A Bombyx mori (B. mori) cDNA was isolated from silkworm pupae cDNA library encoding a homologue of translationally controlled tumor protein (BmTCTPk). BmTCTPk was expressed in E. coli; SDS–PAGE and Western blot showed the molecular weight of recombinant and native BmTCTPk is approximately 28 and 25 kDa, respectively; they are larger than the theoretical molecular weight. Immunohistochemical studies showed that BmTCTPk is uniformly distributed throughout the cytoplasm of BmN cells. In silkworm pupae, BmTCTPk is expressed in the midgut wall, the midgut cavity, and some fat body tissues lying between the midgut wall and body wall. Western blot and ELISAs performed on total protein extracts isolated from silkworm pupae at different development stages showed that, although BmTCTPk is expressed during all pupae stages, its expression level increases dramatically during late pupae stages, suggesting that BmTCTPk may play an important role during the developmental transition from pupa to imago.  相似文献

We have cloned a homologue of the translationally controlled tumor protein (TCTP) from the buccal gland of Lampetra japonica according to information from a cDNA library and primary analysis of expressed sequence tags. Sequence analysis of L. japonica TCTP showed that it had two signature regions of high sequence homology termed TCTP-1 and TCTP-2, respectively. TCTP is highly conserved in evolution. It showed more than 40% identification similarities with parasite TCTPs that had effect on immune responses of host. Phylogeny of 31 TCTP sequences showed that lamprey was closer to jawed vertebrates than to Amphioxus and was a sister group of gnathostomes. TCTP gene from L. japonica was expressed in a pET23b vector and purified by using His Bind affinity chromatography. Polyclonal antibody to recombinant protein was generated in New Zealand Rabbit. Immunoblot analysis to localize the recombinant protein in buccal gland secretion proves that recombinant TCTP is a secretion protein, which may be secreted through a non-classical secretion pathway. A characterization study shows that recombinant TCTP has histamine-releasing function in vitro. It mediated histamine release from rat basophilic leukemia (RBL-2H3) cells. TCTP links both the innate and the adaptive immune responses by modulating the secretion of cytokines from mast cells, basophils, eosinophils, and T and B lymphocytes. These may indicate a potential role of TCTP in the inflammatory process and immune regulation between L. japonica and host.  相似文献

A cDNA encoding a putative translationally controlled tumor protein (TCTP) was isolated from a cDNA library made with mRNA isolated from red ripe strawberry fruits. This protein is highly conserved in all species analyzed. Expression of strawberry TCTP increased along the ripening of strawberry fruits, and is constitutively expressed in vegetative tissues. Background

Human N-Myc downstream regulated gene2 (NDRG2), a novel gene has been cloned and shown to be related to a number of cellular processes, including proliferation, differentiation, stress, and apoptosis. NDRG2 has also been linked to age-related Alzheimer''s disease. Since the role of this gene in senescence is limited, we have investigated the potential role of NDRG2 in human lens epithelial cells (HLECs), a paradigm implicated in age-related cataract.

Methodology/Principal Findings

Cultured HLECs (SRA01/04) were subjected to prolonged exposure to low dose of H2O2 to simulate senescence. After being exposed to 50 µM H2O2 for 2 weeks, HLECs senescent-morphological changes appeared, cell viability decreased dramatically, cell proliferation reduced from 37.4% to 16.1%, and senescence-associated β-galactosidase activity increased from 0 to 90.3%. Ndrg2 protein expression was also significantly increased in these senescent cells. To induce overexpression of NDRG2, SRA01/04 cells were infected with the adenoviral vector of NDRG2. In these cells, overexpression of NDRG2 resulted in a fibroblast-like appearance and the cell viability decreased about 20%. In addition, the NDRG2-overexpression cells demonstrated 20% lower viability when exposed to 50–200 µM H2O2 for acute oxidative stress. Furthermore, the expression of NDRG2 from age-related cataracts was up-regulated 2-fold at both mRNA and protein levels compared with the clear lenses.

