Two special areas involving membranous components in strain KB cells were studied by electron microscopy. The first area described is that of the subsurface regions of two apposing cells in which flattened cisternae (one cisternae in each subsurface region) with membranes spaced 110–230 A apart were found in a confrontation alignment. The long dimension of the profiles of these cisternae ranges from 0.5 to 2 µ. At these intercellular contact areas, each cisterna is closely applied to the adjacent plasma membrane; the intervening space is 60–100 A. We have named the cisternae in these roughly symmetrical areas of cell contact the subsurface confronting cisternae. Communications between these cisternae and those of the rough-surfaced endoplasmic reticulum also were observed. The second area described is that of the intracytoplasmic confronting cisternae. These cisternae were observed as oval or round images about 0.3–1.4 µ in diameter, each image being composed of a pair of concentrically arranged confronting cisternae with membranes spaced 200–400 A apart. The apposing membranes of the two confronting cisternae are electron opaque, smooth, and free of ribosomes, whereas the unapposed membranes are less dense, scalloped, and associated with ribosomes. The spacing between the two intracytoplasmic confronting cisternae is 70–110 A.  相似文献

The Nissl substance of these neurons consists of masses of endoplasmic reticulum showing various degrees of orientation; upon and between the cisternae, tubules, and vesicles of the reticulum lie clusters of punctate granules, 10 to 30 mµ in diameter. 3. A second system of membranes can be distinguished from the endoplasmic reticulum of the Nissl bodies by shallower and more tightly packed cisternae and by absence of granules. Summary Membrane-bounded organelles possessing cisternae, i.e., rough endoplasmic reticulum and Golgi apparatus, in immature rat central neurons were examined by quick-freeze and deep-etch techniques to see how their intracisternal structures are organized and how ribosomes are associated with the membrane of the endoplasmic reticulum. Cisternae of endoplasmic reticulum, 60–100 nm wide, were bridged with randomly-distributed strands (trabecular strands, 12.5 nm in mean diameter). Luminal surfaces of cisternae of the endoplasmic reticulum were decorated with various-sized globular particles, some as small as intramembrane particles, and others as large as granules formed by soluble proteins seen in the cytoplasm. A closer examination revealed much thinner strands (3.3. nm in mean diameter). Such thin strands were short, usually winding toward the luminal surface, and sometimes touching the luminal surface with one end. Ribosomes appeared to be embedded into the entire thickness of cross-fractured membranes of endoplasmic reticulum, that is, their internal portions appeared to be situated at almost the same level as the cisternal luminal surface. From the internal portion of ribosomes, single thin strands occasionally protruded into the lumen, suggesting that these thin strands were newly synthesized polypeptides. A horizontal separation within ribosomes appeared to occur at the same level as the hydrophobic middle of the membrane of the endoplasmic reticulum. Interiors of the Golgi apparatus cisternae, which were much narrower than cisternae of endoplasmic reticulum, were similarly bridged with trabecular strands, but the Golgi trabecular strands were thinner and more frequent. Their cisternal lumina were also dotted with globular particles. No identifiable profiles corresponding to the thin strands in the endoplasmic reticulum were observed. Golgi cisternae showed a heterogeneous distribution of membrane granularity; the membrane in narrow cisternal space was granule-rich, while that in expanded space was granule-poor, suggesting a functional compartmentalization of the Golgi cisternae.  相似文献

Ultrastructural changes of hepatocytes were examined in the sand lamprey,Lampetra reissneri, during various phases of the life cycle. In hepatocytes of ammocoetes, the rough endoplasmic reticulum was composed of short cisternae and the Golgi apparatus were scarcely developed, showing no sexual differences at this stage of life cycle. In hepatocytes of female lampreys at the metamorphic stages 4 to 5, the rough endoplasmic reticulum was developed to form long parallel cisternae and the Golgi apparatus were well-developed. The rough endoplasmic reticulum developed further to form stacks of long parallel cisternae extending over the cytoplasm in hepatocytes of females at the young adult stage, and became composed of both long parallel and vesicular cisternae in the cells of females at the adult stage. The Golgi apparatus were invariably welldeveloped in hepatocytes of young adult and adult females. No consipcuous development was observed in profiles of the rough endoplasmic reticulum and the Golgi apparatus in hepatocytes of males during and after metamorphosis. The ultrastructural changes of the rough endoplasmic reticulum and the Golgi apparatus observed in hepatocytes of female sand lampreys are considered to have an intimate relation to the activity of vitellogenin synthesis in the liver, and it is suggested that the hepatocytes begin to rapidly synthesize vitellogenin in the sand lamprey at the metamorphic stages 4 to 5.  相似文献

