高粱种质材料幼苗期耐盐碱性评价   总被引:7,自引:0,他引:7  
采用Hoagland营养液砂培法,以NaCl和Na2CO3组成的混合盐碱对高粱幼苗进行胁迫处理,建立高粱幼苗期耐盐碱评价方法,并评价了66份高粱种质材料的耐盐碱性.结果表明:盐浓度在8.0~12.5 g·L-1时,高粱耐盐碱品种‘TS-185’与盐碱敏感品种‘Tx-622B’在幼苗期的耐盐碱性差异明显,表明进行高粱幼苗期耐盐碱性评价时适宜的盐浓度范围为8.0~12.5 g· L-1.在10.0和12.5 g·L-12个盐浓度下,66份高粱种质材料的相对存活率、相对地上部鲜质量和相对株高的差异均达显著水平,表明不同品种的耐盐碱性不同.其中,‘三尺三’为高度耐盐碱品种,‘MN-2735’等16个品种为耐盐碱品种,‘EARLY HONEY’等32个品种为中等耐盐碱品种,‘Tx-622B’等16个品种为盐碱敏感品种,‘MN-4588’为高度盐碱敏感品种.苏丹草类型高粱一般具有较高的耐盐碱性,而保持系对盐碱较为敏感.  相似文献

Tolerance to Zn deficiency in wheat germplasm may be inversely related to uptake and transport of Fe to shoots. The present study examined eight bread (Triticum aestivum) and two durum (T. turgidum L. conv. durum) wheat genotypes for their capacity to take up and transport Fe when grown under either Fe or Zn deficiency. Bread wheat genotypes Aroona, Excalibur and Stilleto showed tolerance to Zn and Fe deficiency, while durum wheat genotypes are clearly less tolerant to either deficiency. Roots of bread wheats tolerant to Zn deficiency exuded more phytosiderophores than sensitive bread and durum genotypes. Greater amounts of phytosideophores were exuded by roots grown under Fe than Zn deficiency. A relatively poor relationship existed between phytosiderophore exudation or the Fe uptake rate and relative shoot growth under Fe deficiency. At advanced stages of Zn deficiency, genotypes tolerant to Zn deficiency (Aroona and Stilleto) had a greater rate of Fe uptake than other genotypes. Zinc deficiency depressed the rate of Fe transport to shoots in all genotypes in early stages, while advanced Zn deficiency had the opposite effect. Compared with Zn-sufficient plants, 17-day-old Zn-deficient plants of genotypes tolerant to Zn deficiency had a lower rate of Fe transport to shoots, while genotypes sensitive to Zn deficiency (Durati, Yallaroi) had the Fe transport rate increased by Zn deficiency. A proportion of total amount of Fe taken up that was transported to shoots increased with duration of either Fe or Zn deficiency. It is concluded that greater tolerance to Zn deficiency among wheat genotypes is associated with the increased exudation of phytosiderophores, an increased Fe uptake rate and decreased transport of Fe to shoots. Sorghum [Sorghum bicolor (L.) Moench] is the fifth most important cereal crop of the world. In South America, it is grown mainly on acid soils, and its production on these soils is limited by deficient levels of available P, Ca, Mg, and micronutrients, and toxic levels of Al and Mn. A greenhouse experiment was undertaken to evaluate the genotypic differences in sorghum for uptake (U), inhibition (IH), influx (IN) into roots, and transport (TR) to shoot for nutrients at three levels of soil Al saturation (2, 41, 64%). Overall shoot nutrient U, IN, and TR showed a significant inverse correlation with soil Al saturation and shoot Al concentration, and a significant positive correlation with shoot and root dry weight. The nutrient uptake parameters differentiated genotypes into most and least efficient categories at various levels of soil Al saturation. The nutrient uptake parameters showed significant differences with respect to soil Al saturation, genotypes, and their interactions. In the current study, Al tolerant genotypes recorded higher IN and TR for P, K, Ca, Mg, Zn, and Fe than Al-sensitive genotypes. Therefore, these U, IN, and TR traits could be used in selection of sorghum plants adaptable to acid soils. Sorghum genotypes used in this study showed intraspecific genetic diversity in U, IN, and TR for essential nutrients. This study was undertaken to investigate oxidative stress tolerant mechanisms in chilli (Capsicum annuum L.) under drought genotypes through evaluating morphological, physiological, biochemical and stomatal parameters. Twenty genotypes were evaluated for their genetic potential to drought stress tolerant at seedling stage. Thirty days old seedlings were exposed to drought stress induced by stop watering for the following 10 days and rewatering for the following one week as recovery. Based on their survival performance, two tolerant genotypes viz. BD-10906 and BD-109012 and two susceptible genotypes viz. BD-10902 and RT-20 were selected for studying the oxidative stress tolerance mechanism. Drought reduced root and shoot length, dry weight, ratio, petiole weight and leaf area in both tolerant and susceptible genotypes, and a higher reduction was observed in susceptible genotypes. Lower reduction of leaf area and photosynthetic pigments were also found in tolerant genotypes. Moreover, tolerant genotypes showed higher recovery than susceptible genotypes after the removal of stress. A higher reduction of relative water content (RWC) may cause an imbalance between absorbed and transpirated water in susceptible genotypes. Higher accumulation of proline in tolerant genotypes might be helpful to for better osmotic maintenance than that in susceptible genotypes. Tolerant genotypes showed higher antioxidant activity as they showed DPPH radical scavenging percentage than the susceptible genotypes. Moreover, closer stomata in tolerant genotypes than susceptible ones helped to avoid dehydration in tolerant genotypes. Thus, the above morphological, physiological, biochemical and stomatal parameters helped to show better tolerance in chilli under drought stress.  相似文献

