Recent studies on parotid gland flow rate and composition in healthy subjects do not support the conventional wisdom that there is a gradual deterioration in salivary gland function with aging. With gustatory stimulation a diminution in flow rate was observed only in post-menopausal women taking medications. Studies of whole saliva and preliminary studies of submandibular saliva flow rate, however, suggest that these glands may exhibit functional changes not seen in the parotid. This would be consistent with histological findings. Reports to date on age effects on composition suggest modest selective changes in electrolytes (e.g. sodium) and only subtle changes in proteins (e.g. amylase isoenzymes). Clinical concerns (rampant caries, stomatitis, periodontal disease) arise largely because of the high number of elderly on medications or therapeutic interventions that affect salivary flow and the increase in incidence of diseases of the salivary gland (e.g. sialadeneitis, Sjogren's, sarcoidosis). Reduction in flow rate and alterations in composition diminish salivary protective mechanisms, i.e. antibacterial activity, lubrication and protection of soft tissues, and maintenance of hard tissue integrity. Recent research on structure of mucins, the nature of the salivary lipids and interactions among salivary proteins should stimulate a second generation of studies on both the effects of aging per se and aberrations resulting from disease. Stimulation of compromised function and development of more effective salivary substitutes are also important areas of research.  相似文献

In a survey of Streptomyces species, methods were designed and followed that would specifically select strains capable of producing heat-stable, nonpolyenic, antifungal antibiotics. Of 500 strains grown in shaken flasks, 240 of the culture liquors contained active factors as demonstrated by paper-disc assay against Mucor ramannianus. Culture filtrates and mycelial extracts of the active strains were examined by ultraviolet spectrophotometry; 166 were nonpolyenic as determined by absorption spectra. Heat-stability tests of the nonpolyenic antibiotics over a broad pH range revealed that 15 were stable under all test conditions, 70 moderately stable, and 81 unstable. Culture liquors containing stable, nonpolyenic antifungal agents were chromatographed with eight solvent systems in an attempt to identify the antibiotics. The producing cultures were studied by cross-antagonism tests to discover similarities with producers of known antibacterial antibiotics. Two of the antibiotics produced by promising strains were identified as cycloheximide and musarin. Six antibiotics, presumably new, were detected.  相似文献

Several metabolites responsible for the toxic manifestations of Valsa ceratosperma (Toda et Fries) Maire, a phytopathogenic fungus of the Japanese apple canker, have been isolated from its culture filtrate after growth on apple branch extract. Chemical and spectrometric studies revealed the products to be degradation products of phlorizin which is a dominant component distributed in leaves, stems, fruits and roots of apple. The toxic substances were identified as 3-(p-hydroxyphenyl) propionic acid, phloroglucinol, p-hydroxyacetophenone, p-hydroxybenzoic acid and protocatechuic acid. All of these compounds except p-hydroxyacetophenone were detected in the lesions of apple trees infected by V. ceratosperma. The fungus cultivated in a medium containing added phlorizin also produced the five toxic substances mentioned above. These results suggest that phlorizin is involved in the specific relationship between the host and the pathogen, indicating that the degradation products of phlorizin play important roles in the production of symptoms of infected apple trees.  相似文献

Disease susceptibility and resistance are important factors for the conservation of endangered species, including elephants. We analyzed pathology data from 26 zoos and report that Asian elephants have increased neoplasia and malignancy prevalence compared with African bush elephants. This is consistent with observed higher susceptibility to tuberculosis and elephant endotheliotropic herpesvirus (EEHV) in Asian elephants. To investigate genetic mechanisms underlying disease resistance, including differential responses between species, among other elephant traits, we sequenced multiple elephant genomes. We report a draft assembly for an Asian elephant, and defined 862 and 1,017 conserved potential regulatory elements in Asian and African bush elephants, respectively. In the genomes of both elephant species, conserved elements were significantly enriched with genes differentially expressed between the species. In Asian elephants, these putative regulatory regions were involved in immunity pathways including tumor-necrosis factor, which plays an important role in EEHV response. Genomic sequences of African bush, forest, and Asian elephant genomes revealed extensive sequence conservation at TP53 retrogene loci across three species, which may be related to TP53 functionality in elephant cancer resistance. Positive selection scans revealed outlier genes related to additional elephant traits. Our study suggests that gene regulation plays an important role in the differential inflammatory response of Asian and African elephants, leading to increased infectious disease and cancer susceptibility in Asian elephants. These genomic discoveries can inform future functional and translational studies aimed at identifying effective treatment approaches for ill elephants, which may improve conservation.  相似文献
