Changes in the levels of protein and free amino acids in theseeds and placentae of Nicotiana tabacum were studied duringseed development. Seed maturation was completed 24 days afteranthesis. During maturation, protein rapidly accumulated inthe seeds between the 6th and 18th day, along with an appreciablecompositional change in the protein amino acids as the proportionsof glutamic acid and arginine increased. The amount of freeamino acids in the seeds gradually decreased throughout maturation.The major free amino acid on the 6th day after anthesis wasglutamine, which then drastically decreased between the 6thand 12th day with increases of glutamic acid, proline, arginineand alanine. The latter amino acids decreased thereafter untilthe 24th day. On the other hand, the amount and composition of the proteinsin the placentae did not change significantly throughout seedmaturation. In the early stage of development, the major freeamino acids in the placentae were glutamine, asparagine andglutamic acid, while in the later stage asparagine was mostabundant. The level of/various amino acids in rice embryos rose sharply during embryo differentiation (7–9 days after anthesis). Then it increased steadily or tended to become stable at mid-maturation stage (13–18 days after anthesis), thereafter continuing to increase, except that the contents of aspartate and glutamate decreased significantly. The total free amino acid pool expanded rapidly during the differential stage. There after the pool capacity showed only a slightly increase until the end of embryogenesis. Both on embryo cell and dry weight bases, the capacity reached the maximum at the 9th day after anthesis, then decreased at the 13th day, and later remained stable. We deemed that the establishment of the free amino acid pool is one of the events which occur in the process of rice embryo differentiation. By the fulfillment of the differentiation (the 13th dray after anthesis), the pool capacity within the embryo cells remained stable on the whole. The free amino acid pool was dominated by serine, alanine, aspartate and glutamate during the differentiation stage. In the maturation stage, serine, alanine, arginine and lysine were the main components. These predominant; amino acids may play an important role in regulating the availability of the whole amino acid pool.  相似文献

During the seed development of Nicotiana tabacum, appreciable accumulation of the soluble protein fraction started to occur at around the 6th day after anthesis and finally reached 12% on the basis of dry weight when seed maturation was accomplished. In the soluble fraction of mature seeds, four protein fractions were observed on analytical ultracentrifugation, and the protein having a sedimentation coefficient of 11.7S was the major one. The 11.7S protein was isolated and SDS-polyacrylamide gel electrophoresis indicated that the protein consisted of at least five subunits with molecular weights of 49,000, 31,000, 29,000, 21,000 and 19,000. The 11.7S protein was rich in glutamic acid or glutamine and arginine, and the presence of carbohydrate was confirmed.

During development, all of the five subunits started to appear during the period between the 12th and 15th day after anthesis.  相似文献

Hydroxycinnamic acid esters and photoperiodic flowering induction in vitro of Cichorium intybus.
In root tissues of Cichorium intybus L. cv. Witloof cultured in vitro, the photoinductive period occurs between the 8th and the 16th day after the start of the culture. The promotive light conditions are either long days (16 h photoperiod) or daily cycles of 9 h white + 15 h red light applied during the photoinductive period in an otherwise short day treatment. During the photoinductive period and under suitable irradiation conditions, the endogenous hydroxycinnamic acid esters (especially chlorogenic acid) show a regular development as from the 8th day, reaching a maximal level by the 16th day. The amount of these molecules then decreases rapidly, preceding the external expression of floral induction. When we apply non-inductive conditions [short days (9 h) or daily cycles of 9 h white + 15 h (far-red + red) light supplied from the 8th to the 16th day in otherwise short day conditions], the metabolic changes indicate the same pattern during the inductive period as mentioned above. However, a fundamental difference exists between the inductive and non-inductive conditions, so that the production of hydroxycinnamic acid esters is particularly high towards the end of the second week of in vitro development in the induced state. This increase is not primarily due to increased photosynthetic activity in long day conditions, since it occurs both in the long day treatments and in the treatments with daily cycles of 9 h white + 15 h red light, thus revealing the morphogenetic action of light via phytochrome. This accumulation of hydroxycinnamic acid esters is correlated with floral induction, which appears to be ineffective unless there is a certain minimum amount of these molecules in the tissues.  相似文献

