The infection of apples by Phytophthora syringae   总被引:1,自引:0,他引:1  
Contamination with infected soil has led to high wastage of apples in storage due to rotting by Phytophthora syringae. At 3-3 oC lesions formed 3–4 wk after inoculation with zoospores; the percentage infection fell if the suspensions dried after 48 h at this temperature and after 22 h at 15 oC. Infected soil rotted fruit only if kept moist; at 3-3 oC a 3-day period of wetness resulted in 37-5% rotting after 8 wk. Fruit dipped in soil slurry remained wet in some parts of a 4361(12 bushel) bin for at least 3 wk. There was a 10-fold increase in rotting by contact between sound and rotting fruit after 11 wk at 3-3 oC. Captan gave effective protection against rotting derived from zoospores or infected soil; it had no eradicant action.  相似文献

In laborator experiments germinating conidia penetrated leticels and wounds but not the intact surfaces of apples. Date of harvest had no significant effect on the numbers of apples infected with Nectria galligena but the earliest picks rotted first in barn store. Inoculations of unpicked apples resulted in small arrested lesions which only developed into progressive rots after a considerable period in store. Rots developed mosy quickly from inoculations made between mid-August and mid-September. The size of rot increased with spore number and many inoculations with 10–100 conidia remained as arrested lesions. Arrested lesions developed 10–15 days after unripe apples were inoculated and consisted of a zone of fungal colonization surrounded by suberized, necrotic cells in which compounds toxic to both N. galligena and Penicillium expansum were detected. No antifungal compounds were found in progressive rots to mature apples or in healthy apples of any age. Antifungal activity, measured by inhibition of P. expansum, was greatest 15–20 days from inoculation of unripe apples with N. galligena but decreased after a total of 35 days incubation at 20 d? C. Much less antifungal activity was produced in ripe or desert apples.  相似文献

Yam is an important crop which serves as a source of income to small-holder farmers as well as a foreign exchange earner. Among the constraints faced by yam farmers are pests and diseases, especially deterioration caused by microbes during storage. Since over dependence on pesticides is being discouraged, neem seed and ginger extracts were evaluated as potential control agents against rot-causing fungi. The study was conducted in the Spanish laboratory, at the Faculty of Agriculture, University for Development Studies, Nyankpala Campus. Isolations were made from rotted yam tubers sampled from the Tamale Central market with potato dextrose agar (PDA) as the growth medium. Growth inhibition of the isolates was determined by growing pure cultures on PDA plates amended with 2?ml each of ethanol and aqueous extracts of neem seed and ginger as well as carbendazim. A pathogenicity test proved that Aspergillus flavus, A. niger, A. ochraceus and Penicillium digitatum isolated from the rotted tubers were responsible for the rot. A. niger had a significantly higher (p?<?0.05) occurrence (40%) than the others. Growth inhibitions by carbendazim and ethanol extracts of neem seed and ginger were comparable but significantly higher (p?<?0.05) than the aqueous extracts of neem seed and ginger as well as the control. However, the aqueous extracts of neem seed and ginger had a significantly higher (p?<?0.05) inhibition than the control. For instance, growth inhibition of A. niger by carbendazim, ethanol neem seed and ginger extracts were 70.5, 69.6 and 65.5%, respectively, while inhibition by aqueous neem seed and ginger extracts were 33.9 and 24.8%, respectively. Since aqueous extracts of neem seed and ginger significantly inhibited (p?<?0.05) growth of the rot-causing fungi, they can be used as surface protectants of stored tubers.  相似文献