Conclusions/Significance

NDRG2 is up regulated not only in the ageing process of HLECs in vitro but also in the cells from human age-related cortical cataract in vivo. Up-regulation of NDRG2 induces cell morphological changes, reduces cell viability, and especially lowers cellular resistance to oxidative stress. NDRG2-mediated affects in HLECs may associate with age-related cataract formation.  相似文献

Translationally controlled tumor protein (TCTP) expression is suppressed during cancer cell reversion to a non-malignant phenotype. We identified a primary sequence of TCTP with homology to ADF/cofilin. We confirm that a synthetic peptide corresponding to this sequence binds specifically to actin and is displaced from actin by cofilin. TCTP peptide has higher affinity for G-actin than F-actin and does not block actin-filament depolymerization by cofilin. These results suggest that TCTP may channel active cofilin to F-actin, enhancing the cofilin-activity cycle in invasive tumor cells. Loss of TCTP may result in sequestration of active cofilin by a monomeric pool of actin.  相似文献

Translationally controlled tumor protein (TCTP), also termed P23 in human, belongs to a family of calcium- and tubulin-binding proteins, and it is generally regarded as a growth-regulating protein. Recently, Arabidopsis TCTP (AtTCTP) has been reported to function as an important growth regulator in plants. On the other hand, plant TCTP has been suggested to be involved in abiotic stress signaling such as aluminum, salt, and water deficit by a number of microarray or proteomic analyses. In this study, the biological functions of AtTCTP were investigated by using transgenic Arabidopsis plants overexpressing AtTCTP. Interestingly, AtTCTP overexpression enhanced drought tolerance in plants. The expression analysis showed that AtTCTP was expressed in guard cells as well as in actively growing tissues. Physiological studies of the overexpression lines showed increased ABA- and calcium-induced stomatal closure ratios and faster stomatal closing responses to ABA. Furthermore, in vitro protein-protein interaction analysis confirmed the interaction between AtTCTP and microtubules, and microtubule cosedimentation assays revealed that the microtubule binding of AtTCTP increased after calcium treatment. These results demonstrate that the overexpression of AtTCTP confers drought tolerance to plants by rapid ABA-mediated stomatal closure via the interaction with microtubules in which calcium binding enhances the interaction. Collectively, the present results suggest that the plant TCTP has molecular properties similar to animal TCTPs, such as tubulin- and calcium-binding, and that it functions in ABA-mediated stomatal movement, in addition to regulating the growth of plants.  相似文献

Summary Although several proteases have been identified in homogenates of cultured epithelial cells of the eye lens and in lens tissues, there is little information regarding intracellular protein degradation in intact lens cells in vitro. Cultured lens cells may be useful in the study of intracellular protein degradation in the lens, a tissue with a wide range of protein half-lives. This is of interest because alterations in protein turnover in the lens have been implicated in cataract formation. This study examines intracellular protein degradation in cultured bovine lens epithelial cells (BLEC). Cell cultures were incubated with radiolabeled leucine to label intracellular proteins. Protein degradation was measured by monitoring the release of trichloroacetic-acid-soluble radioactivity into the culture medium. The average half-life of long-lived proteins (half-life >50 h) was typically about 57 h in serum-supplemented medium. Average rates of degradation of long-lived proteins increased by up to 73% when fetal bovine serum was withdrawn from the culture medium. Serum had no effect on the degradation of short-lived proteins (half-life <10 h). Degradation of long-lived proteins in the presence and absence of serum was further studied in cultured BLEC from population doubling level (PDL) 2 to 43. Average half-life of proteins in serum-supplemented medium was 52 to 58 h and did not vary significantly as a function of PDL. Degradation rates in serum-free medium increased approximately twofold up to PDL 7, but returned by PDL 25 to original levels, which were maintained through PDL 43. Cotton aphid (Aphis gossypii Glover) is one of the most important economic pests in the world. Long-term unreasonable usage of insecticides has made cotton aphid developing insecticide-resistance, which frequently leads to serious occurrences of cotton aphid in many regions. It is regarded effective and environmentally friendly to control aphids through utilizing plant resistance. In this study, a translationally controlled tumor protein gene, GhTCTP1, was isolated in cotton. It belongs to TCTP subfamily and encodes a protein of 168 amino acids. GhTCTP1 expression was suppressed in cotton plants under cotton aphid attack, but its expression level was up-regulated in the wounded cotton leaves. The choice test and no-choice test demonstrated that overexpression of GhTCTP1 in Arabidopsis enhanced plant resistance to green peach aphid (Myzus persicae). Quantitative RT-PCR analysis revealed that the defense response genes related to salicylic acid signaling pathway were activated in the GhTCTP1 overexpressing transgenic plants. Content of total amino acids was decreased, and phenylalanine ammonialyase activity was altered in leaves of the transgenic Arabidopsis plants, compared with those in wild type. Furthermore, the callose amount in transgenic Arabidopsis leaves was more than that of wild type. These data suggested that GhTCTP1 might be involved in regulation of plant tolerance to aphids, and can be potentially applied in improving aphid-resistance of crops by genetic manipulation.  相似文献

