The circannual pupation rhythm of Anthrenus verbasci is entrained to an environmental cycle by changes in photoperiod. Exposure of larvae reared under short-day conditions to 4 weeks of long days can induce phase-dependent phase shifts. In the present study, we examined the range of photoperiodic changes effective for phase shifts at 20°C. For larvae under light/dark (LD) 12:12 conditions, 4-week exposure to LD 14:10 caused a clear phase delay, as great an extent as that brought about by exposure to LD 15:9 and LD 16:8. In contrast, the delay brought about by exposure to LD 13:9 was slight. For larvae under LD 10:14, exposure to LD 14:10 and LD 16:8 for 4 weeks induced a phase delay, but exposure to LD 12:12 did not. These results indicate that a clear phase delay is induced when the photoperiodic change exceeds a critical value in the photophase between 13 and 14 h, regardless of the amplitude of the change. Although phase advances were smaller than phase delays, they depended on the amplitude of photoperiodic changes rather than the absolute photophase duration in contrast to the case of the phase delay.  相似文献

We know that entrainment, a stable phase relationship with an environmental cycle, must be established for a biological clock to function properly. Phase response curves (PRCs), which are plots of phase shifts that result as a function of the phase of a stimulus, have been created to examine the mode of entrainment. In circadian rhythms, single-light pulse PRCs have been obtained by giving a light pulse to various phases of a free-running rhythm under continuous darkness. This successfully explains the entrainment to light-dark cycles. Some organisms show circannual rhythms. In some of these, changes in photoperiod entrain the circannual rhythms. However, no single-pulse PRCs have been created. Here we show the PRC to a long-day pulse superimposed for 4 weeks over constant short days in the circannual pupation rhythm in the varied carpet beetle Anthrenus verbasci. Because the shape of that PRC closely resembles that of the Type 0 PRC with large phase shifts in circadian rhythms, we suggest that an oscillator having a common feature in the phase response with the circadian clock, produces a circannual rhythm.  相似文献

In circadian rhythms, the shape of the phase response curves (PRCs) depends on the strength of the resetting stimulus. Weak stimuli produce Type 1 PRCs with small phase shifts and a continuous transition between phase delays and advances, whereas strong stimuli produce Type 0 PRCs with large phase shifts and a distinct break point at the transition between delays and advances. A stimulus of an intermediate strength applied close to the break point in a Type 0 PRC sometimes produces arrhythmicity. A PRC for the circannual rhythm was obtained in pupation of the varied carpet beetle, Anthrenus verbasci, by superimposing a 4-week long-day pulse (a series of long days for 4 weeks) over constant short days. The shape of this PRC closely resembles that of the Type 0 PRC. The present study shows that the PRC to 2-week long-day pulses was Type 1, and that a 4-week long-day pulse administered close to the PRC’s break point induced arrhythmicity in pupation. It is, therefore, suggested that circadian and circannual oscillators share the same mode in phase resetting to the stimuli.  相似文献

The mammalian circadian clock lying in suprachiasmatic nucleus (SCN) is synchronized to about 24 h by the environmental light-dark cycle (LD). The circadian clock exhibits limits of entrainment above and below 24 h, beyond which it will not entrain. Little is known about the mechanisms regulating the limits of entrainment. In this study, we show that wild-type mice entrain to only an LD 24 h cycle, whereas Clock mutant mice can entrain to an LD 24, 28, and 32 h except for LD 20 h and LD 36 h cycle. Under an LD 28 h cycle, Clock mutant mice showed a clear rhythm in Per2 mRNA expression in the SCN and behavior. Light response was also increased. This is the first report to show that the Clock mutation makes it possible to adapt the circadian oscillator to a long period cycle and indicates that the clock gene may have an important role for the limits of entrainment of the SCN to LD cycle.  相似文献

