The ionic requirement for generating action potentials in ventral longitudinal muscle fibers dissected from beetle larvae was examined by conventional electrophysiological techniques. Muscle fibers that generated only graded responses in physiological saline were able to generate an all-or-none action potential when the potassium permeability of the membrane was inhibited by tetraethylammonium+ added to the saline. The peak of the action potential thus elicited was intimately related to the external Ca++ concentration. The action potential was blocked by Co++ which is known as a competitive inhibitor of Ca-spikes. Neither tetrodotoxin (3 μM) nor a Na-free condition effectively blocked the generation of the action potential. Mg++ induced a shift in the peak of the action potential; this was, however, due to the stabilizing action of Mg++ but not due to the penetration of Mg++ through the muscle membrane. No action potential was elicited in the muscle fiber when immersed in a Ca-free, EGTA saline even when a high concentration of either Mg++, Na+, or tetraethylammonium+ was present. The action potential of the larval muscle fiber was thus concluded to be a Ca-spike, through the channel of which Na+ or Mg++ did not penetrate.  相似文献

Abstract Background: During myocardial ischemia, accumulation of end products from anaerobic glycolysis (hydrogen ions (H+), lactate) can cause cellular injury, consequently affecting organ function. The cells' ability to buffer H+ (buffering capacity (BC)) plays an important role in ischemic tolerance. Age related differences in myocardial lactate and H+ accumulation (one hour of ischemia) as well as differences in BC, myoglobin (Mb) and histidine (His) contents in the left (LV) and right (RV) ventricles were assessed in neonatal compared to adult pigs. The BC of the septum was also compared. Methods and Results: Neonatal RV and LV had lactate accumulations of 43% and 63% and significantly greater H+ (p < 0.004) compared to the adult. In the neonate LV, BC was 17% significantly poorer (p = 0.0001), had 33% lower Mb (p = 0.0002) and 15% lower His content (p = 0.0004) when compared to the adult. In the RV, despite similar BC between the neonate and adult, myoglobin content was 36% (p = 0.0004) lower in the neonate. The neonate septum had a BC that was 11% lower than that of the adult. With maturation, the adult LV had a BC that was 10% greater (p < 0.01) than the RV while the septum mirrored that of the LV. Conclusions: During maturation to adulthood, the BC of the septum begins to closely resemble the LV. Neonatal hearts have a potentially greater vulnerability to acid-base disturbances during ischemia in both ventricles when compared to hearts of adults. This is due to lower levels of myoglobin and histidine in the young, which could render them more susceptible to injury during ischemia.Condensed Abstract During myocardial ischemia, H+ and lactate accumulation may pose deleterious effects on the heart. The ability to buffer H+ (buffering capacity, BC) affects ischemic tolerance. Although lactate accumulation during 1 h of global ischemia was similar between ventricles of neonatal and adult swine, H+ accumulation was greater and BC, Mb and His content were lower. With maturation, LV BC was higher than the RV while septum developmentally resembled the LV. Thus, hearts of neonates may be at a greater risk of ischemic injury compared to hearts of adults. Intravenous heparin administration caused a marked inhibition of Mg++-dependent (Na++K+)-stimulated ATPase activity of sarcolemmal (SL) membranes prepared from rabbit heart, whereas basal Mg++-ATPase was not affected. The inhibition depended on K+ concentration and was reversed only in the presence of albumin. Plasma free fatty acid (FFA) concentrations were raised in all animals, after heparin administration. The results obtained support the concept that FFA or other lipids originating in the plasma by the action of lipolytic enzymes released by heparin are involved in the mechanism of inhibition.  相似文献