以转反义硫氧还蛋白基因(anti-trxs)株系01TY70-1-17-5及其对照小麦品种‘豫麦70’为试验材料,以小麦中稳定表达的肌动蛋白基因actin为内标,用半定量反转录聚合酶链式反应(semi-QRT-PCR)方法,对转基因株系及其对照种子中trxh基因时空表达情况进行了检测。检测结果表明,花后15~30d转基因株系trxh基因转录量平均比对照下降了20.1%,花后25d显著低于对照(P<0.05);胚乳trxh基因转录量最低,平均比对照低19.4%;种子吸涨24h时间内,转基因株系trxh基因转录量较对照均略低,但差异不显著。表明,外源trxs基因的导入直接干扰了内源基因的表达。  相似文献

Trends in the time course, changes in the moisture, soluble amino acids, proline, abscisic acid contents and nitrate reductase activity determined byin vivo method in the developing seeds of wheat were studied. Maximum dry matter augmentation in the seed took place in the period between 10–30 days after anthesis. Per cent moisture and moisture content started declining 15 days and 25 days after anthesis, respectively. Levels of soluble amino acids, proline and nitrate reductase activity were higher during initial stages of seed development, but decreased with increasing magnitude of dehydration and accumulation of abscisic acid (ABA) in the maturing seeds.  相似文献

Summary Seed set and fruit development in cucumber (Cucumis sativus L.) were studied in relation to female flower receptivity from day — 2 before anthesis to day + 2 after anthesis. The female cucumber flower is protogynous. The pistil was receptive 2 days before anthesis. The iso-electric focusing (IEF) patterns of the stigma/style proteins, were identical from day -5 to day +2. In pollinated flowers in vivo germination and pollen-tube growth in the ovary were affected by pistil age from day -2 to day +2. In addition, differences in sectorial filling in full seeds were observed within the fruits. A negative correlation was observed between the frequency of fertilized ovules in the pedoncular part of the fruit and ovary length at the time of pollination. In the whole fruit, significant differences in the number of full seeds and fruit size at maturity were found, and these were observed to be correlated with the various stages of female flower maturation at pollination. The day -2 and day +2 stages yielded the smallest fruits with few full seeds compared to the day -1, day 0 and day +1 stages, which had the biggest fruits and a large number of full seeds. A strong positive correlation was found between total seed number (including full and empty seeds), fruit length and weight at maturity. All these results suggest that both seed set in the different parts of the fruit and fruit development are controlled by ovular receptivity rather than by stigma/style receptivity.  相似文献

以百合(Lilium)品种‘Pollyanna’为材料,研究了KCl处理对柱头中SOD、POD及CAT等3种保护酶活性、可溶性蛋白含量以及电阻率的影响。结果表明,处理后3种酶的活性均表现出不同程度的应激性升高,其中SOD活性在花开当天和花开1d显著升高,在蕾期与花开2d极显著升高;花开当天及花开1d的POD活性极显著升高;CAT活性在花开1d升高显著,花开2d及3d时升高极显著。柱头可溶性蛋白含量及电阻率无显著变化,表明KCl处理削弱了细胞膜的膜脂过氧化作用,延缓了柱头衰老,提高了授粉受精作用,且与结实率升高表现出一定相关性。  相似文献

The developmental changes in morphology and germinability of Jatropha curcas seeds, effects of temperature and light on seed germination , and changes in desiccation tolerance of mature seeds were studied in this paper. The results indicated that J. curcas seeds reached a physiological maturation at 58 days after anthesis, and that seed germination percentage reached a peak at physiological maturation, and then decreased. The optimal germination temperature was 25 to 30℃. J. curcas seeds were insensitive to dehydration at physiological maturation . There was not a notable effect of light on seed germination. Therefore, J. curcas seed was a non-photoblastic and orthodox seeds .  相似文献

Cotton embryos from 34 to 54 days after anthesis were analyzed for organic acids, and enzymes associated with organic acid metabolism. During this developmental period, embryos accumulated citrate. Malate synthase activity appeared at 46 days after anthesis and increased rapidly to 54 days. Of other enzymes examined, only citrate synthase activity increased during this period. As isocitrate lyase activity was absent from cotton embryos during maturation, an alternative source of glyoxylate would be required for in vivo malate synthase activity. Of several metabolic sources tested, glycine was converted to glyoxylate via a transamination reaction.  相似文献

