The activities of organ- and tissue-specific diagnostically significant enzymes, including aspartate aminotransferase, alanine aminotransferase, γ-glutamyl transferase, alkaline phosphatase, total and prostatic acid phosphatase, glutamate dehydrogenase, choline esterase, amylase, lactate dehydrogenase, oxybutyrate dehydrogenase, and creatin phosphokinase (muscular and cardiac isoenzymes), as well as the concentrations of total cholesterol and the cholesterol of high- and low-density lipoproteins, the ratios of their concentrations, and the level of triglycerides, were studied during an experiment with 120-day antiorthostatic (?7°) hypokinesia (ANOH) in the blood of six volunteers (men, 21 to 36 years old). In addition, the indices of lipid peroxidation (LPO) and antioxidant activity were determined, including the concentrations of diene conjugates, malonic dialdehyde, Schiff bases and tocopherol, and the total antioxidant activity. It was found that a 120-day ANOH in this experiment did not result in the formation of deep metabolic shifts accompanied by unfavorable changes in organs and tissues. The changes in cholesterol metabolism during hypokinesia were expressed in the redistribution of cholesterol fractions with the dominance of atherogenic forms and a higher risk of atherogenesis. All of the observed metabolic changes were easily reversible and eliminated during the first month of the recovery period. No signs of a higher intensity of free-radical processes were observed during any of the examination periods due to activation of the antioxidant defense system.  相似文献

Data are summarized on changes in the human cardiovascular system associated with the use of cuffs during seven days of antiorthostatic hypokinesia simulating weightlessness. Eight subjects participated in two series of experiments, of which one was carried out with and the other (the control) without cuffs wrapped snugly around the upper third of the thighs. The parameters of the systemic hemodynamics, the cardiac function, and the hemodynamics of the cervicocephalic region and the lower limbs recorded under control and experimental conditions were analyzed. Without cuffs, changes in the hemodynamics during antiorthostatic hypokinesia were caused by displacement of body fluids in the cranial direction. The subjects responded favorably to the use of cuffs during antiorthostatic hypokinesia: most of their hemodynamic parameters remained at the baseline level, and signs of venous stasis in the cervicocephalic region were alleviated. Although the leg veins were distended in subjects wearing thigh cuffs during antiorthostatic hypokinesia, no pathological changes in the veins were detected during or after the experiment. Cuff usage during antiorthostatic hypokinesia lasting for seven days did not produce a cumulative effect on the cardiovascular system. These results justify the use of thigh cuffs in the initial period of adaptation to simulated or real weightlessness.  相似文献

The effect of 120-day antiorthostatic hypokinesia on human physical working capacity and functional state was studied. The study revealed two adaptation phases: the first phase is characterized by a significant decrease in working capacity and the second by relative readaptation. However, the unfavorable effect of 120-day antiorthostatic hypokinesia was evident as a decrease in the effectiveness of function of the cardiorespiratory system that increased the physiological cost of physical work.  相似文献

The content of lipid peroxidation (LPO) products (diene conjugates (DC), malondialdehyde (MDA), Schiff bases (SB), and tocopherol (TP, a main lipid antioxidant) were measured in blood serum of 17 astronauts taking part in long-term (125–217 days) missions on board the International Space Station (ISS) during the preflight period, on the day of the landing, and on the 7th and 14th days after landing (the rehabilitation period, RP). A decrease in the DC and MDA levels against a background of an increase in TP has been found in a group of eight astronauts after landing on board the Space Shuttle spacecraft and a group of eight astronauts after a space flight on board the Soyuz TM in the course of RP. The changes in measured indices were more pronounced in the group of astronauts after the space flight on board the Space Shuttle spacecraft. Inhibition of LPO during RP was regarded as an adequate response to readaptation stress to the conditions on earth. The possible mechanisms of differences in the efficiency of LPO inhibition between groups are discussed: the changes in the biomembrane phase state under the conditions of deceleration load during disorbiting and the stressful reaction to landing on board different spacecrafts.  相似文献

