为了解乙二胺二琥珀酸(EDDS)诱导植物耐受硒(Se)胁迫的生理机制,以彩叶草(Coleus blumei)为材料,采用营养液培养的方法、借助傅里叶变换-衰减全反射红外光谱(FTIR-ATR)和扫描电子显微镜-X-射线能量色散谱(SEM-EDXS)分析方法及生理指标的变化,研究1.0 mg/L Se胁迫条件下添加0、0.5、1.0、1.5、2.5、5.0 mmol/L EDDS对彩叶草根系化学成分变化的影响。利用FTIR-ATR图谱分析发现,随着EDDS处理浓度的提高,彩叶草根系透射峰所对应峰形基本不变,而参与Se吸附的基团如羟基、酰胺基和指纹区等的透射峰发生了不同程度的位移。FTIR-ATR的特征峰与彩叶草根系响应Se胁迫的各生理指标变化趋势基本一致,且FTIR-ATR比传统的生理指标测定更敏感、便捷。SEM-EDXS扫描还发现随着EDDS处理浓度的升高,根系中K、Mg、Fe、Si等元素的含量升高,而营养元素Ca含量降低。该研究结果可为深入了解EDDS处理下彩叶草对Se胁迫的响应机理提供科学依据。  相似文献

以一种具有较高Se富集能力的植物——斜茎黄芪(Astragalus adsurgens)为试验材料,在水培条件下研究不同浓度Se处理对其生长发育、光合参数和初生代谢产物积累的影响。结果表明:5μmol·L-1Se处理显著促进了斜茎黄芪根系和地上部的伸长生长;100μmol·L-1Se处理则具有相反效果。光合色素含量和叶片叶绿素荧光参数研究结果与生长指标相一致,低浓度Se对斜茎黄芪叶片的光合作用具有促进作用,高浓度Se则对其具有明显的抑制作用。随着处理浓度的升高,斜茎黄芪根系和地上部Se含量逐渐增加,但Se的转运系数明显降低,100μmol·L-1处理下Se转运系数比5μmol·L-1处理降低了83.5%。通过对初生代谢产物进行检测和分析,发现低浓度Se处理主要上调了斜茎黄芪中与氨基酸代谢相关途径的代谢水平,而高浓度Se处理主要上调了与次生代谢物质合成关系密切的初生代谢产物的代谢水平。我们的研究结果表明,低浓度Se处理提高了斜茎黄芪光合作用水平以及与生长相关的初生代谢水平,从而促进了植株生长;高浓度S...  相似文献

镉对超富集植物滇苦菜抗氧化系统的影响   总被引:14,自引:1,他引:13  
采用水培实验,研究镉(Cd)对一种新发现的Cd超富集植物——滇苦菜抗氧化系统的影响。结果表明,中、低浓度Cd(1-10μmol.L-1)处理下滇苦菜生物量与对照无显著差异,而高浓度Cd(50-100μmol.L-1)抑制了滇苦菜的生长,整株生物量比对照降低了72%和86%。10μmol.L-1Cd处理下地上部Cd浓度达到270mg·kg-1(干重),转运系数为1.41,符合超富集植物的特征。滇苦菜地上部Cd富集浓度最高达3919mg·kg-1,且64%-87%的Cd分布在地上部。中、低浓度Cd(1-10μmol.L-1)胁迫下,滇苦菜的丙二醛(MDA)和H2O2含量、超氧阴离子自由基(O2.-)产生速率及各抗氧化酶活性与对照没有显著差异。但高浓度Cd(50-100μmol.L-1)胁迫下,地上部MDA、H2O2含量和O2.-产生速率比对照分别提高了5-17、1.6-6倍和2.9-7.2倍,地下部分别提高了1-5、1.4倍和9.5-11倍。同时,高浓度Cd处理下地上部和地下部超氧化物歧化酶(SOD)和过氧化物酶(POD)活性以及地上部谷胱甘肽(GSH)含量显著上升,但谷胱甘肽还原酶(GR)和过氧化氢酶(CAT)活性变化不明显。研究表明,滇苦菜对Cd有较强的富集和转运能力。中、低浓度Cd处理未对滇苦菜造成明显的氧化胁迫,高浓度Cd处理时滇苦菜地上部与地下部均受到氧化胁迫,但对抗氧化胁迫的响应方式不同。  相似文献

