The pre-release risk assessment of parasitoids for classical biological control generally involves non-target testing to define the agent’s host range. To ensure that no suitable host species are falsely rejected in these tests, it has been suggested that the physiological and informational state of parasitoids be manipulated to enhance their “motivation to oviposit”. However, the effects of such factors on host acceptance are not consistent across parasitoid species, making it laborious to identify the conditions necessary to maximise host acceptance. Our objective was to determine whether changes in parasitoid state could alter host acceptance behaviour sufficiently to affect host range expression. In addition, we tested the assumption that a state-dependent shift in motivation to oviposit on the target host will translate to a similar change in responsiveness to lower-ranked host species. Three-day-old and 10-day-old females of the candidate classical biological control agent, Diadromus pulchellus, were offered 12 non-target species of varying relatedness to the target pest, Acrolepiopsis assectella, in a series of no-choice and choice oviposition trials. Younger D. pulchellus females had previously demonstrated greater motivation to oviposit in the target pest and were, therefore, predicted to express a broader host range than older females. Parasitoid age had a minor effect on host range expression that was contrary to expectations. Older females more readily attacked one of the non-target species in no-choice tests and inflicted higher mortality in one of the choice tests. Ultimately however, young and old parasitoids still attacked the same four non-target species and their offspring emerged from the same three. There was an interaction between the effects of parasitoid condition and experimental design on responsiveness to low-ranked hosts: increasing non-target density in choice tests significantly altered attack rates by 10-day-old, but not by 3-day-old, parasitoids. The implications of these findings for host specificity testing depend largely on the specific aims of a host range assessment. Parasitoid state influenced the frequency of non-target attack but did not affect which non-target species were attacked.  相似文献

The polyphagous leafroller moth, Epiphyas postvittana, is a pest of many fruit crops in New Zealand. Since the larva is highly mobile, host selection in this insect may involve both the adult female and the larva. In order to test the relative importance of the adult female and the neonate larva in the selection of host plants, the ovipositional preferences of females, and the preferences or acceptances of neonate larvae towards 26 plant species, consisting of 15 plants considered hosts and 11 not considered hosts, were investigated. In the ovipositional tests, the mean preferences of females for hosts and non-hosts were very similar. In contrast, larvae showed a significantly greater mean preference or acceptance towards hosts than to non-hosts, in both choice and no-choice bioassays, respectively. There were highly significant correlations between the preferences and acceptances of larvae for plants in the choice and no-choice tests. In the no-choice tests, there was a highly significant correlation between the acceptances of neonate larvae towards plants after one and three days (i.e., acceptances changed little over time). Moreover, in these no-choice tests, there was a significant negative correlation between larval acceptance at 1 day and larval mortality after 3 days; that is, the less acceptable a given plant at 1 day, the more likely larvae would fail to establish, feed, and survive on it by three days. Female and larval preferences towards the various plants were also negatively correlated. Together, these data suggest that the selection of a plant for the neonate larva to feed on is largely governed by the preferences of the larva, rather than by the preferences of the female. However, selection of a plant for oviposition by the female, may be important in host selection for reasons unrelated to larval preferences, for example, by encouraging dispersal, perhaps to other plant species, of the neonate larvae and thereby decreasing intersibling competition.  相似文献

Adult oviposition preferences are expected to correlate with host plant suitability for the development of their offspring. For most lepidopteran species, this is particularly important as the hatching neonate larvae of many species are relatively immobile. Thus, the site of oviposition chosen by a female adult can greatly influence the probability of survival for her offspring. In the present study, we investigated the oviposition preference of adult Trichoplusia ni moths for six plant species to determine whether they could accurately rank the suitability of the plants for larval development. We also compared oviposition preferences to neonate larval acceptance and preference to determine whether the adult host range matched that of larval diet breath. Our results indicate that in two-choice and no-choice tests adult T. ni were able to rank the plants accurately, with the exception of anise hyssop. However, when given a choice of all six plants together, they laid more eggs on a plant that was not suitable for larval survival. Larvae accepted and fed on all plants in no-choice tests, and accurately ranked them according to larval performance. We conclude that neonate larvae are better able than adults to rank plants according to larval performance, and that larval diet breadth is wider than the range of plants accepted by adults. We also provide a discussion of the reduced accuracy of adult oviposition preference with increased plant choices.  相似文献

