Our previous studies suggested that salmon gonadotropin‐releasing hormone (sGnRH) neurons regulate both final maturation and migratory behavior in homing salmonids. Activation of sGnRH neurons can occur during upstream migration. We therefore examined expression of genes encoding the precursors of sGnRH, sGnRH‐I, and sGnRH‐II, in discrete forebrain loci of prespawning chum salmon, Oncorhynchus keta. Fish were captured from 1997 through 1999 along their homing pathway: coastal areas, a midway of the river, 4 km downstream of the natal hatchery, and the hatchery. Amounts of sGnRH mRNAs in fresh frozen sections including the olfactory bulb (OB), terminal nerve (TN), ventral telencephalon (VT), nucleus preopticus parvocellularis anterioris (PPa), and nucleus preopticus magnocellularis (PM) were determined by quantitative real‐time polymerase chain reactions. The amounts of sGnRH‐II mRNA were higher than those of sGnRH‐I mRNA, while they showed similar changes during upstream migration. In the OB and TN, the amounts of sGnRH mRNAs elevated from the coast to the natal hatchery. In the VT and PPa, they elevated along with the progress of final maturation. Such elevation was also observed in the rostroventral, middle, and dorsocaudal parts of the PM. The amounts of gonadotropin IIβ and somatolactin mRNAs in the pituitary also increased consistently with the elevation of gene expression for sGnRH. These results, in combination with lines of previous evidence, indicate that sGnRH neurons are activated in almost all the forebrain loci during the last phases of spawning migration, resulting in coordination of final gonadal maturation and migratory behavior to the spawning ground. © 2005 Wiley Periodicals, Inc. Serum thyroid hormone levels were determined in adult Atlantic salmon ( Salmo salar ) of both sexes caught in the ocean and at a sequence of locations on their return migration to spawn. Tri-iodothyronine (T3 ) levels were greatest in fish caught in coastal or estuarine waters or in a river near head-of-tide. T3 levels were lower in fish caught in rivers throughout the angling season and lowest in those captured entering a tributary near spawning. Thyroxine (T4 ) levels were lowest in immature fish captured in the ocean in winter but raised in fish captured in spring; many of the latter group showed endocrine evidence of their becoming sexually mature. T4 levels were greatest in fish captured in coastal waters and progressively lower in fish captured in an estuary, near head-of-tide and in rivers. T4 levels in fish captured at tributary entry near spawning exceeded those in fish caught in rivers earlier in the year. In general, these data support the hypothesis that motor activity level in migrant fish is a determinant of thyroid status. 相似文献

The lamprey (Lampetra fluviatilis L.) is an extant representative of the ancient vertebrate group of Agnathans. During the prespawning migration (the river period of life from autumn until spring) lamprey hepatocytes exhibit widely different energy states: a high-energy state in autumn and spring, corresponding to a normal physiological standard, and a low-energy state in winter, which is provoked by prolonged starvation and profound metabolic arrest. In spring the restoration of energy status (return to an active state) is associated with hormonally induced lipolysis of the lipid droplets stored in the cells. Lamprey hepatocytes demonstrate an aerobic metabolism based on oxidation of free fatty acids. The dynamics of mitochondrial membrane potential (MMP) were measured throughout the prespawning migration. Pharmacological inhibition of the electron transport chain decreased the MMP and caused extensive depletion of cellular ATP without loss of cell viability. The potential molecular mechanisms responsible for winter metabolic depression in lamprey hepatocytes are discussed. 相似文献

