Abstract

We investigated the structure of the hemal node in six healthy hair goats using histological and enzyme histochemical methods. After processing, tissue sections were stained with Crossman's trichrome, Gordon-Sweet's silver and Pappenheim's panoptic stains. Alpha-naphthyl acetate esterase (ANAE) and acid phosphatase (ACP-ase) were demonstrated in frozen sections. Hemal nodes were encapsulated by connective tissue and few smooth muscle cells. Several trabeculae originated from the capsule and extended into the hemal node. A subcapsular sinus was present beneath the capsule and was continuous with the deeper sinuses. Subcapsular and deep sinuses were filled with erythrocytes. The parenchyma consisted of lymphoid follicles, diffuse interfollicular lymphocytes and irregular wide lymphoid cords. Cortical and medullary regions were not distinct. ANAE (+) and ACP-ase (+) cells were located mainly in the germinal centers of the lymphoid follicles and also were scattered equally in the interfollicular region and lymphoid cords. Monocytes, macrophages and reticular cells displayed a diffuse positive reaction, whereas localized granular positivity was observed in lymphocytes. We demonstrated that the general structure of the hair goat hemal nodes is similar to that of other ruminant species.  相似文献

Caprine hemal nodes were studied by light microscopy after glutaraldehyde fixation and epoxy resin embedding. A node consisted of a capsule, subcapsular and other sinuses, cortex, medulla and hilus. Elements of circulating blood filled the interstices of the reticular meshwork and associated macrophages which traversed the lumina of subcapsular and medullary sinuses. The latter were rare in 1-month-old goats, progressively increased in number and size in 2- to 4-month-old goats and coalesced with each other and the subcapsular sinus in adult animals. The cortical tissue appeared as lymphoid nodules. Circumferential lymphatic vessels abutted on outer margins of the nodules and gave origin to several radial lymphatics which branched and anastomosed between the medullary blood sinuses. Medullary cords were organized around the radial lymphatics. A single efferent lymphatic was formed at the hilum by confluence of the radial lymphatics. Our study, in contrast to earlier reports, shows that caprine hemal nodes possess efferent lymphatics. The present data suggest that the hemal nodes are involved, in addition to classical functions, in blood storage by hemoconcentration.  相似文献

Blood vessels, that bring blood to various areas of the human superficial inguinal lymph nodes are predominantly arterioles and precapillaries. They are often arranged radially from the hilus to the capsule and from the capsule towards the portal thickening. The arteries and arterioles of the portal and capsular trabeculae reach the paracortical zone, occupying an intermediate position between the medullary cords and the cortex of the lymph node. The arterioles of the paracortical zone, passing between the cortex and the medullary cords, acquire an arcuate appearance. In both directions from them (into central and peripheral areas of the node) precapillaries branch off at a right angle. The cortex is supplied with blood by the arteriolar branches of the paracortical zone and the capsule of the node. The cortical precapillaries branch into capillaries either within the lymphoid nodules, or along their periphery. In the medullary cords those arterioles branch, that get from the portal thickening, portal trabeculae and paracortical zone.  相似文献

Summary A casting technique has been employed to display in three dimensions, the lymphatic microcirculation within the human lymph node. The casting compound filled the marginal sinus, and diffusely permeated the cortical lymphoid parenchyma. However, deep within the lymph node in the medullary region, the medium remained within the limits of the sinus walls. The casts showed well-defined channels appearing similar to vessels. These converged into larger vessels, which drained into efferent lymphatics leaving the node at the hilus.Electron microscopic examination showed that the outer wall of the marginal sinus and the trabecular side of trabecular sinuses had an intact, continuous endothelium with a basement membrane. However, gaps were present in the inner wall of the marginal sinus, as well as in the parenchymal wall of the trabecular sinus. In the medulla, the sinuses were lined by endothelial cells which appeared similar to macrophages. The sinus lining was incomplete and possessed numerous perforations. These observations indicated that sinus walls adjacent to connective tissue served as a barrier to cell movement, but those adjacent to a large lymphoid cell population had gaps, with cells in apparent transit between sinus lumen and parenchyma.  相似文献

We investigated the structure of hemal nodes in Saanen goats using immunohistochemical staining. We examined the distribution of CD3 positive T lymphocytes, CD79a positive B lymphocytes, CD68 positive macrophages and S100 protein positive follicular dendritic cells. Hemal nodes of six healty adult female goats were used. Hemal nodes were removed from the thoracic and abdominal cavities. The oval to round hemal nodes were observed especially between the abdominal aorta and vena cava, and near the kidneys and adrenal glands. Tissue sections were stained with Crossmon’s modified triple stain to demonstrate general histological structure. The avidin-biotin-peroxidase technique using anti-CD3, anti-CD79a, anti-CD68 and anti-S100 primary antibodies was used for immunohistochemistry. Many CD3 positive T lymphocytes were found in the germinal center of the lymph follicles and in the lymphatic cords of hemal nodes; CD3 positive cells also were observed in the sinuses. CD79a and CD68 positive cells were found at the germinal center of the lymph follicles. In the lymph follicles near the subcapsular sinuses, CD79a and CD68 positive cells were found especially in e areas bordering the mantle zone. S100 positive cells were found in the lymph follicles, lymphatic cords and sinuses.  相似文献

