Background
Independent studies have shown that several single nucleotide polymorphisms (SNP) in the human FTO (fat mass and obesity associated) gene are associated with obesity. SNP have also been identified in the pig FTO gene, among which some are associated with selected fat-deposition traits in F2 crosses and commercial populations. In this study, using both commercial pig populations and an experimental Meishan × Pietrain F2 population, we have investigated the association between one FTO SNP and several growth and carcass traits. Association analyses were performed with the FTO polymorphism either alone or in combination with polymorphisms in flanking loci.Methods
SNP (:g.400C>G) in exon 3 of porcine FTO was genotyped by PCR-RFLP and tested for associations with some growth, carcass and fat-related traits. Linkage analysis placed FTO on the p arm of pig chromosome 6, approximately 22 cM from RYR1. In the commercial populations, allele C of the FTO SNP was significantly associated with back fat depth and allele G with muscling traits. In the Meishan × Pietrain F2 pigs, heterozygotes with allele C from the Pietrain sows and allele G from the Meishan boar were more significantly associated with fat-related traits compared to homozygotes with allele G from the Pietrain and allele G from the Meishan breed. In single- and multi-locus models, genes RYR1, TGFB1 and FTO showed high associations. The contribution in genetic variance from the polymorphism in the FTO gene was highest for back fat depth, meat area on the musculus longissimus lumborum et thoracis tissues and metabolite glucose-6-phosphate dehydrogenase.Conclusions
Our results show that in pig, FTO influences back fat depth in the commercial populations, while in the Meishan × Pietrain F2 pigs with a CG genotype, heterosis occurs for several fat-related traits. 相似文献2.

The apolipoprotein E (ApoE) gene is an important component of plasma lipoprotein, and Fas apoptosis inhibitory molecule (FAIM) is a novel anti-apoptotic gene. In this study, we researched and discussed seven genes in eight different tissues in Qinchuan cattle by quantitative Real-time PCR. The result of analysis showed that ApoE and FAIM 2 genes had a correlation with muscle and fat. PCR–RFLP was applied to analyze the genetic variations of the ApoE and FAIM 2 genes and verify the effect on growth and carcass traits in a total of 365 Qinchuan cattles. The result of haplotype analysis showed that nine different haplotypes were identified among the four SNPs in ApoE and FAIM 2 genes. The statistical analyses indicated that the four SNPs were significant association with growth and carcass traits (P < 0.05, N = 365); and the four SNPs were significant association between nine combined genotypes of candidate genes and growth and carcass traits. Taken together, our results provide the evidence that polymorphisms in candidate genes are associated with growth and carcass traits in Qinchuan cattle, and may be used as a possible candidate for marker-assisted selection and management in beef cattle breeding program. 相似文献

MYH3, whose function is to convert chemical energy to mechanical energy through ATP hydrolysis, is mainly expressed in skeletal muscle at various stages and is indispensable in the procedure of development of skeletal muscle and heart. In the study, genetic variations and genotypes of MYH 3 gene in a total of 365 Qinchuan cattles were analyzed by polymerase chain reaction-restriction fragment length polymorphism, as well as verified the effect on growth and carcass traits. After PCR products were digested by restriction enzymes, eight SNPs were identified and individuals were genotyped. It showed that the SNPs at nucleotides were all in low linkage disequilibrium, therefore no dominated haplotype was found in the population. The result of statistic analysis indicated seven SNPs were significantly associated with growth and carcass traits (P < 0.05, N = 365) except locus G13791A. To sum up, the result of the study proved that polymorphisms in MYH3 gene are associated with the growth performance of Chinese Qinchuan cattle, so the variations of the gene could be used as possible molecular assisted-makers in the beef cattle breeding program and management. 相似文献

The aim of the conducted study was to evaluate correlation between genotypes and PRKAG3 compound genotypes and milk production traits (yield of milk, milk fat and milk protein, and protein and fat content in milk). The study covered a herd of 180 Jersey cows. PCR-RFLP method was used for genotyping. The frequencies of alleles that occur mostly and combined genotypes were as follows: T1526G G - 0.57, G 1609A G - 0.92 and for T1526G/G1609A TG/GG - 0.54. The results obtained in the study demonstrated the correlation between analyzed genotypes and selected milk production traits; however they are not statistically significant. 相似文献

