Summary Nitrogen fixation by strains of Azospirillum isolated from several rice soils and rice cultivars was investigated by15N2 incorporation and C2H2 reduction. C2H2 reducing ability markedly varied among the strains obtained from soils differing widely in their physico-chemical properties. Large variations in15N2 incorporation by Azospirillum isolated from the roots of several rice cultivars were also noticed. The present study reveals that rice cultivars harbour Azospirillum with differential N2-fixing ability and that plant genotype is of importance for optimal associations.  相似文献

Nitrogen (N) is often the most limiting nutrient in organic cropping systems. N2 fixing crops present an important option to improve N supply and to maintain soil fertility. In a field experiment, we investigated whether the lower N fertilization level and higher soil microbial activity in organic than conventional systems affected symbiotic N2 fixation by soybean (Glycine max, var. Maple Arrow) growing in 2004 in plots that were since 1978 under the following systems: bio-dynamic (DYN); bio-organic (ORG); conventional with organic and mineral fertilizers (CON); CON with exclusively mineral fertilizers (MIN); non-fertilized control (NON). We estimated the percentage of legume N derived from the atmosphere (%Ndfa) by the natural abundance (NA) method. For ORG and MIN we additionally applied the enriched 15N isotope dilution method (ID) based on residual mineral and organic 15N labeled fertilizers that were applied in 2003 in microplots installed in ORG and MIN plots. These different enrichment treatments resulted in equal %Ndfa values. The %Ndfa obtained by NA for ORG and MIN was confirmed by the ID method, with similar variation. However, as plant growth was restricted by the microplot frames the NA technique provided more accurate estimates of the quantities of symbiotically fixed N2 (Nfix). At maturity of soybean the %Ndfa ranged from 24 to 54%. It decreased in the order ORG > CON > DYN > NON > MIN, with significantly lowest value for MIN. Corresponding Nfix in above ground plant material ranged from 15 to 26 g N m-2, with a decreasing trend in the order DYN = ORG > CON > MIN > NON. For all treatments, the N withdrawal by harvested grains was greater than Nfix. This shows that at the low to medium %Ndfa, soybeans did not improve the N supply to any system but removed significant amounts of soil N. This study investigate the potential contribution of nitrogen fixation by indigenous cyanobacteria to rice production in the rice fields of Valencia (Spain). N2-fixing cyanobacteria abundance and N2 fixation decreased with increasing amounts of fertilizers. Grain yield increased with increasing amounts of fertilizers up to 70 kg N ha-1. No further increase was observed with 140 kg N ha-1. Soil N was the main source of N for rice, only 8–14% of the total N incorporated by plants derived from 15N fertilizer. Recovery of applied 15N-ammonium sulphate by the soil–plant system was lower than 50%. Losses were attributed to ammonia volatilization, since only 0.3–1% of applied N was lost by denitrification. Recovery of 15N from labeled cyanobacteria by the soil–plant system was higher than that from chemical fertilizers. Cyanobacterial N was available to rice plant even at the tillering stage, 20 days after N application. Summary Accurate estimates of N2 fixation by legumes are requisite to determine their net contribution of fixed N2 to the soil N pool. However, estimates of N2 fixation derived with the traditional15N methods of isotope dilution and AN value are costly.Field experiments utilizing15N-enriched (NH4)2SO4 were conducted to evaluate a modified difference method for determining N2 fixation by fababean, lentil, Alaska pea, Austrian winter pea, blue lupin and chickpea, and to quantify their net contribution of fixed N2 to the soil N pool. Spring wheat and non-nodulated chickpea, each fertilized with two N rates, were utilized as non-fixing controls.Estimates of N2 fixation based on the two control crops were similar. Increasing the N rate to the controls reduced AN values 32, 18 and 43% respectively in 1981, 1982 and 1983 resulting in greater N2 fixation estimates. Mean seasonal N2 fixation by fababean, lentil and Austrian winter pea was near 80 kg N ha–1, pea and blue lupin near 60 kg N ha–1, and chickpea less than 10 kg N ha–1. The net effects of the legume crops on the soil N pool ranged from a 70 kg N ha–1 input by lentil in 1982, to a removal of 48 kg N ha–1 by chickpea in 1983.Estimates of N2 fixation obtained by the proposed modified difference method approximate those derived by the isotope dilution technique, are determined