Abstract

Major efforts are currently underway to dissect the phenotype–genotype relationship in plants and animals using existing populations. This method exploits historical recombinations accumulated in these populations. However, linkage disequilibrium sometimes extends over a relatively long distance, particularly in genomic regions containing polymorphisms that have been targets for selection. In this case, many genes in the region could be statistically associated with the trait shaped by the selected polymorphism. Statistical analyses could help in identifying the best candidate genes into such a region where an association is found. In a previous study, we proposed that a fragment of the PHYTOCHROME C gene (PHYC) is associated with flowering time and morphological variations in pearl millet. In the present study, we first performed linkage analyses using three pearl millet F2 families to confirm the presence of a QTL in the vicinity of PHYC. We then analyzed a wider genomic region of ~100 kb around PHYC to pinpoint the gene that best explains the association with the trait in this region. A panel of 90 pearl millet inbred lines was used to assess the association. We used a Markov chain Monte Carlo approach to compare 75 markers distributed along this 100-kb region. We found the best candidate markers on the PHYC gene. Signatures of selection in this region were assessed in an independent data set and pointed to the same gene. These results foster confidence in the likely role of PHYC in phenotypic variation and encourage the development of functional studies.  相似文献

Climate change and nitrogen deposition affect biodiversity and ecosystem functioning, but interactive effects of these global change drivers are poorly understood. We analysed single and interactive effects of nitrogen (N) fertilisation and drought on the growth performance of Calluna vulgaris. We measured biomass production and allocation, tissue nutrient (N, phosphorus (P) and carbon (C)) concentrations, N allocation patterns (using 15N tracer) and plant's water status (using δ 13C signatures) as response variables in a 2-year greenhouse experiment. N fertilisation increased biomass production and biomass shoot:root ratios. 15N allocation patterns indicated an increasing aboveground N allocation following N fertilisation. Tissue δ 13C signatures were higher in N-fertilised plants. Plant responses to drought were weak. We found strong antagonistic interaction effects of N fertilisation and drought for biomass production. δ 13C values peaked when N-fertilised plants were subjected to drought, indicating that N fertilisation increased the evaporative demands of Calluna plants, likely due to increased biomass shoot:root ratios, which in turn resulted in higher drought susceptibility. As an important consequence, even slight drought events may weaken the competitiveness of Calluna when interacting with enhanced airborne N loads. Single-factor studies, thus, need to be complemented by multi-factor analyses to assess conceivable impacts of co-occurring global change drivers.  相似文献

In the Sahel of Africa, farmers often modify their cultivation practices to adapt to environmental changes. How these changes shape the agro-biodiversity is a question of primary interest for the conservation of plant genetic resources. We addressed this question in a case study on pearl millet in south western Niger where farmers used to cultivate landraces with different cycle length in order to cope with rain uncertainty. Early and late landraces were previously grown on distant fields. Nowadays, mostly because of human population pressure and soil impoverishment, it happens that the two types of landraces are grown on adjacent fields, opening the question whether gene flow between them may occur. This question was tackled through a comparative study among contrasting situations pertaining to the spatial distribution of early and late landraces. Observations of flowering periods showed that pollen flow between the two landraces is possible and has a preferential direction from early to late populations.  相似文献

Summary The effects of-irradiation and changes in the macromolecular structure on the water proton resonance spectra observed in frozen and liquid solutions have been compared for the DNA and polynucleotide solutions, using H2O or mixed H2O/D2O solvents. Rheumatoid and psoriatic arthritis are chronic inflammatory diseases, with massive increase of cardiovascular events (CVE), and contribution of the cytokines TNF-α and IL-17. Chronic inflammation inside the joint membrane or synovium results from the activation of fibroblasts/synoviocytes, and leads to the release of cytokines from monocytes (Tumor Necrosis Factor or TNF) and from T lymphocytes (Interleukin-17 or IL-17). At the systemic level, the very same cytokines affect endothelial cells and vessel wall. We have previously shown [1], [2] that IL-17 and TNF-α, specifically when combined, increase procoagulation, decrease anticoagulation and increase platelet aggregation, leading to thrombosis. These results are the basis for the models of interactions between IL-17 and TNF, and genes expressed by activated endothelial cells. This work is devoted to mathematical modeling and numerical simulations of blood coagulation and clot growth under the influence of IL-17 and TNF-α. We show that they can provoke thrombosis, leading to the complete or partial occlusion of blood vessels. The regimes of blood coagulation and conditions of occlusion are investigated in numerical simulations and in approximate analytical models. The results of mathematical modeling allow us to predict thrombosis development for an individual patient.  相似文献

