本文介绍一种新技术──微波加强抗体洗脱法。在酸洗脱液中微波辐射(最低能量档)3分钟.完全除去经PAP法染色后的垂体前叶切片上的抗体复合物;双重染色显示两种形态和颜色不同的细胞。切片在Tris缓冲盐液中微波辐射3分钟,或分别浸入室温和约50℃的酸洗脱液中2小时和3分钟,抗体复合物不能被洗脱,或洗脱不完全;双重染色时出现假性双标记细胞。结果表明微波具有加强抗体洗脱的作用,是一种快速、有效的除去免疫酶双重染色中抗体复合物的方法。  相似文献

运用ABC—爱先蓝—PAS混合染色及PAP技术对大鼠肝、胃、肠及DAB诱发的大鼠肝癌中肥大细胞进行了染色对比实验,结果表明:Carnoy及福尔马林等固定液固定的组织内肥大细胞均能被爱先蓝染色成蓝色,其染色时间不同,该种染色方法可作为一种常规的染色方法,用于确定肥大细胞在组织中的分布及数量。ABC—爱先蓝—PAS混合染色方法及PAP技术均能准确、有效地确定肥大细胞(MC)性质。ABC—爱先蓝—PAS混合染色使结缔组织肥大细胞(CTMC)呈棕褐色,周边略蓝色。粘膜肥大细胞(MMC)则呈蓝色,通过不同颜色的显示,可清楚地将CTMC与MMC区分开来。PAP方法具更高的特异性。但有关的抗体来源缺乏,因此在无特异Ⅰ抗体的情况下,ABC—爱先蓝—PAS混合染色方法亦可视为鉴别两类不同性质MC的一种较为理想的方法。  相似文献

目的比较ABC法和SP法两种免疫组织化学染色方法的染色效果。方法通过免疫组化检测一氧化氮合酶的三个亚型的蛋白i NOS、e NOS和n NOS的表达,观察三种蛋白的染色结果。结果 SP法检测i NOS和e NOS的敏感性要明显强于ABC法;SP法蛋白染色结果明显优于ABC法。结论 SP法检测NOS优于ABC法。  相似文献

目的 比较冰冻切片技术中几种防冰晶方法对切片保存效果的影响。方法 分别使用液氮骤冷组织、高渗透压脱水法、单纯OCT胶包埋等几种方法后行冰冻切片、HE染色后,分别于当天、1、3、6个月后比较其染色效果,选出最佳保存方法。结果 单纯OCT包埋法在1个月后染色明显变浅,冰晶数量最多,而在3、6个月后染色基本接近背底色,冰晶面积已占据近半个视野,组织结构破坏极其严重;高渗透压脱水法在1个月后染色深度及冰晶数量上无明显变化,但3、6个月后则明显变浅,冰晶数量明显增加;而低温骤冷法在染色后的几个月,染色深度及冰晶的数量均无明显的变化,结构基本维持染色当天的状态。结论 低温骤冷法是这三种防冰晶法中最利于保存切片的方法。  相似文献

目的比较石蜡和冰冻两种不同切片用于检测牛蛙肝糖原的效果。方法采用石蜡和冰冻两种切片方法制作牛蛙肝脏切片,高碘酸希夫氏(periodicacid-Schiff's,PAS)染色法对肝糖原进行组织化学染色,光密度分析糖原含量。结果PAS染色显示肝糖原为紫红色或红色颗粒,冰冻切片中糖原颗粒明显大于石蜡切片,光密度分析显示,石蜡切片中糖原流失明显,与冰冻切片相比,糖原流失了约28%。结论两种切片均可用于糖原检测,冰冻切片制作环节较少、耗时短且染色过程中糖原不易流失。  相似文献

免疫组化染色技术以其操作简单、敏感性高、特异性强,能将形态和抗原定位结合起来等优点广泛应用于肿瘤临床病理诊断和鉴别诊断[1].但是影响免疫组化的因素较多[2-5],存在经验和判断上的差异.石蜡切片在免疫组化染色中的脱片问题由来已久.切片经抗原修复和缓冲液的长时间浸泡后,往往会部分或全部从载玻片上脱落,影响染色结果的判断.不同的粘附剂和粘附剂不同的涂胶方法防脱片效果不尽相同[6-7].我们在工作中也经常遇到石蜡切片脱落问题,为此探讨了多聚赖氨酸较好的涂胶方法,又进一步对三种粘附剂即APES(氨醛三已氧基硅烷),多聚赖氨酸和白乳胶的防脱片效果和染色情况作了比较,成功地总结出多聚赖氨酸双涂胶法.现介绍如下:  相似文献

