A panel of twelve monoclonal antibodies (MAbs), designated FS1 to FS12, have been raised against surface antigens of Fucus serratus sperm. The antibodies were selected on the basis that they show region-, gamete-, species- or genus-preferential binding. Indirect immunofluorescence shows that the antigens bound by the MAbs are distributed non-randomly over the cell surface. Seven MAbs (FS1, FS3, FS4, FS6, FS8, FS9, FS10) bind antigens located primarily on the cell body, while the others (FS2, FS5, FS7, FS11, FS12) bind antigens located primarily on the anterior flagellum. Of the MAbs that label the anterior flagellum, FS2, FS5, FS7 and FS12 form a halo at the perimeter of the flagellum. Electron microscopic-immunogold studies indicate that the halo results from labelling of the mastigonemes, as opposed to the flagellar plasmamembrane. Gamete-preferential binding of antibodies was detected using an enzyme-linked immunosorbent assay with egg membrane vesicles. Eight of the MAbs bind sperm antigens not common to eggs, though FS2, FS4, FS5 and FS9 bind antigens present on both sperm and eggs. In studies of species- and genus-specificity FS2, FS3, FS5, FS6, FS7, FS8, FS10, FS11 and FS12 exhibit genus-preferential binding, labelling sperm of F. serratus and F. vesiculosus more intensely than that of Ascophyllum nodosum. The electrical properties of unfertilized eggs of Fucus serratus L. were characterized using voltage clamp and current clamp with single electrodes. The plasma membrane of the unfertilized egg is excitable. Depolarizing the egg in current clamp induced a transient depolarizing voltage response, the amplitude of which was dependent on the presence of external Ca2+ or Ba2+ and was blocked by La3+. The repolarizing phase was blocked by tetraethylammonium ions. Repeated stimulation at frequencies greater than 0.5 Hz caused a transient loss of excitability. Voltage-clamp experiments revealed that an inward current with an activation threshold of -35 mV underlies the depolarizing phase of the voltage response. This current showed rapid activation and slow inactivation. The current was blocked by La3+ and could be carried by Ca2+ and Ba2+ but not by Sr2+ or Na+. Further depolarization to values more positive than-5 mV induced a slowly activating outward K+ current in addition to the inward current, which corresponded to the repolarizing phase of the voltage response. This K+ current showed little or no inactivation during stimulation and slow deactivation on return to the resting potential. Hyperpolarizing the egg elicited an inward current. On fertilization, the Fucus egg generates a depolarizing fertilization potential. Voltage-clamp experiments revealed an inward fertilization current underlying the fertilization potential. Within 15 min of fertilization a dramatic, irreversible increase in resting K+ permeability developed. Sperm of the brown alga Fucus serratus are highly differentiated, biflagellate, naked cells. Immunolocalisation studies, employing monoclonal antibodies (MAbs — designated FS1 to FS12) raised against antigens of these sperm cells, have revealed that some sperm surface components are distributed over the entire cell, whereas others are restricted to, or occur preferentially on, the surface of the anterior flagellum or cell body. This report describes the use of these MAbs in Western-blot procedures and antigen-modification binding assays to determine the nature of these sperm surface components. Monoclonal antibodies which bind to antigens found on the cell body and both flagella (FS3, FS4, FS6, FS8, FS10) recognise carbohydrate epitopes of a high-molecular-weight glycoprotein (Mr=205 kDa). These MAbs were initially chosen at random from a much larger number of antibodies which bound to sperm in a similar fashion, indicating that this glycoprotein is an immunodominant antigen. Though these MAbs compete under conditions of limited antigen availability, differences in the effects of periodate on antibody binding and differences in other binding data indicate that the MAbs recognise epitopes of this glycoprotein which are neighbouring or overlapping, rather than common. The MAb FS9, which has a similar binding pattern to the above antibodies, also seems to bind to carbohydrate epitopes, but the antigen recognised by this antibody could not be identified in Western-blotting procedures. The MAbs FS7 and FS12, which bind to the mastigonemes on the anterior flagellum and to the cell body and posterior flagellum, recognise a set of glycoproteins in the molecular-weight range 40–250 kDa. The evidence indicates that the antibodies are binding to N-linked carbohydrate side chains of these glycoproteins. Three MAbs that bind to the anterior flagellum (FS2, FS5 and FS11) recognise protein antigens in the molecular-weight range 90–250 kDa; it is not known whether these antigens are glycosylated. The macroalgal belt in the southern Baltic Sea may be partly structured by the interaction of physical and biological factors. A field study, spanning the 1990s, describes a rapid decline of the Fucus spp. stands along the wave-exposed Swedish southeast coast. During this period, a relative dominance of Fucus vesiculosus L. shifted to a relative dominance of Fucus serratus L. The decline of F. vesiculosus coincided with observations of large numbers of the grazing isopods Idotea baltica (Pallas) and Idotea granulosa Rathke, or with field observations of frequent grazing marks on Fucus fronds. I. baltica, but not I. granulosa, tended to aggregate in the declining Fucus spp. stands, indicating a strong preference for Fucus spp. In a mesocosm experiment I. baltica, when given a choice, grazed both Fucus species at weak water motion. At strong water motion grazing was concentrated on F. vesiculosus. It is hypothesized that one of the reasons I. baltica preferred F. vesiculosus to F. serratus at strong water motion may have been differences in habitat quality, like width of thallus, influencing the ability to cling to the plant. Smaller thallus, as in F. vesiculosus, thus is the preferred habitat for grazing of I. blatica. We postulate that the existence of F. serratus in the area may be favoured by strong wave action and moderate but not strong grazing by I. baltica, relaxing the interspecific competition from F. vesiculosus.  相似文献

