The amino acid composition of the vascular sap of a high lysine maize mutant was determined during kernel development. With the exception of proline and cystine, all amino acids that occur in the endosperm were found in the vascular sap of the ear peduncle. Glutamine is the major amino acid transported to the endosperm varying from 30.6 to 20.6 μmol at 7 and 42 days after pollination, respectively. Aspartic acid, the second most important nitrogen form translocated to the seeds, was ca 10 μmol% during kernel filling. Glutamine and arginine content decreased with maturity, while valine, methionine, isoleucine, leucine, tyrosine and phenylalanine increased with kernel development. The remaining N forms were constant during endosperm growth.  相似文献

Tracer studies with aspartic acid-[4-14C], alanine-[1-14C] acetate-[2-14C] and diaminopimelic acid-[1,(7)-14C] injected into the developing endosperm of maize revealed that the biosynthesis of lysine and other amino acids occurs in this organ. The data suggest that lysine is synthesized via the diaminopimelic acid pathway.  相似文献

Incorporation of leucine and changes in different protein fractions have been studied during Sorghum grain development. Most of the label from the injected leucine-[14C] was found in glutelin and residue fraction towards later stages of maturity. The label in albumin, globulin and prolamin decreased with a concomitant increase in label in glutelin and residue proteins. The concentration of lysine, aspartic acid and glycine decreased while that of leucine, proline, alanine, tyrosine, phenylalanine, and cystine increased during grain development. Increase in serine, methionine, valine and isoleucine was only marginal. The proportion of glutamic acid was high at all stages of grain development. Glutelin fraction resolved into two peaks on gel chromatography, only one of which with higher MW was labelled, while in albumin both the peaks were found to be labelled. A comparative study of free amino acids and protein fractions of normal with a double mutant (su1 o2) was made, during endosperm development in segregating ears of a maize synthetic. Zein content showed striking differences in the two genotypes, being 7.7 and 6 times greater in the normal endosperm at 24 and 47 days after pollination respectively. This observed decrease in zein synthesis, coded by sugary-1/opaque-2 genes, causes an accumulation of alanine, glutamic and aspartic acids, glutamine and asparagine in the high lysine endosperm mutant.  相似文献

On the basis of leaf dry wt, the protein content of six varieties of cassava varied from 29.3 to 38.6% and the estimated leaf protein production ranged from 242 to 953 kg per ha. On the basis of fr. wt of leaf, the total amino acids ranged from 8.42 to 9.4% while the essential amino acids averaged 4.21% and the sulphur-containing amino acids only 0.25%. The amino acid composition profiles for the six varieties was similar.  相似文献

Zein extracted from maize endosperm has been fractionated into four polypeptide chains, having the following MWs 23 000, 21 000, 13 500 and 9600. By amino acid analysis the two smaller MW chains (representing 30% of total zeins) have been found to be zein-type molecules. These two chains are thought to be responsible for zein granule formation via -S-S- bridges. Zein is also highly heterogeneous in charge, and is resolved into at least 15 components, with pI's in the pH range 5–9. As demonstrated by amino acid analysis, part of this heterogeneity is due to spot mutations in some of the genes responsible for zein synthesis.  相似文献

The pedicel-placenta-chalazal (PPCh) region is a heterogeneous maternal tissue located at the base of the endosperm. Its active role in the transport and metabolism of nitrogen compounds supplied to the endosperm has been shown by incorporation of labelled compounds, as well as by analysis of appropriate enzyme activities and the free amino acid pool. Within 30 min incubation, 60–70% of the label given in [14C]aspartate or [14C]glutamate was recovered in the organic acid fraction of the PPCh. Although the dry matter and protein content of the PPCh region was practically unchanged between days 15 and 30 after pollination, glutamate transaminase, alanine transaminase, NADP-malic enzyme and NAD-malate dehydrogenase were increased substantially. The PPCh region contains glutamine at a significantly higher concentration and alanine at a lower concentration than the endosperm. In contrast to glutamate synthase, the activity of glutamine synthetase per endosperm and per mg protein is significantly higher in the PPCh region. This implies involvement of these tissues in the transport of nitrogen compounds, mainly in the form of glutamine, as well as possible compartmentation of the glutamine-glutamate cycle in the developing maize kernel.  相似文献

There is indirect evidence that the amino acid composition of proteins depends on their dimension. The amino acid composition of a nonredundant set of about 550,000 proteins was determined and it was observed that, in the range of 50-200 residues, the percentage of occurrence of most of the residue types significantly depends on protein dimension. This result should prove useful in analyzing protein sequences and genomics.  相似文献

The complete amino acid sequence of one of the major wheat protein iso-inhibitors of α-amylase was determined. The sequence was deduced from analysis of fragments and peptides derived from the protein by cleavage with cyanogen bromide and by digestion with trypsin, chymotrypsin, thermolysin and the Staphylococcus aureus V8 protease. The molecule consists of a single polypeptide chain of 123 residues. Both serine and alanine were found in position 65, and further minor examples of microheterogeneity were observed in four other residues.  相似文献

