In the rat hypothalamus, antibodies to ovine CRF41 stain neurons of a paraventriculoinfundibular neuroglandular pathway. CRF like immunoreactivity (CLI)-containing perikarya are mostly packed in the parvocellular division of the paraventricular nucleus. Their morphology and topography differ from that of other peptidergic neurons. However a few CLI perikarya are also stained with vasopressin antibodies. CLI neurons project massively to the external layer of the median eminence (ELME). Adrenalectomy induced a total depletion of ELME CLI 12 to 24 h after surgery, followed by a secondary accumulation already conspicuous 5 days later. This biphasic evolution, identical to that of ELME vasopressin, is totally prevented by a replacement therapy with dexamethasone. Reserpine also induces an acute depletion of ELME CLI and vasopressin, that can be prevented by a monoamine oxidase inhibitor pretreatment. These results indicate the involvement of CLI neurons in the corticotropic axis, suggesting that they are indeed corticoliberin neurons. Among the extrahypothalamic locations of CLI neurons their abundance in the amygdala central nucleus is of interest since it is involved in the corticotropic axis. A similar pattern of CLI was noticed in several mammalian brains and also in lower vertebrates (birds, reptiles, amphibians, fishes). Species adaptations of CLI neurons were observed: CLI neurons are of the cerebrospinal fluid contacting type in the turtle. CLI fibres terminate close to corticotrophs in the fish pituitary. This suggests a direct excitosecretory role of CRF on these cells and concurs with a CRF function of CLI peptide even in fishes. CLI processes and terminals appear in the human fetal ELME at the 16th week of development and increase in number during the following weeks. Perikarya are seen at 19 weeks. In the rat CLI fibers and perikarya were detected as early as the 18th day of fetal development. Thus, paraventriculo-infundibular CLI system develops later than corticotrophs. This chronology perfectly concurs with the results of previous physiological and experimental studies.  相似文献

The anatomical relationships between endorphinic neurons and dopaminergic neurons were evaluated in the rat hypothalamus using a combination of immunocytochemistry and autoradiography. In the arcuate nucleus, endorphinic endings were seen making contacts with dopaminergic cell bodies and dendrites. No synapsis could be observed at the sites of contacts. These results strongly suggest that the endorphinic neurons are directly acting on dopaminergic neurons to modify the release of dopamine into the pituitary portal system.  相似文献

The localization of human GRF1-44-immunoreactive neurons was studied in the rat brain. A dense accumulation of GRF-containing fibers was noted in the external layer of the median eminence. Cell bodies were observed in colchicine-treated rats. The most intensely fluorescent cluster of cells was contained in the arcuate nucleus. Other cells were seen on the base of the hypothalamus, within the median forebrain bundle, dorsal and ventral aspects of the ventromedial nucleus, zona incerta and dorsal part of the dorsomedial nucleus. These cells may influence the pulsatile release of pituitary growth hormone.  相似文献

Neurotensin (NT) injected intracerebroventricularly in rat increases dopamine (DA) turnover in the corpus striatum and nucleus accumbens. Significant increases in 3,4-dihydroxyphenylacetic acid (DOPAC) levels occurred within 15 minutes after injection with peak levels at 60 minutes. The effect on NT on DOPAC and homovanillic acid (HVA) accumulation was dose-dependent at 3–100 μg. NT, like haloperidol, stimulated 3,4-dihydroxyphenylalanine (DOPA) accumulation in striatal neurons, in the presence of DOPA decarboxylase inhibitor, after injection of gamma-butyrolactone (GBL). NT had a similar stimulatory effect on DOPA levels in the accumbens while haloperidol (0.25 mg·kg?1) had no significant effect in this brain region. NT did not block the inhibitory effect of apomorphine on DOPA accumulation in both the striatum and accumbens, while haloperidol inhibited apomorphine effect in both regions. NT also failed to displace 3H-spiperone from DA receptors and the presence of NT in the binding assay did not alter the ability of DA to displace 3H-spiperone in either brain region. These experiments demonstrate that NT increases DA turnover in both the nigrostriatal and mesolimbic pathways.  相似文献