A complex of adaptive changes occurring in the Pacific salmon fry in the process of migration to the sea is described, including behavior, ion content in carcasses, and morphological changes in Stannius bodies, gill epithelium, and nephron tubular epithelium. Participating in experiments with transfer from fresh water into a two-layer aquarium (the lower layer—sea water, the upper layer—fresh water) were smolts of chum salmon and underyearlings of cherry salmon as well as the trachurus and leiurus forms of the three-spined stickleback Casterosteus aculeatus. All fish, regardless of their salt preference, at once after placement into the two-layer aquarium, occupied the sea water zone, at the very bottom of the aquarium. After 1 h, there started brief excursions of cherry salmon and chum salmon to the upper, fresh water layer; however, both forms of the three-spined stickleback did not participate in these excursions. After 12 h, the chum salmon settled down in the lower, sea water layer, while the cherry salmon—in the upper, fresh water layer. Both forms of the three-spined stickleback never left the sea water layer and felt quite comfortably on the aquarium bottom. It seems that the high tolerance of the both stickleback forms to wide salinity limits allows them to choose the convenient position regardless of the water salt composition. By analyzing the material obtained for three years (2001–2003) on structure and functions of the gill epithelium chloride cells (CC), we have come to the conclusion that the fresh water fry of two salmon species, chum and cherry salmons, caught at the same time and practically in the same water reservoirs can be divided into three groups. The underyearlings of the cherry salmon as a rule are characterized by the thickened epithelium of secondary gill lamellae, but by a very small number of CC. In smolts of chum salmon, on the contrary, the epithelium is sufficiently thin, but enriched in the CC that demonstrate an active structure in the very beginning of migration to sea. However, with approaching the sea (and with an increase of terms of migration) the CC activity drops, but their amount does not change. And only after migration to the sea the CC activity rises again, although their amount seems to remain unchanged. The described peculiarities of behavior and of the ion composition regulation in the migrating salmon fry confirm the hypothesis that the salmons evolutionized in fresh water, that the Oncorhynchus genus appeared in large spaces of saltish waters, such as the Japan sea at the period of the early Pleistocene, and that learning of fry of the Oncorhynchus genus (for instance, of O. gorbuscha and O. keta) is the most specialized in the salmons migration to the sea, whereas the fresh water species of chars (Salvelinus) and of trouts (Salmo) are more primitive. 相似文献

Pacific salmon (Oncorhynchus spp.) play an important role as a keystone species and provider of ecosystem services in the North Pacific ecosystem. We
review our studies on recent production trends, marine carrying capacity, climate effects and biological interactions between
wild and hatchery origin populations of Pacific salmon in the open sea, with a particular focus on Japanese chum salmon (O. keta). Salmon catch data indicates that the abundance of Pacific salmon increased since the 1976/77 ocean regime shift. Chum and
pink salmon (O. gorbuscha) maintained high abundances with a sharp increase in hatchery-released populations since the late 1980s. Since the 1990s,
the biomass contribution of hatchery returns to the total catch amounts to 50% for chum salmon, more than 10% for pink salmon,
and less than 10% for sockeye salmon (O. nerka). We show evidence of density-dependence of growth and survival at sea and how it might vary across spatial scales, and we
provide some new information on foraging plasticity that may offer new insight into competitive interactions. The marine carrying
capacity of these three species is synchronized with long-term patterns in climate change. At the present time, global warming
has positively affected growth and survival of Hokkaido populations of chum salmon. In the future, however, global warming
may decrease the marine carrying capacity and the area of suitable habitat for chum salmon in the North Pacific Ocean. We
outline future challenges for salmon sustainable conservation management in Japan, and recommend fishery management reform
to sustain the hatchery-supported salmon fishery while conserving natural spawning populations. 相似文献

Animal migrations can affect disease dynamics. One consequence of migration common to marine fish and invertebrates is migratory allopatry-a period of spatial separation between adult and juvenile hosts, which is caused by host migration and which prevents parasite transmission from adult to juvenile hosts. We studied this characteristic for sea lice (Lepeophtheirus salmonis and Caligus clemensi) and pink salmon (Oncorhynchus gorbuscha) from one of the Canada's largest salmon stocks. Migratory allopatry protects juvenile salmon from L. salmonis for two to three months of early marine life (2-3% prevalence). In contrast, host diversity facilitates access for C. clemensi to juvenile salmon (8-20% prevalence) but infections appear ephemeral. Aquaculture can augment host abundance and diversity and increase parasite exposure of wild juvenile fish. An empirically parametrized model shows high sensitivity of salmon populations to increased L. salmonis exposure, predicting population collapse at one to five motile L. salmonis per juvenile pink salmon. These results characterize parasite threats of salmon aquaculture to wild salmon populations and show how host migration and diversity are important factors affecting parasite transmission in the oceans. 相似文献