Among colonies with different foraging distances, central-place-foraging seabirds may change their foraging and reproductive efforts. We compared the body condition, meal frequency, and diving behavior of male and female Adélie penguins at two locations: Dumont d'Urville, where there was little sea ice and they foraged in open waters far from the colony; and Syowa, where there was heavy, fast sea ice and they foraged in ice cracks close to the colony. The parental mass decrease rate during the chick-rearing period was similar between colonies and between sexes. A large individual variation in meal frequency positively affected the brood growth rate, but daily underwater time did not. A weak but significant positive effect of body condition on brood growth rate was found only in males at Syowa. It was suggested that males work with better body condition than females. We propose the hypothesis that the regional difference in the distance to the feeding sites and the sex difference in body energy reserve might constrain the capacity to regulate reproductive effort.  相似文献

The autolysis of trypsin and α-chymotrypsin is accelerated in the presence of colloidal silica and glass surfaces. It is proposed that adsorption of the enzymes (favoured by electrostatic factors) results in a conformational change that renders the adsorbed enzyme more susceptible to proteolytic attack. Although the adsorbed enzymes are more susceptible to proteolysis, their activity towards low-molecular-weight substrates is not affected, indicating a relatively minor conformational change on adsorption. The rates of autolysis in solution (i.e. in `inert' vessels) are second-order for both trypsin and α -chymotrypsin, with rate constants of 13.0mol−1·dm3·s−1 for trypsin (in 50mm-NaCl at pH8.0 at 25°C) and 10.2mol−1·dm3·s−1 for α-chymotrypsin (in 0.1m-glycine at pH9.2 at 30°C). In glass vessels or in the presence of small areas of silica surface (as colloidal silica particles), the autolysis of both trypsin and α-chymotrypsin can show first-order kinetics. Under these conditions, saturation of the surface occurs and the fast surface proteolytic reaction controls the overall kinetic order. However, when greater areas of silica surface are present, saturation of the surface does not occur, and, since for a considerable portion of the adsorption isotherm the amount adsorbed is approximately proportional to the concentration in solution, second-order kinetics are again observed. A number of negatively charged macromolecules have been shown similarly to increase the rate of autolysis of trypsin: thus this effect, observed initially with glass and silica surfaces, is of more general occurrence when these enzymes adsorb on or interact with negatively charged surfaces and macromolecules. These observations explain the confusion in the literature with regard to the kinetics of autolysis of α-chymotrypsin, where first-order, second-order and intermediate kinetics have been reported. A further effect of glass surfaces and negatively charged macromolecules is to shift the pH–activity curve of trypsin to higher pH values, as a consequence of the effective decrease in pH in the `microenvironment' of the enzyme associated with the negatively charged surface or macromolecule.  相似文献

Background and aims

Soil carbon storage is an important component of global carbon cycling. Andean Andisols have high carbon content and are vulnerable to erosion because of agricultural intensification and deforestation. This study examines the effects of land use on erosion and soil carbon storage in the Río Chimbo watershed of Ecuador.

Methods

Soil carbon content, age, and erosion estimated from 137Cs inventories was measured along an elevational transect under annual cropping, natural forest, páramo, pasture, and tree plantations.

Results

Land use, particularly annual cropping, affected 137Cs levels in the upper soil layers, but did not have an impact on total carbon storage to a depth of 1 m. Relative erosion rates estimated from 137Cs inventories at sites under annual cropping averaged 27 t ha?1?y?1 over the erosion rate of non-cultivated sites. A linear relationship was observed between soil carbon age (determined by 14C levels) and 137Cs levels, where pasture sites had lower 137Cs and older carbon compared to natural forest sites.

Conclusions

The effects of land use on soil loss in the Río Chimbo watershed suggest a loss and/or removal of soil carbon, particularly under annual cropping.  相似文献

A selected series of racemic α-methylene-γ-butyrolactones (AMGBL) were synthesized via allylboration and screened against three human pancreatic cancer cell lines (Panc-1, MIA PaCa-2, and BxPC-3). This systematic study established a discernible relationship between the substitution pattern of AMGBL and their anti-proliferative activity. β,γ-diaryl-AMGBLs, particularly those with a trans-relationship exhibited higher potency than parthenolide and LC-1 against all three cell lines.  相似文献