The expression of a nuclear (57 kDa) protein statin has been previously characterized as a specific marker of quiescent or senescent aging human fibroblasts in vitro. In these studies we have shown that the expression of statin is associated specifically with the postreplicative and terminally differentiating lens epithelial cell. By monitoring the synthesis of specific lens crystallin proteins, and the morphological and cellular changes associated with this differentiated system, we have demonstrated a close correlation between statin expression and cell commitment to the G0 nonreplicative cell cycle state.  相似文献

Translationally controlled tumor protein (TCTP) has both extra- and intracellular functions. Our group recently reported that TCTP interacts with Na,K-ATPase and suppresses its activity. Our studies led to the identification of sorting nexin 6 (SNX6) which binds with TCTP as a potential negative regulator of TCTP. SNX6 does not interact directly with any cytoplasmic domains of Na,K-ATPase. However, when overexpressed, it restores the Na,K-ATPase activity suppressed by TCTP. This was confirmed by measurements of purified plasma membrane Na,K-ATPase activity after incubation with recombinant TCTP and SNX6. SNX6 alone has no effect on Na,K-ATPase activity, but activates Na,K-ATPase via inhibition of TCTP. Inhibition of endogenous TCTP by the overexpression of SNX6 or knockdown of TCTP expression by siTCTP increased Na,K-ATPase activity above the basal level. The interaction between SNX6 and TCTP thus appears to regulate Na,K-ATPase activity.  相似文献

Schistosomiasis is a globally important helminthic disease of both humans and animals, and is the second most common parasitic disease after malaria. Although praziquantel is extensively used for treatment of parasitic diseases, drug resistance has been reported. Therefore, new drugs and effective vaccines are needed for continuous control of schistosomiasis. Eggs produced by schistosomes are responsible for the occurrence and spread of schistosomiasis. Revealing the reproductive mechanism of schistosomes will help to control this disease. In this study, the proteomic profiles of single-sex infected female worms and bisexual infected mature female worms of Schistosoma japonicum at 18, 21, 23 and 25 days p.i. were identified with isobaric tags for relative quantitation-coupled liquid chromatography–tandem mass spectrometry. Differentially expressed proteins were subsequently used for bioinformatic analysis. Six highly expressed differentially expressed proteins in mature female worms were selected and long-term interference with small interfering RNA (siRNA) was conducted to determine biological functions. SiRNA against S. japonicum translationally controlled tumour protein (SjTCTP) resulted in the most significant effect on the growth and development of MF worms. Sjtctp mRNA expression gradually increased over time with a high level of expression maintained at 25–42 days p.i., while levels were significantly higher in mature female worms than male and SF worms. The subsequent animal immune protection experiments showed that recombinant SjTCTP (rSjTCTP) reduced the number of adults by 44.7% (P < 0.01), average egg burden per gram of liver by 57.94% (P < 0.01), egg hatching rate by 47.57% (P < 0.01), and oviposition of individual females by 43.16%. rSjTCTP induced higher levels of serum IgG, IL-2, and IL-10 in mice. Collectively, these results show that SjTCTP is vital to reproduction of female worms and, thus, is a candidate antigen for immune protection.  相似文献

The accessibility of tryptophanyl fluorophores in crystalline proteins to water molecules was estimated by measuring the enhancement of the fluorescence of lens homogenates in 70% D2O as compared to 100% H2O. Assuming that two sorts of fluorophores exist in the proteins, one entirely accessible to H2O and D2O and the other--absolutely not, we have calculated the portion of either group in the protein fluorescence (alpha and 1-alpha, correspondingly). Measurement of murine lens homogenates fluorescence at different stages of radiation-induced cataract, initiated with total gamma irradiation in a dose 5 Gy have shown an increased accessibility of tryptophanyl for water with cataract development. At earlier stages of cataract (appearance of scattered dot opacities) the portion of water-accessible tryptophanyl increased from 0.14 to 0.18, i.e. by a factor of 1.3. The data obtained suggest that protein globules unfold in the coarse of cataract development.  相似文献