The motility of demembranated bull sperm was found to be governed by the concentrations of cyclic adenosine 3', 5'-monophosphate (cAMP) and Ca2+ at low pH (6.6-7.1), and was less sensitive to these variables at higher pH (7.4-7.8). Although motility was generally found to increase with increasing pH in the range from 6.6 to 7.8, the addition of exogenous cAMP markedly and selectively improved the motility at the lower end of the range (pH 6.6-7.1). In the presence of 10 microM cAMP, low Ca2+ (8.0 X 10(-8) M), and a high concentration of Mg-adenosine 5'-triphosphate (ATP, 8 mM), demembranated sperm at pH 6.8 and 7.1 exhibited swimming similar to that of live ejaculated sperm. At a free Ca2+ concentration of 4.4 X 10(-5) M, the motility was rapidly inhibited at pH 6.8-7.1, whereas at pH 7.4-7.8, the activity was not greatly affected. Since calcium is known to antagonize the cAMP pathway by activating Ca2+-dependent phosphodiesterase and Ca2+-dependent phosphatase, this further supports the idea that cAMP-dependent activation is crucial for motility at low pH. Our results demonstrate that the flagellar axoneme can function normally at relatively acidic pH, and produce vigorous swimming at high levels of ATP. Gamete activation in fish is an important step in terms of artificial fertilization of oocytes, cryopreservation studies and other experimental manipulations. Salinity and pH differences in activation media affect to sperm motility and fertilizing ability. These experiments were therefore designed to investigate the combined effects of pH (range 5.0–9.0) and salinity (20, 30, 37, and 45‰) of activation media on sperm motility of European sea bass Dicentrarchus labrax. The best results were obtained at salinity 37‰ and a pH of 9.0. Our results also demonstrated that non-progressive motility at salinity 45‰ was observed in the range of 5.0–9.0 pH. In conclusion, spermatozoa can be motile at a wide range of pH and salinity values although the percent of motile spermatozoa and motility duration are negatively affected by low pH values. 相似文献

Tracking 47 post-spawned adult Atlantic salmon Salmo salar L. in a hydropower-regulated river through autumn, winter and spring revealed that winter survival was 56% and 75% in two study years, respectively, with higher mortality of males than females (50% vs. 33% and 100% vs. 13%, respectively). Some kelts (n = 7) displayed nondirected movements that were interpreted as a reconditioning period for an average of 9–17 days prior to directed downstream movements indicating the initiation of migration. Survival after the initiation of migration in spring was 83% and 94% to the hydropower dam in the first and second study years, and decreased to 60 and 63%, respectively, after dam passage. There were no further losses in the downriver reach in the second year, with the first year having a cumulative survival estimate of 53% to the river mouth. Kelts approached the dam when the spillway gates were available as a passage option most of the time (64%–75%), but some kelts arrived at the dam or had not yet passed when spillways were closed (n = 6) and the only remaining passage option was restricted to the turbines. However, all but one kelt that must have passed via turbine were successful in reaching the river mouth. Migratory delay presumably due to searching behaviour caused by low water flow was estimated at approximately 6 days as migration rates were significantly slower in the reservoir (median ± s.e. 8.5 ± 2.5 km day−1) than up- (29.7 ± 5.0 km day−1) or downriver (22.1 ± 3.1 km day−1). The proportion of time (median 30%) that kelts spent swimming upstream (searching behaviour) in the reservoir was a significant variable for migration success. 相似文献

Characterization of sperm motility in sea bass: the effect of heavy metals and physicochemical variables on sperm motility 总被引:1,自引:0,他引:1
Computer assisted sperm analysis (CASA) was used to characterize the motility of sea bass Dicentrarchus labrax spermatozoa and to study the effect of several physicochemical variables and heavy metals on sperm swimming performance. Duration of sperm motility in sea bass was very short (<50 s). During the first 20 s all the motility variables measured remained approximately constant, the velocity and linearity of the movement being maximum during this period, while both variables decreased sharply later. While slight variations in pH did not significantly modify sperm swimming performance, changes in osmolality affected all the measured motility variables. Two of the heavy metals tested, Cu2+ and Pb2+ , did not affect sperm motility when the activating media contained up to 100 ppm of the metal salts. In contrast, Hg2+ modified the morphology of post-swimming spermatozoa at 0·4–1 ppm (sperm dilution rate 1:39) and completely arrested sperm motility at concentrations as low as 0·1 ppm (sperm dilution rate 1:2500). Assuming a covalent binding to sperm cells, this revealed a finite number of c. 10 million Hg2+ binding sites per spermatozoon. Complementary results using demembranated spermatozoa suggested that the main target of HgCl2 would be located in the plasma membrane and that HgCl2 would inhibit water channels, hence preventing sperm motility. 相似文献

The behavior of a ciliate protozoan, Paramecium, is known to represent the electrical state of the cell membrane, and regulation of the membrane potential and ciliary motion
are known to involve cAMP and cGMP. The present study shows the synchrony of circadian changes in motility, resting membrane
potential and cyclic nucleotides in P. multimicronucleatum. Using an automated system for tracking isolated single microorganisms, the isolated Paramecium cells are confirmed to swim fast and straight during the day (and subjective day) and slowly, with frequent turning, at night
(and subjective night). The resting membrane potential is more negative during the day than at night. cAMP and cGMP concentrations
oscillate in a manner, such that both cAMP and cGMP are higher during the day (or subjective day) than at night (or subjective
night). The ratio of cGMP to cAMP during the light and dark cycle (LD) fluctuates, paralleling the fluctuation of the resting
membrane potential measured during the LD. These results suggest that the Paramecium will provide an excellent model to explore daily and circadian orchestration of second messengers mediating signals from
ambient light/dark cycles and circadian pacemaker to ion channels and cilia, directly involved in daily and circadian cellular
outputs of resting membrane potential and motility.

