Reverse-phase high-performance liquid chromatography was used to analyse 14C-labelled metabolites of indole-3-acetic acid (IAA) formed in the cortical and stelar tissues of Zea mays roots. After a 2-h incubation in [14C]IAA, stelar segments had metabolised between 1–6% of the methanol-extractable radioactivity compared with 91–92% by the cortical segments. The pattern of metabolites produced by cortical segments was similar to that produced by intact segments bathed in aqueous solutions of [14C]IAA. In contrast, when IAA was supplied in agar blocks to stelar tissue protruding from the basal ends of segments, negligible metabolism was evident. On the basis of its retention characteristics both before and after methylation, the major metabolite of [14C]IAA in Zea mays root segments was tentatively identified by high-performance liquid chromatography as oxindole-3-acetic acid.Abbreviations HPLC High-performance liquid chromatography - IAA Indole-3-acetic acid  相似文献

A concentration of 10–5 M tomatine had no effect on leakagefrom, or elongation of, wheat coleoptile segments, but consistentlyreduced IAA-enhanced extension growth by c. 50 per cent. Therewas no evidence of chemical interaction between the alkaloidand the auxin in solution, and IAA action was not affected bypre-treatment for up to 3 h with 10–5 M tomatine. Studieswith [2-14C]IAA revealed that 10–5 M tomatine did notinhibit uptake of auxin into segments. The effect of pre-treatingsegments for up to 3 h with IAA could be virtually nullifiedby 10–5 M tomatine, as could also IAA-induced changesin properties of coleoptile cell walls. Results are discussedin relation to the ability of tomatine to disrupt membrane functionand to current hypotheses implicating membranes in the primaryaction of auxin.  相似文献

The effects of the morphactin 2-ehloro-9-hydroxyfluorene-9-carboxylicacid methyl ester [CFM] on growth, geotropic curvature and transportand metabolism of indol-3yl-acetic acid [IAA-5-3H] in the coleoptilesof Zea mays and A vena saliva have been investigated. A strongcorrelation has been found to exist between the inhibition ofthe geotropic response and the inhibition of auxin transport.CFM supplied at concentrations sufficient to abolish auxin transporthas been shown to promote the elongation of Zea, but not ofAvena, coleoptile segments. CFM does not change the patternof metabolism of IAA in Zea coleoptile segments. In these segmentsIAA is metabolized when its concentration is high, but the radioactivitytransported basipetally, or laterally in geotropically stimulatedcoleoptiles, is virtually confined to the IAA molecule. Radioactivityexported into the basal receiver blocks is wholly confined toIAA. It is concluded that CFM inhibits the geotropic responsein coleoptiles by suppression of the longitudinal and lateralauxin transport mechanisms. The growth-promoting propertiesof this substance cannot be linked with its effects on eitherauxin metabolism or transport.  相似文献

When indole-3-acetic acid (IAA) is applied to the cotyledonsof broad bean seedlings (Vicia faba L. cv Chukyo), the majormetabolites found in the roots are 3-(O-ß-glucosyl)-2-indoIone-3-acetylaspartic acid (Glc-DIA-Asp) and 3-hydroxy-2-indolone-3-acetylasparticacid (DIA-Asp). In this report, the metabolic pathway from IAAto the two dioxindole-3-acetic acid (DIA) conjugates was investigatedby using [14C]IAA, [14C]DIA, [14C]indole-3-acetylaspartic acid(IAA-Asp), and [14C]IAA-[3H]Asp. The precursor of DIA-Asp wasfound to be IAA-Asp but not DIA. Incorporation of the doublelabeled IAA-Asp into the DIA conjugates demonstrated that hydrolysisof IAA-Asp was not involved in the formation of the DIA conjugates.DIA-Asp was further metabolized to Glc-DIA-Asp in the cotyledons,while formation of Glc-DIA-Asp in the roots was very low. Pretreatment of 2?0 mm segments of etiolated zucchini (Cucurbitapepo L.) hypocotyl with cycloheximide (CH) or 2-(4-methyl-2,6-dinitroanilino)-N-methylpropionamide(MDMP) eliminated the stimulation by N-1-naphthylphthalamicacid (NPA) of net uptake of [1-14C]indol-3yl-acetic acid ([1-14C]IAA),but had relatively little effect on the net uptake of IAA inthe absence of NPA. The efflux of [1-14C]IAA from preloadedsegments was not substantially affected by inhibitor pretreatmentin the absence of NPA, but CH pretreatment significantly inhibitedthe reduction of efflux caused by NPA. Pretreatment with CHor MDMP did not affect net uptake by segments of the pH probe[2-14C]5,5-dimethyl-oxazolidine-2,4-dione ([2-14C]DMO), or thenet uptake of [14C]-labelled 3-O-methylglucose ([14C]3-0-MeGlu),suggesting