We have been successful in building an energy-level model thatdescribes seed germination. We used the autocatalytic reactionrate equation to fit the germination rate for seed germination.The two parameters [A]0 and [F]0 were found by fitting the integratedgermination rate equation to the data. The values of [A]0 and[F]0 obey the Arrhenius equation and give activation energiesfor the second and third stage of the four-compartmental modelof seed germination. The thermodynamics of isothermal seed germinationis proposed and the enthalpy, entropy, and free energy are calculatedfor the transition from state A to state F. The time delay isa function of temperature and it leads to a rate constant thatcan be used to get the activation energy for the total germinationprocess. We believe the model is universal. Use of the Weibull Function to Calculate Cardinal Temperatures in Faba Bean   总被引:1,自引:0,他引:1  
The onset of germination of faba bean seeds at constant temperaturewas progressively delayed as that temperature diverged froman optimum of 25.5 ?C. At temperatures below 10 ?C, or above28 ?C, the maximum germination percentage fell to below 90%.There was no germination at 39 ?C. Positive and negative linearrelationships were established between the constant temperaturesand the rates of progress of germination to different percentiles,at sub-optimal and supra-optimal temperatures, respectively.Like germination rates, base temperature (Tb) declined from3.71 to –0.83 ?C as the percentile value increased from10% to 80%. Caution was urged in extrapolating beyond the experimentaldata set. Differences in the ceiling temperature (Tc) with percentilecould not be discerned. Cumulative germination progress curves at each temperature weremodelled by the Weibull, logistic, and cumulative normal distributionfunctions. Cardinal temperatures (Tb and Tc) calculated fromthese data reasonably approximated the actual data. The Weibullfunction demonstrated a good approximation at all percentilelevels, while the logistic and cumulative normal distributionfunctions, as a result of their inherent symmetry, deviatedat the extreme percentiles. It was concluded that the Weibullfunction not only accurately modelled cumulative germinationbut could also be used in the calculation of cardinal temperatures. Salinity is one of the environmental factors that has a criticalinfluence on the germination of halophyte seeds and plant establishment.Salinity affects imbibition, germination and root elongation.However, the way in which NaCl exerts its influence on thesevital processes, whether it is through an osmotic effect ora specific ion toxicity, is still not resolved. Dimorphic seedsof the halophytesAtriplex prostrataandA. patulawere treatedwith various iso-osmotic solutions of NaCl and polyethyleneglycol (PEG). For each treatment, imbibition, germination rate,percent germination, germination recovery and nuclear area ofroot tip cells were compared. Higher concentrations of NaCl(-1.0 MPa) were more inhibitory to imbibition, germination andseedling root elongation than iso-osmotic PEG solutions. Allseeds recovered from a pre-treatment with -2.0 MPa NaCl andPEG solutions, except large seeds ofA. prostratawhich failedto germinate following transfer from -2.0 MPa NaCl. A quantitative model for change in germination rate of barley(Hordeum vulgare L.) during and after loss of primary dormancyis presented. Change in mean germination time on a logarithmicscale is normally distributed within the period of after-ripeningand the standard deviation of this distribution is shown tobe a quantitative function of after-ripening temperature. Therate of change of mean germination time is in inverse proportionto the product of the standard deviation and a parameter whichis characteristic of the seed population. The latter parameteris the rate constant for change in probit cumulative germinationas a negative linear function of the logarithm of mean germinationtime. A model based solely on dormancy loss is combined withan existing model of change in probit viability as a functionof mean germination time to produce a model which predicts thetime to and optimum value of mean germination rate of a populationas it after-ripens. The model provides a quantitative link betweenthe effect of pre-germination and germination environments ontotal and rate of germination of an initially dormant population.Experimental data from dormant barley (cv. Triumph) stored at27, 38, 45, 50 and 60 °C, and germinated at 18 °C wereused to validate the model. The data show that germination ratecontinues to increase after primary dormancy is lost until itreaches an upper limit determined by the