Fucus and Laminaria species, dominant seaweeds in the intertidal and subtidal zones of the temperate North Atlantic, experience tidal cycles that are not synchronized with light:dark (L:D) cycles. To investigate how nutrient assimilation is affected by light cycles, the activity of nitrate reductase (NR) was examined in thalli incubated in outdoor tanks with flowing seawater and natural L:D cycles. NR activity in Laminaria digitata (Huds.) Lamour. showed strong diel patterns with low activities in darkness and peak activities near midday. This diel pattern was controlled by light but not by a circadian rhythm. In contrast, there was no diel variation in NR activity in Fucus serratus L., F. vesiculosus (L.) Lamour., and F. spiralis L. either collected directly from the shore or maintained in the outdoor tanks. In laboratory cultures, transfer to continuous darkness suppressed NR activity in L. digitata, but not in F. vesiculosus; continuous light increased NR activity in L. digitata but decreased activity in F. vesiculosus. Furthermore, 4 d enrichment with ammonium (50 μmol · L?1 pulses), resulted in NR activity declining by >80% in L. digitata, but no significant changes in F. serratus. Seasonal differences in maximum NR activity were present in both genera with activities highest in late winter and lowest in summer. This is the first report of NR activity in any alga that is not strongly regulated by light and ammonium. Because light and tidal emersion do not always coincide, Fucus species may have lost the regulation of NR by light that has been observed in other algae and higher plants.  相似文献

