The ultrastructure and distribution of gonial and somatic cells in the ovary of Dermatobia hominis was studied during the 3rd larval instar. In larvae weighing between 400 and 500 mg, the ovary is partially divided into basal and apical regions by oblong somatic cells that penetrate from the periphery; these cells show ovoid nucleus and cytoplasm full of microtubules. In both regions, gonial cells with regular outlines, large nucleus and low electron-density cytoplasm are scattered among the interstitial somatic cells. These later cells have small nucleus and electrondense cytoplasm. Clear somatic cells with small nucleus and cytoplasm of very low electron-density are restrict to the apical region of the gonad. Degenerating interstitial somatic cells are seen in the basal portion close to the ovary peduncle. During all this larval period the morphological features of the ovary remain almost the same. At the end of the period there is a gradual deposition of glycogen in the cytoplasm of the somatic cells, increase in the number and density of their mitochondria plus nuclear modification as membrane wrinkling and chromatin condensation in masses.  相似文献

The gonads of eight gobioid species were examined histologically, including Pandaka pygmaea and Schindleria praematura , in order to investigate the manifestations of miniaturization and paedomorphosis in the gonads. Rearrangements of reproductive structures were found only in S. praematura , and included only the gonad tissues, not the gametes. Both sexes of S. praematura maintain basic germinal components: the spermatocyst composed of Sertoli cells and spermatogonia in the testis and the follicle and oocyte complex in the ovary. The stromal ovarian tissue and testicular interstitial tissue, however, is reduced compared to other gobies, and the number and location of gonial cells is restricted. This sequestered pattern of gonial cells is known as a restricted spermatogonial type in the testis, and has been reported only in atherinomorphs. The restriction of gonial cells in the ovary is extremely rare among teleosts, known only from one other species. These restricted gonial patterns in S. praematura are probably related to the overall reduction of morphological complexity in this genus, due to its extreme paedomorphosis.  相似文献

Two conditional lethals which change the developmental fate of gonial cells in male Drosophila melanogaster have been isolated. 1(1)63 arrests mitoses in gonial cysts (and other tissues) before the four division cycles have been completed, but allows meiotic divisions and normal spermatid differentiation to proceed. Gonial divisions in testes of 1(1)55, on the other hand, continue well after the usual four divisions, and as a result, cysts with hundreds of cells, many of which later degenerate, are formed. It is suggested that four gonial divisions are not a prerequisite genetically and metabolically for spermatocyte differentiation and that the activation of the spermatocyte program is not predetermined but rather requires the arrest of the gonial program. Other implications of the findings for our understanding of the genetic control of spermiogenesis are briefly discussed.  相似文献

Recombination-defective female meiotic mutants representing 7 loci in Drosophila melanogaster have been examined for effects on gonial recombination in males. These loci were chosen for study because they represent a broad range of the known types of defects in processes necessary for meiotic recombination and somatic chromosome stability. Alleles at 6 of the loci studied did not increase the frequency of gonial recombination in males, whereas a mutant at one locus was associated with an increase (about 10-fold) in gonial recombination. These results suggest that the defects in chromosomal metabolism caused by these recombination, and in some cases repair, defective mutants are distinct from those of the male-recombination promoting elements (Mr) recently isolated from many natural populations. Analysis of the spontaneous events detected in this study showed that a third to a half of the events detected are actually of mutational rather than recombinational origin.  相似文献

Efferent duct (ED) differentiation was examined histologically during female-to-male sex change in the honeycomb grouper Epinephelus merra . During natural sex change, ED differentiation began with the appearance of slit-like structures between the stromal tissue and the tunica ovary and small oval-shaped spaces within the wall of the ovarian cavity, accompanied by oocyte degeneration and the initiation of spermatogenesis, i.e. the early transitional phase. In the late transitional phase, ED structure formation expanded and further ED differentiation occurred, including the rapid multiplication of spermatogonial germ cells. In sex-changed males, the slit-like structures increased in size, fused with each other and finally formed a well-developed ED. The oval-shaped spaces also increased in size and fused to form an ED. In contrast, during artificial sex change, induced by aromatase inhibitor (AI, 1 mg kg−1), ED differentiation in E. merra was first observed as the appearance of slit-like structures and small oval-shaped spaces in the restructuring gonads in the third week after AI treatment. These were accompanied by oocyte degeneration and the proliferation of gonial germ cells into spermatogonia. In the fifth week, the rapid multiplication of spermatogonial germ cells, increases in 11-ketotestosterone (11-KT), and further differentiation of EDs were observed. Sex-changed males had testes containing sperm in the completely differentiated EDs; the significantly highest levels of 11-KT were observed in the sixth week. Simultaneous increases in 11-KT and initiation of ED differentiation were observed, suggesting a role of 11-KT in ED differentiation during sex change. There were no basic differences in the mechanisms of natural and artificially induced ED differentiation. Two types of structure led to the formation of EDs in two different areas of the newly formed testis during sex change.  相似文献