Chinese privet, Ligustrum sinense Lour., is a perennial semi-evergreen shrub that is a serious invasive weed in the United States. Classical biological control offers the best hope for controlling it in an economic, effective, and persistent way. Host specificity of one of the most promising biological control agents of Chinese privet, a flea beetle, Argopistes tsekooni Chen (Coleoptera: Chrysomelidae), was evaluated in China by using laboratory no-choice and choice tests on 13 species of Oleaceae and eight species in other families that have important economic value. In adult no-choice survival and oviposition tests, the flea beetle fed and survived for 30 d on Syringa oblata Lindl., Jasminum nudiflorum Lindl., and three species in the genus Ligustrum. Females also oviposited on these species, but only larvae from eggs laid on S. oblata and Ligustrum spp. developed successfully. In addition, the beetles did not feed or oviposit on the species of economic importance. In choice tests, adults preferred L. sinense for feeding and oviposition. These results show that A. tsekooni is relatively host specific and warrants further testing as a biocontrol agent of Chinese privet in the United States.  相似文献

Abstract:  Cat's claw creeper, Macfadyena unguis-cati , a major environmental weed in coastal and sub-coastal areas of Queensland and New South Wales, Australia is a target for classical biological control. Host specificity of Hypocosmia pyrochroma Jones (Lep., Pyralidae), as a potential biological control agent was evaluated on the basis of no-choice and choice larval feeding and survival, and adult oviposition preference tests, involving 38 plant species in 10 families. In no-choice tests, larval feeding and development occurred only on cat's claw creeper. In choice tests, oviposition and larval development was evident only on cat's claw creeper. The results support the host-specificity tests conducted in South Africa, and suggest that H. pyrochroma is a highly specific biological control agent that does not pose any risk to non-target plants tested in Australia. This agent has been approved for field release by relevant regulatory authorities in Australia.  相似文献

Various plant models were used in both choice and no-choice tests in the laboratory to assess landing and oviposition preferences of the cabbage root fly,Delia radicum (L.). The main factor governing the site most suitable for landing was the conspicuousness of the object and not its shape. Oviposition was influenced considerably by the pre-conditioning of the females. Deprived females laid eggs even when denied access to both host plant chemicals and host-plant models. The dominant role of contact chemical stimuli in host acceptance was reconfirmed, but only a combination of physical and chemical stimuli appeared capable of eliciting normal oviposition. The combination of contact chemical stimuli and the presence of a stem on the test model had a synergistic effect on the numbers of eggs laid in both choice and no-choice situations. In choice bioassays, female cabbage root flies distinguished between models of different shapes, heights and sizes. The size and shape of the models appeared to be perceived in part after the flies had landed.  相似文献

The solitary larval endoparasitoid Eadya daenerys Ridenbaugh (Hymenoptera: Braconidae) is a proposed biocontrol agent of Paropsis charybdis Stål (Coleoptera: Chrysomelidae, Chrysomelinae), a pest of eucalypts in New Zealand. Eadya daenerys oviposition behaviour was examined in two assay types during host range testing, with the aim of improving ecological host range prediction. No‐choice sequential and two‐choice behavioural observations were undertaken against nine closely related species of New Zealand non‐target beetle larvae, including a native beetle, introduced weed biocontrol agents, and invasive paropsine beetles. No behavioural measure was significantly different between no‐choice and two‐choice tests. In sequential no‐choice assays the order of first presentation (target–non‐target) had no significant effect on the median number of attacks or the attack rate while on the plant. Beetle species was the most important factor. Parasitoids expressed significantly lower on‐plant attack rates against non‐targets compared to target P. charybdis larvae. The median number of attacks was always higher towards target larvae than towards non‐target larvae, except for the phylogenetically closest related non‐target Trachymela sloanei (Blackburn) (Coleoptera: Chrysomelidae, Chrysomelinae). Most non‐target larvae were disregarded upon contact, which suggests that the infrequent attack behaviour observed by two individual E. daenerys against Allocharis nr. tarsalis larvae in two‐choice tests and the frass of Chrysolina abchasica (Weise) was probably abnormal host selection behaviour. Results indicate that E. daenerys is unlikely to attack non‐target species apart from Eucalyptus‐feeding invasive paropsines (Chrysomelinae). Non‐lethal negative impacts upon less preferred non‐target larvae are possible if E. daenerys does attack them in the field; however, this is likely to be rare.  相似文献