This study examined the effects of estrogen (17β-estradiol) and testosterone on the growth of long bones in male and female mice, with and without gonadectomy. Weight and nose-to-tail length were determined at 3 weeks of age at time of gonadectomy, 7 days later at the onset of hormone therapy, and throughout the treatment period. Gonadectomized mice exhibited an initial weight gain during the pretreatment period but length was unaffected. Hormone treatment altered weight gain in surgical and intact animals in a gender- and hormone-dependent manner. Estradiol enhanced weight gain in intact mice, but inhibited weight gain in ovariectomized mice. Lower doses of estradiol increased weight gain in orchiectomized mice at early time points. Testosterone increased weight in intact females and males, but not in gonadectomized mice. Estradiol increased nose-to-tail length in intact females at early time points, but inhibited length in ovariectomized females at later times, and it decreased length in intact males. Testosterone increased length in normal females and normal males. Serum Ca was unaffected by ovariectomy, but orchiectomy resulted in decreased levels. Estradiol reduced serum Ca in gonadectomized animals; serum Ca was increased by estradiol treatment in intact females. Changes in tibial bone weight, ash weight and mineral composition, and relative sizes of epiphyseal and metaphyseal bone were gender-, gonadectomy- and hormone-specific. Bone weight was greater in ovariectomized mice. Ash weight per bone was comparable, but there was an increase in Ca and P content with ovariectomy. Estradiol increased bone weight, ash content, and bone Ca and P in ovariectomized and intact females. Orchiectomy alone did not alter bone weight, ash content, or Ca and P, but orchiectomized mice were sensitive to estradiol; all parameters were increased in the orchiectomized animals treated with estradiol. Analysis of the ash content and Ca and P per mg bone, rather than per bone, demonstrated estradiol and testosterone alter net bone formation, but not the amount of mineral per unit bone. Ovariectomy increased hypertrophic cartilage. While estradiol did not alter tibial area in ovariectomized mice, it caused an increase in intact females. The total amount of growth plate cartilage in ovariectomized animals was decreased by estradiol to levels typical of intact animals due to a greater decrease in the hypertrophic cartilage in the ovariectomized mice, as well as a greater increase in metaphyseal bone area. Testosterone had no effect on these parameters in the females. Orchiectomy decreased the amount of growth plate cartilage, but increased the hypertrophic zone. Estradiol increased growth plate cartilage in intact male mice, but decreased it in orchiectomized mice. This difference was also seen in the hypertrophic zone. Total growth plate cartilage and hypertrophic cartilage were increased by testosterone in intact males, whereas metaphyseal and epiphyseal bone area were decreased. The results show for the first time that there is a gender-specific response in both male and female mice to both estradiol and testosterone, whether or not the animals have been gonadectomized. For many parameters, orchiectomized mice behave like females in response to both sex steroids, indicating that the male gonad is needed for mouse bone to exhibit the male phenotypic response to estradiol and testosterone.  相似文献

Intracerebroventricular (icv) injection of purified recombinant human tumour necrosis factor (TNF , 4–8g) in conscious rats, produced increases in colonic temperature (1.0°C) and resting oxygen consumption (VO2, 14%) which were maximal after 80–90 minutes. Pretreatment with propranolol (10mg/kg s.c) significantly inhibited the rise in VO2, and prevented the increase in body temperature. Icv injection of an antagonist to corticotropin releasing factor (-helical CRF 9-41, 25 g), which prevents the pyrogenic and thermogenic actions of interleukin-1, did not influence the effects of TNF on temperature or VO2. Injection of a fragment of TNF (113–130 amino acid sequence) did not affect body temperature or VO2. TNF injection (icv) significantly increased brown adipose tissue (BAT)in vitro mitochondrial GDP binding, and this effect was slightly inhibited, but not prevented, by surgical denervation of the tissue, and was unaffected by pretreatment with -helical CRF 9-41. These data indicate that TNF can stimulate thermogenesis by a direct central action. The effects are largely, but not totally, dependent on the sympathetic nervous system but, unlike the thermogenic actions of interleukin they do not require release of CRF.  相似文献

  • 1.1. The unidirectional transepithelial fluxes of L-phenylalanine, β-methyl-D-glucoside and sodium ions across emusculated sheets of tench mid-intestine were determined in flux chambers.
  • 2.2. No net sodium flux was detectable, but phenylalanine was preferentially transferred from the mucosal to the serosal fluid.
  • 3.3. There was also a net movement of β-methyl-glucoside towards the serosal medium, but it was much smaller than that of phenylalanine.