Elevation of the content of lipid peroxidation (LPO) products in blood serum of patients with acute virus hepatitis (VH) is caused by an increase in the patients' blood serum lipids rather than by the intensity of peroxide reactions in lipids. There is a reverse correlation between the content of LPO products and bilirubin level and a direct correlation between lipid antioxidant activity (AOA) and bilirubin level. Marked antioxidant action of bilirubin that compares very favourably with the action of ionol (4-methyl-2,6-ditretbutylphenol) was demonstrated in the model of oxidation of methyl oleate. It was shown that the rise of lipid AOA during VH might be completely attributed to the antioxidant properties of bilirubin. It is suggested that elevation of bilirubin level and associated increase of lipid AOA during VH can be viewed as a reaction aimed at a decrease of the level of toxic products of LPO and intensification of reparative processes in the liver.  相似文献

The results of changes in the physiological cost of 30-min submaximal aerobic bicycle ergometric exercise and characteristics of the mitochondrial apparatus of m. vastus lateralis were assessed comparatively during 120-day (–6°) antiorthostatic hypokinesia either without prophylactic measures or with low-intensity resistance exercise training for 60 days using a Penguin exercise suit. Hypokinesia was accompanied by an increase in the working heart rate and lactate accumulation in the blood during the test exercise, as well as by a decrease in the myofibril size and the volume density of mitochondria in the m. vastus lateralis fibers. The patterns of dynamic changes in the lactate concentration in the blood during exercise training and in the volume density of central mitochondria were found to be similar. A correlation between the rate of lactate accumulation in the blood during the test exercise and the volume density of mitochondria in the working muscle appeared after long-term (60 days) exposure to hypokinesia. The use of the Penguin exercise suit in dynamic mode during prolonged (60-day) exposure to hypokinesia completely prevented the following effects: atrophy of slow-type fibers, a decrease in the volume density of central mitochondria, and an increase in the level of lactate accumulation in the blood under conditions of a standard submaximal aerobic exercise load. The correlation links between the oxidative potential of working muscle and the energy supply of muscular work are discussed.  相似文献

The changes in activity of antioxidant enzymes of LPO products and lipophilic bioantioxidants of pheasants during the period from 1 day up to 60-day age was investigated for the first time. It is shown, that insufficiently generated enzymatic antioxidant systems is a cause and result of high intensity of processes of free-radical LPO in 1-day pheasants tissues. The compensated increase of antioxidant enzymes (SOD, CAT) activity during the early period of postnatal ontogenesis does not prevent from the fast exhaustion of a pool of the main lipophilic bioantioxidants that in its turn provokes the inhibition of antioxidant system enzymes reactions. This results in the decrease of adaptability of wild birds in conditions of artificial breeding.  相似文献

Problems of adaptation of functional systems of the human body to conditions of continuous weightlessness are considered (prolonged stay under conditions of antiorthostatic hypokinesia and in an immersion medium). It was revealed that, during adaptation to these conditions, polymorphic clinicofunctional disorders develop, transforming into clinicophysiological syndromes, the most frequently observed being autonomic vascular malfunction, asthenoneurotic syndrome, detraining of the blood circulatory system, trophic and neuromuscular disorders, statokinetic syndrome, pain syndrome, and metabolic and hormonal disorders. The severity of the specified disorders and the duration of the recovery period depend on the duration of hypokinesia and the intensity and regularity of application of preventive measures during hypokinesia. The most probable pathophysiological mechanisms of adverse effects of continuous hypokinesia (maladaptation) on functional systems of the human body are described.  相似文献

Hydration status of humans was assessed by means of bioimpedancemetry on board the space station or under the conditions of antiorthostatic hypokinesia (AOSH). Water compartments of the body were decreased in a cosmonaut at the seventh day of a ten-day-long flight to the same degree as in a group of six testers by the seventh day of AOSH (?8°): the amount of total body fluids and intracellular and extracellular volumes were decreased by 5.6–6.5% as compared to the baseline level. The changes in body composition of a cosmonaut during flight were similar to the changes observed in testers during AOSH: lean body weight, which was determined by bioimpedancemetry, was insignificantly decreased, whereas the adipose component of body weight was, on the contrary, increased. It was concluded that the hydration level of the human body was decreased and the amount of body fat was increased during a short-term space flight. It was also shown that the hydration status and composition of the human body were changed in a similar way under the conditions of both AOSH and space flight, which indicates that this ground-based model is adequate for simulation of hydration changes caused by microgravity.  相似文献