Summary As different structural states of the (Na+–K+)-ATPase (EC 3.6.1.3) may lead to a changed reactivity to antibodies, the influence of Na+, K+, Mg++, Pi and ATP on the reaction between highly purified (Na+–K+)-ATPase and antibodies directed against the membrane-bound enzyme was measured. The antigen antibody reaction was registered by measuring the antibody inhibition of (Na+–K+)-ATPase activity.In themembrane-bound but not in thesolubilized enzyme four different degrees of antibody inhibition were obtained at equilibrium of the antigen antibody reaction if different combinations of Na+, K+, Mg++ and ATP were present during the incubation with the antibodies. Corresponding to the different degrees of inhibition, different rates of enzyme inhibition were measured. (a) The smallest degree of enzyme inhibition was obtained when (i) only Mg++, (ii) Mg++ and Na+ or (iii) Mg++ and K+ were present during the antigen antibody reaction. (b) The enzyme activity was inhibited more strongly if Na+, Mg++ and ATP were present together. (c) It was inhibited even more if only (i) Na+, (ii) K+, (iii) ATP or both (iv) ATP and Na+, (v) ATP and K+, (vi) ATP and Mg++, or if (vii) no ATP and activating ions were present. (d) The highest degree of antibody inhibition was obtained if Mg++, ATP and K+ were present together.In the presence of Mg++ plus ADP and in the presence of Mg++ plus the ATP analog adenylyl (--methylene) diphosphonate, Na+ and K+ did not influence the degree of antibody inhibition as they did in the presence of Mg++ plus ATP. It was further found that the degree of antibody inhibition in the presence of Mg++, ATP and K+ was affected by the sequence in which K+ and ATP were added to the enzyme prior to the addition of the antibodies.It is suggested that by antibody inhibition different conformations of the (Na+–K+)-ATPase could be detected. These conformations may possibly not occur in the solubilized enzyme and therefore do not seem to be necessarily linked to the intermediary steps of the ATP hydrolysis of the enzyme. The structural changes which are induced by Na+ and K+ in the presence of Mg++ plus ATP are proposed to occur during the Na+–K+ transport.  相似文献

The temperature sensitivity of the ATPase enzyme systems in a muscle microsomal preparation from the crayfish, Astacus pallipes, was studied. Preincubation of the enzyme preparation in the range 33–36°C produced a marked inactivation of the ATPases; the Mg++-dependent ATPase was very much more sensitive to this treatment than the Na+-K+-Mg++-dependent ATPase. Thus, the Arrhenius μ for the inactivation of the Mg++-dependent ATPase produced by eight minute preincubation is > 100 Kcals. These results are compared with the changes that are observed during the heat death of the whole animal, where exposure to 35°C produces a dramatic change in Na+ permeability within five minutes. Arrhenius μ for heat death is also > 100 Kcals and operates over the identical critical temperature range. It is suggested that the Mg++-dependent ATPase controls passive permeability in these excitable cells and the results also confirm the view that Mg++ and Na+-K+-Mg++ ATPases are separate enzymes.  相似文献

The leak fluxes of Na+, K+, Mg++ and Ca++ in mouse thymocytes are increased by Concavaline A (Con A), within minutes after mitogen addition. The intracellular Mg++ and K+ concentrations were decreased and the Na+ and Ca++ contents were increased by Con A in mouse thymocytes and spleen cells.  相似文献

