Fine structural details of the parasitic yeastlike phase of Sporothrix schenckii contained in biopsy tissue from a naturally-occurring case of disseminated feline sporotrichosis are described and illustrated by electron microscopy. Both free and phagocytosed fungal cells were observed. The fungal cells were contained within an extracellular, electron transparent vacuolar area which was bounded by a limiting membrane of probable host origin. The yeastlike cells were characterized by a conspicuous layer of osmiophilic microfilaments which occurred along the outermost surface of the cell wall. In many yeastlike cells, scattered, membranebound vacuoles containing electron opaque material were observed in the cytoplasm. Asteroid bodies were not observed. 相似文献

Most cell junctions of Sycon are of the simple kind in which the plasma membranes are separated by an intercellular space of about 15 mmu. True septate junctions are found between sclerocytes. They are similar to those of coelenterates and echinoderms, with a parallel arrangement of septa as seen in tangential sections. The functional implications of the restriction of this type of junction to the sclerocytes are discussed. 相似文献

Summary In the epidermis of turbellarians septate junctions of the pleated sheet type have been demonstrated in conventional thin sections and freeze fractured preparations. Septate junctions have been studied in the hind-gut of Periplaneta americana, Incisitermes schwartzi and Thermobia domestica. The topographical disposition of intercellular septa revealed by lanthanum impregnation corresponds well with that of particle rows seen in freeze fracture preparations. However, there is no precise correspondence between the undulations of septa and the disposition of particles within a single row. The spacing of particles is variable and generally less than that of the undulation periodicity of septa. On the other hand, the disposition of septa, and of the rows of particles that correspond to them, appears to be variable: sometimes rectilinear and in close parallel, these may curve or form series of 'finger-print' loops. Moreover, the septa are evidently not deployed as continuous ribbons around the cell since intrruptions are freuqently observed. In addition to their mechanical role in intercellular cohesion, septate junctions apparently ensure a more or less complete closure of the intercellular space (i.e. provide a permeability barrier) and thus play a role comparable with that of tight junctions in epithelia of vertebrates. 相似文献

Summary Tight junctions were found in the apical junctional complex of the adult rabbit subcommissural organ (SCO) in addition to zonulae adhaerentes and gap junctions of typical ependymal cells. Ventricular perfusion of ruthenium red before fixation was found to give excellent results for distinguishing between gap and tight junctions at the ependymal surface. The implication of tight junctions as a mechanical means of sealing off the SCO area from the cerebrospinal fluid and the use of ruthenium red as a tracer substance are discussed.This work was supported by grants from Statens almindelige Videnskabsfond, Copenhagen. 相似文献

Summary The cell coat picture effect which is usually obtained with the conventional RR method, that is with the RR/OsO4 coupled reaction, is investigated. In this first paper, each of conceivable events which might take place between RR, OsO4 and cell surface membrane is discussed or studied. Various tests are carried out on ascites Ehrlich carcinoma cells and Zajdela ascites hepatoma cells treated with numerous chemical reagents, as also on a few pure proteins. The set of data supports the concept that the staining pattern is due to the combination in surface membranes of RR with a colloidal-like form of OsO2. The latter might occur during the formation of stable cyclic osmic acid diesters between OsO4 and membrane unsaturated lipids. A possibility by which the resulting marker is though also to be in a colloidal-like state is put forward. A next report will deal with this problem. 相似文献

Fine structurally, Olisthodiscus luteus is characterised by possessing a sub-surface layer of electron opaque ‘spheres’ approximately 35 nm in diameter. These ‘spheres’ originate in vesicles surrounding the Golgi apparatus. The flagella are of the heterokont type and are attached, at their bases, by a large complicated root to the nucleus. The mitochondria, besides containing microvilli, contain fine fibrils of material that can be removed by treatment with DNase. The phyletic affinities of Olisthodiscus are discussed with reference to this fine structural study and recent biochemical work. Although no conclusive evidence is available, it is suggested that Olisthodiscus should be transferred from the Xanthophyceae temporarily to the Chrysophyceae. 相似文献

