Surface wettability and microtopography can either enhance or deter larval settlement of many sessile marine organisms. This study quantifies the effect of these surface properties on the settlement of pediveligers of Mytilus galloprovincialis, using polymers spanning a range of wettability and microtextured polydimethylsiloxane (PDMS). Furthermore, the adhesion strength of settled pediveligers on microtextured PDMS surfaces was quantified using a flow chamber. Settlement was enhanced at the hydrophilic end of the wettability spectrum, where mean settlement on nylon reached 33.5 ± 13.1%. In contrast, mean settlement on the most hydrophobic polymer (PDMS) was 4.2 ± 3.2%. Microtopography had a much stronger effect compared to wettability, where 400 μm textured PDMS enhanced settlement above 90%. Settlement preferences were also positively correlated to adhesion strength at flow rates of 4 knots, with all initially settled pediveligers on smooth PDMS detaching, while 79.9 ± 5.7% of pediveligers remained on the 400 μm texture.  相似文献

Fouling-release (FR) coatings minimise the adhesion strength of fouling organisms. This study describes improved technologies to control the settlement and adhesion of the important fouling organism Mytilus galloprovincialis by incorporating the nanofillers titanium dioxide (TiO2) and carbon nanotubes (CNTs) in polydimethylsiloxane (PDMS) matrices. The incorporation of TiO2 prevented larval settlement when photoactivated with UV light, even at the lowest concentration of the nanofiller (3.75 wt%). Notably, there was 100% mortality of pediveligers exposed to photoactivated TiO2. However, plantigrades initially settled to photoactivated TiO2, but their adhesion strength was significantly reduced on these surfaces in comparison to blank PDMS. In addition, plantigrades had high mortality after 6 h. In contrast to the enhanced antifouling and FR properties of PDMS incorporating TiO2, the incorporation of CNTs had no effect on the settlement and adhesion of M. galloprovincialis.  相似文献

Settlement increased in drop assays in a 12 h light:12 h dark cycle, while bottom shade had no effect of any magnitude. In addition, settlement was significantly enhanced by storing pediveligers for between 4 and 24 days at 4 °C. Overall, these data provide the template to optimise and standardise static laboratory settlement assays for mussels in order to develop materials that either enhance settlement for the aquaculture industry, or deter settlement for antifouling applications. Straight-hinge veliger and pediveliger larvae of the mussel Mytilus galloprovincialis were refrigerated for varying periods for use in bioassays. Straight-hinge veliger larvae grew to the umbo-veliger stage after 2 months in the refrigerator, but no pediveligers were observed during the 3-month refrigeration period. The average survival rate of larvae in the refrigerator was 79% after 1 month, but gradually decreased with the refrigeration period, and was as low as 22% after 3 months. All refrigerated larvae grew to the pediveliger stage in the incubator at 17 degrees C at the same rate as that of the control larvae that were not refrigerated. Settlement and metamorphosis of pediveligers from both refrigerated and control groups were facilitated by microbial film and epinephrine and inhibited by phentolamine. Thus, refrigeration can be used as an effective method of storing larvae of M. galloprovincialis for use in assays to assess candidate settlement inducers and antifouling substances.  相似文献

In sessile marine invertebrates, larval settlement is fundamental to population maintenance and persistence. Cues contributing to the settlement choices and metamorphosis of larvae have important implications for the success of individuals and populations, but cues mediating larval settlement for many marine invertebrates are largely unknown. This study assessed larval settlement in two common Great Barrier Reef sponges, Coscinoderma matthewsi and Rhopaloeides odorabile, to cues that enhance settlement and metamorphosis in various species of scleractinian coral larvae. Methanol extracts of the crustose coralline algae (CCA), Porolithon onkodes, corresponding to a range of concentrations, were used to determine the settlement responses of sponge larvae. Cnidarian neuropeptides (GLW-amide neuropeptides) were also tested as a settlement cue. Settlement in both sponge species was approximately two-fold higher in response to live chips of CCA and optimum concentrations of CCA extract compared to 0.2 μm filtered sea water controls. Metamorphosis also increased when larvae were exposed to GLW-amide neuropeptides; R. odorabile mean metamorphosis reached 42.0±5.8% compared to 16.0±2.4% in seawater controls and in C. matthewsi mean metamorphosis reached 68.3±5.4% compared to 36.7±3.3% in seawater controls. These results demonstrate the contributing role chemosensory communication plays in the ability of sponge larvae to identify suitable habitat for successful recruitment. It also raises the possibility that larvae from distinct phyla may share signal transduction pathways involved in metamorphosis.  相似文献

