Antioxidant system's state of erythrocytes and tissues in rats under normal and cold stress conditions was studied. Intraperitoneal injection of exogenic DSIP at the dose of 12 mkg/100 g body weight both, to intact and to cold-exposed animals results in the increase of superoxide dismutase, catalase, glutathione peroxidase, glutathione reductase activities and concentration of glutathione in red blood cells, liver and brain.  相似文献

Effect of stress on the antioxidant enzymes and gastric ulceration   总被引:6,自引:0,他引:6  
The effect of cold-restraint stress on the antioxidant enzymes of the rat gastric mucosa was studied with a view to finding out their role in stress induced gastric ulceration. Histological examination revealed stress induced extensive damage of the surface epithelial cell with lesions extending upto submucosa in some cases. Stress causes time-dependent increase in histamine and pepsin content but decrease in acid content of the gastric fluid with the progress of ulceration (ulcer index) for two hours. The tissue lipid peroxidation was significantly increased as evidenced by accumulation of malondialdehyde. Since lipid peroxidation results from the generation of reactive oxygen species, stress effect was studied on some antioxidant enzymes such as superoxide dismutase, peroxidases and prostaglandin synthetase as a function of time. The time dependent increase in stress ulcer correlates well with the concomitant increase in superoxide dismutase activity and decrease in peroxidase and prostaglandin synthetase activity. This creates a favourable condition for accumulation of endogenous H2O2 and more reactive hydroxyl radical (OH·). Administration of antioxidants such as reduced glutathione or sodium benzoate prior to stress causes significant decrease in ulcer index and lipid peroxidation and protection of gastric peroxidase activity suggesting the involvement of reactive oxygen species in stress induced gastric ulceration. This is supported by thein vitro observation that OH· can also inactivate peroxidase and induce lipid peroxidation. Changes of carotenoids and anthocyanins content, lipid peroxidation, and activity of antioxidant enzymes were studied in wild type and tocopherol-deficient lines vte1 and vte4 of Arabidopsis thaliana subjected to 200 mM NaCI during 24 h. The salt stress enhanced the intensity of lipid peroxidation to different extent in all three plant lines. Salt stress resulted in an increase of carotenoid content and activity of catalase, ascorbate peroxidase, guaiacol peroxidase and glutathione reductase in wild type and tocopherol-deficient vte1 mutant. However, the increase in anthocyanins concentration was observed in vte1 mutants only. In vte4 mutant, which contain gamma-tocopherol instead of alpha-tocopherol, the response to salt stress occurred via coordinative action of superoxide dismutase and enzymes of ascorbate-glutathione cycle, in particular, ascorbate peroxidase, glutathione reductase, dehydroascorbate reductase, and glutathione-S-transferase. It can be concluded, that salt stress was accompanied by oxidative stress in three studied lines, however different mechanisms involved in adaptation of wild type and tocopherol-deficient lines to salt stress.  相似文献

为了解褐环乳牛肝菌Suillus luteus对樟子松Pinus sylvestris var. mongolica和油松Pinus tabulaeformis根际土壤真菌群落结构的影响,采用Illumina MiSeq高通量测序,对两种松树接种S. luteus与对照组的根际土壤真菌群落结构进行研究。结果显示,4个样品共获得原始序列347 681条,归为5个真菌门。综合各土壤样品真菌Alpha多样性指数及OTUs-Venn图,发现接种S. luteus后樟子松和油松根际土壤真菌相对丰度与对照存在一定差异。群落结构分析表明,接种S. luteus提高了樟子松和油松根际土壤担子菌和壶菌的相对丰度,抑制了子囊菌门真菌。优势属由原来的Geopora转变为Suillus。通过PCA分析与NMDS分析发现接种S. luteus后樟子松和油松根际土壤真菌群落差异性增加,且接种S. luteus后樟子松根际土壤真菌群落结构相似性显著高于油松根际土壤真菌群落结构相似性。接种S. luteus的樟子松和油松土壤真菌丰富度降低、多样性增加,接种S. luteus改变了樟子松和油松根际土壤真菌群落结构和优势真菌种群。试验结果为樟子松和油松促生真菌菌种筛选和育苗工作提供了依据。  相似文献