Summary The structure of the avian lymph node (ALN) is characterized by a thin capsule, thin lymphoreticular cords, and an absence of trabeculae. It is not possible to subdivide the ALN into cortex, paracortex and medulla, or to subdivide the system of sinuses into marginal, trabecular and medullary divisions. The lymphoreticular cords contain avian germinal centers (AGC) with B-lymphocytes and the area of T-lymphocytes.Postcapillary venules are responsible for the recirculation of lymphocytes. Sinus reticular cells do not exist in the ALN, but free macrophages are present. The phagocytic capacity of the macrophages was determined by injection of vital dyes (India ink, Berlin blue) and inoculation with Candida cells. Macrophages filled with markers migrate from the lymph sinuses into the lymphoreticular cords and further into the AGC. The mobility of the macrophages is remarkably lower after phagocytosis of Candida cells.This study is dedicated to Prof. Dr. Dr. h.c. A histological investigation of the filtering function of the spleen of the sunfish Lepomis spp. was conducted by light, scanning, and transmission electron microscopy. The parenchyma of the organ is predominantly red pulp, a system of splenic cords and sinuses. The white pulp consists of loose lymphoid tissue which forms a cuff around the pulp arteries. Filtering of particulate matter from the blood occurs in the red pulp by phagocytes of the pulp cords and ellipsoids (periarterial macrophage sheaths). The ellipsoids are pale-staining cuffs of macrophages and reticular cells in a framework of reticular fibres surrounding the arterial capillaries. Destruction of effete blood cells (especially erythrocytes) is confined to the pigment nodules; particulate matter is not taken up by the nodules. These yellow-brown bodies are dispersed throughout the red pulp and are bounded by a reticular capsule. They contain masses of phagocytes and have the appearance of a morula. They are associated with blood vessels and are surrounded by sinusoids. Prussian Blue stain shows the presence of haemosiderin within their phagocytes. The phagocytes of the pigment nodules are filled with inclusions such as residual bodies, siderosomes, and fragments of erythrocytes. The early filtering of particulate matter by the phagocytes of the pulp cords and ellipsoids may allow for a more efficient phagocytosis of erythrocytes by the pigment nodules, followed by storage and reutilization of iron-containing compounds uncontaminated by other phagocytosed material.  相似文献

Segmentary lumbar, posterior pancreato-duodenal and inferior tracheobronchial lymph nodes have been investigated macro- and microscopically. The segmentary lymph nodes reach 12 X 45 X 45 mm, 10 X 25 X 100 mm in size. Most often these nodes are found among the posterior pancreato-duodenal lymph nodes (92%). They represent a conglomerate of smaller lymph nodes growing together and having their own capsule, parenchyma, sinuses, afferent and deferent lymph vessels and are united into one large node.  相似文献

Lymphoid organs from belugas, Delphinapterus leucas, ranging in age from less than one to 16 years, were harvested during a sanctioned hunt to investigate morphology. The spleen is divisible into red and white pulp and a stroma consisting of a reticular network, a collagenous capsule, and trabeculae containing smooth muscle bundles. White pulp areas appear to be devoid of follicles and consist mainly of periarteriolar lymphatic sheaths (PALS), that are larger in younger than in older belugas. Definitive marginal zones between red and white pulp are difficult to discern in older belugas. Lymph nodes are similar to those of other mammals; they possess a follicular cortex surrounding a vascular medulla composed of lymphatic cords and sinuses. Smooth muscle is abundant in the medullary region, usually in close proximity to sinuses. The expansive nodular mass at the root of the mesentery, often referred to as the “pseudopancreas,” is similar to lymph nodes in microscopic architecture. Pharyngeal tonsils and gut-associated lymphoid tissue (GALT) are found along the digestive tract and display an “active” morphology. Tonsils are comprised of lobules of follicles separated by vascular connective tissue. Epithelial-lined crypts communicate with the pharyngeal lumen. GALT consists of diffuse and follicular lymphocytes within the intestinal mucosa and submucosa. The thymus is well developed in the younger belugas, with lobules divisible into densely packed cortical zones of thymocytes and more loosely arranged medullary lymphocytes. Hassall's corpuscles are occasionally visible within the medulla. Cetaceans diverged evolutionarily from other mammals over 55 million years ago. This study investigates changes in lymphoid organ morphology in a species that now inhabits a unique ecological niche. This study also lays the groundwork for functional investigation of the beluga immune system, particularly as it relates to differences between healthy and stranded animals. © 1993 Wiley-Liss, Inc.  相似文献

