Histochemical profiles of muscles were identified based on staining for myosin ATPase activity. They reveal typical arrangement of muscular fibres with a zoned pattern. Tonic fibres have a unique histochemical profile and are mixed with the most oxydative fast fibres to form toxic zones. Muscles show fast profiles in thigh and tonic or mixed profiles in fore-arm.  相似文献

Four selected leg muscles (gastrocnemius, soleus, vastus lateralis and intermedius) from thirty-two humans were autopsied within 25 hr of death and examined histochemically.The results of histochemical myofibrillar adenosine triphosphatase activity demonstrated that the soleus and vastus intermedius muscles have a higher proportion of slow twitch fibres (70%, 47%) than their synergists, gastrocnemius and vastus lateralis, respectively.The gastrocnemius contains about 50% slow twitch fibres and the vastus lateralis about 32%. Similar proportions of slow and fast twitch fibres have been reported for these hindlimb muscles in other mammals. Human muscles, however, differ from other mammalian muscles in that the proportion of slow and fast twitch fibres were similar in the superficial and deep regions of the muscles examined. Fast twitch oxidative glycolytic fibres in sedentary humans were observed less frequently, and they are less prominent in terms ofoxidative enzymatic activity when compared to similar fibres of several laboratory mammals studied previously.  相似文献

Morphometric and histochemical investigation of musculus sartorius was performed in ontogenesis in Rana ridibunda and Rana temporaria. Muscular composition was characterized according to the type of muscular fibres. Spectrum of lactatdehydrogenase isoenzymes was studied at different developmental stages. As the studies demonstrated, musculus sartorius underwent some essential changes in ontogenesis which manifested themselves in increasing number of muscular fibres and their areas, in changing LDG isoenzymic spectrum. Differentiation of the muscular fibres three types takes place at the 30th stage after P. V. Terentiev and depends on the nerve system maturation.  相似文献

The adrenergic and cholinergic innervation of the epididymal duct in the mouse has been investigated using histochemical methods and electron microscopy. This study demonstrates that the epididymal innervation of the mouse does not really differ from that of other mammals. Cholinergic nerves are mainly vascular, even in the cauda where cholinesterase activity is increased within the tubular musculature. Catecholamine fibres ensure the motricity of the wall of the canalicular system, particularly the terminal segment where the smooth muscle fibres are specifically differentiated.  相似文献

Summary The masseter muscles of different mammals were studied by means of histochemical reactions: NADH: Nitro BT oxidoreductase (NADHOX), 3-hydroxybutyrate: NAD+ oxidoreductase (HBOX), glycerol-3-phosphate: menadione oxidoreductase (GPOX), and acid-stable and alkali-stable myosin adenosine triphosphatase (ATPase). The masseter muscles of cattle and sheep consisted only of the fibres that reacted moderately for GPOX and strongly for NADHOX, HBOX, and the acid-stable ATPase. The masseter fibres of rats and guinea pigs reacted uniformly and strongly for GPOX and the alkali-stable ATPase. The fibres of the rats showed a weak to strong reaction for NADHOX and mostly a negative reaction for HBOX, whereas those of the guinea pigs reacted uniformly and strongly for NADHOX and HBOX. The masseter fibres of swine and dogs showed a weak or strong reaction for the alkali-stable and a negative or weak reaction for HBOX. The fibres of the swine were weak to strong in NADHOX activity and those of the dogs uniformly strong; the fibres of the two species gave a moderate to strong reaction for GPOX. The masseter fibres of the ruminant differed from those of the other species in histochemical properties, and appeared to have the histochemical characteristics that meed functional demands for slow, long-term exercise.  相似文献

On the basis of the histochemical activity of succinic dehydrogenase, only two fibre-types are distinguished in pigeon pectoralis major muscle. These are narrow "Red" and broad "White". The histochemical activity of myofibrillar ATPase was studied in these two distinct fibre-types. Both fibre-types showed high activity for the ATPase. "Red" fibres of pigeon pectoralis were not alkali-labile, at incubation pH 9.4, as were the "Type I" fibres of both avian and mammalian muscles. Again unlike "Type I" fibres, the "Red" fibres of pigeon pectoralis lacked the characteristic activation of acid-preincubated ATPase reaction. Pigeon pectoralis "Red" fibres are known to possess some characteristics of fast-twitch fibres (e.g. high fat, considerable phosphorylase, fibrillenstruktur myofibrillar arrangement, focal "en plaque" pattern of nerve endings). It is emphasized, therefore, that the pigeon pectoralis "Red" fibres are not equivalent to "Type I or slow-twitch", muscle fibres, but they are possibly "fast-twitch fatigue resistent or Type II Red" muscle fibres.  相似文献

