Volatiles from wild Tunisian diploid (2n=2x=26) and tetraploid (2n=4x=52) Teucrium polium L. populations from five bioclimatic zones were assessed by GC and GC/MS. Thirty-eight compounds were identified. Main volatiles at the species level were myrcene (6; 15.3%), germacrene D (27; 9.0%), alpha-pinene (6.6%), beta-pinene (3; 5.8%), and alpha-cadinol (38; 5.1%). A significant variation among populations was observed. The tetraploid populations belonging to the sub humid and lower semi arid bioclimates showed a high amount of myrcene (6; 24.2-43.8%), those from the upper arid contain high percentages of alpha-cadinol (38; 50.6%). The dominant compounds in diploid populations, mainly located in the semi arid zone, are alpha-pinene (1) and germacrene D (27). Population chemical structure, assessed by a principal component analysis (PCA) using all identified compounds, is concordant with bioclimate and ploidy level. In situ conservation of populations should be made appropriately according to these two factors.  相似文献

Mentha pulegium L. is an aromatic herb belonging to the Lamiaceae family, a wild plant which is distributed in different areas of Iran. In this research, we evaluated the variability of essential oil content and compositions of 12 M. pulegium populations. Essential oils were analyzed using gas chromatography (GC) and gas chromatography/mass spectrometry (GC/MS) methods. The essential oils content varied from 0.22 to 1.63% w/w within different populations. Twenty-nine compounds were identified which represent 83.4–98.7% of the total essential oil. The most significant essential oil compounds among the studied population were identified using the principal components analysis (PCA-biplot). According to the PCA-biplot, the major compounds were pulegone (2.5–51.7%), menthone (0.2–25.3%), limonene (0.0–35.4%), 1,8-cineol (0.0–33.4%), piperitenone oxide (0.2–55.2%), and trans-piperitone epoxide (0.0–28.5%). Besides, hierarchical cluster analysis indicated that the studied populations were classified into two main clusters based on the essential oil components. The canonical correspondence analysis (CCA) indicated that some environmental factors could influence the phytochemical constituents as well as the antioxidant activity. The temperature and altitude were effective environmental factors with regards to 1–8 cineol, limonene and menthone content, while average rainfall was the most effective factor with respect to trans-piperitone epoxide, piperitenone oxide, and pulegone content. Our results consequently showed that environmental factors had a significant effect on the essential oil content and its antioxidant activity in M. pulegium populations.  相似文献

The water-distilled essential oil the leaves of Coridothymus capitatus were analyzed by GC/MS and also analyzed by direct thermal desorption GC/MS. Comparison was made between two analyses techniques. The essential oil consisted mainly of monoterpenes 98.9%, while oxygenated hydrocarbons were identified as 55.6 % and non-oxygenated hydrocarbons as 43.6%. As major components were found carvacrol (35.6%), p-cymene (21.0%), thymol (18.6%), gamma-terpinene (12.3%), alpha-terpinene (3.2%), beta-myrcene (3.0%) and alpha-thujene (1.3%) by hydrodistillation and by the GC/MS method. The direct thermal desorption GC/MS analysis also showed the same major components, namely carvacrol (51.6%), thymol (21.7%), p-cymene (9.7%) gamma-terpinene (8.2%), alpha-terpinene (1.64%). Leonotis leonurus widely used by traditional healers in southern Africa for treatment of various ailments, is well known for its reported psychoactive properties. The present study was undertaken to investigate the variation in essential oil composition between geographically distinct populations of L. leonurus in South Africa using gas chromatography. Plant material (n = 50) was collected from three provinces of South Africa. Essential oils of the aerial parts were obtained by hydrodistillation and analysed using one and two dimensional gas chromatography. Twenty-six compounds accounted for more than 80% of the total composition of the oil. Eight major constituents in the oil, representing about 50% of the total oil composition, were identified by both GC–MS–FID and GCxGC–ToF–MS. These major compounds were trans-β-ocimene (0.1–5.0%), cis-β-ocimene (0.1–31.5%), β-caryophyllene (0.3–15.0%), caryophyllene oxide (0.1–5.0%), α-humulene (0.4–18.2%), γ-elemene (0.4–10.6%), α-cubebene (0.2–12.0%) and germacrene D (0.1–22.1%). Marked similarities exist in the essential oil composition between populations; differences are mostly quantitative when determined by GC–MS–FID, while GCxGC–ToF–MS data reveals both quantitative and qualitative differences.Untargeted multivariate analysis was performed using SIMCA-P + 14.0 PCA and OPLS-DA methods, identifying two distinct clusters, inland and coastal populations.  相似文献