We evaluated the effects of genotype (Muscovy, Pekin and their crossbreed hinny and mule ducks) and feeding levels (overfeeding between 12 and 14 weeks of age vs ad libitum feeding) on energy metabolism and lipid deposition in breast muscle of ducks. Samples of breast muscle (Pectoralis major) were collected at 14 weeks of age from 8 birds per group. Overfeeding induced an accumulation of lipids in breast muscle (1.5- to 1.7-fold, depending on genotype) mainly induced by triglyceride deposition. It also induced a considerable increase in the amounts (expressed as g/100 g of tissue) of saturated and mono-unsaturated fatty acids (SFA, MUFA), while the amounts of poly-unsaturated fatty acids (PUFA) remained unchanged in hinny and Muscovy ducks or slightly increased in Pekin and mule ducks. In breast muscle, overfeeding decreased the activity of the main enzymes involved in lipogenesis from glucose (glucose-6-phosphate dehydrogenase, G6PDH, malic enzyme, ME, acetyl CoA carboxylase, ACX). Lipoprotein lipase (LPL) activity in Pectoralis major muscle was also significantly decreased (-21%). The ability of muscle tissues to catabolize long-chain fatty acids, as assessed by beta-hydroxyacyl CoA dehydrogenase (HAD) activity, was increased in Pectoralis major muscle, as was cytochrome-c oxidase (COX) activity. Hybrid and Pekin ducks exhibited higher levels of ACX and LPL activity in Pectoralis major muscle than Muscovy ducks, suggesting a greater ability to synthesise lipids in situ, and to take up circulating lipids. Total lipid content in breast muscle of hybrid and Pekin ducks was higher than in that of Muscovy ducks. In hybrid and Pekin ducks, lipid composition of breast muscle was characterized by higher amounts of triglycerides, SFA and MUFA than in Muscovy ducks. Finally, oxidative metabolism was greater in Pectoralis major muscles of hybrid and Pekin ducks than in Muscovy ducks, suggesting an adaptative strategy of muscle energy metabolism according to lipid level.  相似文献

The changes in α-amylase activity and in starch and free sugar content were investigated in correlation with lipid mobilization inHelianthus annuus during the first 15 days of seedling growth in discontinuous light and in darkness. Throughout the seedling development α-amylase activity increased more significantly in light than in darkness. It was always lower in cotyledons than in other tissues of the embryo axis. In both culture conditions, most of the transitory carbohydrates accumulated in germinating cotyledons were very likely synthesized by gluconeogenesis from the stored lipid breakdown. Nevertheless, in light-grown cotyledons, photosynthesis contributes to increase the carbohydrate levels. The study of several soluble sugars indicates that 1) sucrose stored in cotyledons of mature seeds was used at the onset of seedling growth, more rapidly in light than in darkness, 2) galactose and xylose, both involved as precursors of some cell-wall polysaccharides, remained at a very low level throughout the 15 days and 3) glucose, fructose and maltose accumulated in old etiolated cotyledons in contrast to what occurred in the light.  相似文献

The loss of flower-rich habitat in agricultural landscapes is a key factor contributing to bumble bee declines across Europe and North America. Yet, agricultural intensification has not only altered flower abundance in the landscape, but also affected when flowers are available during the season (e.g., mass-flowering crops). While we know that both total pollen and nectar as well as temporal availability can impact bumble bee colony success (growth and reproductive output), we have yet to understand how these two factors combined might manifest. We designed an experiment to decouple the effects of total food abundance and its temporal availability on bumble bee microcolony development by exposing them to either constant or pulsed food availability at a high and low ration level. Microcolonies provided constant, high-rations of food grew the most, while those fed variable, but high rations gained less mass over the course of the experiment. Regardless of the temporal presentation of food, microcolonies fed low rations gained little mass over the experiment. Reproductive output was greatest in microcolonies fed high rations, regardless of the temporal availability of food, while those given low rations produced on average 27% fewer drones. This study highlights the importance of food abundance for both colony growth and reproduction, regardless of how food is presented (e.g., constantly or in a pulse). Together, these results indicate that increasing total food abundance will have the greatest, positive impact on colony fitness.  相似文献