intrinsic germinativevigour of the seed lot. Seeds of Chenopodium album L. were buried under field and controlledconditions. The germination capacity of these seeds was testedover a range of conditions at regular intervals. Seed buriedin the field only showed small seasonal changes in germinationcapacity when tested at constant temperatures in incubators.However, when germination was tested at field temperatures,seasonal changes in germination were more obvious. Nitrate andlight always promoted germination. There was a strong positiveinteraction between the effects of the two factors. When nitrateand light were combined, exhumed seeds germinated over a muchlonger period of the year than in water with or without light.Desiccation only stimulated under particular conditions, forexample, when germination was tested in nitrate in darkness.A regression model was developed with the data from the germinationtests in incubators. The model describes the changes in dormancyand germination and estimates germination at field temperaturesaccurately throughout the year. The different germination behaviour of the seeds of two irises,Iris lorteti and I. atropurpurea was found to be due to thedifferent mechanical resistance of the integument, at the micropylarend, to radicle protrusion. A pressure of 135 atm was necessaryin l. lorteti seeds for radicle protrusion, while in I. atropurpurea77 atm was sufficient. In contrast Pancratium maritimum requireda pressure of only 10 atm. The outer integument of seeds ofI. lorteti was found to contain a compound which was toxic tothe germinated embryo but did not act as a germination inhibitor.Extracts of the endosperm also had a slight germination inhibitingeffect. An interaction between this weak inhibitor and the effectof the testa could not be ruled out completely. A test assayfor germination using excised embryos was developed. A methodfor germination of Iris seeds, by cutting off the outer integumentat the micropylar end, was developed and is being exploitedcommercially. Various daily photon doses (daily net number of photons perunit area), provided by varying both photon flux density andphotoperiod, were applied to dormant seed populations of fourgraminaceous species tested for germination in alternating-temperatureregimes on filter papers moistened with either water or a solutionof potassium nitrate. In all four species germination was dependentupon daily photon dose, the product of photon flux density andduration, and quantified by linear relations between the probitof percentage germination and the logarithm of the daily photondose determined by probit analysis. Photon flux density andphotoperiod only influenced germination indirectly as factorsof daily photon dose, except in Echinochloa turnerana (Domin)J.M. Black at high daily photon doses where lower germinationwas observed in continuous light than in 8 h d–1 photoperiod.The low energy reaction, in which increasing dose promotes seedgermination, was observed in Echlnochloa turnerana and Eragrostistef(Zucc.) Trotter but not in the populations of Brachiariahumidicola (Rendle) Schwcickerdt and Panicum maximum Jacq. usedin this study. The high irradiance reaction, in which increasingthe photon dose reduces germination, was, however, observedin all four species. In Eragrostis tef potassium nitrate influencedthe response of seed germination to light, but in the remainingspecies its influence on this relationship was either additive(Panicum maximum) or nil (Brachiaria humidicola and Echinochloaturnerana) in the alternating-temperature regimes used in thisstudy. The implications of the quantal response to photon dosefor devising optimal light regimes to be applied in laboratorytests intended to maximize seed germination are discussed. Several models have been proposed to describe germination rates,but most are limited in statistical analysis and biologicalmeaning of indices. Therefore, a mathematical model is proposedto utilize the logistic function. The function was defined asan overall response including time, temperature, and the interactionbetween time and temperature. Cumulative germination percentagesover time were used to develop the model. Germination tests were conducted on indiangrass (Sorghastrumnutans (L.) Nash) strain ‘IG-2C-F1’, at constanttemperatures of 9, 12, 15, 20, 25, and 30 °C. The functionfitted the observed data over six temperatures at r2 = 0.99.Time to reach 10% of final germination (Gt10) increased from2.5 d at 30 °C to 44.0 d at 9 °C, and Gt50 (time toreach 50% of final germination) increased from 3.6 d at 30 °Cto 53.8 d at 9 °C. True germination rate (% d–1) foreach temperature was maximum at Gt50. A