The DNA topoisomerase I has been isolated from neurons of rat cerebral cortex. The most homogeneous fraction purified contains only one polypeptide of Mr approx. 100 000. The enzyme relaxes supercoiled DNA in the absence of ATP or Mg2+. The optimum monovalent cation concentration for the relaxation of superhelical DNA under conditions of DNA excess is found to be 175-200 mM. The neuron enzyme is similar to other mammalian type I DNA topoisomerases in that it links to the 3' ends of the broken DNA strands. Like calf thymus DNA topoisomerase I, the neuron topoisomerase can be selectively inhibited by poly(dG) but not by other homopolymerical deoxyribonucleotides.  相似文献

Using the immunoperoxidase technique in conjunction with specific antisera to alpha-atrial natriuretic polypeptide (alpha-ANP), it was shown that immunoreactive cell bodies and varicose fibers are widely distributed throughout the rat brain. The highest concentrations of alpha-ANP-containing neuronal cell bodies and fibers were found in the hypothalamus and septum. This result confirms the radioimmunological determination of alpha-ANP immunoreactivity in the rat brain.  相似文献

1. We recorded impulse activity of thermosensitive hypothalamic neurons in rat brain slices during superfusion with ethanol at constant temperatures and during slow sinusoidal temperature changes.

2. At constant temperatures of 37 °C, ethanol application typically induced a triphasic change of the firing rate: An initial excitation turned into complete inhibition followed by spontaneous recovery to higher firing rates.

3. Ethanol application increased the neurons’ temperature sensitivity remarkably.

4. Our data indicate complex neuromodulatory effects of ethanol with different time delays which interfere with basic mechanisms of temperature transduction.

Metallothionein III (MT-III) is a brain-specific member of the metallothionein family and binds zinc in vivo. In order to confirm the precise localization of MT-III in normal rat brain and the change of MT-III expression after transient whole brain ischemia, we raised a high affinity phagemid-antibody specific for rat MT-III. Immunohistochemical analysis revealed that MT-III in normal brain is localized abundantly in neuronal cell bodies in CA1-3 regions of hippocampus, dentate gyrus, cerebral cortex, olfactory bulb and Purkinje cells in cerebellum. This expression pattern of MT-III was similar to that of MT-III mRNA observed by in situ hybridization studies. ELISA and Northern blot analysis revealed that MT-III protein as well as mRNA levels were up-regulated in cerebrum soon after ischemic stress. Immunohistochemical analysis also demonstrated intense staining in neurons in injured brain after ischemia, which distributed in the same regions as in normal brain. These results suggest that MT-III plays an important role in protecting neurons from ischemic insult by reducing neurotoxic zinc levels and inhibits uncontrolled growth of neurites after ischemia.  相似文献

In experiments on rat brain slices, we carried out intracellular recording from neurons of the red nucleus (RN). Passive electrical properties of these neurons (input resistance, membrane time constant) were evaluated. We detected voltage-dependent rebound depolarization and time dependent inward rectification when passing hyperpolarizing pulses of current through the cell. Injections of depolarizing currents caused rhythmical firing of the neurons; the frequency of these firings depends upon the strength of injected current. Rhythmical firings were also characterized by rapid frequency adaptation when currents of different frequency were injected. Stimulation of regions of slices presumably corresponding to the decussion of the brachium conjunctivum mainly evoked EPSPs with a "fast" rise time in RN neurons. This suggests activation of synaptic input from the cerebellar nucleus interpositus. Stimulation of this same region sometimes evoked EPSP-IPSP mixtures and "pure" IPSPs in RN neurons.L. A. Summary Gonadotropin-releasing hormone (GnRH) neurons and their pathways in the rat brain were localized by immunocytochemistry in 6-to 18-day-old female animals, by use of thick frozen or vibratome sections, and silver-gold intensification of the diaminobenzidine reaction product. GnRH-immunoreactive perikarya were observed in the following regions: olfactory bulb and tubercle, vertical and horizontal limbs of the diagonal band of Broca, medial septum, medial preoptic and suprachiasmatic areas, anterior and lateral hypothalamus, and different regions of the hippocampus (indusium griseum, Ammon's horn). In addition to the known GnRH-pathways (preoptico-terminal, preoptico-infundibular, periventricular), we also observed GnRH-immunopositive processes in several major tracts and areas of the brain, including the medial and cortical amygdaloid complex, stria terminalis, stria medullaris thalami, fasciculus retroflexus, medial forebrain bundle, indusium griseum, stria longitudinalis medialis and lateralis, hippocampus, periaqueductal gray of the mesencephalon, and extracerebral regions, such as the lamina cribrosa, nervus terminalis and its associated ganglia. By use of the silver-gold intensification method we present Golgi-like images of GnRH perikarya and their pathways. The aim was to study changes in brain monoamine neurons in an experimental animal model with an extrapyramidal motor syndrome of the parkinsonian type. The neurological signs were observed in rats after acute cobaltous acetate intoxication under mild ischemic conditions. Histofluorescence studies showed a decrease in catecholamine fluorescence (which signifies a decrease in the amine content) in the hypothalamus and mesencephalic reticular formation, but not in the substantia nigra or basal ganglia. Serotonin fluorescence was increased in nerve cell bodies of the dorsal and median raphe nuclei and in nerve terminals in some thalamic and preoptic regions. Histological staining of sections adjacent to the fluorescent ones showed no neuronal loss and some pathology of myelin. The disturbing effect of cobaltous ions on the neuronal transmission, and/or the imbalance between dopamine and serotonin in the extrapyramidal motor syndrome observed in poisoned rats have been discussed.  相似文献