Previous studies demonstrated a high-amplitude, diel cycle for the hemolymph JH titer in the wing-polymorphic cricket, Gryllus firmus. The JH titer rose and fell in the flight-capable morph (long-winged, LW(f)) above and below the relatively temporally invariant JH titer in the flightless (short-winged, SW) morph. The morph-specific JH titer cycle appeared to be primarily driven by a morph-specific diel cycle in the rate of JH biosynthesis. In the present study, cycles of the JH titer and rate of JH biosynthesis in the LW(f) morph persisted in the laboratory under constant darkness with an approximate 24 h periodicity. The JH titer cycle also shifted in concert with a shift in the onset of the scotophase, was temperature compensated in constant darkness, and became arrhythmic under constant light. These results provide strong support for the circadian basis of the morph-specific diel rhythm of the JH titer and JH biosynthetic rate. Persistence of the JH titer cycle under constant darkness in multiple LW-selected and SW-selected stocks also provides support for the genetic basis of the morph-associated circadian rhythm. The morph-specific JH titer cycle was observed in these stocks raised in the field, in both males and females, in each of 3 years studied. The onset of the cycle in the LW(f) morph, a few hours before sunset, correlated well with the onset of the cycle, a few hours before lights-off, in the laboratory. The morph-specific JH titer cycle is a general feature of G. firmus, under a variety of environmental conditions, and is not an artifact of specific laboratory conditions or specific genetic stocks. It is a powerful experimental model to investigate the mechanisms underlying endocrine circadian rhythms, their evolution, and their impact on life history evolution.  相似文献

In the female turnip moth, Agrotis segetum, a pheromone biosynthesis activating neuropeptide (PBAN) stimulates sex pheromone biosynthesis which exhibits a daily rhythm. Here we show data supporting a circadian rhythm in PBAN release from the corpora cardiaca, which we propose regulates the endogenous rhythm in sex pheromone biosynthesis. This conclusion is drawn as the observed daily rhythm in PBAN-like immunoreactivity in the hemolymph is persistent in constant darkness and is phase-shifted by an advanced light:dark cycle. PBAN-like immunoreactivity was found in the brain, the optic lobe, the suboesophageal ganglion and in the retrocerebral complex. In each hemisphere ca. 10 immunopositive neurons were observed in the pars intercerebralis and a pair of stained somata in the dorso-lateral protocerebrum. A cluster of cells containing PBAN-like immunoreactive material was found in the tritocerebrum and three clusters of such cells were found in the SOG. Their processes reach the corpora cardiaca via nervi corporis cardiaci and the dorsal surface of the corpora allata via the nervi corporis allati.  相似文献

The circadian clock gene period (Gryllus bimaculatus period, Gbper) plays a core role in circadian rhythm generation in adults of the cricket Gryllus bimaculatus. We examined the role of Gbper in nymphal crickets that show a diurnal rhythm rather than the nocturnal rhythm of the adults. As in the adult optic lobes, Gbper mRNA levels in the head of the third instar nymphs showed daily cycling in light-dark cycles with a peak at mid night, and the rhythm persisted in constant darkness. Injection of Gbper double-stranded RNA (dsRNA) into the abdomen of third instar nymphs knocked-down the mRNA levels to 25% of that in control animals. Most Gbper dsRNA injected nymphs lost their circadian locomotor activity rhythm, while those injected with DsRed2 dsRNA as a negative control clearly maintained the rhythm. These results suggest that nymphs and adults share a common endogenous clock mechanism involving the clock gene Gbper.  相似文献

Physiological characteristics of the photoreceptors involved in the photoperiodic induction of diapause were investigated in the flesh fly Sarcophaga similis. Both the early and late phases of scotophase were sensitive to light and a light pulse during each of these phases prevented diapause. Certain physiological differences between the phases were, nevertheless, detected. Compared with early scotophase, late scotophase required a light pulse with a long period and a large number of night interruption photoperiodic cycles in order to effectively prevent diapause. The diapause-averting effects of a light pulse during early scotophase were canceled by an additional long dark period, but those during late scotophase were not. Thus, the diapause-averting effects produced during early scotophase are different to those produced during late scotophase. The early scotophase was sensitive to light at wavelengths of 470 nm or shorter, but not to light of 583 nm or longer. In contrast, the late scotophase was sensitive to light of a broad range of wavelengths, ranging from 395 to 660 nm. Furthermore, the early scotophase was considerably more sensitive to monochromatic light with low photon flux density than the late scotophase. These results suggest that different types of photoreceptor are involved in the photoperiodic response.  相似文献

Expression of the circadian rhythm gene timeless was investigated in the pitcher-plant mosquito, Wyeomyia smithii (Coq.), and was found to vary with time of day, instar of diapause, and latitude of origin. The temporal pattern of timeless expression differed between the two diapausing instars and was significantly higher in southern (38-40 degrees N) than in northern (50 degrees N) populations, when diapausing instar was held constant. Expression of timeless is therefore both developmentally and evolutionarily variable. This result provides the first example of a latitudinal difference in the expression of timeless, suggesting that, along with evidence from other insects, timeless has the potential to affect photoperiodic response and its adaptive evolution in temperate seasonal environments.  相似文献