Summary The electron-dense marker which is thought to produce the ruthenium red surface staining is studied. This stain is prepared under conditions which should give its rise in cell surface membrane, and its nature and charge are tested electrophoretically and by measuring the turbidity, respectively. It is a positive colloid resulting from the recharging of colloidal osmium dioxide by RR polycations. Controls on the affinity are carried out by applying positive sol to gelled agarose sections containing hyaluronic acid, polyvinyl sulfate or polylysine. Controls are also carried out on ascites Ehrlich carcinoma and Zajdela ascites hepatoma cells subjected to prior enzymatic and chemical treatments. It is found that the osmium-RR system visualizes all acidic groups in the outer hydrophilic leaflet, that is the greater part of compounds in this external cell layer. A model is presented for the mechanism underlying its rise in cell surface membrane. 相似文献

The interaction sites of ruthenium red (RR) on the Helix aspersa renal epithelium are described. RR staining was located under septate junctions, at lamina densa of the basement membrane and in formations similar to intracytoplasmic vesicles and canaliculi. It has recently been reported that losses of tight junction material could result from the freeze-fracture process. To verify this assumption, we tried to increase the possibility, if any, of losses of junction material, by inducing an important fragmentation of junctional fibrils by bathing ciliary epithelium in a 0.5M sucrose solution before glutaraldehyde fixation and freeze-fracturing at −160 °C. In spite of a significant redistribution of junctional material on both fracture faces, careful examination of complementary replicas and measurements of junction elements and interruption lengths showed that no loss of junctional material occurred in this tissue. The influence of physical parameters (i.e. temperature) on the preservation of the structural integrity of the tight junction during fracturing is now a problem to be considered. 相似文献

The tight junctions of the choroid plexus epithelium of rats were studied by freeze-fracture. In glutaraldehyde-fixed material, the junctions exhibited rows of aligned particles and short bars on P-faces, the E-faces showing grooves bearing relatively many particles. A particulate nature of the junctional strands could be established by using unfixed material. The mean values of junctional strands from the lateral, third, and fourth ventricles of Lewis rats were 7.5 +/- 2.6, 7.4 +/- 2.2, and 7.5 +/- 2.4; and of Sprague-Dawley rats 7.7 +/- 3.4, 7.4 +/- 2.3, and 7.3 +/- 1.6. Examination of complementary replicas (of fixed tissue) showed that discomtinuities are present in the junctional strands: 42.2 +/- 4.6% of the length of measured P-face ridges were discontinuities, and the total amount of complementary particles in E-face grooves constituted 17.8 +/- 4.4% of the total length of the grooves, thus approximately 25% of the junctional strands can be considered to be discontinuous. The average width of the discontinuities, when corrected for complementary particles in E-face grooves, was 7.7 +/- 4.5 nm. In control experiments with a "tighter" tight junction (small intestine), complementary replicas revealed that the junctional fibrils are rather continuous and that the very few particles in E-face grooves mostly filled out discontinuities in the P-face ridges. Approximately 5% of the strands were found to be discontinuous. These data support the notion that the presence of pores in the junctional strands of the choroid plexus epithelium may explain the high transepithelial conductance in a "leaky" epithelium having a high number of junctional strands. However, loss of junctional material during fracturing is also considered as an alternative explanation of the present results. 相似文献

Microwave fixed liver and kidney tissues were examined by electron microscopy. It was found that the preservation of fine structure of these tissues by this method is equal to that processed by routine methods. No difficulty was encountered in sectioning microwave fixed tissue blocks. It is obvious that microwave fixation is a faster and more efficient method. 相似文献

In sea urchin embryos, blastula formation occurs between the seventh and tenth cleavage and is associated with changes in the permeability properties of the epithelium although the structures responsible for mediating these changes are not known. Tight junctions regulate the barrier to paracellular permeability in chordate epithelia; however, the sea urchin blastula epithelium lacks tight junctions and instead possesses septate junctions. Septate junctions are unique to non-chordate invertebrate cell layers and have a characteristic ladder-like appearance whereby adjacent cells are connected by septa. To determine the function of septate junctions in sea urchin embryos, the permeability characteristics of the embryonic sea urchin epithelia were assessed. First, the developmental stage at which a barrier to paracellular permeability arises was examined and found to be in place after the eighth cleavage division. The mature blastula epithelium is impermeable to macromolecules; however, brief depletion of divalent cations renders the epithelium permeable. The ability of the blastula epithelium to recover from depletion of divalent cations and re-establish a barrier to paracellular permeability using fluorescently labelled lectins was also examined. Finally, septate junction structure was examined in embryos in which the permeability status of the epithelium was known. The results provide evidence that septate junctions mediate the barrier to paracellular permeability in sea urchin embryos. 相似文献