Tecnazene (up to 33 ppm) and dichloran (up to 500 ppm) had little effect on germination of spores or growth of Fusarium solani isolated from and causing a rot of potato tubers; they also did not decrease rotting when applied to wounds later inoculated with the pathogen. Benomyl and thiabendazole (up to 500 ppm) also had little effect on spore germination but did greatly decrease growth at 5 ppm. A pronounced pink coloration developed in cultures growing slowly in the presence of benomyl; a similar though less striking effect appeared in agar cultures containing thiabendazole. Benomyl suspended in water or diluted with Fuller's earth gave good control of rotting when applied to wounds inoculated later with F. solani. Still better control was obtained with thiabendazole; dusts containing 1% a.i. substantially decreased rots and those containing 10 % a.i. gave almost complete control when applied to wounds shortly before inoculation. Thiabendazole was also very effective when used 24 h after inoculation and a fair measure of control was obtained when it was applied 24 h later. Benomyl and thiabendazole placed on apparently intact surfaces of tubers caused tissue 5 mm deep to become toxic to F. solani 10 days later, and, unexpectedly, this tissue prevented spore germination. Fuller's earth alone substantially decreased rotting. The results obtained suggest that dusts containing thiabendazole have some promise for the control of Fusarium rots of potato tubers, especially of early crops.  相似文献

The log of the time interval between inoculation with Nectria galligena in October and the onset of rotting in apples held in air was proportional to the deficit between the temperature of incubation and 25°C, but temperature did not affect the rate of subsequent rot expansion. Rots expanded equally fast whether apples were held in dry or moist air. The quantity of rotted tissue obtained after incubating inoculated apples in atmospheres containing up to 12.5% CO2 increased with increasing concentrations of CO2 greater than 2.5%. The quantity of rotted tissue obtained in apples incubated in 10% CO2 was three times as great as that obtained after incubation in air. The incidence of natural rots was lower in apples stored at 4% CO2 than in those stored in air and rotting increased with increasing concentrations of CO2 higher than 4%. Colonies of N. galligena grew faster on malt agar plates incubated in 5% CO2 than in air, but growth was slower in 10% CO2 than in air. The quantity of benzoic acid per mg hyphae accumulated in developing lesions was similarly related to the CO2 concentrations up to 2.5% but decreased at higher concentrations, and the quantities found in apples stored in CO2 concentrations >5.0% CO2 were less than in those stored in air.  相似文献

Abstract

Crude extracts of lucerne (Medicago sativa) and Lotus pedunculatus root were toxic when administered orally to third-instar Costelytra zealandica larvae. Purified saponins from lucerne root were alone found sufficiently toxic to account fully for the activity of the crude extract. Condensed tannins extracted from L. pedunculatus and L. corniculatus had no effect on larval survival when administered at levels equivalent to those present in crude extracts.  相似文献

Effects of five homologous series of amphoteric, anionic, and nonionic surfactants on growth and anthocyanin production by callus cultures of Dimorphotheca sinuata were examined. The phytotoxicity of amphoteric sulfobetaines increased with alkyl chain length and reached a plateau at 14 to 16 carbon atoms. In the case of the anionic sodium alkyl sulfates, the dodecyl derivative caused the greatest inhibition. Higher molecular weight ionic detergents were less toxic, probably due to their high Krafft temperatures and concomitant reduced solubility. The nonionic higher alcohol ethoxylates were generally less inhibitory than the ionic detergents and had little or no effect below their critical micelle concentrations. Fresh weights of tissues and anthocyanin production by Dimorphotheca callus cultures declined with increasing surfactant concentrations.  相似文献

A biocontrol fungus isolated from rotting wood was identified as a Trichoderma strain (named as Trichoderma sp. ZH1) by internal transcribed spacer (ITS) sequences of rRNA genes. The laccase yield of Trametes sp. AH28-2 in cocultivation with Trichoderma sp. ZH1 reached 6,210 U l−1, approximately identical to those induced by toxic aromatic inducers. Cocultures maintained 60–70 % of their highest laccase activity obtained at 5 days after inoculation of the biocontrol fungus, at least for 20 days. Furthermore, a novel laccase isozyme (LacC) was obtained through the fungal interactions. The molecular weight of LacC is about 64 kDa, and its isoelectric point is 6.6. The temperature and pH optimum for LacC to oxidize guaiacol are 55 °C and 5.0, respectively. LacC is stable both at 60 °C and pH 4.0–8.0. Furthermore, the K m values of LacC for various substrates were also determined. Our work demonstrates a safe strategy for the production of industrial laccases, instead of the traditional method of chemical induction.  相似文献