采用盆栽试验,对彩叶草进行PEG-6000浓度为0(对照)、5%、10%、15%、20%(W/V)模拟干旱胁迫,研究在干旱胁迫下彩叶草的生长、渗透调节能力及抗氧化酶活性的变化。结果表明:与对照相比,随着PEG-6000浓度的增加,鲜质量、干质量、含水量、水势、根系脱氢酶活性、无机离子含量包括K+、Na+、Ca2+、Mg2+等均呈下降趋势;NO-3含量呈先下降后上升趋势;硝酸还原酶活性、可溶性蛋白含量、可溶性糖含量、超氧化物歧化酶活性均呈先上升后下降趋势;脯氨酸含量、游离氨基酸含量、过氧化物酶活性、过氧化氢酶活性、超氧阴离子(O-2·)产生速率、质膜透性则呈上升趋势。因此,模拟干旱胁迫对彩叶草生长有抑制作用,且随着PEG-6000浓度增加,其生长受抑制和水分胁迫程度加重;模拟干旱胁迫下,彩叶草不积累K+、Na+、Ca2+、Mg2+和NO-3等无机离子进行渗透调节,而积累脯氨酸、可溶性蛋白、可溶性糖、游离氨基酸等有机小分子物质进行渗透调节,但这4种小分子物质增加幅度不尽相同;轻度模拟干旱胁迫虽增强彩叶草抗氧化酶活性,但仍表现轻度的氧化伤害;重度模拟干旱胁迫加重彩叶草氧化伤害。研究结果可为彩叶草耐旱生理机制的研究积累资料,也为其节水型栽植和养护提供依据。  相似文献

在实验室条件下用低(0.5 mmol·L-1)、中(8 mmol·L-1)、高(15 mmol·L-1)浓度Al3 溶液胁迫接种和未接种丛枝菌根(AM)的樟树幼苗,10周后测定植株叶片的叶绿素含量、净光合速率(Pn),气孔导度(Gs)、蒸腾速率(Tr)、胞间CO2浓度(Ci)和气孔限制值(Ls).结果表明:未接种和接种樟树幼苗叶片叶绿素含量在低浓度Al3 胁迫时与相应对照无显著差异,而在中、高浓度Al3 胁迫下均显著低于相应对照,并均随Al3 浓度增加而逐渐下降;接种植株的叶绿素含量在对照和中、低浓度Al3 胁迫下均高于相应未接种植株,而在高浓度Al3 胁迫下显著低于未接种植株.未接种和接种樟树幼苗叶片Pn在低浓度Al3 胁迫时均显著高于对照(P<0.05),在中、高浓度时均显著低于对照,且在Al3 浓度间差异显著;末接种和接种樟树幼苗叶片Ci均随Al3 胁迫浓度增加逐渐提高,而其余参数则逐渐下降;在同一Al3 浓度处理下,接种和未接种植株的叶片光合参数均无显著差异.研究发现,中、高浓度Al3 胁迫能显著降低樟树幼苗的净光合速率,且光合机构活性降低是主要原因;接种AM真菌能显著增加中、低浓度Al3 胁迫樟树幼苗的叶片叶绿素含量,但不能显著减轻中、高浓度Al3 胁迫对樟树幼苗光合速率的抑制作用.  相似文献