The amino acid compositions of the proteinaceous components of the gum exudates from Prosopis alba, P. chilensis, P. glandulosa, P. laevigata, P. torreyana and P. velutina, and for a sample of commercial gum mesquite, are presented. In agreement with data published previously for the polysaccharide components of their gums, only minor differences in composition are shown by these species. The amino acid compositions are characterized by very high proportions of hydroxyproline and by high proportions of proline and serine; these three amino acids account for 62.5% of those present in the gum from Prosopis velutina. The amino acid compositions of these Prosopis gums are remarkably similar to that established recently for the gum from Acacia senegal (gum arabic).  相似文献

Leaves of greenhouse grown grasses had free protein amino acid contents of generally less than 5 % total amino acids, while field collected grasses averaged 14.7 % free protein amino acid contents. Taxonomic patterns are detectable in the total leaf amino acid profiles of grasses from both sources, those of pooids being distinguishable from those of chloridoids and panicoids, and those of danthonioids showing an intermediate pattern. Leaf profiles of Oryza, Stipeae, and Ehrharteae resemble one another, and are more like those of pooids than those of panicoids. Variations in Thr and Leu are apparently associated with differences in photosynthetic pathway. Grass leaves are generally low in total amino acid contents (2.2 ? 1.0 g % fr. wt), with Ile, Val and Met + Cys identified as the limiting essential amino acids. However, the nutritional ‘chemical scores’ of grass leaf proteins are high (75 %, based on the WHO scoring pattern).  相似文献

The factorial approach to assess the amino acid (AA) requirements of pigs is based on the assumption that the AA composition of body protein is constant. However, there are indications that this assumption may not be valid because the AA composition of body protein can be affected by the AA supply. The extent to which different tissues are affected by an AA deficiency is unknown. The objective of this study was to investigate the effect of feeding pig diets with a deficient or sufficient total sulfur AA supply (TSAA; Met+Cys) from 6 to 23 weeks of age on tissue composition and meat quality. The deficient diet (TSAA–) provided 24% Met : Lys and 51% TSAA : Lys on a standardized ileal digestible basis, which are 19% and 16% below the recommended requirements, respectively. The sufficient diet (TSAA+) provided 33% Met : Lys and 60% TSAA : Lys. Diets were offered slightly below the ad libitum feed intake capacity of the pigs. Pigs offered diet TSAA– had a lower average daily gain, lower weights of the longissimus dorsi (LM) and rhomboideus muscles (RM), and of selected skin sections (P<0.05). The weight of different sections of the small intestine and the liver was not affected by the diet. The protein content of the LM and RM decreased in pigs offered diet TSAA– (P<0.05), whereas the protein content of other tissues was not affected. The TSAA supply affected the AA composition (g/16 g N) of protein in all tissues, but the Met content was changed only in the liver (P<0.05). Pigs receiving diet TSAA– had a lower Cys content in the RM and in the distal jejunum and ileum (P<0.01). The deficient TSAA supply resulted in a lower carcass weight and higher muscle glycogen stores (P<0.05), but did not affect other meat quality traits. The results of this study indicate that the muscles, jejunum and ileum respond more to a prolonged AA deficiency than the liver. In addition, the observed changes in AA composition of tissue protein question the use of a constant AA profile of retained protein to assess AA requirements.  相似文献

Sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) has been employed to resolve protein subunits of finger millet (Eleusine coracana) varieties according to their MW. These studies have established that varietal differences exist in the protein composition of finger millet varieties. The MW distribution of the protein subunits in the albumin-globulin, prolamin and glutelin fractions show many differences between the parental and cross-bred varieties and these differences are greater in the albumin-globulin and glutelin fractions than in the prolamin fraction. Electrophoretic patterns of soluble proteins, pH 5 enzyme fraction, peroxidase, glutamic dehydrogenase, leucine aminopeptidase in developing endosperm of normal and opaque-2 were studied. Multiple forms were found for all the enzymes studied. The GDH pattern showed considerable differences in normal and opaque-2 maize; the soluble protein pattern also differed, both qualitatively and quantitatively. Protein breakdown during germination of maize at 28° is closely correlated with the appearance of protease activity. In the first 2 days of germination, a slight disaggregation of only G3 glutelins into more simple elements (albumin-globulins) can be observed. Between 2 and 2.5 days, there is extensive breakdown of all protein fractions, the rate of which coincides with the rate of appearance of proteolytic activity. After 2.5 days these phenomena slow down and the bulk of the endosperm proteins disappears. Three acid proteases in endosperm extracts of germinated grain (P11, P21 and P22) have been isolated by affinity chromatography and gel filtration, and partially characterized. P11 (MW 40 000) which is present in the ungerminated grain, cannot hydrolyse prolamins and is insensitive to reducing agents. P21 (MW 36 000) and P22 (MW 12 000), which appear on day 3 of germination, can degrade prolamins in vitro. Reducing agents enhance their activity and prevent their aggregation or denaturation. Comparative assays with different substrates suggest our enzyme preparations are principally endotype proteases with little contaminating carboxypeptidase activity.  相似文献