Behaviour of different water soluble and exchangeable bases in a brackishwater fish pond soil was studied under four levels of water salinity, in combination with and without organic matter application. The results showed average content of water soluble bases to increase with increase in water salinity. The bases were dominated by Na+ followed by Mg++, Ca++ and K+ in decreasing order. SAR values of water increased with increase in water salinity and decreased slightly on organic matter treatment.Total content of exchangeable bases in soils was fairly high and was dominated by Ca++ and Mg++, followed by Na+ and K+ respectively. Amount of exchangeable Ca++ + Mg++ decreased while that of Na+ increased with increase in water salinity levels. Amount of exchangeable K+ did not show any appreciable change. Elementary Na+ currents were recorded in inside-out patches excised from cultured neonatal rat heart myocytes in order to study the influence of cytosolic Mg++ and other bivalent cations present at the cytoplasmic membrane surface on cardiac Na+ channel gating. Exposing the cytoplasmic membrane surface to a Mg++-free environment shortened the open state of cardiac Na+ channels significantly. open declined to 62±2% of the value obtained at 5 mmol/l Mgi ++. Other channel properties including the tendency to reopen and the elementary current size either changed insignificantly within a 10% range or remained completely unchanged. An almost identical change of open can be caused by switching from a Mn++ (5 mmol/l) containing internal solution to a Mn++-free internal solution. But open failed to significantly respond to a variation in internal Ni++ from 5 mmol/l to 0 mmol/l. The same response to internal Mg++ withdrawal was obtained with (–)-DPI-modified, non-inactivating Na+ channels, indicating that the exit rate from the open state remains as sensitive to cytosolic Mg++ variations as in normal Na+ channels with operating inactivation. The partition of sulfate, Ca++, and Mg++ across the membrane of the sartorius muscle has been studied, and the effect of various concentrations of these ions in the Ringer solution on the cellular level of Na+, K+, and Cl- has been determined. The level of the three divalent ions in toad plasma and muscle in vivo has been assayed. Muscle was found to contain an almost undetectable amount of inorganic sulfate. Increases in the external level of these ions brought about increases in intracellular content, calculated from the found extracellular space as determined with radioiodinated serum albumin or inulin. Less of the cell water is available to sulfate than to Cl-, and the Mg++ space is less than the Na+ space. An amount of muscle water similar to that found for Li+ and I- appears to be available to these divalent ions. Sulfate efflux from the cell was extremely rapid, and it was not found possible to differentiate kinetically between intra- and extracellular material. These results are consistent with the theory of a three phase system, assuming the muscle to consist of an extracellular phase and two intracellular phases. Mg++ and Ca++ are adsorbed onto the ordered phase, and increments in cellular content found on raising the external level are assumed to occur in the free intracellular phase.  相似文献

To determine the sequence of alterations in cardiac sarcolemmal (SL) Na+-Ca2+ exchange, Na+-K+ ATPase and Ca2+-transport activities during the development of diabetes, rats were made diabetic by an intravenous injection of 65 mg/kg alloxan. SL membranes were prepared from control and experimental hearts 1-12 weeks after induction of diabetes. A separate group of 4 week diabetic animals were injected with insulin (3 U/day) for an additional 4 weeks. Both Na+-K+ ATPase and Ca2+-stimulated ATPase activities were depressed as early as 10 days after alloxan administration; Mg2+ ATPase activity was not depressed throughout the experimental periods. Both Na+-Ca2+ exchange and ATP-dependent Ca2+-uptake activities were depressed in diabetic hearts 2 weeks after diabetes induction. These defects in SL Na+-K+ ATPase and Ca-transport activities were normalized upon treatment of diabetic animals with insulin. Northern blot analysis was employed to compare the relative mRNA abundances of --subunit of Na+-K+ ATPase and Na+-Ca2+ exchanger in diabetic ventricular tissue vs. control samples. At 6 weeks after alloxan administration, a significant depression of the Na+-K+ ATPase -- subunit mRNA was noted in diabetic heart. A significant increase in the Na+-Ca2+ exchanger mRNA abundance was observed at 3 weeks which returned to control by 5 weeks. The results from the alloxan-rat model of diabetes support the view that SL membrane abnormalities in Na+-K+ ATPase, Na+Ca2+ exchange and Ca2+-pump activities may lead to the occurrence of intracellular Ca2+ overload during the development of diabetic cardiomyopathy but these defects may not be the consequence of depressed expression of genes specific for those SL proteins.  相似文献

The interaction of Na+ with poly A, poly U, poly A·poly U, and Poly A·2 poly U has been investigated by means of potentiometry, by means of potentiometry, by means of a linked-function analysis of its effect on the binding of Mg++ ions, and of K+ by means of an analysis of its effect on the sedimentation coefficients of the polymers. The last method was found to be inapplicable. The results of the other two methods were found to be consistent, except in the case of poly A where the existence of base stacking, influenced by the binding of Mg++, significantly affects the linked-function analysis. The results are also consistent with the effects of the concentration of Na+ ions on the thermally induced conformational transitions of poly A·poly U and poly A·2 poly U, and with the extents of “binding” of Na+ to DNA measured by equilibrium and by transport methods. The interaction of Na+ with polynucleotides appears to be physically quite specific, although its thermodynamic basis is not clear. The extent of binding of Na+, Ψ, was found to be independent of the total Na+ concentration but a quadratic function of the extent of Mg++ binding, θ. In the absence of Mg++, Ψ = 0.35–0.38 for poly U, 0.40 for poly A, 0.59 for poly A·poly U, and 0.66 for poly A·2 poly U.  相似文献