Summary Structures identified as annulate lamellae, lamellar bodies and subsurface cisternae were found in neurons of the hyperstriatum accessorium of the avian forebrain. Annulate lamellar arrays with up to six lamellae were present in the larger somata. The lamellae were made up of fused smooth-surfaced cisternae forming pores or annuli and were surrounded by a dense filamentous to granular material. Stacks of nonfenestrated, parallel, regularly spaced cisternae, designated as lamellar bodies, also appeared in the cytoplasm. When flattened they were reminiscent of the electron dense subsurface cisternae. Continuity could be demonstrated between peripherally located subsurface cisternae and lamellar bodies. The dense filamentous to finely granular substance was also located between these structures. Annulate lamellae, lamellar bodies and subsurface cisternae were always observed in conjunction with the rough endoplasmic reticulum. The functional significance of these structural associations is considered.  相似文献

The production of Golgi complexes was investigated in Amoeba proteus by introducing a nucleus into cells that had been enucleated for 5 days. Golgi complexes were not detected in 5 day enucleates, nor were they observed in amebae fixed 15 min after renucleation. Samples taken at longer intervals after the introduction of a nucleus exhibited an increase in the size and abundance of Golgi complexes. Small curved smooth cisternae, some of which were aligned in parallel to form small Golgi complexes, were observed 30 min after the operation. Aggregations of small Golgi complexes increased in number in amebae fixed 1 to 6 hr after renucleation. Golgi complexes of normal size were present 6 hr after the operation and became more abundant in samples fixed 12 hr, and 1, 2, and 3 days after renucleation. The possible participation of the granular endoplasmic reticulum in the development of Golgi complexes was suggested by two observations. First, the Golgi complexes in renucleates contained a dense material similar to the content of the endoplasmic reticulum in enucleates and early renucleates. Second, examples of continuity between the endoplasmic reticulum and Golgi cisternae were present in renucleates. The possibility that Golgi complexes can be produced in the absence of preexisting Golgi complexes is discussed.  相似文献

In the cell cortex of the parasitic ciliate Ichthyophthirius multifiliis different kinds of cisternae were observed: the alveolar sacs, thick membrane cisternae and the endoplasmic reticulum. The thick membrane cisternae possess coated dilated rims and sometimes could be observed close to the endoplasmic reticulum. Using cytochemical techniques acid phosphatase, thiamine pyrophosphatase and nucleoside diphosphatase activities were detected in the thick membrane cisternae and in the alveolar sacs of trosphozoites. In the endoplasmic reticulum acid phosphatase activity was not detected and only very small amounts of thiamine pyrophosphatase and nucleoside diphosphatase reaction product were observed. After exit from the host, a reduction in acid phosphatase activity was evident in the alveolar sacs. At theront stage acid phosphatase activity is absent from these structures. However, high thiamine pyrophosphatase and nucleoside diphosphatase activities remain in the alveolar sacs during the whole life cycle. On the other hand, acid phosphatase, thiamine pyrophosphatase and nucleoside diphosphatase activities were detected in thick membrane cisternae of theronts. Based on the morphological aspects and enzymatic content the thick membrane cisternae of the cell cortex are designated as golgian-like cisternae. The cytochemical results point out a relationship between the alveolar sacs and the Golgi complex.  相似文献