For some phytophagous insects, egg maturation may be dependent on adult feeding. Accordingly, rates of egg maturation may be dependent on the quality and quantity of available food sources. In turn, oviposition behavior could be affected by diet quality via changes in egg load (number of mature eggs carried by a female). Experiments were conducted to determine whether adult feeding may affect oviposition behavior of the glassy-winged sharpshooter, Homalodisca vitripennis. No-choice tests demonstrated that eggs accumulated in glassy-winged sharpshooter abdomens as time since last oviposition increased largely as a function of feeding plant species. In choice tests, glassy-winged sharpshooter females were observed most frequently on the plant species that imparted the greatest egg maturation rate in no-choice tests. Direct tests of the effects of egg load on glassy-winged sharpshooter oviposition behavior found that females were more likely to deposit eggs as egg load increased. Similarly, acceptance of a low-ranked oviposition plant species by female glassy-winged sharpshooters increased with egg load and time since last oviposition. The results indicate that adult feeding affected glassy-winged sharpshooter egg maturation, plant species varied in quality for providing nutrients for egg maturation, and egg load affected oviposition behavior. Thus, the quantity and quality of available feeding plant species may affect glassy-winged sharpshooter egg maturation rates, which in turn may affect the plant species female glassy-winged sharpshooters select for oviposition.  相似文献

Testing the specificity of candidate agents is a key component of risk analysis in weed biological control. This step is often time-consuming due to the numerous plant species that need to be tested under quarantine conditions in the invaded country of the weed species. Here, we examined whether an abridged phylogenetically based test list could be used in the weed's native range to quickly screen the host specificity of candidate agents. Ten plant species were used to test the host specificity of a promising candidate for the biological control of Sonchus oleraceus in Australia, the gall midge, Cystiphora sonchi. No-choice and choice tests were carried out in the native Mediterranean range of the midge. The results showed the midge has potential to threaten native Australian species, as those species showed high infestation levels in no-choice tests and produced significantly higher numbers of galls in choice tests. As a result of this approach, C. sonchi was rapidly discarded from the list of agents to be imported into Australian quarantines for further tests. This study demonstrates that testing a few key phylogenetically related species in the native range may save cost and effort in a weed biological control programme.  相似文献

In phytophagous insects, oviposition behaviour is an important component of habitat selection and, given the multiplicity of genetic and environmental factors affecting its expression, is defined as a complex character resulting from the sum of interdependent traits. Here, we study two components of egg-laying behaviour: oviposition acceptance (OA) and oviposition preference (OP) in Drosophila melanogaster using three natural fruits as resources (grape, tomato and orange) by means of no-choice and two-choice experiments, respectively. This experimental design allowed us to show that the results obtained in two-choice assays (OP) cannot be accounted for by those resulting from no-choice assays (OA). Since the genomes of all lines used are completely sequenced, we perform a genome-wide association study to identify and characterize the genetic underpinnings of these oviposition behaviour traits. The analyses revealed different candidate genes affecting natural genetic variation of both OA and OP traits. Moreover, our results suggest behavioural and genetic decoupling between OA and OP and that egg-laying behaviour is plastic and context-dependent. Such independence in the genetic architectures of OA and OP variation may influence different aspects of oviposition behaviour, including plasticity, canalization, host shift and maintenance of genetic variability, which contributes to the adoption of adaptive strategies during habitat selection.  相似文献

