Responses of 137 neurons of the rostral pole of the reticular and anterior ventral thalamic nuclei to electrical stimulation of the ventrolateral nucleus and motor cortex were studied in 17 cats immobilized with D-tubocurarine. The number of neurons responding antidromically to stimulation of the ventrolateral nucleus was 10.5% of all cells tested (latent period of response 0.7–3.0 msec), whereas to stimulation of the motor cortex it was 11.0% (latent period of response 0.4–4.0 msec). Neurons with a dividing axon, one branch of which terminated in the thalamic ventrolateral nuclei, the other in the motor cortex, were found. Orthodromic excitation was observed in 78.9% of neurons tested during stimulation of the ventrolateral nucleus and in 52.5% of neurons during stimulation of the motor cortex. Altogether 55.6% of cells responded to stimulation of the ventrolateral nucleus with a discharge of 3 to 20 action potentials with a frequency of 130–350 Hz. Similar discharges in response to stimulation of the motor cortex were observed in 30.5% of neurons tested. An inhibitory response was recorded in only 6.8% of cells. Convergence of influences from the thalamic ventrolateral nucleus and motor cortex was observed in 55.7% of neurons. The corticofugal influence of the motor cortex on responses arising in these cells to testing stimulation of the ventrolateral nucleus could be either inhibitory or facilitatory.A. A. Neuronal responses to stimulation of vestibular motor and orbital cerebral cortex were recorded by extracellular techniques in the lateral and medial vestibular nuclei of the bulbar complex during experiments on unanesthetized, immobilized cats. Both phasic and (mostly) tonic response of predominantly inhibitory type were observed. Horseradish peroxidase was injected into the aforementioned nuclei of the vestibular complex during the course of morphological experiments. Labeled neurons were found in the anterior supra- and ectosylvian cerebral gyri, the region of the cruciform sulcus, and that of the orbital cerebral cortex. Evoked potentials arising in the motor cortex in response to its direct stimulation (dendritic and slow negative potentials), to stimulation of the ventrolateral (primary response) and intralaminar (nonspecific response) thalamic nuclei, and to stimulation of the pyramidal tracts (antidromic response), and also postsynaptic responses of neurons corresponding to them were studied in acute experiments on curarized cats. Evoked potentials arising in response to direct cortical stimulation and also to stimulation of the specific and nonspecific thalamic nuclei and pyramidal tracts were recorded from the same point of the motor cortex, and the corresponding intracellular responses were recorded from the same neuron. Slow negative potentials arising under these conditions of stimulation and the IPSPs corresponding to them were shown to have an identical time course. The results show that slow negative potentials are a reflection of hyperpolarization of pyramidal neurons. It is suggested that the individual components of responses evoked by direct stimulation of the cortex and thalamic nuclei have a common genesis.I. S. Afferent connections of the retrosplenial area of the rat limbic cortex were investigated by the retrograde horseradish peroxidase axon transport method. After injection of horseradish peroxidase (HRP) into area 29 of the cortex, HRP-labeled cells were found in the dorsal part of the lateral geniculate body and the posterolateral, pretectal, and anterior dorsal thalamic nuclei. Connections were found between cortical area 29 and visual projection areas (areas 17 and 18a) and with area 29 on the contralateral side of the brain. The results are evidence that all the principal visual structures of the thalamus and the visual cortical projection area form direct projections to the retrosplenial cortex.I. M. Properties of EPSP, evoked in efferent neurons of the parietal associative cortex by stimulation of the cerebellar nuclei, were studied in acute experiments on anesthetized and immobilized cats; intracellular recording was used. The neurons were identified by their antidromic activation after stimulation of the motor cortex, pontinen.n. proprii, or red nucleus. The effects of stimulation of the cerebellar nuclei were of oligo- and polysynaptic nature. The latencies of cerebellofugal EPSP correlated with the latencies of antidromic activation, and correlations were significant both in the cases when the effects of stimulation of separate efferent projections (cortico-cortical, cortico-pontine, or cortico-rubral) and the effects of stimulation of separate cerebellar nuclei were analyzed. It was shown that the rabbit sensorimotor cortex received afferent fibers from neurons located in the specific, nonspecific, and