Fragmented sarcoplasmic reticulum and transverse tubular system, as isolated in the microsomal fraction from rabbit skeletal muscle, bind H+, Na+, K+, Ca++, Mg++, and Zn++. The binding depends on a cation exchange type of interaction between these cations and the chemical components of the membranous systems of the muscle cell. The monovalent and divalent cations exchange quantitatively for each other at the binding sites on an equivalent basis. Scatchard plots of the H+ binding data indicate that the binding groups can be resolved into two major components in terms of their pK values. Component 1 has a pK value of 6.6 and a capacity for H+ binding of 2.2 meq/g N . The second component has a much higher H+ binding capacity (7–8 meq/g N ), but its pK value, 3.4, is non-physiological. The binding of cations other than H+ at a neutral pH occurs at the binding sites making up component 1. The order of affinity of the cations for the microsome binding sites is H+ » Zn++ > Ca++ > Mg++ » Na+ = K+ as reflected by the apparent respective pKM values: 6.6, 5.2, 4.7, 4.2, 1.3, 1.3. Caffeine, which causes contracture and potentiates the twitch of skeletal muscle, does not interfere with the binding of Ca++ by the microsomes at neutral pH.  相似文献

The influence of K+, Na+, Mg++, Li+, a serotonin, acetylcholine and tubocurarine on calcium-ganglioside-interactions was studied by way of equilibrium dialysis using 45Ca as tracer. Experiments were carried out at 22 °C and 4 °C, respectively. The concentrations of the substances were in the range of physiologically relevant conditions. Cations caused a release of Ca++ from calcium-ganglioside-complexes in the sequence of their molar efficiency: Mg++ ≈ Li+ > K+ ≈ Na+. Tubocurarine, serotonin and acetylcholine also affected calcium-ganglioside-interactions. Ca++ was displaced from ganglioside most effectively by tubocurarine, followed by serotonin, whereas acetylcholine competed considerably more weakly.  相似文献

  • 1.1. The hemolymph osmotic, Na+ K+, Ca++ and Mg++ concentrations were determined for both sexes in crabs from mangrove Ucides cordatus and Goniopsis cruentata and supralittoral Ocypode quadrala after aerial desiccation, for 10 hr.
  • 2.2. There was no difference between sexes in the two mangrove crabs, but in O. quadrata the females were the most significantly affected (P < 0.01).
  • 3.3. Hemolymph osmotic Na+, Ca++ and Mg++ concentrations increased significantly in desiccated ghost crabs, while K + concentration was not significantly changed. In the two mangrove crabs, only hemolymph Ca++ concentration increased significantly, after desiccation.
  • 4.4. The short time desiccation is suggested to be a device to study the steps of osmo-ionic regulation in terrestrial crabs.

The selective adsorption of the cations Na+, K+, Mg++ and Ca++ by the cell wall of the Mediterranean algaValonia utricularis (Siphonocladales, Chlorophyceae) from sea water of 40 %. S was investigated by extraction of cell-wall preparations, eluted before in 1.1 mol methanol (adjusted to pH 8) with 0.1 n formic acid in a Soxhlet apparatus. Na+ and K+ were determined by flame photometry, Mg++ and Ca++ by complexometric titration with EDTA. From calculation of the dry weight:fresh weight ratios and the chloride determinations in the eluates, the Donnan free-space fraction of the total cell-wall volume was calculated to about 35 %, and the analytical results of the cation concentrations in the extracts expressed asVal cm–3 DFS. This calculation is based on the assumption that the acidic groups of the noncellulosic matrix material, carrying negative charges by dissociation at the reaction of sea water (ph about 8) are responsible for the adsorption of cations by exhibition of a Donnan effect. The results obtained show clearly that besides the divalent cations Mg++ and Ca++, which according to the physico-chemical laws of the Donnan distribution must be relatively accumulated to the second power of the monovalent ones, potassium is also enriched by selective adsorption, and the K+:Na+ ratio increased significantly compared with that in sea water. This seems to indicate that the strength of attraction between the cations and the negative sites is dependent on the radii of the ions and the state of hydration and/or polarisation of the ions and binding sites.  相似文献

