The prokaryotic algal symbiont of ascidians, Prochloron sp., was found to exhibit carbonic anhydrase activity which is largely associated with the cell surface. This extracellular carbonic anhydrase activity was inhibited, while the intracellular activity was not affected, by chloride or bromide. Acetazolamide and ethoxyzolamide inhibited carbonic anhydrase activity with I50 values of 7×10-4 and 3×10-4M, respectively. These I50 values are similar to those observed for intracellular carbonic anhydrases of Synechococcus sp. The Gram-positive bacterium Streptococcus pneumoniae is a human respiratory tract pathogen that contributes significantly to global mortality and morbidity. It was recently shown that this bacterial pathogen depends on a conserved ??-carbonic anhydrase (CA, EC 4.2.1.1) for in vitro growth in environmental ambient air and during intracellular survival in host cells. Hence, it is to be expected that this pneumococcal carbonic anhydrase (PCA) contributes to transmission and pathogenesis of the bacterium, making it a potential therapeutic target. In this study, purified recombinant PCA has been further characterized kinetically and for inhibition with a series of inorganic anions and small molecules useful as leads. PCA has appreciable activity as catalyst for the hydration of CO2 to bicarbonate, with a kcat of 7.4 × 105 s−1 and kcat/Km of 6.5 × 107 M−1 s−1 at an optimum pH of 8.4. Inorganic anions such as chloride, bromide, iodide, cyanate, selenocyanate, trithiocarbonate, and cyanide were effective inhibitors of PCA (KIs of 21-98 ??M). Sulfamide, sulfamic acid, phenylboronic, phenylarsonic acid, and diethyldithiocarbamate showed inhibition constants in the low micromolar/submicromolar range (KIs of 0.61-6.68 ??M), whereas that of the sulfonamide acetazolamide was in the nanomolar range (KIs 89 nM). In conclusion, our results show that PCA can effectively be inhibited by a range of molecules that could be interesting leads for obtaining more potent PCA inhibitors. PCA might be a novel target for designing antimicrobial drugs with a new mechanism of action.  相似文献

Oxygen-18 exchange techniques were applied to the dehydration of bicarbonate catalyzed by human carbonic anhydrase C. The rates of depletion of oxygen-18 from labeled bicarbonate were measured for both the catalyzed and uncatalyzed reactions at pH 9.4 and 25 °C. The equilibrium dissociation constant of the enzyme-substrate complex K is 0.321 ± 0.040 m and kenz = k2Km is (8.3 ± 1.9) × 105m?1 sec?1 under these conditions. On the basis of these results it is demonstrated that the oxygen-18 exchange technique is capable of measuring K and kenz for the carbonic anhydrase catalyzed dehydration of bicarbonate at a high pH range in which other kinetic techniques are not effective.It was also shown that the oxygen-18 exchange technique is an effective micromethod for the determination of carbonic anhydrase. Rates of isotopic depletion of labeled bicarbonate (in solutions of the enzyme) which fall outside the limits of error for the uncatalyzed rate of depletion demonstrate that this technique can detect concentrations of human carbonic anhydrase C as low as 5 × 10?11m.  相似文献

Rate and equilibrium constants at 25 °C, pH ∼ 1, and ionic strength 0.10 for hydrolysis of the two non-equivalent chlorides of dichloro[S-methyl-l-cysteine(N,S)]platinum(II) isomers, denoted [PtCl2(SmecysH)], and the resultant chloro-aqua species have been determined by NMR, potentiometric, and spectrophotometric methods. Though hydrolysis constants, Kh, for the two chlorides are similar (pKh = 4-5), the rate of hydrolysis of the chloride trans to coordinated S, kh = 3.4 × 10−3 s−1, is 2-3 orders of magnitude faster than the kh for the other chloride, 2.3 × 10−6 s−1, and for the cancer drug cisplatin, cis-[PtCl2(NH3)2], 5.2 × 10−5 s−1. Relative rates of hydrolysis determined under three different experimental conditions (pH ∼ 1 in 0.10 M HNO3, high pH in 0.10 M NaOH, and at low pH with Ag+ assistance) are consistent: the Cl trans to S is 100-1000 times more labile than the Cl cis to S. Potentiometric and NMR methods were also used to estimate pKa values of all aqua species, which are comparable to values reported for corresponding aqua species derived from cisplatin.  相似文献

