Inhibition of cholinesterases (ChE) has been widely used as an environmental biomarker of exposure to organophosphates (OP) and carbamate (CB) pesticides. Different ChE isoforms may be present in the same tissue and may present distinct sensitivities towards environmental contaminants. The present work characterises the soluble ChE present in mosquitofish (Gambusia holbrooki) total head homogenates, through the use of different substrates and selective inhibitors of cholinesterasic activity. Furthermore, the effects of sodium dodecylsulphate (SDS) on the enzymatic activity were investigated, both in vivo and in vitro. These results showed that acetylcholinesterase (AChE) seemed to be the predominant form present in head homogenates of G. holbrooki, despite the inhibition by tetraisopropylpyrophosphoramide (iso-OMPA) found at high concentrations. SDS was responsible for in vitro, but not in vivo, inhibitory effects. The in vitro AChE inhibitory effects of SDS was partially prevented by the use of increasing amounts of ethanol, suggesting that the inhibition was induced by an emulsion effect, which may explain the lack of effect in vivo. 相似文献

The empirical reasons for the persistent rarity of a genotype are typically complex and tedious to identify, particularly in nature. Yet rare morphs occur in a substantial fraction of phenotypic polymorphisms. A colour polymorphism has persisted for decades in the eastern mosquitofish, yet why this is so remains obscure. Here, I report the results of (1) intensive sampling at 45 natural sites to obtain the frequency distribution of the melanic (black) mosquitofish morph in Florida, (2) predation trials, conducted independently in mesocosms, with three different predatory species and (3) two mark-recapture studies, conducted in nature. This work (1) documents the rarity of melanic mosquitofish in nature, (2) demonstrates that melanic males experience a selective advantage over silver males in the presence of predators, (3) indicates no difference in the colour morphs' survival at a natural site essentially devoid of these predators, although suggesting a higher rate of recapture for melanic males at a site rife with predators. Overall, selective predation appears to contribute to the persistence of the melanic morph, despite its rarity in nature. 相似文献

In many poeciliid fishes, large males which court females coexist with small males which sneak-copulate. It is unclear whether these two tactics represent two evolutionarily stable strategies or if sneaking is a conditional strategy adopted by small, unattractive males. We studied the success of sneaky copulation by looking for sperm in the gonoduct of females after they were kept for 48 h with a male. A logistic regression analysis showed that the probability of a female being inseminated increased with female length and decreased with male length. The length of the male relative to that of the female was the best predictor of success. This result was confirmed using virgin females, thereby excluding any possible confounding effect due to the release of sperm from previous copulations. Sperm counts suggested that large males do not compensate for their reduced copulatory success by releasing larger sperm numbers. Behavioural data indicate that the advantages to small males are twofold: they have a greater chance to approach females from behind without being detected, and manoeuvre better when inserting the gonopodium into the female''s gonoduct. The selective advantage of small size might explain male dwarfism in poeciliids. Our results also suggest that small males adopting the sneaky tactic may be as successful as large males adopting courtship, and that alternative mating strategies may be maintained by negative density-dependent selection. 相似文献

Identifying targets of selection is key to understanding the evolution of sexually selected behavioral and morphological traits. Many animals have coercive mating, yet little is known about whether and how mate choice operates when these are the dominant mating tactic. Here, we use multivariate selection analysis to examine the direction and shape of selection on male insemination success in the mosquitofish (Gambusia holbrooki). We found direct selection on only one of five measured traits, but correlational selection involving all five traits. Larger males with longer gonopodia and with intermediate sperm counts were more likely to inseminate females than smaller males with shorter gonopodia and extreme sperm counts. Our results highlight the need to investigate sexual selection using a multivariate framework even in species that lack complex sexual signals. Further, female choice appears to be important in driving the evolution of male sexual traits in this species where sexual coercion is the dominant mating tactic. 相似文献

