Germination of non-dormant seeds under variable-temperature conditions can be predicted from constant-temperature germination data if it is assumed that instantaneous germination rate is independent of thermal history. Thermal-response models of this type have not been validated under simulated field-variable temperature conditions that vary in diurnal pattern, diurnal range and longer-term trends in mean–daily temperature. The purpose of this experiment was to evaluate germination response of thickspike wheatgrass (Elymus lanceolatus), bluebunch wheatgrass (Pseudoroegneria spicata), Sandberg bluegrass (Poa sandbergii) and bottlebrush squirreltail (Elymus elymoides) under both constant and field-variable temperature regimes in the laboratory. It was hypothesized that the thermal history assumption was valid and that constant-temperature data could be used to accurately estimate field-variable temperature response. Seeds were germinated at seven constant temperatures between 5 and 35°C, and under 18 variable-temperature regimes simulating six planting dates at three field sites. Predictions of germination time under variable-temperature conditions were accurate to within a fraction of 1 day up to a cumulative germination percentage of 70% for thickspike wheatgrass, 60% for bluebunch wheatgrass, 55% for Sandberg bluegrass and 70% for bottlebrush squirreltail. It was concluded that, for the variable-temperature regimes tested in this experiment, the thermal-history assumption was valid for earlier-germinating subpopulations.  相似文献

Elymus nutans has been widely planted together with other perennial grasses for rebuilding degraded alpine meadow atop the Qinghai‐Tibetan Plateau. However, the rebuilt sown pastures begin to decline a few years after establishing. One of the possible causes for the degradation of sown grassland may come from allelopathy of planted grasses. The purpose of this study was to examine allelopathic potential of Elymus nutans. Three types of aqueous extract from Elymus nutans and its root zone soil were prepared, and 5 highland crops and 5 perennial grasses were used as recipient plants. Elymus nutans exhibited strong allelopathic potential on germination and seedling growth of 5 crops, but different crops or perennial grasses respond to the extract differently. The pieces aqueous extract have stronger inhibition than whole plant extract and root zone soil extract. Hordeum vulgar var. nudum, Avena sativa, and Festuca sinensis were the most affected, while Chenopodium quinoa and Elymus sibiricus were the least affected. Elymus nutans presented less influence on Poa pratensis and Poa crymophylla than on Festuca sinensis. It is recommended that the species combination of mixture for restoration should be considered for allopathic effects on the coseeding to decrease the seeding rate ratio of Elymus nutans. The annual dicot crop seeds of Chenopodium quinoa and Brassica napus can be used as alternative subsequent crop for the seed field of Elymus nutans monoculture.  相似文献

Seed priming may enhance establishment success of cool-seasonrange grasses which must compete with annual weeds for earlyspring moisture. Previous priming studies have confirmed germinationrate enhancement for these species but relative treatment effectsunder field-temperature conditions have not been assessed. Weprimed seeds of thickspike wheatgrass [Elymus lanceolatus(Scribn.and J. G. Smith) Gould], bluebunch wheatgrass [Pseudoroegneriaspicata(Pursh) Löve], Sandberg bluegrass (Poa sandbergiiVasey.) and bottlebrush squirreltail [Elymus elymoides(Raf.)Swezey] and evaluated their relative emergence rate in threesoil types as a function of spring-planting date. Germinationresponse was simultaneously evaluated in laboratory germinatorsthat were programmed to simulate the field-temperature regimeat planting depth. Seed priming enhanced both germination andemergence rate with the greatest effect occurring during theearlier, cooler planting dates. Total emergence and emergencerate in the field were lower than for the equivalent germinationresponse in the laboratory. Thermal-germination response wasmodelled and predictions developed for evaluating potentialgermination under late winter/early spring soil-temperatureregimes. Soil disturbances can alter microbial communities including arbuscular mycorrhizal (AM) fungi, which may in turn, affect plant community structure and the abundance of exotic species. We hypothesized that altered soil microbial populations owing to disturbance would contribute to invasion by cheatgrass (Bromus tectorum), an exotic annual grass, at the expense of the native perennial grass, squirreltail (Elymus elymoides). Using a greenhouse experiment, we compared the responses of conspecific and heterospecific pairs of cheatgrass and squirreltail inoculated with soil (including live AM spores and other organisms) collected from fuel treatments with high, intermediate and no disturbance (pile burns, mastication, and intact woodlands) and a sterile control. Cheatgrass growth was unaffected by type of soil inoculum, whereas squirreltail growth, reproduction and nutrient uptake were higher in plants inoculated with soil from mastication and undisturbed treatments compared to pile burns and sterile controls. Squirreltail shoot biomass was positively correlated with AM colonization when inoculated with mastication and undisturbed soils, but not when inoculated with pile burn soils. In contrast, cheatgrass shoot biomass was negatively correlated with AM colonization, but this effect was less pronounced with pile burn inoculum. Cheatgrass had higher foliar N and P when grown with squirreltail compared to a conspecific, while squirreltail had lower foliar P, AM colonization and flower production when grown with cheatgrass. These results indicate that changes in AM communities resulting from high disturbance may favor exotic plant species that do not depend on mycorrhizal fungi, over native species that depend on particular taxa of AM fungi for growth and reproduction.  相似文献