that neither inhibitor affected intracellular pHor the general function of proton symporters in the plasma membrane.Both compounds reduced the incorporation of label from [35S]methionineinto trichloroacetic acid (TCA)-insoluble fractions of zucchinitissue, confirming their inhibitory effect on protein synthesis. The steady-state association of [3H]IAA with microsomal vesiclesprepared from zucchini hypocotyl tissue was enhanced by theinclusion of NPA in the uptake medium. The stimulation by NPAof [3H]IAA association with microsomes was substantially reducedwhen the tissue was pretreated with CH. However, CH pretreatmentdid not affect the level of high affinity NPA binding to themembranes indicating that treatments did not result in lossof NPA receptors. It is suggested that the auxin transport site on the effluxcarrier system and the receptor site for NPA may reside on separateproteins linked by a third, rapidly turned-over, transducingprotein. The distribution of exogenously-supplied radioactive labelledindol-3-yl-acetic acid (IAA) and gibberellin A1 (GA1) in geotropicallystimulated roots of Norway spruce (Picea abies (L.) Karst.)has been demonstrated. Seedlings were positioned with theirroot tips in 2.1 x 10–6 M [14C]IAA or 1.3 x 10–8m 3H-GA1 for 4 and 20 h, respectively. After geotropic stimulationfor 90 min in the horizontal position the root tips were cutlongitudinally in 50 µm thick sections, using a freeze-microtome.The radioactivity in the 14C-IAA treated roots occurred in higherconcentration in the lower than in the upper halves (ratio 1.25:1). A similar trend was observed in the [3H]GA1-treated rootswhere the ratio lower: upper halves was 2.04: 1. The ratio ofradioactivity in right and left halves of vertical roots wasapproximately the same in roots supplied with [14C]IAA and [3H]GA1(1.09: 1). The supplied radioactive compounds were analysed chromatographicallyafter extraction in methanol of 6 mm apical root segments. Onlya small fraction (7–8 per cent) of the supplied [14C]IAAwas revealed unchanged in the segments. The major part of thechromatographed, labelled compound has not been identified,but on basis of its RF value it is suggested that it may beindol-3-acetyl-aspartic acid (IAAasp). The chromatographic analysis of the [3H]GA,-treated segmentsshowed that only small fractions of this gibberellin has beenconverted to other compounds. These results have been discussed and correlated with knowledgeof plant growth regulators and their participation in root geotropism. Either 5-[3H]indole-3-acetic acid (IAA) or 5-[3H]indole-3-acetyl-myo-inositol was applied to the endosperm of kernels of dark-grown Zea mays seedlings. The distribution of total radioactivity, radiolabeled indole-3-acetic acid, and radiolabeled ester conjugated indole-3-acetic acid, in the shoots was then determined. Differences were found in the distribution and chemical form of the radiolabeled indole-3-acetic acid in the shoot depending upon whether 5-[3H]indole-3-acetic acid or 5-[3H]indole-3-acetyl-myo-inositol was applied to the endosperm. We demonstrated that indole-3-acetyl-myo-inositol applied to the endosperm provides both free and ester conjugated indole-3-acetic acid to the mesocotyl and coleoptile. Free indole-3-acetic acid applied to the endosperm supplies some of the indole-3-acetic acid in the mesocotyl but essentially no indole-3-acetic acid to the coleoptile or primary leaves. It is concluded that free IAA from the endosperm is not a source of IAA for the coleoptile. Neither radioactive indole-3-acetyl-myo-inositol nor IAA accumulates in the tip of the coleoptile or the mesocotyl node and thus these studies do not explain how the coleoptile tip controls the amount of IAA in the shoot.  相似文献

The transport of [2-14C]IAA and its derived products has beeninvestigated in segments of the leaf sheath base of Echinochloacolonum. Segments orientated vertically in the normal uprightposition transport 14C basipetally at rates approximately 10times higher that acropetally. In horizontal segments the basipetalrates are lower, being only 1–4 times higher than acropetalrates. Segments orientated vertically in the inverted positiontransport 14C basipetally at approximately 1/10 of the acropetalrate. 14C in the receiver discs from vertically orientated segmentswas found to be present predominantly as [2-14C]IAA and itsderived metabolite. The levels of each of these compounds from upright and invertedvertical segments followed the same pattern as that of theirtotal 14C content.  相似文献