Biometrical studies on ancient skeletal series and comparison with modern people by using radiological methods are quite limited in Turkey. Previous studies showed that measurements obtained from orthopantomographs are highly correlated with the actual size of the bones. The aim of the present study is to determine the possible change in gonial angle over time in ancient Anatolian populations with the present. Also an aim was attempted to demonstrate the symmetry of the gonial angle in the jaws and the sexual dimorphism. Gonial angle values (right and left) were taken from 267 Turkish adults with no craniomandibular disorders, orthodontic history or treatment by using panoramic radiographs. Data of the past populations were collected from previous studies. Comparison between right and left sides and the sexual differences were tested by paired student t-test and discriminant analyses were conducted. The results showed that there were no significant differences between the right and left gonial angles of the individuals but there was a significant difference at the left gonial angle between sexes (p < 0.01). Furthermore, no statistically significant difference was found for the gonial angle between the selected past populations with the present sample.  相似文献

Comparative study of ovary development and oogenesis in the dipterans revealed significant differences between the Nematocera (lower dipterans, midges) and the Brachycera (true flies). The occurrence of these differences emphasizes well the phylogenetic division of the Diptera into these major subgroups. Basic discrepancies were found in the course of ovary development and in the mode of follicular cell differentiation. In contrast to more advanced flies, in midges the initial stages of germ cell differentiation, i.e. divisions of gonial cells, germ cell cluster formation and diversification of cystocytes within clusters take place exclusively in the larval and early pupal stages. Moreover, the formation of cystocyte clusters precedes that of ovarioles. Differences in the behaviour of some follicular cells found between the ovarian follicles of midges and advanced flies suggest that both major dipteran subgroups may differ in the scenario and/or the mechanisms of terminal signalling leading to the determination of the anteriormost part of the body.  相似文献

Prostaglandin F-2alpha (1.5 mg over 10 h) was infused into the anterior uterine vein of pigs on Days 6, 8, 10, 12, 14 and 15 of the oestrous cycle. At each stage of the cycle PGF-2alpha suppressed luteal function although the fall in progesterone secretion was much greater and statistically significant when the infusion was performed on Days 12, 14 and 15 of the cycle than on Days 6, 8 and 10. The concentrations of cAMP was depressed on Days 15 and 17 and fatty degeneration of luteal cells on Days 6--8 or 14 was more pronounced in the ovary ipsilateral to the PGF-2alpha infusion than in the contralateral ovary. The results are compatible with the local perfusion of PGF-2alpha from the anterior uterine vein to the ipsilateral ovary, but a systemic effect was also apparent.  相似文献

Using a 2-day brood pattern, the effect of 5-bromodeoxyuridine (BUdR) or 5-bromodeoxycytidine (BCdR) pre-treatment on the radiation-induced yield of sex-linked recessive lethals and translocations was studied in the spermatocytes and late gonial cells (p.i. DNA synthesis cells) of D. melanogaster. The p.i. DNA synthesis cells were irradiated (I.2 kR γ-radiation) in the pre-meiotic or post-meiotic stage. Irradiation of p.i. DNA synthesis cells in the pre-meiotic stage resulted in enhanced lethal frequency with BUdR (3.0%) and BCdR (2.9%) over the other pre-treatment conditions: saline (S), thymidine (TdR) and deoxycitydine (CdR) in the spermatocytes but not in the late gonial cells. The radiosensitizing property was evident with BCdR even when the p.i. DNA synthesis cells were irradiated in the post-meiotic stage; but not with BUdR pre-treatment. Probable reasons for the contradicting results reported in the literature were discussed.  相似文献