The levels of the lipid peroxidation products (LPO), different forms of protein SH-groups and their oxidation rate in the homogenates of the mesencephalon, hypothalamus and sensorymotor cortex of normal and GSH-deficient rats under 3-day food starvation were studied. It was shown, that the basic level of LPO products--lipid hydroperoxides and malonic dialdehyde (MDA) in hypothalamus and sensorymotor cortex of normal animals are by 20-30% (p < 0.05) higher and reduced glutathione (GSH) content is 2 times higher, than these values in mesencephalon. Under 3 day starvation of normal animals activation of the LPO observed only in the hypothalamus and sensorymotor cortex, whereas under 3 day starvation of the GSH-deficient rats formed by the intraparenteraly injection of diethylmaleate in a dose of 2.5 mmol/kg of body weight in all investigated structures the lipid hydroperoxides and MDA increased many times (2-3 times), the content of the surface and masked protein SH-groups decreased and essentially increased the oxidation rate of these functional groups. It was proposed that GSH and its enzymes participate in the LPO regulation and protection of protein SH-groups from oxidative damage at this event the intensity of this prosesse depends on structural and functional organization of nervous tissues.  相似文献

Summary

The antioxidant potential of the brain in developing fetuses was assessed at gestational days (GD) 16, 18 and 20 and postnatal day (PND)1. Higher activities of superoxide dismutase (SOD) and glutathione peroxidase (GPx) were noticed during fetal development which were reduced to about half and one-quarter, respectively, at PND 1. Glutathione reductase (GR) activity remained stationary throughout the experiment and the values were very high compared to those reported for weanling rats. In contrast, catalase (CAT) activity increased with development. Glutathione (GSH) and total sulfhydryls (TSH) were maximum in 16-day fetal brains and declined subsequently. Brain lipid peroxidation (LPO) was found to increase with age. A group of animals was exposed to 20 ppm cadmium (Cd) in drinking water from the day of conception up to PND 1. Cd was found to increase the activities of brain SOD, CAT, and GR significantly at all the time intervals. The metal exposure decreased fetal brain GPx at GD 18 and 20, whereas GPx activity declined precipitously in both groups on PND 1. Cd caused both increments and decrements in the GSH and TSH levels (depending on gestational day) and increased the LPO in brain. It may be concluded that the Cd-intoxicated fetal brain undergoes significant changes in antioxidant defense parameters which, overall, may be sufficient to permit near-normal development and prevent substantial oxidant damage.  相似文献

The efficacies of the standard procedure for application of negative pressure on the lower part of the body (NPLB) and a combination of NPLB and negative-pressure respiration (NPR) during a simulated last stage of a spaceflight under conditions of antiorthostatic hypokinesia (ANOH, –6°) and isolation for 7 days have been compared in the course of three series of tests involving six volunteers in an EU-100 pressurized chamber. After the end of the 7-day ANOH and isolation, episodes of orthostatic disorders were observed in all six subjects in the first series of tests (the control series) and in four subjects in the second series (NPLB). In the third series (NPLB + NPR), orthostatic disorders, if any, were slight. Two main conclusions have been made. First, ANOH combined with isolation in a small chamber may be used to simulate the effects of the combined factors of spaceflight on humans in order to obtain model gravitational circulatory disorders. Second, the combined use of NPLB + NPR under these conditions may be effective for the prevention of orthostatic circulatory disorders in humans.  相似文献

In the blood serum of seventeen members of crews which participated in 14 orbital expeditions to the International Space Station with the duration of 125 to 217 days, during the pre-flight period and on the day of landing on the 1st, 7th and 14th days of the rehabilitation period (RP) the content of LPO products was determined, namely diene conjugates (DC), malon dialdehyde (MDA), shiffbases (SB) and the main lipid oxidant - tocopherol (TP). The group of astronauts who made landing in the Space Shuttle spacecraft (8 persons) and the group of astronauts who accomplished space mission in the Soyus TM spacecraft (9 persons) demonstrated a decrease in DC and MDA levels with a rise in TF concentration in the course of the rehabilitation period. Changes in the group of the American spacecraft astronauts were more pronounced. LPO inhibition during the rehabilitation period is recognized [treated] as an adequate reaction to the stress caused by re-adaptation to the ground conditions. Also are discussed probable mechanisms of intergroup differencies in LPO intensity degree: biomembrane phase state changing under the influence of overloads during de-orbiting and stress response intensity during landing in different types of spacecraft.  相似文献