Pathogenic isolates of Periconia circinata produce a host-specific toxin (PC-toxin) and cause a root and crown rot in susceptible genotypes of sorghum. Treatment with PC-toxin leads to selective development of disease symptoms and an increase in synthesis of a group of acidic, low molecular weight proteins only in susceptible genotypes. Treatment of sorghum seedlings or excised root tips with HgCl2 resulted in responses indistinguishable from those produced by treatment with PC-toxin, but the effects were not genotype specific.  相似文献

In experiments with potted plants, the relationships between soil matric potential, plant water potential and production of water droplets (leaf surface wetness) on the folded central whorl leaf of seedlings of sorghum genotypes that are either resistant or susceptible to shoot fly (Atherigona soccata) damage were investigated. Differences in soil matric potentials in the pots affected the plant water status, which in turn had profound effects on the production of water droplets on the central whorl leaf of the sorghum genotype susceptible to shoot fly. There was no consistent variation in the relationship between plant water potential and soil matric potential of resistant and susceptible sorghum genotypes. However, there was very little or practically no water droplets on the central whorl leaf of the resistant genotypes, indicating that the production of water droplets is not solely the result of internal water status of the plant. It is suggested that leaf surface wetness is genetically controlled and that an understanding of the mechanism by which water is transferred to the leaf surface will enhance breeding for resistance to shoot fly.  相似文献

In sorghum (Sorghum bicolor [L.] Moench), the impact of heat stress during flowering on seed set is known, but mechanisms that lead to tolerance are not known. A diverse set of sorghum genotypes was tested under controlled environment and field conditions to ascertain the impact of heat stress on time-of-day of flowering, pollen viability, and ovarian tissue. A highly conserved early morning flowering was observed, wherein >90% of spikelets completed flowering within 30 min after dawn, both in inbreds and hybrids. A strong quantitative impact of heat stress was recorded before pollination (reduced pollen viability) and post pollination (reduced pollen tube growth and linear decline in fertility). Although viable pollen tube did reach the micropylar region, 100% spikelet sterility was recorded under 40/22°C (day/night temperatures), even in the tolerant genotype Macia. Heat stress induced significant damage to the ovarian tissue near the micropylar region, leading to highly condensed cytoplasmic contents and disintegrated nucleolus and nucleus in the susceptible genotype RTx430. Whereas, relatively less damages to ovarian cell organelles were observed in the tolerant genotype Macia under heat stress. Integrating higher tolerance in female reproductive organ will help in effective utilization of the early morning flowering mechanism to enhance sorghum productivity under current and future hotter climate.  相似文献