Volatiles from twelve wild Tunisian populations of Lavandula multifida L. growing in different bioclimatic zones were assessed by GC (RI) and GC/MS. Thirty‐six constituents, representing 83.48% of the total oil were identified. The major components at the species level were carvacrol (31.81%), β‐bisabolene (14.89%), and acrylic acid dodecyl ester (11.43%). These volatiles, together with α‐pinene, were also the main compounds discriminating the populations. According to these dominant compounds, one chemotype was revealed, a carvacrol/β‐bisabolene/acrylic acid dodecyl ester chemotype. However, a significant variation among the populations was observed for the majority of the constituents. A high chemical‐population structure, estimated both by principal component analysis (PCA) and unweighted pair group method with averaging (UPGMA) cluster analysis based on Euclidean distances, was observed. Both methods allowed separation of the populations in three groups defined rather by minor than by major compounds. The population groups were not strictly concordant with their bioclimatic or geographic location. Conservation strategies should concern all populations, because of their low size and their high level of destruction. Populations exhibiting particular compounds other than the major ones should be protected first.  相似文献

Four populations of charophytes (including three species), Chara inconnexa T. F. Allen (populations 1 and 2), C. vulgaris L., and Nitellopsis obtusa (Desv.) J. Groves, were studied for effective chemicals as oviposition deterrents of Culex pipiens pallens. The charophyte volatile organic compounds (VOCs) were retained in Tenax GR, subsequently desorbed using a thermal desorption cold trap injector (TCT), and analyzed by gas chromatography/mass spectrometry (GC/MS) to elucidate that charophytes have repellent properties. C. inconnexa (1) and C. inconnexa (2) exhibited strong repellent activities, and C. vulgaris showed some repellent activity against C. pipiens pallens with terpenes and benzothiazole playing major roles, while N. obtusa lacked those compounds and did not have an effect. These results suggest that charophytes have potential application as pesticides, but there are interspecific differences. In addition, benzene hydrocarbons were among the volatiles in Chara but not in N. obtusa, implying that some charophytes could be used to absorb these compounds.  相似文献

Genetic and essential oil variations from 14 Tunisian natural populations of Rosmarinus officinalis L. were assessed using eight isozymes and 25 terpenoids. Isozymes were revealed by 13% gel electrophoresis. Volatiles were analysed by GC and GC–MS. The effect of the collection sites and phenophase on yield and chemical composition of Salvia verbenaca essential oils was evaluated. The essential oil constituents were assessed by gas chromatography (GC) and gas chromatography–mass spectrometry (GC–MS). The highest essential oil yields were observed for samples of the higher semi-arid bioclimate and at the flowering period. Eighty-five volatile constituents were identified and their percentages varied significantly (p < 0.05) depending on the collection site and the phenological stage. According to the plants origin, essential oils were dominated by monoterpene hydrocarbons, oxygenated monoterpenes and sesquiterpene hydrocarbons. The monoterpene hydrocarbons (31.9%) predominate at the flowering stage whereas oxygenated sesquiterpenes (27.5%) at the early fruiting stage. The sesquiterpene hydrocarbons (28.2%) was the most represented chemical class at late fruiting. On the basis of GC-MS data, the major identified volatile constituents were viridiflorol (3.4–17.7%), α-pinene (0.7–15.9%), β-caryophyllene (1.0–15.3%) and p-cymene (1.3–14.2%). S. verbenaca contains a diversity of bioactive constituents which shows large variations as affected by the collection sites and phenophase.  相似文献

