Activité oxydasique de l''oxydoréductase de la xanthine, spécifique de la classe des mammifères

In every species other than mammals xanthine oxidoreductase behaves as a dehydrogenase, never as an oxidase. In three mammalian species, the enzyme acts intracellularly as a dehydrogenase, but its class-specific ambivalence allows its extracellular conversion into an oxidase.

En dehors de la classe des Mammifères, l'oxydoréductase de la xanthine ne se comporte jamais comme une oxydase. Chez les Mammifères, une ambivalence de la molécule permet l'expression d'une activité déshydrogénasique (NAD) dans la cellule, et d'une activité oxydasique en dehors de la cellule.     

Bois plio-pléistocènes du gisement de saropée plateau de khorat, est de la Thaïlande

Alluvial deposits from the Mue Nam Mum river at Saropée, Khorat Region, Thailand, yielded about 60 wood specimens, attributed to Araucarioxylon sp., Shoreoxylon thaïlandense n.sp., Careyoxylon pondicherriense Awasthi, Terminalioxylon coriaceum Prakash and Awasthi, Terminalioxylon burmense Mädel-Angeliewa and Müller-Stoll, Pahudioxylon sahnii Ghosh and Kazmi, Cynometroxylon schlagintweitii Müller-Stoll and Mädel, and Albizzinium eolebekkianum Prakash. This association, attributed to a Plio-Pleistocene age, evokes a mixed deciduous forest, probably not far from a river and ancient volcanoes. The presence of Araucariaceae in South-Eastern Asia at a relatively recent epoch is pointed out. A comparison with other fossil wood localities reveals close floristic resemblance with Burma, Western Bengal and Kachchh.

Une nappe fluviatile de la rivière Mue Nam Mum, gisement de Saropée, région de Khorat, en Thaïlande, a livré une soixantaine de bois fossiles, déterminés comme: Araucarioxylon sp.,Shoreoxylon thaïlandense n.sp., Careyoxylon pondicherriense Awasthi, Terminalioxylon coriaceum Prakash et Awasthi, Terminalioxylon burmense Mädel-Angeliewa et Müller-Stoll, Pahudioxylon sahnii Ghosh et Kazmi, Cynometroxylon schlagintweitii Müller-Stoll et Mädel et Albizzinium eolebekkianum Prakash. Cette association, évoquant une forêt semi-dense, s'est vraisemblablement développée à proximité d'un cours d'eau et d'anciens volcans. Elle est attribuée au Pliocène ou au Quaternaire ancien. La question de la présence d'Araucariaceae dans le Sud-Est asiatique à une époque relativement récente est évoquée. Une comparaison avec d'autres gisements à bois fossiles révèle des ressemblances floristiques avec la Birmanie, le Bengale occidental et le Kachchh.     

Deux nouvelles tortues de l'Éocène inférieur de Saint-Papoul (Aude, France)

Two new turtles from the Lower Eocene of Saint-Papoul (Aude, France). Among the fossil turtles from of the Lower Eocene of Saint-Papoul (Aude, France), two new taxa of freshwater Testudinoidea are described. The first is a new species of the genus Palaeoemys, known before in the Lutetian of Germany. The second taxon is a new genus, also described on material of Pépieux-Cesseras and Eygalayes. Le système protéolytique de Penicillium roqueforti: II - Purification et propriétés de la protéase acide

An extracellular protease has been isolated from the culture medium of Penicillium roqueforti. The enzyme was purified by precipitation with ammonium sulfate, filtration on Bio-gel P 100 columns and chromatography on D.E.A.E.-cellulose columns The purified preparation was homogenous by gel filtration on Bio-gel P 60 and electrophoretical analysis at pH 9.0 and 5.0.The protease exhibited the properties of an acid protease: the optimum pH was 3.5 for casein or hemoglobin hydrolysis and for bovine trypsinogen activation; at 40°C, the enzyme is most stable in the range of pH 3.5 to 5.5; the optimum temperatures was 50°C.E.D.T.A., D.F.P. and sulfhydryl reagents induced no inhibition.The enzyme exhibited a milk clotting activity that was fifty times weaker that the activity of rennin. Its molecular weight, estimated by gel filtration was 33 400. Amino acid composition is: Lys₁₅, His₂, Arg₁, Trp₅, Asp₃₃, Thr₂₇, Glu₁₆, Pro₁₀, Gly₄₀, Ala₂₅, Cys₂, Val₂₁, Ile₂₀, Leu₂₁, Tyr₁₄, Phe₁₉.The properties of this protease were closely similar to that of P. janthinellum and Aspergillus oryzae.     

