本文用电子显微镜、蛋白质凝胶电泳和蛋白水解酶测定的方法,比较研究了三种蚊幼虫在感染苏云金杆菌以色列变种后中肠组织的病变,经苏云金杆菌以色列变种处理10分钟后,中肠细胞的细胞器即呈现病变,最明显的是线粒体呈现极度的电子致密状态,细胞质的粗糙内质网也开始瓦解,处理16小时后,细胞核膨胀,各种细胞器进一步瓦解,而围食膜仍然完好。此时细胞的盐溶性蛋白含量减少,图谱上明显缺少一条蛋白区带。另方面,感染10分钟后,中肠的蛋白水解酶活性便下降,不同蚊种的酶活性相差比较大,幼虫中肠的蛋白水解酶主要为胰蛋白酶。 根据结果分析,我们初步认为,苏云金杆菌以色列变种,对蚊幼虫的毒效是苏云金杆菌以色列变种的毒素对中肠细胞能量代谢和中肠蛋白酶活性产生抑制的结果。  相似文献

从神农架原始森林土壤中分离出苏云金芽孢杆菌 9株。经过生理生化和血清学鉴定 ,此 9株苏云金芽孢杆菌分属于H7、H6和H14。生物测定结果表明 :两株H7型菌株对棉铃虫幼虫有较高的毒力 ;另两株对致倦库蚊幼虫和白纹伊蚊幼虫有很强的毒杀作用 ,此两株属苏云金芽孢杆菌H14。  相似文献

本文报道棉铃虫、苏云金芽孢杆菌库斯塔克变种和单宁三者间的相互作用。苏云金芽孢杆菌芽孢晶体混合粉以6个不同的浓度(0%,0.005%,0.01%,0.015%,0.02%,0.025%湿重)分别加入含0.025%单宁和不含单宁的人工饲料中。初孵幼虫分别在上述饲料中饲养48h后,转入相应的不含苏云金芽孢杆菌的人工饲料上饲养,直到留存的幼虫化蛹。结果表明,单宁与苏云金芽孢杆菌混用可提高苏云金芽孢杆菌对棉铃虫的毒力,LD50降低45%,并可显著抑制该虫的生长发育,但二者在抑制生长发育方面无交互作用。选择取食试验显示,苏云金芽孢杆菌对五龄幼虫有显著的抑食性,但单宁无此作用,单宁与苏云金芽孢杆菌混用不会增强抑食性,但单宁能抑制苏云金芽孢杆菌菌落的生长,浓度高于15mg/100ml还能抑制芽孢的萌发。  相似文献

球形芽孢杆菌C3-41是我国分离的一株对蚊幼虫有毒杀作用的高毒力菌株,对库蚊、按蚊幼虫的毒性高于2362菌株,Southern杂交证明C\-3\|41总DNA中35Kb HindIII片段上带有419和514kD二元毒素基因,该片段由3479个核苷酸组成,核苷酸序列同2362菌株的二元毒素基因序列完全相同。含二元毒素基因的重组质粒pCW\|1和pCW\|2能在大肠杆菌中表达产生二元毒蛋白,但表达量低,重组子杀蚊毒性低。无晶体型苏云金芽孢杆菌以色列亚种重组子在其芽孢形成中能产生以晶体形式存在的二元毒素蛋白,其全发酵液和纯化晶体蛋白的杀蚊活性与C\-3\|41相近。  相似文献

苏芸金杆菌以色列变种(Bacillus thuringtensis var.tsraelensis de Barjac)对尖音库蚊(Culex pipiensvar. pallens Coq uillett)三龄幼虫具有极高的毒性,在浓度为1.5×104芽孢/毫升时,死亡率达100%。其他几个变种B. T. var. thuringiensis(E—009)var. kurstaki(HD—1)、var.kenyae(7404)、var. ostriniae(006)浓度达4×105芽孢/毫升时仍未见毒效。 用液体双相与等密度离心法将以色列变种的晶体与芽孢分离,分别研究晶体(纯度达99.5%以上)、芽孢(纯度达99.5%以上)对尖音库蚊幼虫的毒性。发现使蚊幼虫致死主要是晶体LC50为0.023微克/毫升,芽孢也具有毒性LC50为0.86微克/毫升,晶体与芽孢混用制剂的LC50为(0.014微克晶体+0.014微克芽孢)/毫升。 晶体制剂在30℃和21℃的温度条件下毒力没有变化,但芽孢制剂在21℃毒力显著下降;芽孢制剂致死幼虫,立即用相差显微镜镜检,发现肠道内有大量正在萌发的芽孢,对芽孢致死的原因进行了初步分析。  相似文献