Various cadmium (Cd) concentrations (0, 50, 100, 200 and 300 ??mol L?1) affected Elymus dahuricus seed germination, seedling growth, antioxidative enzymes activities (AEA), and amounts of malondialdehyde (MDA) and proline present. These influences were determined for separate E. dahuricus cohorts known to be either infected (E+) or non-infected (E?) by a Neotyphodium endophyte. Under high Cd concentrations (100, 200 and 300 ??mol L?1), E+ specimens showed a significantly (P<0.05) higher germination rate and index, as well as higher values for shoot length, root length and dry biomass. However, the germination rate and index, root length and dry weight did not show a significant (P<0.05) difference under the low Cd concentrations (0 and 50 ??mol L?1). AEA and proline content increased, as did MDA content, in the E+ (vs. E?) specimens under high Cd concentrations. There was no significant (P>0.05) difference under low Cd concentrations. Endophyte infection was concluded to be of benefit to E. dahuricus exposed to high Cd concentrations.  相似文献

The protective effects of 5-aminolevulenic acid (ALA) on germination of Elymus nutans Griseb. seeds under cold stress were investigated. Seeds of E. nutans (Damxung, DX and Zhengdao, ZD) were pre-soaked with various concentrations (0, 0.1, 0.5, 1, 5, 10 and 25 mg l−1) of ALA for 24 h before germination under cold stress (5°C). Seeds of ZD were more susceptible to cold stress than DX seeds. Both seeds treated with ALA at low concentrations (0.1–1 mg l−1) had higher final germination percentage (FGP) and dry weight at 5°C than non-ALA-treated seeds, whereas exposure to higher ALA concentrations (5–25 mg l−1) brought about a dose dependent decrease. The highest FGP and dry weight of germinating seeds were obtained from seeds pre-soaked with 1 mg l−1 ALA. After 5 d of cold stress, pretreatment with ALA provided significant protection against cold stress in the germinating seeds, significantly enhancing seed respiration rate and ATP synthesis. ALA pre-treatment also increased reduced glutathione (GSH), ascorbic acid (AsA), total glutathione, and total ascorbate concentrations, and the activities of superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX) and glutathione reductase (GR), whereas decreased the contents of malondialdehyde (MDA) and hydrogen peroxide (H2O2), and superoxide radical (O2 •−) release in both germinating seeds under cold stress. In addition, application of ALA increased H+-ATPase activity and endogenous ALA concentration compared with cold stress alone. Results indicate that ALA considered as an endogenous plant growth regulator could effectively protect E. nutans seeds from cold-induced oxidative damage during germination without any adverse effect.  相似文献

High molecular weight (HMW) glutenin subunits (GS) play a key role in the determination of end-use quality of wheat and other cereal crops. In this study, we report the isolation and characterization of both promoter region and ORF of novel HMW-GS allele 1St1.3 from a perennial Triticeae species, Elymus canadensis. The amino acid (AA) sequences of E. canadensis 1St1.3 were deduced as 434 aa. Its protein primary structure comprises a signal peptide with a conserved N-terminal domain, a central repetitive domain and a C-terminal domain. E. canadensis 1St 1.3 possesses several distinct characteristics which are different from those of wheat HMW-GSs. The N-terminal domains of E. canadensis 1St 1.3 resemble that of y-type subunits, while their C-terminal domains are more similar to x-type subunits. The deletion of 85 bp fragment has been observed in promoter region of 1St 1.3, however which has not interrupted the expression of this gene. Our results indicate that 1St 1.3 is novel HMW-GS variants which will be valuable for enhancing our understanding of structural differentiation and the evolutionary relationship among HMW-GSs in Triticeae species.  相似文献