with less cost, and are more reliable than the total plant N procedure.Scientific paper No. 6605. A pot experiment was conducted to study the effect of organic and inorganic nitrogen (N) sources on the yield and N uptake of rice from applied and native soil-N. The residual effect of these N sources on a succeeding wheat crop was also studied. Organic N was applied in the form of 15N-labelled Sesbania aculeata L., a legume, and inorganic N in the form of 15N-labelled ammonium sulphate. The two sources were applied to the soil separately or together at the time of transplanting rice. Recovery of N by rice from both the applied sources was quite low but both sources caused significant increases in biomass and N yield of rice. Maximum increase was recorded in soil treated with organic N. The residual value of the two materials as source of N for wheat was not significant; the wheat took up only a small fraction of the N initially applied. Loss of N occurred from both applied N sources, the losses being more from inorganic N. Both applied N sources caused a substantial increase in the availability of soil-N to rice and wheat; most of this increase was due to organic N and was attributed to the so-called ‘priming’ effect or ANI (added nitrogen interaction) of the applied material.  相似文献

Summary A two-year field study was undertaken using15N isotope techniques to differentiate between stimulation of N uptake and N2 fixation in Western Canadian cultivars of spring wheat (Triticum aestivum L. emend Thell) and durum (T. turgidum L. emend Bowden) in response to inoculation with N2-fixing bacteria. Bacterial inoculation either had no effect or lowered the % N derived from the fertilizer and the fertilizer use efficiency. Despite the depression of fertilizer uptake, inoculants did not alter the relative uptake from soil and fertilizer-N pools indicating that bacterial inoculation did not alter rooting patterns. Nitrogen-15 isotope dilution indicated that N2 fixation did occur. In 1984, % plant N derived from the atmosphere (% Ndfa) due to inoculation with Bacillus C-11-25 averaged 23.9% while that withAzospirillum brasilense ATCC 29729 (Cd) averaged 15.5%. In 1985, higher soil N levels reduced these values by approximately one-half. Cultivar x inoculant interactions, while significant, were not consistent across years. However, these interactions did not affect cultivars ‘Cadet’ and ‘Rescue’. In agreement with previous results, ‘Cadet’ performed well with all inoculants in both years while ‘Rescue’ performed poorly. Among 1984 treatments, the N increament in inoculated plants was positively correlated with % Ndfa but no such correlation existed in 1985. N2 fixation averaged over all cultivars and strains was 17.9 and 6.7 kg N fixed ha−1 in 1984 and 1985, respectively. Highest rates of N2 fixation were estimated at 52.4 kg N ha−1 for ‘Cadet’ in 1984 and 31.3 kg N ha−1 for ‘Owens’ in 1985, both inoculated with Bacillus C-11-25, an isolate from southern Alberta soils. Inoculation with either ofAzospirillum brasilense strain Cd (ATCC29729) or 245 did not result in as consistent or as high N2 fixation, suggesting that these wheats had not evolved genetic compatability with this exogenous microorganism. These agronomically significant amounts of N2 fixation occurred under optimally controlled experimental conditions in the field. It is yet to be determined if N2 fixation would occur in response to bacterial inoculation under dryland conditions commonly occurring in Western Canada. Summary Plants from agricultural and natural upland ecosystem were investigated for15N content to evaluate the role of symbiotic N2-fixation in the nitrogen nutrition of soybean. Increased yields and lower δ15N values of nodulating soybeansvs, non-nodulating isolines gave semi-quantitative estimates of N2 fixation. A fairly large discrepancy was found between estimations by δ15N and by N yield at 0 kg N/ha of fertilizer. More precise estimates were made by following changes in plant δ15N when fertilizer δ15N was varied near15N natural abundance level. Clearcut linear relationships between δ15N values of whole plants and of fertilizer were obtained at 30 kg N/ha of fertilizer for three kinds of soils. In experimental field plots, nodulating soybeans obtained 13±1% of their nitrogen from fertilizer, 66±8% from N2 fixation and 21±10% from soil nitrogen in Andosol brown soil; 30%, 16% and 54% in Andosol black soil; 7%, 77% and 16% in Alluvial soil, respectively. These values for N2 fixation coincided with each corresponding estimation by N yield method. Other results include: 1)15N content in upland soils and plants was variable, and may reflect differences in the mode of mineralization of soil organics, and 2) nitrogen isotopic discrimination during fertilizer uptake (δ15N of plant minus fertilizer) ranged from −2.2 to +4.9‰ at 0–30 kg N/ha of fertilizer, depending on soil type and plant species. The proposed method can accurately and relatively simply establish the importance of symbiotic nitrogen fixation for soybeans growing in agricultural settings.  相似文献