球形芽孢杆菌Csub3-41菌株(Bacillus sphaericus C3-41)对致倦库蚊(Culex puinquefa-seiatus)幼虫有很高毒效,对2龄和3-4龄幼虫的半致死剂量(LD50)分别为63.1 和89.7芽孢/蚊幼虫。处理浓度越高,取食时间越长,蚊幼虫取食到的杀蚊活性物质量越多,死亡率越高。当蚊幼虫取食亚致死剂量杀蚊活性物质后,球形芽孢杆菌在感染的活幼虫体内不增殖;但当蚊幼虫取食致死剂量杀蚊活性物质后,蚊幼死亡,球形芽孢杆菌在死蚊幼虫体内增殖明显,6天内芽孢从感染初期的1.86X102蚊幼虫增加到1.59X106/蚊幼虫。芽孢在死蚊幼虫体内能正常萌发、生长、产孢和形成毒素。增殖的芽孢同样对致倦库蚊幼虫有较高毒力。  相似文献

通过测定棉铃虫4龄幼虫,每隔12 h的摄食量、体重净增量以及丝氨酸蛋白酶活性的变化,揭示了它的取食行为及中肠蛋白酶活性的变化规律。结果表明:棉铃虫4龄幼虫取食及消化呈有规律的变化,蜕皮后4龄初,摄食量、体重净增量、类胰蛋白酶、类胰凝乳蛋白酶和总蛋白酶活性均逐渐增加;进入中期(36~48 h),各项指标达到最高值;其后,随着摄食量的下降,各项指标逐渐下降。同时, 连续观察幼虫取食高峰期的取食行为6 h,结果表明: 其间一次连续取食的平均时间为(3.6±0.4) min/次, 总平均取食时间为(62.4±8.9) min,约占整个时间的1/6。该文还对棉铃虫4龄幼虫不同时段取食行为与蛋白酶活性变化之间的相关性及调控机制作了初步讨论。  相似文献

A strain of Clostridium bifermentans individualized as serovar malaysia (C.b.m.) according to its specific H antigen is toxic to mosquito and blackfly larvae when given orally. The toxicity occurs in sporulated cells which contain, in addition to spores, proteinic parasporal inclusion bodies and feather-like appendages; the amino acid content of the inclusion bodies is similar to that of Bacillus thuringiensis serovar israelensis (B.t.i.) and B. sphaericus crystals. The toxicity to Anopheles stephensi is as high as that of B.t.i. and the best strains of B. sphaericus. Culex pipiens is somewhat less susceptible, and Aedes aegypti much less. Pure parasporal inclusion bodies, isolated by ultracentrifugation on sucrose gradients, are highly toxic to mosquito larvae. The larvicidal power is destroyed by heating at 80 degrees C or by treatment with 50 mM NaOH. It is preserved by freeze-drying. The innocuity to mice of the sporulated cells is shown by different routes of administration: force-feeding, percutaneous, subcutaneous, intraperitoneal or intravenous injections. The potential for the biological control of mosquito and blackfly larvae is suggested.  相似文献

The larvicidal activity of Bacillus thuringiensis var. israelensis against mosquitoes and the blackfly is included in parasporal crystalline bodies which are produced during sporulation. Following ingestion, the crystals are solubilized in the larval midgut and induce death within a short time; the spores germinate in the dead larvae and complete a growth cycle. The fate of the spores in surviving live larvae was elucidated by using a nonlarvicidal B. thuringiensis var. israelensis mutant. When introduced as the only food source, spores of this mutant support development to the adult stage of newly hatched Aedes aegypti larvae at a rate directly related to spore concentration. The conclusion that spores of B. thuringiensis var. israelensis are digested in the larval gut was substantiated by following the incorporation of [35S]methionine-labeled spores into larval tissues.  相似文献

【目的】丝氨酸蛋白酶抑制剂家族蛋白是昆虫中调控自身免疫反应的重要蛋白酶抑制剂,本研究旨在研究家蚕Bombyx mori丝氨酸蛋白酶抑制剂2(Bmserpin2)在家蚕2个重要的自身免疫通路即酚氧化酶原(prophenol oxidase, PPO)激活通路和革兰氏阳性菌诱导抗菌肽的TOLL通路中的调控作用。【方法】PCR扩增家蚕Bmserpin2基因片段后原核表达并通过镍柱纯化。利用纯化后的重组Bmserpin2蛋白分别与胰蛋白酶、胰凝乳蛋白酶、弹性蛋白酶和蛋白酶K反应,检测Bmserpin2对上述蛋白酶活性的影响。通过RT-qPCR检测Bmserpin2在家蚕5龄第3天幼虫头、中肠、脂肪体、血淋巴、丝腺和表皮组织中表达的模式。往家蚕5龄第3天幼虫注射Bmserpin2重组蛋白,检测Bmserpin2对其血淋巴中PPO活性的影响。通过滕黄微球菌Micrococcus luteus诱导家蚕5龄第3天幼虫产生抗菌肽并注射Bmserpin2重组蛋白后,RT-qPCR检测其血淋巴中抗菌肽基因gloverin2和moricin表达量。【结果】成功构建重组质粒并表达纯化目的蛋白Bmserpin2。通过与不同蛋白酶反应得出Bmserpin2可极显著抑制消化酶胰蛋白酶和弹性蛋白酶活性,对胰凝乳蛋白酶和蛋白酶K活性影响不显著,提示Bmserpin2对不同蛋白酶具有生物学活性和催化特异性。基因表达模式显示Bmserpin2在家蚕5龄幼虫血淋巴和脂肪体中表达量最高。家蚕5龄幼虫注射重组Bmserpin2蛋白后发现目的蛋白能有效抑制血淋巴中PPO活性。利用滕黄微球菌诱导家蚕5龄幼虫产生抗菌肽后,滕黄微球菌和Bmserpin2混合注射组中血淋巴中抗菌肽基因gloverin2和moricin的转录表达与只注射滕黄微球菌的比较被显著下调。【结论】Bmserpin2可能参与家蚕酚氧化酶原激活和TOLL途径的胞外级联反应的免疫通路。  相似文献