Eastern mosquitofish (Gambusia holbrooki) are native to the southeastern United States but notoriously invasive elsewhere, and are aggressive predators in ecosystems
they inhabit. Information on dispersal behavior is needed to better understand mosquitofish spread upon introduction and potential
means to mitigate that spread. We experimentally tested the effects of shallow water depths (3–24 mm) and obstacles (leaf
litter) on mosquitofish dispersal behavior, plus a range of conditions relevant to field situations. Mosquitofish dispersed
significantly faster in deeper water (p < 0.001) but some dispersed in only 3 mm water depth (i.e., one-half average body depth). Wetland and upland leaf litter
at natural densities strongly interfered with mosquitofish dispersal behavior. Based on our results, introduced mosquitofish
spread rapidly given unimpeded dispersal corridors (e.g., mowed ditches), and may do so at rates >800 m/day. Also, consistent
lack of sexual dimorphism in dispersal behavior indicates that mosquitofish spread is not strongly dependent on female poeciliid
reproductive biology. Our results support designation of mosquitofish as highly invasive and suggest that barriers to mosquitofish
spread must obstruct dispersal pathways as shallow as 3 mm depth. 相似文献

Multiple paternity (MP) increases offspring's genetic variability, which could be linked to invasive species' evolvability in novel distribution ranges. Shifts in MP can be adaptive, with greater MP in harsher/colder environments or towards the end of the reproductive season, but climate could also affect MP indirectly via its effect on reproductive life histories. We tested these hypotheses by genotyping N = 2,903 offspring from N = 306 broods of two closely related livebearing fishes, Gambusia holbrooki and Gambusia affinis. We sampled pregnant females across latitudinal gradients in their invasive ranges in Europe and China, and found more sires per brood and a greater reproductive skew towards northern sampling sites. Moreover, examining monthly sampling from two G. affinis populations, we found MP rates to vary across the reproductive season in a northern Chinese, but not in a southern Chinese population. While our results confirm an increase of MP in harsher/more unpredictable environments, path analysis indicated that, in both cases, the effects of climate are likely to be indirect, mediated by altered life histories. In both species, which rank amongst the 100 most invasive species worldwide, higher MP at the northern edge of their distribution probably increases their invasive potential and favours range expansions, especially in light of the predicted temperature increases due to global climate changes. 相似文献

The demographic and genetic characteristics of dispersing Gambusia holbrooki were examined relative to changes in stream flow velocity, flow direction, and rapid saltwater inundation. Dispersal of all G. holbrooki sex classes decreased as flow velocity increased, dispersal for a given flow velocity being greater in female, than male and juvenile fish respectively. Juveniles and males dispersed at low (9 mm s−1 ) but not high (109 mm s 1 ) flow. Females dispersed at both flow rates. Dispersing adults were not larger than non-dispersing adults of the same sex. Direction of dispersal was sex and size dependent, but independent of flow velocity. Larger females (>22 mm) moved preferentially downstream, while smaller females (17–22 mm) remained stationary or moved in the upstream direction. Males dispersed equally in both directions. No relationship was found between dispersal characteristics and the presence of high salt concentrations. Movement patterns at high flow were independent of glucose-6-phosphate isomerase ( GPI-2* ) alleles carried by dispersing individuals. Significant non-random dispersal of sex or size cohorts, and the interaction of dispersal with flow velocity have significant implications for the maintenance of genetic diversity in stream ecosystems, particularly where localized selection or genetic drift may alter the genetic constitution of specific sex or size cohorts. The applicability of these results to field populations is discussed. 相似文献

Synopsis Pair formation was studied in a polymorphic population of Gambusia holbrooki in a brackish drainage canal in Southern Florida. All females and the majority of males were gray colored and only 5% of the males were black spotted. A small fraction of the population formed pairs, composed of a female and a male that swam synchronously and close together for several minutes. During pairing the male often achieved gonopodial contact with the female. Black spotted males occurred in pairs ten fold more frequently than expected under random pairing. The major behavioral mechanism accounting for the disproportionate occurrence of spotted males in pairs is, probably, the preference of females for these males. 相似文献

