecules and the species [FeIII(PBMPA)(H2O)2]Cl2 is predominant. The catalase-like activity of the complex was tested in water, and it proved to be active in the hydrogen peroxide dismutation. Kinetics studies were conducted following the initial rates method. The reaction is first order in relation to both the complex and the hydrogen peroxide. Based on the presence of a lag phase that depends on the initial complex concentration, we propose that the active species that shows in situ catalase-like activity, is a binuclear complex.  相似文献

Isocryptomerin is a biflavonoid isolated from Selaginella tamariscina used in traditional medicine. In this study, we investigated novel antifungal properties of isocryptomerin. The results indicated that isocryptomerin exerted antifungal activity in an energy-independent manner without remarkable hemolytic effects. To understand mode of action(s) of isocryptomerin, we conducted experiments on Candida albicans, a noted human pathogenic fungal strain. Flow cytometric analysis with bis-(1,3-dibutylbarbituric acid) trimethine oxonol [DiBAC4(3)], a translational membrane potential dye, regeneration test with fungal protoplasts, and fluorescence analysis with 1,6-diphenyl-1,3,5-hexatriene (DPH), a probe for membrane studies by depolarization, indicated that isocryptomerin could depolarize fungal plasma membrane. In conclusion, the results suggested that the antifungal activities of isocryptomerin might be due to its membrane-disruption mechanism(s).  相似文献

The novel N,N-type bidentate ligand precursors, diethyl, dipropyl esters of ethylenediamine-N,N′-diacetic acid dihydrochloride (HOOCCH2NHCH2CH2NHCH2COOH · 2HCl, H2edda · 2HCl), and the corresponding tetrachloroplatinum(IV) complexes, [PtCl4(R2edda)] · H2O (ROOCCH2NHCH2CH2NHCH2COOR, R = Me, Et, n-Pr), were synthesized. The esters coordinated as bidentate ligands via both N donor atoms. The esters, as well as the complexes, have been characterized by infrared, 1H and 13C NMR spectroscopy and elemental analysis. Solid state structures of both dimethyl and diethyl ester platinum(IV) complexes have been determined by X-ray crystallography. Quantum chemical calculations were performed in order to investigate diastereoselectivity in the formation of the platinum(IV) complexes. The in vitro cytotoxic evaluation of the investigated complexes in human tumor cell lines 1411HP, H12.1 (both testicular germ cell tumors), DLD-1 (colon carcinoma), 518A2 (melanoma), A549 (lung carcinoma) and liposarcoma showed a dose-dependent antiproliferative effect in all cell lines. Remarkably, the highest cytotoxic activity was observed in the cisplatin-resistant cell line 1411HP. In addition, at higher concentrations the treatment with these complexes led to the induction of apoptosis in all cell lines except for DLD-1.  相似文献

The lanthanide complex aquatrichloridobis(1,10‐phenanthroline)cerium(III) [Ce(phen)2(H2O)Cl3] (KP776) was fully characterized by elemental analysis, IR‐, and 1H‐ and 13C‐NMR spectroscopy, as well as TG/DTA measurements, and its behavior in H2O, important for the application as a chemotherapeutic, was studied. In addition, the binding of KP776 to nucleotides and single serum proteins was investigated by capillary electrophoresis, whereas binding to proteins in human plasma was observed by ICP‐MS. The compound shows promising anticancer properties in vitro: proliferation of human cancer cell lines is strongly inhibited with IC50 values in the very low micromolar range.  相似文献

Three novel copper(II) complexes as condensation derivatives of 2-pyridinecarboxaldehyde, 2-acetylpyridine or 2-quinolinecarboxaldehyde with ethyl hydrazinoacetate hydrochloride were synthesized. X-ray crystal structures of all three complexes were determined. Crystallographic data indicate a common basis of the structures with some major differences. Both pyridine derivatives form complexes of the type [CuCl2L] with distorted square-planar geometry around copper(II), but show a decisive difference in the side chain orientation. The quinoline derivative forms a dimeric complex of type [Cu2Cl4L2] with distorted trigonal bipyramidal geometry around copper(II), but a rather short Cu···Cu contact, which is reflected in its magnetic properties. All three complexes showed antibacterial activity.  相似文献