为科学管理和利用池塘长期养殖产生的沉积物,实行池塘养殖和种植适度轮换是利用底泥养分和改善池塘底质的重要途径。将基本情况完全相同的9个池塘停止养鱼,进行种植水稻改良底质试验,以空置为对照(CK)、设种稻不晒田(T1)和晒田(T2)两个处理,检测试验过程沉积物的主要化学性质和氨氧化古菌(Ammonia-oxidizing archaea, AOA)的丰度及水稻收获后AOA的群落多样性和结构差异。结果表明,与CK相比, T1(或T2)处理下沉积物中TN、TP和OM显著减少,分别降低了26.58%(31.04%)、58.84%(58.38%)和11.17%(16.11%); T2与T1相比,显示种稻过程中晒田可以加速沉积物中TP和OM的去除;沉积物硝化强度和AOA丰度整体上呈现先增后减的趋势,其中,与CK相比, T1、T2的沉积物硝化强度在水稻分蘖期和拔节孕穗期显著提高, AOA丰度在分蘖期显著增加;种植水稻可以显著提升沉积物AOA的α多样性和OTU数量,增加许多新的AOA种群, T1主要增加了Candidatus nitrosocosmicus cluster(占总序列的3.84%), T2...  相似文献

Background

We investigated the pathological and diagnostic role of selected markers of inflammation, oxidant/antioxidant status, and cellular injury in human Chagas disease.

Methods

Seropositive/chagasic subjects characterized as clinically-symptomatic or clinically-asymptomatic (n = 116), seronegative/cardiac subjects (n = 102), and seronegative/healthy subjects (n = 45) were analyzed for peripheral blood biomarkers.

Results

Seropositive/chagasic subjects exhibited an increase in sera or plasma levels of myeloperoxidase (MPO, 2.8-fold), advanced oxidation protein products (AOPP, 56%), nitrite (5.7-fold), lipid peroxides (LPO, 12–17-fold) and malondialdehyde (MDA, 4–6-fold); and a decline in superoxide dismutase (SOD, 52%) and glutathione (GSH, 75%) contents. Correlation analysis identified a significant (p<0.001) linear relationship between inflammatory markers (AOPP/nitrite: r = 0.877), inflammation and antioxidant/oxidant status (AOPP/glutathione peroxidase (GPX): r = 0.902, AOPP/GSH: r = 0.806, Nitrite/GPX: 0.773, Nitrite/LPO: 0.805, MDA/MPO: 0.718), and antioxidant/oxidant levels (GPX/MDA: r = 0.768) in chagasic subjects. Of these, MPO, LPO and nitrite biomarkers were highly specific and sensitive for distinguishing seropositive/chagasic subjects from seronegative/healthy controls (p<0.001, training and fitting AUC/ROC >0.95). The MPO (r = 0.664) and LPO (r = 0.841) levels were also correlated with clinical disease state in chagasic subjects (p<0.001). Seronegative/cardiac subjects exhibited up to 77% decline in SOD, 3–5-fold increase in LPO and glutamate pyruvate transaminase (GPT) levels, and statistically insignificant change in MPO, AOPP, MDA, GPX, GSH, and creatine kinase (CK) levels.

Conclusions

The interlinked effects of innate immune responses and antioxidant/oxidant imbalance are major determinants of human Chagas disease. The MPO, LPO and nitrite are excellent biomarkers for diagnosing seropositive/chagasic subjects, and MPO and LPO levels have potential utility in identifying clinical severity of Chagas disease.  相似文献

Changes in the water and sodium balances and in the states of the fluid compartments of the human body observed in experiments performed with healthy subjects exposed to long-term (120 days) antiorthostatic hypokinesia (ANOH) were analyzed. A hypothesis was suggested that the normal dietary consumption of sodium could be associated with the accumulation of osmotically inactive sodium in the body of a healthy person (independently of changes in the total water content). The results agree with the assumption that considerable amounts of osmotically inactive sodium may be stored in the human body. This hypothesis was confirmed by the inversion of the correlation between the cumulative sodium balance and the total water content of the body found both in the group-averaged data and in individual data. This nonsmotic sodium accumulation may take place not only during deviations from its normal consumption, but also during its regular dietary supply. Accumulation of sodium in these stores and its depletion are not associated with any significant changes in the volumes of body fluids. Infradian rhythmic changes in the sodium balance observed in some subjects exposed to the long-term ANOH, which were not caused by any periodic external influences, indicated the existence of a specific mechanism regulating the sodium content of the body. This mechanism must be significantly more inert and less precise than the fast regulation of the volume, osmolality, and ionic composition of extracellular fluids.  相似文献