Proline accumulation and mobilization in roots of 7-d-old seedlings of wheat genotypes varying in sensitivity towards water stress were compared. Water stress was induced by polyethylene glycol (PEG-6000; osmotic potential −1.5 MPa) in the presence of 0.1 mM abscisic acid (ABA), 1 mM calcium chloride, 0.5 mM verapamil (Ca2+ channel blocker), 0.5 mM fluridone (inhibitor of ABA biosynthesis). While both the genotypes did not differ in total proline accumulation, rate of proline accumulation and utilization was higher in tolerant genotype C 306 as compared to susceptible genotype HD 2380. The treatment with ABA and CaCl2 caused further increase in proline accumulation during stress and reduced its mobilization during recovery. The membrane stability and elongation rate of roots was observed to be higher at ABA and calcium treatment in both the genotypes under stress. As was evident from inhibitor studies, the tolerant genotype was more responsive to ABA and the susceptible one to calcium. The reniform nematode (Rotylenchulus reniformis) is an important pathogen of pigeonpea (Cajanus cajan). Forty‐six medium maturity (mature in 151–200 days at Patancheru, India) pigeonpea genotypes were evaluated for resistance and tolerance to the reniform nematode in greenhouse and field tests, over the period 1990–97. Each genotype was screened for number of nematode egg masses on a 1 (no egg mass = highly resistant) to 9 (> 50 egg masses = highly susceptible) scale. Plant biomass production in carbofurantreated plots was compared with that in non‐treated plots in a field naturally infested with R. reniformis. Pigeonpea genotypes C 11, ICPL 87119 and ICPL 270 were used as nematode susceptible checks. Genotypes with good plant growth, both in nematode‐free and nematode‐infested plots, were identified as tolerant and evaluated for plant growth and yield for at least three years. All the tested genotypes were susceptible (7 and 9 egg mass score). Single‐plant‐selections, based on plant vigour and yield, were made from genotypes showing tolerance to nematode infection. The level of tolerance was enhanced by plant‐to‐progeny row selection for plant vigour and seed yield in a nematode‐sick field for at least three years. The most promising nematode tolerant genotypes produced significantly greater yield and biomass than the locally grown pigeonpea cultivars in fields naturally infested with R. reniformis at two locations. Pigeonpea landraces are considered to be the most likely sources of tolerance to the nematode. These reniform nematode tolerant lines represent new germplasm and they are available in the genebank of pigeonpea at ICRISAT bearing accession numbers ICP 16329, ICP 16330, ICP 16331, ICP 16332, and ICP 16333.  相似文献

The present investigation was undertaken to identify the possible mode of mechanism that could provide tolerance to maize (Zea mays L.) seedlings under waterlogging. Using cup method, a number of maize genotypes were screened on the basis of survival of the seedlings kept under waterlogging. Two tolerant (LM5 and Parkash) and three susceptible (PMH2, JH3459 and LM14) genotypes were selected for the present study. Activities of antioxidant and ethanolic fermentation enzymes and content of hydrogen peroxide (H2O2), glutathione and ascorbic acid were determined in roots of these genotypes after 72 h of waterlogging. Waterlogging treatment caused decline in activities of superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX) in all the genotypes. However, only susceptible genotypes showed slight increase in glutathione reductase (GR) activity. Significant reduction in APX/GR ratio in susceptible genotypes might be the cause of their susceptibility to waterlogging. The tolerant seedlings had higher GR activity than susceptible genotypes under unstressed conditions. Stress led to decrease in H202 and increase in glutathione content of both tolerant and susceptible genotypes, but only tolerant genotypes exhibited increase in ascorbic acid under waterlogging conditions. In the tolerant genotypes, all the enzymes of anaerobic metabolism viz. alcohol dehydrogenase (ADH), aldehyde dehydrogenase (ALDH) and pyruvate decarboxylase (PDC) were upregulated under waterlogging, whereas in susceptible genotypes, only ADH was upregulated, suggesting that efficient upregulation of entire anaerobic metabolic machinery is essential for providing tolerance against waterlogging. The study provides a possible mechanism for waterlogging tolerance in maize.  相似文献