Fiber-optic biosensors have been studied intensively because they are very useful and important tools for monitoring biomolecular interactions. Here we describe a fluorescence detection fiber-optic biosensor (FD-FOB) using a sandwich assay to detect antibody-antigen interaction. In addition, the quantitative measurement of binding kinetics, including the association and dissociation rate constants for immunoglobulin G (IgG)/anti-mouse IgG, is achieved, indicating 0.38 × 106 M−1 s−1 for ka and 3.15 × 10−3 s−1 for kd. These constants are calculated from the fluorescence signals detected on fiber surface only where the excited evanescent wave can be generated. Thus, a confined fluorescence-detecting region is achieved to specifically determine the binding kinetics at the vicinity of the interface between sensing materials and uncladded fiber surface. With this FD-FOB, the mathematical deduction and experimental verification of the binding kinetics in a sandwich immunoassay provide a theoretical basis for measuring rate constants and equilibrium dissociation constants. A further measurement to study the interaction between human heart-type fatty acid-binding protein and its antibody gave the calculated kinetic constants ka, kd, and KD as 8.48 × 105 M−1 s−1, 1.7 × 10−3 s−1, and 2.0 nM, respectively. Our study is the first attempt to establish a theoretical basis for the florescence-sensitive immunoassay using a sandwich format. Moreover, we demonstrate that the FD-FOB as a high-throughput biosensor can provide an alternative to the chip-based biosensors to study real-time biomolecular interaction.  相似文献

The steady-state kinetic parameters for the hydration of CO2 catalyzed by membrane-bound carbonic anhydrase from the renal brush-border of the dog are compared with the same parameters for water-soluble bovine erythrocyte carbonic anhydrase. For the membrane-bound enzyme, the turnover number kcat is 6.5 × 105 s?1 and the Michaelis constant is 7.5 mm for CO2 hydration at pH 7.4 and 25 °C. The corresponding constants for bovine carbonic anhydrase under these conditions are 6.3 × 105 s?1 and 15 mm (Y. Pocker and D.W. Bjorkquist (1977)Biochemistry16, 5698–5707). The rate constant for the transfer of a proton between carbonic anhydrase and buffer was determined from the dependence of the catalytic rate on the concentration of the buffers imidazole and N-2-hydroxyethylpiperazine-N′-2-ethanesulfonic acid (Hepes); the value of 2 × 108m?1s?1 describes this constant for both forms of carbonic anhydrase at pH 7.4. Furthermore, the pH dependence of the initial velocity of hydration of CO2 in the range of pH 6.5 to 8.0 is identical for the membrane-bound and soluble enzyme at low buffer concentration (1–2 mm imidazole). We conclude that the membrane plays no detectable role in affecting the CO2 hydration activity and that the active site of the renal, membrane-bound carbonic anhydrase is exposed to the aqueous phase.  相似文献

Nucleotide binding properties of two vacant noncatalytic sites of thioredoxin-activated chloroplast coupling factor 1 (CF1) were studied. Kinetics of nucleotide binding to noncatalytic sites is described by the first-order equation that allows for two nucleotide binding sites that differ in kinetic features. Dependence of the nucleotide binding rate on nucleotide concentration suggests that tight nucleotide binding is preceded by rapid reversible binding of nucleotides. ADP binding is cooperative. The preincubation of CF1 with Mg2+ produces only slight effect on the rate of ADP binding and decreases the ATP binding rate. The ATP and ADP dissociation from noncatalytic sites is described by the first-order equation for similar sites with dissociation rate constants k−2(ADP)=1.5×10−1 min−1 and k−2(ATP)≅10−3 min−1, respectively. As follows from the study, the noncatalytic sites of CF1 are not homogeneous. One of them retains the major part of endogenous ADP after CF1 precipitation with ammonium sulfate. Its other two sites can bind both ADP and ATP but have different kinetic parameters and different affinity for nucleotides.  相似文献

