Summary Plants were regenerated from cotyledon tissue of greenhouse grown seedlings of common buckwheat (Fagopyrum esculentum Moench.). Maximum callus regeneration was induced on Murashige and Skoog (MS) medium containing 2,4-D (2.0 mg l−1) and kinetin (KIN) (0.2 mg l−1) and either 3 or 6% sucrose. Friable callus was transferred to MS media containing KIN and benzylaminopurine (BAP) at varied concentrations for embryogenic callus induction. The optimum medium for embryogenic callus induction was found to be MS medium supplemented with 0.2 mg l−1 KIN, 2.0 mg l−1 BAP and 3% (w/v) sucrose. Variation of sucrose from 3 to 6% did not show any significant effect on callus induction or embryogenesis. Regeneration of embryonic callus varied from 13 to 32%. Whole plants were obtained at high frequencies when the embryogenic calluses with somatic embryos and organized shoot primordia were transferred to half-strength MS media with 3% sucrose. Regenerated plants after acclimation were transferred to greenhouse conditions, and both vegetative and floral characteristics were observed for variation. This regeneration system may be valuable for genetic transformation and cell selection in common buckwheat.  相似文献

Anther culture for haploid induction of buckwheat was studied over a period of five years. Approximately 24,000 anthers were isolated and cultured on different culture media. The regeneration capacity was generally very low. Data are presented for experiments that included 7278 anthers on which 99 calluses were formed and 20 buds regenerations were noted. Regeneration occurred most readily on gellan-gum solidified media, with 90 g l-1 maltose, 2.5 mg l-1 BA, 0.5 mg l-1 IAA, and preferably in darkness. Haploid cells, as established by chromosome counts, were observed in eight regenerants. Several abnormalities of pollen development in vitro were detected. Starch presence in pollen as a possible sign of androgenic capacity was studied. Genetic transformation of buckwheat (Fagopyrum esculentum Moench.) and regeneration of transgenic plants were obtained by using Agrobacterium tumefaciens strains as vectors. Buckwheat cotyledons were excised from imbibed seeds, co-cultivated with A. tumefaciens and subjected to previously reported protocols for callus and shoot regeneration. The transformation with oncogenic strains was confirmed by opine and DNA analyses of tumour tissue extracts. Plants were regenerated on cotyledon fragments incubated with strain A281, harboring pGA472, which carries the neomycin phosphotransferase II gene for kanamycin resistance. The transformation of resistant shoot clones was confirmed by NPTII enzyme assay and DNA hybridization. A large number of transformed shoots were rooted and fertile plantlets were raised in the greenhouse. Transgenic plants comprised pin and thrum clones, which were allowed to cross-pollinate. Common buckwheat grown in Pb-contaminated soil was found to accumulate a large amount of Pb in its leaves (8,000 mg/kg DW), stem (2,000 mg/kg DW), and roots (3,300 mg/kg DW), without significant damage. This indicates that buckwheat is a newly recognized Pb hyperaccumulator, which is defined as a plant containing over 1,000 mg/kg of Pb in its shoots on a dry-weight basis. Moreover, it was shown that application of the biodegradable chelator methylglycinediacetic acid trisodium salt at concentrations of up to 20 mmol/kg resulted in a more than five times higher concentration of Pb in the shoot without notable growth inhibitation at up to 10 mmol/kg. These results indicate that buckwheat is a potential phytoremediator of Pb-contaminated soils.  相似文献

A compound that inhibited angiotensin-I converting enzyme (ACE) activity was isolated from buckwheat powder. This compound is thought to be the hydroxy derivative of nicotianamine and its chemical structure is 2'-hydroxynicotianamine. This compound showed a very high inhibitory activity toward ACE, and the IC(50) was 0.08 microM. Only this hydroxy analog was found in buckwheat powder, at about 30 mg/100g, and no nicotianamine was detected. However, nicotianamine was detected in the buckwheat plant body. 2'-hydroxynicotianamine was also found in other polygonaceous plants.  相似文献

