Glucan production from sucrose by Streptococcus mutans OMZ 176 was stimulated approximately threefold in the presence of 0.1% Tween 80. When OMZ 176 was grown in a medium containing glucose, the glucosyltransferase level in the medium was also increased about fivefold in the presence of 0.1% Tween 80. The glucosyltransferase level increased in proportion to the logarithm of the concentration of Tween 80 in the glucose medium. Tween 80 affected neither bacterial growth nor the activity of glucosyltransferase. The appearance of glucosyltransferase in the glucose medium was inhibited immediately by chloramphenicol and actinomycin D and, after a lag, by rifampin as well. It was observed that the fatty acid composition of the cells grown with Tween 80 was altered. These results suggest that Tween 80 stimulates glucosyltransferase synthesis either directly, or indirectly by promoting glucosyltransferase secretion.  相似文献

The rate of expression of the cell-associated fructosyltransferase (FTFm) activity of Streptococcus salivarius ATCC 25975 grown in continuous culture was linearly related to the rate of octadecenoic acid (C18:1) incorporation into the membrane lipids irrespective of the presence or absence of Tween 80 in the growth medium. This observation was confirmed with data obtained from cells grown in the presence of a series of n-alkanols. The results suggested that cosynthesis of lipids containing C18:1 residues was necessary for FTFm expression and accounted for the slight stimulation of enzyme expression by Tween 80 at all growth rates. In contrast, addition of Tween 80 to the growth medium resulted in several-fold increases in extracellular glucosyltransferase (GTFe) production irrespective of the growth rate. Following the addition of the surfactant to the growth medium, an exponential relation between the increased rate of GTFe production and the concomitant net increase in the rate of C18:1 incorporation was noted. The results obtained in continuous culture emphasized the underlying effect growth rate had on GTFe production, especially when Tween 80 was added to the growth medium. In the presence of n-alkanols, the rate of GTFe production plotted as a single 'U'-shaped curve with respect to the rate of C18:1 incorporation irrespective of the chain length of the n-alkanol studied. Rapid analyses of the extracellular proteins by SDS-PAGE suggested that hexan-1-ol and Tween 80 specifically stimulated the synthesis and secretion of GTFe and no other extracellular protein.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

When Streptococcus salivarius was grown in batch culture in the presence of various Tween detergents, the fatty acid moiety of the detergent was incorporated into the lipids of its membrane. Tween 80 (containing primarily oleic acid) markedly stimulated the production of extracellular glucosyltransferase and also increased the degree of unsaturation of the membrane lipid fatty acids. The possibility that an increase in membrane unsaturated fatty acids promoted extracellular glucosyltransferase production was examined by growing cells at different temperatures in the presence or absence of Tween 80. The membrane lipids of cells grown at 30 degrees C, 37 degrees C and 40 degrees C without Tween 80 exhibited unsaturated/saturated fatty acid ratios of 2.06, 1.01 and 0.87 respectively. A significant increase in the production of extracellular glucosyltransferase was observed at 30 degrees C compared to cells grown at 40 degrees C. However, cells produced much more exoenzyme at all temperatures when grown with Tween 80. The results indicated that an increase in the unsaturated fatty acid content of the membrane lipids was not by itself sufficient to account for the stimulation of extracellular glucosyltransferase production by Tween 80, but that the surfactant also had to be present.  相似文献

Streptococcus salivarius (ATCC 25975) produced very low or nondetectable amounts of the extracellular enzyme glucosyltransferase (GTase) when grown in a chemically defined medium. The addition of Tween 80 to this medium resulted in the production of markedly enhanced levels of the enzyme. Oleic acid, the methyl ester of oleic acid, and sucrose each could not substitute for Tween 80 in this regard. The surfactant had no direct activating effect on performed enzyme activity. Tween 80 also stimulated the production of GTase by concentrated cells suspended in defined medium during a time when no measurable growth occurred. Under these conditions, the stimulatory effect of Tween 80 was blocked by chloramphenicol. It was further found that the surfactant dramatically stimulated the differential rate of GTase synthesis. These and other data strongly suggest that Tween 80 stimulates the production of extracellular GTase by acting either directly or indirectly at the level of enzyme synthesis.  相似文献

Thermostable lipase production by Geobacillus thermoleovorans was optimized in shake-flask cultures using Box-Behnken experimental design. An empirical model was developed through response surface methodology to describe the relationship between tested variables (Tween 80, olive oil, temperature and pH) and enzyme activity. Maximum enzyme activity (495 U l–1) was attained with Tween 80 at 5 g l–1; olive oil at 60 g l–1; 70 °C and pH 9. Experimental verification of the model showed a validation of 95%, which is more than 4-fold increase compared to the basal medium.  相似文献

