Liquid-stored red blood cells and washed, previously frozen red blood cells were studied to determine whether a correlation existed between morphology and post-transfusion survival. Red cell concentrates were stored at 4 °C in citrate-phosphate-dextrose (CPD) for 21 days or in CPD-adenine (CPDA-1, CPDA-2, or CPDA-3) for as long as 35 days as liquid-preserved red cells. Both nonrejuvenated and rejuvenated red blood cells were frozen with 40%wv glycerol at ?80 °C and were washed prior to testing.Samples of fresh, liquid-stored, and washed, previously frozen red blood cells were fixed with a 2% veronal glutaraldehyde solution. Phase, light, and electron microscopy were used to measure the numbers of discocytes, discoechinocytes, echinocytes, echinospherocytes, and spherocytes in each sample. A morphology score was assigned, with 100 representing all discocytes and 500 all spherocytes. In all samples phase and light microscopy gave nearly identical scores (r = 0.94), and phase and electron microscopy gave highly similar scores (r = 0.83).The morphology score proved to be a good indicator of 24-hr post-transfusion survival in liquid-stored red blood cells but not in washed, previously frozen red blood cells. Red blood cells stored in the liquid state at 4 °C in CPD, CPDA-1, CPDA-2, or CPDA-3 showed a significant inverse correlation between morphology and 24-hr post-transfusion survival (r = ?0.611) and a significant correlation between red cell ATP and 24-hr post-transfusion survival (r = 0.742). We saw no significant correlation between morphology scores and 24-hr post-transfusion values or between ATP levels and post-transfusion survival values in nonrejuvenated or rejuvenated washed, previously frozen red blood cells.  相似文献

The nucleotide profile of rat liver Golgi vesicles isolated using sucrose gradients has been determined by high-pressure liquid chromatography. The nucleotide composition of this Golgi preparation, probably modified by osmotic shock, differs from that of liver supernatant fraction and from isolated rough microsomes. The major nucleotides present in the Golgi have been tentatively identified as uridine diphosphate and a peak containing uridine monophosphate plus cytidine monophosphate at 1.6 and 0.5 nmol/mg protein, respectively. In order to minimize osmotic shock, we have modified the isolation of Golgi using D2O-sucrose gradients. Intact Golgi from these gradients were extracted directly and analyzed. Higher levels of nucleotides were found in the unshocked preparation, and the profile was also altered, although it was still distinct from that of liver supernatant. Four major peaks were found, tentatively identified as uridine monophosphate plus cytidine monophosphate, adenosine monophosphate, UDP, and uridine diphosphogalactose plus uridine diphosphoglucose, at 6.4, 6.4, 6.1, and 3.3 nmol/mg protein. These results indicate that the membrane of the Golgi apparatus is not freely permeable to nucleotides but that selective mechanisms exist for the uptake or exclusion of specific nucleotides from this organelle. The fact that UDP is selectively retained in shocked Golgi vesicles may indicate the presence of a binding protein which would prevent interference of Golgi function by UDP, a highly inhibitory product of galactosyltransferase.  相似文献

The ability of hemoglobin (Hb) to transport respiratory gases is directly linked to its quaternary structure properties and reversible changes between T (tense) and R (relax) state. In this study we demonstrated that packed red blood cells (pRBCs) storage resulted in a gradual increase in the irreversible changes in the secondary and quaternary structures of Hb, with subsequent impairment of the T↔R transition. Such alteration was associated with the presence of irreversibly settled in the relaxed form, quaternary structure of Hb, which we termed R′. On the secondary structure level, disordered protein organization involved formation of β-sheets and a decrease in α-helices related to the aggregation process stabilized by strong intermolecular hydrogen bonding. Compensatory changes in RBCs metabolism launched to preserve reductive microenvironment were disclosed as an activation of nicotinamide adenine dinucleotide phosphate (NADPH) production and increased reduced to oxidized glutathione (GSH/GSSG) ratio. For the first time we showed the relationship between secondary structure changes and the occurrence of newly discovered R′, which through an artificial increase in oxyhemoglobin level altered Hb ability to bind and release oxygen.  相似文献