The colonial ascidian Didemnum candidum (Savigny) is more abundant at shallow depths on floating docks than at greater depths along pilings in Pearl Harbor, Oahu, Hawaii. To compare the effects of selective settlement and postsettlement mortality on adult distribution, I determined if settlement was nonrandom relative to depth, if differences in adult abundance were responsible for the depth distribution of settlement, and if juvenile mortality varied with depth. A plankton pump was used to measure variation in larval abundance with depth. Acrylic settling plates were suspended at different depths and sampled nondestructively to measure settlement intensity and juvenile mortality. Settlement and mortality of the didemnid ascidians Diplosoma listerianum Milne-Edwards and Diplosoma sp. were also measured in the field and compared to that of D. candidum. Settlement of D. candidum was indeed nonrandom over depth. Both planktonic larvae and settled juvenils were more numerous at 0.5 than 3 or m. Settlement intensity on vertical plates was greatest within 1–2 cm of the water surface in the laboratory and within 2–3 cm in the field. Directly adjacent to the floating dock, where adults were equally abundant, settlement on horizontal plates was greater at shallow depths, suggesting directed movement of larvae upward. However, the ratio of settled juveniles to planktonic larvae (number of settlers: number of larvae) did not significantly differ with depth, suggesting that larvae were not more likely to settle at a particular depth. Settlement of the Diplosoma species was also heaviest near the surface. Juvenile mortality was greater at 0.5 than at 3 or 6 m for both D. candidum and the Diplosoma species. Experimental settlement showed that mortality, per se, of D. candidum was independent of depth. Rather, mortality was density-dependent, and the higher mortality near the surface was due to the greater number of larvae settling there. At this location, nonrandom settlement appears to determine the adult distribution of D. candidum, despite greater juvenile mortality at shallow depths. The pattern of settlement over depth is largely determined by adult proximity, rather than active larval behavior.  相似文献

Early post-settlement mortality is difficult to measure in mobile benthic taxa such as mussels. Field studies typically employ settlement collectors that often contain empty shells of dead individuals, which are usually disregarded. Here, empty shells of pediveligers, i.e. recent settlers that died before beginning to grow as juveniles, were used to assess temporal and spatial patterns of early mortality in mussels, whether size at settlement determined mortality, and whether mortality was density-dependent. This study took place at two intertidal sites in a large harbour in central New Zealand where monthly cohorts of mussels were collected over a 2-year period. Monthly mortality varied substantially, ranging from 0% to 42.6%, but not across the two sites that were 27?km apart. Although there was no density-dependent mortality evident within cohorts, the proportion of pediveligers which were dead on collection was highly positively correlated across sites. Live settlers were on average larger than dead settlers, and the size distribution of dead settlers was shifted toward smaller sizes compared to live settlers. At the extremes, of all settlers with shell length of 260?µm or less, 72% were dead on collection. By contrast, for settlers 310?µm or greater in length, 6% were dead. Together, these results (1) suggest that in this system size at settlement may be an important determinant of very early, peri-metamorphic mortality in mussels, with smaller settlers at higher risk, and (2) demonstrate the value in evaluating dead pediveligers for assessing natural early post-settlement mortality in mussels.  相似文献

Understanding of factors influencing settlement(attachment and metamorphosis) of marine invertebratelarvae is of great importance in aquaculture andcontrol of biofouling. The influence of two factors onsettlement of larvae was assessed from two separateinvestigations: 1, the influence of age (endogenousfactor) on cyprids of the barnacle Balanusamphitrite; and 2, the influence of a microbial film(exogenous factor) on pediveligers of the mussel Mytilus galloprovincialis.The settlement response of cypris larvae of B.amphitrite was found to be age-dependent. Oldercyprids responded more readily to settlement factorsthan newly molted ones. In M.galloprovincialis, competent pediveligers settled inresponse to a microbial filmed surface but not toan unfilmed surface. Moreover, a factor with MW of lessthan 5000 dalton, derived from culture medium of abacterial strain C1.1 (Pseudomonas-Alteromonasgroup), induced the settlement of M. galloprovincialis larvae.Thus, marine invertebrate larvae may require a periodof competence acquisition, during which they arepoorly responsive to settlement inducers. To understand the role of surface wettability in adhesion of cells, the attachment of two different marine algae was studied on hydrophobic and hydrophilic polymer surfaces. Adhesion of cells of the diatom Navicula and sporelings (young plants) of the green macroalga Ulva to an underwater surface is mainly by interactions between the surface and the adhesive exopolymers, which the cells secrete upon settlement and during subsequent colonization and growth. Two types of block copolymers, one with poly(ethylene glycol) side-chains and the other with liquid crystalline, fluorinated side-chains, were used to prepare the hydrophilic and hydrophobic surfaces, respectively. The formation of a liquid crystalline smectic phase in the latter inhibited molecular reorganization at the surface, which is generally an issue when a highly hydrophobic surface is in contact with water. The adhesion strength was assessed by the fraction of settled cells (Navicula) or biomass (Ulva) that detached from the surface in a water flow channel with a wall shear stress of 53 Pa. The two species exhibited opposite adhesion behavior on the same sets of surfaces. While Navicula cells released more easily from hydrophilic surfaces, Ulva sporelings showed higher removal from hydrophobic surfaces. This highlights the importance of differences in cell-surface interactions in determining the strength of adhesion of cells to substrates.  相似文献