tolerate the colder winters of northern and central Europe. Understanding the effects of latitude on its life-history traits
is essential to predict the potential for its invasion of central Europe in current scenarios of climate change. We studied
the variation of life-history traits and parasite load in the eastern mosquitofish along a latitudinal gradient from southern
France to southern Spain, sampling mosquitofish populations in eight Mediterranean river mouths ranging 5° in latitude. Southern
mosquitofish populations displayed higher catch rates, allocated more energy to reproduction (gonadosomatic index and gonadal
weight after accounting for fish size) and had a lower condition (total weight and eviscerated weight after accounting for
fish size) than in northern populations. Despite variability among populations, size-at-maturity (L50) significantly varied with latitude and northern individuals matured at smaller size (lower L50). Parasite prevalence ranged from 0.0 to 26.7% but parasite richness was very low; all the parasites identified were larvae
of pleurocercoid cestodes belonging to the order Pseudophyllidea. The abundance of mosquitofish parasites decreased with latitude
and the presence and number of parasites infecting the mosquitofish had a significant negative effect on fish condition. The
significant effects of latitude on the catch rates, life history and parasites of mosquitofish highlight the importance of
latitudinal studies of invasive species to understand the interactive mechanisms of climate change and biological invasions. 相似文献

Collections in two natural populations of the mosquitofish,Gambusia holbrooki, were made to determine whether the sexesassociated nonrandomly based on body size, and laboratory experimentswere performed to identify mechanisms that could generate theobserved field patterns of association. In both populationslarger females were associated with larger males. However, thesize distributions of males caught with and without femaleswere not different. Both gravid females and females that hadreleased broods within the previous 24 h spent more time nearlarger males in laboratory choice experiments. In contrast,males did not discriminate between females on the basis of sizeor reproductive status in laboratory choice experiments, andmales did not attempt relatively more copulations with largerfemales when one male was allowed to freely interact with twofemales. Body size differences and aggressive interactions betweenmales did influence the access of males to females, with largermales attempting more copulations with females when two maleswere allowed to freely interact with one female: aggressiveinteractions between males were frequent when males differedby < 1.0 mm standard length but were rare when males differedby>5.0 mm standard length. These results indicate that femalechoice for larger males could contribute to generating the nonrandomassociations between the sexes that were observed in these twopopulations and that interactions between males in the vicinityof females may additionally increase the mating advantages oflarger males. 相似文献

The interactive effects of an aryl hydrocarbon receptor (AhR) agonist and of a xenoestrogen on biomarker responses were studied in the liver of male mosquitofish (Gambusia holbrooki). Hepatic 7-ethoxyresorufin O-deethylase (EROD) enzymatic activity was measured as a biomarker of exposure to the model AhR agonist beta-naphthoflavone (bNF). Hepatic proteins indicating the exposure of males to the synthetic oestrogen 17alpha-ethynylestradiol (EE2) were monitored by Western blot analysis using immunoserum prepared for this study. After a semi-static exposure only to waterborne EE2, Western blot analysis of liver homogenate revealed the induction of two protein bands (a double band at 205 kDa and a single band at 125 kDa). The interaction between bNF and EE2 was investigated by analysing, on the one hand, EROD activity and, on the other hand, immunoreactivity corresponding to the two oestrogen-dependent protein bands in the liver of fish exposed to different concentrations of bNF for 2 days, then to the same concentrations of bNF plus 0.1 µg l-1 EE2 for 5 days. EE2 changed neither the basal activity of EROD nor its rate of induction with 1.0 and 4.0 µg l-1 bNF. On the other hand, the induction of oestrogen-dependent proteins with 0.1 µg l-1 EE2 was inhibited by exposure to 4.0 µg l-1 bNF. These results together with literature data suggest that field monitoring of xenoestrogen contamination through the analysis of oestrogen-dependent protein in male fish as a biomarker should take into account the possible negative interference of AhR agonists. 相似文献