A flavonol-3-O-beta-heterodisaccharide glycosidase (FHG I) was isolated from dried aerial tissues of Fagopyrum esculentum Moench (Fagopyri herba). It has a specific enzyme activity of ca. 3.5 nkat mg(-1) protein in buffered extracts when rutin (quercetin-3-O-rutinoside) was used as substrate and an optimal enzyme activity was seen at around pH 4.8 and 30 degrees C. FHG I was purified about 156-fold to apparent homogeneity by hydrophobic interaction, anion exchange and size exclusion chromatographic steps. The apparent molecular mass of FHG I was 74.5+/-2 kDa as determined by SDS-PAGE and it is a monomeric glycoprotein with a carbohydrate content of 23%. The isoelectric point as determined by isoelectric focusing was 5.7 and the energy of activation was 32 kJ mol(-1). FHG I exhibits a high substrate specificity, preferring flavonol 3-O-glycosides comprising the disaccharide rutinose. The K(m) and V(max) values for the natural substrate rutin were calculated to be 0.561 microM and 745 nkat mg (-1) protein, respectively. Two oligopeptide fragments obtained after enzymatic digestion of FHG I were sequenced and showed similarities to sequences of beta-glucohydrolases from other plant species. Polyclonal antibodies were raised and their specificities determined. Another flavonol 3-O-beta-heterodisaccharide glycosidase (FHG II) could also be detected in buckwheat herb, having a molecular mass of 85.3+/-2 kDa and an isoelectric point between pH 6.0 and 6.5.  相似文献

荞麦无菌苗下胚轴切段在不同激素配比的MS培养基上诱导愈伤组织,出愈率均为100%。在2.0mg/L2,4-D和1.5mg/L 6-BA组合下诱导产生的愈伤组织;转入2.0mg/L 6-BA和1.0mg/L KT的MS培养基,再生苗分化率在80%以上。根尖色体分析表明再生植株具一定的遗传稳定性。发根农杆菌A4转化荞麦下胚轴和子叶获得发状根,纸电泳检测所有随机取样测定的发状根均有相应冠瘿碱的存在。  相似文献

Agriculture productivity is severely affected by soil salinity. One possible mechanism by which plants could survive salt stress is to compartmentalize sodium ions away from the cytosol. In the present work, transgenic buckwheat plants overexpressing AtNHX1, a vacuolar Na+/H+ antiporter gene from Arabidopsis thaliana, were regenerated after transformation with Agrobacterium tumefaciens. These plants were able to grow, flower and accumulate more rutin in the presence of 200 mmol/l sodium chloride. Moreover, the content of important nutrients in buckwheat was not affected by the high salinity of the soil. These results demonstrated the potential value of these transgenic plants for agriculture use in saline soil.  相似文献

The effects of supplemental UV-B radiation on crop growth, morphology, reproduction and physiology were studied in three cultivars of Fagopyrum esculentum Moench (buckwheat) originating from high elevation (Qinghai-Tibet plateau) and lower altitudes (The Sichuan Basin). Our results showed that common buckwheat was sensitive to UV-B stress. Plant growth, development, and reproduction were inhibited by elevated UV-B radiation. Plant lipid oxidation and polyphenol oxidase (PPO) activity increased with increasing UV-B radiation, along with the concentration of phenylpropanoid compounds, superoxide dismutase (SOD) activity and ascorbic acid (Asa) concentration were also enhanced at the lowest level of supplemental UV-B radiation but decreased at the higher level of enhanced UV-B. While, a cultivar originating from elevated locations had lower dry matter accumulation and was more tolerant to UV-B radiation than cultivars originating from lower elevations. The effects on leaf thickness and increased antioxidant capacity could be linked with the improved performance of cultivar originating from high elevation when exposed to enhanced UV-B radiation. We conclude that UV-B tolerance should be considered prior to introducing or breeding common buckwheat cultivars from lowland cultivation to regions at high elevation such as the Qinghai-Tibet plateau.  相似文献