The variation of the essential‐oil composition among ten wild populations of Stachys lavandulifolia Vahl (Lamiaceae), collected from different geographical regions of Iran, was assessed by GC‐FID and GC/MS analyses, and their intraspecific chemical variability was determined. Altogether, 49 compounds were identified in the oils, and a relatively high variation in their contents was found. The major compounds of the essential oils were myrcene (0.0–26.2%), limonene (0.0–24.5%), germacrene D (4.2–19.3%), bicyclogermacrene (1.6–18.0%), δ‐cadinene (6.5–16.0%), pulegone (0.0–15.1%), (Z)‐hex‐3‐enyl tiglate (0.0–15.1%), (E)‐caryophyllene (0.0–12.9), α‐zingiberene (0.2–12.2%), and spathulenol (1.6–11.1%). For the determination of the chemotypes and the chemical variability, the essential‐oil components were subjected to cluster analysis (CA). The five different chemotypes characterized were Chemotype I (germacrene D/bicyclogermacrene), Chemotype II (germacrene D/spathulenol), Chemotype III (limonene/δ‐cadinene), Chemotype IV (pulegone), and Chemotype V (α‐zingiberene). The high chemical variation among the populations according to their geographical and bioclimatic distribution imposes that conservation strategies of populations should be made appropriately, taking into account these factors. The in situ and ex situ conservation strategies should concern all populations representing the different chemotypes.  相似文献

This study reports the results of gas chromatography–mass spectrometry (GC–MS) analyses of the essential oil of Angelica archangelica L. (Apiaceae) roots, as well as its in vitro antifungal activity against 10 plant pathogenic fungi. Moreover, the essential oil was evaluated for its antifungal activity using the agar dilution method, and also minimum inhibitory concentrations and minimum fungicidal concentrations were determined. The major compounds identified by GC–MS were α-pinene (21.3%), δ-3-carene (16.5%), limonene (16.4%), and α-phellandrene (8.7%). The oil showed in vitro antifungal activity against some species of the Fusarium genus, Botrytis cinerea, and Alternaria solani. Our study indicates that the oil of A. archangelica could be used as a control agent for plant pathogenic fungi in natural formulations.  相似文献

The compositions of the essential oils obtained from ten populations of Thymus caespititius grown on S. Jorge (Azores) were studied by GC and GC-MS. All the oils analysed were dominated by their monoterpene fraction (69-9%). The sesquiterpene fraction was rather small (4-17%) and consisted mainly of oxygen-containing compounds (3-13%). Enantiomeric ratio of sabinene and alpha-terpineol was investigated in the oils from some populations. (+)-Sabinene was detected with high enatiomeric purity (96 99%). Although ( +)-alpha terpineol was the predominant enantiomer, its purity varied considerably (66-97%). Cluster analysis of all identified components grouped the oils into four main clusters that corresponded with their major components: alpha-terpineol (43-68%), carvacrol (32-52%), thymol (44-58%) and sabinene (41%). The essential oil from Th. caespititius showed a clear chemical polymorphism, that was particularly evident among four populations growing along a 200 m distance at Pico Verde.  相似文献

Essential oils of three different Salvia species [Salvia aucheri var. aucheri (endemic), Salvia aramiensis and Salvia pilifera (endemic)] were screened for their possible antioxidant and antimicrobial properties as well as their chemical compositions. According to the gas chromatography (GC)/EIMS (gas chromatography/electron impact mass spectrum) analysis results; 41 (97.2%), 51 (98.5%) and 83 compounds (98.2%) were identified, respectively. While 1,8-cineole (30.5%), camphor (21.3%) and borneol (8.50%) are the major compounds for S. aucheri var. aucheri oil, beta-pinene (10.3%), was the main constituent for S. aramienesis together with 1,8-cineole (46.0%) and camphor (8.7%). In the case of S. pilifera oil, alpha-thujene (36.1%) and alpha-pinene (13.8%) determined as the major compounds. Antioxidant activity was employed by two complementary test systems namely 2,2'-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging and beta-carotene/linoleic acid systems. Antioxidant activity of S. aramiensis was found to be higher than those of the others for the both systems (12.26+/-1.09 and 92.46%+/-1.64 microg mg(-1), respectively). Additionally, antioxidant activities of BHT, curcumin, ascorbic acid and alpha-tocopherol were determined in parallel experiments. In the case of antimicrobial activity, similar activity pattern was obtained (both in disc diffusion and MIC tests). Antimicrobial activity of S. aramiensis was followed by S. aucheri var. aucheri and S. pilifera, respectively. In these experiments, the most sensitive microorganism Acinetobacter lwoffii was followed by Candida albicans.  相似文献

