Omitting the far‐red in LED lighting for bioreactors is inexplicable because it affects photosynthetic efficiency and photomorphogenetic activity. Therefore, this work compares three light sources (fluorescent—white light (WL), and LED: blue + red—BRL and blue + red + far‐red—BRFRL) for cultures of Chlorella vulgaris. Metabolic activity was determined by isothermal calorimetry. Changes in the chemical composition of biomass were examined by spectrofluorimetry and Raman spectroscopy. Maximum quantum yield of photosystem II was determined on the basis of chlorophyll a fluorescence parameters. The algae grown under BRL produced significantly more thermal energy than those cultured under BRFRL and WL. The Raman spectra of cultures showed characteristic bands for carotenoids, chlorophyll, phenolics, lipids, aliphatic carbohydrates, pectins, and disaccharides. According to the cluster analysis, the chemical composition of cultures grown under BRL and BRFRL was very similar, whereas the WL represented a distinct group. BRL and BRFRL stimulated the biosynthesis of an unidentified compound(s) with fluorescence maximum at 614 nm. At the beginning of the cultivation, photosystem II had very weak photochemical activity. Under BRFRL, ratios of Fv/Fm reached the maximum after 4 days, whereas under BRL and WL, after 6 days of cultivation. The results point to the favorable influence of the far‐red on the metabolism of microalgae.  相似文献

The stimulatory effect of aqueous suspensions of Spirulina platensis dry biomass extracted at pH 6.8 and 5.5 was studied on four lactic acid bacteria (LAB) grown in milk. The addition of dry S. platensis to milk (6 mg/ml) stimulated growth of Lactococcus lactis by 27%. The growth of other strains was also promoted. NPK‐10:26:26 complex fertilizer based culture medium was studied for the mass production of Spirulina platensis using different light emitting diodes (LEDs). First, cultivation was carried out under white LED to formulate the optimum fertilizer loading for which Spirulina growth was maximized. Optimum composition for newly formulated fertilizer medium was NPK fertilizer ?0.76 g L?1 and sodium bicarbonate ?10.0 g L?1 and corresponding biomass productivity was found to be 76.67 mg L?1 day?1. The effect of different LEDs (for example, blue, white, red, green and yellow) on Spirulina growth kinetics and the accumulation of chlorophyll, protein and lipid content was determined using the optimum NPK fertilizer medium. Kinetic parameters (i.e., biomass productivity, maximum specific growth rate, maximum biomass concentration, nitrogen‐to‐biomass conversion factor and phosphorus‐to‐biomass conversion factor) and chlorophyll accumulation were affected by the use of different LEDs and follow the following trend: blue > white > red > green > yellow, whereas protein and lipid accumulation was almost independent of LEDs used. Elemental C, N, P and K concentrations were measured to find the effects of nutrients for the growth of Spirulina platensis. Physicochemical parameters (pH and conductivity) were also monitored during biomass growth under different LEDs. Finally, biomass growth using NPK‐10:26:26 fertilizer under different LEDs was compared with standard Zarrouk medium and better growth results were obtained using optimally formulated NPK‐10:26:26 fertilizer medium.  相似文献

Two green algae (Chlorella vulgaris and Scenedesmus obliquus) and four blue-green algae (Anacystis nidulans, Microcystis aeruginosa, Oscillatoria rubescens and Spirulina platensis) were grown in 81 batch cultures at different nitrogen levels. In all the algae increasing N levels led to an increase in the biomass (from 8 to 450 mg/l), in protein content (from 8 to 54 %) and in chlorophyll. At low N levels, the green algae contained a high percentage of total lipids (45 % of the biomass). More than 70 % of these were neutral lipids such as triacylglycerols (containing mainly 16:0 and 18:1 fatty acids) and trace amounts of hydrocarbons. At high N levels, the percentage of total lipids dropped to about 20 % of the dry weight. In the latter case the predominant lipids were polar lipids containing polyunsaturated C16 and C18 fatty acids. The blue-green algae, however, did not show any significant changes in their fatty acid and lipid compositions, when the nitrogen concentrations in the nutrient medium were varied. Thus the green but not the blue-green algae can be manipulated in mass cultures to yield a biomass with desired fatty acid and lipid compositions. The data may indicate a hitherto unrecognized distinction between prokaryotic and eukaryotic organisms.  相似文献

Marine Spirulina platensis may potentially influence the metabolic process in animal cells, and the effect of marine Spirulina platensis in normal and alloxan-induced diabetic rats was therefore investigated. Normal and diabetic rats (albino Wistar strain) were orally administered marine Spirulina platensis for 30 days and their blood levels of glucose and insulin and body weight changes were determined. Pancreatic histopathology was also noted. Treatment with marine Spirulina platensis caused significant alterations in the content of these indicators and therefore in the antidiabetic capacity of the treated animals compared to control rats.  相似文献