We investigated the influence of inorganic phosphate concentration on the production of curdlan by Agrobacterium species. A two-step culture method was employed where cells were first cultured, followed by curdlan production under nitrogen-limiting conditions. In the curdlan production step, cells did not grow but metabolized sugar into curdlan. Shake-flask experiments showed that the optimal phosphate concentration for curdlan production was in the range of 0.1–0.3 g l−1. As the cell concentration increased from 0.42 to 1.68 g l−1 in shake-flask cultures, curdlan production increased from 0.44 to 2.80 g l−1. However, the optimal phosphate concentration range was not dependent upon cell concentration. The specific production rate was about 70 mg curdlan g-cell−1 h−1 irrespective of cell concentration. When the phosphate concentration was maintained at 0.5 g l−1 under nitrogen-limiting conditions, as high as 65 g l−1 of curdlan was obtained in 120 h. Statistically-based experimental designs were applied to optimize the fermentation for the production of glucosyltransferase by Leuconostoc dextranicum NRRL B-1146. Eleven medium components were examined for their significance on enzyme production using Plackett-Burman factorial design. Tween 80, sucrose and K2HPO4 significantly improved the enzyme production process. The combined effect of these nutrients on glucansucrase production were studied using a 2 2 full-factorial central composite design, a second-order polynomial was established to identify the relationship between the enzyme output and the three medium components. The optimal concentration of variables for maximum glucansucrase production were Tween 80 (0.55%, v/v); sucrose (5.6%, w/v) and K2HPO4 (1%, w/v). The maximum enzyme activity by predicted model was 6.53 U/ml that was in perfect agreement with the actual experimental value (6.40 U/ml).  相似文献

Nine bacterial strains grew at 50°C on a mineral medium containing Tween 80 as sole carbon source and three of them produced significant lipolytic activities. The strains showed optimal growth at 50°C and thus, they are moderate thermophiles. Lipolytic activity were maxima at 57–67°C suggesting high thermostability. Lipolytic activity production was promoted by the addition of Tween 80 in culture medium and inhibited by glucose addition. Tween 80 enhanced TNT mineralization by Phanerochaete chrysosporium   总被引:1,自引:0,他引:1  
The effect of a nonionic surfactant (Tween 80) on 2,4,6-trinitrotoluene (TNT) mineralization by the white-rot fungus Phanerochaete chrysosporium strain BKM-F-1767, was investigated in a liquid culture at 20, 50, and 100 mg TNT.L-1. The presence of 1% (w/v) Tween 80, at 20 mg.L-1 TNT, added to a 4-d-old culture, allowed the highest TNT mineralization level, that is 29.3% after 24 d, which is two times more than the control culture, without Tween 80 (13.9%). The mineralization of TNT resumed upon additional Tween 80 supplementation, consequently, 39.0% of the TNT was respired on day 68. Orbital agitation of the fungal culture was found detrimental to TNT mineralization, with or without Tween 80 in the culture medium. The surfactant also stimulated the growth of P. chrysosporium without any notable effect on either the glycerol consumption rate or the extracellular LiP and MnP activity levels. Respirometric assays highlighted some differences between the oxygen uptake rate of the fungal culture supplemented with or without Tween 80.  相似文献

Primary conidia of the entomopathogens Erynia (subgenus Neopandora) delphacis (1 isolate) and Erynia ( Neopandora) neoaphidis (3 isolates) were stimulated to form germ-tubes with Tween 20 and with free, long-chain fatty acids, each incorporated into Entomophthora complete medium (ECM). When combined with other basal media (three tested), these compounds did not stimulate germ-tube formation. Triacylglycerols and vegetable oils, added to the same media, allowed almost complete resporulation in the fungi. In both species, Tween 20 (0.1%) encouraged greater germ-tube production (41–69%) than the fatty acids (0.1%) (≤36%). For E. delphacis, Tween 20 and the fatty acids differed significantly, but for E. neoaphidis the differences were almost always insignificant. Myristic and oleic acids stimulated germ-tube formation in both species. Palmitic acid allowed almost complete resporulation of the fungi, except for one isolate of E. neoaphidis that formed germ-tubes. Linoleic acid, tested only for E. delphacis, was fungistatic to most conidia. Higher concentrations of the fatty acids (≤1%) did not increase germ-tube formation, except 1% oleic acid which affected E. delphacis alone (>80% germination and germ-tubes). Linoleic acid, and sometimes also myristic and oleic, were fungistatic and/or toxic, depending on their concentration and on medium composition. Addition of fatty acids to ECM usually extended the lag period, and altered the morphology of the conidia and germ-tubes. These phenomena were not observed with Tween 20. Colonies were formed by E. delphacis alone, stimulated by ECM supplemented with Tween 20 or fatty acids. The results are discussed with respect to biological and physiological aspects of germination, and with respect to the mode of action of the fatty acids and the surfactant. 1. Both Tween 80 and sodium fluoride significantly enhanced total extracellular glucosyltransferase activities of Streptococcus mutans. 2. Water-insoluble and water-soluble glucan formation were uniformly increased by Tween 80, whereas fluoride stimulated only water-soluble glucan formation. 3. Elevated glucan formation was due to an increase in enzymes secreted from bacterial cells. 4. Fatty acid composition and phospholipid content in bacterial membrane were changed by Tween 80, although sodium fluoride scarcely showed these changes. 5. Comparative results suggest that modulation of membrane lipids participates in mutansucrase production but not in dextransucrase production of S. mutans.  相似文献