Currently available radioimmunoassay methods for estradiol in serum lack sufficient sensitivity and precision to monitor estradiol levels in patients placed on third generation aromatase inhibitors. We recently validated a gas chromatography/tandem mass spectrometry assay (GC/MS/MS) for estradiol and determined estrogen levels in normal post-menopausal women and in women with breast cancer before and during administration of aromatase inhibitors. Validation of the GC/MS/MS assay in human plasma and human serum included determination of assay sensitivity (<0.63 pg/ml), precision (all CVs less than 17.8%), recovery (98-103%), and linearity of recovery (R=0.998). Levels of estradiol were lower when assayed by GC/MS/MS compared to RIA under all conditions (7.26+/-4.82 pg/ml versus 11.9+12.0 pg/ml in normal post-menopausal women; 5.88+/-3.43 pg/ml versus 13.8+/-7.5 pg/ml in breast cancer patients prior to treatment; and<0.63 pg/ml versus 5.8+/-4.1 pg/ml during aromatase inhibitor therapy). Fifty-five women treated either with atamestane/toremiphene or letrozole/placebo were monitored for estradiol levels at 4, 8 and 12 weeks of therapy. The mean levels of estradiol during aromatase inhibitor therapy was 5.8+/-4.1 pg/ml as measured by RIA and <0.63 pg/ml by GC/MS/MS. The degree of suppression with the aromatase inhibitors as detected by RIA was 58% versus >89% by GC/MS. These results suggest that most RIA methods detect cross-reacting estrogen metabolites and yield higher measured levels than GC/MS/MS. Several pharmacological and clinical considerations suggest that GC/MS/MS should become the preferred method for monitoring aromatase inhibitor therapy.  相似文献

用水蒸气蒸馏法从曼陀罗中提取挥发油,并用气相色谱/质谱联用技术(GC-MS)对挥发油的化学成分进行分离鉴定,用气相色谱峰面积归一化法确定各组分的相对含量.结果从曼陀罗挥发油中鉴定出58种化合物,占总挥发油量的92.37%.其中主要成分为5,6-二氢-6-戊基-2H-吡喃-2-酮(44.29%)、二苯胺(12.50%)、四十四烷(10.41%)、二十烷(4.19%)、(E)-3-己烯-1-醇(2.38%)、3,7,11,15-四甲基-2-十六碳烯-1-醇(2.28%)等.  相似文献

Volatile compounds from two South-East Asian fermented soybean foods, Chungkuk-jang (CKJ) and Itohiki-natto (natto), were analysed by gas chromatography-mass spectrometry (GC-MS), gas chromatography (GC), and GC-sniffing. A total of 112 compounds were identified. A large amount of ethanol was detected from CKJ, while acetone and methyl isobutyrate were major components of natto. The characteristic odor compounds of CKJ were some ethyl esters of short chain fatty acids, diallyl disulfide, and several natto-like odor compounds were identified as ammonia, 2,5-dimethylpyrazine, and 2-methylbutanoic acid.  相似文献

Vepris macrophylla is an evergreen tree occurring in sub‐humid forest of Madagascar and traditionally used in the Island to treat several complaints as well as to prepare aromatic teas and alcoholic drinks. In the present work, the essential oil distilled from the leaves was analyzed for the first time by gas chromatography (GC‐FID) and gas chromatography/mass spectrometry (GC/MS). The major compounds were citral (56.3%), i.e., mixture of neral (23.1%) and geranial (33.2%), citronellol (14.5%), and myrcene (8.3%). The essential oil exhibited antimicrobial activity against S. aureus, P. aeruginosa, and C. albicans as determined by vapor‐diffusion assay, supporting the traditional use of the plant for preparing steam bath for the treatment of infectious diseases. The essential oil was evaluated for cytotoxic activity on human tumor cell lines by MTT (=3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyl‐2H‐tetrazolium bromide) assay, showing inhibitory effects comparable to those of cisplatin, notably on MDA‐MB 231 (human breast adenocarcinoma) and HCT116 (human colon carcinoma) cell lines. Finally, the essential oil was also subjected to screening for its antioxidant activity and the free radical scavenging capacity.  相似文献