The effects of age (from 1 day post-hatch to 98 days of age) and feeding levels (feed restriction followed by overfeeding v. ad libitum feeding) on lipid deposition in breast muscle (quantity and quality, localisation) of mule ducks were determined in relation to muscle energy metabolism (glycolytic and oxidative), plasma levels of lipids, glucose and insulin, and muscle capacity for lipid uptake (characterised by lipoprotein lipase (LPL) activity). Two periods were defined for age effects on intramuscular lipids in breast muscle: − 1 to 42 days of age when lipids (mainly phospholipids and cholesterol provided by egg yolk) stored in the adipocytes during embryonic life were transferred to the muscle fibres and used for growth and energy requirements, − 42 to 98 days of age when the muscle again stored lipids (mainly triglycerides provided by liver lipogenesis), first in fibres and then in adipocytes.Plasma glucose and insulin levels were not affected by age. Plasma levels of lipids and LPL activity in breast muscle were high at 1 and 14 days of age and then decreased, remaining stable until 98 days of age. Energy metabolism activity in the breast muscle (mainly glycolytic activity) increased with age.Feed restriction, corresponding to 79% of ad libitum intake, applied between 42 and 75 days of age only resulted in decreases in plasma insulin concentration and total lipid content of breast muscle, mainly affecting triglyceride and mono-unsaturated fatty acid (MUFA) levels. Overfeeding increased plasma levels of insulin and lipids while glycaemia remained stable. LPL activity and total lipid levels increased in breast muscle, mainly induced by deposition of triglycerides and MUFA occurring particularly during the 2nd week of this period. Glycolytic energy metabolism decreased.In response to age or feeding levels, muscle lipid levels and composition reflect plasma lipid levels and composition and high muscle lipid levels stimulate oxidative energy metabolism.  相似文献

Candida albicans is one of the most common fungal pathogens, and causes systemic and invasive infections in humans. C. albicans biofilms are composed of yeast and hyphal and pseudohyphal elements, and the transition of yeast to the hyphal stage could be a virulence factor. In this study, diverse essential oils were initially investigated for anti-biofilm activity against C. albicans strains, and cascarilla bark oil and helichrysum oil and their components α-longipinene (a major constituent of both) and linalool were found to markedly inhibit biofilm formation without affecting planktonic cell growth. Moreover, α-longipinene and linalool were found to synergistically reduce biofilm formation. Notably, treatments with cascarilla bark oil, helichrysum oil, α-longipinene, or linalool clearly inhibited hyphal formation, and this appeared to be largely responsible for their anti-biofilm effect. Furthermore, the two essential oils, α-longipinene and linalool, reduced C. albicans virulence in Caenorhabditis elegans.  相似文献

Bowé (hardened ferricrete soils formed by erosion, drought or deforestation) are often associated with termite mounds, but little is known about these mounds and their role in the restoration of soils and plant biodiversity on bowé. This study examined termite mounds on bowé and their effects on soil depth and plant richness. Sixty-four sampling plots were laid out randomly on bowé sites with mounds and on adjacent bowé sites without mounds. The height and circumference of each mound were measured. Species inventories were made and soil depth measured in each plot. Linear mixed effects and generalised mixed effects models with Poisson error distribution were used to assess the variation in soil depth and plant species richness in mound and nonmound microsites. Two types of mounds (small vs. large) associated with different termite species were observed on bowé, with the small mounds being most common. Plots with either large or small mounds had deeper soils and higher plant richness than the adjacent plots without mounds. Conservation of termite mounds is important for restoring soils and plant richness on bowé, and termite mounds should be taken into consideration in biodiversity and soil management strategies for bowé.  相似文献

The efficacy of ERr 731(?), a commercially available extract isolated from Rheum rhaponticum, in terms of menopausal complaints like hot flushes, depression, anxiety and vaginal dryness has been proven in a two-year clinical study. Further a recent preclinical study excluded unwanted side effects on the endometrium by showing a lack of stimulation of proliferation marker genes by ERr 731(?) or its constituents in the 3-day uterotrophic assay. The present study aimed at further substantiating the safety of ERr 731(?) in terms of endometrial hyperplasia and at the same time test for potential estrogenic effects in the bone. Therefore, ovariectomized (ovx) rats were treated in a dietary long-term administration for 90 days. Hence, the modulation of proliferation in the uterus was investigated by examining the effects on the mRNA expression of proliferation marker genes (Mki67, Pcna), on the estrogen-responsive gene C3 and on the estrogen receptors ERα and ERβ. We additionally performed densitometry analysis of the proximal tibia metaphysis using peripheral computed tomography (pQCT) and quantified bone homeostasis markers in the serum to examine potential effects on the bone. In this study design, neither an uterotrophic response nor a modulation of proliferation marker genes on mRNA level has been observed as response to the long-term application of the rhapontic extract. Furthermore, no impact of the two administered ERr 731(?) doses on the E2 deprivation-induced bone loss has been evident at the end of the study. In conclusion, the observations from previous trials regarding the endometrial safety of ERr 731(?) have been supported by our experimental findings that exclude a stimulatory activity on proliferation in the uterus in a long-term administration in the young adult rat but no effect on the bone mineral density could be observed.  相似文献