In skeletal muscle from a patient with a mitochondrial myopathy and muscular carnitine deficiency, histochemical analysis demonstrated that mitochondrial ATPase showed activation with loss of latency even before addition of the uncoupler dinitrophenol (DNP). According to combined histochemical and biochemical studies by Meijer and Vloedman (1980), this finding indicates loosely coupled oxidative phosphorylation. After the addition of DNP the reaction intensity was markedly increased, but there were scattered enzyme-deficient fibres in which some residual activity was shown by ultracytochemistry. No defect in mitochondrial enzymes was found in biochemical studies. The enzyme histochemical changes and carnitine deficiency are probably both secondary to an unknown mitochondrial defect. Both the carnitine deficiency and the mitochondrial myopathy remained unchanged following long-term carnitine substitution therapy despite clinical improvement.  相似文献

The muscular coat of the rat esophagus in health consists of a homogenous population of cross-striated muscle fibers having moderate succinate dehydrogenase activity and ultramicroscopic signs characteristic of both red and white myons of skeletal muscles. Experimentally induced functional loads of the esophagus lead to structural changes in the muscles. Initially occurring signs of injury are replaced by the development of muscle hypertrophy. It has been also found that cross-striated muscles of the esophagus are capable of myosatellitogenesis and segregation of nucleosarcoplasmic areas. Longitudinal splitting of muscle fibers was observed in some cases.  相似文献

Summary The purpose of this investigation was to examine critically the validity of a computerized quantitative microphotometric histochemical technique for the determination of succinate dehydrogenase (SDH) activity in skeletal muscle fibres. Sections from the anterior costal diaphragm were removed from Fischer-344 rats (n = 12) and assayed histochemically to determine SDH activity. The SDH activity in individual muscle fibres was computed using a computerized microphotometric histochemical technique which involves measurement of the optical density of deposited diformazan derived from nitroblue tetrazolium within the fibres. To validate the histochemical technique, whole muscle SDH activities were calculated from the histochemical procedure and were compared to SDH activities determined from whole muscle homogenates via a standard quantitative biochemical assay. The mean within-day variability of the computerized microphotometric histochemical technique of determining SDH activity was 6% (range = 0.5–10.9%) for an area containing ~50 fibres and 6.1% (range = 1.05–14.9%) for an individual muscle fibre. Similarly, the mean between-day variability of the microphotometric histochemical technique of determining SDH activity was 5.9% (range = 2.6–13.9%) for an area containing ~50 fibres and 6.6% (range = 2.2–13.9%) for an individual muscle fibre. The inter-class correlation coefficient between biochemically determined SDH activity and histochemically determined SDH activity was r = 0.83 (p < 0.05). Collectively, these data demonstrate that the quantitative histochemical technique of Blanco et al. (1988) is both valid and reliable in the determination of SDH activity in skeletal muscle fibres.  相似文献

Using a variety of histochemical methods -mATPase staining after alkaline and acid preincubations, NADH-TR and alpha-MGPDH- we have investigated the fibre types in porcine skeletal muscle. The results reveal that four major fibre types -I, IIA, IIB and II*- can be separated histochemically in Longissimus lumborum muscle of Landrace pigs. The histochemical properties of the muscle fibre type II* are very similar to that of type IIX described in other mammals. The existence of IIX fibres in pig muscle has been recently demonstrated by molecular biology techniques and our results validate the use of histochemistry (mATPase) as an easy methodology to differentiate the three fast myosins (type II fibres) in pig muscle.  相似文献

Summary Maximum assayable hexokinase activities vary with the proportion of red, fast-twitch, oxidative-glycolytic and intermediate, slow-twitch, oxidative fibres in different rat skeletal muscles. The major isoenzymic form, type II hexokinase, is present throughout the intermyofibrillar sarcoplasm in all fibres but a proportion of the total activity appears to be weakly associated with mitochondria. Variations in the histochemical staining intensity between fibre types correlate with their mitochondrial content and seem to be due mainly to differences in mitochondrially-associated hexokinase activity. Changes in the strength of this association may be important in controlling increases in glucose metabolism in response to prolonged increased muscular activity while regulation of the equilibrium between free and loosely-bound forms may be an important control feature in all skeletal muscle. Type I hexokinase is a minor isoenzymic component of skeletal muscle and occurs mainly in blood vessels and nerves in the perimysia and endomysia. The majority of this isoenzyme is tightly bound to mitochondria and is not detectable in homogenates prepared in the absence of Triton X-100.  相似文献

Myoglobin content is found to be higher in skeletal than in cardiac muscle of Tursiops truncatus and Phocaena phocaena and much higher than that in skeletal muscles of terrestrial mammals. According to the myoglobin content muscle fibres are devided into five types: red, white and three intermediate types. Deep muscles contain more red fibres and less intermediate fibres than superficial ones. White fibres compose almost one half of all fibres of the superficial skeletal muscles of the dolphins. The role of myoglobin distribution and higher content in oxygen supply of muscular tissue is discussed in relation to the peculiarities of dolphin breathing and blood circulation.  相似文献