The invasive weed, parthenium (Parthenium hysterophorus L.) (Asteraceae: tribe Heliantheae), damages agriculture, adversely impacts biodiversity and is hazardous to human and animal health in Ethiopia. The host range of two natural enemies, a leaf-feeding beetle, Zygogramma bicolorata (Coleoptera: Chrysomelidae) and a stem-boring weevil, Listronotus setosipennis (Coleoptera: Curculionidae) was evaluated for biological control of the weed in Ethiopia. The specificity of Z. bicolorata and L. setosipennis was assessed against 29 and 31 non-target plant species, respectively. The host range of Z. bicolorata and L. setosipennis was first assessed using no-choice tests to examine their oviposition and feeding response on non-target plants. Although oviposition by Z. bicolorata occurred on six non-target species in four Asteraceae species in no-choice tests, it was significantly lower than on parthenium and no larvae developed. Zygogramma bicolorata nibbled the leaves of one of the five niger seed (Guizotia abyssinica L. – an oil seed crop closely related to parthenium) cultivars tested, but feeding and oviposition were significantly less than on parthenium. Furthermore, choice tests indicated that Z. bicolorata did not oviposit nor feed on G. abyssinica when parthenium was present. In no-choice tests, L. setosipennis did not oviposit on any of the non-target species assessed. Mean oviposition on parthenium was 39.0?±?3.4 eggs per plant whereas no eggs were laid on any of the 31 species tested. Based on these and other host range tests, permission was obtained to field release Z. bicolorata and L. setosipennis in Ethiopia.  相似文献

This paper describes a series of host-specificity tests carried out to evaluate the safety of eight insects for release in Australia as biological control agents of Onopordum spp. thistles; Larinus latus L., Lixus cardui Ol. and Trichosirocalus briesei Alonso-Zarazaga & Sanchez-Ruiz (Coleoptera: Curculionidae), Tephritis postica Loew and Urophora terebrans Loew (Diptera: Tephritidae), Botanophila spinosa Rondani (Diptera: Anthomyiidae), Tettigometra sulphurea Mulsant & Rey. (Hemiptera: Fulgoroidae) and Eublemma amoena (Hbn.) (Lepidoptera: Noctuidae). Preliminary tests were first carried out against key test plants in the country of origin to determine whether it was worth proceeding with formal testing under quarantine conditions in Australia. In the formal testing procedure, the test plant list was based solely on phylogenetic relationships. The life-stage of each insect tested was selected with respect to its biology and behaviour, e.g. only oviposition was tested for insect species with immobile endophagous larvae, while larval host utilization was tested for those with mobile larval stages. The initial laboratory experiments for each species used were caged no-choice tests, as confinement without choice can elicit extreme behavioural responses, making negative results extremely robust. Where positive results occurred, insects where re-exposed to the plant, but this time given a choice between the target and non-target plants. The results of these tests, plus data from open-field preliminary tests in the country of origin, helped interpret results and determine the risk posed by each candidate biological control agent to non-target plant species. Following submission, all eight candidates were approved for release by the Australian plant biosecurity and conservation authorities. Based on a comparison of the eight species tested, it is recommended that host-specificity testing be kept flexible and pragmatic, rather than moving towards a formulaic risk assessment procedure. Moreover, given the improved state of knowledge of plant phylogenies and the evolution of host utilization, it is time to base testing procedures purely on phylogenetic grounds, without the need to include less related test species solely because of economic or conservation reasons.  相似文献

1. Acceptance of hosts for oviposition is often hardwired in short‐lived insects, but can be dynamic at the individual level due to variation in physiological state determinants such as ageing and prior oviposition. However, the effect of the oviposition history of resources together with time taken to accept less preferred hosts in ageing insects has rarely been investigated. 2. The time taken by parasitic fig wasps to accept resources with different oviposition histories was recorded in order to investigate the effect of wasp physiological state and resource oviposition history on oviposition behaviour. These wasps, which differ in life‐history traits, oviposit at specific developmental stages of enclosed fig inflorescences called syconia. 3. Behavioural assays were performed with naive wasps and wasps aged with and without prior oviposition experience. Syconia at the same developmental stage but differing in oviposition history were offered in no‐choice assays and the time taken to first oviposition attempt was recorded. 4. One short‐lived pro‐ovigenic galler species exhibited a decline with age in time taken to accept a syconium for oviposition. The exact timing of the transition from non‐acceptance to acceptance of less preferred syconia was determined in terms of the proportion of elapsed life span at the transition; this occurred at 25% of elapsed life span. 5. Longer‐lived parasitoids did not show any decline in specificity despite being aged for 50% of their life span. Therefore, host quality, trophic position, egg load and age may individually affect oviposition decisions or have interaction effects.  相似文献