采用室内试验和田间试验相结合方式,研究了不同浓度(0、0.01、0.05、0.10、0.50和1.00 mmol·L-1)2,4-二叔丁基苯酚对番茄叶霉菌及番茄种子萌发幼苗生长的影响.结果表明:2,4-二叔丁基苯酚在叶霉菌培养的前阶段极显著地抑制了菌丝的生长,培养7d时,0.1 mmol· L-1的抑制作用最大,抑制率为40%;在田间抗病性调查期间各浓度处理均提高了番茄幼苗的抗病性,其中以0.1 mmol· L-1处理抗病性最好,病情指数比对照降低了54%;低浓度2,4-二叔丁基苯酚促进了番茄种子的萌发,增加了幼苗的株高、茎粗、地上和地下部鲜质量、叶绿素含量、根系活力和幼苗体内保护酶活性,随着浓度增大,促进作用减弱甚至表现出抑制作用;而低浓度的2,4-二叔丁基苯酚降低了叶片丙二醛(MDA)含量和细胞相对电导率,且随浓度增加作用强度增大;浓度为0.1 mmol·L-1时,抑制作用最强.综合分析表明,2,4-二叔丁基苯酚对番茄植株及叶霉病具有调控作用,可以确定0.1mmol·L-1为田间施加最适浓度.  相似文献

利用水培法研究了Cd胁迫对狭叶香蒲(Typha angustifolia L.)某些生理指标的影响。结果表明,Cd浓度在0.4和1.0 mg.L-1时,狭叶香蒲的叶绿素含量呈上升趋势;Cd浓度高于1.0 mg.L-1时,叶绿素含量有所下降。当Cd浓度达1.5 mg.L-1时,狭叶香蒲的根系活力明显降低。随Cd浓度的增加,超氧化物歧化酶(SOD)和过氧化物酶(POD)活性基本呈现低浓度时上升高浓度时下降的变化趋势;丙二醛(MDA)含量则呈上升趋势。  相似文献

以小白菜品种'秦白2号'为材料,采用盆栽试验研究了不同浓度亚硒酸钠[Se(IV)]和硒酸钠[Se(VI)]胁迫对小白菜生长生理特性的影响及其生理机制,为土壤硒污染修复及其合理开发利用提供理论依据.结果表明,Se(IV)≤10.0 mg·kg-1时,小白菜的叶长、叶宽显著下降,而生物量没有受到显著影响;Se(VI)≤1.0 mg·kg-1时,叶长、叶宽、生物量没有显著变化;更高浓度处理时,叶长、叶宽、生物量均随外源Se(IV)和Se(VI)处理浓度的增大而急速下降.Se(IV)≤40.0 mg·kg-1和Se(VI)≤20.0 mg·kg-1处理均对小白菜叶片叶绿素含量无显著影响,但更高浓度外源Se(IV)和Se(VI)却显著抑制了叶绿素合成.低浓度外源Se(IV)和Se(VI)均使小白菜叶片谷胱甘肽过氧化物酶(GSH-Px)活性上升,膜质过氧化物(MDA)含量下降,对超氧化物歧化酶(SOD)活性、过氧化氢酶(CAT)活性及脯氨酸含量无显著影响;高浓度硒使MDA含量、脯氨酸含量及SOD活性上升,而使GSH-Px活性和CAT活性下降;外源Se(IV)和Se(VI)均使过氧化物酶(POD)活性降低.研究发现,低浓度外源Se(IV)和Se(VI)均提高了小白菜的抗氧化作用,从而促进小白菜叶片叶绿素的合成和生长,高浓度时则相反;低浓度硒的抗氧化作用和高浓度硒的过氧化作用均以Se(VI)大于Se(IV).说明硒酸钠的有效性和毒害作用均大于亚硒酸钠.  相似文献