 应用 3种方法 (肿瘤细胞膜表面和胞内、裸鼠体内和组织切片 ) ,从全人源化的抗大肠癌噬菌体初级抗体库中筛选肿瘤特异性的噬菌体单链抗体 (Sc Fv) .在肿瘤细胞经过 3轮亲和选择 ,回收结合胞膜和内化进入胞内的噬菌体 ,得到抗肿瘤噬菌体单链抗体的富集倍数为 430倍 ;荷瘤裸鼠体内注入初级抗体库后 ,在不同时刻点处死裸鼠 ,回收肿瘤组织内的噬菌体 ,其回收率在 2 4 h时最高 ;初级抗体库与大肠癌组织切片亲和选择后 ,从冰冻组织切片上比从石蜡组织切片上回收得到的噬菌体高出约 1 .6倍 .从上述方法挑选单克隆 ,经 ELISA筛选抗大肠癌阳性噬菌体克隆株 ,分离得到 5个对大肠癌细胞反应较好的单克隆噬菌体单链抗体 .进一步用细胞 ELISA检测对各种肿瘤细胞的特异性反应 ,其中 4个对大肠癌细胞有很好的特异性 ,1个克隆对所有肿瘤细胞均有反应 .因此 ,3种方法用于筛选抗大肠癌噬菌体初级抗体库是有效的 ,具有推广和应用价值 .  相似文献

目前,在HE染色、特殊染色或免疫组织化学染色研究方面多采用普通石蜡包埋切片,其切片厚度可薄达4μm,若更薄的切片,切割有一定的难度.对于各种染色而言,越薄的切片,细胞与组织结构越清晰,染色效果越好.环氧树脂(Epon)包埋的组织块不仅用于透射电子显微镜超薄切片的制备,也可用于光镜半薄切片的制备[1].经典的脱树脂的方法是将半薄切片置于氢氧化钠的无水乙醇饱和溶液中浸泡24h,然后再充分水洗.该法耗时长,容易脱片,本实验中摸索出一种简单而快捷的脱树脂的方法,现介绍如下.  相似文献

探索一种不用在卵壳上手工打孔、适用于果蝇胚胎石蜡切片制作的方法。实验前期对果蝇胚胎进行渗透化处理,后期用琼脂糖、明胶、石蜡3种包埋辅佐剂对胚胎进行聚集、包埋、切片,最后进行HE及PASM染色。渗透化处理后的胚胎用琼脂糖或石蜡聚集并包埋后所制切片HE染色均显示胚胎形态结构正常,背景干净,而琼脂糖聚集法所制切片PASM染色背景呈灰色,明胶聚集法所制切片HE染色和PASM染色背景均显杂色且胚胎出现形变。3种包埋辅佐剂中,琼脂糖和石蜡均适于果蝇胚胎结构的保存,石蜡聚集法所制切片2种染色背景均无杂色,因此最适合做包埋辅佐剂。渗透化处理和石蜡聚集法结合后的改良方法适合于大样本且高脂含量的果蝇胚胎石蜡切片的制备。  相似文献

In the present study we developed an immunoenzymatic double staining technique allowing the simultaneous detection of two neuroactive substances with primary antibodies of the same species and their simultaneous visualization in semithin sections of epoxy-embedded material. For this purpose, primary antibodies against glutamate, GABA, and serotonin were either biotinylated or labeled with the trinitrophenyl (TNP) group. The latter was visualized by a detection system here referred to as the hapten-anti-hapten bridge (HAB) technique. The HAB technique consists of anti-TNP antibodies, serving as bridges between the TNP-ylated primary antibody, and a TNP-ylated marker enzyme, such as alkaline phosphatase. The single components of the HAB technique were optimized by use of a dot-blot assay and an "artificial tissue" system. The optimal staining sequence consisted of TNP-ylated primary antibody with a molar TNP:antibody ratio of 12:1, followed by anti-TNP antibody and TNP-ylated alkaline phosphatase (molar TNP:enzyme ratio of 20:1). No further improvement of detection sensitivity could be obtained when soluble immunocomplexes between anti-TNP antibody and TNP-ylated alkaline phosphatase on the side of phosphatase excess were prepared and used instead of simple TNP-ylated alkaline phosphatase. When compared with other established procedures, such as avidin-conjugated alkaline phosphatase or the ABC method, the HAB technique revealed a similar detection sensitivity. The TNP-ylated primary antibody, however, had to be used at higher concentration than the corresponding unlabeled primary antibody. The suitability of the HAB technique in combination with a modified three-step ABC technique for the simultaneous demonstration of glutamate-like and GABA-like immunoreactivity in the rat brain was demonstrated. The advantages of the new technique in comparison with existing double staining methods are discussed.  相似文献