An F2 population, consisting of 231 individuals derived from a cross between rice cultivars with a similar growing duration, Palawan and IR42, was utilized to investigate the genetic nature of rice varietal ability to stimulate N2 fixation in the rice rhizosphere. To assess rhizospheric N2 fixation, an isotope-enriched 15N dilution technique was employed, using 15N-stabilized soil in pots. IR42, an indica variety, had 23% higher N derived from fixation (Ndfa) than Palawan, a javanica genotype. Normal segregation of atom% 15N excess was obtained in the F2 population, with an average of 0.218 with 8% of plants below IR42 (0.188) and 10% of plants above Palawan (0.248). One-hundred-and-four RFLP markers mapped on 12 chromosomes were tested for linkage to the putative QTLs. Significant (P<0.01) associations between markers and segregation of atom% 15N excess were observed for seven marker loci located on chromosomes 1, 3, 6 and 11. Four QTLs defined by the detected marker loci were identified by interval-mapping analysis. Additive gene action was found to be predominant, but for at least one locus, dominance and partial dominance effects were observed. Significant (P<0.01) epistatic effects were also identified. Individual marker loci detected between 8 and 16% of the total phenotypic variation. All four putative QTLs showed recessive gene action, and no phenotypic effects associated with heterozygosity of marker loci were observed. The results of this study suggest that rice genetic factors can be identified which affect levels of atom% 15N excess in the soil by interacting with diazotrophs in the rice rhizosphere.  相似文献

Effects of light on the short term competition for organic and inorganic nitrogen between maize and rhizosphere microorganisms were investigated using a mixture of amino acid, ammonium and nitrate under controlled conditions. The amount and forms of N added in the three treatments was identical, but only one of the three N forms was labeled with 15N. Glycine was additionally labeled with 14C to prove its uptake by maize and incorporation into microbial biomass in an intact form. Maize out-competed microorganisms for during the whole experiment under low and high light intensity. Microbial uptake of 15N and 14C was not directly influenced by the light intensity, but was indirectly related to the impact the light intensity had on the plant. More was recovered in microbial biomass than in plants in the initial 4 h under the two light intensities, although more 15N-glycine was incorporated into microbial biomass than in plants in the initial 4 h under low light intensity. Light had a significant effect on uptake by maize, but no significant effects on the uptake of or 15N-glycine. High light intensity significantly increased plant uptake of and glycine 14C. Based on 14C to 15N recovery ratios of plants, intact glycine contributed at least 13% to glycine-derived nitrogen 4 h after tracer additions, but it contributed only 0.5% to total nitrogen uptake. These findings suggest that light intensity alters the competitive relationship between maize roots and rhizosphere microorganisms and that C4 cereals such as maize are able to access small amounts of intact glycine. We conclude that roots were stronger competitor than microorganisms for inorganic N, but microorganisms out competed plants during a short period for organic N, which was mineralized into inorganic N within a few hours of application to the soil and was thereafter available for root uptake.  相似文献

Yields of above ground biomass and total N were determined in summer-grown maize and cowpea as sole crops or intercrops, with or without supplementary N fertilizer (25 kg N ha−1, urea) at an irrigated site in Waroona, Western Australia over the period 1982–1985. Good agreement was obtained between estimates of N2 fixation of sole or intercrop cowpea (1984/85 season) based on the15N natural abundance and15N fertilizer dilution techniques, both in the field and in a glasshouse pot study. Field-grown cowpea was estimated to have received 53–69% of its N supply from N2-fixation, with N2-fixation onlyslightly affected by intercropping or N fertilizer application. Proportional reliance on N2-fixation of cowpea in glasshouse culture was lower (36–66%) than in the field study and more affected by applied N. Budgets for N were drawn up for the field intercrops, based on above-ground seed yields, return of crop residues, inputs of fixed N and fertilizer N. No account was taken of possible losses of N through volatilization, denitrification and leaching or gains of N in the soil from root biomass. N2-fixation was estimated tobe 59 kg N ha−1 in the plots receiving no fertilizer N, and 73 kg N ha−1 in plots receiving 25 kg N ha−1 as urea. Comparable fixation by sole cowpea was higher (87 and 82 kg N ha−1 respectively) but this advantage was outweighed by greater land use efficiency by the intercrop than sole crops.  相似文献