Analysis of cell-wall polymers during cotton fiber development   总被引:2,自引:0,他引:2  
Although the fibers of cotton (Gossypium hirsutum L.) are single cells with a secondary wall composed primarily of cellulose, the cell-wall polymers of the fibers are technically difficult to characterize with respect to molecular weights. This limitation hinders understanding how the fiber wall composition changes during development, particularly with respect to genotypic variations, and how the molecular composition is related to physical properties. We analyzed cell-wall polymers from cotton fibers (cultivar, Texas Marker-1) at several developmental stages (8–60 days post-anthesis; DPA) by gel-permeation chromatography of components soluble in dimethyl acetamide and lithium chloride. This procedure solubilizes fiber cell-wall components directly without prior extraction or derivatization, processes that could lead to degradation of high-molecular-weight components. Cellwall polymers from fibers at primary cell-wall stages had lower molecular weights than the cellulose from fibers at the secondary wall stages; however, the high-molecularweight cellulose characteristic of mature cotton was detected as early as 8 DPA. Fungi associated with crown rotting were isolated from 3 year-old lucerne plants growing in sandy and loam soils in an irrigated stand, declining in productivity, at Langhorne Creek, South Australia. The method of isolation markedly affected the range of species recovered. Fusarium spp. and Phomopsis sp. were isolated from washed and surface sterilized crowns, whereas a larger range of fungi, especially Acrocalymma medicaginis, was isolated from unwashed crowns. A. medicaginis was most frequently isolated from rotted tissue flecked with red, while a species of Phomopsis was the dominant fungus in whitish tissue containing zone lines, especially in crown branches. A. medicaginis and Phomopsis sp. caused crown rotting of mature, wounded lucerne plants at temperatures equivalent to summer conditions at Langhorne Creek. There was no evidence that Fusarium oxysporum or F. solani, two of the most frequently isolated species, were pathogens of mature lucerne plants.  相似文献

【目的】近年来,纳米银由于其自身独特的抗菌活性而受到越来越多的关注,有研究表明纳米银是一种广谱的抗菌剂,其对数十种致病微生物都有强烈的抑制和杀灭作用。相较于传统的合成方法而言,具有反应条件温和、环境友好等优势的生物合成法是目前的研究热点。【方法】本研究利用真菌Mariannaea sp. HJ的胞内提取物合成纳米银,并对其合成条件进行优化调控,还进一步考察了合成的纳米银颗粒的抗菌性能。【结果】胞内提取物浓度350 mg/L、AgNO_3浓度5 mmol/L、pH 7.0为菌株HJ胞内提取物合成纳米银的最优条件;TEM图像表明合成的纳米银颗粒主要为球形和伪球形,分散性良好,无明显的团聚现象;XRD表明合成的纳米银晶体结构为面心立方体结构;通过FTIR分析结果推测提取物中的羟基、羧基等官能团可能参与了纳米银的还原和稳定过程。此外,在本实验条件下合成的纳米银颗粒对革兰氏阴性菌Escherichia coli BL21和革兰氏阳性菌Arthrobacter sp. W1都有较好的抗菌活性。【结论】真菌Mariannaea sp. HJ胞内提取物能合成尺寸均一且分散性良好的球形纳米银颗粒,合成的纳米银颗粒在抗菌方面具有潜在的研究价值。  相似文献

An inhibitor of collagenase was released from cultured cervical tissue of sheep and the amounts released were greatest from tissue in late pregnancy (145-146 days). The molecular weights of material with inhibitory activity, estimated by gel filtration of extracted inhibitor, at different stages of pregnancy were different, i.e. 20,000 from extracts of late-pregnant and post-partum samples and 29,000 from extracts of non-pregnant samples. Inhibitor from culture supernatants had a molecular weight of 42,000. We conclude that the inhibitor has binding characteristics and molecular weights similar to those of the tissue inhibitor of metalloproteinase.  相似文献