association thalamic nuclei using the retrograde axonal transport technique. The distribution, dimensions, and shape of the somata of relay neurons spread through the thalamic nuclei were analyzed. The total number of neurons sending out thalamo-sensorimotor-cortical fibers was calculated and the coordinates of loci with the highest density of these cells in each thalamic nucleus were identified. Multipolar and stellate cells with somata measuring 12–20 µm and 10–15 µm in diameter, respectively, prevailed amongst relay neurons. Amongst the specific nuclei, the majority of afferent fibers are sent out by the ventrolateral, ventral anterior, and anterior ventral nuclei. A comparable number of afferent fibers are sent out by the mediodorsal and paracentral nuclei; these split up among the association nuclei and paracentral nuclei, respectively. Of 103 neurons in the rostral part of the posterior sigmoid gyrus of the cat cortex 30 responded to stimulation of the ventro-posterolateral and ventrolateral nuclei of the thalamus (VPL and VL), 42 responded to stimulation of VL only, and 31 to stimulation of VPL only. It was shown by intracellular recording that stimulation of VPL induces a spike response with or without subsequent IPSPs in some neurons and an initial IPSP in others. The spike frequency of single neurons reached 60/sec, but the IPSP frequency never exceeded 10–20/sec. Stimulation of VL was accompanied by: a) antidromic spike responses; b) short-latency monosynaptic EPSPs and spikes capable of following a stimulation frequency of 100/sec; c) long-latency polysynaptic EPSPs and spikes appearing in response to stimulation at 4–8/sec; d) short-latency IPSPs; e) long-latency IPSPs increasing in intensity on repetition of infrequent stimuli. It is concluded that the afferent inputs from the relay nuclei to neurons of the somatosensory cortex are heterogeneous. An important role is postulated for recurrent inhibition in the genesis of the long-latency IPSPs arising in response to stimulation of VL, and for direct afferent inhibition during IPSPs evoked by stimulation of VPL. It is shown that the rostral part of the posterior sigmoid gyrus performs the role of somatic projection and motor cortex simultaneously.A. A. Connections of the retrosplenial cortex with the hippocampus in rats were investigated by the method of retrograde axonal transport of horseradish peroxidase (HRP). If a crystal of HRP was introduced into the dorsocaudal part of the retrosplenial cortex, HRP-labeled cells were found in the rostral pole of hippocampal area CA3 and in the presubiculum. After iontophoretic injection of HRP into the rostral pole of the hippocampus, HRP-labeled cells were found in layers V and VI of the retrosplenial cortex and in the presubiculum. The results are evidence of reciprocity of direct connections between the rostral pole of the hippocampus and the retrosplenial cortex, and also of the existence of direct efferents of the dorsal subiculum to the rostral pole of the hippocampus.I. The effect of excitation of the contralateral nuclei of the cerebellum on the activity of anti-dromically-identified fast and slow pyramidal neurons in the sensorimotor region (SMR) of the cerebral cortex was investigated in delicate experiments on unnarcotized cats immobilized with procurane. It was found that excitation of the above nuclei could evoke peak responses in the neurons of the pyramidal tract (PT), but more often it modified their background activity. These responses were more pronounced on excitation of the intermediate nucleus (IN) or the dentate nucleus (DN), although they also often arose on excitation of the fastigial nucleus (FN). Qualitatively different effects (stimulative and inhibitory) were observed in some neurons on excitation of different nuclei in the cerebellum. It was found that the nature of the responses by fast neurons in the PT to excitation of the central nuclei in the cerebellum did not differe substantially from the nature of the responses by slow neurons, but that the responses of the latter arose with a longer latent period, and in most cases were less pronounced.L. In acute experiments on cats anesthetized with thiopental (30–40 mg/kg, intraperitoneally) and immobilized with D-tubocurarine (1 mg/kg) responses of 145 neurons of the reticular and 158 neurons of the ventral anterior nuclei of the thalamus to electrical stimulation of the centrum medianum were investigated. An antidromic action potential appeared after a latent period of 0.3–2.0 msec in 4.1% of cells of the reticular nucleus and 4.4% of neurons of the ventral anterior nucleus tested in response to stimulation. The conduction velocity of antidromic excitation along axons of these neurons was 1.7–7.6 m/sec. Neurons responding with an antidromic action