Summary The localization of endogenous peroxidase was studied in the glandula orbitalis (lacrimalis) externa of the rat by the method of Graham and Karnovsky (1966). Reaction product is visible in all cisternae of the rough endoplasmic reticulum including the perinuclear cisternae, in condensing vacuoles, and in all secretion granules. The Golgi cisternae seldom are peroxidase positive. Intercalated duct cells rarely contain reaction product in a few scattered cisternae of the rough endoplasmic reticulum and in secretion granules.After the injection of beef liver catalase reaction product is found in the capillary lumen. Both the injected catalase and the endogenous peroxidase are completely inhibited by 10–2M aminotriazole, while the pseudoperoxidatic activity within the erythrocytes persists. After injection of horseradish-peroxidase reaction product is visible within the capillary lumen and also in the intercellular spaces between lacrimal gland cells. 10–2M aminotriazole completely inhibits the endogenous peroxidase while the exogenous horseradish-peroxidase remains unaffected. The inhibitory effect of aminotriazole is not specific for catalase since lacrimal gland peroxidase is also inhibited.  相似文献

Summary The occurrence of stacked annulate lamellae is documented for a plant cell system, namely for pollen mother cells and developing pollen grains of Canna generalis. Their structural subarchitecture and relationship to endoplasmic reticulum (ER) and nuclear envelope cisternae is described in detail. The results demonstrate structural homology between plant and animal annulate lamellae and are compatible with, though do not prove, the view that annulate lamellar cisternae may originate as a degenerative form of endoplasmic reticulum.  相似文献

The distribution of endogenous peroxidase activity in the lacrimal gland of the rat during postnatal development was investigated by electron microscope cytochemistry Peroxidase activity is first found 6 hr after birth in only a few acinar cells At this stage, reaction product fills only localized segments of the scant rough endoplasmic reticulum and of the perinuclear cisternae. Peroxidase activity thus develops asynchronously in a given cell as well as in the secretory cell population as a whole 2 days after birth, all cisternae of the rough endoplasmic reticulum of a peroxidase-positive cell contain reaction product, but the majority of the acinar cells is still negative During the next days, the number of peroxidase-positive cells and the amount of the rough endoplasmic reticulum increase rapidly. By 15 days postparturition, all secretory cells are peroxidase-positive. Reaction product is then found in all cisternae of the rough endoplasmic reticulum including the perinuclear cisternae, in smooth surface vesicles located mainly between the rough endoplasmic reticulum and the Golgi stacks, in condensing vacuoles, and in all secretory granules The Golgi cisternae rarely contain reaction product In total homogenates and in fractions of glandular tissue of adult rats, peroxidatic and catalatic activities are demonstrable. The microsomal fractions and the postmicrosomal supernatants were used to separate peroxidase from catalase by precipitation with ammonium sulfate, and the following parameters were determined: substrate (H2O2-) optimum (∼ 2.0 x 10-4 M), pH-optimum (pH 6 5), temperature-optimum (42°C), and the absorption maximum (415 nm before and 425 nm after addition of H2O2) The same parameters were obtained from lacrimal fluid peroxidase. Both peroxidase from lacrimal gland and that from lacrimal fluid are almost completely inhibited by 10-3 M aminotriazole and are possibly identical enzymes. Peroxidase is secreted into lacrimal fluid, which does not contain catalase.  相似文献

The participation of the Golgi complex—endoplasmic reticulum system during spermiogenesis in 3 species of phytophagous bugs (Acrosternum aseadum, Euchistus heros and Nezara viridula) were analysed at the ultrastructural level, after postfixation with osmium tetroxide\potassium iodide. Intense staining was found in the nuclear envelope, endoplasmic reticulum cisternae, Golgi complex, and in a meshwork of tubules scattered throughout the cytoplasm. It is shown that these compartments present a reducing environment. The presence of this kind of staining on the spermatid suggests an important participation of these structures on the differentiation process of this cell.  相似文献