Abstract. Settlement timing and differential settlement for the larval stage of the green sea urchin, Strongylocentrotus droebachiensis , in the southern Gulf of Maine was studied during the summer of 1996. Settlement densities on astroturf panels were highest in June and early July (13 to 37 m −2 d−1), and peaked in mid-June (199 m−2 d−1). Settlement was low to nonexistent from mid-July through August (0 to 2 m−2 d−1). During the peak in settlement, no selection for substrate type was observed. In the remainder of the settlement period, differential settlement occurred, with a preference for substrate covered with live coralline algae. Test diameter of newly settled urchins varied among the substrates, with urchins settling on live coralline algae having the largest test diameter (0.43 ± 0.01 mm). There were no differences in test diameter among the different weeks in which sampling was done. Sustained onshore winds occurred only during peak settlement, suggesting that wind drift currents may concentrate larvae and influence patterns of larval settlement.  相似文献

Enteromorpha , the most important macroalga that fouls ships, produces very large numbers of swimming spores that respond to a number of settlement cues. Responses to topographic cues have been investigated using surfaces with defined microtopographies fabricated from polydimethyl siloxane elastomer (PDMS). The topographic features were based on two designs, (a) a series of 5 or 1.5 w m deep valleys with valley floors and ridges varying between 5 and 20 w m, and (b) pillars of 5 w m diameter and 5 or 1.5 w m height, spaced 5-20 w m apart. The features were arranged in blocks to provide the swimming spores with a 'choice' of where to settle. Swimming spores settled preferentially in the valleys and against the pillars. The number of spores that settled increased very substantially as the width of the valley decreased. The majority of spores settled in the angle between the valley floor and sidewall. Lower numbers settled on the surfaces with lower profile features. Silica beads of similar dimension to the spore body were used to determine whether the spatial relationships between settled spores and the topographic features were a consequence of active settlement behaviours. The results are discussed in relation to the energy needs for spores to adhere to various surface features.  相似文献

The settlement responses of Pomatoceros lamarkii (Polychaeta: Serpulidae) larvae to biofilms of varying age on slate surfaces and to dried biofilms on slate surfaces were investigated in the laboratory. Settlement experiments were performed as multi-treatment, still water assays. Larvae did not settle on clean, non-biofilmed slates but settled on biofilms up to 28 days old. Settlement intensity was closely related to the bacterial density of a biofilm. Drying a biofilmed surface for 1-2 h at 20 degrees C to simulate a single tidal emersion completely negated the former inductive effect of the biofilm. Drying also negated the larval settlement-inducing effect that normally results from the presence of conspecific adults. The settlement inhibition lasted for approximately 5 days following a single drying event. Larvae settled readily on biofilms exposed to formalin and antibiotics. Treating biofilms with formalin or antibiotics before or after drying had no effect upon larval avoidance of dried biofilms. Freeze-drying a biofilm had the same effect as aerial drying. The biofilm drying effect could not be mimicked by exposing biofilms to hyper-saline seawater. The finding that P. lamarkii larvae do not settle on dried biofilms could have significance in explaining the natural distribution of this species in the intertidal.  相似文献

The global mussel aquaculture industry uses specialised spat catching and nursery culture ropes made of multi-filament synthetic and natural fibres to optimise settlement and retention of mussels for on-growing. However, the settlement ecology and preferences of mussels are poorly understood and only sparse information exists in a commercial context. This study quantified the settlement preferences of pediveligers and plantigrades of Mytilus galloprovincialis on increasingly complex surfaces and settlement locations at a micro spatial scale on and within ropes under commercial hatchery operating conditions using optical microscopy and X-ray micro-computed tomography (µCT). M. galloprovincialis has clear settlement preferences for more complex materials and high selectivity for settlement sites from the pediveliger through to the plantigrade stage. Pediveligers of M. galloprovincialis initially settle inside specialised culture ropes. Larger pediveligers were located close to the exterior of ropes as they increased in size over time. In contrast, smaller individuals were located deeper inside of the ropes over time. This study demonstrates that X-ray µCT is an excellent non-destructive technique for mapping settlement and attachment sites of individuals as early as one day post settlement, and quantifies the number and location of settled individuals on and within ropes as a tool to understand and optimise settlement in complex multi-dimensional materials and environments.  相似文献