On the basis of the fact that selenium from selenite binds to hemoglobin (Hb), we investigated the missing process in the selenium export from red blood cells (RBCs), i.e., the transfer of selenium bound to Hb to RBC membrane proteins. To elucidate the molecular events of the Hb-associated selenium export from RBC, a Hb–Se complex was synthesized from thiol-exchange of Cys-β93 in Hb with penicillamine-substituted glutathione selenotrisulfide, as a model of major metabolic intermediates, and then interactions between the Hb–Se complex and RBC inside-out vesicles (IOVs) were examined. Selenium bound to Hb was transferred to the IOV membrane on the basis of the intrinsic interactions between Hb and the cytoplasmic domains of band 3 protein (CDB3). The observed selenium transfer was inhibited by the pretreatments of IOVs with iodoacetamide and the α-chymotrypsin digestion, indicating that the Hb mediates the selenium transfer to the thiol groups of CDB3. In addition, it was found that deoxygenated Hb, with a high binding affinity for CDB3, more favorably transferred selenium to the IOV membranes than oxygenated Hb, with a low affinity. When selenium export from RBC to the plasma was examined by continuously introducing nitrogen gas, the selenium export rate was promoted with an increase in the rate of deoxygenated Hb. Overall, these data suggested that Hb could possibly play a role in the selenium export from RBC treated with selenite in an oxygen-linked fashion. Summary The cell membrane K+-activated phosphatase activity was measured in reconstituted ghosts of human red cells having different ionic contents and incubated in solutions of varying ionic composition. When K+-free ghosts are suspended in K+-rich media, full activation of the phosphatase is obtained. Conversely, very little ouabainsensitive activity is detected in K+-rich ghosts suspended in K+-free media. These results, together with the fact that Na+ competitively inhibits the effects of K+ only when present externally, show that the K+ site of the membrane phosphatase is located at the outer surface of the cell membrane. The Mg++ requirements for K+ activation of the membrane phosphatase are fulfilled by internal Mg++. Addition of intracellular Na+ to ATP-containing ghosts raises the apparent affinity of the enzyme for K+, suggesting that the sites where ATP and Na+ produce this effect are located at the inner surface of the cell membrane. The applied method to measure the filtrability of red blood cells is accurate enough for the study of the effects of the conditions of storage on their deformability. In this work the standard deviation of the method, the mean biological deviation of the deformability of human red blood cells and their deformability in the course of storage in the ACD-AG-medium have been investigated. Considerable loss of deformability occurs towards the end of storage time of the cells, which is not reversible be restitution of ATP alone. The applicability of this method as a simple test of the vitatity of preserved erythrocytes if to be tested.  相似文献

It is shown that erythrocytes in packed red blood cell preparations can be maintained in native state for a long time using bubbling air ionization (BAI). The BAI procedure proposed to prolong the storage period of donor erythrocytes makes it possible to reduce the level of destructive processes in packed cells stored until use, as indicated by a decrease in the lipid peroxidation intensity and an increase in the antioxidant activity in them.  相似文献

The transfusion of red blood cells from umbilical cord blood (cord RBCs) is gathering significant interest for the treatment of fetal and neonatal anemia, due to its high content of fetal hemoglobin as well as numerous other potential benefits to fetuses and neonates. However, in order to establish a stable supply of cord RBCs for clinical use, a cryopreservation method must be developed. This, in turn, requires knowledge of the osmotic parameters of cord RBCs. Thus, the objective of this study was to characterize the osmotic parameters of cord RBCs: osmotically inactive fraction (b), hydraulic conductivity (Lp), permeability to cryoprotectant glycerol (Pglycerol), and corresponding Arrhenius activation energies (Ea). For Lp and Pglycerol determination, RBCs were analyzed using a stopped-flow system to monitor osmotically-induced RBC volume changes via intrinsic RBC hemoglobin fluorescence. Lp and Pglycerol were characterized at 4 °C, 20 °C, and 35 °C using Jacobs and Stewart equations with the Ea calculated from the Arrhenius plot. Results indicate that cord RBCs have a larger osmotically inactive fraction compared to adult RBCs. Hydraulic conductivity and osmotic permeability to glycerol of cord RBCs differed compared to those of adult RBCs with the differences dependent on experimental conditions, such as temperature and osmolality. Compared to adult RBCs, cord RBCs had a higher Ea for Lp and a lower Ea for Pglycerol. This information regarding osmotic parameters will be used in future work to develop a protocol for cryopreserving cord RBCs.  相似文献

Effective methods for long-term preservation of cord red blood cells (RBCs) are needed to ensure a readily available supply of RBCs to treat fetal and neonatal anemia. Cryopreservation is a potential long-term storage strategy for maintaining the quality of cord RBCs for the use in intrauterine and neonatal transfusion. However, during cryopreservation, cells are subjected to damaging osmotic stresses during cryoprotectant addition and removal and freezing and thawing that require knowledge of osmotic tolerance limits in order to optimize the preservation process. The objective of this study was to characterize the osmotic tolerance limits of cord RBCs in conditions relevant to cryopreservation, and compare the results to the osmotic tolerance limits of adult RBCs. Osmotic tolerance limits were determined by exposing RBCs to solutions of different concentrations to induce a range of osmotic volume changes. Three treatment groups of adult and cord RBCs were tested: (1) isotonic saline, (2) 40% w/v glycerol, and (3) frozen–thawed RBCs in 40% w/v glycerol. We show that cord RBCs are more sensitive to shrinkage and swelling than adult RBCs, indicating that osmotic tolerance limits should be considered when adding and removing cryoprotectants. In addition, freezing and thawing resulted in both cord and adult RBCs becoming more sensitive to post-thaw swelling requiring that glycerol removal procedures for both cell types ensure that cell volume excursions are maintained below 1.7 times the isotonic osmotically active volume to attain good post-wash cell recovery. Our results will help inform the development of optimized cryopreservation protocol for cord RBCs.  相似文献