Sequences from the two ribosomal DNA internal transcribed spacers (ITS1 and ITS2) were compared among five species of Fucus. Based on the present taxon sampling, parsimony analysis showed that Fucus serratus is the sister-group of the remaining Fucus species when Ascophyllum nodosum was used as an outgroup. The topology of the tree was (Fucus serratus (F. lutarius (F. vesiculosus (F. spiralis+F. ceranoides)))). The extremely low variation observed suggests a very recent radiation of the genus which supports the view widely accepted that the Fucales are among the most evolutionarily advanced of the brown algae. We further note that sequence differences between Fucus and Ascophyllum were 28%: this does not rule out the utility of ITS sequences within the Fucaceae. The very low number of informative positions allows to demonstrate empirically that distance matrix methods group on the basis of symplesiomorphies. We have analysed in detail the Na+ content and Na+ influx during fertilization and first divisions of the sea urchin egg (Paracentrotus lividus) using a filtration technique devised to eliminate rapidly contamination by the Na+ of external sea water. In the first 5 min following fertilization the egg fills up with Na+ (+ 30%). Thereafter Na+ is extruded and the Na+ content stabilizes at about 60% of the unfertilized egg level by the second cleavage (2 h). The initial increase in Na+ content is due to a large increase in Na+ influx already detected at 20 sec. The Na+ influx reaches its maximum at 1 min and its minimum at 5 min. H+ excretion follows the same kinetics. A second increase in Na+ influx is noted 5–10 min after fertilization; it reaches its maximum at prophase metaphase (30 min) and its minimum during cleavage (60 min). These oscillations in Na+ influx were observed for the first three divisions. Fertilization also immediately stimulates the Na+ efflux which remains elevated throughout the cell cycle and is responsible for the depletion of the Na+ content of the embryos. Activation of the eggs by weak amine bases (5 mM NH4Cl) which bypasses the early cortical reaction produces only a depletion in the Na+ content of the egg similar to that produced by fertilization. NH4Cl also increases the Na+ influx soon after fertilization, although no transient variations are noted.  相似文献

Patterns of nuclear and organelle inheritance among artificial hybrids of the seaweeds Fucus serratus and F. evanescens were detected using single-strand conformation polymorphism (SSCP). Three alleles were identified in the 231?bp rDNA-ITS1 gene (nuclear): two in F. serratus and one in F. evanescens. Alleles differed by 1–2?bp and all hybrids possessed one allele from each parent. Two haplotypes were present in the 288?bp Rubisco spacer (chloroplast), differentiated by a 33?bp indel. Two haplotypes differing by a single nucleotide were found in a 135?bp region of nad11 gene (mitochondrion). Both organelles are maternally inherited, as all hybrids contained the haplotypes of the parent contributing the egg. Although laboratory hybrids among Fucus spp. have been produced previously, this is the first time that both nuclear and cytoplasmic genetic markers have been used to document inheritance patterns. SSCPs analysed on an automated sequencer offer a rapid and powerful approach for identifying suspected hybrids from field samples, as well as a screen for intraspecific and intra-individual variation in DNA regions prior to confirmation of variations by sequencing.  相似文献

In the ascidian Ciona intestinalis the species-specific interaction between the spermatozoon and the egg occurs between the vitelline coat (VC) of the egg and the plasma membrane of the apical part of the head of the spermatozoa. Concanavalin A (Con A)-binding sites are present on this area of the sperm surface. We used Con A to identify and isolate the spermatozoon plasma membrane components that may be involved in the interaction with the VC. These glycoproteins have been identified on SDS-PAGE of a sperm membrane fraction (SMF) enriched with the extermal proteins, after incubation of the gel with 3H-Con A. Affinity chromatography on Con A-agarose has been used for the purification of sperm plasma membrane proteins with and affinity for the lectin. The biological activity of the Con A-retained fraction was determined with binding and fertilization assays.  相似文献