本研究在室内评价了在济宁采集的捕食性刘氏中剑水蚤单用及与球形芽孢杆菌(B.s.)或苏云金杆菌以色列变种(B.t.i.)合用对蚊幼虫的控制效果。在室内,刘氏中剑水蚤对白蚊伊蚊Ⅰ龄幼虫的捕食效果好于对中华按蚊和淡色库蚊幼虫的效果。蚊笼模拟试验表明,刘氏中剑水蚤可分别于9、10和7周内消灭淡色库蚊、中华按蚊和白蚊伊蚊的幼虫种群。该剑水蚤增长速率快,大量培养经济易行。现场试验证明,刘氏中剑水蚤虽可在蚊虫孳生地存活时间长于蚊虫季节,但单用该剑水蚤不能象化学杀虫剂一样快速完全地控制三种蚊虫。刘氏中剑水蚤与B.s.或B.t.i.合用,应用刘氏中剑水蚤的长持效和生物杀虫剂的速效,取得了高效、持久的防治效果。因此二者合用可用于蚊虫防治并可作为蚊虫综合防治的一个组成部分。  相似文献

Carcasses of mosquito larvae killed by Bacillus thuringiensis var. israelensis allow its complete growth cycle (germination, vegetative growth, and sporulation), thus becoming toxic themselves to scavenging larvae. In this study, we demonstrate that the bacterium is capable of inducing death of Aedes aegypti pupae and of recycling in the resulting carcasses. B. thuringiensis var. israelensis-killed pupae were obtained by treating 40-hr-old synchronized fourth instar larvae with a low dose of spores (8000/ml). The fraction of dead pupae was reduced by higher or lower spore concentrations as well as by treating younger or older larval populations (both fourth instar): Increased proportions of dead larvae were obtained at higher concentration or by earlier treatment, whereas lower concentrations or later treatment resulted in more living pupae. Multiplication of B. thuringiensis var. israelensis is shown to occur in the carcasses of dead pupae. The number of spores in each pupal carcass followed a similar kinetic as in larval carcasses, but the final yield was about 10-fold higher, apparently reflecting the difference in dry weight between the two mosquito developmental stages (426 micrograms vs 83 micrograms, respectively). The specific larvicidal activity in a homogenized dead pupa was similar to that of B. thuringiensis var. israelensis powder, LC50 of about 600 spores/ml.  相似文献

Immediate (24 hours exposure) and long-term (until the emergence of the adults) effects of different doses of a primary powder of Bacillus thuringiensis subsp. israelensis (B.t.i.) against first and second instar Aedes aegypti larvae were monitored. The long-term effect was dose-dependent and was materialized by a prolongation of the preimaginal development and continuous cumulative mortality until the emergence of the adults. Mortality values were higher during the fourth larval instar and the pupal stage. Some of the larvae reaching the fourth instar were smaller in size and remained in this state a 2-4 times longer period than in the control and finally died as larvae or very small pupae. The long-term effect was more intense as the treatment was applied earlier during the larval development. The correlation of the immediate lethal effect with the late effect allows the evaluation of the total impact of a larvicidal treatment.  相似文献

Abstract  The control efficacy of a predatory copepod, Mesocyclops leukarti , collected in Jining, and its compatibility with Bacillus sphaericus (B.s. ) and B. thuringiensis var. csraefensis H-14 ( B. t. i. ) against mosquito larvae were evaluated in the laboratory and in the field. In the laboratory, M. leukarti showed higher predatory activity toward first instar larvae of Aedes albopictus than toward that of Anopheles sinensis and Culex pipiens pallens. Cage simulation test showed that larvae of Cx. pipiens pallens, An. sinensis and Ae. albopictus could be eliminated within 9, 10 and 7 weeks after interaction with the copepods respectively. M. leukarti population can grow quickly, and its mass production is easy and economic. However, pilot field trials indicated that M. leukarti alone could not eradicate mosquito larvae of the three species rapidly and completely as by chemical insecticides, although they could persist in mosquito breeding habitats longer than the mosquito season. When Mesocyclops was used in combination with B. s. and B. t. i. , higher and sustainable control effects were achieved. So this combination, which achieved the rapid larvicidal and sustainable control effect, was presumed to be practical and could be integrated into mosquito control program.  相似文献