The dynamics of the glycemic curve profile was investigated in 8 healthy women during performance of the glucose tolerance test (GТТ) in 120-day antiorthostatic hypokinesia (–6°) (АNОH) simulating the effects of weightlessness. Glycemic profiles were characterized by more marked hyperglycemic response at 30 min of GТТ in the first month of stay under ANOH conditions and had a flattened appearance three months later. Glycemic profiles characteristic of delayed restoration of the blood glucose concentration appeared in the fourth month, which persisted over the first week after the end of АNОH. The glycemic profile changes developed against the background of the signs of progressive congestion of blood in the venous system of the abdominal cavity. The expansion of diameters of the main veins of the abdominal cavity was noted at the beginning of АNОH; an increase in the size of the organs and the signs of expansion of venous plexuses at the site of portocaval anastomoses, after three months; the occurrence of the signs of transudation and free fluid in the abdominal cavity was revealed at the end of hypokinesia. The countermeasures against hypodynamia did not prevent transformation of the glycemic profile nor did they influence the progression of blood flow congestion in the venous system of the abdominal cavity whose features determined the character changes in the glycemic profile in different periods of ANOH.  相似文献

Diazinon is one of the most widely used organophosphate insecticides (OPIs) in agriculture and public health programs. Reactive oxygen species (ROS) caused by OPIs may be involved in the toxicity of various pesticides. The aim of this study was to investigate how diazinon affects lipid peroxidation (LPO) and the antioxidant defense system in vivo and the possible ameliorating role of vitamins E and C. For this purpose, experiments were done to study the effects of DI on LPO and the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and catalase (CAT) in adult rat heart. Experimental groups were: (1) control group, (2) diazinon treated (DI) group, (3) DI+vitamins E and C-treated (DI+Vit) group. The levels of malondialdehyde (MDA) and the activities of SOD and CAT increased significantly in the DI group compared with the control group. The activity of SOD and the levels of MDA decreased significantly in the DI+Vit group compared with the DI group. The differences between the DI+Vit and control groups according to the MDA levels and the activities of both SOD and CAT were statistically significant. These results suggest that treating rats with a single dose of diazinon increases LPO and some antioxidant enzyme activities in the rat myocardium and, in addition, that single-dose treatment with a combination of vitamins E and C after the administration of diazinon can reduce LPO caused by diazinon, though this treatment was not sufficiently effective to reduce the values to those in control group.  相似文献

Malate- and isocytratedehydrogenase activity in mitochondrial and cytoplasmic fractions and lactate dehydrogenase activity in hindlimb muscles have been studied at different stages after 18.5-day flight on a biosatellite "Cosmos-1129" and after 20-day hypokinesia. A decrease in dehydrogenase activity has been found on the first postflight day. The enzyme activities returned to the control values in mitochondria, and in the cytoplasm they were greater by day 6 postflight. It was concluded that hypokinesia did not reveal all the effects of microgravity on the whole system but some enzyme alterations in the muscle resembled those observed during the flight. The effects may be caused by the inhibition of both aerobic and anaerobic metabolic pathways under the effect of microgravity.  相似文献

Radio frequency wave (RFW) generated by base transceiver station (BTS) has been reported to make deleterious effects on reproduction, possibly through oxidative stress. This study was conducted to evaluate the effect of RFW generated by BTS on oxidative stress in testis and the prophylactic effect of vitamin C by measuring the antioxidant enzymes activity, including glutathione peroxidase, superoxide dismutase (SOD) and catalase, and malondialdehyde (MDA). Thirty-two adult male Sprague–Dawley rats were randomly divided into four experimental groups and treated daily for 45 days as follows: sham, sham+vitamin C (l-ascorbic acid 200 mg/kg of body weight/day by gavage), RFW (exposed to 900 MHz RFW) ‘sham’ and ‘RFW’ animals were given the vehicle, i.e., distilled water and the RFW+vitamin C group (received vitamin C in addition to exposure to RFW). At the end of the experiment, all the rats were sacrificed and their testes were removed and used for measurement of antioxidant enzymes and MDA activity. The results indicate that exposure to RFW in the test group decreased antioxidant enzymes activity and increased MDA compared with the control groups (p < 0.05). In the treated group, vitamin C improved antioxidant enzymes activity and reduced MDA compared with the test group (p < 0.05). It can be concluded that RFW causes oxidative stress in testis and vitamin C improves the antioxidant enzymes activity and decreases MDA.  相似文献