The effects of structure and concentration of surfactants on the biodegradation of fluoranthene, a three rings polycyclic aromatic hydrocarbon in the aqueous phase, as well as their effects on the biodegradation and enzyme activity were investigated. The toxicity ranking of studied surfactants is: non-ionic Tween 80 <anionic sodium dodecyl sulfate <cationic Tetradecyltrimethylammonium bromide. The maximum growth of Armillaria sp. F022 (>4,500 mg/L) was showed by Tween 80 (10 mg/L) culture, manifesting that the non-ionic surfactant present in the culture were beneficial to the fungal growth. Laccase showed the highest enzymes activity in all surfactants culture. Non-ionic Tween 80 showed a significant result for laccase activity (1,902 U/L) in the Armillaria sp. F022 culture. The increased enzymes cumulative activity may stem directly from the rising fluoranthene biodegradability as addition of appropriate surfactants. The biotransformation of fluoranthene was greatly improved by Tween 80, and totally fluoranthene degradation was obtained as Tween 80 was 10 mg/L. Two fluoranthene metabolites were isolated from the culture medium and analyzed by a thin layer chromatography, UV visible spectrometer and gas chromatography–mass spectrometry (GC–MS). The oxidation of fluoranthene is initiated by oxygenation at the C-2,3 positions resulting 9-fluorenone. At the end of experiment, one metabolite was detected in the culture extract and identified as phthalic acid. Evidently, Armillaria sp. F022 seems efficient, high effective and deserves further application on the enhanced bioremediation technologies for the treatment of fluoranthene-contaminated soil.  相似文献

Summary Maximum xylanase production byChaetomium cellulolyticum was obtained in the culture supernatant after 30 h of growth at 37°C in basal medium containing 1% xylan at pH maintained between 6.5 and 7.5. Addition of 0.05% Tween 80 to the medium increased the enzyme production considerably. Xylanase production was found to be growth associated. The optimal conditions for enzymatic hydrolysis of xylan were found to be pH 6.0 and 50°C. During enzymatic hydrolysis, xylose, xylobiose and other xylooligosaccharides were liberated from xylan. The pH values for xylanase production and for xylan hydrolysis were closely related to the utilization of hemicelluloses of aspen wood for fungal protein production by this organism as reported in our earlier work.  相似文献

Tween 80 (0.1%, v/v) added to Thermomonospora curvata growing in minimal medium caused a transient lowering of the dry cell mass, decreased the optimal growth temperature of the thermophile from 62 to 54°C, and increased extracellular esterase activity. Cells grown in the presence of Tween 80 had decreased concentrations of branched chain fatty acids and increased concentrations of oleic acid. The detergent removed surface protuberances from mycelia and increased the liberation of enzymes active against crystalline cellulose, but did not stimulate liberation of enzymes active against carboxymethylcellulose, starch or pectin.  相似文献

A diverse surfactant, including the nonionic Tween 80 and Brij 30, the anionic sodium dodecyl sulphate, the cationic surfactant Tetradecyltrimethylammonium bromide, and biosurfactant Rhamnolipid were investigated under fluorine-enriched medium by Armilaria sp. F022. The cultures were performed at 25 °C in malt extract medium containing 1 % of surfactant and 5 mg/L of fluorene. The results showed among the tested surfactants, Tween-80 harvested the highest cell density and obtained the maximum specific growth rate. This due Tween-80 provide a suitable carbon source for fungi. Fluorane was also successfully eliminated (>95 %) from the cultures within 30 days in all flasks. During the experiment, laccase production was the highest among other enzymes and Armillaria sp. F022-enriched culture containing Non-ionic Tween 80 showed a significant result for laccase activity (1,945 U/L). The increased enzyme activity was resulted by the increased biodegradation activity as results of the addition of suitable surfactants. The biotransformation of fluorene was accelerated by Tween 80 at the concentration level of 10 mg/L. Fluorene was initially oxidized at C-2,3 positions resulting 9-fluorenone. Through oxidative decarboxylation, 9-fluorenone subjected to meta-cleavage to form salicylic acid. One metabolite detected in the end of experiment, was identified as catechol. Armillaria sp. F022 evidently posses efficient, high effective degrader and potential for further application on the enhanced bioremediation technologies for treating fluorene-contaminated soil.  相似文献