Keddy's competitive hierarchy model describes species distribution patterns along gradients under equilibrium conditions and can potentially serve as an explanation for zonation patterns of intertidal seaweeds on rocky shores. One of the assumptions of the model is a competitive hierarchy with the top competitor occupying the benign end of the gradient. Another assumption is the consistency of competitive ranks of species in all environmental conditions included in the shared parts of species' fundamental niches. In laboratory replacement series experiments, the competitive ranks of pairs of Fucus species that occupy adjacent zones in the field were analysed and compared to ranks found in previous field experiments. Unattached thalli of Fucus serratus versus F. vesiculosus or F. vesiculosus versus F. spiralis, respectively, were held in aerated beakers to establish the competitive ranking of the three congeners. Each replacement series was conducted at three total densities. F. vesiculosus was clearly competitively dominant over F. serratus. In competition with F. spiralis, F. vesiculosus was only dominant at its lowest absolute input frequencies, but at higher frequencies dominance was reversed. At high densities, the total ranking was F. spiralis > F. vesiculosus > F. serratus, which is the opposite order to that which would be expected from Keddy's model. Although all three species thrived well under the laboratory conditions, the results did not reflect in situ competitive dominances, which may be an effect of nutrient competition in the laboratory. Three species of Fucus inhabiting different zones at the littoral and sublittoral of the White Sea (F. vesiculosus L., F. serratus L., and F. distichus L.) were compared with regard to performance of the photosynthetic apparatus (PSA) and PSA changes upon prolonged desiccation of algae. The content of chlorophyll a and the total content of carotenoids were significantly higher in F. serratus than in F. vesiculosus. The potential quantum yield (F v/F m) of photosystem II (PSII) was 0.69–0.74 in all species. The highest effective quantum yield of PSII (Y II) was observed in F. vesiculosus plants inhabiting zones with the most intense insolation. The ratio of non-photochemical quenching to photochemical quenching (qN/qP) increased with the depth of algal habitats in the row from F. vesiculosus to F. serratus. The fluorescence parameters F. serratus deviated during desiccation from their normal values and did not recover upon the subsequent return to water. By contrast, the fluorescence parameters of F. vesiculosus and F. distichus recovered gradually after the return of algae from air to sea water. It is supposed that F. serratus plants, unlike other species, are characterized by comparatively low physiological plasticity.  相似文献

The Fucus populations on rocky shores along 300 km of the coastal waters of southeast Sweden in the Baltic proper have been studied since 1984 at a number of fixed sites as part of monitoring programmes. This paper describes changes in distribution and abundance of F. vesiculosus and F. serratus during the period 1984–2001. Sheltered sites showed a consistent temporal and spatial pattern of Fucus spp. distribution over a coastline of 300 kilometres. The depth penetration and abundance of Fucus spp. increased during the 1980s. Around 1990 the development reversed as a consequence of grazing and in 1997 many sites were almost devoid of Fucus spp. Since 1998 both abundance and depth penetration have increased again, possibly as a result of local measures against eutrophication. Exposed sites, on the other hand, lost their Fucus populations at the beginning of the 1990s, and they have not recovered. Extended field studies lead us to deduce that the fixed sites referred to above were representative of the Fucus populations in the area investigated. Major declines, both at sheltered and exposed sites, are attributed to grazing by the isopod Idotea baltica. The populations of I. baltica may have been favoured by the continuing eutrophication of the Baltic, a series of mild winters in the 1990s, and a contemporary decline in some potential predators.  相似文献

In this study, effective gamete concentrations, egg viability, and fertilization volumes were evaluated for Haliotis midae (L.). Sperm concentrations between 5?×?103 and 5?×?104?mL?1 (p?>?0.05) consistently resulted in high hatch-out rates (96?±?1%). At concentrations higher than 5?×?105?mL?1, hatch-out rates decreased to 69?±?7% (p??1 resulted in high fertilization rates, with 50?eggs?mL?1 being the ideal concentration for fertilization in H. midae. Egg viability was consistently high up to 100?min post-spawning, with a decrease in hatch-out success, when eggs were fertilized 120?min post-spawning. Fertilization volumes did not affect successful hatch-out. The results from this study can be implemented by South African abalone farms to increase hatch-out rates and subsequent culture. It can also be used as basis for the development of fertilization protocols in other marine invertebrate species.  相似文献

