The ripening of raspberry fruit (Rubus ideausL. cv Glen Clova)is associated with a climacteric rise in ethylene production.As the fruit pigments change from green to red there is a progressivesoftening, loss of skin strength and a breakdown of cell wallsin the mesocarp. An increase in cellulase (endo-1,4-ß-D-glucanase)in both drupelets and receptacles accompanies these changes.The localization of cellulase in the regions of the fruit associatedwith abscission zones suggest the enzyme may be involved infruit separation as well as softening. A polygalacturonase-inhibiting protein (PGIP) was purified fromimmature raspberry fruits using ion exchange chromatography.The protein was composed of a single polypeptide chain withMr of 38·5 kDa and a pI residing above pH 10. Kineticstudies suggested that the inhibition was of a non-competitivenature. The PGIP inhibited two endopolygalacturonases (endo-PG)purified from Botrytis cinerea and an endo-PG produced by Aspergillusniger to varying degrees but did not inhibit two exo-PGs purifiedfrom B. cinerea, bacterial endopectate lyases and bacterialendo-PGs. The concentration of PGIP at various stages of flowerand fruit development was determined. The inhibitor was notdetected in the flower, but reached a maximum of 69 units g–1in the immature green fruit decreasing to 9 units g–1as fruits matured. The outer suberized phellemlayer of the polyderm and a primary protective layer of suberizedand lignified cells across the adaxial cortex of the petioleprecluded infection of the axillary buds by hyphae growing froma saprophytic base in the leaf. No protective layer formed throughthe abaxial cortex at the petiole base; consequently both fungicolonized the epidermis, primary cortex and outermost non-suberizedphelloid cells of the polyderm beneath the node. Two modes of cell separation occur within raspberry abscissionzones. In the cortex, fracture of the middle lamella, partialdissolution of the cell wall, and growth stresses in the abscissionzone cause existing intercellular spaces to develop into largecavities. Cells remain intact during this process. In the vascularbundle, dissolution of the middle lamella and the resultantformation of cavities in the cell wall is followed by cell wallbreakdown and cell disintegration. The production of ethylene by red raspberry (Rubus idaeus L.cv. Glen Clova) fruit increased climacterically during development.The concentration of ethylene within green fruit was low butincreased substantially as fruit abscission and ripening commenced.The receptacle contained higher concentrations than the drupeletsat all stages measured. In the mature ripening fruit the ethyleneconcentrations were found to be physiologically significant,and would accelerate the abscission of large green non-abscisingfruit if supplied as a fumigant. The addition of ethylene toripe fruit did not accelerate abscission, probably because saturatinglevels occurred naturally within these fruit. Reduction of ethylenesynthesis rates using the inhibitor of ethylene production aminoethoxyvinylglycine(AVG) reduced the rate of abscission zone weakening which occursin detached large green fruit. The rate of ethylene productionwas found to be dependent on the supply of the precursor l-aminocyclopropane-l-carboxylicacid (ACC). This only accumulated to any extent in those ripefruit with high rates of ethylene production. The activities of four fungal polygalacturonases (endo-PGI,endo-PGII, exo-PGI, exo-PGII), detected when Botrytis cinereawas grown on immature fruits of red raspberry (Rubus idaeus),were fractionated into soluble and wall-bound fractions. Westernblots and plate-trapped antigen ELISA showed that endo-PGI andendo-PGII were most abundant in the cell wall-bound fractionsof the host. Immuno-inhibition studies using a polyclonal antiserumagainst polygalacturonase-inhibiting protein (PGIP), purifiedfrom immature raspberry fruits, showed that the low level offungal PG activity detected in fractions containing endo-PGIwas due to the presence of PGIP. When a purified preparationof endo-PGI and endo-PGII from B. cinerea was allowed to reactin vitro with either a crude host cell wall preparation, orone which had previously been treated to remove cell wall-boundproteins, both endo-PG isozymes had a greater binding capacitytowards the former wall preparation. Endo-PGI and endo-PGIIalso had an affinity for fungal cell walls. Exo-PGI and exo-PGIIbound to both fungal and host cell walls. Greater quantitiesof fungal endo-PGs were detected by ELISA in fruits previouslyfrozen and thawed (‘freeze-thawed’) and inoculated,than in fresh inoculated fruit. This result paralleled the extentof fungal growth in these tissues as assessed by chitin assayand suggests that the resistance shown by raspberries is dependenton continual replacement of inhibitory substances or inducedresistance mechanisms. The time-course of flower development of Rubus idaeus L. cv.Glen Clova was studied on detached buds opened in the laboratory.After sepal and petal opening petal abscission occurred withthe petals from an individual flower being shed over 3-4 h.Abscission was accompanied by a peak in ethylene production.Treatment of flowers with aminoethoxyvinylglycine eliminatedthe peak in ethylene production but did not prevent petal abscission.However, petal loss was much slower, taking place over a periodof days rather than hours. Abscission was more effectively retardedby silver thiosulphate. Exogenous ethylene accelerated the rateof petal abscission and senescence. The increase in ethyleneproduction coincident with petal abscission appears to accelerateand co-ordinate the shedding of the separate petals on an individualflower. Ovules of the raspberry, Rubus idaeus L., resembled those ofother Rubus species in being unitegmic, crassinucellate, andanatropous. In one cultivar, ‘M. 69/139’, both eggcell and embryo were inserted slightly laterally to the micropyle.Of the tetraploid forms, 4n M. 69/139 showed a proportion ofembryo sacs with delayed differentiation, and 4n ‘M. Jewel’some with four antipodal cells. Embryo development showed littleuniformity in timing, but followed the Asterad type. Occasional delayed endosperm development seemed to be a varietalpeculiarity of M. 69/139. At maturity, the ergastic materialin healthy endosperms consisted of oil globules and aleurongrains. The hypostase formed a projecting pedestal below theendosperm at 8 days. The inner epidermal layer of the testaappeared specialized, both in the early stages of seed growth,when it is suggested that it functioned as an endodermis, andat maturity, when it appeared to form an osmotic barrier.  相似文献