Ultrastructural cytochemical techniques were used for the localization of phosphatases in spermatid and spermatozoon of the mosquito, Culex quinquefasciatus (Diptera : Culicidae). Acid phosphatase was found mainly in the trans-most portion of the Golgi complex. Thiamine pyrophosphotase was preferentially located in the cis-most portion of the Golgi complex. Glucose-6-phosphatase was located in the endoplasmic reticulum and cisternae of the transition zone between the endoplasmic reticulum and the Golgi complex. The complex membrane of the anterior acrosomal region and the axoneme showed acid phosphatase activity. Reaction products indicating the presence of acid phosphatase, thiamine pyrophosphatase, and glucose-6-phosphatase, were observed on the spermatozoon surface at the head and tail regions. These observations support the idea that various phosphatases may play some role in spermatid differentiation.  相似文献

Summary In three cell types of slug tissue-the crypt, mucous and storage cell-ultrastructural alterations of the endoplasmic reticulum (ER) can be induced by oral application of the pesticides Cloethocarb, metaldehyde, or Dimilin. In the crypt cells of the hepatopancreas, the narrow-luminar cisternae of the rough endoplasmic reticulum which are parallelly arranged in controls get slightly dilated, vesiculated and form circular arrays. Intermediate stages between narrow luminar, vesiculated and circularly arranged ER can be observed. In the mucous cells of the skin and the stomach, the wideluminar cisternae of the rough endoplasmic reticulum the lumen of which contains tubular-like structures become heavily dilated. Also in this cell type, intermediate stages between dilated cisternae without tubular-like structures and non-dilated cisternae can be observed. In the storage cells of the crop, in which lipid storage is reduced after molluscicide application, the formation of a special type of ER characterized by locally enlarged ER-cisternae, broken through by several cytoplasmic strings, becomes obvious.  相似文献

The gastrodermal Golgi apparatus of adult Schistosoma mansoni displays two distinct morphologies. In one type, there is an identifiable cis (forming) face where vesicles from the endoplasmic reticulum fuse to form the cisternae. A morphological change occurs in the cisternae as the trans (emitting) face is approached with the cisternae becoming progressively flattened. The cisternae at the emitting face produce a membrane-bound secretory granule with moderately electron-dense contents and a vacuolar structure that may be analogous to a condensing vacuole as reported in several vertebrate secretory cells. In a second type, vesicles possessing a thicker membrane than those of the transfer vesicles are observed at the emitting face. They are not observed when the secretory granules are present. Several cytochemical markers were used to aid in studying the polarity of the Golgi apparatus. Enzymes studied were thiamine pyrophosphatase (TPPase) (EC 3.6.1.1), nucleoside diphosphatase (NDPase) (EC 3.6.1.6) using uridine diphosphate as a substrate, and nicotinamide adenine dinucleotide phosphatase (NADPase) (EC 3.1.3.2). Reaction products from all enzyme markers were observed in the cisternae and, to some extent, in the transfer vesicles. At times, NADPase and TPPase reaction products were observed in all cisternae and in the transfer vesicles of the Golgi. When this distribution was evident, the latter vesicles were observed in clusters occasionally fusing with lipid-like globules dispersed throughout the gastrodermis. Heterogeneity in cisternae was observed when NDPase, TPPase, and osmium reduction techniques were used. NDPase activity was limited to the middle cisternae while reduced osmium was observed in the outer two cisternae and in some transfer vesicles. TPPase reaction product was also observed in the secretory granules and in the condensing vacuoles. It is hypothesized that a functional bipolarity may be demonstrated by the Golgi. Under certain stress conditions, the forming face of the Golgi may package lysosomal enzymes while the emitting region of the Golgi appears to be responsible for the packaging of the secretory granules. The fusion of transfer vesicles and, at times, secretory granules with lipid-like globules is postulated to represent a mechanism by which enzymes may be transported to the lumen of the cecum.  相似文献

Both the apical and the basal cell poles of the sensory cells in trigger hairs of Dionaea muscipula are structured identically. A complex of concentrically arranged endoplasmic reticulum cisternae occupies each of the poles. One to four vacuoles are enclosed within the central cisterna and contain polyphenols (deposits of “tannin”). Structural polarity, whether symmetric or asymmetric, as well as the occurrence of abundant endoplasmic reticulum and numerous mitochondria are characteristics of the perception cells of most animals and plants.  相似文献