potential to stimulation both of the centrum medianum and of other formations were discovered, electrophysiological evidence of the ramification of such an axon. Altogether 53.8% of neurons of the reticular nucleus and 46.9% of neurons of the ventral anterior nucleus responded to stimulation of the centrum medianum by orthodromic excitation. Among neurons excited orthodromically two groups of cells were distinguished: The first group generated a discharge consisting of 6–12 action potentials with a frequency of 130–640 Hz (the duration of discharge did not exceed 60 msec), whereas the second responded with a single action potential. Inhibitory responses were observed in only 0.7% of neurons of the reticular nucleus and 4.4% of the ventral anterior nucleus tested. Connections of the somatosensory cortex surrounding the postcruciate fossa with the lateral region of the motor area (in the cruciate sulcus) were established by the Nauta-Gygax and Fink-Heimer methods and also by the retrograde horse-radish peroxidase transport method. A high degree of differentiation was found in the organization of transcortical sensomotor projections. The pyramidal, stellate, and inverted pyramidal neurons in the third layer of the cortex were shown to take part in the formation of these pathways. Results obtained by the experimental degeneration method combined with electron microscopy showed that afferentation from the first somatosensory area of the cortex reaches mainly cells in layers III and V. Responses of caudate neurons to stimulation of the anterior sigmoid and various parts of the suprasylvian gyrus were studied in acute experiments on cats. The experiments consisted of two series: on animals with an intact thalamus and on animals after preliminary destruction of the nonspecific thalamic nuclei. Stimulation of all cortical areas tested in intact animals evoked complex multicomponent responses in caudate neurons with (or without) initial excitation, followed by a phase of inhibition and late activation. The latent periods of the initial responses to stimulation of all parts of the cortex were long and averaged 14.5–25.5 msec. Quantitative and qualitative differences were established in responses of the caudate neurons to stimulation of different parts of the cortex. Considerable convergence of cortical influences on neurons of the caudate nucleus was found. After destruction of the nonspecific thalamic nuclei all components of the complex response of the caudate neurons to cortical stimulation were preserved, and only the time course of late activation was modified.A. The effect of auditory cortex blockade on response patterns of parietal association cortex neurons responding to different frequency tones was investigated in the cat. Blockade was produced by two methods: bilateral isolation and application of a 6% Nembutal solution to the auditory cortex surface. Frequency threshold curves were plotted for all test neurons. The majority of test neurons (84%) displayed one or two characteristic frequencies before blockade, as against only 63% of all neurons responding following blockade. Changes also affect the range of frequencies at which the cells could respond. Virtually all test neurons responded to application of a broad spectrum of frequencies under normal conditions. After blockade of the auditory cortex 69% of neurons no longer responded to tones above 8–10 kHz. This would suggest that mainly information on high frequency tones is transmitted via the auditory cortex. The question of where acoustic information for parietal association cortex neurons mostly originates is also discussed; association thalamic nuclei are thought to be the main source.A. A. Activity of 112 neurons of the precruciate motor cortex in cats was studied during a forelimb placing reaction to tactile stimulation of its distal parts. The latent period of response of the limb to tactile stimulation was: for flexors of the elbow (biceps brachii) 30–40 msec, for the earliest reponses of cortical motor neurons about 20 msec. The biceps response was observed 5–10 msec after the end of stimulation of the cortex with a series of pulses lasting 25 msec. Two types of excitatory responses of the neurons were identified: responses of sensory type observed to each tactile stimulation of the limb and independent of the presence or absence of motion, and responses of motor type, which developed parallel with the motor response of the limb and were not observed in the absence of motion. The minimal latent period of the responses of motor type was equal to the latent period of the sensory responses to tactile stimulation (20±10 msec). Stimulation of the cortex through the recording microelectrode at the site of derivation of unit activity, which increased during active flexion of the forelimb