To determine the relative importance of seed tubers and stems as sources of inoculum for potato gangrene in progeny tubers, different levels of inoculum of Phoma exigua var. foveata were established in field experiments by planting rotting or contaminated seed tubers and by inoculating stems shortly before haulm desiccation. The pathogen was only occasionally detected by isolation from inside green stems in June and July on plants growing from contaminated but unrotted seed. The incidence of pycnidia on desiccated stems increased with increasing concentration of inoculum contaminating seed tubers and with increasing time interval between desiccation and harvest. Stem infection was probably derived from inoculum on seed tubers spreading via the soil to the stem bases. Stem inoculation of plants growing from uninoculated or uncontaminated seed greatly increased both the gangrene potential of progeny tubers (defined as % wounds with gangrene after uniformly damaging tubers and storing them at 5°C for 12 wk), and tuber contamination (defined as % wounds with gangrene after spreading tuber-borne soil onto test tuber slices and storing them at 5°C for 8 wk). However, when stems of plants growing from rotting or contaminated seed were cut at ground level and removed before desiccation, gangrene potential of progeny tubers was only slightly less than that of tubers from untreated plots. There was no evidence that soil inoculum or aerial spread played a significant role in disease development. Gangrene potential and contamination of progeny tubers were related to Contamination levels on seed tubers. Some transmission also occurred from rotted seed tubers to progeny. Inoculum levels around progeny tubers increased rapidly after desiccation even in plots where stems had previously been cut at soil level and removed to eliminate pycnidial development above ground as a source of inoculum. Both gangrene potential and contamination of progeny tubers were greater on cv. Ulster Sceptre than on cv. Pentland Crown. The results showed that the inoculum on seed tubers, whether from rots or surface contamination, contributed more to the contamination of progeny tubers at harvest than did the inoculum from pycnidia on stems following desiccation of the haulm.  相似文献

Wood is an organic material that is a source of carbon of organisms called Wood-decay fungi, and to preserve the wood, various toxic compounds to man and the environment have been used. To analyze the effect of N,N-Dimethylhexadecylamine (DMHDA) on wood attacked by the rotting fungus Trametes versicolor L. We used an in vitro system to expose the fungus T. versicolor to different concentrations of the DMHDA (50, 150 and 450 μM). We quantified the diameter of mycelial growth and laccase activity, also, under these experimental conditions we studied morphological details of the organisms using different scanning equipment including scanning electron microscopy. The growth of T. versicolor exposed to DMHDA for 60 days, showed a concentration-dependent dose behavior, also, the electron microscopy analysis revealed that the morphology and mycelial density was affected by the DMHDA, showing a formation of atypical morphological and thickener folds. Finally, the pieces of wood treated with DMHDA and exposed to the fungus had a lower mass loss, after a period of 60 days of exposure, the values obtained were 0.7, 1.0 and 0.5 g of mass lost for the control, LoC and LoDMHDA treatments respectively. Wood-rot fungi have represented economic losses worldwide, the strategies used have been supported by toxic compounds for the environment. The DMHDA both in the Petri dish system and as a wood preservative was shown to significantly inhibit the growth of T. versicolor.  相似文献

Abstract

An experiment was conducted to study the effects of interaction between Meloidogyne incognita and Fusarium solani on plant length, fresh and dry weights, number of pods, chlorophyll, carotenoid, nitrogen and phosphorus contents and nitrate reductase activity in lentil plants. The results reveal a maximum damage occurring in all the plant growth, biochemical and nutrient parameters, in plants inoculated with M. incognita 10 days prior to F. solani (Mi?→?Fs). This was followed by simultaneous (Mi?+?Fs) inoculations, fungus inoculation 10 days prior to nematode (Fs?→?Mi), M. incognita alone and F. solani alone treatments. Nematode reproduction factor and root galling were highest in individual inoculation of M. incognita, while root rotting percentage was highest when nematode was inoculated 10 days prior to fungus followed by simultaneous inoculation with both nematode and fungus.  相似文献