Elongation of coleoptile segments, having or not having a tip,excised from rice (Oryza sativa L. cv. Sasanishiki) seedlingswas promoted by exogenous ethylene above 0.3 µl l–1as well as by IAA above 0.1 µM. Ethylene production ofdecapitated segments was stimulated by IAA above 1.0µM,and this was strongly inhibited by 1.0 µM AVG. AVG inhibitedthe IAA-stimulated elongation of the decapitated segment witha 4 h lag period, and this was completely recovered by ethyleneapplied at the concentration of 0.03 µl l–1, whichhad no effect on elongation without exogenous IAA. The effectsof IAA and ethylene on elongation were additive. These factsshow that ethylene produced in response to IAA promotes ricecoleoptile elongation in concert with IAA, probably by prolongingthe possible duration of the IAA-stimulated elongation, butthat they act independently of each other. Moreover, AVG stronglyinhibited the endogenous growth of coleoptile segments withtips and this effect was nullified by the exogenous applicationof 0.03 µl l–1 ethylene. These data imply that theelongation of intact rice coleoptiles may be regulated cooperativelyby endogenous ethylene and auxin in the same manner as foundin the IAA-stimulated elongation of the decapitated coleoptilesegments. The movement and metabolism of kinetin-8-14C in the mesocotyland coleoptile of Zea mays have been investigated. The segments (10 mm) of coleoptile with and without an apexwere used. The presence of apex decreases the quantity of countswhich move basipetally in the segments. The quantity transported acropetally in the segments (apex incontact with receiver blocks) was about three times higher thanthat transported basipetally (apex in contact with donor blocks). Using 10 mm coleoptile segments excised I mm below the apex,there was no appreciable difference between acropetal and basipetalmovement. The transport of kinetin in the mesocotyl was studied with andwithout vascular tissue. The quantity transported in segmentswithout vascular tissue was always lower than in the segmentswith vascular tissue. The presence of the node between the coleoptile and mesocotyldoes not disturb the movement of kinetin in the basipetal position(node in contact with donor blocks). Thin layer chromatography of plant extracts after a 48-hourtransport period revealed several radioactive substances whichappeared to be adenine with Rf 0.5 and other components appearedat Rf 0.6.  相似文献

Greater concentrations of auxin at nodes than in internodes,resulting from some nodal barrier to basipetal transport, havelong been postulated as the cause of early differentiation ofinitially isolated xylem and cambium at the nodes. However,this study, using [14C] indole-3-acetic acid (IAA) applied apicallyand gas chromatography-mass spectrometry, found that in stemsofImpatiens sultanithe IAA concentrations (per unit f. wt) atnodes were similar to those in adjacent internodes, though alittle greater at nodes if expressed per unit length of stemand a little less per unit d. wt. By contrast, in decapitatedshoots and in stem explants of dicotyledons, loss of the apicalsource of basipetally flowing auxin can result in auxin drainagewith some auxin retention in the uppermost remaining nodes.When [14C]IAA was applied apically to shoots for 4 h and stemexplants were excised, the explants had no nodal accumulationinitially whereas comparable explants incubated for 20 h revealedsignificant nodal accumulation. If decapitation leads both tonodal auxin accumulation and to adventitious abscission justabove the node, this fits the hypothesis that abscission sitesare positioned where auxin concentration decreases locally inthe apical direction. Coleoptile tips (about 2.5 mm in length) were excised from 3-day-olddark-adapted maize (Zea mays L.) seedlings and incubated indarkness in potassium phosphate buffer that contained 14C-L-tryptophan(Trp). Subsequent analysis by gas chromatography-mass spectrometryindicated that a significant portion of endogenous indole-3-aceticacid (IAA) had been labeled with 14C. About 8% of the IAA thatdiffused from the tissue into the medium during incubation from0.5 to 2 h was labeled, and 12% of the IAA extracted from thetissue after a 2-h incubation was labeled. On the other hand,30% of the Trp extracted from the tissue after a 2-h incubationwas 14C-Trp, which was more than those determined for IAA. Sincethe experiments were carried out under the non-steady-stateconditions in which the tissue content of 14C-Trp increasedwith time, and since the extracted Trp included the 14C-Trpin the apoplastic space, it seemed that synthesis de novo fromTrp was the major