We present here the syntheses of two dinuclear iron(III) complexes with the polydentate N,O-donor ligand H2BPClNOL (N-(2-hydroxybenzyl)-N-(2-pyridylmethyl)[(3-chloro)(2- hydroxy)]propylamine). The reaction between FeIII(ClO4)3 · 9H2O, the title ligand and two equivalents of NaOAc · 3H2O resulted in the complex . When the synthesis was performed with a lesser amount of NaOAc · 3H2O (half equivalent), the complex without bridging acetate, was obtained. The complexes were characterized by X-ray structural analysis, magnetochemistry, Mössbauer and UV-Vis spectroscopies, and electrochemistry. Complex 2 was also characterized in solution through potentiometric titration. Both complexes crystallize in the monoclinic system. Complex 2 has one water molecule coordinated to each iron centre. Their pKa values are 5.00 and 7.03 for the first protonation/deprotonation equilibrium of each coordinated water molecule. The UV-Vis and electrochemical techniques showed that the absence of an acetate bridge in 2 results in a significant difference in the Lewis acidity of both iron centres, when compared with 1. The lack of an acetate bridge in 2 also results in changes in the anti-ferromagnetic coupling as revealed by magnetic measurements. Complex 2 is an interesting structural model for the active site of iron containing PAPs, since it has an Fe(-alkoxo)2Fe core with an FeIII?FeIII distance of 3.122(1) Å, containing phenolate and water molecules coordinated to the iron centres and is soluble in aqueous solutions. Furthermore, the UV-Vis properties of 2 are similar to those of PAPs, since the complex absorbs at 580 nm in the oxidized form (550-570 nm for PAPs) and at 499 nm in the mixed-valence form (505-510 nm for PAPs) as revealed through spectroelectrochemical studies. Finally, complex 2 successfully promoted the hydrolytic cleavage of plasmid DNA under aerobic and anaerobic conditions, producing single and double DNA strand breaks at biological pH values.  相似文献

The measurement of acyl-CoA dehydrogenase activities is an essential part of the investigation of patients with suspected defects in fatty acid oxidation. Multiple methods are available for the synthesis of the substrates used for measuring acyl-CoA dehydrogenase activities; however, the yields are low and the products are used without purification. In addition, the reported characterization of acyl-CoAs focuses on the CoA moiety, not on the acyl group. Here we describe the synthesis of three medium-chain acyl-CoAs from mixed anhydrides of the fatty acids using an aqueous-organic solvent mixture optimized to obtain the highest yield. First, cis-4-decenoic acid and 2,6-dimethylheptanoic acid were prepared (3-phenylpropionic acid is commercially available). These were characterized by gas chromatography/mass spectrometry (GC/MS), 1H nuclear magnetic resonance (NMR), and 13C NMR. Then cis-4-decenoyl-CoA, 3-phenylpropionyl-CoA, and 2,6-dimethylheptanoyl-CoA were synthesized. These were then purified by ion exchange solid-phase extraction using 2-(2-pyridyl)ethyl-functionalized silica gel, followed by reversed-phase semipreparative high-performance liquid chromatography with ultraviolet detection (HPLC-UV). The purified acyl-CoAs were characterized by analytical HPLC-UV followed by data-dependent tandem mass spectrometry (MS/MS) analysis on the largest responding MS mass (HPLC-UV-MS-MS/MS) and 13C NMR. The yields of the purified acyl-CoAs were between 75% and 78% based on coenzyme A trilithium salt (CoASH). Acyl-CoA dehydrogenase activities were measured in rat skeletal muscle mitochondria using, as substrates, the synthesized cis-4-decenoyl-CoA, 3-phenylpropionyl-CoA, and 2,6-dimethylheptanoyl-CoA. These results were compared with the results using our standard substrates butyryl-CoA, octanoyl-CoA, and palmitoyl-CoA.  相似文献

Two new copper(II) complexes of the type [Cu(L)X2), where L = (E)-N-phenyl-2-[phenyl (pyridine-2-yl)methylene]hydrazinecarboxamide X = Cl/Br have been synthesized and characterized by elemental analyses, FAB (fast atomic bombardment) magnetic measurements, electronic absorption, conductivity measurements cyclic voltammetry (CV) and Electron paramagnetic resonance (epr) spectroscopy. The structures of these complexes determined by single crystal X-ray crystallography show a distorted square based pyramidal (DSBP) geometry around copper(II) metal center. The distorted CuN2OX (X = Cl/Br) basal plane in them is comprised of two nitrogen and one oxygen atoms of the meridionally coordinated ligand and a chloride or bromide ion and axial position is occupied by other halide ion. The epr spectra of these complexes in frozen solutions of DMSO showed a signal at g ca. 2. The trend in g-value (g|| > g > 2.00) suggest that the unpaired electron on copper(II) has dx2-y2 character. Biological activities in terms of superoxide dismutase (SOD) and antimicrobial properties of copper(II) complexes have also been measured. The superoxide dismutase activity reveals that these two complexes catalyze the fast disproportionation of superoxide in DMSO solution.  相似文献