 Microsatellites are highly variable DNA sequences that can be used as markers for the genetic analysis of plants. The potential of microsatellite markers for use in a genetic diversity study in Elymus species was evaluated. Genomic libraries of Elymus caninus were constructed. The libraries were screened with two dinucleotide, (GA)n and (GT)n, and two trinucleotide repeats, (TCT)n and (CAC)n. A total of 19 positive clones were found for the two dinucleotide repeats; no positive clone was found for the trinucleotide repeats. Positive clones were sequenced to confirm the presence of microsatellites and to generate polymerase chain reaction (PCR) primers based on the sequences flanking the microsatellite. All sequenced (GA)n clones have repeats of n>10; over half of the (GT)n microsatellites have n<10 repeats. Primer pairs were designed and evaluated for 8 selected microsatellites. PCR products were amplified from 15 Elymus caninus accessions. The number of alleles found for the eight loci varied from 1 for ECGA89 and ECGT35 to 13 for ECGA22, as determined by non-denaturing polyacrylamide electrophoresis. Six microsatellite loci were found to be polymorphic in E. caninus. The eight primer pairs were tested on three other species; seven were successful in amplifying DNA from Elymus alaskanus and E. mutabilis, and four amplified DNA from E. caucasicus. Recent molecular phylogenetic studies on Elymus have added to our understanding of the origination of Elymus species. However, evolutionary dynamics and speciation of most species in Elymus are unclear. Molecular phylogeny has demonstrated that reticulate evolution has occurred extensively in the genus, as an example, the largest subunit of RNA polymerase II (rpb2) and phosphoenolpyruvate carboxylase (pepC) data revealed two versions of the St genome, St1 and St2contributing to speciation of E. caninus. Phylogenetic analyses of E. pendulinus uncovered additional genome-level complexity. Our data indicated that both chloroplast and nuclear gene introgression have occurred in the evolutionary process of E. pendulinus. Non-donor species genomes have been detected in severalElymus species, such as in allohexaploid E. repens (StStStStHH), a Taeniatherum-like (Ta genome in Triticeae) GBSSI sequence, Bromus- (Bromeae) and Panicum-like (Paniceae) ITS sequences have been detected. The chloroplast DNA data indicated that Pseudoroegneria is the maternal genome donor to Elymus species, but whether different Elymus species originated from different St donors remains an open question. The origin of the Y genome in Elymus is puzzling. It is clear that the Ygenome is distinct from the St genome, but unclear on the relationships of Y to other genomes in Triticeae. Introgressive hybridization may be an important factor complicating the evolutionary history of the species in Elymus. The extent of introgression and its role in creating diversity in Elymus species should be the objective of further investigations.  相似文献

Comparative observations of Bipolaris sorokiniana and Curvularia geniculata conidia germination as influenced by culture age and temperature showed some distinct differences, but generally established the ability of these organisms to function under similar conditions. Total germination of B. sorokiniana conidia was favored by increasing culture age from 20 to 60 days and temperature to 25°C; total conidia germination of C. geniculata was favored by increasing temperature to 25°C, but increasing culture age decreased germination. These reactions seem associated with conidia age. Maximum proportional intra-population germination of conidia of each organism also varied with culture age and temperature. At temperatures of 5°C and 15°C, amplitude of maximum proportional germination of both organisms increased as culture age was increased from 20 to 40 days and then decreased among 60-day-old cultures. At 25°C and above, amplitude of maximum proportional germination of conidia of both organisms decreased from each older culture. Progressively increasing temperature at a given culture age increased the amplitude of maximum proportional germination up to 25°C for conidia of B. sorokiniana, but generally decreased it for conidia of C. geniculata (except 20-day-old cultures). Frequency (specific 2 h interval) of maximum proportional intrapopulation germination of B. sorokiniana shifted from 6 h to 2 h in response to increasing temperature and culture age; conidia from youngest cultures of C. geniculata shifted to intervals of 4 h and 2 h in response to increasing temperature to 25°C, but among conidia from 60-day-old cultures, frequency shifted to 6 h intervals at all temperatures. Above 25°C, maximum proportional germination of C. geniculata conidia from cultures of all ages occurred at 6 h. It was concluded that the germination response of B. sorokiniana and C. geniculata conidia to temperature and culture age (and, subsequently, conidia age) are enough similar that these organisms could function in a potential ‘disease complex’ on Poa pratensis and Agrostis palustris.  相似文献