In our study, genetic variation in coding region of cattle CCAAT enhancer binding protein alpha(namely CEBPA)gene was detected by PCR-RFLP and DNA sequencing methods in 215 individuals from Qinchuan cattle breed. Two haplotypes (A and B) and three observed genotypes (AA, AB, and BB) were detected. The result of DNA sequence showed one mutation by comparisons with NC_007316. The mutation at nt963 (T>G) were located in coding region of the CEBPA gene. Associations between the CEBPA gene genetic variation and the carcass traits were revealed in Qinchuan cattle. Least squares analysis revealed a significant statistical effect of the CEBPA gene different genotypes on slaughter weight and carcass weight in Qinchuan cattle. Individuals with BB genotype showed higher slaughter weight and carcass weight than individuals with AA and AB genotypes. Therefore, these results suggest that the CEBPA gene is a strong candidate gene that affects carcass traits in Qinchuan cattle. 相似文献

The insulin-induced gene 1 (Insig-1) is a regulator of lipid metabolism and plays an important role in the sterol-mediated regulation of SREBP, SCAP and HMG-CoA reductase. We used PCR-RFLP and DNA sequencing to detect polymorphisms of the Insig-1 gene in 215 individuals of the Qinchuan cattle breed. Four SNPs [4366(A>G), 4534(T>C), 5001(T>C), and 5235(G>A)] were indentified. The association of the genetic viariation with growth and carcass traits (body length, withers height, hip width, slaughter weight, and carcass weight) was analyzed. The individuals with better performance had the GG genotype at locus A4366G, and CC genotypes at locus T4534C and locus T5001C. These could be used for beef cattle breeding improvement in China. Additionally, linkage disequilibrium analysis reflected that all mutations were in low linkage disequilibrium with each other. We concluded that polymorphisms in the Insig-1 gene are associated with growth and carcass traits and could be used for marker-assisted selection and management in beef cattle breeding programs. 相似文献

The objectives of the present study were focused on detecting deletion mutation in bovine AMPD1 gene, and analyzing its effect on body measurement and carcass traits in Qinchuan cattle by using DNA sequencing and agarose electrophoresis methods. The 198-bp PCR products of AMPD1 gene exhibited three genotypes and two alleles were revealed: A and B. The frequencies of genotype AA/AB/BB in Qinchuan populations was 0.7163, 0.2233 and 0.0605. The χ2-test analysis demonstrated that the breed was not in agreement with Hardy–Weinberg equilibrium (P < 0.05). The association of the 18-bp deletion mutation of AMPD1 gene with body measurement and carcass traits of Qinchuan cattle were analyzed. The cattle with AA genotype had slaughter weight and carcass weight than those with genotype AB (P < 0.01 or P < 0.05). These results suggest that the 18-bp deletion mutation may influence the carcass traits in Qinchuan cattle. 相似文献

为探讨鸡视网膜母细胞瘤基因1(Retinoblastoma1,RB1)多态性对体重性状的影响,文章以东北农业大学高、低脂双向选择品系肉鸡为实验材料,采用MALDI-TOF-MS、PCR-RFLP方法进行基因多态性检测和个体基因型分析,共获得27个SNP位点的基因型数据。采用滑动窗口法构建单倍型,进而利用单位点和单倍型分别与鸡体重性状进行关联分析。结合单位点和单倍型分析结果,确定了RB1基因上4个显著影响1周龄体重的SNP位点,2个显著影响1、3周龄体重的SNP位点。研究结果表明RB1基因是影响鸡早期体重性状的重要候选基因。 相似文献

The growth hormone receptor (GHR) is a membrane transmitter for the growth hormone signal transduction pathway that regulates various metabolic activities, including cell growth and expressions of cytokine genes. The presence or absence of a genetic polymorphism for the LINE-1 retroposon in the PI promoter, which specifically regulates theGHR gene expression in the liver, was screened by a novel detection method and examined for its relationships with carcass traits in Hanwoo cattle. Han woo cattle had taurine type LINE-1 present (alleleI) as well as incidine type LINE-1 absent (alleleA) promoter sequences. Three genotypes,I/I, I/A andA/A, showed frequencies of 49.1, 36.7 and 14.2%, respectively. The effects of allele A were significant on mean differences for final weight, eye muscle area, marbling score and fat color (p<0.05), but not for carcass weight, backfat thickness, final meat quality grade or meat color (p>0.05). Most 30-month old Hanwoo steers bearing the LINE-1 absent promoter had whiter fat color, heavier live weight and higher marbling score, reflecting intramuscular fat deposition inM. longissimus dorsi, compared to animals bearing a LINE-1 present promoter. This suggests that aGHR polymorphism could be a potential genetic marker for improving beef production of Hanwoo cattle. 相似文献