linear model of 1/Gt50versus temperature was used to estimate the base temperatureof 8.3 °C for germination. An Arrhenius plot indicated achange occurred between 20 °C and 25 °C for temperatureresponse of germination. Published data on hypocotyl growthof Cucumis melo L. were recalculated using the model. Absolutegrowth rates showed a temperature response similar to the publishedweighted-mean elongation rates. Base temperature for hypocotylgrowth of C. melo was estimated as 8.8 °C. A method of quantifying the heterogeneity of dormancy withinseed populations is described. The method compares germinationresponses of unstratified and stratified seeds of Pinus taedaL. (loblolly pine). Mathematical functions were fitted to cumulativegermination curves of unstratified and stratified seeds andfunctions were integrated to determine areas beneath respectivecurves. The area between respective curves was calculated bydifference and was considered to be a measure of dormancy withinseed lots. Allenrolfea occidentalis(Chenopodiaceae) is a highly salt tolerantplant species that is widely distributed in inland salt marshesand salt playas of the western United States. We investigatedthe influence of dormancy-relieving compounds (fusicoccin, ethephon,nitrate and thiourea) in alleviating salinity stress on theseed germination ofA. occidentalis. Seed germination decreasedwith an increase in salinity and no seed germinated at 800 mMNaCl.Fusicoccin (5 µM), ethephon (10 mM) and nitrogenous compounds(20 mMnitrate and 10 mMthiourea) were able to counteract theinhibition produced by salinity treatments. All dormancy relievingcompounds significantly (P<0.0001) promoted germination atall salinity concentations. Fusicoccin completely reversed theinhibitory effects of salinity on seed germination ofA. occidentalis.Ethephon application significantly promoted germination at allsalinities. Background and Aims: Seed physiological dormancy (PD) limits the use and conservationof some of Queensland's (Qld) native forb species. It was hypothesisedthat optimum dormancy-alleviating treatments would reflect environmentalconditions that seeds experience in situ, and this premise wastested for PD seeds of four species native to south-west Qld. Methods: High temperatures and increased rainfall during summer are characteristicof this semi-arid tropical environment. Ex situ treatments weredesigned to mimic conditions that seeds dispersed in springexperience during the summer months before germinating in coolerautumn temperatures. Seeds received between 4 and 20 weeks ofa dry after-ripening (DAR), warm stratification or dry/wet cyclingtreatment (DAR interspersed with short periods of warm stratification),in darkness, before being transferred to germination test conditions.In addition, natural dormancy alleviation of one of the Goodeniaceaespecies was investigated in situ. Key Results: Dry/wet cycling resulted in higher levels of germination ofActinobole uliginosum (Asteraceae), Goodenia cycloptera andVelleia glabrata (Goodeniaceae) when compared with constantDAR or stratification, while Goodenia fascicularis (Goodeniaceae)responded better to short durations of warm stratification.Long durations of DAR partially alleviated PD of A. uliginosum;however, stratification induced and maintained dormancy of thisspecies. Modifications to the dry/wet cycling treatment andgermination test conditions based on data collected in situenabled germination of G. cycloptera and V. glabrata to be furtherimproved. Conclusions: Treatments designed using temperature, relative humidity andrainfall data for the period between natural seed dispersaland germination can successfully alleviate PD. Differences betweenthe four species in conditions that resulted in maximum germinationindicate that, in addition to responding to broad-scale climatepatterns, species may be adapted to particular microsites and/orseasonal conditions.  相似文献

Seeds of the obligate parasitic plants, Orobanche spp., wereconditioned in water or GA3for 2 or 12 weeks and then stimulatedto germinate by the synthetic stimulant GR24. Temperature treatmentsduring the germination tests comprised 169 different constantand alternating temperature regimes on a two-dimensional gradientplate. Optimum temperatures for germination of seeds of O. aegyptiacaand O. crenata were 18–21 °C and 18 °C, respectively.However, longer conditioning periods slightly lowered the optimain both species, and the maximum germination percentage wasalso reduced due to an induction of secondary dormancy. At agiven mean temperature, more seeds germinated at constant thanat alternating temperatures. Results were analysed in termsof characteristics