To examine the effect of organic amendment application on the fate of inorganic-N accumulated in a vegetable field soil during conversion from inorganic to organic input, a pot experiment using 15N-labeled soil was conducted. The soil was labeled with 15N through addition of urea-15N (98 atom % 15N) and was then incubated for 1 year resulting in inorganic soil-N concentration and 15N abundance of 211 mg kg–1 soil and 4.950 atom %, respectively. Chinese cabbage [Brassica campestris (L.) Samjin] plants were grown in the labeled soil for 30 and 60 days after application of organic amendment at the rates of 0 (control), 200, 400, and 600 mg N kg–1 soil. Although organic amendment application did not show any significant effect on the uptake efficiency of inorganic-N by Chinese cabbage during the first 30 days, it significantly (P<0.05) increased inorganic-N uptake efficiency as well as total-N uptake and dry matter yield at the end of the 60-day growth period. Application of the organic amendment also increased microbial immobilization of inorganic-N in both growth periods. Between 30 and 60 days of growth, however, the amount of immobilized N from the inorganic-15N pool decreased, indicating re-mineralization of previously immobilized N. Although the amount of inorganic-15N lost was virtually the same among treatments at day 30, increased immobilization of inorganic-15N caused by organic amendment application led to the higher retention of inorganic-N in the soil and less loss of N at day 60 as compared to the control. These results indicate that increased immobilization by organic amendment application in the early growth season and the subsequent gradual re-mineralization may play an important role in increasing plant uptake of inorganic-15N, while minimizing N loss.  相似文献

Nitrogen (N) transfer from one species to another is important for the N cycling in low-input grassland. In the present work, estimates obtained by an indirect 15N dilution technique were compared with estimates obtained by a direct 15N leaf feeding technique over two complete growing seasons in red clover-ryegrass and white clover-ryegrass mixtures under field conditions. The direct technique confirmed that N transfer between clovers and ryegrass is a bi-directional process. The transfer of N from both clovers to ryegrass occurred within 25 days upon the first labelling event. A very high N transfer occurred from white clover to the associated ryegrass, 4.5 and 7.5 g m−2 in the 1st and 2nd production year, respectively. The corresponding values for transfer from red clover to the associated ryegrass were 1.7 and 3.6 g m−2. Quantified relatively to the total above-ground N content of white clover- ryegrass and red clover-ryegrass mixtures, the N transfer exceeded 50% and 10%, respectively, in three out of seven harvests. The N transfer from 15N labelled grass to associated clovers constituted a relatively constant proportion of approx. 8% of the above-ground N content of the mixtures. Estimates based on the soil 15N dilution technique generally underestimated the net N transfer by more than 50% compared to the direct 15N labelling technique. Furthermore, the indirect 15N dilution technique estimated only marginal differences between red and white clover in the quantities of N transferred, whereas the direct 15N labelling technique showed the N transfer from white clover to the associated ryegrass to be significantly higher than that involving red clover. It is concluded that N transfer is a much more dynamic and quantitatively important process in grassland than previously recognised. Summary The kinetic of15N2 diffusion has been measured in a system similar to that for the estimation of N2 fixation in plant microorganism associations cultivated in soil. The15N2 enrichment of the soil atmosphere reached an homogenous value one hour after injection of15N2 and is identical to that obtained by calculation, indicating that no adsorption occurs in the soil particles.