A field experiment involving two planting densities (83,333 and 166,666 plants per ha), two cropping systems (monoculture and mixed culture) and five cowpea [Vigna unguiculata L. (Walp.)] genotypes was conducted at Nietvoorbij (33°54S, 18°14E), Stellenbosch, South Africa, to select cowpea material with superior growth and water-use efficiency (WUE). The results showed significantly higher photosynthetic rates, stomatal conductance and transpiration in leaves of plants at low density and in monoculture due to greater chlorophyll (Chl) levels relative to those at high density and in mixed culture. As a result, C concentration in leaves and the amount of C, P, K, Ca, Mg, Fe, Cu, Zn, Mn, and B accumulated in shoots at low density and under monoculture were also much higher. Even though no marked differences in photosynthetic rates were found between and among the five cowpea genotypes, leaf C concentration and shoot C, P, K, Ca, Mg, Fe, Cu, Zn, Mn, and B contents differed considerably, with Sanzie exhibiting the highest C concentration and C, P, K, Ca, Mg, Fe, Cu, Zn, Mn, and B contents in shoots, followed by Bensogla and Omondaw, while ITH98-46 and TVu1509 had the lowest shoot concentration and contents of C, P, K, Ca, Mg, Fe, Cu, Zn, Mn, and B. WUE (calculated as photosynthate produced per unit water molecule transpired) was significantly greater in plants at low density and monoculture relative to those at high density and in mixed culture. Isotopic analysis revealed significant differences in δ13C values of sorghum [Sorghum bicolor L. (Moench.)] and cowpea, with higher δ13C values being obtained for plants at low density and in monoculture relative to those at high density or in mixed culture. The five cowpea genotypes also showed significant differences in δ13C values, with Sanzie exhibiting the most negative value (i.e. low WUE) and ITH98-46, the least negative δ13C value (i.e. high WUE). Whether measured isotopically or from gas-exchange studies, sorghum (a C4 species) exhibited much higher WUE relative to cowpea (a C3 species). Both correlation and regression analyses revealed a positive relationship between WUE from gas-exchange studies and δ13C values from isotopic analysis of cowpea and sorghum shoots.  相似文献

Heterodera cajani is an important nematode pest of pigeonpea in India and a simple and reliable greenhouse procedure has been developed to screen pigeonpea genotypes for resistance to it. In pot experiments, white cysts of H. cajani were counted on the roots of the susceptible genotype ICPL 87 at 15, 30 and 45 days after seedling emergence in soils infested with different levels of H. cajani. The seedlings were rated for the number of white cysts per root system on a one (highly resistant, no cysts) to nine (highly susceptible, more than 30 cysts) scale. White cysts were not easy to see on wet roots but were clearly visible on slightly dried roots. Cyst counts and ratings were more uniform when roots of 30 day old seedlings were evaluated than when 15 or 45 day old seedlings were examined. Effects of different H. cajani infestation levels on the ratings were not significant although the use of higher inoculum densities (16 to 27 eggs and juveniles/cm3 soil) was effective in reducing variability. This procedure was used to screen 60 pigeonpea genotypes and all of them were rated seven or nine. Ten accessions of Atylosia spp. and Rhynchosia spp. were rated three.  相似文献

The effects of hypoxia on the levels of essential macroelements and trace elements (K, Na, Ca, Mg, Cu, Zn, Fe, and Mn) in the heart muscles of Wistar rats and plateau pikas (Ochotona curzoniae) were studied by atomic absorption spectrometry. Unlike the rat, the plateau pika is tolerant to hypoxia. The levels of K, Na, and the trace element Mn were not significantly changed in rat or pika hearts after exposure to hypoxia for 1, 10, or 25 d at simulated altitudes of 5000 and 7000 m. Other minerals (Ca, Mg, Cu, Zn, and Fe) were significantly affected by hypoxia and the levels followed different time-courses under different hypoxic regimes in these two animals. There were marked differences between the rat and pika in myocardial accumulation of essential elements such as Ca, which was increased to high levels in the rat but not affected in the pika. The results suggest that hypoxia affects animal physiological mechanisms by regulating the levels of essential elements.  相似文献