In weed biological control programs, pre-release host-specificity testing relies traditionally on no-choice and choice feeding, oviposition, and development tests. Rarely have they included detailed examination of behavioral responses to olfactory and visual cues of biological control candidates, although a better understanding of the mechanisms underlying host recognition may explain potential discrepancies between choice and no-choice tests, and/or between tests conducted in the lab versus field conditions. We investigated how the seed-feeding weevil, Mogulones borraginis, distinguishes its host plant, Cynoglossum officinale, from three native confamilial non-target species in North America. In behavioral bioassays, M. borraginis responded to olfactory and visual cues individually and, to an even greater extent, to both plant cue modalities when offered simultaneously. In tests with the combined cues, M. borraginis was attracted to C. officinale but responded with indifference or was repelled by non-target plants. In electrophysiological experiments, we identified that M. borraginis responded to ten volatile compounds and four wavelengths of lights from inflorescences of C. officinale. We propose that studies of responses to multimodal plant cues can advance our understanding of how biocontrol candidate species discriminate among host plants and closely related non-target species, thereby increasing the accuracy of environmental safety assessments pre-release.  相似文献

The South American egg parasitoid Gonatocerus sp. near tuberculifemur “Clade 1” (G. sp. “Clade 1”) (Hymenoptera: Mymaridae) is a new association of the glassy-winged sharpshooter (GWSS), Homalodisca vitripennis (Germar) (Cicadellidae) and a candidate for its biological control in California, USA. In Argentina, G. sp. “Clade 1” was screened in the laboratory (no-choice tests) and in the field (multiple choice tests) against eggs of 32 Auchenorrhyncha host species and other four potential hosts unrelated to sharpshooters. In no-choice assays, it parasitized only eggs within the leafhopper tribe Proconiini. In contrast, in the long term field tests, it emerged not only from eggs of the Proconiini but also from two species of Cicadellini at low numbers (five wasps out of 698 exposed eggs). Two interpretations arise from the results: (1) Host associations of G. sp. “Clade 1” are restricted to the Proconiini whereas field parasitization of the Cicadellini species were false positive, or (2) G. sp. “Clade 1” parasitizes also some Cicadellini species and its rejection in the laboratory was a false negative. Both interpretations are discussed. Insect motivation could be the explanation for the negative results in the no-choice tests. On the other hand, in the more natural field situations, the host selection process and oviposition behavior should not have been affected and host range would be more realistic. The parasitism of the Cicadellini species would be indicative of a potential non-target effect on the sharpshooters in the USA.  相似文献

Abstract.  1. Previous studies have produced conflicting results with respect to the genetic lability of host preference in the seed beetle Callosobruchus maculatus .
2. In this study, replicate lines of an Asian population were kept on an ancestral host (mung bean) or switched to a novel host (cowpea). After 40+ generations, lines were assayed for host preference (in choice tests) and host acceptance (under no-choice conditions), and were compared to African lines chronically associated with cowpea.
3. Host preference diverged in the expected direction. When presented a mixture of cowpeas and mung beans, females from the cowpea lines laid a greater fraction of their eggs on cowpea than did females from the mung bean lines. Preference for cowpea was nearly as strong in the cowpea lines as it was in the cowpea-adapted African lines.
4. In contrast, the experimental host shift did not affect long-term host acceptance. African females laid more eggs if given cowpeas than if given mung beans, but realised fecundities in the cowpea and mung bean lines were similar on the two hosts. Females from all lines laid more eggs if they were reared on cowpea than on mung bean, but rearing host had no effect on either relative host acceptance or host preference.
5. Comparisons with earlier studies suggest that the lability of host preference varies among beetle populations, which precludes generalisation at the species level. Because lines were maintained under no-choice conditions, modification of host preference probably occurred via a lower acceptance threshold for the novel host, without a concomitant change in the long-term acceptance of the ancestral host.  相似文献