A two variable model with delay in both the variables, is proposed for the circadian oscillations of protein concentrations in the fungal species Neurospora crassa. The dynamical variables chosen are the concentrations of FRQ and WC-1 proteins. Our model is a two variable simplification of the detailed model of Smolen et al. (J. Neurosci. 21 (2001) 6644) modeling circadian oscillations with interlocking positive and negative feedback loops, containing 23 variables. In our model, as in the case of Smolen's model, a sustained limit cycle oscillation takes place in both FRQ and WC-1 protein in continuous darkness, and WC-1 is anti-phase to FRQ protein, as observed in experiments. The model accounts for various characteristic features of circadian rhythms such as entrainment to light dark cycles, phase response curves and robustness to parameter variation and molecular fluctuations. Simulations are carried out to study the effect of periodic forcing of circadian oscillations by light-dark cycles. The periodic forcing resulted in a rich bifurcation diagram that includes quasiperiodicity and chaotic oscillations, depending on the magnitude of the periodic changes in the light controlled parameter. When positive feedback is eliminated, our model reduces to the generic one dimensional delay model of Lema et al. (J. Theor. Biol. 204 (2000) 565), delay model of the circadian pace maker with FRQ protein as the dynamical variable which represses its own production. This one-dimensional model also exhibits all characteristic features of circadian oscillations and gives rise to circadian oscillations which are reasonably robust to parameter variations and molecular noise.  相似文献

Photoperiodic time measurement of Diatraea grandiosella, a Pyralid moth, was investigated for its role in the determination of diapause by using various night-interruption protocols. The photoperiodic-response curve showed a temperature dependence under short days, whereas at long days it was stable in the range between 20 and 30°C. A light pulse averted diapause most effectively when it was placed 6 h after lights-off. Earlier pulses were less effective but gradually increased in effectiveness as the time of the pulse approached the critical time, i.e. 6 h after lights-off. A strong conversion in response appeared as the pulse passed the critical time. The same response pattern was observed both in diapause induction and termination. The species required a long light pulse to avert diapause completely, even if the light pulse was placed during the critical time; 45 min was required to reverse diapause in 50% of the individuals. The most crucial event for photoperiodic time measurement in this species was whether a night phase 6–7.5 h after lights-off was illuminated or not. This hourglass-like feature was also shown in more complex night-interruption protocols with 2–3 light pulses. A possible relation of this phenomenon to the cricadian pacemaker was sought. The Dual System Theory failed to account for most of the features for the photoperiodic time measurements of D. grandiosella found in this study.  相似文献

Cryptochrome (CRY) is a blue-light-absorbing protein involved in the photic entrainment of the circadian clock in Drosophila melanogaster. We have investigated the locomotor activity rhythms of flies carrying cryb mutant and revealed that they have two separate circadian oscillators with different responsiveness to light. When kept in constant light conditions, wild-type flies became arrhythmic, while cryb mutant flies exhibited free-running rhythms with two rhythmic components, one with a shorter and the other with a longer free-running period. The rhythm dissociation was dependent on the light intensities: the higher the light intensities, the greater the proportion of animals exhibiting the two oscillations. External photoreceptors including the compound eyes and the ocelli are the likely photoreceptors for the rhythm dissociation, since rhythm dissociation was prevented in so1;cryb and norpAP41;cryb double mutant flies. Immunohistochemical analysis demonstrated that the PERIOD expression rhythms in ventrally located lateral neurons (LNvs) occurred synchronously with the shorter period component, while those in the dorsally located per-expressing neurons showed PER expression most likely related to the longer period component, in addition to that synchronized to the LNvs. These results suggest that the Drosophila locomotor rhythms are driven by two separate per-dependent clocks, responding differentially to constant light.  相似文献