The effect of morphine on ATPase of synaptic plasma membranes (SPM) and synaptic vesicles isolated from the mouse brain was studied. The activity of synaptic vesicle Mg++-dependent ATPase from mice rendered morphine tolerant and dependent by pellet implantation was 40% higher than that from placebo implanted mice. However, the activities of Mg++-dependent ATPase and Na+, K+ activated ATPase of SPM of tolerant and nontolerant mice were not significantly different. The activity of synaptic vesicular Mg++-dependet ATPase was dependent on the concentration of Mg++ but not of Ca++; maximum activity was obtained with 2 mM MgCl2. On the other hand, Mg++-dependent ATPase activity of SPM was dependent on both Mg++ and Ca++, activity being maximum using 2 mM MgCl2 and 10?5 M CaCl2. It is suggested that this stimulation of ATPase activity may alter synaptic transmission and may thus be involved in some aspects of morphine tolerance and dependence.  相似文献

Abstract

A procedure for purification of the bungarotoxin-binding fraction of sarcolemma from rabbit skeletal muscle is described. Muscle is homogenized in 0.25M sucrose without high salt extraction and membrane fractions separated initially by differential centrifugation procedures. An ultracentrifugation pellet enriched in cell surface and sarcoplasmic reticulum markers is further fractionated on a dextran gradient (density = 1.0 to 1.09). Two fractions are identified as sarcolemma according to high specific activities for lactoperoxidaseiodination, Na+, K+-ATPase and α-bungarotoxin-binding. No Ca++, Mg++-ATPase activity is found in these fractions. A third fraction, the dextran gradient pellet, is enriched in Ca++, Mg++-ATPase activity and lactoperoxidase iodinatable material and characterized by low bungarotoxin binding. This fraction represents a mixture of sarcoplasmic reticulum and transverse tubules with some sarcolemma contamination.  相似文献

A Na+,K+-ATPase has been isolated from canine heart with a specific activity as high as 200 μmoles of inorganic phosphate/mg protein/hour. Activity is not due to simple detergent activation since specific ouabain binding (i.e., [Mg++,Na+,ATP] or [Mg++,Pi]-ligand dependent) ranged from 200–450 pmoles/mg protein. Specific ouabain binding activities are up to ten times greater than heretofore reported.  相似文献

Lysophosphatidylcholine (LPC) added at 30 μM or 10 μM to sarcolemma (SL) membranes of rabbit or dog causes about 50% inhibition of the Mg++ dependent Na+ + K+ stimulated ATPase activity. Higher concentration (100 μM) of free fatty acid (FFA) is needed to produce the same inhibition. Lysophosphatidyl ethanolamine, phosphorylcholine, phosphorylethanolamine also inhibit the enzyme activity. The inhibition of LPC is competitive for the Na+ site of the enzyme and it also affects the K+ site. The inhibitory effect of LPC is not additive to that produced by FFA or ouabain, on the contrary at low concentrations of these inhibitors the inhibition of LPC is reversed. The possible effects of increased levels of blood or tissue LPC on heart cell functions related to the Na+ + K+ ATPase activity are discussed. The possible interference of LPC with cardiac glycoside action is also discussed.  相似文献

Food restriction or decreasing the ratio of [Na+, K+] to Ca++, Mg++J in the diet of female rats before conception favoured the production of female offspring. Seven days of food deprivation decreased uterine fluid GPC diesterase activity in female rats, whereas long-term food restriction (21 days), rather than decreasing the enzyme activity, apparently stimulated it. Dietary Ca and Mg supplementation, likewise, produced significant decrease in GPC di esterase activity. A significantly positive correlation was observed between the levels of uterine GPC diesterase and secondary sex ratio change, which indicates that these dietary techniques of sex-ratio manipulation may modulate the uterine fluid GPC diesterase activity and influence sex determination in utero.  相似文献