【背景】环境细菌可培养率只有1%,菌间的信息交流是限制培养效率的一个重要原因。细胞间信息传递的"第二信使"——环磷酸腺苷(cAMP)是细菌感知和应对外界变化时使用广泛的物质。【目的】研究cAMP诱导培养下的细菌种群组成变化,以探讨对微生物可培养性的影响,为提高细菌可培养率和菌种资源开发提供参考。【方法】以太湖沉积物为对象,结合高通量测序和分离培养方法解析样品中细菌的多样性。【结果】高通量测序的物种组成分析结果表明,cAMP作用下诱导出60个不同于对照组的OTU(Operational taxonomic unit),其中显著差异的是变形菌门和拟杆菌门,前者提高4.08%,后者降低3.36%,非常显著差异的是疣微菌门。在属分类水平有8个显著差异的菌属,2个非常显著差异的菌属(气单胞菌属和Trichococcus)。分离培养结果显示,cAMP诱导下CFU(Colony-forming unit)提高至1.6倍,诱导培养出微小杆菌属(Exiquobacterium)、微杆菌属(Microbacterium)、热单胞菌属(Thermomonas)、Lacibacter、Pedobacter、Massilia、Kocuria和Arthrobacter。【结论】从高通量结果可知cAMP对变形菌门的菌株生长有显著促进作用,而分离培养结果表明,cAMP提高微小杆菌属、微杆菌属、热单胞菌属、Lacibacter、Pedobacter、Massilia、Kocuria和Arthrobacter的可培养性。 相似文献

A boar sperm integral plasma membrane protein (APz) involved in the adhesion of uncapacitated and capacitated sperm to the porcine zona pellucida (ZP) has been characterized by two-dimensional polyacrylamide gel electrophoresis (PAGE) and tested for its ability to bind to various zona glycopeptides. APz shows microheterogeneity and focuses over a wide pH range, with predominant forms focusing above pH 7. The protein, when excised from nonreducing polyacrylamide gels, inhibited sperm-egg binding and bound heat-solubilized zonae preventing these zonae from blocking sperm binding to eggs. In an indirect assay, a polyclonal monovalent antibody, which blocks sperm-egg binding and which is absorbed by APz, was used to determine the ability of zona glycopeptides to prevent the sperm-egg blocking activity of the antibody from being absorbed by intact sperm. When whole heat-solubilized ZP was added to sperm at doses that block sperm-egg binding and the excess ZP was removed, the sperm-egg blocking activity of the antibody was not absorbed by these sperm, and antibody-containing supernatants blocked the binding of untreated sperm to eggs as effectively as antibody that was not mixed with fresh sperm. When alpha ZP3 was used in the same manner, sperm-egg blocking activity again was not absorbed by antibody-treated cells. Beta ZP3, however, failed to block sperm-egg binding and failed to absorb the sperm-egg blocking activity of the antibody. These findings support the argument that the action of APz is physiologically significant and involves specific binding sites on the ZP3 component of the ZP. 相似文献

The objective of this study was to evaluate the effects of six activating solutions on duration of sperm motility, fertilization rate (FR), and hatching rate (HR) of Prochilodus lineatus (Valenciennes, 1837). The activating solutions (SA) used were: SA0 (199 mOsm kg?1, pH 8.5), SA1 (138 mOsm kg?1, pH 7.5), SA2 (256 mOsm kg?1, pH 7.5), SA3 (131 mOsm kg?1, pH 10), NaCl (92 mOsm kg?1, pH 7.5) and distilled water (32 mOsm kg?1, pH 7.5). SA1 induced the highest motility, FR and HR, compared with the other activating solutions. The lowest motility was obtained with SA0, with no fertilization or hatching, whereas motility was zero with SA2 and SA3. It is possible to conclude that the solution SA1 can be used for the activation of gametes during fertilization in induced reproduction of curimba to achieve higher fertilization and hatching rates. Thus, it was found that the osmolality and pH of activating solutions, probably with the participation of dissolved substances therein, are the main factors acting on semen motility after activation. 相似文献