Shoot fly [Atherigona soccata (Rondani)] is a destructive pest of sorghum at the seedling stage and causes huge losses to grain yield and green fodder. The host-plant resistance mechanism is the best approach to reduce the attack of insects in plants. The damage parameters, morphophysiological traits, and biochemical metabolites had been investigated in the leaves and stem of contrasting sorghum genotypes, viz., resistant (IS18551, ICSV705, ICSV700), moderately resistant (PSC-4), and susceptible (SWARNA and SL-44) at 15 and 21 days after emergence (DAE) against shoot fly infestation. The resistant genotypes recorded lowest shoot fly oviposition and incidence (0.3–0.7 eggs plant−1 and 10–15%) than the susceptible genotypes (2.4–3.0 eggs plant−1 and 70–80%), respectively. The susceptible genotype SWARNA recorded 50% and 80% higher deadheart formation than the resistant genotype IS18551 at 15 and 21 DAE, respectively. Resistant genotypes exhibited higher trichome density at adaxial and abaxial part of leaf (118–145 and 106–131) with pink colored leaf sheath (scale 1.50–3.25), glossy leaves (scale1.00–1.25), and lower leaf surface wetness (scale1.25–2.00) compared with susceptible genotype with 49.3–73.3 and 25.3–64.0, scale 2.50–4.00, scale 2.75–3.50, and scale 3.25–4.25 for the respective parameters. Another defense response of sorghum toward the insect attack was modulation of plant metabolism. The infested genotypes responded to insect attack by upregulation of total soluble sugar, total phenol, prussic acid, and chlorophyll content by 1.2–2.1-fold, 1.5–2.0-fold, 1.2–1.3-fold, and 1.2–3.9-fold with more induction in susceptible genotypes at 21 DAE. On the whole, the present study indicates that morphophysiological and biochemical attributes contribute toward the resistance mechanism in sorghum against shoot fly infestation.

Drought susceptibility and low genetic variability are the major constraints of lentil (Lens culinaris Medik.) production worldwide. Development of an efficient pre-field drought phenotyping technique and identification of diversified drought tolerant lentil genotype(s) are therefore vital and necessary. Two separate experiments were conducted using thirty diverse lentil genotypes to isolate drought tolerant genotype(s) as well as to assess their diversity. In both of the experiments, significant (p ≤ 0.01) variation in genotype (G), treatment (T) and G X T was observed for most of the studied traits. In experiment I, genotypes were examined for drought tolerance at the seedlings stage under hydroponic conditions by assessing root and shoot traits. Among the 30 genotypes studied, BM-1247, BM-1227 and BM-502 were selected as highly tolerant to drought stress as they showed maximum seedling survivability and minimum reduction in growth parameters under drought stress. In experiment II, the genotypes were assayed for diversity and drought stress tolerance based on morphological traits grown under field condition. Drought stress caused a substantial reduction in yield attributing traits, however, the genotypes BM-1247, BM-981, BM-1227 and BM-502 were categorized as drought tolerant genotypes with less than 20% yield reduction. The field screening result of drought stress tolerance was coincided well with the results of laboratory screening. Genetic divergence study reflected the presence of considerable diversity among the genotypes. Considering laboratory and field screening results, the genotypes, BM-1247, BM-1227, BM-981 and BM- 502 were selected as the best drought tolerant genotypes. This information can be exploited for further breeding in developing drought tolerance in lentil.  相似文献