Cadmium stress in sugar cane callus cultures: Effect on antioxidant enzymes   总被引:5,自引:0,他引:5  
Catalase (CAT) and superoxide dismutase (SOD) are antioxidant enzymes which are important in the metabolism of reactive oxygen species (ROS), and can be induced by environmental stresses including cadmium (Cd), a heavy metal toxic to living organisms. Sugar cane (Saccharum officinarumL.) in vitro callus cultures were exposed to CdCl2 and the activities of CAT and SOD were analysed. Lower concentrations of CdCl2, such as 0.01 and 0.1 mM caused a significant increase in callus growth, whereas 0.5 and 1 mM CdCl2 strongly inhibited growth of the callus cultures, but only after 9 days of CdCl2 treatment. Red-brown patches were also observed in calluses exposed to 0.5 and 1 mM CdCl2. Calluses grown in 0.01 and 0.1 mM CdCl2 did not exhibit any changes in CAT activity even after 15 days of growth in the presence of CdCl2. However, for calluses grown in higher concentrations of CdCl2 (0.5 and 1 mM), a rapid increase in CAT activity was detected, which was 14-fold after 15 days. Furthermore, up to five CAT isoforms were observed in callus tissue. Total SOD activity did not exhibit any major variation. One Mn-SOD and two Cu/Zn-SOD isoenzymes were observed in callus cultures and none exhibited any variation in response to the CdCl2 treatments. The results suggested that in sugar cane callus cultures, CAT may be the main antioxidant enzyme metabolizing H2O2.  相似文献

I. p. administration of exogenous delta-sleep-inducing peptide (DSIP) decreased the amount of diene conjugates and Schiff bases in the liver and brain in rats. The xanthine oxidase activity, at that, did not change. Cold stress enhanced the xanthine oxidase activity well as the amount of diene conjugates and Schiff bases. Preliminary administration of the delta-sleep-inducing peptide to cold-exposed animals diminished the xanthine oxidase activity and lipid peroxidation in the liver and brain. Protective effects of the DSIP under stress is discussed.  相似文献

Ethanolic sprays of gibberellins were applied to developing shoots of about 12-year-old Scots pine (Pinus sylvestris L.) grafts during the shoot elongation period in two consecutive years. Both male and female flowering was increased by these treatments in both years. The effect was particularly distinct in male flowering. However, different clones showed varying responses to the treatment. This variation was associated with both the genotype and the environmentally determined year-to-year fluctuation in flowering. Differences among clones were analysed further by introducing a model earlier developed for comparisons of the growth rhythm in various woody and herbaceous species.  相似文献

Particulate membrane preparations have been isolated from cambial cells, and from differentiating and differentiated xylem cells of the main stem of pine trees. These preparations synthesise a 14 glucomannan from guanosine 5-diphosphate-mannose. The polysaccharide and the synthase have been characterized and the Km and Vmax for the synthase determined as 85 M and 52.9 M·min-1, respectively. The enzymic activity was inhibited by the addition of guanosine 5-diphosphate-D-glucose so that the presence of an epimerase on the particulate fraction in conjunction with the synthase probably allowed the heteropolymer to be formed with the optimal ratio of the concentrations of the nucleoside-diphosphate sugar donors. No evidence for a polyprenyl-phosphate derivative as an intermediate during the polymer synthesis was obtained. Part of the control mechanism for the deposition of the large amounts of the glucomannan during the secondary thickening of the tracheids of the vascular system is by an increase in the amount of synthase activity at the endomembrane system of the cells. 目的 研究我国南海区某海洋钻井水基钻井液对诸氏鲻虾虎鱼抗氧化酶的影响.方法在19.75~158 mg/L浓度下,对3月龄诸氏鲻虾虎鱼染毒,并用试剂盒法测定超氧化物歧化酶(SOD)和过氧化氢酶(CAT)的活性.结果 SOD对水基钻井液的毒性暴露不太敏感,在染毒第1天只有最低浓度组受到一定程度的抑制,染毒第2 天各浓度组酶活性均受到一定程度的诱导(12.4%~25.5%),这种诱导作用在第4天即有所减弱,之后酶活性的变化变缓.CAT则对水基钻井液表现出较强的敏感性.染毒第1 天,各组酶活性均受到明显的诱导,并且各组酶活性表现出明显的浓度效应关系.最高浓度组酶活性最大诱导率达到97%(P>0.05).随后虾虎鱼CAT酶活性逐渐下降,第7 天,各染毒组酶活性的抑制率达到58.4%~89.1%.结论 CAT在中毒反应中表现出了敏感性,具备了作为生物标志物的重要条件,有望在今后的实际应用中发挥作用.  相似文献