The in vitro antibacterial and antifungal activities of five different amino acid Schiff bases derived from the reaction of 2-hydroxy-1-naphthaldehyde with glycine, L-alanine L-phenylalanine, L-histidine, L-tryptophane and the manganese(III) complexes of these bases were investigated. Structures of the Schiff bases were proven by 1H-NMR. In vitro activities against some Gram-positive (Staphylococcus aureus and Bacillus polymyxa) and Gram-negative (Escherichia coli) bacteria and the fungus Candida albicans were determined. The antimicrobial activities tended to decrease with the increasing size of the amino acid residues.  相似文献

Abstract

A new series of anti-bacterial and anti-fungal mono- and di-substituted triazoles (L1)–(L6) have been synthesized and characterized on the basis of their physical, spectral and analytical data. The ligands (L1)–(L6) on reaction with vanadyl(IV) sulphate led to the formation of vanadyl(IV) metal complexes (1)–(4). The structure of the complexes has been established on the basis of their physical, spectral and elemental analyses data. The synthesized ligands and their vanadyl(IV) complexes have been screened in vitro for anti-bacterial activity against six bacterial species such as, Escherichia coli (ATCC 25922), Shigella flexneri (ATCC 12022), Pseudomonas aeruginosa (ATCC 27853), Salmonella typhi (ATCC 14028), Staphylococcus aureus (ATCC 25923) and Bacillus subtilis (ATCC 6051) and for in vitro anti-fungal activity against six fungal strains, Trichophyton longifusus, Candida albicans, Aspergillus flavus, Microsporum canis, Fusarium solani and Candida glabrata. The screening results showed the vanadyl complexes to be more bactericidal/fungicidal against one or more bacterial/fungal species. The synthesized compounds were also subjected to brine shrimp bioassay for scrutinizing their cytotoxicity.  相似文献

1‐phenyl‐3‐methyl‐4‐benzoyl‐5‐pyrazolone 4‐ethyl‐thiosemicarbazone (HL) and its copper(II), vanadium(V) and nickel(II) complexes: [Cu(L)(Cl)]·C2H5OH·( 1 ), [Cu(L)2]·H2O ( 2 ), [Cu(L)(Br)]·H2O·CH3OH ( 3 ), [Cu(L)(NO3)]·2C2H5OH ( 4 ), [VO2(L)]·2H2O ( 5 ), [Ni(L)2]·H2O ( 6 ), were synthesized and characterized. The ligand has been characterized by elemental analyses, IR, 1H NMR and 13C NMR spectroscopy. The tridentate nature of the ligand is evident from the IR spectra. The copper(II), vanadium(V) and nickel(II) complexes have been characterized by different physico‐chemical techniques such as molar conductivity, magnetic susceptibility measurements and electronic, infrared and electron paramagnetic resonance spectral studies. The structures of the ligand and its copper(II) ( 2 , 4 ), and vanadium(V) ( 5 ) complexes have been determined by single‐crystal X‐ray diffraction. The composition of the coordination polyhedron of the central atom in 2 , 4 and 5 is different. The tetrahedral coordination geometry of Cu was found in complex 2 while in complex 4 , it is square planar, in complex 5 the coordination polyhedron of the central ion is distorted square pyramid. The in vitro antibacterial activity of the complexes against Escherichia coli, Salmonella abony, Staphylococcus aureus, Bacillus cereus and the antifungal activity against Candida albicans strains was higher for the metal complexes than for free ligand. The effect of the free ligand and its metal complexes on the proliferation of HL‐60 cells was tested.  相似文献

Three novel organotin(IV) complexes with 2-(9H-carbazol-9-yl) acetic acid (HL), of the formulae {[nBu2SnOL]2O}2 (1), [nBuSn(O)OL]6 (2) and [nBu3SnOL]6 (3) were prepared. All compounds were characterized by X-ray crystallography, confirming that complex (1) is tetranuclear one with ladder framework, complex (2) is a hexanuclear organotin(IV) complex with drum structure and complex (3) is a macrocycle with 24-membered stannoxane ring. Furthermore, all complexes were tested in vitro for their cytotoxic activity, using human hepatocellular carcinoma cell line (BEL-7402) and human hepatocellular liver carcinoma cell line (HepG2). Complex (1) displayed the best cytotoxicity and can be pointed out as a promising substrate to be subject of further investigations.  相似文献

