Transgenic rice plants in which the content of dienoic fatty acids was increased as a result of co-suppression of fatty acid desaturase were more tolerant to high temperatures than untransformed wild-type plants, as judged by growth rate and chlorophyll content. When untransformed wild-type and transgenic rice seedlings were incubated at 35 °C, seedlings of the transgenic rice lines showed approximately 1.6 and 2.1 times the growth of untransformed wild-type seedlings, as assayed by shoot and root mass, respectively. The chlorophyll content of the transgenic leaves after 9 d at 35 °C was also higher than that of wild-type rice. The maximum photochemical efficiency of photosystem 2 was also higher in transgenic plants than in wild-type plants upon high temperature stress.  相似文献

Trifoliate orange (Poncirus trifoliata L. Raf.), a rootstock widely used for citrus species, is salt-sensitive. Worldwide, salinity is a major abiotic stress affecting citrus growth and yield. Glycinebetaine (GB) is an important osmoprotectant involved in responses to salt stress. However, current evidence regarding the effect of salt stress on GB accumulation in trifoliate orange is limited, and the GB synthesis gene has not yet been shown to confer enhanced salt stress tolerance to this species in a transgenic context. In the current study, we first examined the change in GB level of trifoliate orange seedlings exposed to salt stress, and found that salt increased endogenous GB level in a concentration-dependent manner. A betaine aldehyde dehydrogenase gene (AhBADH) cloned from Atriplex hortensis was introduced into the trifoliate orange by means of Agrobacterium-mediated transformation. RT-PCR analysis on three selected transgenic lines showed that the AhBADH gene was overexpressed in each of them. GB levels in these lines were also higher than those in untransformed wild-type (WT) plants. In the transgenic lines, exposure to 200 mM NaCl resulted in significantly less serious leaf burning and defoliation, lower MDA accumulation, and higher chlorophyll contents than those in the WT plants. Moreover, when exposed to salt, shoots of transgenic plants contained lower levels of Na+ and Cl, higher levels of K+, and a higher K/Na ratio, while the same was true for the roots in most cases. Taken together, the data suggest that overexpression of the AhBADH gene in transgenic trifoliate orange enhanced salt stress tolerance. This may be correlated with the low levels of lipid peroxidation, protection of the photosynthetic machinery, and increase in K+ uptake.  相似文献

Phytochelatin synthase (PCS) is key enzyme for heavy metal detoxification and accumulation in plant. In this study, we isolated the PCS gene TcPCS1 from the hyperaccumulator Thlaspi caerulescens. Overexpression of TcPCS1 enhanced PC production in tobacco. Cd accumulation in the roots and shoots of TcPCS1 transgenic seedlings was increased compared to the wild type (WT), while Cd translocation from roots to shoots was not affected under Cd treatment. The root length of the TcPCS1 transgenic tobacco seedlings was significantly longer than that of the WT under Cd stress. These data indicate that TcPCS1 expression might increase Cd accumulation and tolerance in transgenic tobacco. In addition, the malondialdehyde content in TcPCS1 plants was below that of the wild type. However, the antioxidant enzyme activities of superoxide dismutase, peroxidase and catalase were found to be significantly higher than those of the WT when the transgenic plant was exposed to Cd stress. This suggests that the increase in PC production might enhance the Cd accumulation and thus increase the oxidative stress induced by the cadmium. The production of PCs could cause a transient decrease in the cytosolic glutathione (GSH) pool, and Cd and lower GSH concentration caused an increase in the oxidative response. We also determined TcPCS1 in Thlaspi caerulescens was regulated after exposure to various concentrations of CdCl2 over different treatment times. Expression of TcPCS1 leading to increased Cd accumulation and enhanced metal tolerance, but the Cd contents were restrained by adding zinc in Saccharomyces cerevisiae transformants.  相似文献

The endosomal LeNHX2 ion transporter exchanges H+ with K+ and, to lesser extent, Na+. Here, we investigated the response to NaCl supply and K+ deprivation in transgenic tomato (Solanum lycopersicum L.) overexpressing LeNHX2 and show that transformed tomato plants grew better in saline conditions than untransformed controls, whereas in the absence of K+ the opposite was found. Analysis of mineral composition showed a higher K+ content in roots, shoots and xylem sap of transgenic plants and no differences in Na+ content between transgenic and untransformed plants grown either in the presence or the absence of 120 mm NaCl. Transgenic plants showed higher Na+/H+ and, above all, K+/H+ transport activity in root intracellular membrane vesicles. Under K+ limiting conditions, transgenic plants enhanced root expression of the high‐affinity K+ uptake system HAK5 compared to untransformed controls. Furthermore, tomato overexpressing LeNHX2 showed twofold higher K+ depletion rates and half cytosolic K+ activity than untransformed controls. Under NaCl stress, transgenic plants showed higher uptake velocity for K+ and lower cytosolic K+ activity than untransformed plants. These results indicate the fundamental role of K+ homeostasis in the better performance of LeNHX2 overexpressing tomato under NaCl stress.  相似文献