means by which IAA was produced in the coleoptiletip. The conversion of Trp to IAA was not detected in sub-apicalsegments (5–7.5 mm from the top) that were incubated similarly,an indication that synthesis of IAA occurs specifically in thetip region. When intact seedlings were irradiated with a pulseof red light 2 h before excision of tips and the applicationof 14C-Trp, the amounts of extractable and diffusible IAA werereduced by 40–60% without a change in the rate of 14Cincorporation. The inhibitory mode of action of jasmonic acid (JA) on the growth of etiolated oat (Avena sativa L. cv. Victory) coleoptile segments was studied in relation to the synthesis of cell wall polysaccharides using [14C]glucose. Exogenously applied JA significantly inhibited indoleacetic acid (IAA)-induced elongation of oat coleoptile segments and prevented the increase of the total amounts of cell wall polysaccharides in both the noncellulosic and cellulosic fractions during coleoptile growth. JA had no effect on neutral sugar compositions of hemicellulosic polysaccharides but substantially inhibited the IAA-stimulated incorporation of [14C]glucose into noncellulosic and cellulosic polysaccharides. JA-induced inhibition of growth was completely prevented by pretreating segments with 30 mm sucrose for 4 h before the addition of IAA. The endogenous levels of UDP-sugars, which are key intermediates for the synthesis of cell wall polysaccharides, were not reduced significantly by JA. Externally supplied [3H]myo-inositol and [14C]glucose were incorporatedin cell-wall fractions of segments of etiolated squash hypocotyl.The extent of incorporation of [14C]glucose into cell-wall fractionswas very much greater than that of [3H]myo-inositol. Radioactivityfrom [14C]-glucose was effectively incorporated into hemicelluloseB and cellulose fractions and was incorporated uniformly intohexose, pentose and uronic acid residues, but radioactivityfrom [3H]myo-inositol was incorporated predominantly into uronicacid and pentose residues in the pectin and hemicellulose Bfractions. Exogenously applied ABA significantly suppressed the elongationof segments of squash hypocotyl and the incorporation of radioactivityfrom [l4C]glucose and [3H]myo-inositol into the segments. Furthermore,ABA significantly inhibited the distribution of incorporatedradioactivity from [14C]glucose into the cellulose fraction,but did not affect distribution into the pectic fraction. Bycontrast, ABA only slightly inhibited the distribution of theincorporated radioactivity from [3H]myo-inositol into the pecticfraction. These results suggest that most of the cell-wall polysaccharidesin segments of squash hypocotyl are synthesized via the UDP-sugarpathway, and that ABA significantly inhibits the synthesis ofcellulose but not the synthesis of pectic polysaccharides whenABA suppresses the elongation of the segments. Cd accumulation, its effects on elongation growth of maize coleoptile segments, pH changes of their incubation medium and the membrane potential of parenchymal cells were studied. The Cd content increased significantly with exposure to increasing cadmium concentrations. Coleoptile segments accumulated the metal more efficiently in the range 10–100 μM Cd, than in the range 100–1000 μM Cd. Cd at concentrations higher than 1.0 μM produced a significant inhibition of both growth and proton extrusion. 100 μM Cd caused depolarization of the plasma membrane (PM) potential in parenchymal cells. The simultaneous treatment of maize coleoptile segments by indole-3-acetic acid (IAA) and Cd, counteracted the toxic effect of Cd on growth. Moreover, our data also showed that 100 μM Cd suppressed the characteristic IAA-induced hyperpolarization of the membrane potential, causing membrane depolarization. These results indicate that the toxic effect of Cd on growth of maize coleoptile segments might be, at least in part, caused via reduced PM H+-ATPase activity.  相似文献