Epileptic foci are associated with locally reduced taurine (2-aminoethanesulfonic acid) concentration and Na+, K+-ATPase (EC 3.6.1.3) specific activity. Topically applied and intraperitoneally administered taurine can prevent the development and/or spread of foci in many animal models. Taurine has been implicated as a possible cytosolic modulator of monovalent ion distribution, cytosolic “free” calcium activity, and neuronal excitability. Taurine may act in part by modulating Na+, K+-ATPase activity of neuronal and glial cells. We characterized the requirements for in vitro modulation of Na+, K+-ATPase by taurine. Normal whole brain homogenate Na+, K+-ATPase activity is 5.1 ± 0.4 (4) μmol Pi± h?1± mg?1 Lowry protein. Partial purification of the plasma membrane fraction to remove cytosolic proteins and extrinsic proteins and to uncouple cholinergic receptors yields a membrane-bound Na+, K+-ATPase activity of 204.6 ± 5.8 (4) mol Pi± h?1± mg?1 Lowry protein. Taurine activates the Na+, K+-ATPase at all levels of purification. The concentration dependence of activation follows normal saturation kinetics (K1/2= 39 mM taurine, activation maximum =+87%). The activation exhibits chemical specificity among the taurine analogues and metabolites: taurine = isethionic acid > hypotaurine > no activation =β-alanine = methionine = choline = leucine. Taurine can act as an endogenous activator/modulator of Na+, K+-ATPase. Its action is mediated by a membrane-bound protein.  相似文献

Isolation and identification of substances having an activity to stimulate the fruiting body formation of Schizophyllum commune were attempted. The active principles in its mycelia were divided into four fractions by sequential purification with silica gel column and reverse-phase HPLC column chromatography. By infrared spectra and thin-layer chromatography, the active substances in these four fractions were revealed as cerebrosides. About 0.1 μg of the cerebroside fractions showed a discriminative stimulating activity on S. commune when tested by the method these authors adopted. The active substance in the fraction II was N-2′-hydroxypalmitoyl-1-O-glucosyl-nonadecasphingadienine. The cerebrosides from pea seeds and Fusicoccum amygdali showed the similar activity on S. commune, but some commercial synthetic cerebrosides and cerebrosides from bovine and porcine brains exhibited no stimulating activity. Only definite cerebrosides with special structures seem to be able to induce the fruiting of S. commune.  相似文献

A Drosophila melanogaster mutant, fs(1)pyr Su(b) , carrying a mutation that maps to the tip of the X chromosome, has been isolated. The mutation, when present alone, does not confer a detectable phenotype. However, this mutation causes female sterility and reduces embryonic viability when combined with mutations which deregulate the pyrimidine and β-alanine pools. Embryos that are homozygous for the mutations fs(1)pyr Su(b) , r Su(b) [previously designated as Su(b)] and b, and originate from a female parent homozygous for the three mutations show severely reduced viability. Cu(Ph2P(o-C6H4C(O)H))2(NO2) (3) has been prepared in high yield by treating [Cu(Ph2P(o-C6H4C(O)H))2(NCMe)]BF4 (2) with [Ph2PNPPh2]NO2 at ambient temperature. The nitrite ligand of 3 is coordinated to the Cu(I) center in an O,O-bidentate mode. Protonation of 3 releases NO molecule, which mimics the reactivity of the Type 2 Cu-NiRs. In contrast, reaction of [Pd(NCMe)4](BF4)2 and Ph2P(o-C6H4C(O)H) affords cis-[Pd(Ph2P(o-C6H4C(O)H))2](BF4)2 (4) with the Pd2+ ion chelated by two phosphino-aldehyde moieties. The hemilabile formyl ligands of 4 can be displaced by NO2 to produce trans-Pd(Ph2P(o-C6H4C(O)H))2(NO2)2 (5), of which the nitrite ligands present an N-monodentate bonding feature. Protonation of 5 with HBF4, however, regenerates compound 4, likely via elimination of nitrous acid. The structures of 3-5 have been determined by an X-ray diffraction study.  相似文献

[C20H17N3O2] and cobalt (II) complex [Co(L2)(MeOH)2].ClO4, (L2 = 4-((E)-1-((2-(((E)-pyridin-2-ylmethylene) amino) phenyl) imino) ethyl) benzene-1, 3-diol) novel Schiff base has been synthesiszed and chracterized by Fourier transform infrared, UV–vis, 1H-NMR spectroscopy, and elemental analysis techniques. The interaction of Co(II) complex with DNA and BSA was investigated by electronic absorption spectroscopy, fluorescence spectroscopy, circular dichroism, and thermal denaturation studies. Our experiments indicate that this complex could strongly bind to CT-DNA via minor groove mechanism. In addition, fluorescence spectrometry of BSA with the complex showed that the fluorescence quenching mechanism of BSA was of static type. The complex exhibited significant in vitro cytotoxicity against three human cancer cell lines (JURKAT, SKOV3, and U87). The molecular docking experiment effectively proved the binding of complex to DNA and BSA. Finally, antibacterial assay over gram-positive and gram-negative pathogenic bacterial strains was studied.  相似文献