Developmental changes and effects of various abiotic stresses on peptidyl prolyl cis-trans isomerase (PPIase) activity were studied in the seedlings of sorghum [Sorghum bicolor (L.) Moench cv. CSH-6]. The PPIase activity of sorghum seedlings markedly decreased after two days of germination. Up to 90 % of the PPIase activity was inhibited by cyclosporin-A. Maximal increase in specific PPIase activity in the 3-d-old seedlings was observed in response to osmotic stress and it was transient in nature. The stress-induced enhancement in PPIase activity, depending upon tissue and stress treatment, was due to induction of cyclophilins as well as other PPIases. Osmotic stress-induced enhancement in PPIase activity in the drought susceptible cv. SPRU-94008B was maximal in roots, as compared to shoots in the drought tolerant cv. ICSV-272. An efficient plant regeneration system from leaf-derived callus in 6 genotypes of Avena sativa L. has been established. Regenerable callus was induced in the basal 1–2 mm region of 2 to 5 day old seedlings. Plants were regenerated from the regenerable callus and grown to maturity. The frequency of regenerable callus formation and plant regeneration was correlated with the position, developmental stage and genotype of the expiant. The regeneration capacity of the first one mm of the leaf basal region from three day old seedlings was comparable to that of immature embryos. Furthermore, the leaf regenerable calli were subcultured for 8 months without loss of their regeneration capabilities.  相似文献

Zinc deficiency and salinity are well-documented soil problems and often occur simultaneously in cultivated soils. Usually, plants respond to environmental stress factors by activating their antioxidative defense mechanisms. The antioxidative response of wheat genotypes to salinity in relation to Zn nutrition is not well understood. So, we investigated the effect of Zn nutrition on the growth, membrane permeability and sulfhydryl group (–SH groups) content of root cells and antioxidative defense mechanisms of wheat plants exposed to salt stress. In a hydroponic experiment, three bread wheat genotypes (Triticum aestivum L. cvs. Rushan, Kavir, and Cross) with different Zn-deficiency tolerance were exposed to adequate (1 μM Zn) and deficient (no Zn) Zn supply and three salinity levels (0, 60, and 120 mM NaCl). The results obtained showed that adequate Zn nutrition counteracted the detrimental effect of 60 mM NaCl level on the growth of all three wheat genotypes while it had no effect on the root and shoot growth of ‘Rushan’ and ‘Kavir’ at the 120 mM NaCl treatment. At the 0 and 60 mM NaCl treatments, Zn application decreased root membrane permeability while increased –SH group content and root activity of catalase (CAT) and superoxide dismutase (SOD) in ‘Rushan’ and ‘Kavir’. In contrast, Zn had no effect on the root membrane permeability and –SH group content of ‘Rushan’ and ‘Kavir’ exposed to the 120 mM NaCl treatment. At all salinity levels, ‘Cross’ plants supplied with Zn had lower root membrane permeability and higher –SH group content compared to those grown under Zn-deficient conditions. At the 0 and 60 salinity levels, Zn-deficient roots of Kavir and Rushan genotype leaked significantly higher amounts of Fe and K than the Zn-sufficient roots. In contrast, at the 120 mM treatment, Zn application had no effect or slightly increased Fe and K concentration in the root ion leakage of these wheat genotypes. For ‘Cross’, at all salinity levels, Zn-deficient roots leaked significantly higher amounts of Fe and K compared with the Zn-sufficient roots. The differential tolerance to salt stress among wheat genotypes examined in this study was related to their tolerance to Zn-deficiency, –SH group content, and root activity of CAT and SOD. Greater tolerance to salinity of Zn-deficiency tolerant genotype ‘Cross’ is probably associated with its greater antioxidative defense capacity.  相似文献

We sought to explain rice (Oryza sativa) genotype differences in tolerance of zinc (Zn) deficiency in flooded paddy soils and the counter‐intuitive observation, made in earlier field experiments, that Zn uptake per plant increases with increasing planting density. We grew tolerant and intolerant genotypes in a Zn‐deficient flooded soil at high and low planting densities and found (a) plant Zn concentrations and growth increased with planting density and more so in the tolerant genotype, whereas the concentrations of other nutrients decreased, indicating a specific effect on Zn uptake; (b) the effects of planting density and genotype on Zn uptake could only be explained if the plants induced changes in the soil to make Zn more soluble; and (c) the genotype and planting density effects were both associated with decreases in dissolved CO2 in the rhizosphere soil solution and resulting increases in pH. We suggest that the increases in pH caused solubilization of soil Zn by dissolution of alkali‐soluble, Zn‐complexing organic ligands from soil organic matter. We conclude that differences in venting of soil CO2 through root aerenchyma were responsible for the genotype and planting density effects.  相似文献