of alternating temperatures that appearedto control germination, i.e. mean temperature, maximum temperature,amplitude (difference between daily maximum and minimum temperatures)and thermoperiod (the time spent at the maximum temperatureeach day). Final germination was modelled on the basis of therebeing two prerequisites for germination: a minimum mean temperaturewhich must be exceeded and a maximum temperature above whichthe seed will not germinate. These two requirements were assumedto be independent and to be normally distributed in the seedpopulation so that final germination could be described by amultiplicative probability model. Because of the response tomaximum temperature, inhibitory effects were more evident atalternating temperatures. Amplitude and thermoperiod influencedthis effect of maximum temperature. The implications of thedetrimental effect of alternating temperatures for germinationofOrobanche spp. in the field are discussed. Positive linear relationships were shown between constant temperaturesand the rates of progress of germination to different percentiles,G, for single populations of each of five genotypes of chickpea(Cicer anetinum L.). The base temperature, Tb, at which therate of germination is zero, was 0·0°C for all germinationpercentiles of all genotypes. The optimum temperature, To(G),at which rate of germination is most rapid, varied between thefive genotypes and also between percentiles within at leastone population. Over the sub-optimal temperature range, i.e.from Tb to To(G), the distribution of thermal times within eachpopulation was normal. Consequently a single equation was appliedto describe the influence of sub-optimal temperatures on rateof germination of all seeds within each population of each genotype.The precision with which optimum temperature, Tb(G), could bedefined varied between populations. In each of three genotypesthere was a negative linear relationship between temperatureabove Tb(G) and rate of germination. For all seeds within anyof these three populations thermal time at supra-optimal temperatureswas constant. Variation in the time taken to germinate at supra-optimaltemperatures was a consequence of normal variation in the ceilingtemperature, To(G)—the temperature at or above which rateof progress to germination percentile G is zero. A new approachto defining the response of seed germination rate to temperatureis proposed for use in germplasm screening programmes. In two populations final percentage germination was influencedby temperature. The optimum constant temperature for maximumfinal germination was between 10°C and 15°C in thesepopulations; approximately 15°C cooler than the optimumtemperature for rate of germination. Cell division patterns during germination of spores of Cyatheaaustralis, C. cooperi and Dicksonia antarctica were examinedby light microscopy of sectioned materials and by the scanningelectron microscope. In C. australis and C. cooperi the rhizoidwas traced to a small cell formed by an asymmetric divisionof the spore by a wall parallel to its equatorial plane. Incontrast, the rhizoid was formed by a division of the sporeparallel to its polar axis in D. antarctica. In spores of bothgenera, a second division wall oriented in a plane perpendicularto the first gave rise to the protonemal cell. Certain aspectsof germination described here in spores of Cyathea and Dicksoniaare in conflict with the published accounts of spore germinationin these genera. Stigma-surface esterase activity and stigma receptivity througha sequence of developmental stages of the pistil have been studiedin four taxa characterized by having wet stigmas — Petuniahybrida, Nicotiana tabacum, Crinum defixum and Amaryllis vittata.The style is solid in the first two and hollow in the lattertwo taxa. In all the taxa, stigma—surface esterase couldbe detected in a thin surface layer (pellicle) from a very earlystage of pistil development, irrespective of the presence orabsence of the exudate. However, the taxa showed variation instigma receptivity. In Petunia and Nicotiana, stigmas from pistilsof all the stages supported pollen germination and tube growth.In Amaryllis and Crinum, stigmas of only the mature pistils,when the exudate is present on the stigma, supported normalpollen germination and tube growth. It is inferred that in taxacharacterized by a wet stigma and solid style, the factors requiredfor pollen germination are present from an early stage of pistildevelopment and the exudate per se is not involved in pollengermination. In taxa characterized by a wet stigma and hollowstyle, however, the pellicle does not carry the factors requiredfor pollen germination