Phycobilisomes (PBS) were isolated from blue-green algaSpirulina platensis. Scanning tunneling microscope was used to investigate the three-dimensional structure of PBS deposited on freshly cleaved highly oriented pyrolytic graphite (HOPG) in ambient condition at room temperature. The results showed that the rods of PBS radiated from the core to different directions in the space other than arrayed in one plane, which was different from the typical hemi-discoidal model structure. The diameter of PBS was up to 70 nm, and the rod was approximately 50 nm in length. Similar results were observed in Langmuir-Blodgett (LB) film of PBS. The dissociated PBS could reaggregate into rod-like structures and easily form two-dimensional membrane while being absorbed on HOPG, however, no intact PBS was observed. The filling-space model structure of PBS inSpirulina platensis with STM from three-dimensional real space at nanometer scale was found, which showed that this new structural model of PBS surely exists in blue-green algae and red algae. The function of this structural model of PBS was also discussed.  相似文献

The biochemical properties of Spirulina platensis in an internally illuminated photobioreactor (IlPBR) were investigated under different light-emitted diode (LED) wavelengths; blue (λmax= 450 and 460 nm), green (λmax= 525 nm), red (λmax = 630 and 660 nm), and white (6,500K), with various light intensities (200, 500, 1,000, and 2,000 μmol/m2/sec) were examined. The highest specific growth rate, maximum biomass, and phycocyanin productivity occurred under the red LEDs (0.39/day, 0.10 g/L/day, and 0.14 g/g-cell/day, respectively) at 1,000 μmol/m2/sec; the lowest growth rate was obtained under blue LEDs. Indeed, the size of trichomes was changed into short form under blue LEDs at all light intensities or all LEDs at 2,000 μmol/m2/sec for the first 2 days after inoculation, and S. platensis did not grow in the IlPBR under the dark condition. These results provide a base for different approaches for designing the pilot scale photobioreactor and developing cost-effective light sources.  相似文献

The effects of initial glucose concentration and light intensity on specific growth rate, phycocyanin concentration and cell dry weight concentration in mixotrophic batch cultivation of Spirulina platensis using both shake flask and fermenter were investigated. Based on experimental results in shake flask culture, a number of mathematical models were constructed, and the optimal initial glucose concentration and the optimal light intensity were calculated to be Sopt=2.4471 g liter−1 and Lopt=3.8632 klx. Finally, a time-dependent kinetic model for mixotrophic batch cultivation of Spirulina platensis in fermenter was also proposed. This was in good agreement with the experimental results and could be employed to predict the production of biomass and phycocyanin, and the consumption of glucose in fermenter culture.  相似文献

Diabetes mellitus (DM), a metabolic disorder, is becoming a major health problem worldwide. Insulin is the single hope for management of type 1 diabetes, but it is not always available or suitable. For finding additional bioresources, the present study was performed. ELISA‐based preliminary screening of cyanobacterial biomass using antihuman insulin antibody have detected an insulin‐like antigen in Spirulina platensis S‐5, Spirulina NCCU482, and Spirulina NCCU483. Their similarity with insulin‐like antigen was further confirmed by electrophoretic mobility using bovine insulin as marker.  相似文献

The ultrastructure of Spirulina platensis, a cyanobacterium with a helical morphology, has been studied in relation to temperature and light intensity. An increase in temperature gives rise to a more tightly coiled trichome, an increase in sheath material formation and a decrease in cyanophycin (above 17°C) and polyglucan (above 20°C) granule concentration. An increase in light intensity leads to an increase in gas vesicle concentration while the phycobilisome content decreases. Furthermore, cylindrical bodies have been observed with a somewhat different ultrastructure from those found in other species of cyanobacteria. The occurrence, size and ultrastructure of polyhedral bodies, photosynthetic lamellae, mesosomes, lipid deposits and an unknown kidney-shaped inclusion in relation to temperature and light intensity are described.  相似文献

The paper describes the composition of extracellular free fatty acids (FFAs) and intracellular fatty acids (FAs) in the enrichment cultures of the prokaryotic alga Spirulina platensisand the eukaryotic alga Chlorella vulgarisgrown at optimal, supraoptimal, and suboptimal growth temperatures. With increasing growth temperature, the degree of unsaturation of the intracellular FAs of both algae decreased, while that of the extracellular FFAs of S. platensisincreased. The influence of nutrient addition on the growth rate of Spirulina platensis in the Mangueira Lagoon water was studied in order to investigate the feasibility of using this water for biomass production. The addition of urea and sodium bicarbonate was studied through surface response methodology, over concentration ranges from 0.0 to 0.01170 M, and 0.0–19.70 gl–1 respectively. The growth of Spirulina platensis in Mangueira Lagoon water with no addition of nutrients was carried out and compared with the biomass growth after nutrient addition. The results indicated that the optimal level of nutrients was 0.00585 M urea and without the addition of sodium bicarbonate. The biomass concentration was 1.4 gl–1 in 780 h of cultivation and the doubling time (t d) was 3.85 days. In 300 h, the biomass concentration in the medium without nutrient addition was 0.9 gl–1, with a doubling time of 3.80 days.  相似文献