Despite new information concerning functional morphology of spleen, there are still some inaccuracies mostly regarding the spleen blood circulation. Billroth’s (splenic) cords are formed from three-dimensional network of fibroblastic reticular cells located among branched sinuses. Results from our study using scanning electron microscopy confirm an intimate contact between adjacent reticular cells and erythrocytes. Arterial terminals can be observed in the Billroth’s cords. The wall of sinuses reminds a sieve and it is lined with a special type of endothelium. In electron microscope, endothelial cells look like rods oriented parallel to the longitudinal axis of sinuses. Based on our observations fibroblastic reticular cells change to fixed phagocytes under no circumstances, hence they do not participate in phagocytosis. They may have a recognition function for cells circulating around them. According to our opinion, the open and the closed blood circulation are present in the human spleen simultaneously. Blood flowing in the closed circulation can help “absorption” of extra-vascular liquid and the blood elements into the vascular lumen. Due to sporadic occurrence of smooth muscle cells in the capsule and trabeculae, we assume that human spleen is not a blood reservoir, unlike the spleen in some other animals.  相似文献

Many aspects of the blood vascular network of the lymph node are unknown, and others need confirmation. We have studied the blood vasculature of rat peripheral lymph nodes by means of carbon perfusion and vascular cast corrosion techniques. At the hilus of the node, an artery gives off arterioles running in medullary cords towards the cortex. Some reach the peripheral cortex directly, branching there into slender cortical vessels. Other arterioles enter the periphery of the deep cortex units, and then head towards the peripheral cortex. Upon reaching it, they curve part way above the center of the deep cortex units and provide slender branches to the overlying peripheral cortex. Dense plexuses of capillaries arise from arterioles in the medullary cords, in the periphery of the deep cortex units, and in the outermost stratum of the extrafollicular zone of the peripheral cortex. In the cortex, the draining high endothelial venules are restricted to the extrafollicular zone and to the periphery of the deep cortex units. At the cortico-medullary junction, these peculiar venules transform into regular medullary venules which form the hilar veins. In contrast, the folliculo-nodules and center of the deep cortex units are little vascularized by a loose capillary network, while no vessels occur in the subsinus layer. These features of the node vascular network are of interest in relation to the node architecture.  相似文献

We defined the somatic environment in which female germinal cells develop, and performed ultrastructural analyses of various somatic cell types, with particular reference to muscle cells and follicle cells, that reside within the ovary at different stages of oogenesis. Our findings show that ovarian wall of the crayfish is composed of long muscle cells, blood cells, blood vessels and hemal sinuses. The follicle and germinal cells lie within a common compartment of ovarian follicles that is defined by a continuous basal matrix. The follicle cells form branching cords and migrate to surround the developing oocytes. A thick basal matrix separates the ovarian interstitium from ovarian follicles compartment. Transmission electron microscopy shows that inner layer of basal matrix invaginates deeply into the ovarian compartment. Our results suggest that before being surrounded by follicle cells to form follicles, oogonia and early previtellogenic oocytes reside within a niche surrounded by a basal matrix that separates them from ovarian interstitium. We found coated pits and coated vesicles in the cortical cytoplasm of previtellogenic and vitellogenic oocytes, suggesting the receptor mediated endocytosis for transfer of material from the outside of the oocytes, via follicle cells. The interstitial compartment between the inner muscular layer of the ovarian wall and the basal matrix of the ovarian follicle compartment contains muscle cells, hemal sinuses, blood vessels and blood cells. Granular hemocytes, within and outside the vessels, were the most abundant cell population in the ovarian interstitium of crayfish after spawning and in the immature ovary. J. We have produced a panel of monoclonal antibodies directed against nonlymphoid cells in central and peripheral lymphoid organs. In this paper we present the reactivity of one of these antibodies, ER-TR7. This antibody detects reticular fibroblasts, which constitute the cellular framework of lymphoid and nonlymphoid organs and their products. In frozen sections of the spleen incubated with this antibody, the red pulp and white pulp are clearly delineated. Furthermore, the major white pulp compartments--the follicles and periarteriolar lymphoid sheath as well as the marginal zone--are recognized by their characteristic labeling patterns. In lymph nodes, the capsule, sinuses, follicles, paracortex, and medullary cords are clearly delineated. In the thymus and bone marrow no such specialized compartments were demonstrated. ER-TR7 reacts with an intracellular component of fibroblasts. Since ER-TR7 does not react with purified laminin, collagen types I-V, fibronectin, heparan sulfate proteoglycan, entactin, or nidogen, it detects a hitherto uncharacterized antigen. The possible role of the ER-TR7 positive reticular fibroblasts in the cellular organization of peripheral lymphoid organs will be discussed.  相似文献