Pairing between two nonhomologous chromosomes, one a free X-duplication and the other a free fourth chromosome, has been observed cytologically with high frequencies in the oögonial cells of Drosophila melanogaster. The frequencies of nonhomologous pairing ranged from 27 to 47% and showed a positive correlation with the similarity in size between the two participating nonhomologues. Partial homology increased pairing frequency between nonhomologues in the oögonial cell, in contrast to the behavior of the same nonhomologues at distributive pairing in the oöcyte, where pairing is strictly size-dependent. Pairing between homologues in the same oögonial cells occurred at a frequency of only 71% and was higher for the autosomes (73%) than for the sex chromosomes (66%). An increased frequency of homologous pairing was found for older gonial cysts (4-cell, 72.0% ; 8-cell, 76.1%) as compared with younger cysts (1-cell, 59.1% ; 2-cell, 53.1%).Research sponsored by the U.S. Atomic Energy Commission under contract with the Union Carbide Corporation.  相似文献

The X chromosome can be identified with the light microscope throughout all stages of the gonial cell cycle (including interphase) in the grasshopper Brachystola magna. At gonial mitotic stages the X chromosome gives the appearance of being undercondensed or negatively heteropycnotic. At interphase the X projects out from the body of the nucleus. — Examination with the electron microscope reveals that the X is compartmentalized at least two gonial cell cycles prior to the entry of the cells into meiotic prophase. The membrane layers that envelope the X chromatin at interphase remain associated with the X chromosome throughout gonial mitotic stages providing the ultrastructural basis for the apparent negative heteropycnosis observed with the light microscope. — The X chromosome is inactive in RNA synthesis during gonial mitotic stages but is hyperactive in RNA synthesis when compared to autosomes at gonial interphase. — X chromosome condensation which reaches its maximum at premieotic interphase is initiated at or prior to the pre-pentultimate gonial division.  相似文献

The effects of aminoglutethimide phosphate (AGP) on ovulation, ovum maturation, fertilizability, and steroid production were studied with the use of an isolated perfused rabbit ovary preparation. AGP (10(-3) or 10(-4) M) was added to the perfusate of one ovary. The contralateral control ovary was perfused in medium alone. Thirty minutes later human chorionic gonadotropin (hCG) (50 IU) was added to the perfusate of all ovaries. No difference was observed in time of ovulation or ovulatory efficiency between controls and AGP-treated ovaries. The degree of ovum maturity and degeneration was also comparable in the two groups. Progesterone and estradiol production were significantly reduced by AGP treatment. A second experiment examined fertilizability of ova ovulated in vitro after perfusion with 10(-3) M AGP. AGP significantly reduced the rate of normal fertilization as observed 12 h after insemination. The percentage of inseminated ova with evidence of degeneration was greater in ova from AGP-treated ovaries than in those from controls, however, this difference was not significant. The study indicates that AGP affects neither hCG-induced ovulation nor meiotic resumption; however, fertilizability of ova from ovaries treated with AGP is impaired. These data suggest that the intrafollicular steroid environment may participate in cytoplasmic maturation of ovulated ova.  相似文献

Summary Synergid degeneration was examined in the isolated embryo sac and egg apparatus of Nicotiana tabacum using quantitative cytology, fluorochromatic reaction (FCR) and chlorotetracycline (CTC). Most synergid degeneration occurs after pollen tubes (PT) arrive in the ovarian chamber between 42 and 48 h after pollination; synergid degeneration was precluded when PT were prevented from entering the ovary by stylar excision indicating that the signal that triggers synergid degeneration travels only relatively short distances in this plant. There was no evidence for any preferentiality between right or left synergids with regard to cell size or degeneration pattern. FCR staining confirms that synergid degeneration involves the loss of membrane integrity and is a reliable indicator of the onset of degeneration. CTC labeling of the degenerated synergid reveals that a concentrated reserve of membrane-bound calcium is present in the receptive synergid, possibly aiding in the attraction, arrest and discharge of the PT, releasing the sperms into the receptive ES.  相似文献

Effects of exogenous estradiol and testosterone on the gonad and pituitary were examined in the hagfish. Estradiol caused degeneration of the ovary. Testosterone had no effect on the ovary. Neither steroid had a significant effect on the testis. Neither estradiol nor testosterone provoked a conspicuous change in adenohypophysial cells. Therefore, a feedback mechanism between gonadal steroids and gonadotropin either does not exist or plays a very minor role. The degeneration of the ovary may be due to a pharmacological effect of estradiol.  相似文献