The effects of hyperoxia on the status of antioxidant defenses and markers of oxidative damage were evaluated in goldfish tissues. The levels of lipid peroxides, thiobarbituric acid reactive substances, carbonyl proteins and the activities of some antioxidant enzymes were measured in brain, liver, kidney and skeletal muscle of goldfish, Carassius auratus L., over a time course of 3-12 h of hyperoxia exposure followed by 12 or 36 h of normoxic recovery. Exposure to high oxygen resulted in an accumulation of protein carbonyls in tissues throughout hyperoxia and recovery whereas lipid peroxides and thiobarbituric acid reactive substances accumulated transiently under short-term hyperoxia stress (3-6 h) but were then strongly reduced. This suggests that hyperoxia stimulated an enhancement of defenses against lipid peroxidation or mechanisms for enhancing the catabolism of peroxidation products. The activities of principal antioxidant enzymes, superoxide dismutase and catalase, were not altered under hyperoxia but catalase increased during normoxic recovery; activities may rise in anticipation of further hyperoxic excursions. In most tissues, the activities of glutathione-utilizing enzymes (glutathione peroxidase, glutathione-S-transferase, glutathione reductase) as well as glucose-6-phosphate dehydrogenase, were not affected under hyperoxia but increased sharply during normoxic recovery. Correlations between some enzyme activities and oxidative stress markers were found, for example, an inverse correlation was seen between levels of thiobarbituric acid reactive substances and glutathione-S-transferase activity in liver and catalase and glucose-6-phosphate dehydrogenase in kidney. The results suggest that liver glutathione-S-transferase plays an important role in detoxifying end products of lipid peroxidation accumulated under hyperoxia stress.  相似文献

We investigated whether cold acclimation leads to increased activity of the antioxidant defense enzymes and muscle injury. Comparisons were between short track skaters (n=6) and inline skaters (n=6) during rest and at submaximal cycling (65% VO2max) in cold (ambient temperature: 5+/-1 degrees C, relative humidity: 41+/-8%) and warm conditions (ambient temperature: 21+/-1 degrees C, relative humidity: 35+/-5%), during 60 min, respectively, and during the recovery phase. Erythrocyte superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSHpx), reduced glutathione (GSH), thiobarbituric substance acid (TBARS), serum creatine kinase (CK), lactate dehydrogenase (LDH), plasma myoglobin (Mb) and cortisol were determined. Activities of CAT and GSHpx and the level of GSH and TBARS in erythrocyte and the level of LDH in serum were elevated in cold acclimated subjects. We suggested that the compensatory increase in antioxidative defense enzymes resulting from long-term cold exposure may reflect the elevated reactive oxygen species (ROS) production and muscle injury at this environment acclimation.  相似文献

Interactions between the ectomycorrhizal fungus Laccaria laccata and the soil fungus Trichoderma virens in co-culture and in the rhizosphere of Pinus sylvestris seedlings growing in vitro were investigated by light and scanning electron microscopy. The growth of T. virens was inhibited in co-culture. Shortened, more branched and sometimes deformed or injured hyphae of T. virens were observed in the zone of inhibition. Two-month-old mycorrhizae of P. sylvestris/ L. laccata were inoculated with a conidial suspension of T. virens and examined at intervals of 7-24 h and 2, 3, and 6 days post-inoculation (p.i.). On non-mycorrhizal roots, conidia germination was high and long hyphae formed 3 days p.i. On mycorrhizal roots, short germ tubes were observed only sporadically. At 3 days p.i., the mantle hyphae of L. laccata grew towards the conidia and coiled around them. Extremely dense coils of hyphae were found around clusters of conidia. Deformation of conidia, breaks in conidial walls and their partial degradation were observed 6 days p.i.  相似文献

The effect of pretreatment with 0.05 mM salicylic acid (SA) on the activity of superoxide dismutase (SOD) and peroxidase in the roots of four-day-old seedlings of wheat (Triticum aestivum L.) was studied under conditions of salination. The level of the stress-induced accumulation of active oxygen species and, therefore, activities of SOD and peroxidase in seedlings pretreated with SA were significantly lower than in untreated seedlings, which indicates that these enzymes contribute to the protective effect of SA on plants under conditions of salination.  相似文献