In many taxa, temporary nutritional shortage early in development can favour compensatory strategies that include elevated growth (‘compensatory growth’) and/or extension of the usual period of development (‘catch-up growth’) once conditions improve. The net gains from each strategy depend on the extent to which larger body size increases fitness relative to associated costs (e.g. long-term effects on adult performance, or a greater risk of juvenile mortality). These costs and benefits are likely to differ between the sexes due to sex-specific selection. We documented the responses of male and female mosquitofish (Gambusia holbrooki) to 3 weeks of low food availability (7–28 days old) that restricted subsequent growth and morphology compared to control fish continuously reared on a high food diet (N = 635 fish total). Neither sex elevated their growth rate immediately after being returned to a normal diet compared to control fish. When measured over the entire period until maturation, however, females showed compensatory growth. Males did not. Both sexes also exhibited catch-up growth but the delay until maturation was significantly longer for males. Despite early growth restriction, both sexes eventually matured at almost the same size as control fish, although males had a significantly smaller gonopodium (a sexually selected trait) than that of control males. Salinity-induced mortality was examined in the mosquitofish Gambusia holbrooki to determine if natural selection associated with the tidal inundation of lower reach populations could potentially maintain glucose-6-phosphate isomerase ( GPI -2; E.C. 5.3.1.9) allele frequency variation observed at field sites. In vitro experiments established if survival was independent of GPI -2 genotype, and/or standard body length (S.L.). Bioassay results at 17%0 were consistent with lower survival of GPI -2*88/88 homozygote genotypes, GPI -2*100/100 genotypes in juvenile size classes, and greater survival of GPI -2*109/100 genotypes; 100% mortality was observed at salinities of 30%0 . In the early stages of the 30%0 bioassay, juvenile fish carrying each GPI -2 allele suffered similar, high rates of mortality. Following 19 weeks exposure to 4–8%0 NaCl the frequency of GPI -2*88 alleles decreased in adult G. holbrooki (> 17 mm SL). Salinity-induced mortality was found to be a selective influence capable of creating and maintaining GPI -2 allele frequency differences among G. holbrooki subpopulations, and/or between sex/size cohorts within subpopulations, but only at intermediate salinities. Whether saltwater inundation at field sites maintains inter-population diversity and produces local adaptation will be determined by the age structure of the population during periods of exposure, fluctuations in salinity as determined by the relationship between rainfall and tidal regime, and the level of immigration from non-affected sites. 相似文献

The two coefficients of the von Bertalanffy growth model [maximal standard length (LS∞) and initial growth rate (a)] were determined in eastern mosquitofish Gambusia holbrooki in the most northern area where it is established (Rennes, Brittany, France). The estimated mean LS∞ was 57·7 and 25·0 mm in females and males, respectively. The value of a was identical for both males and females before the males diverged from the females by slowing down their growth, probably because of the onset of puberty. The sexual competition reported in males would be responsible for higher interindividual variability of LS∞ in males than in females. a was positively correlated with water temperature and could be modulated by fish density and length at birth. Moreover, interindividual variability of a was influenced in a complex way by interactions between water temperature, length at birth and fish density. When compared to the data from studies in G. holbrooki inhabiting their native range or the southern introduced range, the present results suggest that the thermal tolerance domain for growth was altered and that males exhibited a shorter length at sexual maturity and a shorter LS∞ in the population established in Rennes. 相似文献

In social animal groups, an individual's spatial position is a major determinant of both predation risk and foraging rewards. Additionally, the occupation of positions in the front of moving groups is generally assumed to correlate with the initiation of group movements. However, whether some individuals are predisposed to consistently occupy certain positions and, in some instances, to consistently lead groups over time is as yet unresolved in many species. Using the mosquitofish (Gambusia holbrooki), we examined the consistency of individuals' spatial positions within a moving group over successive trials. We found that certain individuals consistently occupied front positions in moving groups and also that it was typically these individuals that initiated group decisions. The number of individuals involved in leading the group varied according to the amount of information held by group members, with a greater number of changes in leadership in a novel compared to a relatively familiar environment. Finally, our results show that the occupation of lead positions in moving groups was not explained by characteristics such as dominance, size or sex, suggesting that certain individuals are predisposed to leadership roles. This suggests that being a leader or a follower may to some extent be an intrinsic property of the individual. 相似文献

Quantification of metallothioneins (MTs) is classically associated with a cellular response to heavy metal contamination and is used in the monitoring of disturbed ecosystems. Despite the characterization of several MT genes in marine bivalves, only a few genetic studies have used MT genes as potential biomarkers of pollution. The aim of this study was to assess whether MT gene polymorphism could be used to monitor exposure of the Pacific oyster Crassostrea gigas to heavy metals and to develop specific genetic markers for population genetic studies in relation to environmental stress. The polymorphism of two exons of the C. gigas MT gene CgMT1 were studied using polymerase chain reaction single-strand conformation polymorphism (PCR-SSCP) in both field populations exposed to various metals concentrations and in experimentally exposed populations. High frequencies of two SSCP types in exons 2 and 3 of the CgMT1 gene have found to be significantly associated with tolerance to metals in experimental and field oyster populations. The use of MT1 gene polymorphism in C. gigas as in the present study should therefore be of high ecological relevance. In conclusion, the analysis of the types in these two CgMT1 gene exons, which can confer a greater tolerance to heavy metals, can constitute a good biomarker of effect of the presence of heavy metals in ecosystems. 相似文献