Upon plotting of areas against optical densities in immunocytochemically stained tissue sections, hyperbolic curves were obtained which could be reduced to two straight lines, one representing variations in stained structures, and the other variations in background. The slopes of the stained structure lines reflected staining intensity independently of total area of stained structure in a section. The ratio of slopes of the stained structure and background lines reflected immunocytochemical sensitivity. A comparison of the peroxidase-antiperoxidase (PAP) method with the avidin-biotin complex (ABC) method showed that at usual antibody dilutions the PAP method was much more sensitive than the ABC method, while at impractically high antibody dilutions it was moderately more sensitive. Once sufficient dilutions of antibodies were reached, staining intensities dropped sharply with the PAP method. On the other hand, the dilution curves were flat with the ABC method. The ABC method consequently appeared unsuitable for estimating variations in concentration of antigen or for distinguishing high or low concentrations of antigen. The ABC method provided a stain for myelin even in the absence of any antibodies.  相似文献

The protooncogene product HER-2/neu is the target of the humanized monoclonal antibody trastuzumab (Herceptin). Several tests are used clinically to identify patients with HER-2/neu overexpression based on evaluation by pathologists of gene amplification by fluorescence in situ hybridization or protein expression using immunohistochemistry (IHC). A simple technique has been developed for staining formalin-fixed, paraffin-embedded breast cancer tissue using unmodified Herceptin/trastuzumab as the primary antibody. Results were compared with staining with the commercial kit, HercepTest, as well as with polyclonal anti-HER-2/neu antibodies and with biotinylated trastuzumab. These procedures were tested using four breast cancer microarrays. There were 854 cores that were stained with all four antibodies, representing 325 cases. A standard 4-point scoring system (0-3) was used. A total of 156 cases (48%) were scored as 0 by all the methods used and 31 (9.5%) were positive (3+) by all methods. Of interest, three cases scored negative using polyclonal anti-HER-2/neu antibodies but were positive using unmodified trastuzumab. To clarify this discrepancy, whole sections of tumors were examined with both antibodies using double labeling. There were some tumors that demonstrated a mosaic pattern of staining with neighboring cells or groups of cells stained exclusively with one antibody or the other. These results demonstrate that unmodified humanized or human therapeutic antibodies could be used for preclinical testing or in a clinical laboratory setting for IHC-based selection of patients for treatment, and results of such selection could be different from those obtained using polyclonal antibody-based IHC procedure.  相似文献

Summary A combination of the PAP- and ABC-techniques was developed to enhance the intensity of the immunocytochemical staining with monoclonal antibodies at light and electron microscopical levels. This amplification technique could be performed in 4 (single PAP + ABC) or 6 (double PAP + ABC) sequential steps depending on the quality of the primary antibodies used and the processing of the tissue before the immunocytochemical reaction: First step — Incubation of the tissue sections with the monoclonal primary antibodies; Second step — biotinylated anti-rat or anti-mouse IgG; Third step — monoclonal PAP complex; Fourth step — ABC complex which binds to the biotinylated secondary antibody. If stronger enhancement of the immunostaining has required the steps 2 and 3 could be repeated followed by the 6th step — the ABC complex. Choline acetyltransferase-like immunoreactivity of the rat hypoglossal nucleus and desmin- and vimentin-like immunoreactivity of human testis were studied. After the 4-and more pronounced the 6-step reaction a significant increase of the staining intensity was observed for all the reactions under study. ChAT-like immunoreactivity was observed to longer distances of the nerve cell dendrites after their emerging from the perikarya and within a greater number of structures in the neuropil as compared to the standard techniques. At electron microscopical level the technique permits longer fixation of the tissue which is important for the better preservation of the ultrastructure as well as for the easier recognition of the reaction product even in the smallest dendrite branches and the axons of the nerve cells. Stronger staining intensity was obtained for desmin-like immunoreactivity (LI) (within myofibroblasts of the lamina propria and muscle cells of the blood vessels)-and vimentin-LI (within Sertoli cells, myofibroblasts of the lamina propria and fibroblasts of the interstitium) of the human testis. The full combination (6 step-reaction) permits the detection of smallest quantities of an antigen in sections of different tissues using highly diluted primary antibodies. The technique represents a further alternative among the immunocytochemical amplification methods.  相似文献