To ultimately determine whether different levels of soil nitrogen (N) deposition can modify the detrimental effects of cadmium (Cd), the seedlings of Toxicodendron vernicifluum (Strokes) F. A. Barkley were exposed to soil Cd stress (0, 5 and 15 mg kg?1 dry soil), N deposition (0, 13 and 40 mg kg?1 dry soil) and their combinations. Soil Cd stress caused damage in plant growth, photosynthesis and other physiological indexes, and in the ultrastructure of mesophyll cells. The effects of N deposition on growth, lipid peroxidation and enzyme activities depended on the relative amounts of N supplied. The combination of low N deposition and Cd stress was positive to plant growth, photosynthesis and enzyme activities, and it caused lower levels of Cd accumulation and lipid peroxidation compared with the effect of Cd stress alone. The combination of high N deposition and Cd stress led to a higher Cd accumulation and lipid peroxidation, and to lower enzyme activities, as compared with the effect of Cd stress alone. T. vernicifluum was found to be sensitive to soil Cd stress. Soil Cd had detrimental effects on T. vernicifluum seedlings, but the tolerance of T. vernicifluum to Cd increased under low N deposition.  相似文献

While growing in the field, plants may encounter several different forms of abiotic stress simultaneously, rather than a single stress. In this study, we investigated the effects of calcium (Ca) deficiency on cadmium (Cd) toxicity in rice seedlings. Calcium deficiency alone decreased the length, fresh and dry weight, and the Ca concentration in shoots and roots. Also, the content of glutathione (GSH), the ratio of GSH/oxidized glutathione, and the activity of catalase were lower in Ca-deficient leaves compared to control leaves. Exogenous Cd caused a decrease in the contents of chlorophyll and protein, and induced oxidative stress. Based on these stress indicators, we found that Ca deficiency enhanced Cd toxicity in rice seedlings. Under exogenous Cd application, internal Cd concentrations were higher in Ca-deficient shoots and roots than in the respective controls. Moreover, we observed that Ca deficiency decreased heat-shock (HS) induced expression of HS protein genes Oshsp17.3, Oshsp17.7, and Oshsp18.0 in leaves thereby weakening the protection system and increasing Cd stress. In conclusion, Ca deficiency enhances Cd toxicity, and Ca may be required for HS response in rice seedlings.  相似文献

Recently our findings have shown that the integration of the gene coding for the rat gluco-corticoid receptor (GR receptor) in Nicotiana langsdorffii plants induced morphophysiological effects in transgenic plants through the modification of their hormonal pattern. Phytohormones play a key role in plant responses to many different biotic and abiotic stresses since a modified hormonal profile up-regulates the activation of secondary metabolites involved in the response to stress. In this work transgenic GR plants and isogenic wild type genotypes were exposed to metal stress by treating them with 30 ppm cadmium(II) or 50 ppm chromium(VI). Hormonal patterns along with changes in key response related metabolites were then monitored and compared. Heavy metal up-take was found to be lower in the GR plants. The transgenic plants exhibited higher values of S-abscisic acid (S-ABA) and 3-indole acetic acid (IAA), salicylic acid and total polyphenols, chlorogenic acid and antiradical activity, compared to the untransformed wild type plants. Both Cd and Cr treatments led to an increase in hormone concentrations and secondary metabolites only in wild type plants. Analysis of the results suggests that the stress responses due to changes in the plant's hormonal system may derive from the interaction between the GR receptor and phytosteroids, which are known to play a key role in plant physiology and development.  相似文献

Environmental pollution by heavy metals is a severe issue worldwide. Microbe-assisted phytoremediation is a safe, inexpensive, and promising strategy in refinement of metal-polluted regions. Current in vitro work was installed to study effects of the endophytic fungus Serendipita indica on some physiological traits and cadmium (Cd) bioaccumulation of Alyssum montanum and Helianthus annuus seedlings grown in MS medium, under varying levels of Cd (0, 20, 40 and 60 mg Cd/l medium). Even though Cd stress induced phyto-toxicity in both tested species, but a significant improvement was found in biomass accumulation, photosynthetic pigments content, and chlorophyll fluorescence indicators in inoculated seedlings by S. indica under different doses of Cd in media. The non-infected A. montanum plantlets accumulated more Cd in shoot than root, and illustrated the properties of an accumulator species as evidenced by translocation factor (TF) and bioaccumulation factor of shoot (BFS) higher than 1. Contrary to this, un-colonized H. annuus seedlings had higher amount of Cd in root than shoot and showed a phyto-stabilizer feature, as evidenced by TF˂1 and bioaccumulation factor of root (BFR) higher than 1. Presence of S. indica significantly enhanced Cd accumulation in root, while it noticeably diminished Cd amounts of shoot in both A. montanum and H. annuus seedlings, so that inoculated plants had higher values for BFR against lower values for BFS and TF, in compare to non-inoculated ones. These findings indicated that S. indica can be considered as a bio-fertilizer to improve the physiological characteristics of tested species under Cd stress, as well as a bio-stabilizer of Cd in the roots of A. montanum and H. annuus in the regions exposed to toxic levels of Cd.  相似文献