Hexachlorobenzene (HCB) is a fungicide and a pollutant of increasing concern in industrialized regions of the world. Reproductive failure is one of the effects of HCB upon mammals. Alteration of cell shape in the ovary surface epithelium (OSE) of Cynomolgus monkeys following oral administration of HCB was observed in this study. At the lowest dose used, 0.1 mg/kg body weight, a dosage that is associated with follicular degeneration, HCB caused quantifiable changes in length-to-width ratios of OSE. Measurement of cell shape by light microscopy offers a reliable indication of OSE changes induced by HCB.  相似文献

Histological changes in the perch ovary during development are described and related to changes in gonad weight and to a macroscopic scale of maturity stages after Kesteven (1960). A single organ formed by the fusion of two primordial ovaries, a central internal oviduct, and an extremely thick chorion surrounding oocytes are characteristic features of the perch ovary. Resting oocytes can be differentiated from developing oocytes which are larger and possess a granulosa layer which permits formation of the chorion and yolk acquisition. In the maturing ovary, oocyte diameter increases rapidly and the chorion, which possesses four layers, the tunica propria, two middle zones forming a fine network, and the zona radiata, also expands. The spent ovary is disorganized with follicles and tunica wall contracted, and a number of residual oocytes may be apparent. There was no evidence of pre-ovulatory degeneration although autolysis of a small number of residual oocytes was observed. The gonadosomatic index was highest for males in September and at a maximum in females immediately prior to spawning. After spawning the index fell rapidly with lowest values recorded in June and July. Variations in the gonadosomatic index are related to developmental stages in females.  相似文献

This study was designed to classify and differentiate the population of oocytes released from the mouse ovary by mechanical means. Liberated oocytes were classified on the basis of their number, size and by the nature of the attachment of follicle cells to these oocytes. Microscopical examination of oocytes mechanically released from the ovaries revealed three distinct morphological classifications of oocytes: (1) those completely devoid of follicle cells, (2) those encased in follicle cells and (3) those in the process of degeneration. Ooxytes ranged in size from approximatley 30mu to 119mu. Those oocytes surrounded by follicle cells could be further subdivided into two groups depending on whether the follicle cells could be mechanically removed from the oocyte. These data demonstrate that the adult ovary contains a variety of classes of oocytes which differ in size and in the extent to which the follicle cells are attached to the oocytes. It is suggested that the metabolic activities and meiotic potnetials of the various oocyte populations differ and as a result, care should be employed to insure a uniform population of oocytes when conducting further studies sith mechanically liberated oocytes.  相似文献

The effect of chemosterilant, apholate, on-oogenesis has been studied in Aedes aegypti. Treatment of larvae to 20 ppm of the chemical induced ultrastural changes in the presumptive and primary follicles of the adult ovary. These changes comprised condensation of chromatin, disruption of nuclear envelope and extensive degeneration as evidenced by numerous myelin figures and residual bodies. In some primary follicles, where cellular degeneration was restricted to epithelial cells, no arrest in development was observed. However, in comparison with controls, these follicles also were retarded. Larval treatment with 30 ppm apholate completely supressed ovariolar development. High incidence of autophagy was observed in tissues at both dose levels.  相似文献

The honeycomb grouper shows protogynous hermaphroditism. The endocrine mechanisms involved in gonadal restructuring throughout protogynous sex change are largely unknown. In the present study, we investigated changes in the gonadal structures and levels of serum sex steroid hormones during female to male sex change in the honeycomb grouper. On the basis of histological changes, entire process of sex change was assigned into four developmental phases: female, early transition (ET), late transition (LT), and male phase. At the female phase, the oocytes of several developmental stages were observed including gonial germ cells in the periphery of ovigerous lamellae. At the beginning of ET phase, perinucleolar and previtellogenic oocytes began degenerating, followed by proliferation of spermatogonia toward the center of lamella. The LT phase was characterized by further degeneration of oocytes and rapid proliferation of spermatogenic germ cells throughout the gonad. At the male phase, no ovarian cells were observed and testis had germ cells undergoing active spermatogenesis. Serum levels of estradiol-17beta (E2) were high in females in the breeding season, but low in the non-breeding female, transitional and male phase, and those of 11-ketotestosterone (11-KT) and testosterone (T) were low in females and gradually increased in the transitional and male phase. The present results suggest that low serum E2 levels and degeneration of oocytes accompanied by concomitant increase in the 11-KT levels and proliferation of spermatogenic germ cells are probably the events mediating protogynous sex change in the honeycomb grouper.  相似文献