Ribosomal bodies (RB), constituted of crystalline sheets of ribosomes interposed between flat parallel cisternae, are present in the oocyte and follicular cells of Lacerta sicula during the winter rest. During spring, the RB undergo progressive transformation into rough endoplasmic reticulum and free ribosomes, so that these formations are completely lacking during summer. The mechanism underlying the formation of RB and the possible significance that they may have for protein synthesis during oogenesis are discussed.  相似文献

The Golgi apparatus of plant cells is engaged in both the processing of glycoproteins and the synthesis of complex polysaccharides. To investigate the compartmentalization of these functions within individual Golgi stacks, we have analyzed the ultrastructure and the immunolabeling patterns of high-pressure frozen and freeze-substituted suspension-cultured sycamore maple (Acer pseudoplatanus L.) cells. As a result of the improved structural preservation, three morphological types of Golgi cisternae, designated cis, medial, and trans, as well as the trans Golgi network, could be identified. The number of cis cisternae per Golgi stack was found to be fairly constant at approximately 1, whereas the number of medial and trans cisternae per stack was variable and accounted for the varying number of cisternae (3-10) among the many Golgi stacks examined. By using a battery of seven antibodies whose specific sugar epitopes on secreted polysaccharides and glycoproteins are known, we have been able to determine in which types of cisternae specific sugars are added to N-linked glycans, and to xyloglucan and polygalacturonic acid/rhamnogalacturonan-I, two complex polysaccharides. The findings are as follows. The β-1,4-linked d-glucosyl backbone of xyloglucan is synthesized in trans cisternae, and the terminal fucosyl residues on the trisaccharide side chains of xyloglucan are partly added in the trans cisternae, and partly in the trans Golgi network. In contrast, the polygalacturonic/rhamnogalacturonan-I backbone is assembled in cis and medial cisternae, methylesterification of the carboxyl groups of the galacturonic acid residues in the polygalacturonic acid domains occurs mostly in medial cisternae, and arabinose-containing side chains of the polygalacturonic acid domains are added to the nascent polygalacturonic acid/rhamnogalacturonan-I molecules in the trans cisternae. Double labeling experiments demonstrate that xyloglucan and polygalacturonic acid/rhamnogalacturonan-I can be synthesized concomitantly within the same Golgi stack. Finally, we show that the xylosyl residue-linked β-1,2 to the β-linked mannose of the core of N-linked glycans is added in medial cisternae. Taken together, our results indicate that in sycamore maple suspension-cultured cells, different types of Golgi cisternae contain different sets of glycosyl transferases, that the functional organization of the biosynthetic pathways of complex polysaccharides is consistent with these molecules being processed in a cis to trans direction like the N-linked glycans, and that the complex polysaccharide xyloglucan is assembled exclusively in trans Golgi cisternae and the trans Golgi network.  相似文献

Affinity-purified, monospecific rabbit antibodies against rat pancreatic alpha-amylase and bovine pancreatic alpha-chymotrypsinogen were used for immunoferritin observations of ultrathin frozen sections of mildly fixed exocrine pancreatic tissue from secretion-stimulated (pilocarpine) rats and from overnight-fasted rats and guinea pigs. The labeling patterns for both antibodies were qualitatively alike: Labeling occurred in (a) the cisternae of the rough endoplasmic reticulum (RER) including the perinuclear cisterna, in (b) the peripheral area between the RER and cis-Golgi face, and (c) all Golgi cisternae, condensing vacuoles, and secretory granules. Labeling of cytoplasmic matrix was negligible. Structures that appeared to correspond to rigid lamellae were unlabeled. Differences in labeling intensities indicated that concentration of the zymogens starts at the boundary of the RER and cis-side of the Golgi complex. These data support the view that the Golgi cisternae are involved in protein processing in both stimulated and unstimulated cells and that Golgi cisternae and condensing vacuoles constitute a functional unit.  相似文献