In monocropped cereal systems, annual N inputs from non-fertilizer sources may be more than 30 kg ha-1. We examined the possibility that these inputs are due to biological N2 fixation (BNF) associated with roots or decomposing residues. Wheat was grown under greenhouse conditions in pots (34 cm long by 10 cm diameter) containing soil from a plot cropped to spring wheat since 1911 without fertilization. The roots and soil were sealed from the atmosphere and exposed to a15N2-enriched atmosphere for three to four weeks during vegetative, reproductive or post-reproductive stages. This technique permitted detection of as little as 1 μg fixed N plant-1 in plant material and 40 μg fixed N plant-1 in soil. No fixation of15N2 occurred during either of the first two labelling periods. In the final labelling period, straw returned to the soil was significantly enriched in15N, especially in a pot with a higher soil moisture content. Total BNF in this pot was 13 μg N plant-1, or about 30 g N ha-1. In a separate experiment with soil from the same plot, we detected BNF only when soil was amended with glucose at a high soil moisture content. Two experiments have been conducted, one in semi-solid Hoagland nutrient medium and the other in shallow pots containing saline soil. N2-fixing bacteria belonging toAzospirillum, Azotobacter, Klebsiella andEnterobacter were inoculated separately on kallar grass grown in semi-solid nutrient medium. It was shown that inoculation affects root proliferation and also results in15N isotopic dilution. The % Ndfa ranged from 47–70 whereas no significant effect on the total nitrogen uptake was observed. The bacterial colonization of the root surface and the presence of enteric bacteria inside the root hair cells is reported. In a soil pot experiment, non-N2-fixingPolypogon monspeliensis was used as a reference plant (control). A treatment receiving a high rate of nitrogen was also used as a non-N2-fixing control.15N-labelled ammonium sulphate at 20 kg N ha–1 and 90 kg N ha–1 was used. The % Ndfa in the aerial parts of kallar grass was 12–15 whenP. monspeliensis was used as reference plant whereas 37–39% Ndfa was estimated when the treatment receiving high nitrogen fertilizer was used as a non-N2-fixing control. The effects of planted fallows of Sesbania sesban (L.) Merr. and Calliandra calothyrsus (Meissner) on soil inorganic nitrogen dynamics and two subsequent maize crops were evaluated under field conditions in the highlands of eastern Kenya. Continuous unfertilised maize, maize/bean rotation and natural regrowth of vegetation (weed fallow) were used as control treatments. The proportion of symbiotic N2-fixation was estimated by measuring both leaf 15N enrichment and whole-plant 15N enrichment by the 15N dilution technique for Sesbania and Calliandra, using Eucalyptus saligna (Sm.) and Grevillea robusta (A. Cunn) as reference species. Above- and below-ground biomass and N contents were examined in Sesbania, Calliandra, Eucalyptus and Grevillea 22 months after planting. Both the content of inorganic N in the topsoil and the quantity of N mineralised during rainy seasons were higher after the Sesbania fallows than after the other treatments. Compared to the continuous unfertilised maize treatment, both residual crop yields were significantly higher when mineral N (one application of 60 kg N ha–1) was added. Furthermore, the second crop following the Sesbania fallow was significantly higher than the continuous maize crop. The above-ground biomass of the trees at final harvest were 31.5, 24.5, 32.5 and 43.5 Mg ha–1 for the Sesbania, Calliandra, Grevillea and Eucalyptus, respectively. For the total below-ground biomass the values for these same tree species were 11.1, 15.5, 17.7, and 19.1 Mg ha–1, respectively, of which