Wnt7a is a member of the WNT gene family, which encodes secreted signaling proteins and responds to many biological processes. Specifically Wnt7a influences satellite stem cells and regulates the regenerative potential of the muscle. However, similar researches about the bovine Wnt7a gene are lacking. Therefore, in this study, polymorphisms of the bovine Wnt7a gene were detected in 488 individuals from Chinese Qinchuan cattle by DNA pooling, forced PCR-RFLP, and DNA sequencing methods. 3 novel SNPs were identified, two SNPs (g.T4926C and g.A21943G) were in the intron and the last one (g.C63777T) was in the exon. Five haplotypes involved in these three variant sites in the Wnt7a gene were identified and their effects on growth traits were analyzed. The results revealed that haplotype 1 had the highest haplotype frequencies and was highly significantly associated with body height (P < 0.01), body weight (P < 0.05), chest width (P < 0.05) and height at hip cross (P < 0.01) respectively. 相似文献

We analyzed 20 exons, with their intron-exon boundaries, of the bovine Flt-1 gene, using a strategy combining PCR amplification and single-strand conformational polymorphism analysis (PCR-SSCP), followed by nucleotide sequence analysis, in 675 cattle. We then looked for associations between polymorphisms and growth traits. Twelve novel SNPs (ss#184956516, ss#184956517, ss#184956518, ss#184956519, ss#251343993, ss#251343994, ss#251343995, ss#251343996, ss#251343997, ss#251343998, ss#251343999, and ss#251344000) were detected in the bovine Flt-1 gene in all three breeds. We observed no significant associations between these polymorphisms and birth weight, body weight and average daily gain during different growth periods (6, 12, 18, and 24 months old) (P > 0.05), or in body height, body length, heart girth, or height at the hip in Nanyang cattle breeds. 相似文献

DNA methylation is essential for adipose deposition in mammals. We screened SNPs of the bovine DNA methyltransferase 3b (DNMT3b) gene in Snow Dragon beef, a commercial beef cattle population in China. Nine SNPs were found in the population and three of six novel SNPs were chosen for genotyping and analyzing a possible association with 16 meat quality traits. The frequencies of the alleles and genotypes of the three SNPs in Snow Dragon beef were similar to those in their terminal-paternal breed, Wagyu. Association analysis disclosed that SNP1 was not associated with any of the traits; SNP2 was significantly associated with lean meat color score and chuck short rib score, and SNP3 had a significant effect on dressing percentage and back-fat thickness in the beef population. The individuals with genotype GG for SNP2 had a 25.7% increase in lean meat color score and a 146% increase in chuck short rib score, compared with genotype AA. The cattle with genotype AG for SNP3 had 35.7 and 24% increases in dressing percentage and 28.8 and 29.2% increases in back-fat thickness, compared with genotypes GG and AA, respectively. Genotypic combination analysis revealed significant interactions between SNP1 and SNP2 and between SNP2 and SNP3 for the traits rib-eye area and live weight. We conclude that there is considerable evidence that DNMT3b is a determiner of beef quality traits. 相似文献

A total of 129 Limousin calves were used to investigate how leptin gene polymorphisms affect growth traits, such as body weight,
average daily gain, wither height, sacrum height and chest girth in beef cattle. Two single nucleotide polymorphisms (SNPs)
were genotyped, including the Sau3AI polymorphism in intron 2 and the A59V polymorphism in exon 3. The genotype and allele frequencies for each SNP and the
haplotype frequencies for both SNPs were estimated in the studied herd. Statistical analysis revealed that the A59V polymorphism
significantly affected the body weight at 210 days of age (P ≤ 0.01) and the average daily gain between 3 and 210 days of age (P ≤ 0.05) with T as a desirable allele. No associations were observed between the Sau3AI polymorphism and the growth traits mentioned above. However, the average daily gain between 3 and 210 days of age was significantly
higher in the CT/CT haplotype animals compared with the CC/CC and CC/CT individuals. These results indicated that selection for the A59V TT animals might contribute to an improved body weight in Limousin cattle.