Etude experimentale de L′auto-association des molécules modèles dipeptidiques. III. Influence de la dimerisation stéréosélective sur les spectres de résonance magnétique protonique

Proton magnetic resonance spectra of model dipeptide molecules R₁–C′₁O₁–N₂H₂–CHR₂–C′₂O₂–N₃H₃–R₃ in CCl₄ solutions exhibit splited signals when investigating on mixtures of L and D enantiomers differing from the racemic composition. The major effect is observed on amide proton signals which are the ones most sensitive to the ratio of aggregation. The stereoselective dimerization of enantiomeric molecules in the so-called C₅ conformational state is shown to be responsible for such a phenomenon, the intensity of which depends on the bulkiness of the side chain R₂. A theoretical approach is proposed which gives predictions in close agreement with our own experimental findings.     

Étude expérimentale de l'auto-association des molécules modèles dipeptidiques. II. Association stéréosélective des molécules énantiomères

Modes of aggregation fo alanine-, norvaline- and valine-contaiing dpepetides with the general formula R₁? C₁O₁? N₂H₂? CHR₂? C₂O₂? N₃H₃? R₃ have been studied in CCl₄ solution by using infrared and nuclear magnetic resonance spectroscopies. Solutions of the pure L isomer and of the racemic mixture do not give identical data. At a given concentration, the racemic mixtrue is more aggregated than the pure enantiomer, and the difference, negligible in the case of alanine derivative, increases wiht the bulkiness of the side cahin R₂. The results show that a selective interaction takes place between enantiomeric molecules, resulting ina dimer associating tow inverse configurated C₅ conformers. The stabilizaion of this dimer proceeds from two symmetrical and intermolecular H₃ … O₁ hydrogen bondings.     

Une nouvelle voie de biosynthèse de l'acide oléique dans les végétaux

Oleic acid biosynthesis in Cauliflower slices has been studied from the following radioactive precursors: ¹⁴C-acetate, ¹⁴C-decanoate, ¹⁴C-laurate, ¹⁴C-palmitate, ¹⁴C-stearate and ¹⁴C-hydroxyacids derived from ¹⁴C-decanoate. The higher incorporation yield was obtained withe ¹⁴C-decanoate or the hydroxyacids derived from it. A new biosynthetic pathway for oleic acid, implying the intervention of a β-γ-hydroxylaurate-dehydrase is discussed.     

Activité DNA-polymérase nucléaire dans les spermatozoides de souris et évolution de cette activité au cours de la spermatogenèse

The presence of a nuclear DNA polymerase activity has been demonstrated in the mouse testis by a cytochemical method. The enzyme was present in the nuclei of the various cell types of mouse spermatogenesis, including spermatozoa. The nuclear activity increased during meiotic prophase and then decreased in spermatids and spermatozoa. A peculiar attention was devoted to the properties of the nuclear DNA polymerase present in spermatozoa. All four deoxyribonucleoside-triphosphates were needed for optimal activity; the acid-insoluble radioactive product obtained by polymerization in situ was completely removed by DNase, but not by RNase nor by pronase. The reaction of polymerization was DNA-dependent, sensitive to high concentrations of KCl and not affected by NEM nor by PHMB.     

Régulation de la synthèse du DNA dans la glande séricigène de Bombyx mori: II. Etude de l''activité DNA-polymérasique présente dans la fraction soluble brute

In the silkglands of Bombyx mori larvae, DNA synthesis suddenly stops in the middle of the fifth larval instar, when cellular growth is over and cell activity is turned on to massive silk synthesis. Of much interest is to determine what factors are implied in the quantitative and temporal control of DNA synthesis in this tissue.Previous work on thymidine metabolism in the intact organ lead us to look for several enzymatic activities in the soluble fraction of the cells during this period.The results presented here concern the kinases of the thymidylic pathway. As measured in vitro from crude cellular extracts, thymidine-kinase is present at a lower level than thymidylate-kinase but both activities vary in connection with the pattern of in situ DNA synthesis, reaching their highest levels on the fourth day of the instar and decreasing as DNA synthesis definitively stops in the glands.     