Candidate weed biocontrol agents must be screened to exclude those that could threaten desirable plants . Traditionally , this has been done by rejecting species that develop on economically important plants in laboratory no - choice tests . However , because congeneric plants often support development in these tests , even when they are not utilized in nature , the tests do not meet legislated requirements for rare plant species or the increasing public concern for native plants . Plant suitability for larval development is a poor predictor of host range because insects use a sequence of steps in which the early steps , such as host finding and acceptance for oviposition , tend to be stronger than the later ones , such as suitability for development . This study is a trial of a new approach to screening insects as weed biocontrol agents that uses risk analysis to quantify the suitability of a plant as a host on the basis of inset performance at various stages in its life cycle . The insects used for the study was a NW Chinese biotype of the leaf beetle , Altica carduorum, which in terms of climate adaptation and damage it inflicts on the weedy thistle Cirsium arvense, is a promising biocontrol agent for Canada . However , its ability to develop on all North American Cirsium spp . in laboratory no - choice tests currently excludes its release . We show by risk and factor analyses , with five sequential host - selection parameters , that the suitability of these thistles to A. carduorum is so low that the beetle would not jeopardize the continued existence of rare native thistles , so its release should not be a problem .  相似文献

The oviposition and feeding preferences of the pollen beetle, Meligethes aeneus, were determined in choice and no-choice tests in field, semi-field and greenhouse trials. Plant species used were Brassica napus, B. campestris, B. juncea, B. nigra, B. carinata, Sinapis alba and Crambe abyssinica. With respect to number of eggs laid, S. alba and C. abyssinica were inferior to the other species. Pollen beetles laid fewer eggs on B. nigra than on the other Brassica spp. in no-choice tests, however this difference was partly due to fewer eggs laid per bud rather than fewer buds used for oviposition. Most eggs, for all plant species, were deposited in buds sized 2–3 mm. Feeding damage on all plant species was relatively similar. Pollen beetles seem to have a wider host range for feeding than for oviposition. There was good agreement in plant species ranking as oviposition hosts between the field, semi-field and greenhouse trials.  相似文献

The effect of adult learning through an oviposition experience, and pre-adult learning through development inside a host was investigated in two strains of an Australian egg parasitoid,Trichogramma nrivelae (Hymenoptera: Trichogrammatidae). Host response was measured in two types of laboratory preference tests. In single host tests, females reared on three lepidopteran hosts (Heliothis punctigera (Noctuidae),Papilio aegeus (Papilionidae), andHypolimnas bolina (Nymphalinae)) were presented host eggs individually, and allowed two ovipositions. Pre-adult experience affected host preference in only one strain, while acceptance of a host was increased in both strains when they had previously oviposited in this host species. An oviposition experience had a stronger effect on host preference than pre-adult experience. In choice tests, pairwise combinations of hosts were arranged in an alternating grid. The ratio of host acceptances to host contacts was computed for the whole test and up to the first oviposition. These indicated that effects of rearing host were weak or absent, but the host chosen initially had a strong effect on host preference. Relative size of the hosts had a strong effect on choice of the first host. The implications of learning inTrichogramma are discussed in relation to host preference testing procedures and the selection of candidate strains for mass rearing and inundative release.  相似文献

The innate preference of Trichogramma platneri for the eggs of six host species (Chrysoperla carnea (Stephens), Cydia pomonella (L.), Ephestia kuehniella (Zeller), Helicoverpa zea (Boddie), Manduca sexta (L.), and Sitotroga cerealella (Olivier)) was evaluated as the first host accepted by an individual female parasitoid when eggs were presented simultaneously. Selective exploitation of the same hosts by T. platneri was evaluated in no-choice, paired-choice, and multiple-choice tests, using number of parasitoids emerged, proportion of female offspring, and proportion of time spent on host eggs as different measures of preference. T. platneri showed a strong innate preference for C. pomonella and M. sexta. H. zea and M. sexta were the most preferred hosts and S. cerealella the least preferred, in the no-choice and choice tests. The number of progeny emerged and proportion of female offspring were equally suitable measures of host preference for T. platneri. Although no-choice and choice tests provided very similar preference ranking for T. platneri, a polyphagous species, the no-choice tests appeared poorer at separating hosts that were close in rank and multiple-choice tests were considered likely to be more realistic for more specialized control agents.  相似文献