Rice SPX domain gene, OsSPX1, plays an important role in the phosphate (Pi) signaling network. Our previous work showed that constitutive overexpression of OsSPX1 in tobacco and Arabidopsis plants improved cold tolerance while also decreasing total leaf Pi. In the present study, we generated rice antisense and sense transgenic lines of OsSPX1 and found that down-regulation of OsSPX1 caused high sensitivity to cold and oxidative stresses in rice seedlings. Compared to wild-type and OsSPX1-sense transgenic lines, more hydrogen peroxide accumulated in seedling leaves of OsSPX1-antisense transgenic lines for controls, cold and methyl viologen (MV) treatments. Glutathione as a ROS scavenger could protect the antisense transgenic lines from cold and MV stress. Rice whole genome GeneChip analysis showed that some oxidative-stress marker genes (e.g. glutathione S-transferase and P450s) and Pi-signaling pathway related genes (e.g. OsPHO2) were significantly down-regulated by the antisense of OsSPX1. The microarray results were validated by real-time RT-PCR. Our study indicated that OsSPX1 may be involved in cross-talks between oxidative stress, cold stress and phosphate homeostasis in rice seedling leaves.  相似文献

Phosphoenolpyruvate carboxylase (PEPC) is known to play a key role in the initial fixation of CO2 in C4 photosynthesis. The PEPC gene from sugarcane (a C4 plant) was introduced into indica rice (Hang2), a process mediated by Agrobacterium tumefaciens. Integration patterns and copy numbers of the gene was confirmed by DNA blot analysis. RT-PCR and western blotting results showed that the PEPC gene was expressed at both the mRNA and protein levels in the transgenic lines. Real-time PCR results indicated that expression of the sugarcane PEPC gene occurred mostly in green tissues and changed under high temperature and drought stress. All transgenic lines showed higher PEPC enzyme activities compared to the untransformed controls, with the highest activity (11.1 times higher than the controls) being observed in the transgenic line, T34. The transgenic lines also exhibited higher photosynthetic rates. The highest photosynthetic rate was observed in the transgenic line, T54 (22.3 μmol m?2 s?1; 24.6 % higher than that in non-transgenic plants) under high-temperature conditions. Furthermore, the filled grain and total grain numbers for transgenic lines were higher than those for non-transgenic plants, but the grain filling (%) and 1,000-grain weights of all transgenic lines remained unchanged. We concluded that over-expression of the PEPC gene from sugarcane in indica rice (Hang2) resulted in higher PEPC enzyme activities and higher photosynthesis rates under high-temperature conditions.  相似文献

The purpose of this study was to clone S-adenosylmethionine synthase (BtSAMS) from Balsas teosinte (Zea mays ssp. parviglumis) and investigate its function via heterologous expression in Arabidopsis plants. The BtSAMS coding sequence was cloned and its length is 1185 bp. The expression of BtSAMS in Z. mays ssp. parviglumis was a 12.2-fold increase after aphid infestation. Transgenic Arabidopsis plants heterologously expressing BtSAMS were generated and their morphological characters were observed. A prolonged vegetative growth period was displayed by BtSAMS-OX plants. The more and longer trichomes were observed on the upper surface of BtSAMS seedlings. The ethylene content in BtSAMS-OX plants was not significantly different compared with the untransformed controls. These BtSAMS-OX transgenic plants showed greater resistance to peach aphids compared with the untransformed control plants. Peach aphids appeared to prefer the untransformed controls in that aphids number reduced by more than 50% on BtSAMS plants compared to controls. When sprayed with the insecticide avi-imidacloprid, the number of peach aphids decreased on BtSAMS-OX plants and untransformed controls by 91.6% and 80.5%, respectively. Peach aphids also showed lower reproduction ability when forced to feed on the transgenic plants. The number of newly born nymphs on BtSAMS plants was less than 50% of that on the untransformed control. We therefore propose that BtSAMS may be a suitable candidate gene to confer resistance to peach aphids in plants.  相似文献

Abiotic stresses are the major concern in recent years as their effect on world food production is constantly increasing. We have obtained salt tolerant Arabidopsis lines overexpressing SaRBP1 (Suaeda asparagoides RNA binding protein 1) of a Korean halophyte, S. asparagoides. Homozygous T3 Arabidopsis transgenic lines were developed and used for salt stress tolerance studies. The transgenic seedlings displayed tolerance to salt and mannitol compared to the wild type (WT) seedlings. Transgenic lines produced longer primary roots, more fresh weight, and higher number of lateral roots than WT. In planta stress tolerance assay results showed that the survival rates of transgenic plants were significantly higher than WT plants. Transgenic lines showed delayed germination under 200 mM NaCl stress. In addition, the transgenics showed higher water retention ability than WT. Subcellular localization results revealed that SaRBP1 was targeted to the cytoplasm. Northwestern blot analysis results confirmed the RNA binding property of SaRBP1. Quantitative Real-Time Polymerase Chain Reaction results revealed that many stress marker genes were upregulated by SaRBP1 overexpression. Thus, our data demonstrate that SaRBP1 overexpression lines are tolerant to salt stress. Hence, this is the first report for the functional characterization of SaRBP1, a novel RBP gene isolated from S. asparagoides cDNA library.  相似文献