Présence des enzymes catalysant la biosynthèse d'UDP glucose dans une fraction légère de l'appareil de Golgi des hépatocytes de chat

A Golgi-rich fraction is prepared from cat hepatocytes by the means of a four-step sucrose density gradient. The material applied to this gradient is composed either of smooth microsomes prepared from healthy animals, or of total microsomes prepared from cat treated by 50 per cent ethanol (0.6 g/100 g body weight, administered by stomach tube).A light fraction (d : 1.10) is obtained by the two procedures. It does not show any glucose-6-phosphatase activity, but is enriched in sialyltransferase, known as a marker enzyme for Golgi apparatus. It also contains the three enzymes implicated in the biosynthetic pathway for UDP-glucose (glucokinase, phosphoglucomutase and UTP : glucose-1-phosphate uridylyltransferase).UDP-glucose being the ultimate substrate in membranous glucosylation reactions, these results could support the hypothesis that sugar-nucleotides necessary for the glycoprotein biosynthesis are produced in the Golgi vesicles directly.     

Etude,par spectroscopie infra-rouge,de la conformation de quelques composés peptidiques modèles

Some experimental data are given on the infrared spectra between 3300 and 3500 cm^?1 of dilute solutions in carbon tetrachloride of three types of model compounds: CH_3?CONH-CH(R₁)-CONH(R₂), (I); CH₃-CON(CH₃)-CH(R₁)-CONH(R₂), (II) and CH₃-CONH-CH(R₁)-CON(R₂)₂, (III). In studying the N-H stretching bands, it was found that there are two types of intramolecular hydrogen bonds in these molecules; these result in two different cyclized conformations, C₅ and C₇, which contain respectively, five and seven atoms in the ring. By using model substances I, II, and III, in which the nitrogen atoms are unequally substituted, it is possible to identify the N-H stretching bands which are to be ascribed to the N-H oscillators included in the two different chelated conformations. It is found also that the stretching frequency of a free N-H oscillator depends upon the substituent on the nitrogen atom. Thus, it is possible to observe, with some of the model compounds I, four different absorption bands located at 3340, 3420, 3440, and 3460 cm^?1. The first two are ascribed to the N-H oscillators included in the H? bonds which lock the C₇ and C₅ conformations; the last two correspond to free N-H which differ with the substituent on the nitrogen atom.     

Mesure des déclins de l'intensité et de l'anisotropie de fluorescence du pérylène en présence de membranes d'érythrocyte de pigeon: Comparaison avec un milieu isotrope visqueux

Using synchrotron radiation as the excitation light, we studied the fluorescence parameters of perylene incubated with pigeon erythrocyte membranes and with an isotropic viscous medium, the Primol 342 oil.From 4 to 37°C, we observed a single lifetime of 4.5 ns in the oil and two with the membrane (τ₁ = 1−1.4 ns and τ₂ = 5.4−6.1 ns). The dependence upon temperature of the rotation correlation time of perylene () in the oil was characteristic of an isotropic medium, whereas the limiting value of anitropy (r ∞) was zero. With the membrane, γ ∞ decreased from 0.14 to 0.06 and from 2.9 to 0.5 ns, indicating a greater amplitude and frequency of molecular motions.The addition of chlorpromazine, indomethacine, tetracaine, n-octylamine, octanol or octanoic acid to the membrane decreased the τ₁ and τ₂ values. This would stem from the desorganization of the membrane induced by the drugs.     

Réponses des adultes de Lasiomma melania,ravageur des cônes de Larix decidua, à des pièges colorés de différents types

La réponse visuelle des adultes de Lasiomma melania Ackl. (Diptera, Anthomyiidae) à des carrés englués de diverses couleurs ou combinaisons de couleurs a été étudiée durant 5 années consécutives dans plusieurs mélèzins (Larix decidua) des Hautes-Alpes. Une préférence pour le jaune et le jaune à bandes pourpres a été mise en évidence chez ce ravageur dont la femelle pond sur les cônes de mélèze. L'attractivité de chaque type de piège est fonction à la fois du sexe et du stade de maturation sexuelle des individus. La couleur jaune attire en majorité des insectes émergents immatures. Elle semble représenter, selon le plan dans lequel elle est située, un signal nutritionnel de type fleur (plan horizontal) ou un signal de reconnaissance du feuillage de mélèze (plan vertical). Les individus mûrs sexuellement, recherchant les arbres produisant des cônes, sont en revanche capturés essentiellement sur les pièges verticaux jaunes à bandes pourpres. Ceux-ci miment, en l'amplifiant, le contraste de réflectance existant entre le feuillage des mélèzes et les cônes. Ce contraste paraît correspondre à un signal permettant la reconnaisance des arbres possédant des cônes. La ponte est cependant limitée à un stade particulier de développment de ces derniers alors que le contraste est à peu près stable dans le temps. D'autres signaux émis par les cônes, vraisemblablement d'ordre olfactif, doivent guider le vol d'approche et l'atterrissage des femelles.