Human serum albumin (HSA) participates to heme scavenging, in turn HSA-heme binds gaseous diatomic ligands at the heme-Fe-atom. Here, the effect of abacavir and warfarin on denitrosylation kinetics of HSA-heme-Fe(II)-NO (i.e., koff) is reported. In the absence of drugs, the value of koff is (1.3 ± 0.2) × 10−4 s−1. Abacavir and warfarin facilitate NO dissociation from HSA-heme-Fe(II)-NO, the koff value increases to (8.6 ± 0.9) × 10−4 s−1. From the dependence of koff on the drug concentration, values of the dissociation equilibrium constant for the abacavir and warfarin binding to HSA-heme-Fe(II)-NO (i.e., K = (1.2 ± 0.2) × 10−3 M and (6.2 ± 0.7) × 10−5 M, respectively) were determined. The increase of koff values reflects the stabilization of the basic form of HSA-heme-Fe by ligands (e.g., abacavir and warfarin) that bind to Sudlow’s site I. This event parallels the stabilization of the six-coordinate derivative of the HSA-heme-Fe(II)-NO atom. Present data highlight the allosteric modulation of HSA-heme-Fe(II) reactivity by heterotropic effectors.  相似文献

Although allosteric transitions of GroEL by MgATP2 have been widely studied, the initial bimolecular step of MgATP2− binding to GroEL remains unclear. Here, we studied the equilibrium and kinetics of MgATP2 binding to a variant of GroEL, in which Tyr485 was replaced by tryptophan, via isothermal titration calorimetry (ITC) and stopped-flow fluorescence spectroscopy. In the absence of K+ at 4-5 °C, the allosteric transitions and the subsequent ATP hydrolysis by GroEL are halted, and hence, the stopped-flow fluorescence kinetics induced by rapid mixing of MgATP2 and the GroEL variant solely reflected MgATP2 binding, which was well represented by bimolecular noncooperative binding with a binding rate constant, kon, of 9.14 × 104 M− 1 s− 1 and a dissociation rate constant, koff, of 14.2 s− 1, yielding a binding constant, Kb (= kon/koff), of 6.4 × 103 M− 1. We also successfully performed ITC to measure binding isotherms of MgATP2 to GroEL and obtained a Kb of 9.5 × 103 M− 1 and a binding stoichiometric number of 6.6. Kb was thus in good agreement with that obtained by stopped-flow fluorescence. In the presence of 10-50 mM KCl, the fluorescence kinetics consisted of three to four phases (the first fluorescence-increasing phase, followed by one or two exponential fluorescence-decreasing phases, and the final slow fluorescence-increasing phase), and comparison of the kinetics in the absence and presence of K+ clearly demonstrated that the first fluorescence-increasing phase corresponds to bimolecular MgATP2 binding to GroEL. The temperature dependence of the kinetics indicated that MgATP2 binding to GroEL was activation-controlled with an activation enthalpy as large as 14-16 kcal mol− 1.  相似文献

The retention rate of the spin label 3-isothiocyanto methyl-2,2,5,5-tetramethyl-1-pyrrolidinyl oxyl spin label (proxyl) attached to the porcine N-acetyl-NPY peptide and the porcine N-acetyl-D-Trp32-NPY peptide at Lys4 was investigated using SK-N-MC neuroblastoma cell membranes containing the Y1 receptor. The release rate of the spin labeled peptides was monitored by electron spin resonance and the KD was determined by a direct radiolabeled NPY displacement binding assay. The analyses show that for the porcine [Ac-Tyr1N4-proxyl]-NPY, the KD was 8 × 10–10 M and koff was 2.7 × 10–4 sec–1 yielding a value for kon of 3.3 × 105 sec–1 M–1. The [Ac-Tyr1, N4-proxyl,-D-Trp32]-NPY antagonist ligand had a value of KD equal to 1.35 × 10–7 M and koff was 1.7 × 10–4 sec–1 leading to a value for kon of 1.2 × 103 sec–1 M–1. The difference in the kon rates of two orders of magnitude is interpreted as demonstrating the N-acetyl-N4 proxyl-D-Trp32-NPY ligand binding transition state to be of higher energy then for the unmodified NPY amino acid sequence.  相似文献