The introduced Fucus evanescens (hermaphroditic) and the native F. serratus (dioecious) have been in secondary contact along the Danish coast of the Kattegat Sea for 60–100 years and dioecious hybrids have been observed at Blushøj for several years. Hybridization in Fucus is unusual because it appears to always involve a hermaphroditic and a dioecious parental pair. We determined the degree and spatial patterns of introgression for 286 individuals using 10 microsatellite loci and cpDNA. Hybrids accounted for nearly 13% of the population, yet parental species were well differentiated (FST = 0.633). The presence of F. evanescens chloroplasts in 100% of F1 hybrids revealed asymmetrical hybridization. Fucus evanescens cpDNA was observed in 50% of introgressed and 5.4% of pure F. serratus, but no F. serratus cpDNA was found in F. evanescens. In contrast, nuclear DNA introgression was symmetrical with an equal amount (≈1.5%) of genes introgressed into each parental species. Survivorship and viability data suggest selection against hybrids in the hybrid zone.  相似文献

A field study to determine the precise times of year at which three intertidal species of Fucus start to produce hyaline hairs and cease producing such hairs was conducted on the Isle of Man, U.K. Hairs were first observed during February, and within 6 days of their initial appearance, all tagged plants of all species at all tidal heights on the shore possessed hairs. Hair production continued until the beginning of October, at which time Fucus plants growing at the lowest stations (+ 3.0 m) had glabrous apical growth. Hair production continued later into the year for plants growing higher on the shore, and it was not until mid-November that glabrous apical growth was observed in all plants. Phosphate uptake rates of pilose (hairy) and glabrous (hairless) apical sections were measured in November 1988 for F. spiralis L. and in January 1989 for F. spiralis and F. serratus L., at phosphate concentrations ranging from 0.8 μM (ambient seawater) to 9.0 μM. In ambient seawater, pilose plants of F. spiralis removed phosphate 2–3 times faster than glabrous plants, whereas the uptake rates of pilose plants of F. serratus were about 50% greater than those of glabrous plants. The differences between uptake rates of pilose and glabrous plants of both species were smaller or nonsignificant at higher phosphate concentrations. The field and laboratory data are consistent with the hypothesis that hairs are formed in Fucus as a response to increased nutrient demand and that hairs facilitate the uptake of nutrients from seawater at concentrations typical of natural situations.  相似文献

Summary Methods are described for the collection, treatment and uniform discharge of large quantities of gametes, and a measure of fertilization control in the monoecious brown alga, Fucus distichus. Fertilization was examined using the techniques of blister-formation, gamete separation experiments, and electron microscopy. Sperm enter the freshly-discharged egg packets through a mesochite pore and juxtaposition with the eggs early. However, experimental and thinsection data indicate that fertilization does not occur until the eggs dissociate from the mesochite and round up. Hence, the egg surface appears to undergo three functional changes following its release from the thallus: (1) a pre-dissociation state which inhibits fertilization within the mesochite; (2) a dissociation state when fusion of gametes is possible; and (3) a post-fertilization state characterized by the formation of extraneous coats. As the egg is activated by the sperm a nonmembranous layer appears to detach from the egg surface to form an activation layer. This is augmented by fibrous units to function as a fertilization barrier which ultimately thickens to form the cell wall. The area between the eggs within the egg packet is characterized by the presence of fibrous and particulate substances which are continuously given off through the egg surface. These apparently play a functional role in the sequential stages of fertilization in Fucus. The electrical properties of the egg of the medaka, Oryzias latipes, were studied before, during, and after fertilization. The resting potential of the unfertilized egg averaged ?39 ± 9 mV in Yamamoto's Ringers (Y. Ringers), but 20% of the values were between ?50 and ?60 mV. Fertilization triggers a small depolarization of 4 ± 3 mV in 10% Y. Ringers with an average duration of 20 ± 10 sec. The amplitude of this depolarization is independent of [Na+]o, [Ca2+]o, and [Cl?]o, so it appears to be due to a nonspecific leak triggered by sperm-egg fusion. The depolarization is followed by a longer hyperpolarizing phase with an average amplitude of 31 ± 12 mV. Recovery from this hyperpolarization has a fast phase lasting 155 ± 18 sec, followed by a slower phase which reaches a steady average membrane potential of ?19 ± 1 mV by 9 min after fertilization. The membrane resistance falls 10-fold during the first 2 min after fertilization, from 40 (1520 kΩ-cm2) to 3 MΩ. This is largely due to an increase in the K+ conductance. At the peak of the hyperpolarization, the membrane potential exhibits a 28 mV/decade [K+]o dependence and a 6 mV/decade [Na+]o dependence. The membrane resistance slowly recovers over the next 8 min to a value about 30% larger than before fertilization. The relation of current vs voltage was linear before, during, and after fertilization and indicated a reversal potential of ?98 ± 20 mV for the hyperpolarization peak. The egg's capacitance averaged 0.04 ± 0.01 μF (0.9 μF/cm2) before fertilization and approximately doubles within 90 sec after fertilization. It then decreases over a 9-min period, reaching a value 25% smaller than before fertilization.  相似文献