Summary Twenty-five plant tissues from several species, including thirteen crown gall tissues, were analysed for the full range of unusual compounds (the opines) whose synthesis in crown gall cells and utilization by Agrobacterium tumefaciens are genetically determined by the Ti plasmids found in this bacterial species. A technique for the analysis of the non-guanidino opines by GC and GC/MS is described. None of the opines were detected in any of the various normal tissues analysed. In the crown gall tissues, on the other hand, these compounds were often present at very high levels. The essential oils of Daucus carota L. (Apiaceae) seeds sampled from ten wild populations spread over northern Tunisia were characterized by GC‐FID and GC/MS analyses. In total, 36 compounds were identified in the D. carota seed essential oils, with a predominance of sesquiterpene hydrocarbons in most samples (22.63–89.93% of the total oil composition). The main volatile compounds identified were β‐bisabolene (mean content of 39.33%), sabinene (8.53%), geranyl acetate (7.12%), and elemicin (6.26%). The volatile composition varied significantly across the populations, even for oils of populations harvested in similar areas. The chemometric principal component analysis and the hierarchical clustering identified four groups, each corresponding to a composition‐specific chemotype. The in vitro antimicrobial activity of the isolated essential oils was preliminarily evaluated, using the disk‐diffusion method, against one Gram‐positive (Staphylococcus aureus) and two Gram‐negative bacteria (Escherichia coli and Salmonella typhimurium), as well as against a pathogenic yeast (Candida albicans). All tested essential oils exhibited interesting antibacterial and antifungal activities against the assayed microorganisms.  相似文献

The composition of the essential oil isolated by steam distillation from aerial parts of the Costa Rican herb Lepechinia schiedeana (Schlecht) Vatke (Lamiaceae) collected in El Empalme, Costa Rica, was determined by capillary gas chromatography (GC) and coupled gas chromatography-mass spectrometry (GC-MS) analyses. Fifty-one components were identified corresponding ca. 93% of the oil. The major components were beta-pinene (26.6%), cis-pinocamphone (25.1%), delta-3-carene (6.1%), trans-pinocamphone (4.0%), camphor (3.8%) and beta-caryophyllene (3.7%).  相似文献

Throughout the world, the highly selfing annual common groundsel, Senecio vulgaris (Asteraceae) is a common weed. Recently, it has also colonized ecological compensation areas in agro-ecosystems. We investigated the genetic structure of S. vulgaris using random amplified polymorphic DNA (RAPD) profiles of 80 plants from nine populations representing three habitat types in two regions in Switzerland. RAPD variation among regions (19.8%), among populations within regions (19.2%) and within populations (61.1%) was highly significant (ANOVA; P < 0.001). Gene flow estimated from the observed differentiation among populations (PhiST = 0.382) was low (assuming Wright's island model, Nem = 0.404). Genetic distances between pairs of populations were significantly correlated with geographical distances (Mantel test; r = 0.37, P < 0.03). Molecular variance obtained with AMOVA was lowest in the small populations in compensation areas (1.13), intermediate in vineyard populations (2.49), all located in northern Switzerland and highest in the larger vegetable field populations from western Switzerland (3.41; P < 0.05). Overall, there was a positive correlation of molecular variance and population size (P < 0.05), as expected under genetic drift. However, molecular variance was negatively correlated with population size among populations in ecological compensation areas, suggesting that selection was also important. We also applied triazine herbicide to leaves of three offspring of each of the 80 plants. Plants from populations of compensation areas showed higher mean levels and reduced variation in the resistance to triazine herbicide than plants from vineyards and vegetable fields. This suggests that compensation areas were colonized from adjacent corn fields, in which there has been selection for herbicide resistance. We discuss the implications of our results for the biological control of S. vulgaris.  相似文献