at the elbow (11 stimuli at intervals of 2.5 msec, current not exceeding 25 µA), in 70% of cases evoked an electrical response in the flexor muscle of the elbow.M. In acute experiments on cats evoked potentials (EP) of the orbital cortex were recorded and the electrogenesis and functional purpose of individual components of associative responses (AR) were investigated. It was concluded that the initial negative fluctuation of the AR arises as a consequence of the physical propagation of potentials from the projection somatosensory cortex and the second, positive, component and the following negative component are the result of arrival of an afferent volley into the orbital cortex via specific thalamic nuclei. These two components are due to activation of neurons of the orbital cortex. The afterdischarge, which appears sometimes, develops under the effect of impulses arriving from nonspecific thalamic nuclei. It is shown that during the second, positive, phase of the AR, primarily afferent neurons are activated, and during the negative phase, efferent neurons of the orbital cortex. A comparative study of neuronal response in separate cortical columns of the somatosensory cortex (the barrel field area) was made in unanesthetized partially curarized white rats under various circumstances: during passive deflection of immobile vibrissa, unhindered volitional sweeping movement of the vibrissae, and during movement induced by stimulating the motor cortex and facial muscles. Differences in the response of the same neurons emerged under these different experimental situations. Different groups of neurons — responding before, during, and after volitional vibrissa movements were observed. To study the possible pathways along which visual signals reach a visual zone discovered by the writers in the cat frontal cortex in the ventral bank of the cruciate sulcus, horseradish peroxidase was injected into a site previously identified by physiological experiments. Stained neurons were discovered in visual areas of the cortex (lateral suprasylvian and ectosylvian), in the parietal cortex (areas 5 and 7), and also in small numbers in the prefrontal and limbic cortex. Stained neurons were found in the following nuclei in the thalamus: n. medialis dorsalis, intralaminar nuclei (nn. centralis lateralis, paracentralis, centralis medialis), nn. ventralis medialis, anteromedialis, and reuniens. Many stained neurons were found in the claustrum and a few in the substantia grisea centralis in the midbrain. The responses of motor cortex neurons in the cat to the presentation of a single auditory click and a series of 10 clicks presented with 1,000/sec frequency were studied under conditions of chronic experiments before and after the development of an instrumental food reflex. After reflex development a single presentation of a positive conditioned stimulus (single click) markedly influenced for 7 sec the appearance of instrumental movements. At the same time, the immediate responses of motor cortex neurons to presentation of the conditioned auditory stimulus had no impact on the appearance in the motor cortex of discharges leading to the realization of instrumental movements. Consequently, motor cortex neurons do not require activation from afferent sensory inputs for the generation of such discharges. The immediate neuronal responses to conditioned stimulation did not inhibit the realization of the instrumental reflex. It is proposed that they are associated with the realization of motor function in the unconditioned defensive response evoked by the presentation of an auditory stimulus. The presence or absence of responses to auditory conditioned stimulation was dependent upon the signal meaning of the stimulus, its physical parameters, and the degree of excitability of the animal.A. Extracellular recording techniques were used on cats anesthetized with Nembutal to illustrate antidromic activation of pontine neurons produced by stimulating the medial and occasionally the superior cerebellar peduncle, the cerebellar central nuclei, pyramidal tract, and sensorimotor region of the cortex. Of the pontine nucleus projection, that extending to the lateral cerebellar nucleus was the most clearly defined. Stimulation of the pyramidal tract, central cerebellar nuclei and the superior cerebellar peduncle was found to produce monosynaptic excitation of pontine neurons. Retrogradely labeled thalamic neurons—the sources of afferents in the focus of peak activity induced by radial nerve stimulation—were investigated in adult cats by means of microiontophoretic horseradish peroxidase injection into the first somatosensory zone of the cerebral cortex. Labeled cells were found mainly in the ventroposterolateral and a smaller proportion in the posteroventral medial thalamic nuclei.