Fouling-release coatings were prepared from blends of a fluorinated/siloxane copolymer with a poly(dimethyl siloxane) (PDMS) matrix in order to couple the low modulus character of PDMS with the low surface tension typical for fluorinated polymers. The content of the surface-active copolymer was varied in the blend over a broad range (0.15–10 wt % with respect to PDMS). X-ray photoelectron spectroscopy depth profiling analyses were performed on the coatings to establish the distribution of specific chemical constituents throughout the coatings, and proved enrichment in fluorine of the outermost layers of the coating surface. Addition of the fluorinated/siloxane copolymer to the PDMS matrix resulted in a concentration-dependent decrease in settlement of barnacle, Balanus amphitrite, cyprids. The release of young plants of Ulva, a soft fouling species, and young barnacles showed that adhesion strength on the fluorinated/siloxane copolymer was significantly lower than the siloxane control. However, differences in adhesion strength were not directly correlated with the concentration of copolymer in the blends.  相似文献

Larval behaviour is important to dispersal and settlement, but is seldom quantified. Behavioural capabilities of larval Lutjanus carponotatus in both offshore pelagic and reef environments at Lizard Island, Great Barrier Reef were observed in situ to determine if they were sufficient to influence dispersal. Offshore, larvae swam with higher directional precision and faster on the windward side of the island (28 cm.s−1) than on the leeward side (16 cm s−1). Most larvae swam directionally. Mean swimming directions were southerly in the windward area and northerly in the leeward area. Larvae avoided the surface and remained mostly between 3–15 m. Larvae released near reefs were 2–3 times faster swimming away from reefs (19 cm s−1) than swimming toward or over them (6–8 cm s−1). Speed swimming away was similar to that offshore. Of 41 larvae released near reefs, 73% reached the reef, 59% settled, and 13% of those reaching the reef were eaten. Larvae settled onto hard and soft coral (58%), topographic reef features (29%) and sand and rubble (13%). Settlement depth averaged 5.5 m (2–8 m). Before settling larvae spent up to 800 s over the reef (mean 231 s) and swam up to 53 m (mean 14 m). About half of the larvae interacted with reef residents including predatory attacks and aggressive approaches by residents and aggressive approaches by settling larvae. Settlement behaviour of L. carponotatus was more similar to a serranid than to pomacentrids. Settlement-stage larvae of L. carponotatus are behaviourally capable, and have a complex settlement behaviour.  相似文献

Experiments performed in the field and in the laboratory show that the barnacle, Balanus improvisus, preferentially settles on smooth surfaces. Settlement and recruitment of B. improvisus was evaluated on micro-textured surfaces with scales of surface texture ranging from 1 to 100 μm in profile heights. Surface texture with profile heights within a topographic range of 30-45 μm reduced settlement and recruitment by 92% as compared to smooth surfaces. The reduction in recruitment on micro-textured surfaces is best explained by behavioural responses to surface topography. Behavioural experiments show that cyprids have a higher propensity for smooth surfaces than for micro-textured surfaces. Cyprids spend more time exploring smooth surfaces and more time swimming when exposed to micro-textured surfaces. Micro-textured surfaces are more often rejected by cyprids after exploration than smooth surfaces. It is suggested that some scales of surface texture could be exploited to improve future anti-fouling techniques in geographical areas where Balanus improvisus is a severe fouling problem.  相似文献

The development of novel, fouling‐release surfaces has led to the need for better test methods to evaluate their performance. A water channel has been designed to measure the adhesion strength of microfouling organisms to test surfaces. The apparatus allows six replicate microscope slides to be mounted in a fully‐developed, turbulent channel flow. Wall shear stress in the test section can be varied from 0.9–30 Pa over a Reynolds number range of 2,800 to 27,000 based on the bulk mean velocity and channel height. Calibration of the device indicates that the accuracy and repeatability in the wall shear stress is within 4% throughout the range. Experiments using the fouling diatom Amphora settled on acid‐washed glass slides are presented. The results show significant differences in the shear stress required to remove Amphora cells with settlement time. No significant differences among the replicate slides were observed, indicating flow uniformity in the test section.  相似文献