Human serum albumin (HSA) is a monomeric allosteric protein. Here, the effect of ibuprofen on denitrosylation kinetics (koff) and spectroscopic properties of HSA-heme-Fe(II)-NO is reported. The koff value increases from (1.4 ± 0.2) × 10−4 s−1, in the absence of the drug, to (9.5 ± 1.2) × 10−3 s−1, in the presence of 1.0 × 10−2 M ibuprofen, at pH 7.0 and 10.0 °C. From the dependence of koff on the drug concentration, values of the dissociation equilibrium constants for ibuprofen binding to HSA-heme-Fe(II)-NO (K1 = (3.1 ± 0.4) × 10−7 M, K2 = (1.7 ± 0.2) × 10−4 M, and K3 = (2.2 ± 0.2) × 10−3 M) were determined. The K3 value corresponds to the value of the dissociation equilibrium constant for ibuprofen binding to HSA-heme-Fe(II)-NO determined by monitoring drug-dependent absorbance spectroscopic changes (H = (2.6 ± 0.3) × 10−3 M). Present data indicate that ibuprofen binds to the FA3-FA4 cleft (Sudlow’s site II), to the FA6 site, and possibly to the FA2 pocket, inducing the hexa-coordination of HSA-heme-Fe(II)-NO and triggering the heme-ligand dissociation kinetics.  相似文献

The synthesis and characterisation of cis- and trans-[Co(tmen)2(NCCH3)2](ClO4)3 are described. Solvolysis rates have been measured by both 1H NMR spectroscopy and UV-Vis spectrophotometry in dimethyl sulfoxide at 298.2 K. The cis isomer undergoes solvolysis by consecutive first-order reactions, k1=5.61 × 10−4 and k2=5.35 × 10−4 s−1, each with steric retention. The measured solvolysis rate (single step reaction) for the trans isomer is k=1.54 × 10−5 s−1. The solvent exchange rates have been measured by 1H NMR spectroscopy in CD3CN at 298.2 K: kex(cis)=kct + kcc=2.0 × 10−5 and kex(trans)=ktc + ktt=4.56 × 10−6 s−1. From these data, the measured cis-trans isomerisation rate (1.71 × 10−6 s−1) and equilibrium position in CH3CN (17% trans), the steric course for substitution in the exchange processes has been determined: trans reactant - 69% trans product; cis reactant - 99% cis product. Aquation rates for cis- and trans-[Co(tmen)2(NCCH3)2](ClO4)3 have also been determined spectrophotometrically and by NMR; kcis=1.3 × 10−4 and ktrans=2.7 × 10−5 s−1. In both cases the steric course for the primary aquation step is indeterminate because the subsequent steps are faster. Where data are available, the [Co(tmen)2X2]n+ complexes are found to be consistently much more reactive than their [Co(en)2X2]n+ analogues.  相似文献

An immobilization procedure for protein on surface plasmon resonance sensor (SPR) chips is described. The target protein, cyclophilin D, is thereby genetically linked to a mutant of the human DNA repair protein O6-alkylguanine-DNA-alkyltransferase (hAGT). The procedure includes the immobilization of an alkylguanine derivative on the surface by amine coupling and contact of the surface with a solution of the fusion protein (TCypD-hAGT). TCypD-hAGT could be immobilized using buffer solutions of purified protein or cell extracts. High densities of covalently linked proteins were achieved by either procedure. Binding experiments performed with the ligand cyclosporin A indicate relative binding activities close to 100%. The KD value (12 nM) and the kinetic rate constants kon (3 × 105 M−1s−1) and koff (4 × 10−3s−1) are given and compared to values determined for cyclophilin D linked to the surface by amide coupling chemistry. The KD value is in excellent agreement with the KD value determined in solution by fluorescence titration.  相似文献