Control is exerted on the movement of mammalian spermatozoa at ejaculation and at capacitation. Here the activation of motility in motionless pre-ejaculated sperm was investigated. This was done by isolating quiescent caudal epididymal sperm from the hamster and observing that the addition of either calcium cAMP, cGMP, or cUMP conferred full motility upon them. Other salts, nucleotides, caffeine, sugars, or oxygen did not. Epididymal fluid which contains phosphodiesterase had too little calcium to activate the sperm while seminal plasma had more than enough. The cAMP content of quiescent sperm was low, but ATP levels were high. At the activation of motility, sperm cAMP synthesis became very rapid. It thus appears that sperm are quiescent on the male because they lack cAMP, and that calcium, supplied at ejaculation, initiates rapid cAMP synthesis to produce motility. 相似文献

Histamine, bradykinin and prostaglandin E1 increase the permeability coefficient of albumin; this effect is mimicked by dibutyryl-cAMP and by theophylline. They also cause a significant rise in the cAMP levels of the incubated tissue. Serotonin has lesser activities on these parameters. Presumably, the first three mediators control the transcellular passage of albumin by increasing the cAMP content of the tissue, serotonin being less effective.These findings on the mesentery seem to have a couterpart in vascular tissue as shown by others using different methods. 相似文献

The European eel''s singular spawning migration from European waters towards the Sargasso Sea remains elusive, including the early phase of migration at sea. During spawning migration, the movement of freshwater resident eels from river to sea has been thought to be irreversible. We report the first recorded incidents of eels returning to the river of origin after spending up to a year in the marine environment. After migrating to the Baltic Sea, 21% of the silver eels, tagged with acoustic transmitters, returned to the Narva River. Half returned 11–12 months after moving to the sea, with 15 km being the longest upstream movement. The returned eels spent up to 33 days in the river and migrated to the sea again. The fastest specimen migrated to the outlet of the Baltic Sea in 68 days after the second start—roughly 1300 km. The surprising occurrence of returning migrants has implications for sustainable management and protection of this critically endangered species. 相似文献

A perturbation of zinc metabolism has been noted in subjects with obesity. Zinc may also participate in the intracellular
signal cascade by affecting cellular calcium influx and a change in the calcium-calmodulin (CaM)-cyclic adenosine monophosphate
(cAMP) pathway. The possible effects of zinc on cellular concentrations of CaM, a major cytosolic calcium-binding protein,
in the adipocytes derived from obese (ob/ob) mice and their lean counterparts were studied. Adipocytes derived from both phenotypes
of mice were treated either with 0.2 mM of zinc sulfate or without any additive for 1 h of incubation; the cellular levels of CaM and cAMP were then determined.
The results showed that the obese mice had lower CaM and cAMP levels in their adipocytes compared to the lean mice. Zinc treatment
reduced CaM and increased cAMP levels in all mice, although this effect was more pronounced in the lean mice. This study indicated
that there was an inverse interaction between CaM and cAMP in their cellular levels in the mouse adipocytes and that might

Cold transport of epididymides from genetically modified mice is an efficient alternative to the shipment of live animals between research facilities. Mouse sperm from epididymides cold-stored for short periods can maintain viability. We previously reported that cold storage of mouse epididymides in Lifor® perfusion medium prolonged sperm motility and fertilization potential and that the sperm efficiently fertilized oocytes when reduced glutathione was added to the fertilization medium. Cryopreservation usually results in decreased sperm viability; an optimized protocol for cold storage of epididymides plus sperm cryopreservation has yet to be established. Here, we examined the motility and fertilization potential of cryopreserved, thawed (frozen-thawed) sperm from previously cold-stored mouse epididymides. We also examined the protective effect of sphingosine-1-phosphate (S1P) on sperm viability when S1P was added to the preservation medium during cold storage. We assessed viability of frozen-thawed sperm from mouse epididymides that had been cold-transported domestically or internationally and investigated whether embryos fertilized in vitro with these sperm developed normally when implanted in pseudo-pregnant mice. Our results indicate that frozen-thawed sperm from epididymides cold-stored for up to 48 h maintained high fertilization potential. Fertilization potential was reduced after cold storage for 72 h, but not if S1P was included in the cold storage medium. Live pups were born normally to recipients after in vitro fertilization using frozen-thawed sperm from cold-transported epididymides.