Over 3 successive seasonal cycles (April 1986 to October 1988), field experiments were established within 3 intertidal levels in the sheltered rocky intertidal of Helgoland (North Sea, German Bight). Competitors for space (Mytilus edulis, macroalgae), herbivores (Littorina spp.) and predators (Carcinus maenas) were either excluded from areas (0.25 m2) covered by undisturbed communities or enclosed at natural densities on areas that were cleared before of animals and plants. All the experimental fields (each 0.25 m2) were covered by cages with 4 mm gauze at the sides and a plexiglas top. The results of the experiments in the upper intertidal (occupied byLittorina spp. andEnteromorpha) showed that a natural density of herbivores could not prevent algal settlement and had only little influence on algal growth. Instead abiotic factors (storms, algae washed ashore) decreased the stock of the green algae. Experiments in the mid intertidal, dominated byMytilus (50% cover),Fucus spp. (20%) and grazingL. littorea (100 ind. m?2) showed that community structure was directly changed both by grazing periwinkles and by competition for space between mussels and macroalgae. WheneverLittorina was excluded, the canopy ofFucus spp. increased continuously and reached total cover within two years. In addition to the increase ofFucus spp., the rock surface and the mussel shells were overgrown byUlva pseudocurvata, which covered the experimental fields during parts of the summer in the absence of herbivores. As soon as perennial species (fucoids) covered most of the experimental areas, the seasonal growth ofUlva decreased drastically. Presence and growth of macroalgae were also controlled by serious competition for space with mussels. EstablishedMytilus prevented the growth of all perennial and ephemeral algae on the rocks. However, the shells of the mussels provided free space for a new settlement ofFucus andUlva. In the lower intertidal (dominated by total algal cover ofF. serratus, herbivores such asL. littorea andL. mariae, and increasing number of predators such asCarcinus), the feeding activity of herbivores can neither prevent the settlement of the fucoid sporelings nor reduce the growth of macroalgae.F. serratus achieved a total canopy on the rock within one year. Doubled density of herbivores prevented the settlement ofFucus and most of the undercover algae. Predation byCarcinus onLittorina spp. had little influence on the herbivore community patterns. However, the crabs supported the establishment of macroalgae by excluding the mussels from the lower intertidal. In summary, the community organization and maintenance in the mid and lower intertidal is influenced to a high degree by biological interactions. Whereas both the relatively important herbivory byL. littorea and competition for space between mussels and macroalgae dominate in the mid intertidal, predation reaches its highest relative degree of importance for community structure in the lower intertidal.  相似文献

Einige Algen (fädige Grünalgen,Bangia, Enteromorpha, Punctaria, Ceramium, Polysiphonia undLaminaria) konnten in gürtelförmigen Zonen auftreten; diese sind schmäler als am natürlichen Standort.