Fatty acid composition and lipid synthesis in developing safflower seeds   总被引:1,自引:0,他引:1  
Linoleic acid predominated in every lipid class during the whole period of seed development of safflower, while linolenic acid decreased with increasing maturation and it was not detected in mature seeds. Just before the initiation of triacylglycerol accumulation, the fatty acid composition of triacylglycerols changed more rapidly than those of phospholipids and glycolipids. Saturated fatty acids tended to accumulate at the 1- and 3-positions of the glycerol molecule and the more highly unsaturated acids at the 2-position. The fatty acid compositions at the 1- and 3-positions were similar in all cases investigated, but in none of the triacylglycerols was the distribution completely symmetrical. The positional distribution of linolenic acid in triacylglycerols prepared from the immature seeds 2 days after flowering and from the leaves was unusual; in spite of its highest degree of unsaturation, it was preferentially esterified at the 1- and 3-positions. When triacylglycerol was most rapidly accumulated (14–18 days after flowering), the incorporation of acetate-[U- 14C] into total lipids was also maximum and dienoic fatty acids were the principal acids labelled. Diacylglycerols and compound lipids reached the highest rate of synthesis 15 days after flowering, and then a maximum incorporation into triacylglycerol occurred 18 days after flowering. Incubation temperature affected the synthesis of individual lipid classes. Triacylglycerol was more rapidly synthesized at 32° than at 10°, while diacylglycerols and compound lipids were accumulated under the low-temperature condition. A rise of incubation temperature caused a depression in dienoic acid synthesis.  相似文献

以转反义硫氧还蛋白基因株系01TY34-73-9及其对照品种‘豫麦34’为材料,运用PCR检测和酶活性测定的方法,对转基因株系遗传稳定性以及转基因与对照种子中脱支酶活性进行测定。结果显示:(1)外源基因已经稳定遗传至后代;(2)转基因种子在不同成熟时期和不同萌发过程中的脱支酶活性与对照相比均有不同程度的降低平均降低10.3%,但仅花后25 d到收获后5 d脱支酶活性显著低于对照,其中最低值出现在花后30 d,平均比对照下降了12.0%;(3)在花后30 d和后熟5 d萌发过程中,转基因种子脱支酶活性始终低于对照,平均下降6.2%和22.2%。表明反义trxs基因的导入干扰了小麦trxh基因的表达,使trxh转录量减少,小麦籽粒中脱支酶的活性受抑。  相似文献

用SDS-PAGE方法研究了假萌发素(ψG)在小麦种子成熟和萌发过程中活性的变化.结果表明:在种子成熟过程中只有ψG表达,扬花后10 d,在颖壳、内外桴、种皮和果皮中皆可检测到ψG的草酸氧化酶活性,随着发育进程的推进,ψG的活性增大.在种子萌发过程中,在小麦品种中育5号的维管束过渡区中除了萌发素G和G'外,还可检测到ψG的草酸氧化酶活性.由于ψG在种子成熟过程中主要存在于颖壳、内外桴、果皮及种皮这些保护组织中,且开始大量表达的时间正是生长接近停止时,于是推测ψG很可能通过降解草酸产生H2O2而推动这些组织细胞壁的木质化.  相似文献

为了研究对烟草中烟碱与绿原酸的快速生物降解,筛选了能有效降解烟草中烟碱与绿原酸的链霉菌Z6菌株与Z8菌株,考察了Z6和Z8菌株在不同培养基上的培养特征,探讨了所选菌株对烟碱与绿原酸的降解特性。实验结果表明,Z8链霉菌在烟草固体培养基中培养48h后,对培养基中烟碱的降解率达到83.9%;培养72h后,对烟碱的降解率可达到93.7%,此时烟草中的烟碱含量降低到0.38mg/g,达到了欧盟条例的无害化标准。Z6菌对绿原酸的降解程度较高,培养48h后,对绿原酸的降解率为57.1%;培养72h后,降解率可达到67.5%.  相似文献

讨论了从烟草废料中提取绿原酸的优势和甲醇、乙醇、丙酮、水等不同溶剂经超声波辅助提取绿原酸的效果:研究结果表明,用体积分数40%的甲醇得到的浸提液中,绿原酸质量浓度为2.11mg/mL,比以水为溶剂时高出近50%.不同浓度的甲醇溶液中,体积分数50%的甲醇提取绿原酸的浓度最高。对树脂的吸附动力学分析表明,大孔树脂CN-101对烟草浸提液中绿原酸的吸附遵循Freundlich等温方程,吸附和解析分离所得的绿原酸收率为87.6%.在超声辅助条件下,利用甲醇等有机溶剂提取烟草中的绿原酸,进而用大孔树脂进行吸附分离的方法可行。  相似文献