研究了在BG11液体培养基中添加0.1~1.5 mg·L-1 IAA对微藻TH6(Oedocladium sp. )生长和脂肪酸含量的影响.结果表明,随培养时间的延长,微藻TH6的生长量逐渐增加;与对照相比,添加不同浓度IAA对微藻TH6的生长均有不同程度的促进作用,其中添加1.0 mg·L-1 IAA对微藻TH6生长的促进效果最佳,至培养第33天,微藻TH6的生长量比对照提高了44.34%.在培养基中添加IAA均能不同程度提高微藻TH6的总脂肪酸含量,当IAA 浓度为0.1、0.5、1.0和1.5 mg·L-1时,总脂肪酸含量分别是对照的2.09、2.13、2.41和1.73倍.IAA对微藻TH6中软脂酸、硬脂酸、亚油酸和油酸含量的影响作用不同.添加不同浓度IAA均能不同程度提高软脂酸的含量,当IAA浓度为1.0 mg·L-1时,软脂酸的相对含量最高,达到了28.62%;较高浓度的IAA(1.0和1.5 mg·L-1)能促进硬脂酸含量的提高, 低浓度IAA(0.1和0.5 mg·L-1)使硬脂酸含量降低;当IAA浓度为0.1~1.0 mg·L-1时,亚油酸含量较对照有不同程度提高;IAA对油酸含量的影响作用不明显.研究结果显示,含有1.0 mg·L-1 IAA的BG11液体培养基为微藻TH6的最佳液体培养基.在这一培养基中,微藻TH6的总脂肪酸含量最高,饱和及不饱和脂肪酸含量也较高,且微藻TH6生长量最大,可作为生物柴油资源进一步研究和开发.  相似文献

N-Acetylglucosamine-6-sulfate sulfatase activity was assayed by incubation of the radiolabeled monosaccharide N-acetylglucosamine [1-14C]6-sulfate (GlcNAc6S) with homogenates of leukocytes and cultured skin fibroblasts and concentrates of urine derived from normal individuals, patients affected with N-acetylglucosamine-6-sulfate sulfatase deficiency (Sanfilippo D syndrome, mucopolysaccharidosis type IIID), and patients affected with other mucopolysaccharidoses. The assay clearly distinguished affected homozygotes from normal controls and other mucopolysaccharidosis types. The level of enzymatic activity toward GlcNAc6S was compared with that toward a sulfated disaccharide and a sulfated trisaccharide prepared from heparin. The disaccharide was desulfated at the same rate as the monosaccharide and the trisaccharide at 30 times that of the monosaccharide. Sulfatase activity toward glucose 6-sulfate and N-acetylmannosamine 6-sulfate was not detected. Sulfatase activity in fibroblast homogenates with GlcNAc6S exhibited a pH optimum at pH 6.5, an apparent Km of 330 mumol/liter, and inhibition by both sulfate and phosphate ions. The use of radiolabeled GlcNAc6S substrate for the assay of N-acetylglucosamine-6-sulfate sulfatase in leukocytes and skin fibroblasts for the routine enzymatic detection of the Sanfilippo D syndrome is recommended.  相似文献

在极大螺旋藻(S.maxima)和钝顶螺旋藻(S.platensis)不同生长阶段进行硒胁迫处理,分别从接种后第1d至第5d开始添加硒,并不断增加硒含量,至第7d使硒的累计添加量为1000mg·L-1,形成5种不同硒胁迫(硒胁迫Ⅰ~Ⅴ),观察各种硒胁迫下螺旋藻的生物量及对无机硒的生物有机化的影响。结果表明:硒胁迫Ⅰ~Ⅳ对两种螺旋藻的生长影响不明显,而硒胁迫Ⅴ对螺旋藻生长有明显促进作用;藻体含硒总量和螺旋藻对无机硒的有机化率按硒胁迫Ⅰ~Ⅴ依次增加。首次提出硒胁迫强度概念,并用此较好地解释了有关实验结果。  相似文献