and tube growth; they appear to be presentin the exudate. Somatic embryos ofVitis vinifera L. ‘Grenache noir’develop abnormally and form viable plantlets at very low frequencies.They grow continuously and, after the torpedo stage, they formgiant structures which do not undergo further organogenesis.Morphological, histological and cytochemical data were usedto study development from the globular to the giant-embryo stage.Histological organization of somatic embryos until the torpedostage was similar to that of zygotic embryos. Somatic embryosformed bipolar axes, which differentiated precociously and simultaneouslya root and a shoot meristem. However, they differed from theirzygotic homologues by forming a cotyledonary crown or multiplecotyledons and by their rapid cellular differentiation. At theend of the torpedo stage and up to the giant-embryo stage, somaticembryos underwent some characteristic events of germination:the radical grew, tannins accumulated, and protodermal cellssuberized. However the shoot apex was rapidly disorganized anddisappeared. This peculiar behaviour is discussed in comparisonwith the phenomenon of precocious germination often observedfor immature zygotic embryos inin vitro culture. Germination of strawberry seed was accelerated by natural andartificial infections of Ulocladium charatarum and other fungi,including the Arthrinium state of Apiospora montagnei and aCladosporium sp. Germination was variously promoted by fungiboth in light and darkness, on damp filter paper, water andmalt agar plates and on horticultural compost. Germinated seedlingswere apparently undamaged by these infections, although whencultured on malt agar the seedlings were overgrown by the fungus.Several other fungi promoted germination, but three of themlater killed the seedlings. Germination of seed of all threecultivars that were tested was promoted by fungal infectionwhich probably promoted germination by hastening breakdown ofthe seed coat. Dormant E. crus-galli seeds can be stimulated to germinate bycontact with solutions of several anaesthetic-like substances.The order of activity was n-pentanol > benzyl alcohol >n-butanol > n-propanol > ethanol. 2-propanol is nearlyinactive. The relative activity of the substances as germinationstimulants is closely related to their membrane-buffer partitioncoefficients in a manner similar to their effectiveness as anaestheticsin animals. Thus, n-pentanol is a more active stimulant of germinationthan ethanol and its increased effectiveness is correlated witha higher membrane-buffer partition coefficient Similarly, ananaesthetic that stimulated germination (n-propanol) causedgreater leakage of electrolytes, U.V. absorbing substances andamino acids from E. crus-galli seeds than a substance that wasnot active as a germination stimulant (2-propanol). In animals,action of anaesthetics can be prevented by application of externalpressure, an argument for membrane action. Action of n-propanolin stimulating germination can be similarly prevented by pressurein E. crus-galli seeds. The several lines of evidence supporta membrane action for anaesthetic-like substances in seeds and,accordingly, a membrane site as the locus of seed dormancy. Vapour from incubated Pinus radiata D. Don litter caused a depressionin the germination of Lolium perenne L. seeds and responsesvarying from stimulation to depression in germination of Trifoliumrepens L. seeds. The seeds ofNasturtium palustreDC. do not germinate, eitherin the light or darkness, at various constant temperatures,but require for their full germination a certain period of alow temperature (5°) applied immediately after light irradiation.These results indicate the existance of at least two processes,a light-dependent process and a low temperature-requiring process,in the initiation of germination ofNasturtiumseeds. Experimentalevidence indicated further that the light exposure causes twodifferent processes in the seed germination. 2. When a dark period at 23° was inserted between the lightirradiation and the low temperature treatment the germinationwas suppressed. The inhibitory effect of the inserted dark periodat 23° was eliminated by a short irradiation during thedarkness (light-break). 3. Prolonged exposure ofNasturtium seeds to any concentrationof gibberellin brought about no germination when exposure wasgiven in complete darkness. The germination was promoted onlywhen light irradiation was applied to the seeds. A short applicationof gibberellin at a fairly high concentration was, however,remarkably effective for the germination even in the darkness,and the germination was inhibited as the gibberellin applicationwas lengthened.