An on-line measurement technique for estimating biomass production rate in a photosynthetic micro-organism culture was developed and tested experimentally. The technique is based on monitoring O2 production from the increase in pressure inside a closed photobioreactor. The data obtained by this method correlated with the direct measurement of the biomass concentration. A material balance on the components in the system allows the validity domain of the method to be defined. The method was applied to batch cultures of the cyanobacterium, Spirulina platensis, in a cylindrical photobioreactor validating existing physiological and light energy models.  相似文献

This study investigates the effects of shear stress on photosynthesis in dilute suspensions of Spirulina platensis and Chlorella by measuring the oxygen production rate using a coaxial, double-rotating-cylinder apparatus that generates Couette shear flow. Our device enables up to 0.6 Pa shear stress to be applied, which has the hydrodynamic effect of generating the algal motion and acutely augmenting the oxygen production rate of Spirulina, primarily because the surface area of algae exposed to illumination is increased. However, there is shear-flow limitation on any increase in oxygen production, and the shear stress at maximum oxygen production rate tends to decrease with increasing temperature. The comparative study with Chlorella showed the reverse relationship between oxygen production and shear stress, and the cause of this difference is discussed in terms of several factors such as size, shape, hydrodynamic stress capacity and others.  相似文献

The salt tolerance of two representatives of genus Spirulina (Arthrospira) Spirulina platensis and Spirulina maxima has been investigated. They both are the wide-spread objects of photobiotechnology and it has been shown that the content of 5–15 % sea-water in medium has not caused the decreasing of biomass yield more than 15–20% as compared with control. The further decreasing of biomass was proportionate to sea-water content in medium. The investigation of reactivity of native (intravital) exometabolites secreted into cultural medium has showed that the sea-water content influence the oxidative activity (OA) of exometabolites and hour’s rhythmics.  相似文献

The present study suggests that the effect of silver coated polyester film fixed in culture racks serves as a reflector of light intensity on Spirulina platensis cultivation, using of KNO3 and urea as nitrogen sources. The use of light reflector (LR) gave light intensity of 4.8-6.0 klux and the reduction in number of tube light with reflector gave 2.5 klux of light intensity and its effect was studied on S. platensis. Total chlorophyll productions were observed for the cultivation at light intensity of two lights with reflector. This improvement is simple, inexpensive and saves 50% electric energy by reducing the number of lights, and thus contributes to energy conservation.  相似文献

Microalgae provide a wealthy natural resource of bioactive compounds, which have many biological activities. Monosodium glutamate is a food additive that acts either as food preservatives or as tastiness enhancer. It was confirmed that monosodium glutamate poses a serious responsibility in the pathogenesis of anovulatory infertility. Therefore, the idea of this research was directed to reveal efficiency of Chlorella vulgaris and Spirulina platensis extracts against the ovarian dysfunction resulted due to monosodium glutamate consumption. Adult female albino mice were gavages orally monosodium glutamate alone or with either Chlorella vulgaris or Spirulina platensis aqueous extracts for 28?days. Female mice were subjected to superovulation to study the oocytes nuclear maturation stages. Histological and quantitative investigation was carried on ovaries. Biochemical assessment to measure the sex hormones level and ovarian enzymatic antioxidants was done. In addition, ovarian antioxidant mRNA genes were determined using quantitative PCR and Glyceraldehyde-3-phosphate dehydrogenase was used as an internal control. The result revealed that monosodium glutamate reduced the oocytes quality and maturation rate, while, both algae improve the oocyte quality and maturation rate than in monosodium glutamate group. Chlorella vulgaris and Spirulina platensis improved the monosodium glutamate ovarian tissue histological alteration, sex hormones content and raised the ovarian enzymatic antioxidants level. In addition, monosodium glutamate markedly diminished the Glutathione peroxidase, superoxide dismutase and catalase mRNA expressions, However, Chlorella vulgaris or Spirulina platensis upregulated the expression of genes close to control. In conclusion, Chlorella vulgaris and Spirulina platensis showed potential alleviative role against the monosodium glutamate ovarian dysfunction.  相似文献

A specific method was developed for monitoring the concentration of cyanobacteria (blue-green algae) before waterblooms, based on their characteristics ofin vivo fluorescence. The excitation and emission spectra of cyanobacteria are very different from those of eukaryotic algae, due to the importance of phycocyanin, rather than chlorophylla, in determining the fluorescence characteristics. Our results, based on four cyanobacteria:Microcystis aeruginosa, Anabaena cylindrica, Phormidium tenue andSpirulina platensis, indicate that excitation at 620 nm and its emission at 645 nm is a sensitive and specific method for their detection. Furthermore, the addition of 10 M photosynthesis inhibitor 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU) induced only 3% increase in phycocyanin fluorescence, suggesting that this measurement is almost independent of the ongoing rate of photosynthesis.Author for correspondence  相似文献

Summary A rural oriented methodology for the production of the blue green alga Spirulina platensis has been developed and practically evaluated on pilot plant scale (45 m2). The system suggested is almost free of electrical inputs or capital intensive equipment and designed to use maximal manpower. A production rate of 3 kg of dry algae per week can be achieved using 45 m2 area.  相似文献