Lipases are important cuticle degrading enzymes involved in the infection process of entomopathogens by hydrolysing the ester bonds of lipoproteins, fats and waxes present in the insect integument. Production of extracellular lipase by Isaria fumosoroseus (Cordycipitaceae; Hypocreales) isolate IF28.2 was investigated using different combinations of basal medium components. The effect of different vegetable oils added to a basal medium at different concentrations to improve enzyme production was evaluated. Maximum lipase activity (125.33±2.96 U/mL) as well as maximum biomass production (22.36±0.99 mg/mL) was observed for olive oil when used at a concentration of 2% (v/v) of the basal medium. In the presence of surfactants, the highest lipase activity occurred when SDS and Tween 80 were added at the time of fungal inoculation. SDS proved to be the best surfactant having 110.66±3.52 U/mL lipase activity. The effects of the divalent metal ions (iron and magnesium) on lipase activity were also studied. Iron inhibited, whereas magnesium slightly increased lipase activity. The optimum pH for lipase production was 5.7 while 32°C proved to be the best temperature for lipase production.  相似文献

Acinetobacter O16, a psychrophilic species, produced extracellular lipase (measured by hydrolysis of olive oil, tributyrin, or beta-naphthyl laurate) when grown on a complex medium (peptone plus yeast extract). Most lipase was produced during the logarithmic phase of growth. Very little cell-bound lipase was formed. These cells also produced an esterase (measured by the hydrolysis of beta-naphthyl acetate). At first, all esterase was cell bound; significant amounts appeared in the external medium late in growth. Breaking the cells did not increase cell-bound lipase activity. After breaking of the cells, most of the cell-bound lipase and esterase activity was solubilized, even after very high speed centrifugation. No appreciable amounts of these enzymes were released by osmotic shock. Lipase formation was greatly affected by nutrient conditions. Lowering either the yeast extract of the peptone content of the normal complex medium lowered or abolished lipase formation. Esterase activity was lowered to a lesser extent. Cells growing in synthetic amino acid plus vitamin medium or in acid-hydrolyzed casein produced substantial amounts of esterase but no cell-free or cell-bound lipase. However, if sodium taurocholate was added to these media, lipase was produced. Greatest production occurred if a mixture of di- and poly-peptides was also present. Taurocholate also stimulated lipase production in the normal complex medium. Adding Tween 80 or ethanol to the normal complex medium inhibited lipase production. Sodium acetate, oleic acid, olive oil, or Tween 20 added to synthetic media did not affect lipase production. The psychrophile grew more quickly at 30 degrees C than at 15 or 20 degrees C but produced more lipase at the lower temperatures. Esterase production was about the same at 20 and 30 degrees C. A mesophilic Acinetobacter species produced the same amount of lipase and esterase at 20 and 30 degrees C. The best production of lipase by the psychrophile occurred in standing cultures.  相似文献

This study investigated the effects of surfactant additives and medium pH on mycelial morphology and exopolysaccharide (EPS) production in liquid culture of a valuable medicinal fungus Cordyceps sinensis Cs-HK1. In the medium containing 20 g l−1 glucose and 6 g l−1 peptone as the sole nitrogen source, the Cs-HK1 fungal mycelia formed smooth and spherical pellets about 1.8-mm mean diameter. The mycelial pellets became less uniform at pH (4.0–5.0) lower than the optimum (pH 6.0) or turned to filamentous form at higher pH (8–9). Surfactants added to the medium inhibited pellet formation, resulting in smaller and looser pellets. Tween 80 exhibited a remarkable promoting effect on EPS production, increasing the EPS yield more than twofold at 1.5% (w/v), which was most probably attributed to the stimulation of EPS biosynthesis and release from the fungal cells by Tween 80.  相似文献

热凝胶是粪产碱杆菌(Alcaligenes faecalis)在氮源限制条件下生成的水不溶性胞外多糖,分泌到胞外后就附着在菌体外壁,因此在细胞生长期提高生物量对促进热凝胶合成有重要意义。热凝胶分批发酵时, 起始NH4Cl浓度提高到3.6 g/L时能促进菌体生长和热凝胶合成,但是过量NH4Cl会抑制热凝胶合成,且生物量提高不是很明显。为了进一步提高菌体浓度, 在菌体生长期, 氨水取代NaOH溶液进行流加控制pH为7.0, 随后又用2 mol/L NaOH控制pH 5.6。实验表明, 氨水流加使菌体浓度大大提高,流加24 h使菌体浓度达到18.8 g/L。但是菌体浓度过高也会抑制热凝胶的合成,在氨水流加14 h时,菌体浓度在11.9 g/L左右, 热凝胶产量最高(72 g/L)。  相似文献