Variation in fruit and seed development following crosses betweendiploid and autotetraploid forms of raspberry cultivars is described. Drupelet set was sensitive to a large number of influences,and varieties differed in their response to such crosses but,once set, fruit growth was largely independent of seed development.Mailing Jewel had a high drupelet set as a seed parent. Chromosomedoubling reduced the fertility of both male and female gametes,but the effect was greater in the pollen where the ability toinduce set was impaired. This is ascribed to physiological ratherthan chromosomal causes. It is suggested that seed size in infertile crosses betweenplants of different ploidy depended on a balance between earlygrowth following the stimulus of fertilization and later slowergrowth due to a weakness in the endosperm. The rate of fruit maturation was more rapid on tetraploid seedparents, but otherwise showed few consistent features.  相似文献

Black raspberry necrosis virus (BRNV) induces a severe apical necrosis in black raspberry (Rubus occidentalis) but fails to induce diagnostic symptoms in red raspberry. However, BRNV infection of F1, F2 and F3 hybrids from the cross black raspberry × red raspberry induced mosaic symptoms of varying intensity but no typical apical necrosis. In a survey of 28 red raspberry cultivars, a few developed severe angular chlorotic leaf spots when infected with raspberry leaf mottle virus and a few others did so when infected with raspberry leaf spot virus. These reactions were determined by single dominant genes designated Lm and Ls respectively. The value of the different host reactions for controlling the effects and spread of these viruses is discussed.  相似文献

Seed development was studied in crosses made between the diploidforms and autotetraploid mutants of two raspberry (Rubus idaeusL.) cultivars. Drupelet abortion did not appear to be causedby failure of fertilization; it occurred mainly between thefourth and eighth day after pollination, and manifested itselfas simultaneous cessation of growth in all tissues. In healthydrupelets at eight days after pollination, growth appeared tobe proceeding at similar rates in all the tissues except thatthe embryo seemed to be growing faster than the endosperm inthe most advanced crosses. Both diploid and tetraploid ovulesappeared to be more stimulated by pollen from tetraploid plants.Embryo differentiation differed according to the direction ofcross, especially in stunted embryos. Radicles tended to beabnormally large when diploid pollen was used and smaller thanusual with haploid pollen.  相似文献

A commercial serological, multiwell assay kit was used to assess the detection limits of Phytophthora fragariae var. rubi in raspberry roots. Detection limits in time lapse after inoculation, were assessed after inoculation of root systems of raspberry plants by zoospores of P. fragariae var. rubi. In extracts taken 3-9 days after inoculation, the pathogen was detected from the fourth day after inoculation. In a test series of simulated P. fragariae var. rubi infection where 0.25, 0.5, 1.0 and 1.5% of infected root tissue respectively, were mixed with healthy tissue (w/w), it was possible to detect the pathogen at 0.25% of simulated infection level. The results obtained show the possibility of an early detection of small amounts of antigen by the ELISA test procedure used. This enhance possibilities for early diagnosis and thereby more effective prevention of Phytophthora diseases in raspberry.  相似文献

Black raspberry (Rubus occidentalis) is an important specialty fruit crop in the US Pacific Northwest that can hybridize with the globally commercialized red raspberry (R. idaeus). Here we report a 243 Mb draft genome of black raspberry that will serve as a useful reference for the Rosaceae and Rubus fruit crops (raspberry, blackberry, and their hybrids). The black raspberry genome is largely collinear to the diploid woodland strawberry (Fragaria vesca) with a conserved karyotype and few notable structural rearrangements. Centromeric satellite repeats are widely dispersed across the black raspberry genome, in contrast to the tight association with the centromere observed in most plants. Among the 28 005 predicted protein‐coding genes, we identified 290 very recent small‐scale gene duplicates enriched for sugar metabolism, fruit development, and anthocyanin related genes which may be related to key agronomic traits during black raspberry domestication. This contrasts patterns of recent duplications in the wild woodland strawberry F. vesca, which show no patterns of enrichment, suggesting gene duplications contributed to domestication traits. Expression profiles from a fruit ripening series and roots exposed to Verticillium dahliae shed insight into fruit development and disease response, respectively. The resources presented here will expedite the development of improved black and red raspberry, blackberry and other Rubus cultivars.  相似文献

Rostromysis bacescuii gen. et sp. nov. and Megalopsis denlicaudagen. et sp. nov. belonging to the tribe Leptomysini are describedfrom the south-west coast of Australia. The characters uniqueto the genus Rostromysis are the unusually long club-shapedrostrum and the large delicate lamella-like pleopods of thefemale, thus easily distinguishing it from the allied generaTenagomysis and Doxomysis.