The folding mechanism and stability of dimeric formate dehydrogenase from Candida methylica was analysed by exposure to denaturing agents and to heat. Equilibrium denaturation data yielded a dissociation constant of about 10−13 M for assembly of the protein from unfolded chains and the kinetics of refolding and unfolding revealed that the overall process comprises two steps. In the first step a marginally stable folded monomeric state is formed at a rate (k1) of about 2 × 10−3 s−1 (by deduction k−1 is about10−4 s−1) and assembles into the active dimeric state with a bimolecular rate constant (k2) of about 2 × 104 M−1 s−1. The rate of dissociation of the dimeric state in physiological conditions is extremely slow (k−2 ∼ 3 × 10−7 s−1).  相似文献

Three barley xyloglucan endotransglycosylases (HvXETs), known as xyloglucan xyloglucosyl transferases (EC 2.4.1.207), were subjected to kinetic and computational docking studies. The kcat·Km−1 values with the reduced [3H]-labelled XXXG, XXLG/XLXG and XLLG acceptor substrates were 0.02 × 10−2, 0.1 × 10−2 and 3.2 × 10−2 s−1 μM−1, while the Km constants were 10.6, 8.6 and 5.3 mM, obtained for HvXET3, HvXET4 and HvXET6, respectively. Docking of XLLG in acceptor-binding regions revealed that at least two conformational states were likely to participate in all isoforms. The assessments of kinetic and computational data indicated that the disposition of aromatic residues at the entrance to the active sites and the flexibility of proximal COOH-terminal loops could orient acceptors more or less favourably during binding, thus leading to tighter or weaker Km constants. The data suggested that binding of acceptors in HvXETs is guided by contributions from the conserved residues in the active sites and by the of neighbouring loops.  相似文献

The effect of pH on the hydrolysis of α-N-benzoyl-l-arginine ethyl ester (BAEE) and α-N-benzoyl-l-argininamide (BAA) by a proteolytic enzyme component purified from Ficus carica var. Kadota latex has been studied in detail over the pH range of 3 to 9.5. kcat (lim) values for the hydrolysis of BAEE and BAA were essentially identical (5.20 and 5.01 sec−1, respectively at 30°). kcat values for hydrolysis of BAEE and BAA were dependent on prototropic groups with apparent pK values of 4.24 and 8.53 and 4.10 and 8.59, respectively. kcat (lim) values for tht hydrolysis of BAEE and BAA were essentially identical (5.20 and groups of pK 4.33 and 8.60 and 4.55 and 8.51, respectively. Thus the pH optimum is 6.5 for both substrates. Km (app) values for BAEE and BAA were 3.32 × 10−2m and 6.03 × 10−2m respectively over the pH range of 3.9 to 8.0. These data are interpreted in terms of the involvement of a carboxyl and a sulfhydryl group in the active center of the enzyme. The data do not support the concept that deacylation of the acyl-enzyme is completely the rate controlling step in the hydrolyses. Rather, it appears that the magnitude of k2 and k3 are not greatly different.  相似文献

Summary The charge-pulse technique has been used previously for the study of quasistationary processes in membranes which required only a moderate time resolution. It is shown here that a time resolution of about 400 nsec may be achieved with this technique and that it may be applied to the kinetic analysis of carrier-mediated ion transport. By this method we have studied the transport of alkali ions through optically black monoolein membranes in the presence of the ion carrier valinomycin. All three relaxation processes that are predicted by theory have been resolved. From the relaxation times and the relaxation amplitudes the rate constants for the association (k R ) and the dissociation (k D ) of the ioncarrier complex, as well as the translocation rate constants of the complex (k MS ) and the free carrier (k S ) could be obtained. For 1m Rb+ at 25° C the values arek R =3×105 m –1 sec–1,k D =2×105 sec–1,k MS =3×105 sec–1,k S =4×104 sec–1. The activation energies of the single rate constants which have been estimated from experiments at two different temperatures range between 50 and 90 kJ/mol.  相似文献