To investigate whether the photoperiodic clocks of species possessing strongly self-sustaining circadian clocks share identical features, we compared the full response cycle (initiation and termination of the response) in body mass and testes of the non-migratory house sparrow (Passer domesticus) with that of the migratory redheaded bunting (Emberiza bruniceps) under Nanda-Hamner experiments. Birds were exposed to a 36 h day (L:D=6:30 h), controls exposed to a 24 h day (L:D=6:18 h), for a period of 31 weeks. By week 18 of L:D=6:18 h, there was a small increase in body mass among sparrows, but not among buntings, and the testes of bunting did not grow, while those of sparrow grew slightly. The response to L:D=6:30 h is of particular interest. There was a rapid gain and subsequent loss in the body mass of bunting, but not of sparrows. Further, both species underwent a testicular cycle as if they were exposed to long days, but the response of sparrows was slower and hence delayed the attainment of peak testicular size. Such a differential response to exotic light cycles between these two photosensitive species, despite their similar circadian oscillatory properties (strong self-sustainment), could suggest a species-specific adaptation of the endogenous clock involved in photoperiodic regulation of avian seasonality.  相似文献

Temperate zone birds are highly seasonal in many aspects of their physiology. In mammals, but not in birds, the pineal gland is an important component regulating seasonal patterns of primary gonadal functions. Pineal melatonin in birds instead affects seasonal changes in brain song control structures, suggesting the pineal gland regulates seasonal song behavior. The present study tests the hypothesis that the pineal gland transduces photoperiodic information to the control of seasonal song behavior to synchronize this important behavior to the appropriate phenology. House sparrows, Passer domesticus, expressed a rich array of vocalizations ranging from calls to multisyllabic songs and motifs of songs that varied under a regimen of different photoperiodic conditions that were simulated at different times of year. Control (SHAM) birds exhibited increases in song behavior when they were experimentally transferred from short days, simulating winter, to equinoctial and long days, simulating summer, and decreased vocalization when they were transferred back to short days. When maintained in long days for longer periods, the birds became reproductively photorefractory as measured by the yellowing of the birds' bills; however, song behavior persisted in the SHAM birds, suggesting a dissociation of reproduction from the song functions. Pinealectomized (PINX) birds expressed larger, more rapid increases in daily vocal rate and song repertoire size than did the SHAM birds during the long summer days. These increases gradually declined upon the extension of the long days and did not respond to the transfer to short days as was observed in the SHAM birds, suggesting that the pineal gland conveys photoperiodic information to the vocal control system, which in turn regulates song behavior.  相似文献

The mole crab Emerita talpoida migrates with the tide in the swash zone of sand beaches. A circatidal rhythm in vertical swimming underlies movement, in which mature male crabs show peak swimming activity 1-2 h after the time of high tides at the collection site. In addition, there is a secondary rhythm in activity amplitude, in which crabs are maximally active following low amplitude high tides and minimally active following high amplitude high tides. The present study determined the phase response relationship for entrainment of the circatidal rhythm with mechanical agitation and whether the cycle in activity related to tidal amplitude could be entrained by a cycle in the duration of mechanical agitation at the times of consecutive high tides. After entrainment with mechanical agitation on an orbital shaker, activity of individual crabs was monitored in constant conditions with a video system and quantified as the number of ascents from the sand each 0.5 h. Mechanical agitation at the times of high tide, mid-ebb and low tide reset the timing of the circatidal rhythm according to the timing relationship to high tide. However, mechanical agitation during flood tide had no entrainment effect. In addition, a cycle in duration of mechanical agitation entrained the rhythm in activity amplitude associated with tidal amplitude. Both rhythms and entrainment effectiveness over the tidal cycle may function to reduce the likelihood of stranding above the swash zone.  相似文献

The rhythm of carbon dioxide output in Bryophyllum leaves was entrained on exposure to 0.25 h of white light every 24 h. Entrainment also occurred on similar exposure to monochromatic radiation in spectral bands centred at 660 nm and, to a lesser extent, at 730 nm, but a band centred at 450 nm was without effect. A skeleton irradiation programme comprising two 0.25-h exposures to white light per 24 h also entrained the rhythm when the intervening dark periods were either 7.5 h and 16 h, or 10.5 h and 13 h. The rhythm disappeared when the two exposures were separated by 11.5-h and 12-h dark periods. Regular 0.25-h exposures to red light separated by 11.75-h periods of darkness also resulted in loss of the rhythm. Red/far-red reversibility was observed in irradiation schedules having either one or two exposures to red light daily. In the latter case, far-red reversal of the effects of one of the exposures to red light resulted in entrainment of the rhythm by the other, instead of abolition of the rhythm. The occurrence of distinct red/far-red reversibility suggests strongly that phytochrome is the pigment involved in entrainment of this rhythm by cycles of light and darkness.Abbreviation LD light-dark rhythm  相似文献