Decapitated segments from maize (Zea mays L.) coleoptiles orientedvertically in an upright position show a strong spontaneousgrowth response (SGR) 3 h after decapitation. The latent periodof the SGR is markedly reduced when these segments are orientedin an inverted position. Coleoptile segments with intact tipsexhibit a weak and transient SGR in the vertical upright orientation.However, in the inverted orientation, these segments show atypical SGR. The data are inconsistent with the current hypothesisthat the SGR is caused by a time-dependent increase in tissuesensitivity to auxin. The parallel increase in membrane potentialdifference and growth rate during the time-course of the SGRindicates a possible role for PM H+-ATPase in the establishmentof the SGR in maize coleoptile segments. Attempts were made to disrupt the plasmodesmata between oatcoleoptile cells (Avena saliva L. cv. Victory) by severe plasmolysis.Coleoptiles, allowed to regain turgor after plasmolysis, wereable to execute geotropic and phototropic curvatures and segmentswould grow in response to applied auxin. In coleoptiles similarlytreated, studies with [14C]IAA have shown that longitudinal,basipetal transport of auxin still takes place and, as in controls,IAA is preferentially redistributed laterally within coleoptilesorientated horizontally. Physical continuity of the symplast of oat coleoptile cellsmay not always be disrupted by severe plasmolysis. Nevertheless,functional continuity appears to be interrupted. Despite this,all the processes involved in the execution of tropistic curvaturesremain intact, including transport of hormones. Plasmodesmatalcontinuity between oat coleoptile cells appears not to be anecessary requirement for auxin transport.  相似文献

If segments of Impatiens sultani stem are explanted and incubated,separation layers often form across them and lead to abscission.To test the suggested role of auxin concentration in controllingthe position of abscission sites, explants were labelled byapplying [14C]IAA to the shoot tip 4 h prior to explanting;transport of auxin applied in this way seems to resemble thatof endogenous auxin. During subsequent incubation of explantsfor 20 h, basipetal transport resulted in 14C accumulating justabove the base of the explants (nearly 80 % in the bottom 4mm of 24 mm explants). In internodal explants that had beenwounded at explanting by incising one side so as to sever avascular bundle, and in nodal explants with the leaf removed,the 14C also accumulated just above the wound or node to abouttwice the concentration otherwise expected; this accumulationwas probably due to basipetal transport being impeded by vasculardiscontinuity at the wound or node. Accumulation just abovethe base, or above a wound or node, resulted in gradients of14C concentration (presumably reflecting endogenous auxin concentration)decreasing in the morphologically upward direction at each ofthese three positions where abscission sites tend to occur. Auxin transport was investigated in excised stem segments ofNicotiana tabacum L. by the agar block technique using [1-14C]indol-3yl-acetic acid (IAA). The ability of the stems to transportauxin basipetally increased as secondary development proceeded;by contrast the ability of the pith to transport auxin declinedwith age. By separation of the stem tissues it was shown thatthe great majority of auxin transport took place in cells associatedwith the internal phloem and in cells close to the cambium;in both cases similar velocities of transport were found (c.5.0 mm h–1 at 22°C). The effects of osmotic gradientson auxin transport through the internal phloem were investigated.IAA was found by chromatography to account for practically allthe radioactivity in receiver blocks and other extracts of stemsegments. The relationship between coleoptile elongation and alcoholicfermentation of rice under anoxia is examined using seeds either:(a) N2 flushed during submergence, (b) incubated in stagnantdeoxygenated agar at 0·1% w/v to simulate the stagnantconditions of waterlogged soil, or (c) incubated in waterloggedsoil. Coleoptile elongation growth was greater for N2 flushing> stagnant agar > soil; seed survival was also greatestin this order over 1-5 d. Ethanol concentrations in coleoptiles and intact seeds (cv.IR42) were approximately 300 and 100 mol m-3 respectively whenseeds were grown 3 d in stagnant agar, however 92% of the ethanolin seeds diffused into the external medium when solutions weremixed for 5-10 s. Coleoptile growth under anoxia was relatedto rates of ethanol synthesis (RE) in different treatments;there was greater coleoptile growth and RE for seeds in N2 flushedsolutions than in stagnant deoxygenated agar. Coleoptile growthof individual seeds was also related to the RE of each seedat 2-3 d after anoxia (r2 = 0·46). Analysis of different tissues was important in evaluating growthand metabolism of coleoptiles. Although the coleoptile onlyaccounted for 0·7% of seed dry weight at 3 d after anoxia,it contained 21% of the ethanol produced by rice seeds. Therewere also three-fold higher rates of RE on a fresh weight basisin expanding tissues in the base of the coleoptile relativeto the elongated tissues at the apex.