Brains from 1-day-old rats were dissociated with trypsin and the cells were maintained in culture for 3-4 weeks. These primary cultures contained insulin-like immunoreactivity in the limited populations of neurons. Typically, the fluorescent staining pattern observed in the soma was homogeneous and varicosity-like structures were observed on the neurites of the majority of insulin-like immunoreactive neurons. Serum deprivation of brain cell cultures did not reduce immunoreactivity, whereas cycloheximide caused approx. 80% decrease in the number of insulin-like immunoreactive neurons. Incubation of these cultures with [3H]valine resulted in the incorporation of radioactivity into immunoprecipitable insulin. These results suggest that insulin-like immunoreactivity present in the Central Nervous System (CNS) neurons may be synthesized by brain cell cultures.  相似文献

Using the indirect immunofluorescent technique, corticotropin releasing factor (CRF)-like immunoreactive nerve fibers and cell bodies were observed to be widely distributed in rat brain. A detailed stereotaxic atlas of CRF-like immunoreactive neurons was prepared. Large numbers of CRF-containing perikarya were observed in the nucleus paraventricularis, with scattered cells in the following nuclei: accumbens, interstitialis stria terminalis, preopticus medialis, supraopticus, periventricularis hypothalami, amygdaloideus centralis, dorsomedialis, substantia grisea centralis, parabrachialis dorsalis and ventralis, tegmenti dorsalis lateralis, vestibularis medialis, tractus solitarius and reticularis lateralis. The most intense staining of CRF-containing fibers was observed in the external lamina of the median eminence. Moderate numbers of CRF-like fibers were observed in the following nuclei: lateralis and medialis septi, tractus diagonalis, interstitialis stria terminalis, preopticus medialis, supraopticus, periventricularis thalami and hypothalami, paraventricularis, anterior ventralis and medialis thalami, rhomboideus, amygdaloideus centralis, habenulae lateralis, dorsomedialis, ventromedialis, substantia grisea centralis, cuneiformis, parabrachialis dorsalis and ventralis, tegmenti dorsalis lateralis, cerebellum, vestibularis medialis, reticularis lateralis, substantia gelatinosa trigemini and lamina I and II of the dorsal horn of the spinal cord. The present findings suggest that a CRF-like peptide may be involved in a neurotransmitter or neuromodulator role, as well as a hypophysiotropic role.  相似文献

Ultrastructure of mitotically dividing cells in the sensomotor cerebral cortex of mature rats has been investigated, after transplantation into them embryonal nervous tissue of 17-day-old embryos. In 4 days after the operation in the recipient's nervous tissue, arranging around the developing transplant, among various proliferating cells with mitotic figures, some cells with signs of neurons (oval bodies, electron opaque cytoplasm with developed organelles and RNP-particles, specific for the nervous cell, with axonal terminals on the body) have been found. These cells are surrounded with the satellite glia. There are no mitotic figures in the pyramidal neurons.  相似文献