Correlated traits are important from an evolutionary perspective as natural selection acting on one trait may indirectly affect other traits. Further, the response to selection can be constrained or hastened as a result of correlations. Because mating behaviour and body colour can dramatically affect fitness, a correlation between them can have important fitness ramifications. In this work, melanic (black) male mosquitofishes (Gambusia holbrooki) with temperature-sensitive body-colour expression are bred in captivity. Half of the sons of each melanic sire are reared at 19 degrees C (and express a black body colour) and half are reared at 31 degrees C (and express a silver body colour). The two colour morphs are placed in the same social setting and monitored for behavioural differences. Mating behaviour and colour are correlated traits. Mating behaviour differs markedly between the two phenotypes, despite high genetic relatedness. Melanic (black) phenotypes are more aggressive towards females, chasing them and attempting more matings than their silver siblings. Females avoid melanic-male mating attempts more than silver-male mating attempts. When males with temperature-sensitive colour expression are melanic and aggressive, they probably experience a very different selective regime in nature from when they are silver and less aggressive. Under some conditions (e.g. predation), melanic coloration and/or aggression is advantageous compared with silver coloration and/or less aggressive behaviour. However, under different conditions (e.g. high-frequency melanism), melanism and/or aggression appears to be disadvantageous and melanic males have reduced survival and reproduction. Selective advantages to each morph under different conditions may enable the long-term persistence of this temperature-sensitive genotype. 相似文献

The introduction of fish has decimated many amphibian populations through increased predation, primarily on their larvae. Some amphibian species now occupy marginal habitats as a response to the presence of introduced fish predators. Such habitats may include ephemeral water bodies where fish do not usually occur, although breeding in these suboptimal environments may incur some cost to a species if its larvae are not adapted to develop under these conditions. We investigated this scenario of amphibian decline using the endangered green and golden bell frog (Litoria aurea) and the introduced mosquitofish (Gambusia holbrooki) in a factorial experiment to determine the responses of tadpoles to declining water levels and the introduced predator. Tadpoles metamorphosed asynchronously but did not accelerate development in declining water or when housed with mosquitofish. Mass at metamorphosis was 30% less in declining water. Mass increased with time to metamorphosis in constant water-level treatments, but decreased in declining water. Tadpoles did not respond to mosquitofish and were therefore assumed to be naive to this predator. These results suggest that ephemeral habitats may be suboptimal for breeding, and tadpoles appear better suited to develop in permanent water bodies free of introduced fish. Intra-clutch variability in larval development (i.e. bet-hedging) may allow L. aurea to cope with unpredictable pond duration, whereby even permanent water bodies may occasionally dry out. The responses observed in L. aurea suggest that introduced fish may have reduced the suitability of permanent water bodies as breeding sites for other pond-breeding amphibian species. The use of less favourable ephemeral habitats as breeding sites may be responsible for some of the declines reported in amphibians since the 1970s. 相似文献

A species recovery project for the endangered fish Red‐finned Blue‐eye (Scaturiginichthys vermeilipinnis) commenced in 2009 at Edgbaston Reserve in central western Queensland. To date, the project has included relocations and control of alien Eastern Gambusia (henceforth Gambusia, Gambusia holbrooki), using the piscicide rotenone. Preventing Gambusia colonising springs where Red‐finned Blue‐eye occur or have been relocated is crucial to the success of the project. As such, this study tested the use of porous silt fences that were installed around five springs from 2012. Results indicate that silt fencing is a suitable barrier material and that provided installed silt fences are serviced regularly, the material is an effective method of quarantining populations of endangered species in isolated aquatic environments. Four of the five installed fences were effective, and maintenance consisted of small repairs at regular intervals (quarterly) followed by replacement after 2 years. As such, silt fencing may have wider application in the conservation of endangered fish and other organisms, particularly in areas where isolated waterbodies occur and where their size renders exclusion fencing a practical recovery action. 相似文献