Alzheimer’s disease (AD) is a devastating neurodegenerative disease affecting millions of people. β-Secretase-1 (BACE-1), an enzyme involved in the processing of the amyloid precursor protein (APP) to form Aβ, is a well validated target for AD. Herein, the authors characterize 10 randomly selected hydroxyethylamine (HEA) BACE-1 inhibitors in terms of their association and dissociation rate constants and thermodynamics of binding using surface plasmon resonance (SPR). Rate constants of association (ka) measured at 25 °C ranged from a low of 2.42 × 104 M−1 s−1 to the highest value of 8.3 × 105 M−1 s−1. Rate constants of dissociation (kd) ranged from 1.09 × 10−4 s−1 (corresponding to a residence time of close to three hours), to the fastest of 0.028 s−1. Three compounds were selected for further thermodynamic analysis where it was shown that equilibrium binding was enthalpy driven while unfavorable entropy of binding was observed. Structural analysis revealed that upon ligand binding, the BACE-1flap folds down over the bound ligand causing an induced fit. The maximal difference between alpha carbon positions in the open and closed conformations of the flap was over 5 Å. Thus the negative entropy of binding determined using SPR analysis was consistent with an induced fit observed by structural analysis.  相似文献

Association of a sulfated galactosyl ceramide, sulfatide, with the viral envelope glycoprotein hemagglutinin (HA) delivered to the cell surface is required for influenza A virus (IAV) replication through efficient translocation of the newly synthesized viral nucleoprotein from the nucleus to the cytoplasm. To determine whether the ectodomain of HA can bind to sulfatide, a secreted-type HA (sHA), in which the transmembrane region and cytoplasmic tail were deleted, was generated by using a baculovirus expression system. The receptor binding ability and antigenic structure of sHA were evaluated by a hemagglutination assay, solid-phase binding assay and hemagglutination inhibition assay. sHA showed subtype-specific antigenicity and binding ability to both sulfatide and gangliosides. Kinetics of sHA binding to sulfatide and GD1a was demonstrated by quartz crystal microbalance (QCM) analysis. QCM analysis showed that the sHA bound with the association rate constant (k on) of 1.41?×?104 M?1 sec?1, dissociation rate constant (k off) of 2.03?×?10?4 sec?1 and K d of 1.44?×?10?8 M to sulfatide immobilized on a sensor chip. The k off values of sHA were similar for sulfatide and GD1a, whereas the k on value of sHA binding to sulfatide was 2.56-times lower than that of sHA binding to GD1a. The results indicate that sulfatide directly binds to the ectodomain of HA with high affinity.  相似文献

Ferrous human serum heme–albumin (HSA–heme–Fe(II)) displays globin-like properties. Here, the effect of ibuprofen and warfarin on kinetics of HSA–heme–Fe(II) nitrosylation is reported. Values of the second-order rate constant for HSA–heme–Fe(II) nitrosylation (kon) decrease from 6.3 × 106 M−1 s−1 in the absence of drugs, to 4.1 × 105 M−1 s−1 and 4.8 × 105 M−1 s−1, in the presence of saturating amounts of ibuprofen and warfarin, respectively, at pH 7.0 and 20.0 °C. From the dependence of kon on the drug concentration, values of the dissociation equilibrium constant for ibuprofen and warfarin binding to HSA–heme–Fe(II) (i.e., K = 3.2 × 10−3 M and 2.6 × 10−4 M, respectively) were determined. The observed allosteric effects could indeed reflect ibuprofen and warfarin binding to the regulatory fatty acid binding site FA2, which brings about an alteration of heme coordination, slowing down HSA–heme–Fe(II) nitrosylation. Present data highlight the allosteric modulation of HSA–heme–Fe(II) reactivity by heterotropic effectors.  相似文献