To explore the fruit development characteristics of Actinidia chinensis'Hort16A' , fruit shape indexes during 2-154 d after anthesis, contents of soluble sugar and titratable acid in fruit during 30-154 d after anthesis, and contents of organic acid components in fruit during 44-147 d after anthesis were analyzed. The results show that fruit of 'Hort16A' grows rapidly during 30-44 and 58-72 d after anthesis. Content of soluble solid in fruit during 100-140 d after anthesis is slightly higher than that during 2-93 d after anthesis, and increases rapidly during 147-154 d after anthesis. Fruit firmness decreases rapidly during 147-154 d after anthesis. In general, during 30-154 d after anthesis, content of soluble sugar in fruit increases gradually, while content of titratable acid decreases firstly, then increases, and decreases again. In the fruit, tartaric acid is not detected, content of quininic acid is the highest, contents of malic acid and citric acid are higher, and contents of succinic acid and fumaric acid are very low. Total content of organic acids increases firstly and then decreases, and reaches the highest value at 140 d after anthesis. It is suggested that in order to improve fruit quality of'Hort16A' , its cultivation and management plans will be set up based on fruit development characteristics.  相似文献

自花后46d到88d果实成熟.黄皮种子的发芽率由0升至100%.而活力指数逐渐上升,到花后74d达到最大值,之后略有下降.每粒种子的呼吸强度在花后46-67d持续增加,此后则渐渐减弱,但湿藏2d后又回升.黄皮种子的发育明显超前于果实.花后74d时.每粒种子的干重已接近最大值,这时种子活力最大.而果实的鲜重虽然已接近最大值.但其干重却只有成熟时的73%。花后46-53d的种子,其发芽率小于100%,轻微脱水能提高种子的发芽率及活力指数,花后60d至果实成熟.种子发芽率均为100%.这时任何程度的脱水都会引起活力指数的下降,但不同发育时期的黄皮种子耐脱水力有差别.其中以花后67d的耐脱水力最强.花后88d果实成熟时种子的耐脱水力最弱。  相似文献

对影响杜仲细胞悬浮培养及其次生代谢物绿原酸产生的几种主要因子进行了研究。结果表明,在杜仲细胞悬浮培养生产绿原酸的过程中,第15天绿原酸的含量达到最大值。35g/L的蔗糖为最适碳源,MS培养基为最适悬浮培养基,pH为5.3时利于绿原酸的合成,2,4-D、NAA对绿原酸合成的促进效果不大,添加1.0mg/L的6-BA绿原酸的合成效果较好。  相似文献

为了探索绿原酸和芸香苷的新型植物资源和生物活性,同时为低等级烤烟叶的综合利用开辟有效途径,本文采用高效液相色谱技术,对中国和湖南省不同生态烟区以及不同部位烤烟叶中绿原酸和芸香苷的含量进行了研究。结果表明,东北烟区低等级烤烟叶中的绿原酸和芸香苷含量很高;湖南省湘南烟区烤烟叶中绿原酸和芸香苷含量显著高于湘北和湘中烟区;不同部位烤烟叶中绿原酸含量差异不显著,芸香苷含量差异极显著,并且上部叶>中部叶>下部叶。  相似文献

Activity, control and primer requirements of starch phosphorylase in developing barley endosperm were investigated. Phosphorylase was detected in endosperm extracts from 3 days after anthesis. Unprimed activity was predominant between 2 and 10 days after anthesis, when it constituted 70–80% of total activity, but this proportion declined rapidly as the grain developed. The existence of at least 2 isoenzymes was indicated by studies of pH dependence and phosphate inhibition, and was further supported by acrylamide gel electrophoresis and column chromatography using DEAE-cellulose. The two isoenzymes which ere possibly both glyco proteins, appear in barley endosperm soon after anthesis. One appears capable of unprimed activity, and may be associated with the initiation of a-1,2 glucans, which then serve as primers for starch synthetase. This disappears by 13–15 days after anthesis. The other isoenzyme is capable of some unprimed activity but undergoes modification between 15 and 20 days after anthesis, resulting in the loss of unprimed activity. The relevance of the results to initiation of starch synthesis and to starch synthetase in amyloplasts is discussed.  相似文献