This report is a comprehensive fine structural analysis of the morphological changes occurring during metamorphosis of the marine hydrozoan Mitrocomella polydiademata. Five stages are recognized during metamorphosis: planulae just prior to settlement, ball and filiform stages, immature polyps, and primary feeding polyps. We examined morphologically the process of regeneration before and after fission in a sexual strain of the freshwater planarian
Dugesia japonica. Usually fission takes place in the post-pharyngeal region. Decapitation significantly accelerates the rate
of fissioning. When decapitated worms were treated with substance P and neuropeptide K separately, the rate of fission markedly
decreased in both cases. Before the onset of fission, a presumptive region of fission was recognized in the post-pharyngeal
portion where undifferentiated cells, regenerative cells and newly differentiated cells were localized. Moreover a functional
network of fixed parenchyma cells was noted in this region. After fission, cell distribution in the blastema became quite
different from that of artificially amputated worms. This difference seems to be due to the process that occurs in the presumptive
region of fission.

Fluorescent proteins homologous to green fluorescent protein (avGFP) display pronounced spectral variability due to different chromophore structures and variable chromophore interactions with the surrounding amino acids. To gain insight into the structural basis for yellow emission, the 3D structure of phiYFP (λem = 537 nm), a protein from the sea medusa Phialidium sp., was built by a combined homology modeling – mass spectrometry approach. Mass spectrometry of the isolated chromophore-bearing peptide reveals that the chromophore of phiYFP is chemically identical to that of avGFP (λem = 508 nm). The experimentally acquired chromophore structure was combined with the homology-based model of phiYFP, and the proposed 3D structure was used as a starting point for identification of the structural features responsible for yellow fluorescence. Mutagenesis of residues in the local chromophore environment of phiYFP suggests that multiple factors cooperate to establish the longest-wavelength emission maximum among fluorescent proteins with an unmodified GFP-like chromophore. 相似文献

Synopsis Based on the results of investigations into the fine structure of red and white muscle of roach, Rutilus rutilus, and chub, Leuciscus cephalus (Sänger et al. 1990), analysis of axial muscle has been extended to include nine other cyprinid species. Quantitative comparisons were made of myofibrils, mitochondria, lipid and sub-sarcolemmal cytoplasm in relation to muscle fibre type and species. A comparison of these variables between the species shows that for red fibres there are significant differences in all measured variables in at least some cases. The most striking difference is in lipid content, with Danube bleak, Chalcalburnus chalcoides mento, having the highest amount. For white fibres, there were significant differences only in intermyofibrillar mitochondria and myofibrils, which were most significant in Danube bleak. As a preliminary study, intermediate, fibres have been examined qualitatively and compared with the other two fibre types using the above mentioned variables. The results allow cyprinids to be arranged according to their scores from the different measurements. These groupings are discussed in relation to the different life-styles of the fish. 相似文献

Summary The intercellular connections between the epithelial cells of Bowman's capsule were investigated. It could be demonstrated that typical zonulae occludentes (tight junctions) are present in the species (rat, hamster, and Tupaia) studied. Freeze-fracturing shows a network of anastomizing strands; some species variations are described. In the rat two strands are common. In the golden hamster mostly two to four and occasionally five strands occur. In Tupaia regularly three tight junction strands are found and also gap junctions associated with the zonulae occludentes. In thin sections the goniometric analysis confirms the freeze-fracturing results and reveals attachment zones of macular shape, which are classified as intermediate junctions and desmosomes. The functional role of these cell junctions observed in the epithelium of Bowman's capsule is discussed. 相似文献