Summary The effect of microwave antigen retrieval on the immunostaining of human duodenal endocrine cells in formaldehyde-fixed, paraffin-embedded material was investigated. The sections were immunostained by the avidin-biot in complex (ABC) and immunogold-silver autometallography (IGSS) methods with and without prior microwave treatment. Dilutions of up to 1:30 000 of the following antisera/antibodies were used: anti-chromogranin A, anti-chromogranin AB, anti-secretin, anti-gastrin, anti-gastric inhibitory polypeptide, anti-somatostatin and anti-serotonin. The detection threshold for all the antibodies was lower after antigen retrieval, and the primary antibody could be used in higher dilutions. The dilutions varied for different antibodies and were between two and ten times the optimal dilution without antigen retrieval. At extremely high dilutions of, or without, the primary antibody, non-specific staining of some lymphocytes and the mucus of some goblet cells was observed when the avidin method was applied, but not with the immunogold technique. This phenomenon was not observed when optimal dilution or a lower dilution was used. This seems to have been caused by the binding of the avidin-biotin complex to epitopes in these structures unmasked by microwave treatment when competition with specific binding sites was absent. An avidin-biotin alkaline-phosphatase (ABAP) staining method has been developed for the labeling of tissue sections and cell smears. The introduction of alkaline phosphatase as a marker enzyme through an avidin bridge results in excellent immunocytochemical labeling of different antigens using poly- and monoclonal antibodies. This technique avoids problems with endogenous peroxidase activity that sometimes occur using peroxidase staining procedures. The introduction of a preformed avidin-biotin alkaline-phosphatase complex (ABAPC) makes the presented technique as simple to handle as the widely used avidin biotin-peroxidase complex method (ABC). The ABAPC technique could be combined with other enzymatic labelings for double immunoenzymatic staining.  相似文献

Immunohistochemistry (IHC) is a valuable technique utilized to localize/visualize protein expression in a mounted tissue section using specific antibodies. There are two methods: the direct and indirect method. In this experiment, we will only describe the use of indirect IHC staining. Indirect IHC staining utilizes highly specific primary and biotin-conjugated secondary antibodies. Primary antibodies are utilized to discretely identify proteins of interest by binding to a specific epitope, while secondary antibodies subtract for non-specific background staining and amplify signal by forming complexes to the primary antibody. Slides can either be generated from frozen sections, or paraffin embedded sections mounted on glass slides. In this protocol, we discuss the preparation of paraffin-embedded sections by dewaxing, hydration using an alcohol gradient, heat induced antigen retrieval, and blocking of endogenous peroxidase activity and non-specific binding sites. Some sections are then stained with antibodies specific for T cell marker CD8 and while others are stained for tyrosine hydroxylase. The slides are subsequently treated with appropriate secondary antibodies conjugated to biotin, then developed utilizing avidin-conjugated horseradish peroxidase (HRP) with Diaminiobenzidine (DAB) as substrate. Following development, the slides are counterstained for contrast, and mounted under coverslips with permount. After adequate drying, these slides are then ready for imaging.  相似文献

Visualization of rabbit cathepsin B was achieved utilizing monospecific sheep antibodies and the avidin-biotin-peroxidase complex (ABC) method. This technique was applied to stain 1) paraffin sections of the liver, 2) fixed fibroblasts from tissue culture, and 3) fixed mesenteries. Cathepsin B was found to be localized within cells of the lining of the liver sinusoids (most probably Kupffer cells), in perinuclear granules of cultured fibroblasts, and within histiocytes of the mesentery. The results demonstrate that the method permits precise and highly sensitive localization of cathepsin B within cells and tissues. Compared to fluorescent staining of cathepsin B, the ABC method has the advantage that routine paraffin sections can be stained, and that all the orthodox histological staining procedures can still be carried out.  相似文献

Many of the antigens commonly investigated in histopathology can be enhanced by microwave pretreatment (MWPT) of formalin fixed, paraffin embedded tissue sections. We developed a double labeling method using microwave heating to detect otherwise undetectable nuclear antigens combined.with immunohisto-chemistry (IHC) of cytoplasmic or membranous antigens that do not benefit from MWPT. We used the same primary antibody solutions used in single antibody IHC. The staining technique is based on the alkaline phosphatase anti-alkaline phosphatase (APAAP) and the labeled avidin-biotin (LSAB) methods. Four different protocols were tested, each modifying the sequence of MWPT, APAAP and LSAB staining. In this study Ki67, estrogen receptor, progesterone receptor, c-neu, CD68 and desmin primary antibodies were used in routinely formalin fixed, paraffin embedded tissues of 50 tumor specimens. MWPT followed by LSAB for microwave enhanced antigens and APAAP for antigens that cannot be enhanced by MWPT gave the best double staining results. This method improves characterization of tumor cell features from paraffin embedded tissue and should aid analysis of tumor differentiation, receptor status and nuclear proteins in the single cells in archival tissues.  相似文献