The present work was undertaken with a view to study the effect of oral feeding of 2% Aloe vera gel extract (AGE) for 30 days on azoxymethane (AOM)-induced oxidative stress in rats. It was observed that AOM administration resulted in a significant increase in malondialdehyde and conjugated dienes, with reduction in hepatic glutathione (GSH), vitamin A and uric acid contents. AOM-induced reduction in hepatic GSH and uric acid was brought back to normal by AGE. There was a significant raise in hepatic catalase, superoxide dismutase and glucose-6-phosphate dehydrogenase (G-6-PD) activities as a result of feeding of the extract. Ingestion of the extract effected reduction in AOM-induced colonic GSH-peroxidase, G-6-PD and glutathione S-transferase and femur bone marrow micronuclei formation. Hence, it is suggested that Aloe vera gel extract possess the ability to reduce AOM- induced oxidative stress and toxicity in liver.  相似文献

Although some plant responses to salinity have been characterized, the precise mechanisms by which salt stress damages plants are still poorly understood especially in woody plants. In the present study, the physiological and biochemical responses of Broussonetia papyrifera, a tree species of the family, Moraceae, to salinity were studied. In vitro-produced plantlets of B. papyrifera were treated with varying levels of NaCl (0, 50, 100 and 150 mM) in hydroponic culture. Changes in ion contents, accumulation of H2O2, as well as the activities and isoform profiles of superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) in the leaves, stems and roots were investigated. Under salt stress, there was higher Na+ accumulation in roots than in stems and leaves, and Ca2 +, Mg2 + and P3 + content, as well as K+/Na+ ratio were affected. NaCl treatment induced an increase in H2O2 contents in the tissues of B. papyrifera. The work demonstrated that activities of antioxidant defense enzymes changed in parallel with the increased H2O2 and salinity appeared to be associated with differential regulation of distinct SOD and POD isoenzymes. Moreover, SDS-PAGE analysis of total proteins extracted from leaves and roots of control and NaCl-treated plantlets revealed that in the leaves salt stress was associated with decrease or disappearance of some protein bands, and induction of a new protein band after exposure to 100 and 150 mM NaCl. In contrast, NaCl stress had little effect on the protein pattern in the roots. In summary, these findings may provide insight into the mechanisms of the response of woody plants to salt stress.  相似文献

The impact of five Bacillus thuringiensis (Bt) cotton varieties and their respective isogenic non-Bt(NBt) isolines (ANKUR-2534, MECH-6304, RCH-317, ANKUR-651 and MECH-6301) was assessed on the key soil enzymes i.e., dehydrogenase, alkaline phosphatase and urease in their rhizosphere at four growth stages of the crop, namely vegetative, flowering, bolling and harvesting. These varieties were grown on farmer's field in villages 22 miles and 24 miles of Ganganagar District of Rajasthan State in India. Results showed that dehydrogenase, alkaline phosphatase and urease activities were higher in rhizosphere of Bt isolines as compared to NBt isolines of all the varieties. Except phosphatase, differences in dehydrogenase and urease activities in rhizosphere of Bt and NBt isolines of all five varieties were significant (P < 0.05). Maximum enhancement in the three enzymes activities was observed in MECH-6304 Bt isoline rhizosphere. Maximum and minimum activities of dehydrogenase and urease were observed in MECH-6304 and RCH-317 Bt isolines, respectively, whereas phosphatase activity was maximum and minimum in MECH-6304 and ANKUR-651 Bt isolines, respectively. Maximum dehydrogenase and urease activities were observed at boll formation and minimum at flowering and harvesting stage, respectively, while maximum phosphatase activity was observed at vegetative stage and minimum at harvesting stage. In conclusion, all the studied Bt isolines of cotton varieties showed no adverse effect on dehydrogenase, alkaline phosphatase and urease activities in the rhizosphere.  相似文献

The data on the influence of chromium in different tissues of rats at its consumption with mixed fodder in the form of CrCl3 x 6H2O on the intensity of peroxidation processes and activity of antioxidant enzymes are presented. The degree of high chromium content in the studied tissues of rats at its addition to mixed fodder in the amount of 200 microg/kg during 30 days was established. Chromium content in the rat tissues decreased in the order: the spleen, heart, kidneys, lungs, brain, liver, skeletal muscle. In all tissues of rats fed with mixed fodder with chromium addition, except for skeletal muscles, content of lipid peroxidation products--hydroperoxide and TBARS-products decreased. The content of lipid peroxidation products decreased in the spleen, kidneys, liver and lungs. Also in all organs and tissues of rats the activity of glutathione peroxidase, glutathione reductase and catalase increased at the action of chromium. In the brain and kidneys the level of reduced glutathione increased. Superoxide dismutase activity was significantly higher not only in the heart and skeletal muscles of animals and is probably equal in the lungs and liver, and in other organs--the brain, kidneys and spleen in animals of the studied group the enzyme activity was lower as compared to animals of the control group. Obtained results demonstrate the regulatory influence of chromium on free radical process in the rat tissues.  相似文献