以发芽率、苗长、根长、苗干重、根干重变化为种子萌发和幼苗生长参数,研究苗期紫花苜蓿(Medicago sativa)植株浸提液对不同地区垂穗披碱草(Elymus nutans)种子萌发生长的化感作用。结果表明:地上部浸提液对LS、DX垂穗披碱草种子发芽率具有明显的促进作用,而对GS、KMX、LKZ、LZ地区垂穗披碱草种子发芽率均表现为抑制作用,其中对GS垂穗披碱草种子发芽率的抑制作用最强,在14.5%和5.5%浓度处理时抑制率分别为57.89%、55.26%;苗长方面,浸提液5.5%浓度对垂穗披碱草苗长的抑制率顺序为:LZNQDXLSKMXQHGSLKZ,14.5%处理时抑制率顺序为:LZKMXLKZNQGSQHDXLS,其中抑制率最高的为LZ垂穗披碱草,在14.5%和5.5%浓度处理时抑制率分别为33.03%、28.97%;根长方面,5.5%处理对垂穗披碱草根长的抑制率顺序为:QHNQLZKMX、GSLKZDX、LS,14.5%处理时抑制率顺序为:GSQH、NQLZLSKMXDXLKZ,其中在高浓度下抑制率最高的为GS垂穗披碱草,抑制率为57.69%;地上部浸提液对LKZ、LZ垂穗披碱草苗干重均具有促进作用,高浓度浸提液对NQ垂穗披碱草苗干重产生促进作用(RI0),而对KMX、DX垂穗披碱草苗干重均表现为抑制作用;根干重方面,浸提液对LS、QH、GS、NQ、LKZ垂穗披碱草根干重均有明显的抑制作用,而对KMX、DX垂穗披碱草根干重产生促进作用,高浓度浸提液对LZ垂穗披碱草的根干重产生促进作用。从根浸提液的作用来看,根浸提液除对LS、DX垂穗披碱草种子发芽率和根干重、GS垂穗披碱草种子发芽率和苗干重及NQ垂穗披碱草根干重具有促进作用外(P 0.05),对其余地区垂穗披碱草的各项指标均有明显的抑制作用(RI0)。所有以上结果表明,紫花苜蓿植株浸提液对垂穗披碱草种子萌发生长的作用具有一定的浓度效应。不同地区垂穗披碱草对紫花苜蓿地上部浸提液的敏感性趋势总体为:GSQHLSKMXNQLKZLZ,最不敏感或有促进作用的是DX垂穗披碱草;对根浸提液的敏感性趋势总体为:QHNQLZKMX,根浸提液对LS、GS、LKZ、DX垂穗披碱草种子萌发生长具有促进作用。紫花苜蓿植株不同部位浸提液对垂穗披碱草种子萌发生长的化感效应顺序为:地上部根。  相似文献

Background and AimsIn Mediterranean ecosystems, the heat shock of wildfire disrupts physical seed dormancy in many plant species. This triggers germination in the post-fire environment where seedling establishment is optimal due to decreased competition and increased resource availability. However, to maintain the soil seed bank until a fire occurs, the minimum heat capable of breaking seed dormancy (i.e. the lower heat threshold) must be above the maximum temperatures typically observed in the soil during the summer. We therefore hypothesized that summer temperatures have shaped heat requirements for physical dormancy release. Specifically, we predicted that seeds from populations growing under warmer summers will have higher values of the lower heat threshold.MethodsTo evaluate this prediction, we collected seeds from two Cistus species in 31 populations (20 Cistus albidus and 11 Cistus salviifolius) along a climate gradient of summer temperatures on the eastern coast of Spain. For each population, seeds were treated to 10 min heat shocks, from 30 to 120 °C in 5 °C increments (19 treatments), to simulate increasing heat doses from summer to fire-related temperatures. Seeds were then germinated in the lab.Key ResultsFor all populations, maximum germination was observed when applying temperatures associated with fire. Lower heat thresholds varied among populations, with a positive relationship between summer temperatures at seed population origin and the heat dose required to break dormancy.ConclusionsOur results suggest that fire drives maximum dormancy release for successful post-fire germination, while summer temperatures determine lower heat thresholds for ensuring inter-fire seed bank persistence. Significant among-population variation of thresholds also suggests that post-fire seeder species have some potential to modify their dormancy release requirements in response to changing climate.  相似文献