Maintenance of pH 7.0 during the fermentation period favors accumulation of high molecular weight polysaccharide-containing components called lipopolysaccharide–protein and polysaccharide–lipid complexes in the capsules and culture medium. Increased pH of the culture medium to 8.0 reduced the period of exponential growth and the yield of polysaccharide-containing complexes as compared to optimal conditions. Maintenance of pH 5.5 suppressed the culture growth and polysaccharide production. The polysaccharide–lipid complexes obtained when pH was stabilized at the level of 7.0–8.0 had relatively low molecular weights and included only acidic polysaccharides. The use of potassium gluconate instead of sodium malate as a source of carbon in the culture medium changed the polysaccharide composition and increased the content of glucosamine, which increased the affinity of polysaccharides for wheat germ agglutinin. Prolongation of Azospirillum cultivation to five days introduced new glucose-containing polysaccharide components in the capsule.  相似文献

Abstract

The objective of this study was to evaluate the physico-chemical and antimicrobial properties of a dual polymerization experimental endodontic sealer (E) and experimental sealers containing dibutyltin methacrylate (Sn2+) (ETs) or calcium methacrylate (Ca2+) (ECs). The pH and ion release levels of the sealers were measured. The dimensional stability was evaluated in accordance with ISO 6876. Biofilm growth inhibition was evaluated using confocal laser scanning microscopy (CLSM). Biofilm viability analysis was performed using the SYTO 9 technique. The shelf life was evaluated through the degree of conversion and film thickness tests after the sealers had been stored for different periods of time. For statistical analysis, ANOVA and Tukey’s post hoc test were used, with a significance level of 5%. ETs revealed better anti-biofilm potential after 15?days than that of the controls. The degree of conversion was reduced after the shelf-life period. The addition of calcium and dibutyltin methacrylate improved the anti-biofilm properties of the experimental endodontic sealer without impairing their physico-chemical properties.  相似文献

Pure crystals (at least 99% purification) of sigma-endotoxin were isolated from Bac. thuringiensis var. galleriae. The complete dissolution of crystals might be achieved by the increase of pH up to 12 and higher or by a combined action of S = S-reducing and denaturing agents. Electrophoresis of the solubilized crystal proteins in 5% polyacrylamide gels containing 0,1% sodium dodecyl sulfate and 8 M urea reveals two major bands corresponding to molecular weights of 120000--140000 (65%) and 65000 (8-10%), and some minor components whose molecular weights varied from 65000 to 340000. Urea (3--8 M) causes to partial dissolution of the crystals; the component with molecular weight of 65000 is mainly found in the solution (component A). In dithioerythritol extracts at pH 9 the major component of the crystal is the protein with molecular weight 120000--140000 (component B). The crystals, alkali-soluble components and proteins isolated from crystals by selective extraction (3--8 M urea or 0.01 M dithioerythrytol, pH 9) were found toxic for the larvae of Galleria mellonella.  相似文献

A bioassay and an empirically derived formula were developed to quantify fungitoxic effects. This bioassay can be easily performed and objectively read, and it is suitable for low-volume samples originating from aqueous or organic solvents. The formula defines the Inhibition Index (I), a single value that incorporates both the response to concentrations of the inhibitory compound and the persistence of inhibition. Antagonistic efficacy of metabolites produced by biocontrol strains of Trichoderma spp. were measured based on inhibition of growth of Rhizoctonia solani. Although the bioassay itself was not influenced by pH or light conditions, these factors affected metabolite production or activity. Aqueous extracts from light-grown germlings of Trichoderma virens inhibited R. solani more than extracts from germlings grown in the dark. Low pH increased the inhibitory activity of extracts from T. virens. Tests of fungal strains developed for biocontrol demonstrated that the bioassay reflected their activity both in the field and in other in vitro tests. The bioassay and formula are readily adapted for use with other fungi.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献