Summary In this communication, the results of applying various histochemical techniques for the localization of oxidoreductases, transferases, hydrolases and isomerases in the human heart are presented. The Purkinje fibres of the atrioventricular conducting system of the human heart differ from the myocardium proper in containing a slightly higher activity of most of the glycolytic and gluconeogenetic enzymes investigated. The relatively higher activity of 6-phosphofructokinase, the key enzyme in anaerobic carbohydrate metabolism, is especially noteworthy. On the other hand, the activities of some of the enzymes that play a part in the aerobic energy metabolism is slightly less than those in the myocardium fibres.As for the activity of the NADPH regenerating enzymes, the activity of 6-phosphogluconate dehydrogenase and malate dehydrogenase (oxaloacetate-decarboxylating) is somewhat higher, and the activity of glucose-6-phosphate dehydrogenase similar, in the Purkinje fibres compared to that in the myocardial fibres. The activity of myosin ATPase is similar for both types of fibre. Likewise, the fibres of the conducting system and of the myocardium show a similar activity of acid phosphatase, -glucuronidase, non-specific naphthylesterase and peroxidase. The neurogenic function of the conducting system of the human heart was demonstrated by the high activity of acetylcholinesterase in the Purkinje fibres and in the atrioventricular node. All these histochemical findings in Purkinje fibres are similar at widely differing levels of the conducting system.  相似文献

Summary Some histochemical parameters of the three main fibre types of rat vastus lateralis muscle were studied. Succinic dehydrogenase (SDH), creatine kinase (CK), sarcotubular ATPase (SR-ATPase) and mitochondrial ATPase activities were demonstrated in serial sections. The three fibre types, recognised by the distribution pattern of SDH activity, all show high CK activity. However, red Type I oxidative fibres when examined for ATPase and ATPase dependent CK activity, show distinct heterogeneity revealing sub-populations within the same homogeneous fibre type. Three distinct patterns were recognised in the red Type I fibres depending on the distribution of the final reaction product. The present histochemical evidence confirms the fact that subdivision of mammalian skeletal muscle into three fibre types is only approximate and probably more than three types exist.  相似文献

Staining for motor endplates and chemical digestion of five major muscles of the domestic chicken shows that these confirm the short-fibre strap muscle paradigm. The individual fibres are spindle-shaped, terminating in gradually tapering ends. The motor endplates of the individual fibres align in cross-bands along the length of the fascicles. These bands are spaced much more tightly than are comparable bands in mammals; unlike the condition in mammals, many fibres are longer than twice the interband spacing. The spacings between bands differ by more than a factor of five along the length of each muscle. The proportions among bands remain relatively constant. These proportions are not affected by the degree of muscular contraction, nor do they change with ontogeny, suggesting that the arrangement is established before hatching.  相似文献

It is suggested that ABRM, smooth muscle of Mytilus edulis L. and Mytilus galloprovincialis Lmk. (Mollusca Pelecypoda), is composed of one histochemical fibre type. The fibres are characterized by a low myofibrillar ATPase activity. Succinic and nicotinamide adenine dinucleotide oxidoreductase activities are distributed in a reverse pattern than that of the ATPase activity. Glycogen phosphorylase is richly represented in ABRM fibres and this detection is in opposition with the negative detection of alkaline phosphatase activity. These preliminary histochemical observations are similar to those found in some vertebrate smooth muscles. Mitochondrial glycerol-3-phosphate, 6-phosphogluconate, lactate and octopine dehydrogenases are not detected in muscle fibres whereas glio-interstitial tissues show weak but distinct reactivity. These last results especially characterize Mytilus catch fibres and are briefly discussed in relationship with previous physiological, biochemical and morphological observations.  相似文献

Summary When incubated for histochemical demonstration of actomyosin ATPase both the tensor tympani and the stapedius were found to contain two types of muscle fibres, one with high actomyosin ATPase activity, indicating a high speed of contraction, and one with low actomyosin ATPase activity, indicating a low speed of contraction. In the tensor tympani 59% and 41% of the muscle fibres had a high and low ATPase activity, respectively. The corresponding numbers in the stapedius muscle were 82% and 18%. These findings are discussed with reference to previous physiological and morphological studies.  相似文献

Summary Morphological, histochemical and ultrastructural characteristics of intrafusal fibre types were studied in rat muscle spindles. The existence of three intrafusal fibre types, namely the typical bag, the intermediate bag and the chain fibres was confirmed. Intrafusal fibres differ in diameter, length and number of nuclei in the equatorial zone. Histochemically, typical bag fibres exhibit both alkali-and acid-stable ATPase activity and low SDH activity. Intermediate bag fibres possess low alkali-stable ATPase activity; after acid-preincubation, however, they have low activity only in the juxtaequatorial region, whereas in the polar zones they exhibit high acid-stable ATPase activity. The SDH activity varies from moderate to high. The chain fibres exhibit high alkali-stable and low acid-stable ATPase and high SDH activity in the extensor digitorum longus muscle, whereas in the soleus muscle the acid-stable ATPase activity varies from a low one to a high one, either among individual chain fibres in one spindle, and/or repeatedly along the fibre length.Since there are regional differences in morphological characteristics and in staining properties of intrafusal fibres, a reliable identification of intrafusal fibre types can only be achieved by an analysis of serial sections.  相似文献