Novel antimicrobial peptides (AMP), designated Fa-AMP1 and Fa-AMP2, were purified from the seeds of buckwheat (Fagopyrum esculentum Moench.) by gel filtration on Sephadex G75, ion-exchange HPLC on SP COSMOGEL, and reverse-phase HPLC. They were basic peptides having isoelectric points of over 10. Fa-AMP1 and Fa-AMP2 had molecular masses of 3,879 Da and 3,906 Da on MALDI-TOF MS analysis, and their extinction coefficients in 1% aqueous solutions at 280 nm were 42.8 and 38.9, respectively. Half of all amino acid residues of Fa-AMP1 and Fa-AMP2 were cysteine and glycine, and they had continuous sequences of cysteine and glycine. The concentrations of peptides required for 50% inhibition (IC50) of the growth of plant pathogenic fungi, and Gram-positive and -negative bacteria were 11 to 36 μg/ml. The structural and antimicrobial characteristics of Fa-AMPs indicated that they are a novel type of antimicrobial peptides belonging to a plant defensin family.  相似文献

The paper describes the amino acid sequence of a 26 kDa basic subunit of 13S globulin of common buckwheat (Fagopyrum esculentum Moench). The protein has 93 and 75% sequence homology with 11S globulin of Coffea arabica and beta subunit of 11S globulin of Cucurbita pepo respectively. The subunit has the "globally conserved" N-terminal sequence consisting of Gly-Ile-Asp-Glu and the cysteine at P7' from the proteolytic processing site. A conserved 7 residue domain of Pro-His-Trp-Asn-Ile-Asn-Ala, characteristic of basic subunits of legumins from non-leguminous angiosperms, is also present in this protein. A distinguishing features of this subunit is the relatively high level of lysine and methionine.  相似文献

Summary Absorption of nitrate and ammonium was studied in water culture experiments with 4 to 6 weeks old plants of barley (Hordeum vulgare L.), buckwheat (Fagopyrum esculentum L. Moench) and rape (Brassica napus L.). The plants were grown in a complete nutrient solution with nitrate (5.7±0.2 mM) or nitrate (5.6±0.2 mM) + ammonium (0.04±0.02 mM). The pH of the nutrient solution was kept at 5.0 using a pH-stat. It was found that phosphorus deficiency reduced the rate of nitrate uptake by 58±3% when nitrate was the sole N source and by 83±1% when both nitrate and ammonium were present. The reduction occurred even before growth was significantly impeded by P deficiency. The inhibition of the uptake of ammonium was less,i.e. ammonium constituted 10±1% of the total N uptake in the P sufficient plants and 30±5% in the P deficient plants. The reduction of nitrate absorption greatly decreased the difference between the uptake of anions and cations. It is suggested that P deficiency reduced the assimilation of NO 3 into the proteins, which might cause a negative feedback on NO 3 influx and/or stimulate NO 3 efflux.  相似文献

Among pale-green tomato plants heterozygous for the xanthophyllic2 (xa-2) mutation that were transformed with a T-DNA harbouring the NPTII and GUS gene, a plant with a high frequency of green/white twin spots was found. The genetic analysis of this plant indicated that the occurrence of these twin spots was caused by a genetic defect located at the distal end of chromosome 10S, where xa-2 also is located. The genetic analysis of green plants regenerated from leaf expiants of this twin-spot plant revealed that the green sectors derive from non-disjunction of the xa-2 + allele. In an analysis of mitotic chromosome behaviour bridges were observed in approximately 5% of the anaphases, providing arguments that a breakage-fusion-bridge cycle caused by a tissue culture-induced genomic instability is the most likely cause of this aberrant behaviour of chromosome 10.  相似文献