Elucidation of the role of endogenous cytokinins in cambial activity and wood formation requires knowledge of their identity and concentrations in the cambial region. Here, we have used capillary liquid chromatography/frit-FAB mass spectrometry to identify endogenous cytokinins in the vascular cambial region of mature Pinus sylvestris (L.) trees. Full-scan mass spectra were obtained for isopentenyladenine, isopentenyladenosine, zeatin riboside, dihydrozeatin and dihydrozeatin riboside. Of these, isopentenyladenine, dihydrozeatin and dihydrozeatin riboside are demonstrated by rigorous physico chemical methods for the first time in a conifer. In addition, an adenine glycoside was found for the first time in a plant. The identified cytokinins were quantified in active and dormant cambial region tissues by isotope dilution techniques using the appropriate deuterated isotope for each cytokinin species. The concentration of the detected cytokinins ranged between 1.3 and 5.5 pmol g-1 fresh weight, and did not vary greatly between dormant tissues, and in tissues actively dividing and differentiating. This observation indicates that cessation and reactivation of cell division activity in the vascular cambium is controlled by factors other than cytokinin availability.  相似文献

甘蔗是重要的糖能兼用植物,同时也是潜在的重金属土壤修复植物。为了探明甘蔗对铜胁迫的生理响应,该研究采用营养液培养的方法分析了铜胁迫对甘蔗生长、叶片相对电导率、叶绿素含量及根系抗氧化酶活性等的影响。结果表明:甘蔗地上部分生物、叶绿素含量随铜浓度增加而下降,地上部分铜含量和叶片的相对电导率则随铜浓度增加而提高。根系的丙二醛(MDA)含量在胁迫24h开始发生显著的变化,在400μmol·L^-1铜处理下比对照显著增加了25.5%,随着胁迫时间的延长,丙二醛含量增加幅度上升。到胁迫72h,100μmol·L^-1铜处理的MDA含量也显著的提高。根系SOD、POD和CAT酶活性在胁迫24h开始发生显著的变化,但不同酶活性变化存在差异。SOD酶活性除了在胁迫的72h随铜浓度增加而提高,其他时间表现为在100μmol·L^-1铜处理下活性下降,随着铜浓度继续增加SOD酶活性提高,到400μmol·L^-1铜处理SOD酶活性又出现下降。POD酶活性在处理的24、48和72h表现为随浓度增加先提高后下降的趋势,但在处理7d时随铜浓度增加而提高。CAT酶活性随着浓度的升高和胁迫时间的延长而下降,到了胁迫的第7天,三个铜处理下CAT酶活性分别下降了89.98%、96.88%和98.50%。由此可见,铜胁迫严重影响甘蔗的生长,较短时间的处理就已经引起了甘蔗根系的氧化胁迫,根系中SOD、POD和CAT酶活性的变化在甘蔗对抗铜引起的氧化胁迫中起到重要作用。  相似文献

The effects of long term salinity on some morpho-physiological characteristics were studied in strawberry Kurdistan and Queen elisa cultivars. Vegetative and biochemical traits were measured in strawberry cultivars subjected to three levels of salinity including 0, 40 and 80 mM at 20, 40 and 60th days after NaCl addition. Results showed that in both cultivars the dry weight of plant organs decreased in response to NaCl, except of crown weight in cv. Kurdistan. Root to shoot ratio increased due to a greater reduction in above ground biomass under salinity. Strawberry cultivars tended to decrease their stomatal conductance, RWC, proline, soluble carbohydrates and proteins during the different evaluation periods. Compared to the 20th day, peroxidase activity decreased at 80 mM during 40 and 60 days in cv. Queen elisa. On the contrary, ascorbate peroxidase activity elevated until the 40th day and decreased afterwards, in addition application of 40 and 80 mM NaCl increased the ascorbate peroxidase activity of both studied cultivars. Catalase activity increased from 20th until 60th days in cv. Queen elisa, while showed increase in cv. Kurdistan until day 40 and then decreased again at day 60. Application of 40 and 80 mM NaCl resulted in an increase in peroxidase, ascorbate peroxidase and catalase activities of both cultivars. The Queen elisa cv. showed lower tolerance index (45.88%) compared with cv. Kurdistan (67.97%). Finally, higher salinity resistance of cv. Kurdistan is probably associated with its ability to maintain higher RWC and higher activity of antioxidant enzymes.  相似文献