Caprine hemal nodes were studied by transmission electron microscopy after glutaraldehyde fixation and epoxy resin embedding. Hemal node macrophages were observed to be engaged in erythrophagocytosis. In the early stages of endocytosis, intact erythrocytes were contained in some of the heterophagic vacuoles of macrophages. Later, granular, electron-dense material appeared on erythrocytes, presumably as a result of lysosomal degradation of their matrices. Subsequently, the matrix fragmented and probably formed 'myelin-like figures' and residual bodies that dominated the macrophage cytoplasm. In addition, images of sinusoidal endothelium, reticular cells, lymphocytes and, rarely, eosinophils were observed that depicted structures resembling various stages of lysosomal digestion of erythrocyte matrix noted in macrophages. Our study provides evidence to support the fact that effete erythrocytes are filtered, besides known organs, also in caprine hemal nodes. The morphology and location of hemal nodes suggest that the organ can be an efficient blood filter.  相似文献

Testes from 47 juvenile Swamp buffalo bulls were examined for puberty and sexual maturity histologically and daily sperm production per gram of testis parenchyma was determined by enumeration of elongated spermatids in homogenates of testis parenchyma. Puberty was defined as the attainment of a daily sperm production per gram of testis parenchyma >0.5 x 10(6). In most bulls, puberty is attained by 24 mo of age, when scrotal circumference (SC) is approximately 16 cm, and liveweight exceeds 135 kg. Sexual maturity was defined as the attainment of adult levels of daily sperm production per gram of testis parenchyma (14 x 10(6)). In most bulls, this occurs at 30 to 33 mo of age, when SC is in the 17-to 20-cm range, and liveweight generally exceeds 250 kg. There was marked individual variation in age, liveweight and SC at both puberty and sexual maturity.  相似文献

Mesenteric lymph nodes in fetuses and offspring of white rats have been studied during various age periods after indomethacin injection in doses 2.5 mg/kg and 1 mg/kg daily from the 18th up to 21st day of pregnancy. Dose-dependent retardation in formation of main structures of the mesenteric lymph nodes, decreasing amount of the lymphoid type cells have been revealed. Retardation in the capsule, sinuses and reticular fibers of the node takes place, as well as decrease in lympho- and plasmocytopoiesis. It is more pronounced after indomethacin in dose 2.5 mg/kg. Simultaneously, formation of the cerebral substance and appearance of lymphoid nodules and their germinative centers are delayed. After the drug effect (2.5 mg/kg), up to the age of 2 weeks the amount of tissue basophils and eosinophilic granulocytes decreases. When the dose of the drug is 1 mg/kg decreasing amount of these cells is substituted for their compensatory increase on the 2d-3d week. The data obtained demonstrate a decreasing function of the lymph nodes, that depends on the dose of indomethacin.  相似文献

Age-related changes in testosterone, progesterone and estradiol 17-beta were determined by radioimmunoassay (RIA) in the serum of 155 male buffalo calves of varying ages. The calves were classified into 17 age groups. The mean weight of calves increased from 33.6 +/- 9.6 kg at one week of age to 531 +/- 41.4 kg at 42 months. The testosterone levels were less than 100 pg/ml from birth until 15 months of age, followed by peak concentrations of 422 +/- 79 pg/ml at 24 to 30 months and 793 +/- 193 pg/ml at 42 to 48 months (corresponding to puberty and maturity, respectively). The progesterone levels were higher in newly born calves and mature bulls. Otherwise, the levels continued to be low throughout the period of growth and development. Estradiol 17-beta was significantly higher in postnatal calves up to two months of age. The testosterone revealed a positive correlation with weight and age while E2 17-beta showed a negative correlation with age. These results do not support a direct role of peripheral progesterone and estradiol 17-beta in the onset of puberty and sexual maturity of buffalo bulls.  相似文献

The lymphatic nodes of intact albino rats were investigated electron microscopically. It was shown that the lymphatic sinuses were restricted by a layer of flattened cells; the basal membrane was absent. Certain distinctions in the structure of the cell lining sinuses and the reticular cells comprising the reticular base of the lymphoid tissue of the lymphatic node were found. The structure of the "sinus network" strands is shown. The structure of the cells of the sinuses lining is shown to be identical to the structure of cells of the vascular endothelium. It suggests the endothelial nature of the lining of the lymphatic node sinuses.  相似文献