A pot experiment was carried out in a growth chamber to investigate P efficiencies and mycorrhizal responsiveness of modern (Krichauff and Excalibur) and old (Khapstein, Bobin, Comeback and Purple Straw) wheat cultivars (Triticum aestivum). The arbuscular mycorrhizal fungus (AMF) used in this study was Glomus intraradices. The growth medium was a soil/sand mixture with NaHCO3-extractable P of 9.4 mg P kg–1 and no extra P was added. Plant P efficiencies (uptake, utilisation and agronomic) were found to differ significantly between cultivars, but no general trends of changes with the year of release of the cultivar were found. AMF colonisation was found to decrease plant growth under our experimental conditions with low light intensity. Mycorrhizal responsiveness (MR) was measured in terms of the improvement in plant P nutrition (shoot P concentrations). MR was found to be generally lower in modern cultivars than in old cultivars, indicating that modern breeding programs may have reduced the responsiveness of modern wheat cultivars to arbuscular mycorrhizal fungi. MR was also found to decrease in general with increased plant P utilisation efficiency.  相似文献

Results are reported for tomato (Lycopersicon esculentum L. var. Ailsa craig) and wheat (Triticum aestivum L. cv. Mara) which demonstrate that increasing concentrations of Mg in the plant raises plant tolerance to Mn toxicity.Water culture experiments with tomato show that under conditions of high Mn supply (200 µM, Mn), not only does increasing Mg application (0.75 mM to 15 mM) depress Mn uptake, but the higher Mg concentrations in the shoot counteract the onset of Mn toxicity when the concentrations of Mn in the shoot are also high. The ratio of Mg: Mn in the tissues is a better indicator of the appearance of toxicity symptoms than Mn concentration alone. Toxicity symptoms were observed when the Mg:Mn ratio in the shoot tissue was from 1.13 to a value between 3.53 and 6.54. The corresponding Mg: Mn ratio in the older leaves was from 0.82 to between 2.27 and 3.51.For wheat grown in soil, analyses of leaves revealed that growth could be expressed by the following relationship: Y=A+B exp(-kX), where Y=growth, X=Mg:Mn ratio, A, B and k=constants. Growth was significantly reduced when the Mg:Mn ratio fell below 20:1. From a measurement of this ratio it is therefore possible to predict the appearance of Mn toxicity and its influence on growth.  相似文献

The Lizada and Yang method, commonly used for analyzing 1-aminocyclopropane-1-carboxylic acid (ACC), the immediate precursor of the plant hormone ethylene, is subject to interference and lacks internal standards. The use of combined gas chromatography-mass spectrometry (GC-MS) overcomes these shortcomings but the method is expensive and unavailable to many laboratories. We describe an alternative physico-chemical method using a capillary column gas chromatograph fitted with a standard nitrogen/phosphorus detector. After forming the N-benzoyl n-propyl derivative, measurements of ACC concentrations in extracts of leaves and in xylem sap of tomato plants using the nitrogen/phosphorus detector were within 10% of those obtained by GC-MS. Concentrations in plants grown in well-drained soil were approximately 0.16 nmol g–1 fresh weight (leaves) and 0.04–0.01 mmol m–3 (sap). Flooding the soil for 48–72 h increased these values approximately 9-fold.  相似文献

With the aim of improving the safety and nutritional quality of traditional African weaning porridge, the reduction of the viscosity of a high solids fermented pearl millet porridge by addition of sorghum malt (amylase rich flour, ARF) was investigated. The effect of fermentation, cooking, malt addition and recooking on the microflora of, and the survival of an inoculated pathogen were determined. Addition of 5% (w/v) sorghum ARF to the gelatinized millet porridge gave an acceptable viscosity of 2500–3000 cP at a high solid content of 30%. Fermentation inhibited the growth of microorganisms in the porridge and recooking the fermented porridge after sorghum ARF addition further eliminated (<102 c.f.u./g) the moulds and coliforms that were introduced with the sorghum ARF. The recooked, fermented millet plus sorghum ARF porridge prevented the proliferation of the inoculated Escherichia coli and reduced it to <102 c.f.u./g within 18 h. The porridge could supply children under 3 years with the daily required protein using 1.4 feedings per day and required energy with 4 feedings a day.  相似文献