coarse roots (>2 mm), including tap roots, amounted to 70–90%. About 70–90% of the N in Sesbania, and 50–70% in Calliandra, was derived from N2-fixation. Estimates based on leaf 15N enrichment and whole-plant 15N enrichment were strongly correlated. The N added by N2-fixation amounted to 280–360 kg N ha–1 for Sesbania and 120–170 kg N ha–1 for Calliandra, resulting in a positive N balance after two maize cropping seasons of 170–250 kg N ha–1 and 90–140 kg N ha–1, for Sesbania and Calliandra, respectively. All the other treatments gave negative N balances after two cropping seasons. We conclude that Sesbania sesban is a tree species well suited for short duration fallows due to its fast growth, high nutrient content, high litter quality and its ability to fix large amounts of N2 from the atmosphere.  相似文献

A field experiment was carried out over 12 months to determine the effect of animal treading on N2 fixation in a mixed white clover-ryegrass pasture. The experimental site was defoliated by mowing for the duration of the study. A single treading event of moderate or severe pugging intensity was initiated in plots during wet spring conditions by using dairy cows at varying stocking rates (4.5 cows 100 m−2 for 1.5 or 2.5 h, respectively). Inputs of dung and urine onto the plots was avoided by overnight housing of the cows and interception of excreta during the pugging event. Soil air-filled porosity decreased from 21% in the non-pugged control to 15–16% in pugged treatments by day 3. Bulk density of soil was not significantly affected by pugging. Soil inorganic N concentration increased in pugged treatments, and was 4-fold greater on day 28 in severely pugged plots compared to non-pugged plots. White clover plant density and plant size was markedly lower in pugged treatments (up to 85% and 72% reduction, respectively under severe pugging). White clover growth was most affected during the first 156 days after pugging (up to 90% decrease under severe pugging), leading to an annual clover dry matter production loss of 9% and 52%, respectively. The proportion of clover N derived from atmospheric N2 (%Ndfa; estimated by 15N dilution) was initially reduced (to a lower limit of 43%) by severe pugging (days 28–71) before recovery to control levels (90%) by day 91. Annual N2 fixation in clover herbage decreased significantly from 76 kg N ha−1 yr−1 in the non-pugged control, to 66 and 36 kg N ha−1 yr−1 under moderate and severe pugging, respectively. Most of this difference was evident within the first 156 days after pugging. Our data indicates that the major loss in fixed N2 input under pugging was due to reduced clover growth and production resulting from pugging damage and loss of residual white clover biomass by hoof action.  相似文献

Summary Inoculation with root-nodule bacteria had favourable influence on N-uptake and yield of wheat. Since waterlogged root region of rice permits higher nitrogenase activity a pot culture experiment was conducted using same nine strains of rhizobia,Azotobacter chroococcum and bluegreen algae as inoculants.R. leguminosarum in combination with 50 kg N ha−1;R. japonicum and a strain of rhizobium isolated from moong bean increased the yield of paddy cv. Pusa-33. On the other hand an adverse effect of bacterial inoculation and of applied N was observed in case of Azotobacter, and rhizobia isolated from green gram, cicer, soyabean and clover. 土壤有机质与粮食安全和气候变化等重大问题密切相关,也是目前国内外研究热点。通过分析黄淮海平原10个小麦玉米长期定位试验资料,在不同施氮量下建立土壤有机质积累与碳投入的数学模型,探讨了土壤有机质积累与碳氮投入的关系。结果表明:土壤有机质积累与碳投入的关系受氮肥用量的影响显著,无论低量施氮(长期定位试验中的常规施肥量,化肥氮量≤330kg/(hm.2a))还是高量施氮(长期定位试验中的高量施肥量,化肥氮量330kg/(hm.2a)),秸秆根茬碳还田量与土壤有机质变化量之间均呈极显著正相关关系(P0.01);利用土壤有机质积累与碳投入的数学模型计算的维持土壤有机质平衡的碳投入量(MSC),高量施氮时为7254kg/(hm.2a),低量施氮为1297kg/(hm.2a),前者是后者的5.6倍。因此,大量施用氮肥降低了投入碳的积累,不利于土壤有机质的提升。  相似文献