A single nucleotide polymorphism was identified in the coding sequence of the bovine transferrin gene. Two alleles (SSCP1 and SSCP2) were detected by SSCP analysis and the mutation point was identified and confirmed by direct sequencing of the PCR products. The relationship between protein and DNA polymorphism was established. Protein variants A, D1 and E correspond to SSCP allele 1 and variant D2 corresponds to SSCP allele 2. DNA sequences from genotypes AA, AE, AD2, D1E, D2E and D2D2 reveal an A/G substitution at position 1455 of the cDNA which causes a Gly/Glu substitution which could be responsible for the mobility difference between D1 and D2 variants. Because of the number of variants, this suggests that other SNPs exist in the bovine transferrin gene. A linkage analysis between the SSCPs and two microsatellites (UWCA46 and CSSM019) mapped the transferrin gene to BTA1. The objective of this research were to detect bovine Dickkopf 2 (DKK2) gene polymorphism and analyze their associations with body measurement traits (BMT) and meat quality traits (MQT) of animals. Blood samples were taken from a total of 541 Qinchuan cattle aged from 18 to 24 months. Polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) was employed to find out DKK2 single-polymorphism nucleotide (SNPs) and to explore their possible association with BMT and MQT. Sequence analysis of DKK2 gene revealed 2 SNPs (C29 T and A169C) in 5′ untranslated region (5′UTR) of exon 1.C29T and A164T SNPs are both synonymous mutation, which showed 2 genotypes namely (CC, CT) and (AA and AC), respectively. Association analysis of polymorphism with body measurement and meat quality traits at the two locus showed that there were significant effects on CT, BL, RL, PBW, BFT, LMA, and IFC. These results suggest that the DKK2 gene might have potential effects on BMT and MQT in Qinchuan cattle population and could be used for marker-assisted selection. 相似文献

为研究阿片黑皮质素前体(POMC)在动物采食和能量平衡调控中发挥重要作用, 文章采用PCR-SSCP结合DNA测序方法, 对秦川牛、南阳牛、郏县红牛、晋南牛、鲁西牛、安格斯牛和荷斯坦奶牛共计480头个体POMC基因的多态性进行研究, 并分析了多态位点与南阳牛生长性状的相关性。结果表明, 牛POMC基因3个位点中, 在3′侧翼区P3位点新发现3个连锁存在的SNP(811845 C>T、811821 T>C和811797 A>G, 与NW_928357对照)。POMC基因3′侧翼区多态位点与南阳牛6月龄体重和0~6月龄平均日增重显著相关, BB型个体显著大于AA型(P<0.05)。 相似文献

Angiopoietin-like protein 6 (ANGPTL6), which plays an important role in angiogenesis and energy metabolism as a circulating orphan peptide secreted by liver, could produce a potential effect on growth and development of animals. The objective of this study was to detect genetic variations of ANGPTL6 gene in 732 Qinchuan cattle, as well as to analyze their associations with growth traits and carcass weight. Herein, three novel mutations (T2359C, C2403A and G3258T) were identified in cattle for the first time. Chi-square test showed T2359C and C2403A loci were in Hardy-Weinberg equilibrium except G3258T locus. Haplotype with TCG (wild type) was dominant with frequency of 40.0 % among eight different haplotypes. Statistical analysis showed body height (BH) and hucklebone width (HW) of individuals with genotype CC were significant higher than other genotypes in T2359C locus (P < 0.05). In the C2403A locus, chest girth (CG), chest depth (CD), and chest breadth (CB) of individuals with genotype CA were all extremely significant higher (P < 0.01) while BH was significant higher (P < 0.05) than genotype CC. The association analysis of combined sites showed BH, CG, CB and HW of individuals with combined genotype TC-CA were significant higher than other combined genotypes (P < 0.05) in ScaI-VspI site. BH, CG and CB of individuals with genotype CA-GG were significant higher (P < 0.05) as well as CD was extremely significant higher (P < 0.01) than other genotypes in VspI-RsaI site. For ScaI-VspI-RsaI site, CD and HW of individuals with genotype TC-CA-GG were significant higher (P < 0.05) than those of other combined genotypes. Slaughter test also revealed that the effect of the combined genotypes was extremely significant on the carcass weight. Carcass weight of individuals with genotype TT-CA and TT-CA-GT were extremely significant heavier than other combined genotypes (P < 0.01) in the ScaI-VspI and ScaI-VspI-RsaI sites. And those with genotype TT-GT of ScaI-RsaI were significantly heavy (P < 0.05) as well as the individuals with genotype CA-GT of VspI-RsaI. In conclusion, the present results provided evidence that polymorphisms of ANGPTL6 gene were associated with growth and slaughter traits, and may apply to Qinchuan cattle breeding program as a possible candidate for marker-assisted selection (MAS). 相似文献