There is a U-shaped dose-response between selenium (Se) status and health outcomes, but underlying metabolic processes are unclear. This study aims to identify candidate proteins in liver regulated by dietary Se, ranging from deficiency to toxic. Male rats (n=4) were fed graded Se concentrations as selenite for 28 days. Bulk Se analysis was performed by ICP-MS on both soluble and insoluble fractions. Soluble fraction samples were chromatographically separated for identification of selenocompounds by SEC-ICP-MS and protein quantification by LC-MS/MS. Bioinformatics analysis compared low-Se (0 and 0.08 µg Se g−1) and high-Se (0.8, 2 and 5 µg Se g−1) with adequate-Se (0.24 µg Se g−1) diets. Major breakpoints for Se were seen at 0.8 and 2 µg Se g−1 in the insoluble and soluble fractions, respectively. Glutathione peroxidase 1 protein abundance reached a plateau at ≥0.08 µg Se g−1diet; Se bound to selenium binding protein 2 was observed with 2 and 5 µg Se g−1 Se. The extreme diets presented the highest number of differentially expressed (P value <0.05, FC ≥1.2) proteins in comparison to the adequate-Se diet (0 µg Se g−1: 45 proteins; 5 µg Se g−1: 59 proteins); 13 proteins were commonly affected in 0 and 5 µg Se g−1 treatments. Network analysis revealed that the metabolism of glutathione, xenobiotics and amino acids were enriched in both 0 and 5 µg Se g−1 diets, indicating a U-shape effect of Se. This similarity is likely due to down-stream effects of lack of essential selenoproteins in Se deficiency and due to toxic effects of Se that exceeds the capacity to cope with excess Se.  相似文献

The distribution of selenate and selenite selenium in C57L/J mice during the progression of BW7756 murine hepatoma was investigated using intra-ocular injection with the oxyanions labeled with 75Se radioisotope. Comparison is made with the normal distribution of selenium studied by the RIXRF method. The trace elemental profiles, TEP, for the two oxidation states are compared in healthy and disease states. It has been found that the presence of tumor significantly changes the level of tracer in various uninvolved organs. These changes are prominent in the early growth phase of the tumor. The two oxidation states show differences in the TEP for kidney, spleen, stomach and testes.  相似文献

A variety of plant species are being considered for the phytoremediation of selenium (Se) contaminated soils in agricultural regions of central California. Use of this plant-based technology may also attract a wide range of insects to these Se-accumulating plants. The first field study surveyed the diversity of insects attracted to tall fescue, birdsfoot trefoil, kenaf, and Indian mustard. Over 7500 specimens were collected by a sweep net collection technique for one complete growing season. Most of the 84 families identified were associated with beneficial insects, although pestiferous insects, for example, thrips, aphids, lygus, and leafhoppers, were also found. In the second study the bioaccumulation of Se in the cabbage looper [Trichoplusia ni (Hübner)] was investigated on Indian mustard grown in Se-rich water culture solution. Neonate larvae were transferred to plants and fed on Se-treated and no Se treated plants (controls) for 14 days, respectively. Pupae were collected from each treatment and incubated until adult insects emerged. Almost 50% fewer pupae were collected from Se-treated plants compared with “controls”, resulting in fewer adult insects. Selenium concentrations were as high as 3173 μg Se kg-1 DW in adult insects hatched from Se-treated plants compared with <5 μg Se kg-1 DW in insects from “controls”. Based on both studies, we concluded that insect diversity should be determined and insects monitored for bioaccumulation of Se on phytoremediation sites in agricultural regions.  相似文献

Incorporation of intravenously injected 75Se selenomethionine into platelets has been found to vary with alterations in the rate of platelet production. It appears to label newly produced platelets during their formation in megakaryocytes and provides a method by which thrombopoiesis may be studied in vivo. The purpose of the present study was to measure the pattern of uptake of75Se into proteins in normal rat lenses and into the proteins of lenses with selenite-induced cataract. Ten-day-old suckling rats received a single injection of75Se with or without a cataractous dose of cold carrier sodium selenite. Four days after injection, the proteins from excised lenses were counted for75Se radioactivity and subjected to gel permeation chromatography, amino acid analyses, and mass spectrometry. All three soluble crystallin lens proteins took up75Se in both normal and cataractous lenses. However, cataractous lenses did not take up75Se into a soluble protein in which major quantities of75Se were taken up in normal rats. Futhermore,75Se in the gamma-crystallins was associated with an unusual acidic amino acid. It was concluded that selenium metabolism by lens proteins may be unusual compared to other soft tissues.  相似文献