Time-to-event analysis represents a collection of relatively new, flexible, and robust statistical techniques for investigating the incidence and timing of transitions from one discrete condition to another. Plant biology is replete with examples of such transitions occurring from the cellular to population levels. However, application of these statistical methods has been rare in botanical research. Here, we demonstrate the use of non- and semi-parametric time-to-event and categorical data analyses to address questions regarding seed to seedling transitions of Ipomopsis rubra propagules exposed to various doses of constant or simulated seasonal diel temperatures. Seeds were capable of germinating rapidly to >90 % at 15–25 or 22/11–29/19 °C. Optimum temperatures for germination occurred at 25 or 29/19 °C. Germination was inhibited and seed viability decreased at temperatures ≥30 or 33/24 °C. Kaplan–Meier estimates of survivor functions indicated highly significant differences in temporal germination patterns for seeds exposed to fluctuating or constant temperatures. Extended Cox regression models specified an inverse relationship between temperature and the hazard of germination. Moreover, temperature and the temperature × day interaction had significant effects on germination response. Comparisons to reference temperatures and linear contrasts suggest that summer temperatures (33/24 °C) play a significant role in differential germination responses. Similarly, simple and complex comparisons revealed that the effects of elevated temperatures predominate in terms of components of seed viability. In summary, the application of non- and semi-parametric analyses provides appropriate, powerful data analysis procedures to address various topics in seed biology and more widespread use is encouraged.  相似文献

Nucleotide variation in chloroplast Asp(GUC)–Thr(GGU) intergenic region and genetic relationships among this group were examined among Elymus trachycaulus complex, Elymus alaskanus and Elymus caninus. The estimates of nucleotide diversity (π) ranged from 0.00111 for Elymus virescens to 0.03086 for E. caninus. Highest nucleotide diversity was found for E. caninus among the taxa analyzed here and followed by Elymus hyperarcticus. E. virescens accessions are genetically very uniform. Phylogenetic analysis suggested that E. caninus is paraphyletic. Elymus violaceus is genetically distinct from both E. alaskanus and E. trachycaulus. Our result indicates that Asp (GUC)–Thr (GGU) intergenic region has a high rate of evolutionary in Elymus species. Large indels detected in this region appear to have a highly rate of evolution and are thus more prone to homoplasy. We also first reported a minisatellite discovered in Asp (GUC)–Thr (GGU) region in Elymus species. The minisatellite identified here is an excellent candidate marker for studying population structures of Elymus species.  相似文献

Background and Aims

Across their range, widely distributed species are exposed to a variety of climatic and other environmental conditions, and accordingly may display variation in life history strategies. For seed germination in cold climates, two contrasting responses to variation in winter temperature have been documented: first, an increased ability to germinate at low temperatures (cold tolerance) as winter temperatures decrease, and secondly a reduced ability to germinate at low temperatures (cold avoidance) that concentrates germination towards the warmer parts of the season.

Methods

Germination responses were tested for Calluna vulgaris, the dominant species of European heathlands, from ten populations collected along broad-scale bioclimatic gradients (latitude, altitude) in Norway, covering a substantial fraction of the species'' climatic range. Incubation treatments varied from 10 to 25 °C, and germination performance across populations was analysed in relation to temperature conditions at the seed collection locations.

Key Results

Seeds from all populations germinated rapidly and to high final percentages under the warmer incubation temperatures. Under low incubation temperatures, cold-climate populations had significantly lower germination rates and percentages than warm-climate populations. While germination rates and percentages also increased with seed mass, seed mass did not vary along the climatic gradients, and therefore did not explain the variation in germination responses.

Conclusions

Variation in germination responses among Calluna populations was consistent with increased temperature requirements for germination towards colder climates, indicating a cold-avoidance germination strategy conditional on the temperature at the seeds'' origin. Along a gradient of increasing temperatures this suggests a shift in selection pressures on germination from climatic adversity (i.e. low temperatures and potential frost risk in early or late season) to competitive performance and better exploitation of the entire growing season.  相似文献

观察了披碱草属中小颖组、宽颖组和长颖组主要代表种的叶表皮形态学特征,总结了3组植物叶表皮结构的异同,探讨了叶表皮特征的分类学意义。同时,根据3组植物叶表皮性状的演化趋势,分析了各组的演化关系和系统位置。结果表明,小颖组最原始,宽颖组较进化,长颖组最高级;小颖组可能直接派生了较进化的宽颖组,并在宽颖组的基础上进而产生了最高级的长颖组。小颖组、宽颖组和长颖组的这一系统关系与利用外部形态特征所获得的演化趋势基本一致。  相似文献
